以杉木茎尖诱导形成的愈伤组织为试验材料,研究维生素C、柠檬酸、半胱氨酸、硫代硫酸钠4种抗氧化剂和活性炭对杉木愈伤组织培养中褐化的影响。结果表明:维生素C和半胱氨酸能有效地控制褐化的发生,且对愈伤组织生长没有明显的抑制作用,是杉木愈伤组织培养较为适宜的抗褐变剂。  相似文献

The respiratory rate increased in two phases during early stages of callus formation in carrot (Daucus carota)-root phloem slices cultured in vitro, showing separate peaks at about 24 and 96 h of culture. In the first phase (within 24 h of culture), the activities of phosphoglucose isomerase, phosphofructokinase and succinate dehydrogenase did not increase, but active syntheses of RNA and protein, indicated by experiments with incorporation of [14C]-uracil and -leucine, resulted in an active turnover of ATP, to which the first increase in the respiratory rate may be attributable. On the other hand, the activity of phosphoribosylpyrophosphate synthetase, an enzyme related to nucleic acid synthesis, increased in the first 24 h of culture. In the second phase (24–96 h of culture), the activities of the respiratory enzymes investigated increased. This increase was repressed by cycloheximide, indicating de novo syntheses of the respiratory enzymes during this time, which may result in an enlargement of the respiratory capacity, to which the second increase in respiration may be mainly attributable. In the first phase, exogenous supplied 2,4-dichlorophenoxyacetic acid had little or no effect on the respiratory rate and the activities of the respiratory enzymes, but it enhanced synthesis of RNA and the activity of phosphoribosylpyrophosphate synthetase. In the second phase, it increased all the activities of enzymes investigated as well as the respiratory rate and RNA synthesis.  相似文献

以大蒜品种‘三月黄’(Allium sativum L.cv. Sanyuehuang)花序轴为外植体进行离体培养,对其器官发生过程进行了形态学和解剖学观察。结果显示:大蒜花序轴离体培养不经过愈伤组织,通过器官直接发生途径形成不定芽,其不定芽起源于大蒜花序轴维管组织韧皮部一侧周围的皮层薄壁细胞,属于外起源;皮层薄壁细胞经脱分化后,由最先形成的拟分生组织发育为茎尖分生组织,然后环绕其形成叶原基,茎尖和叶共同构成一个完整的不定芽;大蒜花序轴离体培养发生的不定芽与花苞中自然形成的营养芽发生部位一致。不定芽通过壮苗、生根培养可正常生根形成植株,如果继代培养周期超过21 d,鳞茎形成率可达90.56%。  相似文献

The development of callus in vitro was investigated using the isolated explants from the autumn twig of Olea europaea. The callus developed on the cut surface of the stem-segment originate, d from the living cells of different tissues. Two distinct morphological types of the calli were observed. The highly friable callus was generally obtained while a non-friable form might develop occasionally. The change from one form to another was achieved by changing the level of NAA in the culture medium. These results obtainecl from above may be used to explain the possible role of the auxin playing a part in the form of callus. However, another distinct type of the callus in addition was also obtained in the same culture medium. This type of the callus developed in the form of a knob. It originated from cortical parenchyma cells distal from the cut surface of the stem The divisions of the cortex cells induced only were restricted to the inner layers near by the pericyele. The present observation suggests that the interpretation for the variations of the callus forms may be elaborated further when the roles of callus of the different sources will have been explored. There were three stages in the course of callus development from the cut sur- face of stem-segments, namely, activation, division and formation. These three dovelopmental stages were characterized by changes in cell morphology of the population as well as in the cell division, grawth and the relative RNA accomulation in the callus tissue. The results are shown in table 1. The formation stage proposed in this paper differs with the differentiation phase (Yeoman, 1970, 1973) on the differen- tiation of the tissues and it is suggested that appearance of wound-cambinm and differentiation of the parenchyma in this stage may be considered as the characteristics of callus development. The general conceptions of activation, dedifferentiation and differentiation are also briefly discussed.  相似文献

研究了纯氧对荔枝果皮褐变和细胞超微结构的影响。结果表明,100%O2显著抑制了荔枝果皮褐变,在100%O2下贮藏6d的荔枝果实,中果皮细胞的细胞器和膜系统的完整性保持较好,而细胞膜透性较低,因此,认为高氧抑制荔枝果皮褐变与其维持细胞膜的完整性相联系。  相似文献

通过在培养基中逐代增加氯化钠浓度方法获得的适应0.5%到1.0%NaCl的盐适应细胞系,与非盐适应细胞系的蛋白质电泳图谱相比,其蛋白质SDS-PAGE图谱中出现了26kD蛋白质,但量不多;此外,40.3和16.5kD蛋白质量增加,其中16.5kD蛋白质增加显著;而33.8和18kD蛋白质则减少。上述16.5,26和40.3kD蛋白质的诱导产生或含量的增加和细胞在盐环境中表现出的适应性相伴随发生,表明这3种蛋白质可能与抗盐性有关。  相似文献

离体蒜苔构成一个完整的细胞内含物再分配系统。 25 ℃条件下,于黑暗中贮存时,苔茎基部细胞内含物转移到顶端珠蒜中,最后苔茎下部枯萎,顶端形成鲜嫩多汁的珠蒜。适当浓度 GA3处理苔茎基部可以有效抑制上述细胞内含物再分配过程。已有研究表明, H2O2由超氧化物歧化酶(SOD)催化产生,被过氧化物酶(POD)和过氧化氢酶(CAT)催化降解; H2O2对生物个体发育具有重要调节作用。本文主要测定GA3对离体蒜苔H2O2代谢的影响;为进一步探讨H2O2在细胞内含物再分配中的作用提供参考。 取珠蒜未明显膨大的离体蒜苔为供试材料,采用 50μg/mL GA3溶液处理蒜苔基部,用比色法和氧电极法测定珠蒜和苔茎下部H2O2水平和SOD、POD、CAT活性。结果表明:(1)在处理后48h内,珠蒜和苔茎下部H2O2代谢即产生明显差异(Fig.1-4);(2)贮存20d后对照珠蒜明显膨大,而GA3处理珠蒜光显著变化(Table1);(3)GA3处理显著提高了珠蒜H2O2水平和SOD、POD、CAT活性,相反苔茎下部H2O2水平和POD、CAT活性受到显著抑制,而SOD活性提高(Fig.5-8)。GA3处理对珠蒜和苔茎下部H2O2代谢的相反  相似文献

The role of cholinergic neurotransmitters in the changes in ultrastructure of adrenocortical cells remains to a great extent unexplored. We studied the influence of acetylcholine (ACh) and pilocarpine, agonists of muscarinic ACh (mACh) receptors, on the ultrastructure of adrenocortical secretory cells derived from the rat adrenal cortex. Both agonists were found to stimulate fragmentation of the mitochondria and lipid droplets and reconstruction of the peroxisomal compartment. At the same time, assembling of the smooth endoplasmic reticulum (sER) membranes was observed after ACh application, while pilocarpine action was not always prominent. Thus, we conclude that activation of mACh receptors by its agonists influence the ultrastructure of the adrenocortical cells, and the respective ACh-induced effects are more intensive.  相似文献

Understanding neural injury in hydrocephalus and how the brain changes during the course of the disease in-vivo remain unclear. This study describes brain deformation, microstructural and mechanical properties changes during obstructive hydrocephalus development in a rat model using multimodal magnetic resonance (MR) imaging. Hydrocephalus was induced in eight Sprague-Dawley rats (4 weeks old) by injecting a kaolin suspension into the cisterna magna. Six sham-injected rats were used as controls. MR imaging (9.4T, Bruker) was performed 1 day before, and at 3, 7 and 16 days post injection. T2-weighted MR images were collected to quantify brain deformation. MR elastography was used to measure brain stiffness, and diffusion tensor imaging (DTI) was conducted to observe brain tissue microstructure. Results showed that the enlargement of the ventricular system was associated with a decrease in the cortical gray matter thickness and caudate-putamen cross-sectional area (P < 0.001, for both), an alteration of the corpus callosum and periventricular white matter microstructure (CC+PVWM) and rearrangement of the cortical gray matter microstructure (P < 0.001, for both), while compression without gross microstructural alteration was evident in the caudate-putamen and ventral internal capsule (P < 0.001, for both). During hydrocephalus development, increased space between the white matter tracts was observed in the CC+PVWM (P < 0.001), while a decrease in space was observed for the ventral internal capsule (P < 0.001). For the cortical gray matter, an increase in extracellular tissue water was significantly associated with a decrease in tissue stiffness (P = 0.001). To conclude, this study characterizes the temporal changes in tissue microstructure, water content and stiffness in different brain regions and their association with ventricular enlargement. In summary, whilst diffusion changes were larger and statistically significant for majority of the brain regions studied, the changes in mechanical properties were modest. Moreover, the effect of ventricular enlargement is not limited to the CC+PVWM and ventral internal capsule, the extent of microstructural changes vary between brain regions, and there is regional and temporal variation in brain tissue stiffness during hydrocephalus development.  相似文献

测定碳酸钙(CaCO3)胁迫下枳壳愈伤组织的某些生理生化指标的结果表明:除全Fe含量外,CaCO3浓度与过氧化物酶(POD)活性、超氧化物歧化酶(SOD)活性、过氧化氢酶(CAT)活性、活性Fe含量、可溶性蛋白质含量、愈伤组织增重均呈显著或极显著负相关,与培养液pH值呈显著正相关;随着培养的进程,POD活性、活性Fe和可溶性蛋白质含量、愈伤组织增重呈降低趋势,SOD和CAT活性先升后降,含高浓度CaCO3的培养液pH值仅在20日左右略有降低.  相似文献

玉米种子脱水试验表明,25℃下萌发24h种子脱水耐性开始丧失,丧失50%和100%的时间分别为33h、58h。萌发过程中随着吸胀时间增加,玉米种子脱水耐性逐步丧失。显微观察显示,种子吸胀过程中,胚根细胞的贮藏物质逐步减少,线粒体等细胞器的分化程度则不断提高,尤其是脂类物质的分解程度与脱水耐性变化的关系似乎更明显。  相似文献

Gas chromatography (GC) is a highly sensitive method used to identify and quantify the fatty acid content of lipids from tissues, cells, and plasma/serum, yielding results with high accuracy and high reproducibility. In metabolic and nutrition studies GC allows assessment of changes in fatty acid concentrations following interventions or during changes in physiological state such as pregnancy. Solid phase extraction (SPE) using aminopropyl silica cartridges allows separation of the major lipid classes including triacylglycerols, different phospholipids, and cholesteryl esters (CE). GC combined with SPE was used to analyze the changes in fatty acid composition of the CE fraction in the livers of virgin and pregnant rats that had been fed various high and low fat diets. There are significant diet/pregnancy interaction effects upon the omega-3 and omega-6 fatty acid content of liver CE, indicating that pregnant females have a different response to dietary manipulation than is seen among virgin females.  相似文献

The genetic improvement of garlic plants (Allium sativum L.) with agronomical beneficial traits is rarely achieved due to the lack of an applicable transformation system. Here, we developed an efficient Agrobacterium-mediated transformation procedure with Danyang, an elite Korean garlic cultivar. Examination of sGFP (synthetic green fluorescence protein) expression revealed that treatment with 2-(N-morpholino) ethanesulfonic acid (MES), L-cysteine and/or dithiothreitol (DTT) gives the highest efficiency in transient gene transfer during Agrobacterium co-cultivation with calli derived from the roots of in vitro plantlets. To increase stable transformation efficiency, a two-step selection was employed on the basis of hygromycin resistance and sGFP expression. Of the hygromycin-resistant calli initially produced, only sGFP-expressing calli were subcultured for selection of transgenic calli. Transgenic plantlets produced from these calli were grown to maturity. The transformation efficiency increased up to 10.6% via our optimized procedure. DNA and RNA gel-blot analysis indicated that transgenic garlic plants stably integrated and expressed the phosphinothricin acetyltransferase (PAT) gene. A herbicide spraying assay demonstrated that transgenic plants of garlic conferred herbicide resistance, whilst nontransgenic plants and weeds died. These results indicate that our transformation system can be efficiently utilized to produce transgenic garlic plants with agronomic benefits.  相似文献

Changes in ultrastructure of protein bodies in subaleurone cells of rice endosperm during germination were studied by transmission electron microscopy. The subaleurone cells contained two different types of protein bodies: PB-I (spherical) and PB-II (crystalline). Both types of protein bodies were deconstructed during germination. But there was a considerable difference in digestibility between PB-I and PB-II. PB-II which did not have a dense core was easily digested from the central portion when germination began. At 6 days of germination, PB-II was almost deconstructed. On the other hand, PB-I which displayed concentric rings with a dense core was digested from the outside after 3 days of germination. At 9 days of germination, many kernels of the spherical protein bodies remained.

Changes in subunit composition of protein bodies during germination were investigated by SDS-polyacrylamide gel electrophoresis. Protein body fractions were isolated from germinating grains at various stages by enzymatic digestion and two-phase system, then subjected to SDS-polyacrylamide gel. As germination proceeded, 15 (b1), 20 (d1), 24 (e), 35 (f1) and 37 (f3) kdaltons subunits decreased. On the other hand, 16 (b2), 21 (d2) and 36 (f2) k daltons subunits remained at the later stage of germination. We think that PB-I contains b2, d2 and f2 subunits and is attacked only from the outside at middle and later stages of germination by de novo protease. On the contrary, PB-II contains b1 d1 e, f1 and f3 subunits is utilized at an early stage of germination. PB-II may possibly contain latent protease. The breakdown process of PB-I was by exo-type digestion, on the contrary, that of PB-II was by endo-type digestion.  相似文献
