DNA barcoding is used in a variety of ecological applications to identify organisms, including partially digested prey items from diet samples. That particular application can enhance the ability to characterize diet and predator–prey dynamics but is problematic when genetic sequences of prey match those of consumer species (i.e., self-DNA). Such a result may indicate cannibalism, but false positives can result from contamination of degraded prey samples with consumer DNA. Here, nuclear-encoded microsatellite markers were used to genotype invasive lionfish, Pterois volitans, consumers and their prey (n?=?80 pairs) previously barcoded as lionfish. Cannibalism was confirmed when samples exhibited two or more different alleles between lionfish and prey DNA across multiple microsatellite loci. This occurred in 26.2% of all samples and in 42% of samples for which the data were considered conclusive. These estimates should be considered conservative given rigorous assignment criteria and low allelic diversity in invasive lionfish populations. The highest incidence of cannibalism corresponded to larger sized consumers from areas with high lionfish densities, suggesting cannibalism in northern Gulf of Mexico lionfish is size- and density-dependent. Cannibalism has the potential to influence population dynamics of lionfish which lack native western Atlantic predators. These results also have important implications for interpreting DNA barcoding analysis of diet in other predatory species where cannibalism may be underreported.  相似文献

The opportunistic feeder Pterois volitans is a voracious invader, causing large impacts in marine food-webs. We have used a Ecopath-with-Ecosim model to hypothesize an invasion by lionfish and to predict the likely impact of this potential generalist mesopredator in a subtropical food-web model. With thirty-three functional groups, the initial Ecopath model was balanced with a low biomass of lionfish (0.07 t/km2). In Ecosim, three scenarios of different vulnerability settings for the linkages between the introduced fish and its prey were tested, representing the default setting, a top-down control, and an extreme top-down control. The scenarios were tested using different assumptions on the ability of the invasive fish to change the proportions of prey consumed according to prey availability. Our model predicted that the hypothesized lionfish invasion would have a strong impact on this subtropical marine food web: (1) by reducing prey populations and, consequently, food for native predators, and; (2) by predating on key species, causing direct impacts and possibly cascading trophic effects. Reef fish were the most affected, including some groups ecologically and economically important, like lutjanids and groupers. However, some adaptations in the fishing strategy of fishermen are expected which may affect other fish groups. Stakeholders should be warned of the potential ecological and socio-economic impacts that may arise from a lionfish invasion and various strategies and policy options should be immediately developed and applied (1) to prevent the arrival and establishment of the lionfish, and; (2) to make the ecosystem more resilient to this and other possible exotic species.  相似文献

Feeding ecology of the lionfish (Pterois volitans), an invasive species in the Western North Atlantic, was examined by collecting stomach content data from fishes taken throughout the Bahamian archipelago. Three relative metrics of prey quantity, including percent number, percent frequency, and percent volume, were used to compare three indices of dietary importance. Lionfish largely prey upon teleosts (78% volume) and crustaceans (14% volume). Twenty-one families and 41 species of teleosts were represented in the diet of lionfish; the top 10 families of dietary importance were Gobiidae, Labridae, Grammatidae, Apogonidae, Pomacentridae, Serranidae, Blenniidae, Atherinidae, Mullidae, and Monacanthidae. The proportional importance of crustaceans in the diet was inversely related to size with the largest lionfish preying almost exclusively on teleosts. Lionfish were found to be diurnal feeders with the highest predation occurring in the morning (08:00–11:00).  相似文献

Mesophotic coral reefs (30–150 m) have been assumed to be physically and biologically connected to their shallow-water counterparts, and thus may serve as refugia for important taxonomic groups such as corals, sponges, and fish. The recent invasion of the Indo–Pacific lionfish (Pterois volitans) onto shallow reefs of the Caribbean and Bahamas has had significant, negative, effects on shallow coral reef fish populations. In the Bahamas, lionfish have extended their habitat range into mesophotic depths down to 91 m where they have reduced the diversity of several important fish guilds, including herbivores. A phase shift to an algal dominated (>50% benthic cover) community occurred simultaneously with the loss of herbivores to a depth of 61 m and caused a significant decline in corals and sponges at mesophotic depths. The effects of this invasive lionfish on mesophotic coral reefs and the subsequent changes in benthic community structure could not be explained by coral bleaching, overfishing, hurricanes, or disease independently or in combination. The significant ecological effects of the lionfish invasion into mesophotic depths of coral reefs casts doubt on whether these communities have the resilience to recover themselves or contribute to the recovery of their shallow water counterparts as refugia for key coral reef taxa.  相似文献

Less than a decade after being observed off Florida, the invasive Indo-Pacific lionfish is now widely distributed off the southeast coast of the United States. As a step towards measuring invasion impacts to native communities, we examine the magnitude and extent of this invasion by first, compiling reports of lionfish to provide range information and second, estimate lionfish abundance from two separate studies. We also estimate native grouper (epinepheline serranids) abundance to better assess and compare lionfish abundances. In the first study we conducted SCUBA diver visual transect surveys at 17 different locations off the North Carolina coast in water depths of 35–50 m. In the second study, we conducted 27 Remote Operated Vehicle (ROV) transect surveys at five locations from Florida to North Carolina in water depths of 50–100 m. In both studies, lionfish were found to be second in abundance only to scamp (Mycteroperca phenax). Lionfish were found in higher abundance in the shallower North Carolina SCUBA surveys ( ha−1) than in the deep water ROV surveys ( ha−1). Lionfish reports continue to expand most recently into the Bahamas, raising the specter of further spread into the Caribbean and Gulf of Mexico. The potential impacts of lionfish to native communities are likely to be through direct predation, competition and overcrowding. The high number of lionfish present in the ecosystem increases the potential for cascading impacts throughout the food chain. Within the southeast region the combined effects of climate change, overfishing and invasive species may have irreversible consequences to native communities in this region. Larval duration of the non-native Indo-Pacific lionfish was estimated from samples collected along the Bahamian Archipelago using sagittal otoliths. Settlement marks, characterized by daily growth increments with reduced coloration, less definitive margins, and a re-orientation of the growth axes and otolith shape, were determined for 28 individuals. Settlement age was between 20 and 35 days with a mean of 26.2 days. Comparisons of settlement age to other littoral and reef fish species suggest that lionfish settlement age is moderate to relatively low. Lionfish pelagic larval duration is apparently sufficient to allow their rapid establishment and wide geographic range in the western Atlantic and Caribbean.  相似文献

Lionfish (Pterois miles) were observed avoiding coral pinnacles inhabited by the moray eels Gymnothorax flavimarginatus and G. javanicus in the northern Red Sea, Egypt. Release of lionfish (Standard Length 93–104 mm) in such coral pinnacles in November 2016 resulted in almost immediate predation by large moray eels (Total Length > 1 m). Predation by moray eels may be the key control mechanism of population growth in the native biogeographical range of Pterois spp. and may indirectly explain the success of the invasive populations. This is the first video-documented record of moray eels feeding on the lionfish P. miles.  相似文献

Round Goby Neogobius melanostomus invasion of the Grand River (Ontario, Canada) presents an opportunity to assess the role of abiotic gradients in mediating the establishment and impact of nonnative benthic fishes in rivers. In this system, sequential low-head dams delineate uninvaded and invaded river reaches and create upstream gradients of increasing water velocity. We hypothesized that flow refugia created by impounded reservoirs above low-head dams enhance local Round Goby abundance. Round Goby influence on the native fish community was determined by variance partitioning, and we used generalized additive models to identify small-bodied benthic fish species most likely to be impacted by Round Goby invasion. Round Goby abundance declined as the degree of reservoir effect decreased upstream. The distributions of four species (including the endangered Eastern Sand Darter Ammocrypta pellucida) in invaded reaches were best explained by inclusion of both reservoir-associated abiotic variables and Round Goby abundance as model terms. To determine establishment potential of the uninvaded reach immediately upstream, four environmental habitat characteristics were used in discriminant function analysis (DFA) to predict three potential outcomes of introduction: non-invaded and either lower or higher Round Goby abundance (low and high invasion status, respectively) than the median number of Round Goby at invaded sites. Our DFA function correctly classified non-invaded and high-abundance invasion status sites > 85% of the time, with lower (73%) success in classifying low-abundance invasion status sites, and the spatial pattern of our results suggest that likelihood of establishment is greatest in impounded habitat.  相似文献

The enemy release hypothesis (ERH), which has been the theoretical basis for classic biological control, predicts that the success of invaders in the introduced range is due to their release from co-evolved natural enemies (i.e. herbivores, pathogens and predators) left behind in the native range. We tested this prediction by comparing herbivore pressure on native European and introduced North American populations of Hypericum perforatum (St Johns Wort). We found that introduced populations occur at larger densities, are less damaged by insect herbivory and suffer less mortality than populations in the native range. However, overall population size was not significantly different between ranges. Moreover, on average plants were significantly smaller in the introduced range than in the native range. Our survey supports the contention that plants from the introduced range experience less herbivore damage than plants from the native range. While this may lead to denser populations, it does not result in larger plant size in the introduced versus native range as postulated by the ERH.  相似文献

Two repeated DNA sequences isolated from a partial genomic DNA library of Helianthus annuus, p HaS13 and p HaS211, were shown to represent portions of the int gene of a Ty3 /gypsy retroelement and of the RNase-Hgene of a Ty1 /copia retroelement, respectively. Southern blotting patterns obtained by hybridizing the two probes to BglII- or DraI-digested genomic DNA from different Helianthus species showed p HaS13 and p HaS211 were parts of dispersed repeats at least 8 and 7 kb in length, respectively, that were conserved in all species studied. Comparable hybridization patterns were obtained in all species with p HaS13. By contrast, the patterns obtained by hybridizing p HaS211 clearly differentiated annual species from perennials. The frequencies of p HaS13- and p HaS211-related sequences in different species were 4.3x10(4)-1.3x10(5) copies and 9.9x10(2)-8.1x10(3) copies per picogram of DNA, respectively. The frequency of p HaS13-related sequences varied widely within annual species, while no significant difference was observed among perennial species. Conversely, the frequency variation of p HaS211-related sequences was as large within annual species as within perennials. Sequences of both families were found to be dispersed along the length of all chromosomes in all species studied. However, Ty3 /gypsy-like sequences were localized preferentially at the centromeric regions, whereas Ty1/ copia-like sequences were less represented or absent around the centromeres and plentiful at the chromosome ends. These findings suggest that the two sequence families played a role in Helianthusgenome evolution and species divergence, evolved independently in the same genomic backgrounds and in annual or perennial species, and acquired different possible functions in the host genomes.  相似文献

Background

A mismatch has emerged between models and data of host-parasite evolution. Theory readily predicts that parasites can promote host diversity through mechanisms such as disruptive selection. Yet, despite these predictions, empirical evidence for parasite-mediated increases in host diversity remains surprisingly scant.  相似文献

Studying Pneumocystis has proven to be a challenge from the perspective of propagating a significant amount of the pathogen in a facile manner. The study of several fungal pathogens has been aided by the use of invertebrate model hosts. Our efforts to infect the invertebrate larvae Galleria mellonella with Pneumocystis proved futile since P. murina neither caused disease nor was able to proliferate within G. mellonella. It did, however, show that the pathogen could be rapidly cleared from the host.  相似文献

A genetic transformation system has been developed for callus cells of Crataegus aronia using Agrobacterium tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with 5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. Putative transgenic calli were obtained 4 weeks after incubation of the co-cultivated explants onto maintenance medium supplemented with 50 mg l−1 hygromycin. Molecular analysis confirmed the integration of the transgenes in transformed callus. To our knowledge, this is the first time to report an Agrobacterium-mediated transformation system in Crataegus aronia.  相似文献

In the classical taxonomy, three Scomber species are distinguished: S. scombrus, S. australasicus, and S. japonicus. Yet, some fish taxonomists have recently recognized Scomber colias, inhabiting the Atlantic Ocean, as a separate species from S. japonicus, distributed in the Pacific Ocean. Such proposal was based on significant mitochondrial DNA divergence as well as great phenotypic variation among individuals from these two ocean basins. However, in the absence of nuclear DNA data this issue remains still controversial. In this study, a phylogenetic analysis of nuclear 5S rDNA sequences was performed. A total of 30 individuals of S. colias collected in the Atlantic and 34 specimens of S. japonicus from the Pacific were characterized. Moreover, nine individuals of Pacific S. australasicus and eight of Atlantic S. scombrus were included. Maximum likelihood, maximum parsimony, and neighbor-joining analyses revealed the presence of two well-supported distinct clades corresponding to S. colias and S. japonicus, respectively. Altogether, morphologic and genetic data are in agreement with the recognition of two different species, S. colias in the Atlantic, and S. japonicus in the Pacific.  相似文献

The maT clade of transposons is a group of transposable elements intermediate in sequence and predicted protein structure to mariner and Tc transposons, with a distribution thus far limited to a few invertebrate species. We present evidence, based on searches of publicly available databases, that the nematode Caenorhabditis briggsae has several maT-like transposons, which we have designated as CbmaT elements, dispersed throughout its genome. We also describe two additional transposon sequences that probably share their evolutionary history with the CbmaT transposons. One resembles a fold back variant of a CbmaT element, with long (380-bp) inverted terminal repeats (ITRs) that show a high degree (71%) of identity to CbmaT1. The other, which shares only the 26-bp ITR sequences with one of the CbmaT variants, is present in eight nearly identical copies, but does not have a transposase gene and may therefore be cross mobilised by a CbmaT transposase. Using PCR-based mobility assays, we show that CbmaT1 transposons are capable of excising from the C. briggsae genome. CbmaT1 excised approximately 500 times less frequently than Tcb1 in the reference strain AF16, but both CbmaT1 and Tcb1 excised at extremely high frequencies in the HK105 strain. The HK105 strain also exhibited a high frequency of spontaneous induction of unc-22 mutants, suggesting that it may be a mutator strain of C. briggsae.  相似文献
