The economy of carbon in nodulated white lupin (Lupinus albusL.) was studied in terms of consumption of net photosynthatein nitrogen fixation, in maintenance of respiration, and inthe production of dry matter and protein. Net photosynthesisrose to a maximum in early fruiting and then fell abruptly dueto shedding of leaves. Nodulated roots acquired translocateequivalent to 51% of the plant's net photosynthate, 78% of thecarbon of this translocate being respired, 10% entering drymatter, and 12% returning to the shoot attached to productsof nitrogen fixation. Nodules utilized 4?0–6?5 g C infixing 1 g nitrogen. Photosynthate was utilized most effectivelyfor nitrogen fixation in late vegetative growth. Fruits sequestered16% of the plant's net photosynthate, shoot night respiration17%, and dry matter formation in shoot vegetative parts 22%.Averaged over growth, 9?9 g net photosynthate was required toproduce 1 g seed dry matter and 31 g net photosynthate to produce1 g seed protein. Budgets for utilization of the carbon of netphotosynthate were constructed for 10 d intervals of the plant'sgrowth cycle. Feeding of shoots with 14CO2 resulted in radiocarbonbecoming partitioned approximately as predicted by these budgets.The dependence of root respiration on recent photosynthate wasassessed by following the time course of release of 14CO2 tothe rooting medium of the 14CO-labelled plants.  相似文献

An improved method for the measurement of respiratory utilizationof current photosynthate in the nodulated root system of water-culturedsoybean (Glycine max L.) plants was developed using a steady-state13CO2 labelling technique. Well-nodulated plants at the latevegetative stage were allowed to assimilate 13CO2 for 10 h incontinuous light at a constant CO2 concentration with a constant13C abundance. The respiratory evolution of 13CO2 from rootsand nodules was measured continuously throughout the periodof 13CO2 assimilation and during a subsequent 36 h chase periodby using a differential infrared 13CO2 analyser. The plantswere grown with nitrogen-free or (15 mmol dm–3)-containing culture solution for 3 d before13CO2 assimilation. In plants grown without , nodule respiration averaged 69% of the total respiration of the undergroundparts over the full experimental period and the CO2 respiredreached an apparent isotopic equilibrium at 80–85% labellingafter initiating 13CO2 assimilation. By contrast, the CO2 respiredfrom the roots did not reach an isotopic equilibrium and labellingwas only 56% at the end of exposure to 13CO2 These findingsdemonstrated that nodule respiration is strongly dependent onrecently assimilated carbon compared with root respiration. Plants supplied with in the culture solution showed a decreased rate of nodule respirationand a slightly increased rate of root respiration. The extentsand time courses of labelling of respired CO2 from both theroots and nodules were similar in the presence and absence of except that the maximum level of labelling of CO2 derived from nodule respiration in plantswith was significantly higher (about 91%) than for plants growing without . Carbon dioxide output and oxygen uptake by intact nodulatedroots of soyabean and white clover were determined over a rangeof external oxygen concentrations (10 to 60%) in the presenceand absence of 10% acetylene. The responses of both sets ofroot systems were in agreement with the known characteristicsof the variable oxygen diffusion resistance of the nodules.These measurements were used to calculate the respiratory quotient(RQ, CO2/O2) for both total root respiration and nitrogenase-linkedrespiration values for each external oxygen concentration. Inthe absence of acetylene, the RQ values of soyabean total rootrespiration showed no significant response to external oxygen,with a mean of 1.06, but differences in values for white cloverroots were significant at P=0.05, with a mean of 1.09. The sametrend was shown for RQ values of nitrogenase-linked respiration,with mean values of 1.10 and 1.12 for soyabean and clover, respectively.The addition of acetylene caused a small but significant initialreduction in RQ of total root respiration with both soyabeanand white clover, as did the subsequent reduction of externaloxygen to 10%. For white clover, increases in external oxygenconcentration to 50% and 60% also produced significant increasesin the RQ of total root respiration compared to that in air.These variations were more pronounced for RQ values of nitrogenaselinkedrespiration. Results are discussed in relation to the use ofRQ values of unity in calculations of nodular oxygen diffusionresistance and the involvement of physical and biochemical processesin the regulation of oxygen supply to bacteroids. Measurements of rates of net photosynthesis were made on singleBlanca white clover leaves on plants taken from a field-grown,mixed clover/perennial ryegrass sward during two regrowth periods. Net photosynthesis fell by 20 per cent in the first measurementperiod as leaf area index increased and the grass componentof the crop flowered, but did not change significantly in thesecond measurement period during which the grass remained vegetative. Leaves which had been artificially protected from shading inthe sward did not have significantly different photosyntheticcapacities from leaves in the undisturbed sward, even in thefirst measurement period. As leaf area index and sward height increased, successive cloverpetioles were longer, keeping the newly expanded leaves nearthe top of the sward where they received full light. It is suggestedthat it is this which allows successive clover leaves, unlikethose of vegetative grasses, to attain a high photosyntheticcapacity throughout a growth period. Root-infecting nematodes are a major cause of white clover, Trifolium repens, not reaching its potential in New Zealand pastures. Resistance and/or tolerance are the preferred control options. Greenhouse-based, recurrent selection programs have developed resistance to Meloidogyne trifoliophila and Heterodera trifolii, and a field-based program has developed tolerance. Lines from these programs were compared with commercial cultivars as controls in a series of field trials at four sites over 4 years. Resistant lines from the CCN program performed better than susceptible lines and as well as most cultivars, reflecting the high level of resistance developed in this greenhouse-based program. In stained root from Cambridge, numbers of CCN were lower in resistant lines than in cultivars; numbers in susceptible lines were intermediate. CCN resistance was also reflected to a lesser extent in the number of cysts counted in soil under resistant lines in Palmerston North. The root-knot nematode-resistant material performed better than the susceptible and as well as most cultivars. In one trial of CRKN-resistant lines, resistant and susceptible lines had similar numbers of CRKN which were both lower than the numbers in the cultivars; in the second trial, there were fewer CRKN in resistant than in susceptible lines or cultivars. The tolerant selections, developed under field conditions, performed as well as or better than the cultivars. The selections from the breeding programmes have exhibited strong agronomic potential across locations and years, and the best material has been crossed; progeny are being assessed in current field trials.  相似文献

Single, clonal plants of white clover were grown without inorganicnitrogen in four contrasting day/night temperature regimes,with a 12 h photoperiod, in controlled environments. Root andnodule respiration and acetylene reduction activity were measuredin a flow-through system during both day and night for plantsacclimated to day/night regimes of 23/18, 15/10 and 10/5 ?C.Similar measurements were made on plants acclimated to 20/15?C and stepwise at temperatures from 4 to 33 ?C. Peak rate of ethylene production, nitrogenase-linked respirationand basal root + nodule respiration increased approximatelylinearly from 5 to 23 ?C both in temperature-acclimated plantsand in plants exposed to varying measurement temperatures. Themeasured attributes did not vary significantly between day andnight. Temperatures above 23–25 ?C did not further enhancethe rate of ethylene production, which remained essentiallythe same up to the maximum measured temperature of 33 ?C. The measurements of nitrogenase-linked respiration between 5and 23 ?C, during both day and night, demonstrated a constant‘energetic cost’ of acetylene reduction of 2.9 µmolCO2 µmol C2H4–1,. Over the same temperature range,the approximate activation energy of acetylene reduction was60 kJ mol–1. The integrated day plus night nitrogenase-linkedrespiration accounted for 13.4–16% of the plant‘snet shoot photosynthesis in a single diurnal period: there wasno significant effect of temperature between 5 and 23 ?C. Successive sets of cuttings of three white clover genotypeswere raised in a 15 °C growth room and transferred to thefield at 14 d intervals over the course of a year. Rates ofleaf appearance (leaves per stolon growing point per unit time)were found to be closely correlated with 10 cm soil temperatures.Petiole lengths, and weights of the lamina+petiole increasedin May and decreased towards the end of August, but also exhibiteda marked response to a mid-season water deficit. In the conditionsof the experiment (i.e. in the absence of competition from neighbouringplants) the vast majority of axillary buds developed into visiblebranches at all times of the year. There was, however, an increasein the nodal age at which bud development was first observedin winter. Deferred bud development was also observed, particularlyin one genotype, during periods characterized by dry soil surfaceconditions. The results are discussed in relation to observedpatterns of stolon branching in sward conditions. Growth habits and cyanogenesis were studied in a field experimentwith white clover (Trifolium repens L.) cv. Huia. Eighty controlplants propagated by seeds, and 80 clones of an in vivo selectedvariant were examined in mid-late August and late September.The temperature effect on cyanogenic glucoside levels was alsoexamined on in vitro grown plantlets of the variant in growthchambers. Results showed that the upright growth habit dominated in controlplants. The population of the variant plants was mainly composedof prostrate clones and a transition from prostrate to uprightgrowth habit occurred. Considerable variation was observed withregard to all measured morphological characters in both controland variant plants. Great variation was also noted in cyanogenicglucoside content in the leaf laminae of the control plants.Low cyanogenic plants, on the other hand, dominated in the variant.The number of low and high cyanogenic plants increased in thecontrol by the end of the growth season, after the first frosts.A one-way shift of cyanogenic (slight increase) was recordedin the variant. Similar levels of cyanogenic glucoside and linamarasewere determined in the in vitro and field-grown plants. Cyanogenicglucoside content slightly decreased with the age of the invitro plantlets, but variation in temperature did not causeany changes in their level. Single plants of white clover, grown in a controlled environmentand dependent for nitrogen on fixation in their root nodules,were defoliated once by removing approximately half their shoottissue. Their regrowth was compared with the growth of comparableundefoliated plants. Two similar experiments were carried out:in the first, plants were defoliated at 2.5 g, and in the secondat 1.2 g total plant d. wt. Defoliation reduced rate of N2 fixation by > 70 per cent,rate of photosynthesis by 83–96 per cent, and rate ofplant respiration by 30–40 per cent. Nodule weights initiallydeclined following defoliation as a result of loss of carbohydratesand other unidentified components. No immediate shedding ofnodules was observed but nodules on the most severely defoliatedplants exhibited accelerated senescence. The original rates of N2 fixation were re-attained after 5–6or 9 d regrowth, with increase in plant size at defoliation.In general, the rate of recovery of N2 fixation was relatedto the re-establishment and increase of the plant's photosyntheticcapacity. Throughout the growth of both defoliated and undefoliatedplants nodule respiration (metabolism) accounted for at least23 ± 2 per cent of gross photosynthesis. The unit ‘cost’of fixing N2 in root nodules, in terms of photosynthate, appearedto be unaffected by defoliation, except perhaps for plants veryrecently defoliated. Similarly, the percentage nitrogen contentsof shoot, root and nodules of defoliated plants became adaptedwithin a few days to those characteristic of undefoliated plants. Introduction of the pea (Pisum sativum L.) lectin (PSL) gene into white clover (Trifolium repens L.) hairy roots facilitates nodulation by the nitrogen-fixing bacterium Rhizobium leguminosarum biovar viciae, which normally nodulates pea and not white clover (C.L. Diaz, L.S. Melchers, P.J.J. Hooykaas, B.J.J. Lugtenberg, and J.W. Kijne [1989] Nature 338: 579-581). Here, we show that PSL is functionally expressed in transgenic white clover hairy roots transformed with the PSL gene. PSL could be isolated from these roots by affinity chromatography. Immunoanalysis of PSL showed the presence of polypeptides corresponding to the PSL precursor and its [beta] subunits. In addition, we developed a highly sensitive localization technique based on specific binding of a glycan moiety of rat IgE to PSL. Similar to the situation in pea roots, PSL appeared to be localized on the external cell surface of elongated epidermal cells and on the tips of emerging and growing root hairs of transgenic white clover hairy roots. PSL was not observed on normal white clover roots and on hairy roots without the PSL gene. These results show that (a) in transgenic white clover hairy roots, PSL is correctly processed and targeted to root cells susceptible to rhizobial infection, and (b) like in pea roots, PSL is surface bound with at least one of its two sugar-binding sites available for (rhizobial) ligands.  相似文献

A period of growth under shade netting in the glasshouse allowedthe cultivation of white clover stolons with an accumulationof undeveloped axillary buds similar to that often found onstolons from grass/clover swards. The subsequent capacity ofthese nodes to develop branches under different circumstanceswas investigated in three experiments. Removal of the laminaeand petioles subtending sets of four buds along a stolon reducedthe rate at which branches were initiated from the buds. Treatmentsin which petioles, or petioles plus laminae, were retained initiatedbranches more quickly. Shading the stolons reduced both therate of initiation and the percentage of buds which developed,unless both petioles and laminae were retained. There was someevidence that conditions applied to individual buds may actin the same way as the same conditions applied to sets of fourbuds and that illuminated nodes may depress the performanceof neighbouring shaded notes. Fewer buds developed at older nodes than at younger nodes duringthe summer, but during the autumn younger buds initially developedmore slowly than older buds. This suggests that buds can developat a younger nodal age in summer than in winter. When leafless stolons were cut up into component internodesbuds developed faster than on intact stolons, provided the budwas located at the end of the internode nearest the main stolongrowing point. If the bud was at the other end, branch developmentwas slower than on intact stolons. The results are discussedin relation to clover growth in sward conditions. The rate of photosynthesis of leaves of perennial ryegrass (Loliumperenne L.) and white clover (Trifollum pratense L.) grown atdifferent temperatures was measured at a range of temperatures.There was a small effect of the temperature at which a leafhad grown on its photosynthetic rate, but a large effect ofmeasurement temperature, especially in bright light, where photosyntheticrates at 15°C were about twice those at 5°C. Exposure of mature, nodulated plants of white clover (Trifoliumrepens) cv. Blanca to 330 mg dm–3 NO3-N for 8 d causednitrogenase activity per plant to decrease by 80%. Total nodulatedroot respiration was not significantly affected but analysisof its components showed an 81% decrease in nitrogenase-linkedrespiration and a 340% increase in growth and maintenance respiration.Carbon costs of nitrogenase activity (mol CO2 respired per molC2H4 produced) increased by 45% over the exposure period. Sucrosecontent of the nodules decreased, but the pattern of decreasedid not correlate with that of nitrogenase activity. The oxygendiffusion resistance of the nodules was increased by a factorof five. Characterization of this resistance increase suggestsan abnormal modification of the diffusion barrier and it isconcluded that alteration in the oxygen supply to the bacteroidsis involved in the effect of nitrate on nitrogenase activity. Effects of temperature on floral initiation of ten white clovervarieties growing in controlled environments are described.Plants grown under long days (16 h) were subjected to constanttemperatures of 26, 18 and 10 °C. Relationships betweenmorphological and physiological traits and flowering were examined. Most plants flowered at the two higher temperatures but only10 per cent of plants flowered at 10 °C. Larger leaved typestended to produce more reproductive buds per stolon at the highertemperatures than did smaller leaved varieties. Of the floral characters studied, floret number was least affectedby temperature. Ovule number and peduncle length were greatestat 18 °C. A quantitative analysis of the 14C-labelled assimilate suppliedby the expanded leaves on the primary shoot to growing leaves,stem, lateral shoots (branches or stolons) and roots in redand white clover was conducted during vegetative growth. Stem growth of the primary shoot was inhibited in both cloversand utilized no energy resources. The growing leaves at theprimary shoot apex of white clover imported 4 per cent of theshoot's assimilate compared with 10 per cent in red clover.At the basal end of the primary shoot, the tap root of whiteclover imported 16 per cent of the shoot's assimilate comparedwith 22 per cent in red clover. Branches in red clover and stolonsin white clover were by far the largest sinks for primary shootassimilate, importing 39 per cent and 63 per cent of the labelledassimilate, respectively. Analyses of the translocation of assimilate from individualprimary shoot leaves demonstrated that in both clovers olderleaves exported more of their assimilate to branches or stolons,whereas younger leaves exported more of their assimilate toroots, and possibly in white clover, to growing leaves at thetip of the shoot. Of the labelled assimilate exported to branchesor stolons, each primary shoot leaf exported preferentiallyto the branch or stolon in its own axil, but in addition exportedsubstantial quantities of assimilate to all other axillary shoots,particularly those arising from basal axils where the subtendingleaf had died. The growth of white clover (Trifolium repens L.) in conditionstypical of April in Southern England (8 °C day/4 °Cnight, 12 h photoperiod of 90 J m–2 s–1 visibleradiation) was extremely slow, whether the plants were dependentfor nitrogen on fixation by their root nodules or were suppliedwith abundant nitrate; although growth was slower in the nodulatedplants. The reasons for slow growth were a large root: shootratio and a small leaf area, particularly in the nodulated plants,and a low photosynthetic rate in all plants. The probable effectsof these characteristics on the growth of white clover withgrasses in mixed pastures are discussed.
