We tested the hypothesis that the degree of anisotropic expansion of plant tissues is controlled by the degree of alignment of cortical microtubules or cellulose microfibrils. Previously, for the primary root of maize (Zea mays L.), we quantified spatial profiles of expansion rate in length, radius, and circumference and the degree of growth anisotropy separately for the stele and cortex, as roots became thinner with time from germination or in response to low water potential (B.M. Liang, A.M. Dennings, R.E. Sharp, T.I. Baskin [1997] Plant Physiol 115:101–111). Here, for the same material, we quantified microtubule alignment with indirect immunofluorescence microscopy and microfibril alignment throughout the cell wall with polarized-light microscopy and from the innermost cell wall layer with electron microscopy. Throughout much of the growth zone, mean orientations of microtubules and microfibrils were transverse, consistent with their parallel alignment specifying the direction of maximal expansion rate (i.e. elongation). However, where microtubule alignment became helical, microfibrils often made helices of opposite handedness, showing that parallelism between these elements was not required for helical orientations. Finally, contrary to the hypothesis, the degree of growth anisotropy was not correlated with the degree of alignment of either microtubules or microfibrils. The mechanisms plants use to specify radial and tangential expansion rates remain uncharacterized.  相似文献

Clark-type oxygen microelectrodes were used to measure the radial and longitudinal oxygen distribution in aerenchymatous and nonaerenchymatous primary roots of intact maize seedlings. A radial intake of oxygen from the rooting medium was restricted by embedding the roots in 1% agar causing aeration to be largely dependent upon longitudinal internal transport from the shoot. In both root types, oxygen concentrations declined with distance from the base, and were lower in the stele than in the cortex. Also, the bulk of the oxygen demand was met internally by transport from the shoots, but a little oxygen was received by radial inward diffusion from the surrounding agar, and in some positions the hypodermal layers received oxygen from both the agar and the cortex. Near to the base, the oxygen partial pressure difference between the cortex and the center of the stele could be as much as 6–8 kPa. Nearer to the tip, the differences were smaller but equally significant. In the nonaerenchymatous roots, cortical oxygen partial pressures near the apex were becoming very low (< 1 kPa) as root lengths approached 100 mm, and towards the center of the stele values reached 0.1 kPa or lower. However, the data indicated that respiratory activity did not decline until the cortical oxygen pressure was less than 2 kPa. Mathematical modeling based on Michaelis–Menten kinetics supported this and suggested that the respiratory decline would be mostly restricted to the stele until cortical oxygen pressures approached very low values. At a cortical oxygen pressure of 0.75 kPa, it was shown that respiratory activity in the pericycle and phloem might remain as high as 80–100% of maximum even though in the center of the stele it could be less than 1% of maximum. Aerenchyma production resulted in increases in oxygen concentration throughout the roots with cortical partial pressures of ca. 5–6 kPa and stelar values of ca. 3–4 kPa near the tips of 100 mm long roots. In aerenchymatous roots, there was some evidence of a decline in the oxygen permeability of the epidermal–hypodermal cylinder close to the apex; a decline in stelar oxygen permeability near the base was indicated for both root types. There was some evidence that the mesocotyl and coleoptile represented a very significant resistance to oxygen transport to the root.  相似文献

The Radial and Longitudinal Path of Ion Movement in Roots   总被引:4,自引:0,他引:4  
The existence of a barrier to lateral outward diffusion of ions from roots was demonstrated quantitatively and by autoradiography. Ions applied to the apical zone were excreted through the basal cut end with no lateral outward diffusion from the central zone. The ions moving through the conducting tissues showed no leakage to the external solution regardless whether the treatment roots came from seedlings grown under low or high salt conditions. However, when dinitrophenol (DNP) was also applied to the central zone in the external salt solution, leakage of apically applied calcium occurred from the conducting tissues. Autoradiographic studies with labeled calcium suggested that the endodermal layer was an effective barrier preventing the lateral outward diffusion of ions. The ions moved longitudinally through the stele. In the stelar tissues the autoradiographic studies failed to detect the presence of any radioactive calcium in the central duct and in the mature xylem vessels, although high concentrations of labeled ions were found in the living cells of the stele, particularly the xylem parenchyma. It is suggested that xylem parenchyma cells may be involved in the longitudinal transport of ions.  相似文献

Epidermal strips from either well-watered or water-stressedplants of Commelina communis L. were subjected to a range ofABA concentrations (10–6–10–3 mol m–3)in the presence (330 parts 10–6 in air) or virtual absence(3 parts 10–6 in air) of CO2. The stomatal response toCO2 was greater in epidermis from water-stressed plants, althoughthere was a distinct CO2 response in epidermis from well-wateredplants. Additions of ABA via the incubation medium had littleeffect on the relative CO2 response. Stomata responded to ABAboth in the presence and virtual absence of CO2, but the relativeresponse to ABA was greatest in the high CO2 treatment. Whenwell-watered plants were sprayed with a 10–1 mol m–3ABA solution 1 d prior to use, the stomatal response of detachedepidermis to both CO2 and ABA was very similar to that of epidermisdetached from water-stressed leaves. It is hypothesized thata prolonged exposure to ABA is necessary before there is anymodification of the CO2 response of stomata.  相似文献

A root pressure probe was employed to measure hydraulic properties of primary roots of maize (Zea mays L.). The hydraulic conductivity (Lpr) of intact root segments was determined by applying gradients of hydrostatic and osmotic pressure across the root cylinder. In hydrostatic experiments, Lpr was constant along the segment except for an apical zone of approximately 20 millimeters in length which was hydraulically isolated due to a high axial resistance. In osmotic experiments, Lpr decreased toward the base of the roots. Lpr (osmotic) was significantly smaller than Lpr (hydrostatic). At various distances from the root tip, the axial hydraulic resistance per unit root length (Rx) was measured either by perfusing excised root segments or was estimated according to Poiseuille's law from cross-sections. The calculated Rx was smaller than the measured Rx by a factor of 2 to 5. Axial resistance varied with the distance from the apex due to the differentiation of early metaxylem vessels. Except for the apical 20 millimeters, radial water movement was limiting water uptake into the root. This is important for the evaluation of Lpr of roots from root pressure relaxations. Stationary water uptake into the roots was modeled using measured values of axial and radial hydraulic resistances in order to work out profiles of axial water flow and xylem water potentials.  相似文献

We have analyzed the way in which temperature affects leaf elongation rate of maize (Zea mays L.) leaves, while spatial distributions (observed at a given time) of cell length and of proportion of cells in DNA replication are unaffected. We have evaluated, in six growth chamber experiments with constant temperatures (from 13 to 34[deg]C) and two field experiments with fluctuating temperatures, (a) the spatial distributions of cell length and of leaf elongation rate, and (b) the distribution of cell division, either by using the continuity equation or by flow cytometry. Leaf elongation rate was closely related to meristem temperature, with a common relationship in the field and in the growth chamber. Cell division and cell elongation occurred in the first 20 and 60 mm after the ligule, respectively, at all temperatures. Similar quantitative responses to temperature were observed for local cell division and local tissue expansion rates (common x intercept and normalized slope), and both responses were spatially uniform over the whole expanding zone (common time courses in thermal time). As a consequence, faster cell elongation matched faster cell division rate and faster elongation was compensated for by faster cell displacement, resulting in temperature-invariant profiles of cell length and of proportion of dividing cells. Cell-to-cell communication, therefore, was not necessary to account for coordination.  相似文献

Pea roots and maize mesocotyl segments retain polyribosomeswhen subjected to mild and moderate water stress. Even underwater-stress conditions that are lethal, i.e. where there isno recovery of protein synthesis upon return to full hydration,some polyribosomes are retained. Therefore cessation of proteinsynthesis under water stress conditions is not due to the inabilityof cells to retain their polyribosomes. This observation contradictsseveral previous reports. The polynbosome content of PEG-stressedroots is lower, at equivalent water potentials, than that ofair-dried roots. We suggest that the capacity of a higher planttissue to withstand water stress cannot easily be correlatedwith its capacity to retain polyribosomes.  相似文献

The oncogenic roles of PDGF-D and its proteolytic activator, matriptase, have been strongly implicated in human prostate cancer. Latent full-length PDGF-D (FL-D) consists of a CUB domain, a growth factor domain (GFD), and the hinge region in between. Matriptase processes the FL-D dimer into a GFD dimer (GFD-D) in a stepwise manner, involving generation of a hemidimer (HD), an intermediate product containing one FL-D subunit and one GFD subunit. Although the HD is a pro-growth factor that can be processed into the GFD-D by matriptase, the HD can also act as a dominant-negative ligand that prevents PDGF-B-mediated β-PDGF receptor activation in fibroblasts. The active GFD-D can be further cleaved into a smaller and yet inactive form if matriptase-mediated proteolysis persists. Through mutagenesis and functional analyses, we found that the R340R341GR343A (P4–P1/P1′) motif within the GFD is the matriptase cleavage site through which matriptase can deactivate PDGF-D. Comparative sequence analysis based on the published crystal structure of PDGF-B predicted that the matriptase cleavage site R340R341GR343A is within loop III of the GFD, a critical structural element for its binding with the β-PDGF receptor. Interestingly, we also found that matriptase processing regulates the deposition of PDGF-D dimer species into the extracellular matrix (ECM) with increased binding from the FL-D dimer, to the HD, and to the GFD-D. Furthermore, we provide evidence that R340R341GR343A within the GFD is critical for PDGF-D deposition and binding to the ECM. In this study, we report a structural element crucial for the biological function and ECM deposition of PDGF-D and provide molecular insight into the dynamic functional interplay between the serine protease matriptase and PDGF-D.  相似文献

Radial transport of water in primary maize roots has been studiedby following the kinetics of the H2O-D2O exchange using protonmagnetic resonance. The method is based on the use of an impermeantrelaxation agent, dextran-magnetite, which made possible separationof the surrounding proton signal from the root water protonsignal. Two pulsed NMR techniques for continuously followingthe exchange are described. The exchange curve was fitted toseveral theoretical models in order to evaluate the dominantradial water pathway in roots. It was found that the choiceof the appropriate model could not be made solely on the basisof goodness of fit and thus the previously calculated valueof membrane water permeability, which is not dependent on thewater transport pathways, was used as an additional testingcriterion. It is concluded that the major radial water fluxin root occurs through the cells, and not through the intercellularspace, as previously assumed. The relatively low calculatedapparent diffusion coefficient (D' = 3 x 10–6 cm2 s–1)of water is mostly determined by water permeation through aseries of membranes. A minor fraction of water in roots (10–15%),exchanged at a slower rate than the rate at which bulk diffusionwould allow and was assumed to correspond to the water havingrestricted mobility due to the influence of macro-molecules.The NMR measurements of relaxation of water in roots also suggestdifferent water fractions and it is concluded, from both experimentalapproaches, that most of the cytoplasmic water has significantlydifferent mobility from the bulk water. The effects of three ranges of CO2 concentration on growth,carbon distribution and loss of carbon from the roots of maizegrown for 14 d and 28 d with shoots in constant specific activity14CO2 are described. Increasing concentrations of CO2 led toenhancement of plant growth with the relative growth rate (RGR)of the roots affected more than the RGR of the shoots. Between16% and 21% of total net fixed carbon (defined as 14C retainedin the plant plus 14C lost from the root) was lost from theroots at all CO2 concentrations at all times but the amountsof carbon lost per unit weight of plant decreased with time.Possible mechanisms to account for these observations are discussed. The sensitivity of homoserine dehydrogenase (EC 1.1.1.3) to inhibition by the feed-back modifier, l-threonine, was examined in preparations derived from etiolated shoots, roots, and lightgrown tissues of Zea mays L. var. earliking. A progressive decrease in enzyme sensitivity was observed during seedling growth. Enzyme derived from internode tissue retained a greater sensitivity to the effector than enzyme derived from apical portions of etiolated shoots, whereas enzyme from root tips was characteristically more sensitive than that prepared from mature cells of the root. Enzyme desensitization occurred rapidly during culture of excised shoots and the activities of both homoserine dehydrogenase and aspartokinase (EC 2.7.2.4) declined during shoot culture under a variety of conditions. The initial enzyme levels and the characteristic sensitivity of homoserine dehydrogenase were preserved during culture at 5 to 7 C, but desensitization was not prevented by inclusion of cycloheximide in the culture medium.Results of control experiments provide evidence that desensitization occurs in vivo. No alteration of the enzyme properties was detected during extraction or concentration of sensitive or insensitive enzyme or during coextraction of enzyme from mixed populations of different age shoots; nor was a differential distribution of inhibitors or activators indicated during assay of mixed preparations. The change in enzyme sensitivity was apparent under a variety of assay conditions and was not accompanied by changes in the apparent affinity of the enzyme for the substrate, homoserine. It is suggested that systematic changes in the regulatory characteristics of certain enzymes could be an important level of metabolic regulation during cellular differentiation.Three forms of maize homoserine dehydrogenaase were detected after acrylamide gel electrophoresis of samples derived from 72-hr shoots. Similar analysis of samples from older shoots revealed a broad asymmetric band of enzyme activity, suggesting that changes in the relative distribution of specific forms of the enzyme could be related to the growth-dependent changes in the sensitivity of maize homoserine dehydrogenase.  相似文献

A relatively simple and reproducible technique to analyse both longitudinal and transversal biochemical microgradients in roots has been presented. Although no statistically significant differences of acid phosphomonoestherase activity were found between upper and lower faces of geostimulated maize roots, an increase of activity was demonstrated in the two first mm of the horizontal roots in comparison with the vertical roots. The spatial distribution of the root system through the soil profile has an impact on moisture and nutrient uptake by plants, affecting growth and productivity. The spatial distribution of the roots, soil moisture, and fertility are affected by tillage practices. The combination of high soil density and the presence of a soil plow pan typically impede the growth of maize (Zea mays L.).We investigated the spatial distribution coordination of the root system, soil moisture, and N status in response to different soil tillage treatments (NT: no-tillage, RT: rotary-tillage, SS: subsoiling) and the subsequent impact on maize yield, and identify yield-increasing mechanisms and optimal soil tillage management practices. Field experiments were conducted on the Huang-Huai-Hai plain in China during 2011 and 2012. The SS and RT treatments significantly reduced soil bulk density in the top 0–20 cm layer of the soil profile, while SS significantly decreased soil bulk density in the 20–30 cm layer. Soil moisture in the 20–50 cm profile layer was significantly higher for the SS treatment compared to the RT and NT treatment. In the 0-20 cm topsoil layer, the NT treatment had higher soil moisture than the SS and RT treatments. Root length density of the SS treatment was significantly greater than density of the RT and NT treatments, as soil depth increased. Soil moisture was reduced in the soil profile where root concentration was high. SS had greater soil moisture depletion and a more concentration root system than RT and NT in deep soil. Our results suggest that the SS treatment improved the spatial distribution of root density, soil moisture and N states, thereby promoting the absorption of soil moisture and reducing N leaching via the root system in the 20–50 cm layer of the profile. Within the context of the SS treatment, a root architecture densely distributed deep into the soil profile, played a pivotal role in plants’ ability to access nutrients and water. An optimal combination of deeper deployment of roots and resource (water and N) availability was realized where the soil was prone to leaching. The correlation between the depletion of resources and distribution of patchy roots endorsed the SS tillage practice. It resulted in significantly greater post-silking biomass and grain yield compared to the RT and NT treatments, for summer maize on the Huang-Huai-Hai plain.  相似文献

Water flows are described in root segments from which the centralstele had been removed (‘sleeves’). The water flowwas measured either as an exchange flow, using tracers, or asnet osmotic flow. Metabolic inhibitors inhibited both typesof flow. From the above measurements, the over-all hydraulicconductivity (Lp) and permeability coefficient () for waterwere calculated. By measuring the effect of water drag on theflow of sucrose and urea, reflection coefficients () were calculatedfor sucrose and urea. The phytotoxic effects of lead (Pb) on seed germinability, seedling growth, photosynthetic performance, and nutrient accumulation (K(+) and Cu(2+)) in two maize genotypes (EV-1098 and EV-77) treated with varying levels of PbSO(4) (0.01, 0.1, and 1.0 mg L(-1)) were appraised in this study. In the seed germination experiment, lead stress significantly reduced seed germination percentage and index, plumule and radicle lengths as well as fresh and dry weights in both genotypes. In the second experiment, lengths and fresh and dry weights of shoots and roots decreased due to Pb in both genotypes with increase in plant age. Higher Pb levels also decreased photosynthetic rate (A), water use efficiency (A/E), and intrinsic water use efficiency (A/g(s)), but increased transpiration rate (E) and C(i)/C(a) ratio as a result of increase in stomatal conductance (g(s)). The concentrations of K(+) and Cu(2+) decreased in root, stem, and leaves of both genotypes, which could be a direct consequence of multifold increase in Pb concentration in these tissues. Overall, cv. EV-1098 had better Pb tolerance potential than EV-77 because the former genotype showed less reduction in seed germinability parameters, photosynthetic performance, and K(+) and Cu(2+) accumulation in shoot and root under lead stress.  相似文献

The active, far-red light absorbing, form of phytochrome was found to inhibit growth and phytochrome levels in the mesocotyl and coleoptile of 4- to 5.5-day-old seedlings of Zea mays L. Short, low-irradiance red or far-red light treatments were used to produce different proportions of active phytochrome at the end of highdirradiance white-light periods, which left different levels of total phytochrome in the plants. After light treatments which left relatively high levels of spectrophotometrically assayable phytochrome in the seedlings, apparent phytochrome synthesis in the subsequent dark period was low regardless of the proportions of each form of the pigment present at the beginning of the dark period. In light treatments producing relatively low levels of assayable phytochrome, levels of apparent phytochrome synthesis in both red and far-red treatments and differences between apparent synthesis in red and far-red treatments were maximal. No simple correlation was found between growth and apparent phytochrome synthesis. However, growth and total phytochrome levels were positively correlated in both organs. Using a subtractive method of correlation, in which only phytochrome effects were plotted, strong linear relationships between phytochrome levels or longitudinal growth and Pfr levels were found in those light treatments leaving greater than 8% of dark control levels of phytochrome in the tissues. Using this technique non-linear, inverse relationships between Pfr and apparent phytochrome synthesis was found, indicating that modes of phytochrome control over phytochrome synthesis and growth differ. Our results are consistent with the view that in vivo assays of “bulk’ phytochrome reflect levels and states of the physiologically active phytochrome fraction under our experimental conditions in maize.  相似文献

Callus induction and subculture was successful with mature embryos and stem sections of seedlings of Zea mays L. on Linsmaier and Skoog's medium modified to contain 4 mg/I of 2,4-D and 1 g/I of casamino acids. — 2,4-D was superior to NAA and IAA for both callus induction and growth. Callus subcultured on NAA formed abundant roots on agar-solidified media and numerous root-like primordia in liquid cultures. — Kinetin had no effect on callus induction in the presence of 2,4-D and neither kinetin nor gibberellic acid stimulated callus growth during subculture. — Callus grew equally well on the medium of Linsmaier and Skoog, that of Schenk and Hildebrandt, and the B-5 medium of Gamborg and Eveleigh containing 2% sucrose, 4 mg/I of 2,4-D and 1 g/I of casamino acids. — The callus grew more rapidly at 25°C than at 30°C or 35°C. Little difference was noted at any temperature in callus growth in alternating light (16 h) and dark (8 h) or continuous dark. — Sucrose was superior to glucose and maltose in both liquid and agar-solidified cultures. Maize plants (Zea mays L.) were cultured with nutrient solutioncontaining 0.001 or 0.5 mM orthophosphate (Pi). Effects of lowphosphate (low-P) nutrition on growth, leaf phosphate status,photosynthesis, and carbon partitioning were investigated. Withlow-P treatment, the fresh weight of aerial parts decreasedby about 40% by 24 days after planting. Detailed studies ofthe effects of low-P treatment on the other characteristicsof maize leaves-were done using the middle part of the thirdleaf, counting from the base. Low-P treatment had almost noeffect on specific leaf weight or soluble protein content measured13 to 21 days after planting. Low-P treatment decreased Chicontent slightly (by 15% 19 days after planting). Twenty onedays after planting, low-P treatment had greatly decreased thelevels of leaf acid extractable Pi (by 77%) and photosynthesisrates (by 68%). The detrimental effects of low-P treatment onthe rates of photosynthesis and the amounts of acid extractablePi became progressively greater with time. There was a strongcorrelation between levels of leaf acid extractable Pi and ratesof photosynthesis. The minimum level of Pi necessary to sustainthe maximum photosynthesis rate was 0.6 mmol m–2. Belowthis minimum content of Pi, the rate of photosynthesis decreasedsharply with decreasing Pi. To investigate the direct effectof Pi depletion on photosynthate partitioning at equivalentrates of photosynthesis, the rates in controls were reducedto almost the same as those in 18 or 19 day old low-P plants(about 50% of those in controls) by lowering light intensityand/ or ambient CO2 concentration. The data clearly indicatesthat low-P treatment had a direct effect in lowering photosynthatepartitioning into starch. Starch mobilization during the nightwas also inhibited under low-P conditions. The effect of (+) (ABA) and (?)-abscisic acid and nine ABA metabolites, precursors or derivatives on radial water movement through maize roots, was investigated using a suction technique (Freundl and others 1998). (+)-ABA, (+)- and (?)-abscisyl aldehyde, (+)-8?-hydroxymethyl ABA, (+)-8?-methylene, and (+)-8?-acetylene ABA stimulated radial water transport. (?)-ABA, phaseic acid, and (+)-8?-acetylene methyl ABA were ineffective. ELISA analysis for ABA detected and apparent increase of free ABAxyl in xylem sap of excised root systems that were perfused with either (+)-abscisyl aldehyde, (+)-8?-methylene, (+)8?-acetylene-ABA, or ABA-glucose ester. The analogues (+)-8?-hydroxymethyl ABA and (?)-abscisyl aldehyde passed the cortex of maize roots without changing the ABAxyl. The data from this study permit conclusions about the structural requirements for hormonal regulation of hydraulic conductivity.  相似文献
