Cytochalasin B binding proteins in human erythrocyte membranes. Modulation of glucose sensitivity by site interaction and partial solubilization of binding activities

We have previously described three different cytochalasin B binding sites in human erythrocyte membranes, a D-glucose-sensitive site (Site I), a cytochalasin E-sensitive site (Site II), and a site (Site III) insensitive to both D-glucose and cytochalasin E. Ligand bindings to each of these sites were considered to be independent (Jung, C., and Rampal, A. (1977) J. Biol. Chem. 252, 5456-5463). However, we have obtained subsequently the following evidence which indicated that an interaction occurs between Sites II and III, and this modulates sensitivity of Site III to the sugar. The displacement of cytochalasin E greatly exceeds the sum of their independent displacements. This ghosts extracted with EDTA or 2,3-dimethylmaleic anhydride at low ionic strength lack Site II activity but retain Site I and III activities, and both of these activities are displaceable by D-glucose alone. This indicated that the removal of Site II from the membrane confers glucose sensitivity to Site III. These observations are consistent with a model that Sites II and III in the membrane exist in a close association through which unliganded Site II maintains the glucose insensitivity of Site III, and once site II is liganded or removed by extraction this association is disrupted and Site III becomes glucose-sensitive. The ghosts extracted with Triton X-100 retain a cytochalasin B binding activity similar to that of site II (Kd = 1.8 X 10(-7) M, cytochalasin E-sensitive, glucose-insensitive), whereas a binding activity similar to that of Site I (Kd = 4 X 10(-7) M, cytochalasin E-insensitive, glucose-sensitive) is recovered in the Triton extract. A cytochalasin B binding activity similar to that of Site II is solubilized by EDTA at low ionic strength.     

Integrated stratigraphy and 40Ar/39Ar chronology of early Middle Miocene sediments from DSDP Leg 42A,Site 372 (Western Mediterranean)

An integrated magneto-biostratigraphic framework is presented for Middle Miocene sediments of DSDP Site 372 located in the Western Mediterranean. Detailed biostratigraphic analysis shows a nearly complete sequence of early Middle Miocene calcareous plankton bioevents in the Mediterranean, including the LCO (Last Common Occurrence) of the nannofossil Sphenolithus heteromorphus which has been astronomically dated in the Ras il Pellegrin (RIP) section on Malta Island [Abels, H.A., Hilgen, F.J., Krijgsman, W., Kruk, R.W., Raffi, I., Turco, E., Zachariasse, W.J., 2005. Long-period orbital control on middle Miocene global cooling: integrated stratigraphy and astronomical tuning of the Blue Clay Formation on Malta, Paleoceanography, 20, PA4012. doi: 10.1029/2004PA001129. 11 pp]. Thermal demagnetization of discrete samples revealed a characteristic low-temperature component with dual polarities despite a weak paleomagnetic signal. The resultant magnetostratigraphic record, combined with the calcareous plankton biostratigraphy, is straightforwardly correlated to the geomagnetic polarity time scale (CK95) of Cande and Kent [Cande, S.C., Kent, D.V., 1995. Revised calibration of the Geomagnetic Polarity Time Scale for the Late Cretaceous and Cenozoic, J. Geophys. Res., 100, 6093–6095] and the Astronomical Tuned Neogene Time Scale (ATNTS04) of Lourens et al. [Lourens, L.J., Hilgen, F.J., Laskar, J., Shackleton, N.J., Wilson, D., 2004. The Neogene Period. In: Gradstein F.M., Ogg J.G., Smith A.G. (Eds.), A Geologic Time Scale, Cambridge Univ. Press, pp. 409–440]. The subchrons recorded in Site 372 succession range from C5Br up to C5AAr. To confirm the magnetostratigraphic calibration, ⁴⁰Ar/³⁹Ar dating was performed on feldspar of two volcanic ash layers. The radio-isotopic dating indicates a younger age for these two ash layers compared to the magnetostratigraphic calibrated ages according to the CK95 and ATNTS04 age models. However, if the astronomically calibrated age of 28.21 ± 0.04 Ma is used for the Fish Canyon standard (FCs), the age for the older ash layer exactly matches its ATNTS04 age. Ages for bioevents were calculated assuming constant sedimentation rates between magnetostratigraphic age-tie points. The S. heteromorphus LCO has an age of 13.54 Ma and 13.63 Ma according to CK95 and ATNTS04, respectively, which is consistent with the astronomical tuned age of 13.65 Ma determined at RIP section [Abels, H.A., Hilgen, F.J., Krijgsman, W., Kruk, R.W., Raffi, I., Turco, E., Zachariasse, W.J., 2005. Long-period orbital control on middle Miocene global cooling: integrated stratigraphy and astronomical tuning of the Blue Clay Formation on Malta, Paleoceanography, 20, PA4012. doi:10.1029/2004PA001129. 11 pp]. We therefore conclude that the magnetostratigraphic calibration of DSDP Site 372 is correct and that, for the time being, this site can be considered as a reference section for the early Middle Miocene of the Mediterranean region.

Herpes simplex-1 helicase-primase. Identification of two nucleoside triphosphatase sites that promote DNA helicase action.

Herpes simplex virus type 1 (HSV-1) encodes a heterotrimeric helicase-primase comprised of the products of the UL5, UL8, and UL52 genes (Crute, J. J., and Lehman, I. R. (1991) J. Biol. Chem. 266, 4484-4488). A steady state kinetic analysis of the enzyme isolated from HSV-1-infected CV-1 cells or insect cells expressing the enzyme after infection with recombinant baculoviruses has shown it to possess two sites capable of hydrolyzing nucleoside triphosphates in a DNA-dependent manner. One site (Site I) hydrolyzes both ATP and GTP; the second (Site II) hydrolyzes only ATP. These two sites are contained within a subassembly of the helicase-primase formed by coexpression of the UL5 and UL52 genes in insect cells. Sites I and II are activated by separate DNA effector sites, both of which support DNA helicase action. These findings are likely to be of importance in understanding how helicases in general catalyze the unwinding of duplex DNA and, in particular, how the helicase-primase functions at the HSV-1 replication fork.     

A Magnetometer Survey of the Knife River Indian Villages

Abstract

There are several Indian village sites at the confluence of the Knife and Missouri rivers near the town of Stanton, North Dakota. Two of them were examined or tested by D. J. Lehmer in 1965. Four of the sites are now being incorporated into the Knife River Indian Villages National Historic Site. As part of the development of this Historic Site a proton magnetometer survey was carried out in 1976 on parts of Sakakawea Village and the Buchfink Site. In addition, a survey was conducted on part of the Amahami Village in Stanton. This article discusses the theory of magnetic surveying, the varying nature of the regions surveyed, and the extensive results abtained. These results include the identification of at least 12 earth lodge sites with associated features, the correlation of some features in the magnetic data with soil probe information, and a number of suggestive but unidentifed features on which the magnetic measurements focus.     

A Preliminary Report on Two Sites at Piney Creek,Wyoming 48J03Ll and 48J031

Abstract

During the summer of 1964, the University of Wyoming and the Wyoming Archaeological Society conducted exploratory excavations in two sites near Buffalo, Wyoming. Site 48J03ll consists of a number of stone circles and is apparently a camp site and site 48J0312 is a bison kill and butchering site. Artifact assemblages suggest an affiliation with late prehistoric and early historic sites in the Yellowstone River drainage.
