介绍一种马铃薯块茎蛾Phthorimaeaoperculella的室内人工饲养方法。在人工养虫室内用 1 0 %的蜂蜜水喂养成虫 ,用滤纸收集卵块 ,幼虫在薯块上饲养 ,化蛹于牛皮纸卷成的纸筒中 ,饲养结果发现马铃薯块茎蛾完成 1代大约需要 35~ 4 6d ,室内连续饲养块茎蛾 3代 ,卵的孵化率、幼虫化蛹率、成虫羽化率均在 90 %以上。  相似文献

在室内研究了分离自小猿叶甲的一株球孢白僵菌(SCAU-BB 01D)对小猿叶甲的致病力。结果表明,该菌株能感染小猿叶甲的成虫和各龄幼虫,但对不同虫期的致病力存在差异。在105~108孢子/m l的浓度范围内,随着处理浓度的升高,各虫期小猿叶甲的感病死亡率增加,在最高浓度1×108孢子/m l,处理后成虫第14天及1~3龄幼虫第10天的累计死亡率分别为84.7%、94.0%、96.0%和81.0%。用TDM模型对成虫和各龄幼虫的致病力数据进行模拟,所建模型均顺利通过Hosm er-L em eshow拟合异质性检验,表明模型拟合良好,并由模型估计出了该菌株对小猿叶甲各虫期的致死剂量与致死时间。在处理后第10天,成虫和1~3龄幼虫的致死中浓度(LC50)分别为2.68×107、1.07×106、1.63×105孢子/m l和8.31×106孢子/m l,而第14天成虫的LC50为2.38×106孢子/m l。随着浓度的增加,各虫期所需的感病死亡时间缩短,在最高浓度1×108孢子/m l,球孢白僵菌对小猿叶甲成虫及1~3龄幼虫的致死中时(LT50)分别为9.28、4.29、4.40d和5.06 d。综合分析白僵菌对各虫期的致死剂量及致死时间可以看出,不同虫期的小猿叶甲对球孢白僵菌敏感性不同。结果表明该菌株在小猿叶甲生物防治中具有一定的潜力。  相似文献

The tomato leafminer Tuta absoluta (Meyrick 1917) (Lepidoptera:Gelechiidae) is a devastating pest, causing losses of up to 100%. An interesting tool for its control is the use of the insect specific viruses of Baculoviridae family. Despite its high potential, its use on T. absoluta larvae has been poorly studied. In this work, Colombian granuloviruses VG013 and VG003 isolated from T. absoluta and Tecia solanivora (Lepidoptera:Gelechiidae) larvae sampled in tomato and potato crops, respectively, were morphologically, biologically and molecularly characterized. Occlusion bodies showed ovoid shape containing one nucleocapsid. Restriction endonuclease analysis revealed a pattern similar to Phthorimaea operculella granulovirus and bioinformatics studies showed that both isolates are variants of that baculovirus specie. Similar mean lethal concentrations (LC50) on T. absoluta larvae were estimated for both viruses, although VG013 exhibited shorter mean time to death than VG003. The latter reached a higher OBs yield in comparison with VG013. These results demonstrated an interesting potential of evaluated betabaculoviruses to control T. absoluta populations and pointed key features to its use under field conditions.  相似文献

Barley, Hordeum vulgare, one of the important crops in the word, is used in malting, feed and food industries. The red flour beetle, Tribolium castaneum, was found wherever grains or other dried foods are stored. Disinfestations of barley using chemical methods to kill insects, in this research, for the first time we isolated the pathogenic KB512 of entomopathogenic fungi Beauveria bassiana from soil and insects, which produced aerial and submerged conidia and blastospores in laboratory conditions. We investigated the best conditions for the production and utilisation of spore suspension to spray the larvae of T. castaneum, which is one of the important pests in Kerman province (Iran). One hundred and eighty isolates that naturally infected by T. castaneum were reared during spring and summer seasons 2010–2011. The pathogenicity test was carried out with direct spray. To bioassay the isolates, three concentrations of the spore suspension were prepared as follows: 1?×?106 and 1?×?108 conidia/ml. The pests were sprayed by aerial conidial suspension, which was prepared by 0.01% Tween 80 in distilled water, and the controls were sprayed by 0.01% Tween 80 in distilled water. After spraying the pests, the plates were incubated at 25?±?1?°C and 80% of relative humidity. Then, the treated pests were monitored every day for the fungal growth and mortality.  相似文献

Abstract:  Eggs of the potato tuber moth (PTM) Phthorimaea operculella (Zeller) obtained from 150 or 300 Gy-irradiated moths, either inbred or out-crossed to untreated counterparts, were offered as host material to three generalist Trichogramma species; Trichogramma cacoeciae , Trichogramma evanescens , and Trichogramma principium , to determine the ability of these egg parasitoids to develop in F1 sterile eggs. All eggs laid by the different parental crosses at 150 or 300 Gy, were acceptable and suitable for Trichogramma development. However, in no choice and paired comparison tests, eggs from 300 Gy-irradiated parents were significantly less preferred compared with eggs laid by non-irradiated moths. No intra-specific differences for PTM sterile eggs were recorded among the tested Trichogramma species. The mean number of wasps emerging from each egg types was not affected by an interaction between parental crosses and applied doses. Overall, this study demonstrates that release of irradiated and non-irradiated moths at a 5 : 1 over-flooding ratio and Trichogramma over potatoes in small plexiglass boxes, is complementary and more effective in reducing PTM F1-emerged progeny than either technique used alone. The results provide baseline data for developing an integrated control approach by using inherited sterility technique in conjunction with Trichogramma for PTM population suppression.  相似文献

Leptinotarsa decemlineata is a serious potato pest throughout the world that has developed resistance to many insecticides. This study investigated the virulence of two indigenous isolates of Beauveria bassiana, consisting of DEBI007 and IR1217C. Five different concentrations of each isolate, 1 × 107, 3.1 × 107, 1 × 108, 3.1 × 108 and 1 × 109 conidia ml?1, were applied in bioassay using dipping method on second instar larvae. Control insects were treated with distilled water containing 0.01% Tween-80 (Sigma, Germany) as surfactant. The results showed that mortality percentages due to DEBI007 and IR1217C isolates in the highest concentration were 60% and 57.77%, respectively. There was a significant difference between efficacies of different conidial concentrations; therefore, with rise in conidial concentration, the mortality percentage was increased. The DEBI007 isolate that was previously isolated from the soil was more effective than IR1217C isolate. Calculated LC50 for this effective isolate was 1.4 × 107 conidia ml?1, which, respectively, is the highest concentration.  相似文献

ABSTRACT

The potato tuber moth (PTM) Phthorimaea operculella is a critical potato pest. Larvae infest both foliage and tubers and mature larvae pupate in the soil or other safe places. Cordyceps tenuipes, an entomopathogenic fungus, infect lepidopteran pupae. To determine the effectiveness of this fungus as a biocontrol agent for PTM, we evaluated the time-concentration-mortality (TCM) response of PTM pupae to C. tenuipes using the following bioassays: (1) direct immersion in conidial suspensions, (2) incubation in sterilised or (3) unsterilised soilpremixed with conidia, and (4) incubation in unsterilised soil drenched with conidial suspensions to simulate field conditions. Fungal infection caused 100%, 83.3%, 73.3%, and 85.0% mortality of PTM pupae in assays 1–4, respectively. At 108 conidia/mL or conidia/g concentration, assays 1 and 4 had short lethal times (LT50) of 2.2 and 2.6 days compared with 3.7 and 4.8 days for assays 2 and 3, respectively. On day 7 after inoculation, assays 1 and 4 also had low lethal concentrations (LC50) of 1.69 × 103 conidia/mL and 1.10 × 105 conidia/g compared with those of assays 2 and 3, which showed low virulence, with LC50 of 3.50 × 105 and 3.60 × 106 conidia/mL, respectively. Our results demonstrate that C. tenuipes is a promising candidate for PTM biocontrol at the pupal stage. Drenching the soil surface with conidial suspensions may be the most effective method of field application.  相似文献

Aims: Larval stages of Frankliniella occidentalis are known to be refractory to fungal infection compared with the adult stage. The objective of this study was to identify promising fungal isolate(s) for the control of larval stages of F. occidentalis. Methods and Results: Ten isolates of Metarhizium anisopliae and eight of Beauveria bassiana were screened for virulence against second‐instar larvae of F. occidentalis. Conidial production and genetic polymorphism were also investigated. Metarhizium anisopliae isolates ICIPE 7, ICIPE 20, ICIPE 69 and ICIPE 665 had the shortest LT50 values of 8·0–8·9 days. ICIPE 69, ICIPE 7 and ICIPE 20 had the lowest LC50 values of 1·1 × 107, 2·0 × 107 and 3·0 × 107 conidia ml?1, respectively. Metarhizium anisopliae isolate ICIPE 69 produced significantly more conidia than M. anisopliae isolates ICIPE 7 and ICIPE 20. Internally transcribed spacers sequences alignment showed differences in nucleotides composition, which can partly explain differences in virulence. Conclusion: These results coupled with the previous ones on virulence and field efficacy against other species of thrips make M. anisopliae isolate ICIPE 69 a good candidate. Significance and Impact of the Study: Metarhizium anisopliae isolate ICIPE 69 can be suggested for development as fungus‐based biopesticide for thrips management.  相似文献

Abstract

The onion thrips Thrips tabaci is one of the most important pests of greenhouse and open-field broccoli, onion and other crops. However, the current strategy of using synthetic pesticides for its control is inadequate and unsustainable, leading to a growing interest in novel and effective biological control alternatives such as entomopathogenic fungi. Among 20 isolates of Beauveria bassiana tested for virulence against T. tabaci in laboratory bioassays, we found strain SZ-26 as the most potent, causing 83–100% mortality in adults at 1×107 mL?1conidia after 4–7 days. Further experiments in greenhouses showed the strain SZ-26 significantly lowered the numbers of adult and larval stages.  相似文献

House flies are pests of medical and veterinary importance. They are known to be susceptible to fungal conidia presented in sugar baits. The purpose of this study was to evaluate the potential for commercial formulations of entomopathogenic fungi to be used as bait for fly control. Differences in sporulation by product were found. Current formulations may influence fly feeding on baits, but because formulations may protect conidia. Additional research into more attractive formulations is warranted.  相似文献

Abstract The red turpentine beetle (RTB), Dendroctonus valens LeConte, as a destructive invasive pest, has become one of the most economically important forest pest in China. Effective control measures are desperately needed. Entomopathogenic fungi, such as Beauveria bassiana, have shown great potential for the management of some bark beetle species. In this study, 12 isolates of B. bassiana from bark beetle were examined for biological characteristics and virulence, to assess their potential as biocontrol agents for RTB. There were significant differences (at P= 0.05) in colony growth rate, conidial yield, conidial germination, tolerance to UV light and extracellular proteases activity among the tested B. bassiana isolates. Isolates, including Bb1801, Bb1906, Bb789 and Bb773, exhibited the best characteristics, because they have faster hyphal growth rate, higher spore production and faster spore germination, higher UV tolerance and protease (Pr1) production. The results of a pathogenicity test of B. bassiana on RTB larvae showed that most isolates of B. bassiana have demonstrated high efficacy and the highest virulent isolate was Bb1801, which killed 100% of the treated insects and had a median lethal time (LT50) of 4.60 days at a concentration of 1×107 conidia/mL. Therefore, isolate Bb1801 has a great potential for sustainable control of RTB in the forest. The correlation between biological characteristics and virulence of the fungal isolates is discussed and the possibility of combination of entomopathogenic fungi with semiochemicals, as one of the promising strategy for RTB control, is considered.  相似文献

Five Beauveria bassiana strains were evaluated for control of western flower thrips. Strain RSB was the most virulent, causing 69–96% mortality at concentrations of 1×104–1×107 conidia mL−1, 10 days after inoculation of first instars. A significant enhancement of spore production by solid-state culture of Beauveria bassiana was achieved via a statistical modeling and optimization approach. A two-level Plackett–Burman design was implemented to screen the significant medium components for spore production. Among the six variables tested, sucrose, NaNO3, and H3BO3 were identified as the most significant factors (confidence levels above 95%) for spore production. The steepest ascent experiments were adopted to determine the optimal region of the medium composition. The developed second-order polynomial model predicted that the maximal spore production was 6.24×109/g with the following optimal culture conditions: sucrose 34.7 g/L, NaNO3 52.8 g/L, and H3BO3 79.6 µg/L. Finally the verification experiments were carried out and the maximal spore production reached 6.221×109/g at the above optimal conditions. The spore production in the verification experiments was very close to that predicted by the model, indicating that the constructed second-order model had superior modeling and optimization ability and can be applied in the optimal spore production of Beauveria bassiana YP9 as a biological control agent.  相似文献

Orius sauteri (Poppius; Hemiptera: Anthocoridae) is an important predator of western flower thrips, Frankliniella occidentalis (Pergande; Thysanoptera: Thripidae). O. sauteri would be directly exposed to the entomopathogenic fungus Beauveria bassiana (Bals.) Vuillemin in the field should the fungus be used as a biopesticide. If the fungus were to negatively affect O. sauteri in agro-ecosystems, predation of F. occidentalis by O. sauteri may be limited. The present study was undertaken to evaluate the insecticidal activity of strain B.bassiana-RSB of B. bassiana, which is highly virulent to F. occidentalis, on the predator under laboratory conditions. Results showed that, regardless of the concentration applied to first instars, Bb-RSB was not insecticidal against O. sauteri, nor did direct applications affect the developmental rate of the predator. Significant differences in developmental rates and adult longevity were observed between O. sauteri that fed on Bb-RSB-infected F. occidentalis cadavers and those that fed on untreated thrips. Developmental time (from first instar to adult) increased from 0.3 to 0.7 days and adult longevity decreased by 0.8 to 1.2 days for predators fed thrips treated with low and high concentrations of strain Bb-RSB, respectively, compared with predators fed on untreated thrips. However, these differences were only 3–13% of mean values for the controls, suggesting that the effects of Bb-RSB on O. sauteri are relatively minor. These findings highlight the potential use of O. sauteri in combination with B. bassiana for the biocontrol of F. occidentalis, but field tests must be performed to confirm their compatible use.  相似文献

Abstract:  In laboratory bioassays, the efficacy of the entomopathogenic fungus Beauveria bassiana against the spruce bark beetle, Ips typographus , was tested under various conditions. Four of the tested isolates and the commercial product Boverol® caused 99–100% mortality when tested at a concentration of 1.0 × 107 conidia/ml at 25°C. Using B. bassiana isolate 138 at a concentration of 1.0 × 106, the median survival time (MST) was 6.1 d and significantly longer compared with the MST of 4.2 and 4.0 d at 1.0 × 107 and 1.0 × 108 conidia/ml, respectively. In the next experiment, the beetles were maintained on spruce bark, filter paper or artificial diet during the bioassay with Boverol®, and significant differences in the MST of 3.6, 2.5 and 5.3 d, respectively, were noticed. The experiment with Boverol® at different temperatures showed that the beetles lived significantly longer at 15°C (MST 8.7 d) than at 20, 25, 30 and 35°C. At 25°C, the beetles died most rapidly (MST 3.5 d). At different relative humidities (RH) of 40, 70 and 100%, nearly all beetles were dead after treatment with a suspension of Boverol® at 1.0 × 107 conidia/ml. At 40% RH, 49% of the untreated beetles died after 7 d. The best effects were achieved with the following bioassay: beetles were fed for three days on artificial diet, then dipped into a solution of 1.0 × 107 conidia/ml and transferred on a piece of spruce bark in Petri dishes at 25°C and 70% RH.  相似文献

A standard bioassay method for assessing the pathogenicity of Beauveria bassiana (Balsamo) Vuillemin (GHA strain) against second instar tarnished plant bug, Lygus lineolaris (Palisot de Beauvois) (Hem., Miridae) was developed. Several types of inoculation methods, assay containers and incubation times were tested. Our goal was to minimize control mortality and maximize treatment mortality. Five inoculation methods (immersing broccoli florets or bean pods, spraying broccoli florets or bean pods, and immersing insects) and four types of plastic containers (114‐, 171‐, 228‐ and 455‐ml) were tested. Immersing insects directly in a fungal suspension was the most effective inoculation method, which resulted in a treatment mortality of 70–81.3% at a concentration of 1 × 107 conidia/ml. The 114‐ml plastic container was the most suitable assay container when 10 tarnished plant bug nymphs were treated together, resulting in a control mortality of only 6% 12 days after treatment. Within the first 6 days after treatment, 71.1% of the insects were killed, compared with a total mortality of 81.3% after 12 days. Nymphs infected with the test fungus changed colour from green to black. Mycelial outgrowth and sporulation on the cadavers demonstrated that most nymphs died of fungal infection. A total of 61.1 and 80.5% of the cadavers showed signs of mycelial outgrowth 9 days after death among those that were surface sterilized and those that were not, respectively.  相似文献

本文就烟草潜叶蛾幼虫空间分布型及其垂直分布规律进行了探讨,结果表明:烟草潜叶蛾幼虫在田间呈聚集分布,聚集强度不因种群密度的改变而改变,幼虫主要聚集分布在烟草下部第一段(4片叶)上;降集或随机分布在第二段上;随机分布在第三段上,此外,应用虫株率进行田间种群密度的估计,其中Wilson模型和Gerrard模型所配理论曲线的预测值与实测值显著适合,但Gerrard模型的抽样估计误差较Wilson模型的小,最后本文用Taylor式中的参数a,b确定理论抽样数及序贯抽样,其模型分别为。  相似文献

对来源于吉林省不同地区的15株球孢白僵菌Beauveria bassiana(Bals.)Vuill.菌种对大豆蚜Aphis glycines Matsumura毒力进行了室内生物学测定。所有供试菌株对大豆蚜虫均有一定的致病性,杀虫效率差异明显,杀虫效率与孢子浓度和作用时间均呈正相关。通过对大豆蚜毒力室内生物学测定结果的综合分析,菌株S14-X-1为毒力较高菌株,具有进一步研究应用价值。  相似文献

AIMS: To monitor the fate of inundatively applied strains of Beauveria bassiana against Masson's pine caterpillar, Dendrolimus punctatus, in Magushan Forest, southeast China, and evaluate the potential environmental risks of biocontrol introduction. METHODS AND RESULTS: Filed samplings of mycosed cadavers were carried out continuously for more than one year after the inundative applications of two exotic B. bassiana strains. By employing four complementary molecular markers, the purified cultures from collections were characterized and the two released strains Bb17 and Bb13 were recovered with respective frequencies of 9.1 and 5.2%. The released strains were isolated from nontarget hosts. The genetic diversity of the population of B. bassiana in the trial plot changed dynamically over time and no single genotype strain dominated throughout the sampling period or at different seasons. The indigenous strains were observed to be predominant in the local environment. Co-infection and/or genetic recombination might occur between strains under natural conditions. CONCLUSION: Field applications of the biocontrol agent B. bassiana appear to pose no major risk to the environment. There was no evidence of displacement of indigenous strains of B. bassiana. SIGNIFICANCE AND IMPACT OF THE STUDY: The results indicate that the introduction of exotic B. bassiana strains can persist in local environment and infect nontarget insect hosts, highlighting the importance of safety assessment of the large-scale application of fungal biocontrol agents.  相似文献
