The production and regulation of extracellular bacteriolytic enzymes of Agaricus bisporus are being studied to understand better the nutrition of this fungus and to identify factors that regulate the selectivity of mushroom compost as a growth medium. Both muramidase (EC.3.2.1.17) and N -acetyl-β- D -glucosaminidase (β-GlcNAcase, EC.3.2.1.30) have been detected in liquid cultures of A. bisporus , and in cultures fruiting in sterile and non-sterile compost. A turbidometric assay, based on the decrease in optical density of suspended Bacillus subtilis bacterial cell walls, was used to measure muramidase production by A. bisporus . A colorimetric assay was used to measure β-GlcNAcase. Both bacteriolytic enzyme activities were produced on a range of sole carbon sources, including killed freeze-dried B. subtilis cells. Muramidase activity was highest in axenic compost cultures. Bacteriolytic enzyme activity peaked as the first group of fruit bodies was harvested in both sterile and non-sterile compost.  相似文献

The Brazilian almond mushroom is currently cultivated for its medicinal properties but cultivars are suspected all to have a common origin. The objective of this work was to assess the potential of wild isolates of Agaricus subrufescens Peck (Agaricus blazei, Agaricus brasiliensis) as a source of new traits to improve the mushroom yield and quality for developing new cultures under European growing conditions. The wild European strains analysed showed a good ability to be commercially cultivated on wheat straw and horse manure based compost: shorter time to fruiting, higher yield, similar antioxidant activities when compared to cultivars. They have a valuable potential of genetic and phenotypic diversity and proved to be interfertile with the original culture of the Brazilian almond mushroom. Intercontinental hybrids could be obtained and combine properties from both Brazilian and European germplasm for increasing the choice of strains cultivated by the mushroom growers.  相似文献

The fungus Agaricus bisporus is commercially grown for the production of edible mushrooms. This cultivation occurs on compost, but not all of this substrate is consumed by the fungus. To determine why certain fractions remain unused, carbohydrate degrading enzymes, water-extracted from mushroom-grown compost at different stages of mycelium growth and fruiting body formation, were analyzed for their ability to degrade a range of polysaccharides. Mainly endo-xylanase, endo-glucanase, β-xylosidase and β-glucanase activities were determined in the compost extracts obtained during mushroom growth. Interestingly, arabinofuranosidase activity able to remove arabinosyl residues from doubly substituted xylose residues and α-glucuronidase activity were not detected in the compost enzyme extracts. This correlates with the observed accumulation of arabinosyl and glucuronic acid substituents on the xylan backbone in the compost towards the end of the cultivation. Hence, it was concluded that compost grown A. bisporus lacks the ability to degrade and consume highly substituted xylan fragments.  相似文献

Ammonia suppressants are applied to chicken litter to decrease ammonia levels. And mushroom (Agaricus bisporus) producers use poultry litter to increase the nitrogen in the compost. To determine the influence of ammonia suppressants used in poultry litter on compost preparation and mushroom production, four mushroom crops were cultivated from compost prepared using litter treated with PLT, Barn Fresh and Impact-P at 25.22 kg/100 m2, 40 kg/100 m2, and 0.49 kg/100 m2, respectively, during the poultry production process. In general, no significant differences (P>0.05) were noted between treatments for total nitrogen, ammonia, pH, EC, ash, and moisture when compost or the headspace air was sampled during compost preparation throughout all stages. Nor were mushroom yields or counts significantly affected (P>0.05) by the presence of ammonia suppressants in the poultry litter. Thus, the mushroom industry can confidently use poultry litter amended with PLT, Impact-P, and Barn Fresh when used at the recommended rates.  相似文献

Actinomycetes isolated from 10 different soil and compost samples were screened for production of pectinolytic enzyme activities when grown on pectin-containing solid and liquid media. Pectinolytic enzymes, detected by using plate diffusion tests with a medium containing ramie (Boehmeria nivea) plant material as the sole carbon source, were mainly pectate lyases, but low activities of pectinesterases were also observed. Polygalacturonases and polymethylgalacturonases were not produced. Multiple forms of pectate lyases were detected in the culture supernatants of some of the strains by using the zymogram technique of isoelectric focusing gels. Xylanolytic and cellulolytic activities were always found to be associated with pectinolytic activities. None of the pectinolytic enzymes were produced in a medium with glucose as the sole carbon source. Treatment of ramie bast fibers with crude enzyme preparations from a selection of strains showed a good correlation between the pectate lyase activity applied and the degumming effect, resulting in good separation of the bast fibers.  相似文献

目的研究以鸡粪和菌糠为主体物料在高温堆肥发酵过程中纤维素酶、木聚糖酶、淀粉酶以及蛋白酶酶活性的变化。方法通过测定各酶活动态变化,了解各酶在鸡粪堆肥中的变化规律。其中纤维素酶和淀粉酶采用DNS比色法,木聚糖酶活采用还原糖法,蛋白酶活采用福林法进行测定。结果随温度不断变化和酶本身的性质以及物料的作用机制机制不同,酶活动态变化表现出一定的差异性。物料发酵温度在0~3 d时迅速上升,在3~18 d持续高温期,15 d之后下降趋于恒定。各酶活变化基本上符合先上升至峰值再下降,最后保持恒定的规律。其中各酶发生高效反应的时间具有不一致性,纤维素酶活在第18天时达到峰值,淀粉酶活在第9天时达到峰值,木聚糖酶在第12天时达到峰值,而蛋白酶活在第3天时即达到峰值。结论酶对物料腐熟具有重要用,本研究结果为堆肥腐熟提供参考指标,对优化堆肥工艺具有重要意义。  相似文献

柱状田头菇Agrocybe aegerita作为一种新开发的食用菌,越来越受到消费者的喜爱和研究者的关注,但在其栽培生产中遇到的产量相对较低的问题,一直未能得到很好的解决,致使柱状田头菇的栽培难以得到大规模的推广。本研究对不同来源的柱状田头菇在其6个不同生长发育阶段中,9种与培养料中主要组分分解相关的胞外酶活性的变化进行了测定,结果表明:柱状田头菇属褐腐菌,对非木质纤维素的利用能力最强,对纤维素的利用能力较强,对木质素的利用能力较差,但柱状田头菇具有漆酶活性。  相似文献

To evaluate the attractiveness of several mushroom‐growing substrates to the female mushroom fly Lycoriella ingenua (Dufour) (Diptera: Sciaridae), a pest of the cultivated white button mushroom, Agaricus bisporus (JE Lange) Emil J Imbach (Agaricales), we developed a two‐choice, static‐flow olfactometer. Behavioral assays using this olfactometer indicated that mushroom compost with A. bisporus mycelia growing in it was not more attractive than compost lacking growing mycelia. We also found that female flies were more attracted to compost lacking A. bisporus mycelia than to the actual commodity, the white button mushroom fruiting bodies. Flies were not, however, attracted to sterilized compost, suggesting the attraction is due to volatiles produced by microbial metabolism in the compost. We also found that female L. ingenua flies were attracted to the mycoparasitic green mold Trichoderma aggressivum Samuels & W Gams (Hypocreales). Flies preferred mushroom compost that had T. aggressivum growing in it over compost lacking T. aggressivum, providing an experimental outcome consistent with the anecdotal belief that L. ingenua flies are vectors of T. aggressivum spores that can infest mushroom‐growing houses.  相似文献

以农村生活垃圾中的可堆肥腐熟成分和蘑菇渣为堆肥原料,通过添加微生物菌剂进行堆肥试验,研究其在农业废弃物堆肥腐熟过程中的作用,并通过田间试验研究堆肥腐熟后肥料样品对黄瓜和青椒的增产效果,以验证其肥效。结果表明添加微生物菌剂有助于堆肥腐熟后样品的氮、磷、钾的保全和有机质的增加,促进养分均衡。添加微生物菌剂的堆肥腐熟肥料样品在田间试验中对黄瓜和青椒的增产效果最为显著,分别为22.21%和19.87%。  相似文献

Actinobacteria are widely distributed in many environments and represent the most important trigger to the occupant respiratory health. Health complaints, including hypersensitivity pneumonitis of the workers, were recorded in a mushroom compost facility (MCF). The studies on the airborne bacteria were carried out to find a possible microbiological source of these symptoms. Culture analysis of compost bioaerosols collected in different location of the MCF was performed. An assessment of the indoor microbial exposure revealed bacterial flora of bioaerosol in the mushroom compost facility represented by Bacillus, Geobacillus, Micrococcus, Pseudomonas, Staphylococcus spp., and actinobacterial strain with white aerial mycelium. The thermotolerant actinobacterial strain of the same morphology was repeatedly isolated from many locations in MCF: air, compost sample, and solid surface in production hall. On the base of complex morphological, chemotaxonomic, and phylogenetic characteristics, the isolate has been classified as Nocardiopsis alba. Dominant position of N. alba in microbial environment of the mushroom compost facility may represent an indicator microorganism in compost bioaerosol. The bioavailability of N. alba in mushroom compost facility creates potential risk for the health of workers, and the protection of respiratory tract and/or skin is strongly recommended.  相似文献

Evolution of extracellular enzyme activities during manure composting   总被引:7,自引:0,他引:7  
AIMS: The objectives of this work were to determine the extracellular enzyme profiles during composting, relate the activities of these enzymes to the changes in microbial population and compare the enzyme profiles between two manures. METHODS AND RESULTS: API ZYM assay was used to monitor the activities of 19 extracellular enzymes during poultry and pig manure composting. Results showed an overall increase in diversity and relative abundance of enzymes present. The relative abundance and activities of enzymes were higher in poultry manure than in pig manure. Among the 19 enzymes tested, esterase, valine amino-peptidase and alpha-galactosidase were the most abundant enzymes in poultry manure, whereas it was N-acetyl-beta-glucosaminidase for the pig manure. A number of these enzymes correlated with change in numbers of different microbial groups during composting. CONCLUSIONS: The composting process represented a combined activity of a wide succession of environments, as one enzyme/microbial group overlapped the other and each emerged gradually due to the continual change in temperature and progressive breakdown of complex compounds to simpler ones. SIGNIFICANCE AND IMPACT OF THE STUDY: The results presented here show the applicability of the API ZYM test not only in monitoring the quantitative and qualitative fluctuation of the available substrate during composting, but also in revealing differences in composts and compost maturity.  相似文献

Spent steamed compost, phase II compost, and dust emanating from spent compost during dumping of stationary-bed mushroom houses were examined bacteriologically. The total count for spent compost was 16 X 10(8) microorganisms per g. The total count for dust was 333 microorganisms per liter of air. Actinomycetes belonging to the genus Streptomyces often constituted 90% or more of isolates from dust, whereas mold spores constituted approximately 5%. Dust weight averaged 3.4 mg/liter of air and contained approximately 33% inanimate and 67% animate (microbial) particles. Spent compost and casing contained approximately 60% moisture; the average pH of compost was 6.93, and that of casing was 7.70. Ouchterlony precipitin results with antisera from workers afflicted with either farmer's or mushroom worker's lung were positive for Bacillus licheniformis, Micropolyspora faeni, Thermoactinomyces vulgaris, Aspergillus fumigatus, Humicola grisea var. thermoidea, spent compost, and phase II compost. Their usefulness in determining the etiology of this and related forms of allergic alveolitis is questioned and discussed. The relationship of dust particle size; microbial species, prevalence and antigenicity; and compost antigenicity to the etiology of mushroom worker's lung is discussed. The microbial ecology of mushroom compost and moldy hay associated with farmer's lung is compared.  相似文献

Bioconversion of lignocellulosic biomass to fuel requires a hydrolysis step to obtain fermentable sugars, generally accomplished by fungal enzymes. Large-scale screening of different microbial strains would provide optimal enzyme cocktails for any target feedstock. The aim of this study was to screen a large collection of Trichoderma sp. strains for the hydrolytic potential towards switchgrass (Panicum virgatum L.). Strains were cultivated in a small-scale system and assayed in micro-plates for xylanase and cellulase activities. The population distributions of these traits are reported after growth on switchgrass in comparison with cellulose. The distribution profiles suggest that the growth on switchgrass strongly promotes xylanase production. The IK4 strain displayed the highest xylanase activity after growth on switchgrass (133U/mL). Enzymes (10FPU/g substrate) from IK4 were compared with those from 2 cellulolytic Trichoderma strains and a commercial enzyme in saccharification time-course experiments on untreated and pretreated switchgrass and on an artificial substrate. Samples were analysed by DNS assay and by an oxygraphic method for sugar equivalent or glucose concentration. On the untreated substrate, IK4 enzymes even outperformed a 5-fold load of commercial enzyme, suggesting that xylanase or accessory enzymes are a limiting factor on this type of recalcitrant substrate. On the other substrates, IK4 preparations showed intermediate behaviour if compared with the commercial enzyme at 10FPU/g substrate and at 5-fold load. IK4 also nearly halved the time to release 50% of the hydrolysable sugar equivalents (T(50%)), with respect to the other preparations at the same enzymatic load. DNS assay and oxygraphic method gave highly correlated results for the 3 saccharified substrates. The study suggests that accessory enzymes like xylanase play a key role in improving the performance of cellulase preparations on herbaceous lignocellulosic feedstocks like switchgrass.  相似文献

Investigations into the dynamic nature of composting environments are necessary to understand and ultimately optimise the complex processes that occur. In this study, various parameters were measured to investigate physical, chemical and biological changes that occur in compost during the production of Agaricus bisporus. In addition to monitoring the compost samples during mushroom cultivation, uninoculated samples were maintained for comparative purposes. Principal components analysis of the variables measured showed a clear distinction between the thermophilic Phase I composts, uninoculated Phase II composts and mushroom inoculated composts. Leucine assimilation, a novel technique to composting environments, is presented as suitable method for assessing microbial activity in such systems. Strict agreement between leucine assimilation and fluorescein diacetate (FDA) hydrolysis, a rarely used technique in composting environments, was not observed, suggesting that neither should be used as a sole measure of microbial activity in compost. Association of FDA hydrolysis with the culturable heterotrophic count suggests that FDA hydrolysis may indicate bacterial as opposed to total microbial activity.  相似文献

AIMS: To develop strategies for increasing the growth of Lentinula edodes in eucalyptus residues. To this end, we have examined the effects of cereal brans additions on production of mycelial biomass and enzymes. METHODS AND RESULTS: Three isolates of the mushroom shiitake, L. edodes (Berk. Pegler), were evaluated for enzyme and ergosterol production on eucalyptus residue supplemented with 5, 10, 15 and 20% (w/w) of soya, wheat or rice brans. Nitrogen imput on eucalyptus residues accelerated mycelial growth by supplying the L. edodes with this limiting nutrient. High levels of enzymes activities were produced in eucalyptus residues supplemented by soya bran. Comparison of cellulose and xylanase production with manganese peroxidase (MnP) at 20% soya bran indicated that hydrolytic enzymes, but oxidative enzymes were reduced. CONCLUSIONS: Mycelial growth measurements revealed that eucalyptus residues supplemented with cereal brans supported fast growth of L. edodes, indicating that mycelium extension is related to the bioavailability of nitrogen. The type and concentration of nutrient supplement has a considerable effect both on substrate colonization and on the type of hydrolytic and oxidative enzymes produced. These characteristics may be useful for mushroom growing. SIGNIFICANCE AND IMPACT OF THE STUDY: Lentinula edodes is commercially important for edible mushroom production and supplements which enhance growth and enzymes production might also be beneficial for mushroom yields.  相似文献

Rice straw (RS) is an important raw material for the preparation of Agaricus bisporus compost in China. In this study, the characterization of composting process from RS and wheat straw (WS) was compared for mushroom production. The results showed that the temperature in RS compost increased rapidly compared with WS compost, and the carbon (C)/nitrogen (N) ratio decreased quickly. The microbial changes during the Phase I and Phase II composting process were monitored using denaturing gradient gel electrophoresis (DGGE) and phospholipid fatty acid (PLFA) analysis. Bacteria were the dominant species during the process of composting and the bacterial community structure dramatically changed during heap composting according to the DGGE results. The bacterial community diversity of RS compost was abundant compared with WS compost at stages 4–5, but no distinct difference was observed after the controlled tunnel Phase II process. The total amount of PLFAs of RS compost, as an indicator of microbial biomass, was higher than that of WS. Clustering by DGGE and principal component analysis of the PLFA compositions revealed that there were differences in both the microbial population and community structure between RS- and WS-based composts. Our data indicated that composting of RS resulted in improved degradation and assimilation of breakdown products by A. bisporus, and suggested that the RS compost was effective for sustaining A. bisporus mushroom growth as well as conventional WS compost.  相似文献

The influences of Zn and Cu on soil enzyme activities (acid phosphatase, alkaline phosphatase, arylsulfatase, cellulase, dehydrogenase, protease (z-FLase), urease, beta-D-glucosidase and beta-D-fructofuranosidase (invertase)) and microbial biomass carbon were investigated in agricultural soils amended with municipal sewage sludge or compost since 1978. The trace metals in the soils were fractionated using a sequential extraction method. Long-term application of the sewage sludge and composts caused accumulations of Cu and Zn in the soils, ranging from 140 to 144 and from 216 to 292 mg kg(-1), respectively. The percentage of Cu was highest in the NaOH- and HNO3-extractable fractions (44-51% and 38-46%, respectively), while the percentage of Zn was highest in the HNO3- and EDTA-extractable fractions (65-83% and 11-32%, respectively). Although the percentage of the bioavailable fractions (sum of KNO3 + H2O-, NaOH-, and EDTA-extractable amounts) of Cu (53-64%) was higher than that of Zn (15-37%), the percentage of the most labile fractions (KNO3 + H2O) of Zn (2.1-5.9%) was larger than that of Cu (1.1-2.4%). The size of the microbial biomass carbon increased with the application of sewage sludge or compost. For some enzymes, however, the ratio of the enzyme activity to microbial biomass was lower in the soils amended with sewage sludge or compost than that in the control soil. The soil enzyme activities were more adversely affected by Zn than by Cu. From a multiple regression analysis, it was found that dehydrogenase, urease, and beta-D-glucosidase activities were reduced by the KNO3 + H2O-extractable fraction of Zn in the soils. These microbial activities seem to be sensitive to Zn stress, indicating the possibility that they might be useful bioindicators for evaluation of the toxic effects of Zn on microorganisms in the soils.  相似文献

Preliminary studies suggested that the use of compost tea made from spent mushroom substrate (SMS) may be regarded as a potential method for biologically controlling dry bubble disease in button mushroom. The aim of this study was to assess the effect of SMS compost tea on the host, the button mushroom, to ascertain whether the addition of these water extracts has a toxic effect on Agaricus bisporus mycelium growth and on mushroom yield. In vitro experiments showed that the addition of SMS compost tea to the culture medium inoculated with a mushroom spawn grain did not have an inhibitory effect on A. bisporus mycelial growth. The effect of compost teas on the quantitative production parameters of A. bisporus (yield, unitary weight, biological efficiency and earliness) was tested in a cropping trial, applying the compost teas to the casing in three different drench applications. Quantitative production parameters were not significantly affected by the compost tea treatments although there was a slight delay of 0.8-1.4 days in the harvest time of the first flush. These results suggest that compost teas have no fungitoxic effect on A. bisporus so that they can be considered a suitable biocontrol substance for the control of dry bubble disease.  相似文献
