Background and aims

Selenium is an essential micro-nutrient for animals, humans and microorganisms; it mainly enters food chains through plants. This study proposes to explore effect of inorganic Se forms on its uptake and accumulation in Zea mays.

Methods

Zea mays was grown in a controlled-atmosphere chamber for 2 weeks in a hydroponic solution of low-concentration selenium (10 μg/L (i.e.0.12 μM) or 50 μg/L (i.e. 0.63 μM) of Se). For each concentration, four treatments were defined: control (without selenium), selenite alone, selenate alone and selenite and selenate mixed.

Results

At low concentrations, selenium did not affect the biomass production of Zea mays. However, for both concentrations, Se accumulation following a selenite-only treatment was always higher than with selenate-only. Moreover, in the selenate-only treatment, Se mainly accumulated in shoots whereas in the selenite-only treatment, Se was stocked more in the roots. Interactions between selenate and selenite were observed only at the higher concentration (0.63 μM of selenium in the nutrient solution).

Conclusions

Se form and concentration in the nutrient solution strongly influenced the absorption, allocation and metabolism of Se in Zea mays. Selenate seems to inhibit selenite absorption by the roots.  相似文献

Aims

The objective of this study was to determine the relative importance of transpirational pull, Se speciation, sulfate and species on Se accumulation by plants, in order to determine which of these factors must be considered in the future development of models to predict Se accumulation by plants.

Methods

Seedlings of durum wheat (Triticum turgidum L. var durum cv ‘Kyle’) and spring canola (Brassica napus L. var Hyola 401) were grown hydroponically and exposed to SeO 4 2- (selenate) with or without SO 4 2- (sulfate), or to HSeO 3 - (biselenite) under different transpiration regimes altered through ‘low’ (~50%) or ‘high’ (~78%) relative humidity (RH). Plants were harvested after 0, 8, 16, or 24?h exposures, digested, and analyzed for Se by GFAAS.

Results

Accumulation and distribution of Se by plants is dependent on plant species, Se speciation in the nutrient solution, SO 4 2- competition, and transpiration regimes. Canola accumulated and translocated more Se than wheat. In wheat and canola, the greatest accumulation and translocation of Se occurred when plants were exposed to SeO 4 2- without SO 4 2- compared to solutions of SeO 4 2- with SO 4 2- or HSeO 3 2- . Wheat plants exposed to SeO 4 2- and SO 4 2- had an increased Se accumulation and translocation under increased transpiration rates than when exposed to SeO 4 2- without SO 4 2- or HSeO 3 2- . On the other hand, increases in transpiration increased the translocation of Se to canola shoots when exposed to HSeO 3 - more than any other treatments.

Conclusions

Overall, our results suggest that plant species is the most important factor influencing Se accumulation and translocation, but that these endpoints can be modified by climate and specific soil Se or S content. Models to predict accumulation of Se by plants must consider all of these factors to accurately calculate the mechanisms of uptake and translocation.  相似文献

Background and aims

Crop biofortification programs require fast, accurate and inexpensive methods of identifying nutrient dense genotypes. This study investigated energy-dispersive X-ray fluorescence spectrometry (EDXRF) for the measurement of zinc (Zn), iron (Fe) and selenium (Se) concentrations in whole grain wheat.

Methods

Grain samples were obtained from existing biofortification programs. Reference Zn, Fe and Se concentrations were obtained using inductively coupled plasma optical emission spectrometry (ICP-OES) and/or inductively coupled plasma mass spectrometry (ICP-MS). One set of 25 samples was used to calibrate for Zn (19–60?mg?kg–1) and Fe (26–41?mg?kg–1), with 25 further samples used to calibrate for Se (2–31?mg?kg–1 ). Calibrations were validated using an additional 40–50 wheat samples.

Results

EDXRF limits of quantification (LOQ) were estimated as 7, 3 and 2?mg?kg–1 for Zn, Fe, and Se, respectively. EDXRF results were highly correlated with ICP-OES or -MS values. Standard errors of EDXRF predictions were ±2.2?mg Zn kg–1, ±2.6?mg Fe kg–1, and ±1.5?mg Se kg–1.

Conclusion

EDXRF offers a fast and economical method for the assessment of Zn, Fe and Se concentration in wheat biofortification programs.  相似文献

Aims

A comparison was performed between plant species to determine if extractable, rather than total soil Se, is more effective at predicting plant Se accumulation over a full growing season.

Methods

Durum wheat (Triticum turgidum L.) and spring canola (Brassica napus L.) were sown in potted soil amended with 0, 0.1, 1.0, or 5.0 mg kg?1 Se as SeO4 2? or SeO3 2?. In addition, SeO4 2?-amended soils were amended with 0 or 50 mg kg?1 S as SO4 2?. Soils were analyzed for extractable and total concentration of Se ([Se]). Twice during the growing season plants were harvested and tissue [Se] was determined.

Results

Plants exposed to SeO3 2? accumulated the least Se. Fitted predictive models for whole plant accumulation based on extractable soil [Se] were similar to models based on total [Se] in soil (R2?=?0.73 or 0.74, respectively) and selenium speciation and soil [S] were important soil parameters to consider. As well, soil S amendments limited Se toxicity.

Conclusions

Soil quality guidelines (SQGs) based on extractable Se should be considered for risk assessment, particularly when Se speciation is unknown. Predictive models to estimate plant Se uptake should include soil S, a modifier of Se accumulation.  相似文献

Background

The aim of the present paper was to describe the enzymatic antioxidant system in Hymenolepis diminuta collected from rats exposed to chronic cestode invasion.

Methodology

We dissected different tissues of H. diminuta (immature proglottids, genital primordia, hermaphroditic proglottids, early uterus, and gravid uterus) and studied activity of: superoxide dismutases (SOD1 and SOD2), catalase (CAT), glutathione peroxidases (non-Se-dependent GSHPx and Se-dependent GSHPx), glutathione-S-transferase (GST) and glutathione reductase (GSHR), and oxidative stress markers ?? reduced glutathione (GSH), and the lipid peroxidation level (TBARS).

Results

We demonstrated changes in antioxidant enzyme activities and levels of oxidative stress markers in different tissues of the parasite. The levels of TBARS and GSH indicate that oxidative stress occurred in tissues located proximal to the intestine wall. Activity of SOD1 was high in all parts of H. diminuta, but the GST activity was the highest of all studied antioxidant enzymes. SOD2 activity differed significantly in various parts of H. diminuta. Significant differences were observed for nonSeGSHPx and activity of other GSH-dependent enzymes was generally similar in all the tissues.

Conclusions

Our results show that the enzymatic antioxidant system of H. diminuta, allows the parasite to adapt and live under conditions of chronic oxidative stress. It suggests an oxidative-antioxidative balance during interactions between parasite and host.  相似文献

Aims

The study aimed to find soil parameters that are best related to Se plant uptake for low Se soils with predominantly organic Se, and to explore the mechanisms that control Se bioavailability in the soils under study.

Methods

A pot experiment using nineteen soil samples taken from different fields of arable land (potato fields) in the Netherlands was conducted on summer wheat (Triticum aestivum L.). Selenium in wheat shoots and soil parameters, including basic soil properties, C:N ratio, inorganic selenite content, and Se and organic C in different soil extractions (0.01 M CaCl2, 0.43 M HNO3, hot water, ammonium oxalate, aqua regia) were analysed. Regression analysis was performed to identify soil parameters that determine Se content in wheat shoots.

Results

The regression model shows that Se:DOC ratio in 0.01 M CaCl2 soil extraction explained about 88 % of the variability of Se uptake in wheat shoots. Selenium uptake increased with Se:DOC ratio in CaCl2 extraction, which can be interpreted as a measure of the content of soluble Se-rich organic molecules. Selenium:DOC ratio in CaCl2 extraction and Se uptake increased towards higher soil pH and lower soil C:N ratio. The soil C:N ratio is also negatively correlated to Se:organic C ratio in other extractions (0.43 M HNO3, hot water, ammonium oxalate, aqua regia), indicating that at low soil C:N ratio soil organic matter is richer in Se. Contrarily, the soil pH is positively and strongly correlated to Se:organic C ratio in CaCl2 and hot water extractions, but only weakly correlated to Se:organic C ratio in other extractions.

Conclusions

Selenium-rich dissolved organic matter is the source of bioavailable Se in low Se soils with predominantly organic Se. The soil pH and quality of soil organic matter (i.e. soil C:N ratio) are the main soil properties determining Se bioavailability in these soil types.

This is the first report on genetic analysis and genome mapping of major dominant genes for near non-host resistance to barley crown rust ( Puccinia coronata var. hordei ) in common wheat.

Abstract

Barley crown rust, caused by Puccinia coronata var. hordei, primarily occurs on barley (Hordeum vulgare L.) in the Great Plain regions of the United States. However, a few genotypes of common wheat (Triticum aestivum L.) were susceptible to this pathogen among 750 wheat accessions evaluated. To investigate the genetics of crown rust resistance in wheat, a susceptible winter wheat accession PI 350005 was used in crosses with two resistant wheat varieties, Chinese Spring and Chris. Analysis of F1 plants and F2 populations from these two crosses indicated that crown rust resistance is controlled by one and two dominant genes in Chris and Chinese Spring, respectively. To determine the chromosome location of the resistance gene Cr1 in Chris, a set of 21 monosomic lines derived from Chris was used as female parents to cross with a susceptible spring type selection (SSTS35) derived from the PI 350005/Chris cross. Monosomic analysis indicated that Cr1 is located on chromosome 5D in Chris and one of the crown rust resistance genes is located on chromosome 2D in Chinese Spring. The other gene in Chinese Spring is not on 5D and thus is different from Cr1. Molecular linkage analysis and QTL mapping using a population of 136 doubled haploid lines derived from Chris/PI 350005 further positioned Cr1 between SSR markers Xwmc41-2 and Xgdm63 located on the long arm of chromosome 5D. Our study suggests that near non-host resistance to crown rust in these different common wheat genotypes is simply inherited.  相似文献

Genotypic variation in the ability of wheat roots to penetrate wax layers   总被引:1,自引:0,他引:1  

Background and aims

The role of the root system in mediating crop yields has recently been emphasised, resulting in several laboratory approaches for phenotyping root traits. We aimed to determine the existence of, and reasons for, genotypic variation in wheat (Triticum aestivum L.) root penetration of strong wax layers.

Methods

Three contrasting groups (UK elite lines, CIMMYT lines and near-isogenic lines of cv Mercia containing dwarfing and semi-dwarfing Rht alleles) comprising 18 different genotypes with contrasting phenologies were studied. We determined the ability of roots of these genotypes to penetrate strong wax layers and the angular spread of the root systems.

Results

There were no intrinsic differences in root system ability to penetrate strong wax layers (consistent with the similar root diameter of all lines) since greater root penetration was simply related to more root axes. Recording root penetration of concentric zones of the wax layer demonstrated that cv. Battalion had a root system with a smaller angular spread than cv. Robigus, which had the root system with the greatest angular spread.

Conclusions

There was limited genotypic variability in root penetration of strong layers within the wheat cultivars studied. A key challenge will be to determine the physiological and agronomic significance of the variation in root angular spread.  相似文献

Aims

Aluminum-tolerant wheat plants often produce more root exudates such as malate and phosphate than aluminum-sensitive ones under aluminum (Al) stress, which provides environmental differences for microorganism growth in their rhizosphere soils. This study investigated whether soil bacterial community composition and abundance can be affected by wheat plants with different Al tolerance.

Methods

Two wheat varieties, Atlas 66 (Al-tolerant) and Scout 66 (Al-sensitive), were grown for 60 days in acidic soils amended with or without CaCO3. Plant growth, soil pH, exchangeable Al content, bacterial community composition and abundance were investigated.

Results

Atlas 66 showed better growth and lower rhizosphere soil pH than Scout 66 irrespective of CaCO3 amendment or not, while there was no significant difference in the exchangeable Al content of rhizosphere soil between the two wheat lines. The dominant bacterial community composition and abundance in rhizosphere soils did not differ between Atlas 66 and Scout 66, although the bacterial abundance in rhizosphere soil of both wheat lines was significantly higher than that in bulk soil. Sphingobacteriales, Clostridiales, Burkholderiales and Acidobacteriales were the dominant bacteria phylotypes.

Conclusions

The difference in wheat Al tolerance does not induce the changes in the dominant bacterial community composition or abundance in the rhizosphere soils.  相似文献

Yield stability of hybrids versus lines in wheat,barley, and triticale   总被引:1,自引:0,他引:1  

Key message

We present experimental data for wheat, barley, and triticale suggesting that hybrids manifest on average higher yield stability than inbred lines.

Abstract

Yield stability is assumed to be higher for hybrids than for inbred lines, but experimental data proving this hypothesis is scarce for autogamous cereals. We used multi-location grain yield trials and compared the yield stability of hybrids versus lines for wheat (Triticum aestivum L.), barley (Hordeum vulgare L.), and triticale (×Triticosecale Wittmack). Our study comprised three phenotypic data sets of 1,749 wheat, 96 barley, and 130 triticale genotypes, which were evaluated for grain yield in up to five contrasting locations. Yield stability of the group of hybrids was compared with that of the group of inbred lines estimating the stability variance. For all three crops we observed a significantly (P < 0.05) higher yield stability of hybrids compared to lines. The enhanced yield stability of hybrids as compared to lines represents a major step forward, facilitating coping with the increasing abiotic stress expected from the predicted climate change.  相似文献

Background and aims

Production of indole-3-acetic acid (IAA) by Azospirillum brasilense is one of the most important mechanisms underlying the beneficial effects observed in plants after inoculation with this bacterium. This study determined the contribution of the hisC1 gene, which encodes aromatic amino acid aminotransferase-1 (AAT1), to IAA production, and analyzed its expression in the free-living state and in association with the roots of wheat.

Methods

We determined production of IAA and AAT activity in the mutant hisC::gusA-sm R . To study the expression of hisC1, a chromosomal gene fusion was analyzed by following β-glucuronidase (GUS) activity in vitro, in the presence of root exudates, and in association with roots.

Results

IAA production in the hisC mutant was not reduced significantly compared to the activity of the wild-type strain. AAT1 activity was reduced by 50% when tyrosine was used as the amino acid donor, whereas there was a 30% reduction when tryptophan was used, compared to the activity of the wild-type strain. Expression of the fusion protein was up-regulated in both logarithmic and stationary phases by several compounds, including IAA, tryptophan, tyrosine, and phenyl acetic acid. We observed the expression of hisC1 in bacteria associated with wheat roots. Root exudates of wheat and maize were able to stimulate hisC1 expression.

Conclusions

The expression data indicate that hisC1 is under a positive feedback control in the presence of root exudates and on plants, suggesting that AAT1 activity plays a role in Azospirillum–plant interactions.  相似文献

Aims

Typha latifolia L. is an aquatic plant that has been widely exploited for the aims of phytoremediation. The main reason why we have chosen this plant species for the current study is its capacity to accumulate and detoxify heavy metals. The main topic of the investigation focused on the root uptake of copper (II) nitrate and copper (II) sulfate and the impact of different chemical copper species on the excreted organic acids.

Methods

Oxalic, malic, acetic and lactic acids were determined using capillary electrophoresis; a comparison between the concentration and the time course during 7 days of treatment was performed.

Results

There is a correlation between the total copper (II) sulfate concentration in the roots and the total amount of the excreted organic acids. In addition to that organic acids are involved in the detoxification mechanisms of Typha latifolia for copper (II) nitrate and copper (II) sulfate.

Conclusions

Different from so far investigated plant species the highest amounts of organic acids are excreted from T. latifolia roots not in the first hours after treatment, but up to 7 days later.  相似文献

Aims

The purpose of this study was to test the hypotheses that soil nutrient patchiness can differentially benefit the decomposition of root and shoot litters and that this facilitation depends on plant genotypes.

Methods

We grew 15 cultivars (i.e. genotypes) of winter wheat (Triticum aestivum L.) under uniform and patchy soil nutrients, and contrasted their biomass and the subsequent mass, carbon (C) and nitrogen (N) dynamics of their root and shoot litters.

Results

Under equal amounts of nutrients, patchy distribution increased root biomass and had no effects on shoot biomass and C:N ratios of roots and shoots. Roots and shoots decomposed more rapidly in patchy nutrients than in uniform nutrients, and reductions in root and shoot C:N ratios with decomposition were greater in patchy nutrients than uniform nutrients. Soil nutrient patchiness facilitated shoot decomposition more than root decomposition. The changes in C:N ratios with decomposition were correlated with initial C:N ratios of litter, regardless of roots or shoots. Litter potential yield, quality and decomposition were also affected by T. aestivum cultivars and their interactions with nutrient patchiness.

Conclusions

Soil nutrient patchiness can enhance C and N cycling and this effect depends strongly on genotypes of T. aestivum. Soil nutrient heterogeneity in plant communities also can enhance diversity in litter decomposition and associated biochemical and biological dynamics in the soil.  相似文献

Aims

We aimed to identify plant growth-promoting rhizobacteria that could be used to develop a biofertilizer for rice.

Methods

To obtain plant growth-promoting rhizobacteria, rhizosphere soils from different crops (rice, wheat, oats, crabgrass, maize, ryegrass, and sweet potato) were inoculated to rice plants. In total, 166 different bacteria were isolated and their plant growth-promoting traits were evaluated in terms of colony morphology, indole-3-acetic acid production, acetylene reduction activity, and phosphate solubilization activity. Moreover, genetic analysis was carried out to evaluate their phylogenetic relationships based on 16S rRNA sequence data.

Results

Strains of Bacillus altitudinis, Pseudomonas monteilii, and Pseudomonas mandelii formed associations with rice plants and fixed nitrogen. A strain of Rhizobium daejeonense showed nitrogen fixation activity in an in vitro assay and in vivo. Strains of B. altitudinis and R. daejeonense derived from rice rhizosphere soil, strains of P. monteilii and Enterobacter cloacae derived from wheat rhizosphere soil, and a strain of Bacillus pumilus derived from maize rhizosphere soil significantly promoted rice plant growth.

Conclusions

These methods are effective to identify candidate species that could be developed as biofertilizers for target crops.  相似文献

Polyunsaturated fatty acids (PUFAs) occur in phospholipids of synapses of central nervous system (CNS). PUFAs may thus determine the fluidity of synaptosomal membranes and regulate neuronal transmission. It was therefore tempting to suggest an oxidative system in CNS protecting the membrane function, e.g., glutathione peroxidase (GSH-Px). In order to trace GSH-Px Wistar rats were loaded with 4800 kBq of75Se sodium selenite. By means of gradient ultracentrifugation, particulate fractions of CNS were isolated and radioactivity as well as selenium dependent GSH-Px were estimated. The following data were obtained:
  1. All fractions (myelin, synaptic vesicles, synaptosomes, mitochondria, and microsomes) contained75Se.
  2. After acetone precipitation of GSH-Px activity, fractionation on Sephadex G-150 revealed in all particulate fractions at least two peaks of radioactivity with GSH-Px activity.
  3. The two GSH-Px peaks from the Sephadex filtration were freeze dried and applied on a hydrophobic T-gel column and eluted with decreasing molarity of ammonium sulfate from 1.5 to 0.05M. The first Sephadex peak with GSH-Px activity from myelin and the second peak with GSH-Px activity from synaptic vesicles could now be resolved into two different fractions of radioactivity on the T-gel. The remaining Sephadex G-150 peaks could only be resolved into one peak of radioactivity.
  4. SDS-polyacrylamide gel electrophoresis of the T-gel peaks from all fractions showed a protein band with a mobility identical with that of human erythrocyte GSH-Px. The T-gel elution of myelin, synaptic vesicles and mitochondria gave rise to nearly pure CNS GSH-Px activity.
. The data presented support the idea that CNS fractions have membrane bound GSH-Px activity that may function as protecting enzymes towards oxidative stress in the brain.  相似文献

Wheat stem rust resistance gene SrWeb is an allele at the Sr9 locus that confers resistance to Ug99.

Abstract

Race TTKSK (Ug99) of Puccinia graminis f. sp. tritici, the causal fungus of stem rust, threatens global wheat production because of its broad virulence to current wheat cultivars. A recently identified Ug99 resistance gene from cultivar Webster, temporarily designated as SrWeb, mapped near the stem rust resistance gene locus Sr9. We determined that SrWeb is also present in Ug99 resistant cultivar Gabo 56 by comparative mapping and an allelism test. Analysis of resistance in a population segregating for both Sr9e and SrWeb demonstrated that SrWeb is an allele at the Sr9 locus, which subsequently was designated as Sr9h. Webster and Gabo 56 were susceptible to the Ug99-related race TTKSF+ from South Africa. Race TTKSF+ possesses unique virulence to uncharacterized Ug99 resistance in cultivar Matlabas. This result validated that resistance to Ug99 in Webster and Gabo 56 is conferred by the same gene: Sr9h. The emergence of pathogen virulence to several resistance genes that are effective to the original Ug99 race TTKSK, including Sr9h, suggests that resistance genes should be used in combinations in order to increase resistance durability.  相似文献

We report a new stripe rust resistance gene on chromosome 7AS in wheat and molecular markers useful for transferring it to other wheat genotypes.

Abstract

Several new races of the stripe rust pathogen have established throughout the wheat growing regions of China in recent years. These new races are virulent to most of the designated seedling resistance genes limiting the resistance sources. It is necessary to identify new genes for diversification and for pyramiding different resistance genes in order to achieve more durable resistance. We report here the identification of a new resistance gene, designated as Yr61, in Chinese wheat cultivar Pindong 34. A mapping population of 208 F2 plants and 128 derived F2:3 lines in a cross between Mingxian 169 and Pindong 34 was evaluated for seedling stripe rust response. A genetic map consisting of eight resistance gene analog polymorphism (RGAP), two sequence-tagged site (STS) and four simple sequence repeat (SSR) markers was constructed. Yr61 was located on the short arm of chromosome 7A and flanked by RGAP markers Xwgp5467 and Xwgp5765 about 1.9 and 3.9 cM in distance, which were successfully converted into STS markers STS5467 and STS5765b, respectively. The flanking STS markers could be used for marker-assisted selection of Yr61 in breeding programs.  相似文献

Fertile hybrids were produced with genetic material transferred from Th. intermedium into a wheat background and supply a source of genetic variation to wheat improvement.

Abstract

Both symmetric and asymmetric somatic hybrids have been obtained from the combination of wheatgrass (Thinopyrum intermedium) and bread wheat (Triticum aestivum). Two wheat protoplast populations, one derived from embryogenic calli and the other from a non-regenerable, rapidly dividing cell line, were fused with Th. intermedium protoplasts which had been (or not been) pre-irradiated with UV. Among the 124 regenerated calli, 64 could be categorized as being of hybrid origin on the basis of plant morphology, peroxidase isozyme, RAPD DNA profiling and karyological analysis. Numerous green plantlets were regenerated from 13 calli recovered from either the symmetric hybrid (no UV pre-treatment) or the asymmetric one (30 s UV irradiation). One of these hybrid plants proved to be vigorous and self-fertile. The regenerants were all closer in phenotype to wheat than to Th. intermedium. Genomic in situ hybridization analysis showed that the chromosomes in the hybrids were largely intact wheat ones, although a few Th. intermedium chromosome fragments had been incorporated within them.  相似文献
