 In samples from 56 populations of Cardamine amara, representing four diploid subspecies (subspp. amara, opicii, balcanica, and pyrenaea) and two tetraploid subspecies (subspp. austriaca and olotensis) from different parts of the European distribution area, four enzyme systems with 23 alleles were studied. These data, together with previous morphological and karyological data, suggest that the distribution and variation pattern within the species was strongly influenced by the last glacial period and postglacial migrations. Cardamine amara subsp. pyrenaea is monomorphic for a unique allele, and subsp. balcanica has a unique allele, too which, however, is not fixed in all populations of the taxon. Both taxa seem to be relic ones, although otherwise subsp. balcanica in respect of allelic spectrum much resembles subsp. amara. The other two diploid subspecies, subsp. amara and subsp. opicii, are not characterised by presence of unique alleles but differ in allele frequencies. The two tetraploid subspecies have different evolutionary histories. C. amara subsp. austriaca seems to be an autopolyploid derivative of subsp. amara which colonised open space offered by retreating glaciers in the Eastern Alps. C. amara subsp. olotensis from the Iberian Peninsula represents most probably a polyploid of preglacial time. Distribution data on Cardamine amara L. in Serbia and Republic of Macedonia are presented, together with a short review of the history of taxonomic treatment of C. amara in this area. C. amara subsp. balcanica Marhold, Ančev & Kit Tan is reported as a new taxon both for the flora of Serbia and flora of Republic of Macedonia and the occurrence of C. amara subsp. amara in Serbia was confirmed.  相似文献

Hybridization between two diploid (2n = 2x = 16) species ofBrassicaceae, Cardamine rivularis andC. amara, at Urnerboden, Central Switzerland, resulted in the rather unusual triploid hybridC. insueta (2n = 3x = 24), and later on in the amphiploidC. schulzii (2n = 6x = 48). The hybrid and the neopolyploid species colonized successfully some man-made biotopes. Plants ofC. insueta are mostly functional females with non-dehiscent anthers, but true hermaphrodite individuals with partly sterile pollen grains also occur within the population. Analyses of cpDNA and nuclear DNA permitted to establish the parentage of the hybrid: the maternal parent which contributed unreduced egg cells proved to beC. rivularis whereas the normally reduced pollen originated fromC. amara. The pronounced genetic variability inC. insueta revealed by isozyme and RAPD analyses, at variance with the polarized segregation, heterogamy and strong vegetative reproduction of the hybrid, is possibly influenced by recurrent formation ofC. insueta which party results from backcrosses betweenC. insueta andC. rivularis but may also proceed by other pathways. The amphiploidCardamine schulzii has normally developed anthers but its pollen is sometimes highly sterile. The surprisingly uniform genetic make-up of the new amphiploid species might be related to its possible monotopic origin and/or young phylogenetic age but should be further assessed. Site management seems to be very important to a further development of hybridogenous populations and their parent species. In conclusion, the evolution at Urnerboden is discussed in the context of the traditional concept of multiple plant origins.  相似文献

Analyses of mitotic chromosome numbers and nuclear DNA content were performed for 39 populations of 17 perennial Cerastium taxa from south‐eastern Europe. The DNA content ranged from 2C = 2.43 to 8.78 pg, revealing four ploidy levels corresponding to 4x (2n = 36), 8x, 12x and 16x. High‐polyploid cytotypes with a greater range of ploidy (up to 2n = 144) occur mostly in the central mountainous parts of the Balkan Peninsula. The chromosome number was determined for the first time for C. dinaricum (2n = 36 + 1B), C. decalvans subsp. orbelicum (2n = 36), C. decalvans subsp. glutinosum (2n = 36), C. neoscardicum (2n = 144), C. malyi subsp. serpentini (2n = 144) and C. moesiacum s.s. (2n = 144). New chromosome counts were recorded for C. arvense subsp. strictum (2n = 108), C. banaticum subsp. kosaninii (2n = 36) and C. grandiflorum (2n = 36). For the first time, flow cytometry was used to estimate C values for six species (15 taxonomic entities). The intraspecific variation quotient of C values is high, ranging from 1.003 in C. malyi to 1.306 in C. decalvans subsp. decalvans. The variation in chromosome size among both tetra‐ and octoploid members of Cerastium is much more prominent than in most other angiosperm polyploid series. Significant genome downsizing after polyploidization was observed in some investigated taxa. Differences in ploidy levels and monoploid genome size values confirm the taxonomic status of C. decalvans subsp. glutinosum and C. decalvans subsp. leontopodium. 为探讨国产毛茛科(Ranunculaceae)驴蹄草属(Caltha L.)植物的细胞学特征,对驴蹄草(C.palustris L.)3个居群和花葶驴蹄草(C.scaposa Hook.f.&Thoms.)5个居群进行了细胞学研究。驴蹄草贵州纳雍居群的染色体数目为2n=32(四倍体),两个云南中甸居群的染色体数目均为2n=64(八倍体)。花葶驴蹄草四川红原、康定、石渠居群的染色体数目均为2n=32(四倍体),该数目为首次报道;西藏林芝和云南德钦居群的染色体数目均为2n=64(八倍体)。驴蹄草的染色体比花葶驴蹄草大。这两种植物的32或64条染色体分别以4条或8条为单位大致能够排列为8组同源染色体,但同一组内的染色体经常具有明显的异形性(heteromorphy),不同居群的核型组成多少具有差异。同时,还分析了驴蹄草和花葶驴蹄草的不同倍性细胞型在我国的地理分布式样。  相似文献

New chromosome numbers for two species from the Senecio nemorensis group: S. dacicus (2n = 40) and S. ucranicus (2n = 40) have been ascertained. The counts for S. germanicus Wallr. subsp. germanicus (2n = 40), S. hercynicus Herborg subsp. hercynicus (2n = 40), S. ovatus (P. Gaertn. et al.) Willd. subsp. ovatus (2n = 40) occurring in the Carpathians are also reported. The study confirmed only the known tetraploid chromosome number for the taxa of this group. The pollen fertility ranged from 82.09 to 92.99% in all examined species and subspecies, including their hybrids.  相似文献

Nasturtium officinale (2n=4x=32),N. microphyllum (2n=8x=64) andN. ×sterile (2n=6x=48) have been investigated by isozyme analyses to study evolutionary processes withinNasturtium. Four additional species assumed to be involved in the formation of the octoploidN. microphyllum (Rorippa amphibia, R. sylvestris, R. palustris, andCardamine amara) were also examined. A total of 641 individuals were analyzed for six isozyme systems (alcohol dehydrogenase, aspartate aminotransferase, glutamate dehydrogenase, leucine aminopeptidase, malate dehydrogenase, and phosphoglucoisomerase). Eleven gene complexes coding for 43 allozymes were detected. Fifteen alleles were observed inN. officinale, twelve of them being fixed. All alleles fixed inN. officinale were also present and fixed inN. microphyllum. Seven additional fixed alleles were observed inN. microphyllum. The presence of these seven alleles inRorippa taxa provide evidence for an allopolyploid origin ofN. microphyllum withN. officinale and aRorippa taxon involved.C. amara is not a parent species ofN. microphyllum. N. ×sterile showed a fixed banding pattern which was identical to that ofN. microphyllum. It is argued thatN. ×sterile is a hybrid betweenN. officinale andN. microphyllum. Human impact has played a major role in the evolution ofN. ×sterile. The formation and persistence of the hybrid were influenced by introducingN. officinale into the natural distribution area ofN. microphyllum and by creating ditches and ponds where due to its vegetative capabilitiesN.×sterile could establish.  相似文献

The phylogeography of the African four-striped mouse, Rhabdomys pumilio, was investigated using complete sequences of the mtDNA cytochrome b gene (1140 bp) and a combination of fluorescence in situ hybridization (FISH) and conventional cytogenetic banding techniques (G- and C-banding). Two cytotypes (2n=46 and 2n=48) were identified by cytogenetic analysis. There is no evidence of diploid number variation within populations, difference in gross chromosome morphology or of subtle interchromosomal rearrangements at levels detected by ZOO-FISH. Analysis of the mtDNA cytochrome b resulted in two major lineages that correspond roughly to the xeric and mesic biotic zones of southern Africa. One mtDNA clade comprises specimens with 2n=48 and the other representatives of two cytotypes (2n=48 and 2n=46). The mean sequence divergence (12%, range 8.3–15.6%) separating the two mtDNA clades is comparable to among-species variation within murid genera suggesting their recognition as distinct species, the prior names for which would be R. dilectus and R. pumilio. Low sequence divergences and the diploid number dichotomy within the mesic lineage support the recognition of two subspecies corresponding to R. d. dilectus (2n=46) and R. d. chakae (2n=48). Our data do not support subspecific delimitation within the nominate, R. pumilio. Molecular dating places cladogenesis of the two putative species at less than five million years, a period characterised by extensive climatic oscillations which are thought to have resulted in habitat fragmentation throughout much of the species range.  相似文献

This article describes the use of cytogenomic and molecular approaches to explore the origin and evolution of Cardamine schulzii, a textbook example of a recent allopolyploid, in its ∼110-year history of human-induced hybridization and allopolyploidy in the Swiss Alps. Triploids are typically viewed as bridges between diploids and tetraploids but rarely as parental genomes of high-level hybrids and polyploids. The genome of the triploid semifertile hybrid Cardamine × insueta (2n = 24, RRA) was shown to combine the parental genomes of two diploid (2n = 2x = 16) species, Cardamine amara (AA) and Cardamine rivularis (RR). These parental genomes have remained structurally stable within the triploid genome over the >100 years since its origin. Furthermore, we provide compelling evidence that the alleged recent polyploid C. schulzii is not an autohexaploid derivative of C. × insueta. Instead, at least two hybridization events involving C. × insueta and the hypotetraploid Cardamine pratensis (PPPP, 2n = 4x−2 = 30) have resulted in the origin of the trigenomic hypopentaploid (2n = 5x−2 = 38, PPRRA) and hypohexaploid (2n = 6x−2 = 46, PPPPRA). These data show that the semifertile triploid hybrid can promote a merger of three different genomes and demonstrate how important it is to reexamine the routinely repeated textbook examples using modern techniques.  相似文献

Considerable karyotypic differentiation has occurred within the group of taxa comprising the eastern North American members of the genus Claytonia. Patterns of karyotypic differentiation are congruent with evolutionary groupings based on flavonoid chemistry, particularly at the diploid level. The 2n = 16 diploid chemotype found in both C. caroliniana and C. virginica possesses a karyotype composed entirely of metacentric chromosomes, while acrocentric chromosomes predominate in the karyotypes of the 2n = 12 and 2n = 14 diploid chemotypes of C. virginica. The 2n = 16 diploid also has a karyotype significantly larger than those of the other diploid cytotypes. Polyploid karyotypes of both species show varying degrees of divergence from their presumed diploid progenitors.  相似文献

The occurrence of diploid and tetraploid cytotypes in Thymus section Mastichina (Lamiaceae) was investigated by chromosome counting and flow cytometric measurements of DNA content. To cover the entire native distribution of the section across the Iberian Peninsula, plant material was sampled from 29 populations, representing all three taxa (Thymus albicans, T. mastichina subsp. mastichina and T. mastichina subsp. donyanae). We provide first estimates of genome size for taxa of this section. Analyses revealed the existence of new cytotypes in T. mastichina subsp. mastichina (2n?=?2x?=?28, 30) and confirmed the existence of previous ones (2n?=?4x?=?56, 58, 60). We also confirmed the presence of exclusively diploid cytotypes (2n?=?2x?=?30) in the southwestern Iberian endemics T. albicans and T. mastichina subsp. donyanae. We conclude that the southwestern Iberian Peninsula acted as a Pleistocene glacial refugium that may have permitted speciation processes within this section. Several hypotheses concerning these processes and the origin and distribution of the studied taxa are discussed.  相似文献

Cardamine battagliae Cesca & Peruzzi sp. nova is related to Cardamine heptaphylla (Vill.) O. E. Schulz. (2n = 48). On the other hand, the new species is well distinguished both from a morphological and karyological point of view. Because of its distribution and high ploidy level (2n = ca. 160) C. battagliae can be considered as a new apoendemic taxon. Morphological and karyological data are presented and discussed.  相似文献

Crocus speciosus is a widely distributed autumn-flowering species showing considerable variation in its morphological characters. Three subspecies are recognised, two of which are endemic to small areas of Turkey. The third, subsp.speciosus, occurs in a much wider area and contains a number of different cytotypes.C. pulchellus is a distinct but closely related species. The morphological relationships between the species and subspecies are discussed and a key to the taxa is provided. Cytological data are given and a distribution map of the cytotypes is provided. Analyses of meiotic and mitotic chromosomes were undertaken in 16 taxa of Echinocereus belonging to 12 species and all seven taxonomic sections (sensu Taylor). Chromosome numbers are reported for the first time for eight taxa, and previously published chromosome counts are confirmed for the remaining eight. Both diploid and polyploid counts were obtained. Eleven (69%) of the taxa surveyed were diploid (2n = 22); the five varieties of E. engelmannii were polyploid (2n = 44). Overall, chromosome counts are available for 23 of the 48 proposed species (sensu Taylor). Of these, 19 (82%) are diploid, and four (18%) are polyploid. Polyploid cytotypes are most common in the primitive sections, e.g., sections Erecti and Triglochidiatus, which suggests that polyploidy is probably a derived condition in Echinocereus. Polyploid taxa range from medium to high latitudes and elevations relative to the overall distribution of the genus. Polyploidy, hybridization, and cryptic chromosomal rearrangements are thought to be the major causes of the speciation events of the genus.  相似文献

The Poa laxa Haenke species group, comprising four alpinearctic taxa in P. sect. Oreinos, has a complicated and confusing taxonomic history. Here we re‐examine the taxonomy of the group and section based on sequences of three plastid and two nuclear ribosomal DNA markers. Poa laxa s.l. resolved in a clade with species of sections Abbreviatae, Oreinos, Stenopoa, and Tichopoa. In the plastid analyses, Poa laxa s.s. (type of sect. Oreinos) was placed in a subclade with P. glauca and other sect. Stenopoa species, while all other P. laxa s.l. taxa were placed in a subclade with species of sect. Abbreviatae. We maintain P. laxa s.s. (mainly 2n = 28) of west–central Europe in sect. Oreinos, while the other P. laxa s.l. taxa from northern Europe and North America (all 2n = 42 or higher ploid) are referred to P. sect. Abbreviatae. In North America only one collection of the northern European P. flexuosa subsp. flexuosa is accepted (Greenland, Jensen's Nunatakker), and P. flexuosa subsp. fernaldiana is accepted based on populations in eastern Canada and northeast USA. A new subspecies, P. flexuosa subsp. consauliae, is described from eastern Arctic Canada and Greenland; its variable morphological characteristics suggest introgression with P. glauca and it is predicted to be apomictic. Rocky Mountain P. laxa subsp. banffiana is treated as P. banffiana. Oregon material formerly considered to belong to P. laxa s.l. is named P. wallowensis. Poa sect. Oreinos is thus found to be polyphyletic, and is here emended to comprise only three continental European species, including P. laxa. Lectotypes are designated for P. aspera var. laxiuscula, P. fernaldiana, P. flexuosa, and P. laxa var. debilior. New chromosome numbers are reported for P. flexuosa subsp. consauliae (ca 2n = 42), and P. glauca (2n = 63) from Baffin Island. A key to the species and subspecies found in North America is given.  相似文献

To reveal the general cytogeographical pattern of Cyanus section Protocyanus in Europe, DNA ploidy and/or chromosome numbers were newly examined for 160 populations by flow cytometry (450 plants) and/or chromosome counting (30 plants). Furthermore, previously published karyological data were revised (236 records). Our analyses confirmed chromosome counts of 2n = 22 for all newly investigated samples of the C. triumfetti group (the records for C. semidecurrens and C. ternopoliensis are new), C. diospolitanus and C. achtarovii; 2n = 44 for C. montanus and C. mollis; and 2n = 20 for C. lingulatus, C. napulifer, C. nissanus, C. orbelicus, C. thirkei, C. tuberosus and C. velenovskyi. The chromosome count of 2n = 20 is the first report for C. epirotus. The cytotype 2n = 40 was newly recorded for the Crimean endemic C. fuscomarginatus and Calabrian and Greek populations of C. graminifolius. The cytotypes 2n = 20 and 2n = 40 were confirmed for C. pindicola. For the first time triploidy (2n~3x~30) was found in C. nissanus, C. thirkei and in a newly discovered hybrid, C. epirotus × C. graminifolius. Two contrasting ecogeographical patterns emerged: cytotypes derived from the base chromosome number x = 11 (2n = 22, 44) are widespread in northern latitudes and ecologically diverse, whereas cytotypes with x = 10 (2n = 20, 30, 40) are confined to mountains in southern Europe. In general, tetraploids have smaller ranges than diploids. The new combinations Cyanus section Protocyanus (Dobrocz.) Ol?avská comb. nov. and Cyanus ternopoliensis (Dobrocz.) Principal coordinate analysis using Gower’s similarity coefficient and canonical discriminant analysis were performed using morphological data to assess species distinctness in theCardamine concatenata alliance, comprising the eastern North American dentarias. The results of an overall principal coordinate analysis of the alliance using quantitative and qualitative characters revealed distinct and separate clusters that correspond to five previously recognized species (C. angustata, C. concatenata, C. diphylla, C. dissecta, C. maxima). One additional taxon,C. incisa, was found to group closely with theC. diphylla cluster. In a second principal coordinate analysis including only individuals fromC. diphylla andC. incisa, both are supported as distinct morphological entities. Canonical discriminant analysis of only above-ground quantitative characters was less effective in distinguishing previously recognized taxa, in keeping with the emphasis on qualitative characters in previous taxonomic treatments of the group.Cardamine incisa (Eames) Schumann (pro hybr.) is raised to the rank of species. Somatic chromosome numbers have been determined for the followingCerastium taxa:C. eriophorum (2n = 36),C. alpinum (2n = 72),C. transsylvanicum (2n = 108),C. arcticum (2n = 108),C. latifolium (2n = 36),C. carinthiacum (2n = 36),C. banaticum (2n = 36),C. arvense subsp.glandulosum (2n = 36),C. arvense subsp.arvense (2n = 72) andC. fontanum (2n = 144). Karyotypes of three diploid species (C. eriophorum, C. banaticum andC. latifolium), belonging to three different taxonomic groups, were analysed and found to be similar. The relative nuclear DNA contents of all taxa were determined by flow cytometry and, for five species, also by Feulgen cytophotometry. The values obtained by the two methods are similar. A comparison of nuclear DNA contents among diploids shows that values differ significantly between different taxonomic groups, and are correlated with average chromosome size. Within closely related polyploid groups nuclear DNA amounts increase from 2x- to 4x- and 6x taxa as 1 : 1.4 : 2.4 in theC. alpinum complex, whereas DNA amounts are doubled comparing 2x- and 4x-subspecies in theC. arvense complex.  相似文献

Evolution and taxonomic split of the model grass Brachypodium distachyon   总被引:1,自引:0,他引:1  

Background and Aims

Brachypodium distachyon is being widely investigated across the world as a model plant for temperate cereals. This annual plant has three cytotypes (2n =  10, 20, 30) that are still regarded as part of a single species. Here, a multidisciplinary study has been conducted on a representative sampling of the three cytotypes to investigate their evolutionary relationships and origins, and to elucidate if they represent separate species.

Methods

Statistical analyses of 15 selected phenotypic traits were conducted in individuals from 36 lines or populations. Cytogenetic analyses were performed through flow cytometry, fluorescence in situ hybridization (FISH) with genomic (GISH) and multiple DNA sequences as probes, and comparative chromosome painting (CCP). Phylogenetic analyses were based on two plastid (ndhF, trnLF) and five nuclear (ITS, ETS, CAL, DGAT, GI) genes from different Brachypodium lineages, whose divergence times and evolutionary rates were estimated.

Key Results

The phenotypic analyses detected significant differences between the three cytotypes and demonstrated stability of characters in natural populations. Genome size estimations, GISH, FISH and CCP confirmed that the 2n = 10 and 2n = 20 cytotypes represent two different diploid taxa, whereas the 2n = 30 cytotype represents the allotetraploid derived from them. Phylogenetic analysis demonstrated that the 2n = 20 and 2n = 10 cytotypes emerged from two independent lineages that were, respectively, the maternal and paternal genome donors of the 2n = 30 cytotype. The 2n = 20 lineage was older and mutated significantly faster than the 2n = 10 lineage and all the core perennial Brachypodium species.

Conclusions

The substantial phenotypic, cytogenetic and molecular differences detected among the three B. distachyon sensu lato cytotypes are indicative of major speciation processes within this complex that allow their taxonomic separation into three distinct species. We have kept the name B. distachyon for the 2n = 10 cytotype and have described two novel species as B. stacei and B. hybridum for, respectively, the 2n = 20 and 2n = 30 cytotypes.  相似文献
