通过研究荞麦种子萌发期(0~7 d)超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)和抗坏血酸过氧化物酶(ASP)四种抗氧化酶的酶活性变化,以及对自由基等的清除效果,分析荞麦种子内在抗氧化酶系统在萌发期对细胞膜结构的修复及保护作用。实验结果表明:荞麦种子在萌发期产生的活性氧、自由基等对四种抗氧化酶均有激活效应,其中超氧化物歧化酶(SOD)活性最先上升,其他三种酶随其后被激活,四种酶的氧化反应存在一定关联性和协同作用。苦荞的抗氧化酶活性高于甜荞。  相似文献

PAL activity in young fruits of Citrus unshiu was initiallyvery high. It decreased gradually as the fruit grew and afterthe "June drop", declined markedly. PAL behavior appears tobe closely related to the tissue differentiation of fruit. Proteincontent of the fruit showed a pattern of change similar to thatof PAL activity. Also, the amount of hesperidine was high inimmature fruit. Summary Xylem differentiation was induced in cultured Coleus internode slices when grown in the light on a simple agar/sucrose/IAA medium and in darkgrown soybean callus tissue when cultured on a complex defined medium containing 5×10-7 M kinetin. In the Coleus system, the activity of phenylalanine ammonialyase followed the same time course as the formation of lignified wound vessel members. The specific activity of PAL was higher in the soybean callus tissues grown on 5×10-7 M kinetin, which produced tracheary elements, than in the soybean tissue grown on 10-8 M kinetin, which did not produce tracheids. Phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) and tyrosine ammonia-lyase (TAL, 4.3.1.), the key enzymes of the phenylpropanoid pathway, are inducible in response to biotic (such as chitin from fungal cell walls) and abiotic cues. Application of chitin and chitosan to soybean leaf tissues caused increased activity of PAL and TAL enzymes. The elevation of enzyme activity was dependent on the chain length of the oligomers and time after treatment. The hexamer of chitin and pentamer of chitosan produced the maximum activities at 36 h after treatment as compared to controls. Total phenolic content of soybean leaves increased following chitosan and chitin oligomer treatments, showing a positive correlation between enzyme activity and total phenolic content.  相似文献

When dark-grown cell suspension cultures of parsley (Petroselinum hortense) were illuminated for increasing periods of time, increasing amounts of phenylalanine ammonialyase activity were obtained 5 hr after the onset of light.Pulses of [35S]methionine of varying duration from 1 to 150 min were given to cell cultures in the dark period subsequent to a light period of 2.5 hr. The cells were harvested 5 hr after the onset of light. Analysis of the soluble proteins by polyacrylamide gel electrophoresis revealed a distinct peak of radioactivity coinciding with the activity of phenylalanine ammonia-lyase. The results of experiments in which radioactive methionine was administered for 10 min to dark-grown or light-induced cells at different times after the light period were compared. An efficient incorporation of radioactivity into the fractions possessing the enzyme activity was observed 5 hr after induction, while no significant labeling was detected either after 1.5 or 25 hr, or in extracts from nonilluminated cells. The radioactive fractions containing the enzyme activity were further analyzed by sodium dodecyl sulfate-disc gel electrophoresis. Significant amounts of radioactivity at the molecular weight of the subunits of phenylalanine ammonia-lyase (84,000) were found only in the extracts from cells which had been labeled 5 hr after induction. These results suggest that the light-induced increase in phenylalanine ammonia-lyase activity is due to de novo synthesis, but not to an activation of preformed, inactive enzyme.  相似文献

为探究七叶一枝花的种子萌发生理机制,该文从种胚形态上对七叶一枝花种子萌发过程的时期进行了划分,并分析了不同时期种子内源激素含量及相关酶活性的变化规律。结果表明:(1)根据种胚形态可将种子萌发过程细分为8个时期,即种胚未萌动时期(S1期)、心形胚时期(S2期)、种胚膨大时期(S3期)、胚根未突破种皮时期(S4期)、子叶柄伸长和胚根突破种皮时期(S5期)、下胚轴突破种皮时期(S6期)、上胚轴伸长时期(S7期)、胚根伸长时期(S8期)。(2)不同萌发时期种子的α-淀粉酶活性均明显高于β-淀粉酶。(3)超氧化物歧化酶(SOD)活性在S5期最高,S1期最低; 过氧化物酶(POD)和过氧化氢酶(CAT)活性随种子萌发进程总体呈上升趋势,可溶性蛋白含量随种子萌发进程先下降后升高。(4)吲哚乙酸(IAA)、赤霉素(GA3)、脱落酸(ABA)和水杨酸(SA)含量随种子萌发进程整体呈下降趋势; 1-氨基环丙烷羧酸(ACC)、茉莉酸(JA)、油菜素内酯(BRs)随种子萌发进程整体呈上升趋势; 细胞分裂素类(CTKs)含量无显著变化; IAA/ABA和GA3/ABA随种子萌发进程呈先下降后上升趋势,而CTKs/ABA则不断升高。(5)ABA、IAA、GA3含量与胚率呈负相关,而ACC、JA、BRs、POD、CAT、β-淀粉酶活性与胚率呈正相关。综上认为,七叶一枝花在不同时期种子内源激素含量及相关酶活性各不相同,其中α-淀粉酶活性、POD活性可能与种胚胚根伸长有关,GA3可能影响种胚形成,而ABA则可能抑制种胚的生长发育。  相似文献

Microsomal fractions were isolated from livers of rats that been fed p-dimethylaminoazobcnzene (DAB) for 2 months sygnificant decreases were noted in the specific activities of four of the membrane-bound enzymes, Sudies on membrane composition and the interactions between membranc components were carried out in an attempt to explain the mechanisms responsable for these decreases. Changes in the lipid components of the membrane were brought about as a result of DAB feeding. No changes were detected in the protein components of the membrane. 1-Anilinonaphthalene-8-Sulfonic acid (ANS) fluorescence enhancement was decreased by 25 % in membranes delived from DAB-fed animals. The response of the membranebound enzymes to pretreatment with NH4OH and Triton X-100 was examined. The glucose-6-phosphatase of membranes derived from carcinogen-fed animals was more activated by Triton than the enzyme from control. It is concluded that altered membrane conformation is a result of DAB carcinogensus, and that such alterations may be responsible for loss of enzyme activity.  相似文献

The hyperoxia-induced increases in the activity of lung glucose-6-phosphate dehydrogenase (G-6-P) and glutathione reductase (GR) after exposure of rats to greater than 97% O2 for 6 days were accompanied by equivalent increases in the amount of the respective immunoreactive proteins. Hyperoxia also increased lung glutathione (GSH) + oxidized glutathione (GSSG) content and the magnitude of this hyperoxic response of increased GSH + GSSG, G-6-P, and GR (maximal 1.3- to 1.8-fold) declined as a function of age during the first 3 wk of life. Fetal rat lung explants cultured 4 days in 95% O2 showed increased G-6-P and GR activity and increased levels of the specific proteins 1.5-fold those of explants at 2 days of culture. We conclude that the hyperoxic response of increased rat lung G-6-P and GR activity in vivo and in vitro involves not just alteration of enzyme activity but also specific increases in the proteins catalyzing the reactions.  相似文献

The contribution of cytochrome and alternative pathways to respiration during soybean [ Glycine max (L.) Merr. cv. Chippewa 64] seed formation and maturation was measured by oxygen consumption of embryonic axis and cotyledon tissues and of mitochondria isolated from immature seeds. Respiration rates (dry weight basis) declined duing the last month of seed maturation. Cyanide sensitive respiration (cytochrome pathway) accounted for 80 to 90% of total oxygen consumption throughout seed formation and maturation. The proportion of the total respiration resistant to 1 m M cyanide and inhibited by 1 m M salicylhydroxamic acid (alternative pathway) did not exceed 6%. A residual oxygen consumption, accounting for 5 to 15% of total respiration, persisted in the presence of both inhibitors. Likewise, virtually no alternative respiration was found in mitochondria prepared from immature seeds. The alternative respiratory pathway seems absent during soybean seed formation and maturation regardless of whether grown in the greenhouse or field.  相似文献

腊梅 (Chimonanthuspraecox)花两性 ,离心皮雌蕊着生在杯状花托上 ,柱头线形 ,干性。花粉经昆虫传播 ,落在柱头上 1d后萌发 ,第 8d从珠孔进入 ,第 1 4d左右完成双受精 ,为珠孔受精。胚乳为核型胚乳 ;初生胚乳核经短暂休眠进行核分裂 ,位于合点端的游离核首先形成细胞 ,并从合点向珠孔端细胞化 ,第 37d胚乳充满整个囊腔。合子经过近 2周的休眠后开始分裂 ,随着胚的发育 ,大部分胚乳降解 ,为胚的发育提供营养。合点端的胚乳细胞则侵入合点珠心组织 ,为胚进一步发育提供营养。其胚胎发生为柳叶菜型。  相似文献

The course of biosynthesis of fatty acids in the seeds of winter rape (Brassica napus L. ssp.oleifera, f.biennis cv. T?ebí?ská) was investigated. After the termination of flowering seed samples were taken at five intervals, the seeds were divided into 4 fractions according to size, and their weight, water content, oil content and fatty acid composition were determined. The oil content was found to increase in all size categories with time, with the exception of a minute drop when complete maturity is reached. Larger seeds contained more oil. The fatty acid composition changes with time in the individual size fractions almost continuously. The same holds for differences between seed sizes of the same sample. The main change in oil composition consists in the decrease of C18 acids in favour of C22 acids. Greatest decrements during maturation were found with oleic acid, less with linoleic acid. In absolute amounts the quantity of all synthesized acids rises, the greatest rise being observed with C22 acids (i.e. predominantly erucic acid). It follows from the mean rates of synthesis of the individual groups (C16, C18, C20, C22) of fatty acids that the fraction of C22 rate of synthesis increases, while that of the C18 acids decreases with the same speed. The results indicate that the fatty acid synthesis is most intense during the second half of seed maturation, the main role being played by accelerating the synthesis of higher acids, especially of erucic acid.  相似文献

We have used conserved and nonconserved regions of cDNA clones for phenylalanine ammonia-lyase (PAL) and chalcone synthase (CHS) isolated from a soybean-nodule cDNA library to monitor the expression of members of the two gene families during the early stages of the soybean-Bradyrhizobium japonicum symbiosis. Our results demonstrate that subsets of the PAL and CHS gene families are specifically induced in soybean roots after infection with B. japonicum. Furthermore, by analyzing a supernodulating mutant line of soybean that differs from the wild-type parent in the number of successful infections, we show that the induction of PAL and CHS is related to postinfection events. Nodulated roots formed by a Nod+ Fix- strain of B. japonicum, resembling a pathogenic association, display induction of another distinct set of PAL and CHS genes. Our results suggest that the symbiosis-specific PAL and CHS genes in soybean are not induced by stress or pathogen interaction.  相似文献

During maturation of tobacco seed, the amount of chlorogenic acid rapidly increased between days 7 and 11 after anthesis and then decreased until day 15. Chlorogenic acid was not detected in fully mature seed. On the other hand, caffeic acid appeared from day 15, reached a maximum at day 21, and rapidly disappeared by day 25 although the amount was always low in comparison with that of chlorogenic acid. Changes in dry wt, water content, testa color and germination of tobacco seeds during maturation were also investigated. The results suggested that there were rapid biochemical changes in seed between days 11 and 15 after anthesis.  相似文献

The influence of the allelochemicals ferulic (FA) and vanillic (VA) acids on peroxidase (POD, EC 1.11.1.7) and phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) activities and their relationships with phenolic acid (PhAs) contents and root growth of soybean (Glycine max (L.) Merr.) were examined. Three-day-old seedlings were cultivated in nutrient solution containing FA or VA (0.1 to 1 mM) for 48 h. Both compounds (at 0.5 and 1 mM) decreased root length (RL), fresh weight (FW) and dry weight (DW) and increased PhAs contents. At 0.5 and 1 mM, FA increased soluble POD activity (18% and 47%, respectively) and cell wall (CW)-bound POD activity (61% and 34%), while VA increased soluble POD activity (33% and 17%) but did not affect CW-bound POD activity. At 1 mM, FA increased (82%) while VA reduced (32%) PAL activities. The results are discussed on the basis of the role of these compounds on phenylpropanoid metabolism and root growth and suggest that the effects caused on POD and PAL activities are some of the many mechanisms by which allelochemicals influence plant growth.  相似文献
