Chitosan nanoparticles, such as those prepared in this study, may exhibit potential antibacterial activity as their unique character. The purpose of this study was to evaluate the in vitro antibacterial activity of chitosan nanoparticles and copper-loaded nanoparticles against various microorganisms. Chitosan nanoparticles were prepared based on the ionic gelation of chitosan with tripolyphosphate anions. Copper ions were adsorbed onto the chitosan nanoparticles mainly by ion-exchange resins and surface chelation to form copper-loaded nanoparticles. The physicochemical properties of the nanoparticles were determined by size and zeta potential analysis, atomic force microscopy (AFM), FTIR analysis, and XRD pattern. The antibacterial activity of chitosan nanoparticles and copper-loaded nanoparticles against E. coli, S. choleraesuis, S. typhimurium, and S. aureus was evaluated by calculation of minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). Results show that chitosan nanoparticles and copper-loaded nanoparticles could inhibit the growth of various bacteria tested. Their MIC values were less than 0.25 microg/mL, and the MBC values of nanoparticles reached 1 microg/mL. AFM revealed that the exposure of S. choleraesuis to the chitosan nanoparticles led to the disruption of cell membranes and the leakage of cytoplasm.  相似文献

聚羟基脂肪酸酯(polyhydroxyalkanoate)PHA 纳米微球是很多微生物在营养失衡的情况下,在体内合成的一种可生物降解的细胞内聚酯,主要作为微生物的碳源及能量储备。天然 PHA 微球的内部是由疏水的聚酯链构成的疏水核心,其外层是由磷脂界膜及膜上嵌入或附着的包括 PHA合酶 PhaC 和 PHA 颗粒相关蛋白 PhaP 等蛋白构成的边界层。PhaC 通过共价键连接在PHA微球表面,而 PhaP 通过疏水相互作用吸附在 PHA 微球表面。通过将外源性功能蛋白与 PhaC 或 PhaP 进行融合表达,在重组微生物体内就能直接合成表面带有功能蛋白的纳米微球复合体。由于该纳米微球在微生物细胞内是以独立的包涵体形式存在,因此通过细胞破碎及离心等方法就能简便、有效地使其从细胞中分离并得以纯化。鉴于 PHA 微球这种表面易被修饰改造的特性,越来越多的功能蛋白通过与 PHA 微球表面蛋白(PhaC 或 PhaP)的融合表达,呈递在了 PHA 微球表面,使其成为一种廉价、高效的蛋白固定化及呈递的新技术。本文在介绍了 PHA 微球的结构特性及生物合成的基础上,着重综述了目前关于功能化 PHA 微球在蛋白纯化、固定化酶、生物分离、靶向递药、疾病诊断、成像技术及新型疫苗开发方面的研究现状及其未来在生物医药等领域的广泛应用前景。  相似文献

Magnetic core shell nanoparticles are composed of a highly magnetic core material surrounded by a thin shell of desired drug, polymer or metal oxide. These magnetic core shell nanoparticles have a wide range of applications in biomedical research, more specifically in tissue imaging, drug delivery and therapeutics. The present review discusses the up-to-date knowledge on the various procedures for synthesis of magnetic core shell nanoparticles along with their applications in cancer imaging, drug delivery and hyperthermia or cancer therapeutics. Literature in this area shows that magnetic core shell nanoparticle-based imaging, drug targeting and therapy through hyperthermia can potentially be a powerful tool for the advanced diagnosis and treatment of various cancers.  相似文献

Biogenic synthesis of metal nanoparticles has been well proved by using bacteria, fungi, algae, actinomycetes, plants, etc. Among the different microorganisms used for the synthesis of metal nanoparticles, actinomycetes are less known. Although, there are reports, which have shown that actinomycetes are efficient candidates for the production of metal nanoparticles both intracellularly and extracellularly. The nanoparticles synthesized by the members of actinomycetes present good polydispersity and stability and possess significant biocidal activities against various pathogens. The present review focuses on biological synthesis of metal nanoparticles and their application in medicine. In addition, the toxicity of these biogenic metal nanoparticles to human beings and environment has also been discussed.  相似文献

Antimicrobial activity of silver nanoparticles is gaining importance due its broad spectrum of targets in cell compared to conventional antimicrobial agents. In this context, a UV photo-reduction method was used for the synthesis and the nanoparticles were characterized by UV–Visible spectroscopy, transmission electron microscopy, atomic force microscopy and thermogravimetric analysis techniques. The antibacterial activity of the synthesized silver nanoparticles was evaluated both in liquid and solid growth media employing various susceptibility assays on Pseudomonas aeruginosa, a ubiquitous bacterium. The dose dependent growth suppression by nanoparticles was studied with well diffusion method. By broth dilution method, the minimum inhibitory concentration (MIC) was found to be 2 μg/ml. It was observed that the bactericidal effect depends both on nanoparticle concentration and number of bacteria present. In our study, we could demonstrate the complete antibiofilm activity of silver nanoparticles at a concentration as low as 1 μg/ml. Our observations substantiated the association of reactive oxygen species and cell membrane damage in the antibacterial mechanism of silver nanoparticles. Our findings suggested that these nanoparticles can be exploited towards the development of potential antibacterial coatings for various biomedical and environmental applications.  相似文献

Nanotechnology is a field that is burgeoning day by day, making an impact in all spheres of human life. Biological methods of synthesis have paved way for the “greener synthesis” of nanoparticles and these have proven to be better methods due to slower kinetics, they offer better manipulation and control over crystal growth and their stabilization. This has motivated an upsurge in research on the synthesis routes that allow better control of shape and size for various nanotechnological applications. Nanosilver has developed as a potent antibacterial, antifungal, anti-viral and anti-inflammatory agent. The recent advancement in the field includes the enzymatic method of synthesis suggesting enzymes to be responsible for the nanoparticle formation. The biomedical applications of silver nanoparticle can be effective by the use of biologically synthesized nanoparticles which minimize the factors such as toxicity and cost and are found to be exceptionally stable. The targeting of cancer cells using silver nanoparticles has proven to be effective, but neither the exact mechanism of action nor the modes of activation of the downstream signaling molecules have been revealed yet. The review illustrates a probable signaling pathway and mechanism by which silver nanoparticles target the cancer cells. The current review also examines the historical background of nanoparticles, role of silver nanoparticles in various biomedical applications and also focusing on better methods of the synthesis of nanoparticles.  相似文献

The use of nanotechnology in medicine and more specifically drug delivery is set to spread rapidly. Currently many substances are under investigation for drug delivery and more specifically for cancer therapy. Interestingly pharmaceutical sciences are using nanoparticles to reduce toxicity and side effects of drugs and up to recently did not realize that carrier systems themselves may impose risks to the patient. The kind of hazards that are introduced by using nanoparticles for drug delivery are beyond that posed by conventional hazards imposed by chemicals in classical delivery matrices. For nanoparticles the knowledge on particle toxicity as obtained in inhalation toxicity shows the way how to investigate the potential hazards of nanoparticles. The toxicology of particulate matter differs from toxicology of substances as the composing chemical(s) may or may not be soluble in biological matrices, thus influencing greatly the potential exposure of various internal organs. This may vary from a rather high local exposure in the lungs and a low or neglectable exposure for other organ systems after inhalation. However, absorbed species may also influence the potential toxicity of the inhaled particles. For nanoparticles the situation is different as their size opens the potential for crossing the various biological barriers within the body. From a positive viewpoint, especially the potential to cross the blood brain barrier may open new ways for drug delivery into the brain. In addition, the nanosize also allows for access into the cell and various cellular compartments including the nucleus. A multitude of substances are currently under investigation for the preparation of nanoparticles for drug delivery, varying from biological substances like albumin, gelatine and phospholipids for liposomes, and more substances of a chemical nature like various polymers and solid metal containing nanoparticles. It is obvious that the potential interaction with tissues and cells, and the potential toxicity, greatly depends on the actual composition of the nanoparticle formulation. This paper provides an overview on some of the currently used systems for drug delivery. Besides the potential beneficial use also attention is drawn to the questions how we should proceed with the safety evaluation of the nanoparticle formulations for drug delivery. For such testing the lessons learned from particle toxicity as applied in inhalation toxicology may be of use. Although for pharmaceutical use the current requirements seem to be adequate to detect most of the adverse effects of nanoparticle formulations, it can not be expected that all aspects of nanoparticle toxicology will be detected. In this review, we highlight our recent achievements in using colloidal gold nanoparticles as building blocks for fabrication of anisotropic and multicomponent nanoparticles (e.g., nanoshells, semiconductor nanocrystals, and gold nanorods). The use of titanium dioxide (TiO2) in various industrial applications (eg, production of paper, plastics, cosmetics, and paints) has been expanding thereby increasing the occupational and other environmental exposure of these nanoparticles to humans and other species. However, the health effects of exposure to TiO2 nanoparticles have not been systematically assessed even though recent studies suggest that such exposure induces inflammatory responses in lung tissue and cells. Because the effects of such nanoparticles on human neural cells are unknown, we have determined the putative cytotoxic effects of these nanoparticles on human astrocytes-like astrocytoma U87 cells and compared their effects on normal human fibroblasts. We found that TiO2 micro- and nanoparticles induced cell death on both human cell types in a concentration-related manner. We further noted that zinc oxide (ZnO) nanoparticles were the most effective, TiO2 nanoparticles the second most effective, and magnesium oxide (MgO) nanoparticles the least effective in inducing cell death in U87 cells. The cell death mechanisms underlying the effects of TiO2 micro- and nanoparticles on U87 cells include apoptosis, necrosis, and possibly apoptosis-like and necrosis-like cell death types. Thus, our findings may have toxicological and other pathophysiological implications on exposure of humans and other mammalian species to metallic oxide nanoparticles.  相似文献

Mycoendophytes are the fungi that occur inside the plant tissues without exerting any negative impact on the host plant. They are most frequently isolated endophytes from the leaf, stem, and root tissues of various plants. Among all fungi, the mycoendophytes as biosynthesizer of noble metal nanoparticles (NPs) are less known. However, some reports showing efficient synthesis of metal nanoparticles, mainly silver nanoparticles and its remarkable antimicrobial activity against bacterial and fungal pathogens of humans and plants. The nanoparticles synthesized from mycoendophytes present stability, polydispersity, and biocompatibility. These are non-toxic to humans and environment, can be gained in an easy and cost-effective manner, have wide applicability and could be explored as promising candidates for a variety of biomedical, pharmaceutical, and agricultural applications. Mycogenic silver nanoparticles have also demonstrated cytotoxic activity against cancer cell lines and may prove to be a promising anticancer agent. The present review focuses on the biological synthesis of metal nanoparticles from mycoendophytes and their application in medicine. In addition, different mechanisms of biosynthesis and activity of nanoparticles on microbial cells, as well as toxicity of these mycogenic metal nanoparticles, have also been discussed.  相似文献

The purpose of this study was to outline the effects of interactions between a model drug and various acrylic polymers on the physical properties of nanoparticles prepared by an aerosol flow reactor method. The amount of model drug, ketoprofen, in the nanoparticles was varied, and the nanoparticles were analyzed for particle size distribution, particle morphology, thermal properties, IR spectroscopy, and drug release. The nanoparticles produced were spherical, amorphous, and had a matrix-type structure. Ketoprofen crystallization was observed when the amount of drug in Eudragit L nanoparticles was more than 33% (wt/wt). For Eudragit E and Eudragit RS nanoparticles, the drug acted as an effective plasticizer resulting in lowering of the glass transition of the polymer. Two factors affected the preparation of nanoparticles by the aerosol flow reactor method, namely, the solubility of the drug in the polymer matrix and the thermal properties of the resulting drug-polymer matrix.  相似文献

Active application of gold nanoparticles for various diagnostic and therapeutic purposes started in recent decades due to the emergence of new data on their unique optical and physicochemical properties. In addition to colloidal gold conjugates, growth in the number of publications devoted to the synthesis and application of multifunctional nanocomposites has occurred in recent years. This review considers the application in biomedicine of multifunctional nanoparticles that can be produced in three different ways. The first method involves design of composite nanostructures with various components intended for either diagnostic or therapeutic functions. The second approach uses new bioconjugation techniques that allow functionalization of gold nanoparticles with various molecules, thus combining diagnostic and therapeutic functions in one medical procedure. Finally, the third method for production of multifunctional nanoparticles combines the first two approaches, in which a composite nanoparticle is additionally functionalized by molecules having different properties.  相似文献

Despite the vast research being conducted on the development of biosynthetic procedures, the process is limited owing to the unavailability of modes to control the size and shape of the biosynthesized nanoparticles. In this study, we investigate the size and shape control of gold nanoparticles synthesized by leaf extract of Piper betle (PBE). The effects of various counter ions, temperatures, pH and reaction times on the morphology of gold nanoparticles are also scrutinized. Results from this study indicate that the presence of iodine during biosynthesis leads to the formation of spherical gold nanoparticles and induces the presence of bromine-emanating, truncated nanoplatelets. Spherical nanoparticles are formed with increasing incubation temperature. pH 3 was found to be the optimum for nanoparticles synthesis. The presence of phosphates, sulphates and nitrates increases the productivity of nanoparticles. ICP analysis revealed complete reduction of AuCl4 ions within 48 h of the reaction. The use of plant extract for rapid synthesis represents a novel and environmentally friendly approach for the fabrication of gold nanoparticles and nanoplatelets, as an alternative to chemical methods.  相似文献

Nanotechnology is an emerging branch of science, which has potential to solve many problems in different fields. The union of nanotechnology with other fields of sciences including physics, chemistry, and biology has brought the concept of synthesis of nanoparticles from their respective metals. Till date, many types of nanoparticles have been synthesized and being used in different fields for various applications. Moreover, copper nanoparticles attract biologists because of their significant and broad-spectrum bioactivity. Due to the large surface area to volume ratio, copper nanoparticles have been used as potential antimicrobial agent in many biomedical applications. But the excess use of any metal nanoparticles increase the chance of toxicity to humans, other living beings, and environment. In this article, we have critically reviewed the bioactivities and cytotoxicity of copper nanoparticles. We report a new method for multistep peptide synthesis on polymeric nanoparticles of differing sizes. Polymeric nanoparticles were functionalized via their temporary embedment into a magnetic inorganic matrix that allows multistep peptide synthesis. The matrix is removed at the end of the process for obtaining nanoparticles functionalized with peptides. The medicinal and physicochemical properties of nanoscale materials are strong functions of the particle size and the materials used in their synthesis. The nanoparticle shape also contributes significantly to their medicinal properties. Several shapes ranging from oval, spherical, rods, to teardrop structures may be obtained by chemical methods. Triangular and hexagonal nanoparticles have been synthesized by using a pine cone extract (PCE). Here, we report the discovery that PCE, when reacted with silver nitrate ions, yields a high percentage of thin, flat, single-crystalline nanohexagonal and nanotriangular silver nanoparticles. The nanohexagonal and nanotriangular nanoparticles appear to grow by a process involving rapid reduction with assembly at room temperature at a high pH. The nanoparticles were characterized by UV–Vis absorption spectroscopy, SEM-EDS, TEM, FTIR, and X-ray diffraction analyses. The anisotropy of the nanoparticle shape results in large near-infrared absorption by the particles. Highly anisotropic particles are applicable in various fields, including agriculture and medicine. The obtained silver nanoparticles (Ag NPs) had significant antibacterial action on both Gram classes of bacteria associated with agriculture. Because the Ag NPs are encapsulated with functional group-rich PCE, they can be easily integrated in various applications.  相似文献

Gamma-polyglutamic acid (gamma-PGA) is a hydrophilic, biodegradable, and naturally available biopolymer produced by a number of microbial species, most commonly, the Bacillaceae family. Its biological properties such as nontoxicity, biocompatibility, and nonimmunogenicity qualify it as an important biomaterial in drug delivery applications. This review focuses mainly on the development of gamma-PGA nanoparticles as drug delivery carriers for anticancer therapeutics. We discuss various techniques for the production and characterization of gamma-PGA nanoparticles and controlled-release strategies. We also present a brief overview of the tumor physiology that forms the basis for the development of various targeted drug delivery approaches in cancer chemotherapy.  相似文献

Abstract

A great deal of research has been done on various uses of copper oxide. The synthesis of copper oxide nanoparticles was mediated using tea extract. The first sign of the reduction of copper ions to copper oxide was the change in color of extract to dark brown after treating with copper chloride. The resulting nanoparticles were characterized using X-ray diffraction (XRD) and Fourier transform infrared spectrometry (FTIR). Finally, the antimicrobial effects of these nanoparticles on Fusarium solani were studied in vitro by agar dilution method. The TEM images showed the synthesis of copper oxide with size of less than 80?nm. The synthesized copper oxide nanoparticles showed significant inhibitory effects on F. solani cultures so that the concentration of 80?μg/ml prevented approximately 90% of the mycelium growth of the fungus. The results showed that the inhibition zone of silver nanoparticles strongly depends on their concentration and increases by increasing the concentration of copper oxide nanoparticles in the medium.  相似文献

We developed a facile and quick ethanol-based method for preparing silk nanoparticles and then fabricated a biodegradable and biocompatible dual-drug release system based on silk nanoparticles and the molecular networks of silk hydrogels. Model drugs incorporated in the silk nanoparticles and silk hydrogels showed fast and constant release, respectively, indicating successful dual-drug release from silk hydrogel containing silk nanoparticles. The release behaviors achieved by this dual-drug release system suggest to be regulated by physical properties (e.g., β-sheet contents and size of the silk nanoparticles and network size of the silk hydrogels), which is an important advantage for biomedical applications. The present silk-based system for dual-drug release also demonstrated no significant cytotoxicity against human mesenchymal stem cells (hMSCs), and thus, this silk-based dual-drug release system has potential as a versatile and useful new platform of polymeric materials for various types of dual delivery of bioactive molecules.  相似文献

Chitosan nanoparticles have been synthesized as potential anticancer agents, and evaluated, in vitro, against various cancer cell lines. In this study, in vivo antitumor activity of chitosan nanoparticles against Sarcoma-180 and mouse hepatoma H22 was investigated. Chitosan nanoparticles showed significant antitumor activity in vivo.
