Summary Alkaline phosphatase and peroxidase activities were investigated in typical neutrophils and in vacuolated cells, the latter considered to be another type of neutrophil in catfish blood. This study was carried out by light- and electron-microscopy. Two lines of neutrophils were recognized in the peripheral blood of catfish. Alkaline phosphatase activity was present only in the vacuolated neutrophils; peroxidase activity appeared only in the typical neutrophils. The presence of two types of neutrophil in catfish blood suggests that important steps of neutrophil evolution occurred in fish.  相似文献

The variability of the bioelectric field of the electrosensitive catfish, Ictalurus nebulosus, was investigated by recording the potential variation occurring when the fish passed a stationary electrode, and by recording the field of a stationary fish by a 15-electrode array. A good first order approximation of the recorded field of a 20 cm long fish is a dipole dc source with the source and sink about 7 cm apart, carrying a current of about 1 microA in water with a specific resistivity of 3.3 kohm cm. At 5 cm distance from the dipole axis such a source generates an electric potential swing in the order of 50 microV in free space, head negative, tail positive. Superimposed on the basic component are respiration related fluctuations, and fluctuations related to the activity of the alimentary canal, gills, and skin. Novel stimuli, or stressors like investigators approaching the aquarium, evoke sudden increases in field strength. which last about 15 min. Demineralization of the aquarium water causes changes in field strength and reversal of field polarity. The administration of food causes field variations in the vicinity of the anal opening. The bioelectric field shows diurnal fluctuations of 100 microV. The peak is at about 04:00, the dip at 14:00. The fluctuations of the bioelectric field are sufficiently strong and specilic to serve as electrical stimuli to other electrosensitive catfish. It is suggested that the field changes allow a simple form of electrocommunication. i.e. inform conspecifics about some physiological properties of the field source. The cellular mechanisms underlying the fluctuations of the bioelectric field are homeostatic processes mediated by ion pumps and ion channels.  相似文献

We performed the first combined‐data phylogenetic analysis of ictalurids including most living and fossil species. We sampled 56 extant species and 16 fossil species representing outgroups, the seven living genera, and the extinct genus ?Astephus long thought to be an ictalurid. In total, 209 morphological characters were curated and illustrated in MorphoBank from published and original work, and standardized using reductive coding. Molecular sequences harvested from GenBank for one nuclear and four mitochondrial genes were combined with the morphological data for total evidence analysis. Parsimony analysis recovers a crown clade Ictaluridae composed of seven living genera and numerous extinct species. The oldest ictalurid fossils are the Late Eocene members of Ameiurus and Ictalurus. The fossil clade ?Astephus placed outside of Ictaluridae and not as its sister taxon. Previous morphological phylogenetic studies of Ictaluridae hypothesized convergent evolution of troglobitic features among the subterranean species. In contrast, we found morphological evidence to support a single clade of the four troglobitic species, the sister taxon of all ictalurids. This result holds whether fossils are included or not. Some previously published clock‐based age estimates closely approximate our minimum ages of clades.  相似文献

Unit responses in the acoustic-lateral region of the medulla to electrical and mechanical stimulation of the lateral line organs were investigated in acute experiments on curarized catfish. Of the total number of neurons 70% possessed spontaneous activity. An electrical stimulus evoked a tonic response both in spontaneously active and in "silent" cells. Three main types of firing pattern of the neurons were distinguished: fast-adapting, slow-adapting, and grouped. As regards the relation of the neurons to polarity of the stimulus they were subdivided into two groups. The thresholds of unit responses to electrical stimulation varied considerably: from 2.5·10–9 to 5·10–12 A/mm2. The effect of intensity of the electrical stimulation on unit responses in the medulla is analyzed. The precise dependence of on- and off-responses of each neuron on stimulus intensity of any polarity was determined. The neurons were shown to be sensitive to both electrical and mechanical stimuli. It is postulated that this phenomenon is due to convergence of impulses from electrical and mechanical receptors of the lateral line on the neurons. Ampullary electroreceptors of the freshwater catfish, Ictalurus nebulosus, were examined for the effects of bath-applied l-glutamate (l-GLU) and its agonists quisqualate (Q), kainate (KA) and n-methyl-d-aspartate (NMDA). Resting discharge rate and stimulus-evoked activity in single afferent fibers were recorded in an in vitro preparation.2. l-GLU (0.1–1 mM), Q (0.8–40 μM) and KA (10–250 μM) strongly increased both resting activity and stimulus-evoked activity in the afferent fibres. NMDA had no effect, even at a concentration of 5.0 mM.3. The potencies of l-GLU and its agonists, arbitrary defined as the concentrations which gave 50% of maximal frequency increase, were of the order of 7 μM (Q), 25 μM (KA) and 0.6 mM (l-GLU).4. The excitatory effects of l-GLU persisted in receptors suppressed by high Mg2+, indicating that l-GLU was acting at the postsynaptic site.5. The data presented are consistent with our current concept that the action of l-GLU in Ictalurus electroreceptors is mediated via Q/AMPA- and KA-types, but not the NMDA-type, of membrane receptors.  相似文献

By means of recording the cardiac rhythm, respiration rate and motor activity in unrestrained animals, studies have been made on the dynamics of formation of cataleptic-like immobilization in the teleost I. nebulosus. Rhythmic illumination at a rate 3/min. (10 sec light on - 10 sec light off) in the daytime gradually caused the onset of immobilization in the fish. This immobilization is characterized by the specific heart rate, type of respiration and the degree of plastic tone. Under the same experimental conditions, the formation of photogenic catalepsy in the frog Rana temporaria was confirmed. Typical heart rate, respiration pattern and the degree of plastic tone were determined as well. The role of illumination rhythm in the onset of all conditions which are characteristic for the "awakefulness-rest" cycle is discussed.  相似文献

The use of venom to subdue prey or deter predators has evolved multiple times in numerous animal lineages. Catfishes represent one of the most easily recognized, but least studied groups of venomous fishes. Venom glands surround spines on the dorsal and pectoral fins that serve as venom delivery structures. Species of madtom catfishes in the genus Noturus were found to each have one of four venom delivery morphologies: (1) smooth spine with no venom gland; (2) smooth spine with venom gland associated with shaft of spine; (3) serrated spine with venom gland associated with shaft of spine; and (4) serrated spine with venom gland associated with shaft of spine and posterior serrations. Analyses accounting for the phylogenetic history of Noturus species suggest that a serrated pectoral spine with a venom gland is the ancestral condition for the genus. The presence of serrations and a venom gland have been largely conserved among Noturus species, but sting morphology has changed at least five times within the genus. Four of these changes have resulted in a loss of morphological complexity, including the loss of posterior serrations, loss of venom glands associated with the posterior serrations, and one complete loss of the venom gland. © 2010 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 102 , 115–129.  相似文献

The receptor cells of the ampullary electroreceptor organs of Ictalurus nebulosus bear microvilli on the apical membrane. Whereas microvilli in mechanoreceptive hair cells and in chemoreceptor cells have a transduction function, the function of these membrane specializations in electroreceptor cells is not fully understood. To study the role of the microvilli of the electroreceptor cells, the ampullary electroreceptor organs were apically exposed to the microfilament-disrupting agents cytochalasin B and D. Electrophysiological measurements showed that cytochalasin caused a high decrease in sensitivity and a slight decrease in spontaneous activity. Exposure to cytochalasin B resulted in a striking disorganization of the microvilli on the apical membrane of the electroreceptor cells. The most plausible explanation for the results is that treatment with cytochalasin mainly affects the actin filaments of the microvilli causing an increase of the resistance of the apical membrane. A high apical resistance results in a decrease of the voltage over the basal membrane, which in turn reduces the sensitivity. The conclusion is that intact apical microvilli are necessary for proper functioning of ampullary electroreceptor organs. Alterations in microvillar properties, like surface area and ion channel conductancy might play a considerable role in the regulation of the sensitivity.  相似文献

The mitochondrial cytochrome b gene was sequenced for six individuals of the pygmy madtom, Noturus stanauli, a globally imperilled catfish, from both known localities in Tennessee (U.S.A.) separated by over 1055 river km. By means of evoked potential methods, a lateral line center in the thalamus of the bullhead catfish is here identified. The locus of this lateral line center corresponds to the mechanoreceptive thalamic zone and/or torothalamic tract identified by anatomical means. The thalamic lateral line area responds to acoustic stimuli as well as lateral line nerve shock but fails to respond to electroreceptor inputs that do cause responses in their special part of the midbrain torus semicircularis. The latency of the first peak of response to lateral line nerve shock, which is also the main response peak, is 30 ms for the thalamic zone compared to 15 ms for the earliest peak response in the mechanoreceptive part of the torus semicircularis. The thalamic response fatigues much more quickly than the toral response and has different dynamic properties to closely spaced stimulus pairs as well.  相似文献

Mannose-binding C-type lectin (MBL) was isolated from channel catfish (Ictalurus punctatus) NWAC 102 and 103 strains, blue catfish (Ictalurus furcatus) D+B and Rio Grande strains, hybrid catfish (channel catfish female NWAC 103 x blue catfish male D+B) sera, and purified by affinity chromatography from channel catfish Norris strain serum. Reduction of purified channel catfish MBL with 2-ME yielded a single band of 62 kDa by SDS-PAGE and Western blot analysis using guinea pig anti-MBL IgG as primary antibody. Channel catfish NWAC 102 strain, channel catfish NWAC 103 strain and hybrid catfish sera had molecular masses of 63 kDa for MBL. Blue catfish (D+B strain) serum MBL had a molecular mass of 66 kDa. Rio Grande blue catfish serum MBL had a molecular mass of 65 kDa. Amino acid composition analysis (mol%) of the affinity-purified channel catfish MBL found a high content of serine present. Functional binding studies of channel catfish and blue catfish MBLs binding to Edwardsiella ictaluri were done using a dot-immunoblot ELISA method. A dot-immunoblot ELISA binding assay was done to compare nine different strains and species of channel catfish and blue catfish for their levels of serum MBL. Blue catfish had higher levels of MBL than did the various strains of channel catfish tested. MBL could be used as a genetic marker for selection of disease resistance in the different strains of catfish used in aquaculture. This study describes the presence of serum MBL in catfish and evidence for a C-type lectin complement pathway of innate immunity.  相似文献

Channel catfish, Ictalurus punctatus , exhibited passive cutaneous anaphylaxis (PCA) when sensitized with Flexibacter columnaris antiserum produced in channel catfish (the first record of PCA in any fish), but not when sensitized with homologous antiserum against channel catfish virus. Stomach contents of 124 white catfish, Ictalurus catus, ranging from 51 to 431 mm collected from farm ponds of Auburn University, Auburn, Alabama were examined. Fishes in these ponds were given supplemental feed six days a week. Juvenile catfish below 100 mm fed more actively on a variety of food items than larger fish. White catfish measuring 201 mm and above preferred dipterans to other natural food items. In all size groups supplemental feed occupied a dominant position when compared to other food items. Other food items present in the stomachs in varying quantities included detritus, micro-crustaceans, trichopterans, fish remains and miscellaneous items, thus indicating the omnivorous feeding of white catfish.  相似文献
