Three antibiotic-resistance mutations were isolated from strain FL496–2B: two are independent Mendelian genes, one conferring both oligomycin and venturicidin resistance (oliR496) and the other conferring cycloheximide resistance (cyhR496). The third is a mitochondrial mutation, OR9, and confers a low level of oligomycin resistance to cells (in vivo) but not to the extracted mitochondrial ATPase (in vitro). This mutation is located on the mitochondrial DNA at a new locus [OLI4] linked to [OLI2] and independent from [OLI1] and [OLI3] and from the other mitochondrial loci.

All three mutations (O R9, oliR496, cyhR496 ) were found without any selection, in the same prototrophic haploid strain, which contained unknown resistances to antibiotics.

Some physiological, genetical and biochemical properties of the mitochondrial mutation are described.

During a 1-year measurement period, we recorded the CO2 efflux from stems (R S) and coarse woody roots (R R) of 13–20 common tree species at three study sites at 1,050, 1,890 and 3,050 m a.s.l. in an Andean moist forest. The objective of this work was to study elevation changes of woody tissue CO2 efflux and the relationship to climate variation, site characteristics and growth. Furthermore, we aim to provide insights into important respiration–productivity relationships of a little studied tropical vegetation type. We expected R S and R R to vary with dry and humid season conditions. We further expected R S to vary more than R R due to a more stable soil than air temperature regime. Seasonal variation in woody tissue CO2 efflux was indeed mainly attributable to stems. At the same time, temperature played only a small role in triggering variations in R S. At stand level, the ratio of C release (g C m?2 ground area year?1) between stems and roots varied from 4:1 at 1,050 m to 1:1 at 3,050 m, indicating the increasing prevalence of root activity at high elevations. The fraction of growth respiration from total respiration varied between 10 (3,050 m) and 14% (1,050 m) for stems and between 5 (1,050 m) and 30% (3,050 m) for roots. Our results show that respiratory activity and hence productivity is not driven by low temperatures towards higher elevations in this tropical montane forest. We suggest that future studies should examine the limitation of carbohydrate supply from leaves as a driver for the changes in respiratory activity with elevation.  相似文献

The clinical applicability of brain oxygenation mapping using the MOBILE (Mapping of Oxygen By Imaging Lipids relaxation Enhancement) magnetic resonance (MR) technique was assessed in the clinical setting of normal brain and of acute cerebral ischemia as a founding proof-of-concept translational study. Changes in the oxygenation level within healthy brain tissue can be detected by analyzing the spin-lattice proton relaxation (‘Global T 1 ’ combining water and lipid protons) because of the paramagnetic properties of molecular oxygen. It was hypothesized that selective measurement of the relaxation of the lipid protons (‘Lipids T 1 ) would result in enhanced sensitivity of pO2 mapping because of higher solubility of oxygen in lipids than in water, and this was demonstrated in pre-clinical models using the MOBILE technique. In the present study, 12 healthy volunteers and eight patients with acute (48–72 hours) brain infarction were examined with the same clinical 3T MR system. Both Lipids R1 (R1 = 1/T1) and Global R1 were significantly different in the infarcted area and the contralateral unaffected brain tissue, with a higher statistical significance for Lipids R1 (median difference: 0.408 s-1; p<0.0001) than for Global R1 (median difference: 0.154 s-1; p = 0.027). Both Lipids R1 and Global R1 values in the unaffected contralateral brain tissue of stroke patients were not significantly different from the R1 values calculated in the brain tissue of healthy volunteers. The main limitations of the present prototypic version of the MOBILE sequence are the long acquisition time (4 min), hampering robustness of data in uncooperative patients, and a 2 mm slice thickness precluding accurate measurements in small infarcts because of partial volume averaging effects.  相似文献

We estimated respiratory fluxes in a treeline-associated Pinus canariensis forest in Tenerife, Canary Islands, an ecotone with strong seasonal changes in soil water availability. CO2 efflux rates from the foliage, above ground woody tissue and the soil were measured by chamber techniques. Site-specific models obtained from these chamber measurements were then combined with half-hourly measurements of canopy, stem and soil temperature as well as with soil water potential, leaf area and stem sapwood volume data for scaling up component-specific CO2 efflux to ecosystem respiration (R ECO). Integrated over an entire year R ECO was 550 g C m?2 ground surface area (average of 2008 and 2009) and comprised the following component fluxes: 57 % from the soil, 10 % from above ground woody tissue and 33 % from the foliage. Between year differences in R ECO and its components were <3 %. R ECO varied markedly throughout an entire year generally following the seasonal trends in temperature during most of the year. During the dry summer, however, R ECO was significantly reduced due to limited soil water availability in the rooting horizon. Thus, in treeline-associated forests under predicted scenarios of increasing aridity in Mediterranean regions, it is likely that there will be a shift in the contribution of CO2 efflux from the soil (R S), the foliage (R F) and above ground woody tissue (R W) to R ECO from predominately below ground to increasingly above ground. Such changes should be taken into account for predicting the response of treeline forests to a changing climate at their upper distribution limit.  相似文献

ObjectiveTo describe a three-time-point method for estimating kinetic parameters involved in 64Cu-labeled Ramucirumab (64Cu-NOTA-RamAb) trapping of VEGFR-2 positive lung tumors.Materials and methodsPositron emission tomography (microPET) data of tumor-bearing mice for 64Cu-NOTA-RamAb trapping in VEGFR-2 positive HCC4006 tumor were used, involving tissue activity measurements acquired at 3, 24 and 48 h post-injection, without and with administration of RamAb blocking dose. A kinetic model provided an analytical formula describing the tissue time-activity-curve, involving 64Cu-NOTA-RamAb uptake (Ki), release rate constant (kR) and fraction of free tracer in blood and interstitial volume (F).ResultsFitting analytical formula outcomes on mean microPET data yielded values of the kinetic parameters: Ki = 0.0314/0.0123 gram of blood per hour per gram of tissue, kR = 0.0387/0.0313 h−1 and F = 0.2075/0.2007 gram of blood per gram of tissue, without/with RamAb blocking dose, respectively (R = 0.99999 for the graph displaying microPET versus theoretical data; P < .01).ConclusionsThree independent kinetic parameters (Ki, kR and F) can be assessed from three data points acquired at early, mid and late imaging, i.e., at 3, 24 and 48 h post-injection, for further characterization of 64Cu-NOTA-RamAb trapping in VEGFR-2 positive lung tumors.  相似文献

Carcass data were collected from 24 kids (average live weight of 12.5±5.5 kg; range 4.5 to 22.4 kg) of Jarmelista Portuguese native breed, to evaluate bioelectrical impedance analysis (BIA) as a technique for prediction of light kid carcass and muscle chemical composition. Resistance (Rs, Ω) and reactance (Xc, Ω), were measured in the cold carcasses with a single frequency bioelectrical impedance analyzer and, together with impedance (Z, Ω), two electrical volume measurements (VolA and VolB, cm2/Ω), carcass cold weight (CCW), carcass compactness and several carcass linear measurements were fitted as independent variables to predict carcass composition by stepwise regression analysis. The amount of variation explained by VolA and VolB only reached a significant level (P<0.01 and P<0.05, respectively) for muscle weight, moisture, protein and fat-free soft tissue content, even so with low accuracy, with VolA providing the best results (0.326⩽R2⩽0.366). Quite differently, individual BIA parameters (Rs, Xc and Z) explained a very large amount of variation in dissectible carcass fat weight (0.814⩽R2⩽0.862; P<0.01). These individual BIA parameters also explained a large amount of variation in subcutaneous and intermuscular fat weights (respectively 0.749⩽R2⩽0.793 and 0.718⩽R2⩽0.760; P<0.01), and in muscle chemical fat weight (0.663⩽R2⩽0.684; P<0.01). Still significant but much lower was the variation in muscle, moisture, protein and fat-free soft tissue weights (0.344⩽R2⩽0.393; P<0.01) explained by BIA parameters. Still, the best models for estimation of muscle, moisture, protein and fat-free soft tissue weights included Rs in addition to CCW, and accounted for 97.1% to 99.8% (P<0.01) of the variation observed, with CCW by itself accounting for 97.0% to 99.6% (P<0.01) of that variation. Resistance was the only independent variable selected for the best model predicting subcutaneous fat weight. It was also selected for the best models predicting carcass fat weight (combined with carcass length, CL; R2=0.943; P<0.01) and intermuscular fat weight (combined with CCW; R2=0.945; P<0.01). The best model predicting muscle chemical fat weight combined CCW and Z, explaining 85.6% (P<0.01) of the variation observed. These results indicate BIA as a useful tool for prediction of light kids’ carcass composition.  相似文献

Sodium-glucose cotransporter 2 (SGLT2) inhibitors have been shown to increase ketone bodies in patients with type 2 diabetes; however, the underlying mechanisms have not been fully elucidated. Here we examined the effect of the SGLT2 inhibitor dapagliflozin (1 mg/kg/day, formulated in a water, PEG400, ethanol, propylene glycol solution, 4 weeks) on lipid metabolism in obese Zucker rats. Fasting FFA metabolism was assessed in the anesthetized state using a [9,10-3H(N)]-palmitic acid tracer by estimating rates of plasma FFA appearance (Ra), whole-body FFA oxidation (Rox), and nonoxidative disposal (Rst). In the liver, clearance (Kβ-ox) and flux (Rβ-ox) of FFA into β-oxidation were estimated using [9,10-3H]-(R)-bromopalmitate/[U-14C]palmitate tracers. As expected, dapagliflozin induced glycosuria and a robust antidiabetic effect; treatment reduced fasting plasma glucose and insulin, lowered glycated hemoglobin, and increased pancreatic insulin content compared with vehicle controls. Dapagliflozin also increased plasma FFA, Ra, Rox, and Rst with enhanced channeling toward oxidation versus storage. In the liver, there was also enhanced channeling of FFA to β-oxidation, with increased Kβ-ox, Rβ-ox and tissue acetyl-CoA, compared with controls. Finally, dapagliflozin increased hepatic HMG-CoA and plasma β-hydroxybutyrate, consistent with a specific enhancement of ketogenesis. Since ketogenesis has not been directly measured, we cannot exclude an additional contribution of impaired ketone body clearance to the ketosis. In conclusion, this study provides evidence that the dapagliflozin-induced increase in plasma ketone bodies is driven by the combined action of FFA mobilization from adipose tissue and diversion of hepatic FFA toward β-oxidation.  相似文献

Accordingly, substituted enolated malonic dialdehydes (CHO-CR4CR5-OH) react with CoMe3(PMe3)3 to afford 6-coordinate (cis-dimethyl)(2-formyl-enolato)trans-bis(trimethylphosphine)cobalt(III) compounds Co[(CHO-CR4CR5-O)(Me)2](PMe3)2 (3: R4, R5 = (CH2)2C6H4; 4: R4 = R5 = C6H5). In the molecular structure of 4, the cobalt atom is centred in an octahedral coordination geometry brought about by a six-membered chelate ring (O:O-ligand), cis-dimethyl and trans-trimethylphosphine groups. Experiment showed that the response of a genotype to mutation, i.e., the magnitude of mutational change in a phenotypic property, can be correlated with the extent of phenotypic fluctuation among genetic clones. To address a possible statistical mechanical basis for such phenomena at the protein level, we consider a simple hydrophobic-polar lattice protein-chain model with an exhaustive mapping between sequence (genotype) and conformational (phenotype) spaces. Using squared end-to-end distance, RN2, as an example conformational property, we study how the thermal fluctuation of a sequence's RN2 may be predictive of the changes in the Boltzmann average 〈RN2〉 caused by single-point mutations on that sequence. We found that sequences with the same ground-state (RN2)0 exhibit a funnel-like organization under conditions favorable to chain collapse or folding: fluctuation (standard deviation σ) of RN2 tends to increase with mutational distance from a prototype sequence whose 〈RN2〉 deviates little from its (RN2)0. In general, large mutational decreases in 〈RN2〉 or in σ are only possible for some, though not all, sequences with large σ values. This finding suggests that single-genotype phenotypic fluctuation is a necessary, though not sufficient, indicator of evolvability toward genotypes with less phenotypic fluctuations.  相似文献

Manganese(II) complexes [Mn(L)X2] were prepared and characterized, where L is a neutral di-Schiff base ligand incorporating pyridylimine donor arms, including (1R,2R)-N,N′-bis(2-pyridylmethylidene)-1,2-diphenylethylenediimine (L1), (1R,2R)-N,N′-bis(6-methyl-2-pyridylmethylidene)-1,2-cyclohexyldiimine (L2), or (1R,2R)-, (1S,2S)- or racemic N,N′-bis(2-pyridylmethylidene)-1,2-cyclohexyldiimine (L3), and X =  or Cl. Product complexes were structurally characterized, specifically including [Mn(R,R-L1)(NCCH3)3](ClO4)2, [Mn(R,R-L2)(OH2)2](ClO4)2 and racemic [Mn(L3)Cl2]. The first of these complexes features a heptacoordinate ligand field in a distorted pentagonal bipyramid, and the latter two are hexacoordinate, but retain equatorially monovacant pentagonal bipyramidal structures. Complexes [Mn(L3)X2] (X = Cl, ) were reacted with the primary phosphine FcCH2PH2 (Fc = -C5H4FeC5H5), H2O and ethyldiazoacetate (EDA). The first two substrates prompted reactivity at a single ligand imine bond, resulting in hydrophosphination and hydrolysis, respectively. Complexes of the derivative ligands were also structurally characterized. Evidence for EDA activation was obtained by electrospray ionization mass spectrometry, but catalytic carbene transfer was not obtained.  相似文献

Many cardiac arrhythmias are caused by slowed conduction of action potentials, which in turn can be due to an abnormal increase of intracellular myocardial resistance. Intracellular resistivity is a linear sum of that offered by gap junctions between contiguous cells and the cytoplasm of the myocytes themselves. However, the relative contribution of the two components is unclear, especially in atrial myocardium, as there are no precise measurements of cytoplasmic resistivity, Rc. In this study, Rc was measured in atrial tissue using several methods: a dielectrophoresis technique with isolated cells and impedance measurements with both isolated cells and multicellular preparations. All methods yielded similar values for Rc, with a mean of 138 ± 5 Ω·cm at 23°C, and a Q10 value of 1.20. This value is about half that of total intracellular resistivity and thus will be a significant determinant of the actual value of action potential conduction velocity. The dielectrophoresis experiments demonstrated the importance of including divalent cations (Ca2+ and Mg2+) in the suspension medium, as their omission reduced cell integrity by lowering membrane resistivity and increasing cytoplasm resistivity. Accurate measurement of Rc is essential to develop quantitative computational models that determine the key factors contributing to the development of cardiac arrhythmias.  相似文献

Measurements were made of the water proton relaxation rate (T?12 = R2), electron spin resonance (ESR) six-line signal of ‘free’ Mn2+, and O2-evolution activity in thylakoid membranes from pea leaves. The main results are: (1) Aging of thylakoids at 35°C causes a parallel decrease in O2-evolution activity, in R2 and in the content of bound Mn, suggesting that R2 may be related to the loosely bound Mn involved in O2 evolution. (2) Treatment of thylakoids with tetraphenylboron (TPB) at [TPB] > 2 mM produces a 2-fold increase in R2, without release of Mn2+. The titration curve exhibits three sharp end points. The first end point occurs at a [TPB][chlorophyll] of 1.25, at which the O2 evolution is completely inhibited. (3) Treatment of thylakoids with NH2OH also increases R2 by nearly 2-fold, either by the reduction of the higher oxidation states of Mn to Mn2+ and / or by exposing the Mn to solvent protons. Also, progressive release of bound Mn occurs at [NH2OH] ≥ 1 mM as shown by an increase increase in the Mn2+ ESR signal and a decrease in R2. (4) Addition of H2O2 (0.1–1.0%) to thylakoids causes an enhancement of R2 similar to that by NH2OH, but without the release of Mn2+. (5) Heat treatment of thylakoids at 40–50°C releases Mn2+ and increases R2. Conversely, pH values of 7 to 4 release Mn2+ without changing R2 while pH values of 7–9 increase R2 without releasing Mn2+. Thus, both high and low pH values as well as the heat treatment cause structural changes enhancing the relaxivity of the bound Mn or of other paramagnetic species.  相似文献

The R21(TC) factor, obtained by transduction of the R10(TC.CM.SM.SA) factor with phage ε to group E Salmonella, is not transferable by the normal conjugal process. However, when R21(TC)+ transductants are infected with the F13 factor, the nontransferable R21(TC) factor acquires transmissibility by conjugation. R21(TC)+ conjugants of Escherichia coli K-12, to which only the R21(TC) factor was transmitted by cell-to-cell contact from an F′ R+ donor, were still unable to transfer their R21(TC) factor by conjugation. In crosses between Hfr and FE. coli K-12 strains containing R21(TC), the gene responsible for tetracycline resistance was located on the E. coli K-12 chromosome between lac and pro, near lac.  相似文献

Interannual variations in CO2 exchange across Amazonia, as deduced from atmospheric inversions, correlate with El Niño occurrence. They are thought to result from changes in net ecosystem exchange and fire incidence that are both related to drought intensity. Alterations to net ecosystem production (NEP) are caused by changes in gross primary production (GPP) and ecosystem respiration (Reco). Here, we analyse observations of the components of Reco (leaves, live and dead woody tissue, and soil) to provide first estimates of changes in Reco during short-term (seasonal to interannual) moisture limitation. Although photosynthesis declines if moisture availability is limiting, leaf dark respiration is generally maintained, potentially acclimating upwards in the longer term. If leaf area is lost, then short-term canopy-scale respiratory effluxes from wood and leaves are likely to decline. Using a moderate short-term drying scenario where soil moisture limitation leads to a loss of 0.5 m2 m−2 yr−1 in leaf area index, we estimate a reduction in respiratory CO2 efflux from leaves and live woody tissue of 1.0 (±0.4) t C ha−1 yr−1. Necromass decomposition declines during drought, but mortality increases; the median mortality increase following a strong El Niño is 1.1% (n=46 tropical rainforest plots) and yields an estimated net short-term increase in necromass CO2 efflux of 0.13–0.18 t C ha−1 yr−1. Soil respiration is strongly sensitive to moisture limitation over the short term, but not to associated temperature increases. This effect is underestimated in many models but can lead to estimated reductions in CO2 efflux of 2.0 (±0.5) t C ha−1 yr−1. Thus, the majority of short-term respiratory responses to drought point to a decline in Reco, an outcome that contradicts recent regional-scale modelling of NEP. NEP varies with both GPP and Reco but robust moisture response functions are clearly needed to improve quantification of the role of Reco in influencing regional-scale CO2 emissions from Amazonia.  相似文献

Amoebae of the Myxomycete Physarum polycephalum differentiate to yield plasmodia in two ways: in crossing, haploid amoebae of appropriate genotypes fuse to form diploid plasmodia; in selfing, plasmodia form without amoebal fusion or increase in ploidy. Amoebae carrying the mating-type allele matAh (formerly mth) self efficiently, but occasionally give rise to mutants that self at very low frequencies. Such "amoebal-plasmodial transition" mutants were mixed in pairs to test their ability to complement one another in the formation of plasmodia by crossing. The pattern of crossing permitted 33 mutants to be assigned to four complementation groups (aptA-, npfA-, npfB- and npfC-). Similar tests had previously proved only partially successful, as crossing had occurred only rarely in mixtures of compatible strains. The efficiency of complementation was greatly increased in the current work by mixing strains that carried different alleles of a newly-discovered mating-compatibility locus, matB; this locus had no effect on the specificity of complementation. A possible interpretation of the complementation behavior of the mutants is suggested.  相似文献

An epimeric mixture of 24-hydroxy-[24-3H]vitamin D3 was synthesized by the reduction of 24-ketovitamin D3 by sodium borotritide. The epimeric mixture was converted to the trimethylsilylether derivatives and subjected to high-pressure liquid chromatography using silica gel columns to separate the 24-hydroxy-[24-3H]vitamin D3 isomers. The 24R-hydroxy-[24-3H] vitamin D3 induced calcification in rachitic rats while the 24S-hydroxy-[24-3H] vitamin D3 had little or no such activity. As both isomers of 24-hydroxy-vitamin D3 are metabolized to 24,25-dihydroxyvitamin D3, it appears that the 24-hydroxyvitamin D3-25-hydroxylase does not discriminate between the isomers. Only the R-isomer of 24-hydroxyvitamin D3 is metabolized to 1,24-dihydroxyvitamin D3, although only trace amounts of this compound were found 2 days after the administration of 24-hydroxyvitamin D3. The striking difference in the metabolism of the isomers is the high selectivity of the 1-hydroxylase for R-isomer. It is suggested that the high specificity of biological activity for the R-isomer of 24-hydroxyvitamin D3 is because of the specificity of the 1-hydroxylation of 24,25-dihydroxyvitamin D3 for the R configuration.  相似文献

The genome of the rice blast fungus Magnaporthe oryzae codes for two proteins with N-terminal dioxygenase (DOX) and C-terminal cytochrome P450 (CYP) domains, respectively. One of them, MGG_13239, was confirmed as 7,8-linoleate diol synthase by prokaryotic expression. The other recombinant protein (MGG_10859) possessed prominent 10R-DOX and epoxy alcohol synthase (EAS) activities. This enzyme, 10R-DOX-EAS, transformed 18:2n-6 sequentially to 10(R)-hydroperoxy-8(E),12(Z)-octadecadienoic acid (10R-HPODE) and to 12S(13R)-epoxy-10(R)-hydroxy-8(E)-octadecenoic acid as the end product. Oxygenation at C-10 occurred by retention of the pro-R hydrogen of C-8 of 18:2n-6, suggesting antarafacial hydrogen abstraction and oxygenation. Experiments with 18O2 and 16O2 gas confirmed that the epoxy alcohol was formed from 10R-HPODE, likely by heterolytic cleavage of the dioxygen bond with formation of P450 compound I, and subsequent intramolecular epoxidation of the 12(Z) double bond. Site-directed mutagenesis demonstrated that the cysteinyl heme ligand of the P450 domain was required for the EAS activity. Replacement of Asn965 with Val in the conserved AsnGlnXaaGln sequence revealed that Asn965 supported formation of the epoxy alcohol. 10R-DOX-EAS is the first member of a novel subfamily of DOX-CYP fusion proteins of devastating plant pathogens.  相似文献

Small-angle neutron scattering studies of Escherichia coli tyrosyl-tRNA synthetase indicate that in solution this enzyme is a dimer of Mr, 91 (±6) × 103 with a radius of gyration RG of 37.8 ± 1.1 Å.The increase in the scattering mass of the enzyme upon binding tRNATyr has been followed in 20 mm-imidazole · HCl (pH 7.6), 10 mm-MgCl2, 0.1 mm-EDTA, 10 mm-2-mercaptoethanol, 150 mm-KCl. A stoichiometry of one bound tRNA per dimeric enzyme molecule was found. The RG of the complex is equal to 41 ± 1 Å. Titration experiments in 74% 2H2O, close to the matching point of tRNA, show an RG of 38.5 ± 1 Å for the enzyme moiety in the complex. From these values, a minimum distance of 49 Å between the centre of mass of the bound tRNA and that of the enzyme was calculated.In low ionic strength conditions (20 mm-imidazole-HCl (pH 7.6), 10 mm-MgCl2, 0.1 mm-EDTA, 10 mm-2-mercaptoethanol) and at limiting tRNA concentrations with respect to the enzyme, titrations of the enzyme by tRNATyr are characterized by the appearance of aggregates, with a maximum scattered intensity at a stoichiometry of one tRNA per two enzyme molecules. At this point, the measured Mr and RG values are compatible with a compact 1:2, tRNA: enzyme complex. This complex forms with a remarkably high stability constant: (enzyme:tRNA:enzyme)/(enzyme:tRNA)(enzyme) of 0.1 to 0.3(× 106) m?1 (at 20 °C). The input impedance of muscle fibers of the crab was determined with microelectrodes over the frequency range 1 cps to 10 kc/sec. Care was taken to analyze, reduce, and correct for capacitive artifact. One dimensional cable theory was used to determine the properties of the equivalent circuit of the membrane admittance, and the errors introduced by the neglect of the three dimensional spread of current are discussed. In seven fibers the equivalent circuit of an element of the membrane admittance must contain a DC path and two capacitances, each in series with a resistance. In two fibers, the element of membrane admittance could be described by one capacitance in parallel with a resistance. In several fibers there was evidence for a third very large capacitance. The values of the elements of the equivalent circuit depend on which of several equivalent circuits is chosen. The circuit (with a minimum number of elements) that was considered most reasonably consistent with the anatomy of the fiber has two branches in parallel: one branch having a resistance Re in series with a capacitance Ce; the other branch having a resistance Rb in series with a parallel combination of a resistance Rm and a capacitance Cm. The average circuit values (seven fibers) for this model, treating the fiber as a cylinder of sarcolemma without infoldings or tubular invaginations, are Re = 21 ohm cm2; Ce = 47 µf/cm2; Rb = 10.2 ohm cm2; Rm = 173 ohm cm2; Cm = 9.0 µf/cm2. The relation of this equivalent circuit and another with a nonminimum number of circuit elements to the fine structure of crab muscle is discussed. In the above equivalent circuit Rm and Cm are attributed to the sarcolemma; Re and Ce, to the sarcotubular system; and Rb, to the amorphous material found around crab fibers. Estimates of actual surface area of the sarcolemma and sarcotubular system permit the average circuit values to be expressed in terms of unit membrane area. The values so expressed are consistent with the dielectric properties of predominantly lipid membranes.  相似文献

The generation in cyclic fashion of 26 mutable R-Navajo (mRnj) alleles in maize involved transposition of a non-specific repressor of gene action, Modulator (Mp), first away from, and then back to, the R locus represented by the R-Navajo (Rnj) allele on chromosome 10. The mRnj alleles reconstituted in this way varied widely, and continuously, in mutability to Rnj—that is, in transposition of Mp away from the R locus, thus derepressing the Rnj gene. They were alike, or nearly so, however, in activating Ds chromosome breakage and in increasing the stability of variegated pericarp, another unstable compound allele comprising Mp conjoined with Prr on chromosomal 1. These latter two phenomena are based primarily on loci elsewhere in the genome. It is postulated that the 26 reconstituted mRnj alleles carry a common Mp which, however, is intercalated at a different site within each allele. Nucleotide sequence in the regions adjacent to Mp is assumed to determine the frequency with which a form of micro-nondisjunction occurs whereby Mp is released from a donor site. Transposition to a new site is interpreted in terms of a chromosome model that gives effect to nicking, or single strand breaks, occurring throughout the genome as a prerequisite to unwinding, strand separation, and replication, of the DNA double helix.  相似文献
