Integrated biosystem is becoming a major aspect of wastewater management practice. Microbial communities in piggery wastewater sampled from anaerobic (thermophilic and mesophilic) and aerobic digesters (algal tanks) during waste remediation were analyzed by culture-independent techniques based on polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE). The use of Muyzer's 314F-GC, 518R bacterial primers, and archaeal A934F, 1309R primers followed by partial 16s rDNA sequence analysis of the main bands from DGGE revealed the presence of unknown and as yet uncultured microorganisms but also showed functional and ecologically significant denitrifying, acetogenic bacteria along with autotrophic, hydrogenotrophic, and acetoclastic methanogen archaea. Thermophilic digesters were dominated by γ-Proteobacteria, Methanothermobacter sp., while mesophilic digesters showed dominance by Firmicutes, uncultured bacteria, Methanosarcina, and Methanoculleus genera. Under aerobic conditions within algal tanks, pH rose from 7.17 to 9.32, with a significant decrease in total ammonia nitrogen, chemical oxygen demand, and soluble phosphorus levels. PCR-DGGE proved a useful tool for investigating the dynamics of microbial community in the bio-processing of piggery wastewater. Knowledge of the microbial communities involved in digestion of piggery wastewater will allow optimization of integrated biosystem by removing the main pollutants like inorganic ammonium-nitrogen, phosphorus, and pathogens from intensive farming system.  相似文献

Microorganisms exist in nature as members of complex, mixed communities. The microbial communities in industrial wastewater bioreactors can be used as model systems to study the evolution of new metabolic pathways in natural ecosystems. The evolution of microbial metabolic capability in these bioreactors is presumably analogous to phenomena that occur in natural ecosystems. The microorganisms in these bioreactors compete for different carbon sources and constantly have to evolve new metabolic capabilities for survival. Thus, industrial bioreactors should be a rich source of novel biocatalysts.  相似文献

Over the past decades, biological treatment of metallurgical wastewaters has become commonplace. Passive systems require intensive land use due to their slow treatment rates, do not recover embedded resources and are poorly controllable. Active systems however require the addition of chemicals, increasing operational costs and possibly negatively affecting safety and the environment. Electrification of biological systems can reduce the use of chemicals, operational costs, surface footprint and environmental impact when compared to passive and active technologies whilst increasing the recovery of resources and the extraction of products. Electrification of low rate applications has resulted in the development of bioelectrochemical systems (BES), but electrification of high rate systems has been lagging behind due to the limited mass transfer, electron transfer and biomass density in BES. We postulate that for high rate applications, the electrification of bioreactors, for example, through the use of electrolyzers, may herald a new generation of electrified biological systems (EBS). In this review, we evaluate the latest trends in the field of biometallurgical and microbial-electrochemical wastewater treatment and discuss the advantages and challenges of these existing treatment technologies. We advocate for future research to focus on the development of electrified bioreactors, exploring the boundaries and limitations of these systems, and their validity upon treating industrial wastewaters.  相似文献

The length of the favourable season determines voltinism in insect populations. In some insects, there is variation in fecundity and timing of reproduction among females. If the length of the favourable season does not allow all offspring to develop into adults without diapause, the benefits of high early fecundity may outweigh the associated cost of low lifetime fecundity. We tested this by exploring mating frequencies of Pieris napi females along a latitudinal gradient in different generations. Pieris napi is a bivoltine butterfly, and genetically polyandrous females enjoy higher lifetime fecundity than monandrous ones. Polyandry is, however, coupled with a relatively low early fecundity. We found that monandrous females are more likely to produce an additional generation than polyandrous ones under conditions that allow production of only a partial summer generation. Monandrous females were also the first to emerge and slightly over-represented in the summer generation under conditions that allow the development of a complete summer generation. Further, a stochastic model shows that variation in the timing of reproduction between strategies is sufficient to explain the observed patterns. Thus, seasonality may counter-select against polyandry, or more generally against low early reproductive rate, and promote maintenance of polymorphism in life history strategies.  相似文献

We present in this paper a qualitative method to validate and monitor the structure of a non-linear model with respect to experimental data, under some hypotheses. This method is broadly independent of the analytical formulation of the model, and depends only on the qualitative structure (the signs of the Jacobian matrix). The temporal sequences of the extrema of a filtered experimental signal are compared with the transitions allowed by a graph. In particular, we show that the usual moving average of the outputs follows this transition graph. We apply this method to compare models of algal growth in a bioreactor with experimental data.  相似文献

The diversity of grazers on savannahs exerts pressure on the grass to provide nutrition. In this study the concentrations of major and trace animal nutrients in above ground grass tissue as influenced by grazing were examined. Two palatable grass species grazed by both livestock and wild grazers were sampled: Brachiaria nigropedata and Eragrostis lehmanniana. Samples of grass tissue were collected from widely spread sampling sites in both livestock and wildlife grazing sites, at the end of the growing season. Although B. nigropedata had higher nutrient concentrations, there was general covariance in nutrient levels in the two grasses. The covariance indicated that the respective sites generally had location context-dependent low or high nutrient concentrations in all grasses. The adequacy of the nutrient concentrations for grazer nutrition was assessed using literature-sourced minimum and maximum tolerable limit requirements of cattle and the high population gazelle-like species as illustration. The results showed that, in these nitrogen-limited savannahs, the grass generally met grazer requirements in terms of crude protein, potassium, molybdenum, nickel and zinc but that it had deficiencies in phosphorous, sodium, and boron, as well as grazing intensity-related localised toxic levels of potassium, phosphorous, magnesium, iron and manganese. The deficiencies and toxicities were inferred to result in abnormalities in grazer physical condition and reproduction. Given the importance of livestock in semiarid savannahs, these effects on livestock grazers were inferred as threatening human food security. The study shows that high grazing intensity can raise the concentrations of some animal nutrients in above ground grass tissue to levels above grazer tolerable limits. Grazing rotation to reduce grazing intensity is recommended as a proactive grazing management strategy to limit the effects.  相似文献

It has been argued that one of the best ways to conserve biological diversity is to maintain the integrity of functional processes within communities, and this can be accomplished by assessing how much ecological redundancy exists in communities. Evidence suggests, however, that the functional roles species play are subject to the influences of local environmental conditions. Species may appear to perform the same function (i.e. be redundant) under a restricted set of conditions, yet their functional roles may vary in naturally heterogeneous environments. Incorporating the environmental context into ecological experiments would provide a critical perspective for examining functional redundancy among species.  相似文献

Optimization of a fixed-bed bioreactor used in wastewater treatment is addressed. The objective of optimization is to maximize the treatment efficiency of the biofilter by manipulating the feed flow rate while satisfying operational constraints. Numerical results indicate that the optimal input is characterized as being on the boundary of the admissible region. Thus, the characterized optimal solution is implemented using a simple feedback control law, which provides the optimal input profile despite variations in substrate inlet concentration and biomass growth rate.  相似文献

Disparities in nutrient content (nitrogen and phosphorus) between herbivores and their plant resources have lately proven to have major consequences for herbivore success, consumer-driven nutrient cycling, and the fate of primary production in ecosystems. Here we extend these findings by examining patterns of nutrient content between animals at higher trophic levels, specifically between insect herbivores and predators. Using a recently compiled database on insect nutrient content, we found that predators exhibit on average 15% greater nitrogen content than herbivores. This difference persists after accounting for variation from phylogeny and allometry. Among herbivorous insects, we also found evidence that recently derived lineages (e.g., herbivorous Diptera and Lepidoptera) have, on a relative basis, 15%-25% less body nitrogen than more ancient herbivore lineages (e.g., herbivorous Orthoptera and Hemiptera). We elaborate several testable hypotheses for the origin of differences in nitrogen content between trophic levels and among phylogenetic lineages. For example, interspecific variation in insect nitrogen content may be directly traceable to differences in dietary nitrogen (including dilution by gut contents), selected for directly in response to the differential scarcity of dietary nitrogen, or an indirect consequence of adaptation to different feeding habits. From some functional perspectives, the magnitude rather than the source of the interspecific differences in nitrogen content may be most critical. We conclude by discussing the implications of the observed patterns for both the trophic complexity of food webs and the evolutionary radiation of herbivorous insects.  相似文献

We have developed a radiation resistant bacterium for the treatment of mixed radioactive wastes containing ionic mercury. The high cost of remediating radioactive waste sites from nuclear weapons production has stimulated the development of bioremediation strategies using Deinococcus radiodurans, the most radiation resistant organism known. As a frequent constituent of these sites is the highly toxic ionic mercury (Hg) (II), we have generated several D. radiodurans strains expressing the cloned Hg (II) resistance gene (merA) from Escherichia coli strain BL308. We designed four different expression vectors for this purpose, and compared the relative advantages of each. The strains were shown to grow in the presence of both radiation and ionic mercury at concentrations well above those found in radioactive waste sites, and to effectively reduce Hg (II) to the less toxic volatile elemental mercury. We also demonstrated that different gene clusters could be used to engineer D. radiodurans for treatment of mixed radioactive wastes by developing a strain to detoxify both mercury and toluene. These expression systems could provide models to guide future D. radiodurans engineering efforts aimed at integrating several remediation functions into a single host.  相似文献

Variation, and in particular regular pattern in that variation, forms the foundation for evolutionary physiology. Nonetheless, with the exception of seemingly good fits between the tolerances of animals and the environments they live in, this variation is often not well explored. Here, three examples of different forms of such variation (both large- and small-scale) in a range of physiological traits in insects are explored. In the first example, I show that at global, regional, and local scales, variation in insect upper lethal temperatures is far less variable than variation in lower lethal temperatures, and that upper and lower tolerances are partially decoupled. Second, I demonstrate that variation in upper and lower lethal limits, desiccation resistance and tolerance, and respiration rate are often partitioned at taxonomic levels above that of the species. In other words, there is considerable phylogenetic constraint in the evolution of the responses of insects to the environment. These findings suggest that several ideas regarding insect physiological adaptations might have to be re-examined. They also suggest that approaches using both "raw" and corrected data should be adopted where possible. Finally, I demonstrate that there is considerable intra-individual variation in the characteristics of insect discontinuous gas exchange cycles. This is perhaps well-known to researchers in the field, but the implications thereof for arguments in favour of the adaptive nature of these regular cycles have not been carefully examined. Together, these findings suggest that there is still much to be learned about variation in insect physiological traits.  相似文献

Over the last 20 years, there has been a growing requirement by governments around the world for organisations to adopt more sustainable practices. Wastewater treatment is no exception, with many currently used systems requiring large capital investment, land area and power consumption. High rate algal ponds offer a sustainable, efficient and lower cost option to the systems currently in use. They are shallow, mixed lagoon based systems, which aim to maximise wastewater treatment by creating optimal conditions for algal growth and oxygen production—the key processes which remove nitrogen and organic waste in HRAP systems. This design means they can treat wastewater to an acceptable quality within a fifth of time of other lagoon systems while using 50% less surface area. This smaller land requirement decreases both the construction costs and evaporative water losses, making larger volumes of treated water available for beneficial reuse. They are ideal for rural, peri-urban and remote communities as they require minimum power and little on-site management. This review will address the history of and current trends in high rate algal pond development and application; a comparison of their performance with other systems when treating various wastewaters; and discuss their potential for production of added-value products. Finally, the review will consider areas requiring further research.  相似文献

Elasticities in stochastic matrix models are used to understand both population and evolutionary dynamics. We examine three such elasticities: stochastic elasticity E(ij)(S) with respect to the (i, j) matrix element, the elasticity E(ij)(S mu) with respect to the mean mu(ij) of the matrix element, and the elasticity E(ij)(S sigma) with respect to the variability sigma(ij) of the matrix element. We show that the stochastic elasticity E(S) does not accurately describe the effect of variability; one should use E(S sigma) and E(S mu). We establish two general properties of these elasticities: a sum rule that connects them and a limit on the sum of the E(S sigma). We discuss the implications of these properties for the analysis of buffering and selection on the average rates versus the variability of rates.  相似文献

Insect populations vary in the proportion of individuals exhibiting a particular ‘state’ (e.g., developmental stage, sex, egg load, or nutritional status). Because an insect's developmental state often determines both its size and its behavior, it is likely that this will also affect the probability of being sampled. We propose that a comprehensive approach to pest management must consider the interaction between the structure of a pest population (i.e., the relative number of individuals in each state) and any state‐dependent sampling bias. To illustrate the usefulness of this method we sampled populations of the western tarnished pest bug, Lygus hesperus Knight (Heteroptera: Miridae), in cotton fields. Our sampling technique utilized large cages to measure the absolute densities of each L. hesperus stage and adult sex within a population. This technique allowed us to document a wide range of absolute stage structures and sex ratios across 10 L. hesperus populations in California. Using a combination of cage samples and sweep net samples, we quantified the state‐dependent sampling bias by calculating the efficiency of sweep sampling as a function of L. hesperus developmental stage and sex. We found that the efficiency of sweep nets increased steadily with each successive developmental stage (i.e., nymphal instar) of L. hesperus. We also found that sweep nets are slightly more efficient in capturing male vs. female L. hesperus adults. Since other studies have documented that the stage and sex of L. hesperus can affect feeding impact on cotton flower buds, our results suggest that accurate predictions of Lygus damage will need to incorporate stage and sex‐dependent sampling biases.  相似文献

We review the uses of fossil insects, particularly Coleoptera (beetles) and Chironomidae (non-biting midges) from ancient deposits to inform the study of wetland ecosystems and their ecological and restoration processes. In particular, we focus on two contrasting ecosystems, drawing upon research undertaken by us on British raised mire peats and shallow lake systems, one an essentially terrestrial ecosystem, the other aquatic, but in which wetland insects play an important and integral part. The study of raised mire peats suggests that faunal stability is a characteristic of these wetland systems, over what appear to be extensive periods of time (up to several millennia), whilst studies of shallow lake ecosystems over recent timescales indicates that faunal instability appears to be more common, usually driven by increasing eutrophication. Drawing upon a series of fossil Coleoptera records spanning several thousand years from Hatfield Moors, south Yorkshire, we reconstruct in some detail the mire’s ontogeny and fluctuations in site hydrology and vegetation cover, illustrating the intimate association between substrate, topography and peat development. A comparison between fossil and modern beetle populations indicates that the faunal characteristics of this mire and its adjacent neighbour, Thorne Moors, become established during the early phases of peat development, including its rare endemics, and that the faunal biodiversity on the sites today is dictated by complex site histories. The over-riding characteristic of these faunas is of stability over several thousand years, which has important implications for the restoration of degraded sites, especially those where refugial areas are limited. In contrast, analyses of fossil Chironomidae from shallow lakes allow researchers to track changes in limnological status and while attempts have been made to reconstruct changes in nutrient levels quantitatively, the chironomids respond indirectly to such changes, typically mediated through complex ecosystem dynamics such as changes in fish and/or macrophyte communities. These changes are illustrated via historic chironomid stratigraphies and diversity indices from a range of shallow lakes located across Britain: Slapton Ley, Frensham Great Pond, Fleet Pond, Kyre Pool and Barnes Loch. These sites have shown varying degrees of eutrophication over recent timescales which tends to be associated with a decline in chironomid diversity. While complex functional processes exist within these ecosystems, our evidence suggests that one of the key drivers in the loss of shallow lake chironomid diversity appears to be the loss of aquatic macrophytes. Overall, while chironomids do show a clear response to altered nutrient regimes, multi-proxy reconstructions are recommended for a clear interpretation of past change. We conclude that if we are to have a better understanding of biota at the ecosystem level we need to know more of the complex interactions between different insect groups as well as with other animal and plant communities. A palaeoecological approach is thus crucial in order to assess the role of insect groups in ecosystem processes, both in the recent past and over long time scales, and is essential for wetland managers and conservation organisations involved in long term management and restoration of wetland systems  相似文献

We compared phytoplankton and phytobenthos pigment strategies in 17 shallow lakes and ponds from northern Canada and Alaska, sampled during mid to late summer. Benthic chlorophyll a concentrations (8–261 mg m−2) greatly exceeded those of the phytoplankton (0.008–1.4 mg m−2) in all sites. Cyanobacteria dominated the phytobenthos, while green algae and fucoxanthin-groups characterized the plankton. Both communities had higher photoprotection in cold, UV-transparent, high latitude waters. Phytoplankton had higher concentrations of photoprotective carotenoids per unit chlorophyll a than the phytobenthos. The planktonic photoprotective pigments were positively correlated with UV-penetration, and inversely correlated with temperature and coloured dissolved organic matter. A partial redundancy analysis showed that the benthic pigments were related to latitude, area and temperature. The UV-screening compound scytonemin occurred in high concentrations in the phytobenthos and was inversely related to temperature, while benthic carotenoids per unit chlorophyll a showed much lower variability among sites. These differing pigment strategies imply divergent responses to environmental change between the phytobenthos and phytoplankton in high latitude lakes.  相似文献

In this paper we demonstrate how broad scale comparative physiology has an important role to play in informing a variety of assumptions made in macroecology. We do so by examining large-scale geographic variation in insect development, thermal tolerance and metabolic rate. From these studies, and those from the literature on insect water loss and thermoregulation, we show that there is often a bias to the geographic extent of available empirical data. Studies of cold hardiness are most usually undertaken at high latitudes, while investigations of upper thermal tolerances and water loss are most common in warm arid regions. Likewise, we demonstrate that much variation in insect physiological tolerances is partitioned at higher taxonomic levels, which has important implications for comparative physiology. Intriguingly, data on the full range of variables we review are available for only three species. We also show that, despite its importance, body size is regularly reported in only some kinds of investigations (metabolic rate, water loss rate), whereas in others (upper lethal temperature, cold hardiness, development) this variable is often ignored. In short, although large-scale comparative physiology can contribute considerable understanding to both physiology and ecology, there is much that remains to be done.  相似文献
