Morphologically intact structures have been isolated from anaerobically grown yeast cells which have many of the properties of yeast mitochondria. The structures are about 0.5 µ in diameter and contain malate dehydrogenase, succinate dehydrogenase, oligomycin-sensitive ATPase, and DNA of buoyant density 1.683 g/cc, characteristic of yeast mitochondria. The morphology of the structures is critically dependent on their lipid composition. When isolated from cells grown anaerobically in the presence of supplements of unsaturated fatty acid and ergosterol, their unsaturated fatty acid content is similar to that of mitochondria from aerobically grown cells. These lipid-complete structures consist pre-dominantly of double-membrane vesicles enclosing a dense matrix which contains a folded inner membrane system bordering electron-transparent regions which are somewhat different from the cristae of functional mitochondria. In contrast, the structures from cells grown without lipid supplements are much simpler in morphology; they have a dense granular matrix surrounded by a double membrane but have no obvious folded inner membrane system within the matrix. The lipid-depleted structures are very fragile and are only isolated in intact form from protoplasts that have been prefixed with glutaraldehyde  相似文献

Mitochondrial fractions, relatively free from contamination by other cytoplasmic structures, have been isolated by differential centrifugation from homogenates of brown adipose tissue from starved rats. It was possible in such fractions to distinguish two types of mitochondria in this tissue. Type I mitochondria, when morphologically intact, are limited by a bilaminar membrane and show regular parallel cristae. In isolated fractions, a proportion of these mitochondria are swollen, vacuolation occurring within the cristae between their limiting membranes. Type II mitochondria are distinguished from the more numerous type I bodies by the opaque appearance of their matrix. They are limited by a membrane which is in part single, and in part double. They show a few, but crisply outlined internal membranes. Vacuolation of this type of mitochondrion has not been observed. Vacuolation comparable to that in brown fat mitochondria was also observed between the two laminae of the enclosing membrane and within the cristae of liver mitochondria.  相似文献

Summary Axon profiles in thyroid glands obtained from adult male Wistar rats were studied electron-microscopically, using common and serial thin sections.Bouton profiles of nerve fibers, resembling the terminal or en passant type, often appeared closely associated with vascular smooth muscle cells via basement membranes. These structures are probably adrenergic, since they contained mainly small-core vesicles (mean diameter: 41.2 nm), in addition to a few large-core (mean diameter: 88.4 nm) and flattened vesicles.Nerve fibers containing microtubules and sometimes mitochondria and vesicles were seen lying between basement membranes and follicular cells. The incidence of nerve fiber contacts on profiles of follicular cells was 0.0177±0.0092 (S.D.). Using serial sections, follicles were seen to have up to two nerve endings, separated from the plasma membranes of the follicular cells by a gap of 22 nm. They contained mainly flattened vesicles and several large-core vesicles (mean diameter: 95.1 nm). Small-core vesicles were rarely seen in these nerve endings. Furthermore, subsurface cistern-like rough endoplasmic reticulum was found immediately under the plasma membranes of follicular cells facing membranes of nerve endings. These results suggest that the nerve fibers in contact with follicular cells are different from the adrenergic type.  相似文献

Unusual huge organelles together with mitochondria of typical organization were found in a part of population of callus cells of pea. They like mitochondria are surrounded by two biologic membranes, the inner one has invaginations. Inclusions in the matrix are a specific character of structural organization of these unusual mitochondria. These inclusions look as electronically dense bars that extend parallel to longitudinal axis of organelle and consist of fibril-like elements. Organization of inclusions and cristae as well as a shape of organelles are changing during cell development. Possible nature of the unusual mitochondria is discussed.  相似文献

In the sac-like ovary of the polyplacophoran mollusc, Lepidochitona cinerea , nutritive tissue arises from the ventral gonadal wall of the organ as prominent folds which support the oocytes during the various stages of their development. Each oocyte is enveloped by the follicular epithelium. Approximately twenty follicle cells surround one full-grown oocyte and by this late stage are connected to it and to each other by desmosomes. The follicle cells contain glycogen, Golgi dictyosomes, mitochondria, lipid droplets, numerous cisternae and vesicles of the rough endoplasmic reticulum, and various kinds of lysosomes. The nutritional function of these cells and their possible role forming the oocytic hulls is discussed.  相似文献

Between 1950 and 1960 mitochondria were recognized as well‐characterized organelles of animal and fungal cells. They shared more functional autonomy than other cellular structures. The transmission of some mitochondrial characteristics did not obey Mendelian rules and followed cytoplasmic inheritance patterns. Was this situation a consequence of still unknown complexities? We present a personal account on how approaches were set up to test very different hypotheses. In the end, it was shown that mitochondria had their own DNA, mitochondrial DNA, and that this molecule carried information specific to these organelles.  相似文献

Morphologically, the salivary glands of ticks are paired structures consisting of a secretory and an excretory portion, lacking a reservoir for the storage of the secretion. The secretory portion is composed in females by cells that form acini classified into the types I, II, and III. The excretory possess a major duct, from which arise several intermediate ducts that then subdivide to form the canaliculi or acinal tubules, which end at the acini from where they collect the secretion. The present study describes the ultrastructural changes that occur in the mitochondria of cells of the acini I, II, and III in the salivary glands of partially engorged females of the Cayenne tick Amblyomma cajennense. The results show that this organelle exhibits completely disarrayed crests due to the presence of lipidic material inside the matrix and between the crests, thus demonstrating their participation in the production of the lipids that would be used structurally by the cells. These organelles with ultrastructural changes were denominated derived mitochondria.  相似文献

To clarify 24-hr variations in rat thyroid follicular cells under physiological conditions, their subcellular structures were examined at six evenly spaced times during 24 hr by using a morphometric technique. The volume, surface, and numerical densities of subcellular structures varied distinctly over each 24-hr period, with a bimodal pattern. The cellular and nuclear volumes varied also bimodally over 24 hr. A decrease in the surface density of the apical plasmalemma at 1200 and 0000 hr coincided with an increase in volume density of cytoplasmic granules representing colloid droplets and dense bodies. Most granules (colloid droplets) appearing at these times were reduced in electron density. At other times, especially at 1600 and 0400 hr, morphometric parameters of rough endoplasmic reticulum (rER), Golgi complex, and subapical vesicles were prominently increased, although values for rER did not peak at 1600 hr. At these times, the volume densities of cytoplasmic granules, most of which were heterogeneous and of homogeneous electron density, were decreased. These findings coincided with immediate and subsequent reactions of follicular cells after injection of thyroid-stimulating hormone (TSH). From the evidence, it seems likely that variations in follicular cells over a 24-hr period reflect variations in blood TSH concentration. The total membrane areas of membrane components in follicular cells were calculated from the morphometric measurements. These areas fluctuated unimodally during 24 hr over a 65% range. This suggests that the membranes in follicular cells are subjected to cyclic degradation and regeneration during each 24-hr period.  相似文献

Summary The epithelium of the bovine oviduct was studied with respect to its ultrastructural organization and histochemical features.The ciliated cells seem to undergo no changes during the sexual cycle. The nucleus is regular, the mitochondria are small and frequent. Particles resembling lysosomes are present. Except for the free surface, containing alkaline phosphatase, these cells do not present any particular histochemical characteristics.The secretory cells contain more ribonucleic acid than the ciliated cells, and the endoplasmic reticulum is better developed. This also shows changes correlated with the sexual phases, thus the cisternae are much larger during the follicular phase than during the luteal phase. The nucleus is deeply fissured, the cisternae of the ergastoplasm dipping into these fissures. The mitochondria are larger and less frequent than in the ciliated cells. Secretion granules are frequently present. They are basophilic and periodic acid-Schiff reactive. They seem not to contain glycogen. The granules undergo characteristic changes and are subsequently discharged. This process is not paralleled by any changes in the histochemical reactions studied.Tests for the demonstration of lipids, non-specific esterase, and acid phosphatase did not give information that could be correlated to the ultrastructural organization.This investigation was supported by a grant from the foundation Therése och Johan Anderssons Minne  相似文献

MicroRNAs (miRNAs) regulate the expression of many mammalian genes and play key roles in embryonic hair follicle development; however, little is known of their functions in postnatal hair growth. We compared the effects of deleting the essential miRNA biogenesis enzymes Drosha and Dicer in mouse skin epithelial cells at successive postnatal time points. Deletion of either Drosha or Dicer during an established growth phase (anagen) caused failure of hair follicles to enter a normal catagen regression phase, eventual follicular degradation and stem cell loss. Deletion of Drosha or Dicer in resting phase follicles did not affect follicular structure or epithelial stem cell maintenance, and stimulation of anagen by hair plucking caused follicular proliferation and formation of a primitive transient amplifying matrix population. However, mutant matrix cells exhibited apoptosis and DNA damage and hair follicles rapidly degraded. Hair follicle defects at early time points post-deletion occurred in the absence of inflammation, but a dermal inflammatory response and hyperproliferation of interfollicular epidermis accompanied subsequent hair follicle degradation. These data reveal multiple functions for Drosha and Dicer in suppressing DNA damage in rapidly proliferating follicular matrix cells, facilitating catagen and maintaining follicular structures and their associated stem cells. Although Drosha and Dicer each possess independent non-miRNA-related functions, the similarity in phenotypes of the inducible epidermal Drosha and Dicer mutants indicates that these defects result primarily from failure of miRNA processing. Consistent with this, Dicer deletion resulted in the upregulation of multiple direct targets of the highly expressed epithelial miRNA miR-205.  相似文献

Some mitochondria in leukemic cells in two of nine investigated patients with chronic lymphocytic leukemia and in one patient with morphologically less usual form of this disease contained unusual rodlet-like inclusion bodies. These structures were always present in the matrix of frequently dilatated intercristal spaces. The width of rodlet-like inclusions ranged between 10 and 20 nm, their maximal length was 114 nm. All observed rodlet-like inclusion bodies appeared to be composed of fine filaments 2...3 nm in width and resembled the DNA containing structures in mitochondria of few other cell types. In mitochondria of leukemic cells, however, these inclusions represent a further structural abnormality.  相似文献

The zona pellucida is the unique extracellular glycoprotein matrix which is assembled during growth of the mammalian oocyte. The present studies were carried out to examine the formation of this structure in relation to the differentiation of ovarian cell types during follicular development. Specific antibodies were developed against total rabbit ZP proteins as well as against ZP proteins electrophoretically purified by high-resolution two-dimensional polyacrylamide electrophoresis gels (2D-PAGE). Antibodies were characterized by (a) immunoelectrophoresis, (b) a Staphylococcus aureus protein A binding assay, and (c) immunoblotting following 2D-PAGE separation of ZP proteins. Immunoperoxidase localization with these antibodies was used to determine the stage of ovarian follicular development at which ZP antigens first appear as well as to evaluate the cellular and extracellular distribution of these proteins throughout folliculogenesis. The ZP proteins were first observed in the cytoplasm and at the periphery of the oocytes surrounded by a thin squamous follicular cell layer. No staining was observed in the cytoplasm of follicle cells during early folliculogenesis. As the ZP matrix was assembled extracellularly, the intensity of staining of the outer and inner regions could be distinguished. This differentiation of the matrix coincided with the differentiation of the follicular cells into a multilayer cell complex. At this stage, specific ZP proteins are localized within the cytoplasm of the inner layers of these follicular cells. The staining is then diminished in cells of preantral follicles. These studies demonstrate that the formation of the ZP is an excellent model system to study the early stages of follicular development and cell differentiation.  相似文献

Ameloblasts from different regions of upper incisors of rats were examined with the electron microscope. During matrix formation, the cells resemble secretory cells. They are extremely long, tightly packed, and show considerable polarity. Nuclei are at the basal end of the cell. Mitochondria are proximal and the Golgi apparatus distal to the nucleus. Ergastoplasm is found in all levels but mainly in the distal end. A terminal bar apparatus separates the distal end of the cell from Tomes's process. Next to this is soft enamel. The next incisal region is a transitional zone in which the ameloblasts separate easily from the enamel. Endoplasmic reticulum is dilated and very obviously in communication with the perinuclear space. Mitochondria are present not only proximal, but also distal, to the nucleus. The next incisal zone consists of cells related to the maturation of enamel. They no longer resemble secretory cells, but now have more characteristics of transporting cells. Processes from the distal end of the cell are present with mitochondria closely applied to the base of the processes. A considerable amount of intercellular space exists with microvilli projecting into the space. Iron granules appear in these cells, and the ergastoplasmic cisternae are dilated. In the incisal end of this zone, the iron granules form aggregates. The iron finally leaves the cells to enter the enamel. Free RNP particles and fibrils become more evident after the iron leaves the cells. In the most incisal region, the ameloblasts are further reduced in height. Distal processes are no longer present and fibrils are more conspicuous.  相似文献

The following stereological parameters of mitochondria were calculated in rat germ cells during the spermatogenesis: volume density of matrix, outer compartment, outer membrane and inner membrane, surface density of outer membrane and inner membrane. They were the basis to calculate the partition coefficient of matrix and partition coefficient of outer compartment. The matrix volume demonstrated a decreasing in mitochondria of germ cells during spermatogenesis. The relative volume of outer compartment was calculated with the intracristal spaces and revealed increasing from spermatogonia to spermatids. The partition coefficient for the matrix significantly decreased. Our observations suggest that transformation of mitochondrial configuration during spermatogenesis and spermiogenesis is the expression of intensive metabolic processes and activity of membrane transport in germ cells.  相似文献

The structure of the kidney of the Swiss albino mouse changes progressively during the first 2 weeks after birth. Cells proliferate to form new nephrons, cells differentiate by acquiring specialized membranous components, and certain cytological features which are present at birth diminish in abundance or disappear. The differentiation of the cells of the cortical tubules has been studied using the light and electron microscopes. The tubules are partially and variably differentiated at birth. During the first 2 weeks after birth the brush border develops in the proximal tubules by the accumulation of numerous microvilli on the apical cell margins. Basal striations develop in proximal and distal tubules as an alignment of mitochondria, the result of what appears to be progressive interlocking of adjacent fluted cells. The mitochondria increase in number and size, accumulate homogeneous matrix, and acquire small, very dense granules. The collecting ducts develop tight pleating of the basal cell membranes, and dark cells containing numerous small cytoplasmic vesicles and microvilli appear. At birth there are dense irregular cytoplasmic inclusions presumed to be lipide in renal cells, the cytoplasmic granules of Palade are abundant, and there are large round bodies in the cells of the proximal tubules. The lipide inclusions disappear a few days after birth, and the cytoplasmic granules of Palade diminish in abundance as the cells differentiate. The large round bodies in the proximal tubules consist of an amorphous material and contain concentrically lamellar structures and mitochondria. They resemble the cytoplasmic droplets produced in the proximal tubules of adult rats and mice by the administration of proteins. The large round bodies disappear from the proximal tubules of infant mice during the first week after birth, but the concentric lamellar structures may be found in adult mice.  相似文献

This paper reports on the fine structure of rat oocytes at stages before ovulation, during maturation, fertilization, and early cleavage. The study includes parallel observations on light and electron microscope preparations with attempted correlations. The follicular cells of the ovarian egg are described as sending long processes through the zona pellucida to the egg surface where they mingle with thin projections from the egg itself. No open communication between follicle cell cytoplasm and egg cytoplasm was observed. During maturation and fertilization both types of processes are withdrawn from the zona. The germinal vesicle and later the pronuclei of the fertilized egg are characterized by numerous large nucleoli. These have the form of thick walled vesicles with diameters as great as 8 to 10 µ. The wall is dense in the EM image and appears to consist in part of small granules. The cytoplasm shows several inclusions including mitochondria of usual form and a Golgi component which has the typical fine structure and the distribution described by earlier light studies. Small dense particles, presumably RNP particles, are distributed throughout the cytoplasmic matrix and show no preference for membranes. The endoplasmic reticulum of the oocyte is represented by a scattering only of vesicles, but begins a more extensive and elaborate development with the onset of segmentation. One inclusion of the ooplasm, similar in size to mitochondria, receives special attention. It is a vesicular structure, containing a large number of small vesicles (10 to 50 mµ in diameter) and frequently a central density or nucleoid. They are referred to as multivesicular bodies. Such bodies are found in small number in the ovarian egg, but increase greatly in number during maturation and fertilization. It appears from the micrographs of eggs in these latter stages that these vesicular bodies break down and liberate their content of small vesicles to the surrounding ooplasm. Inconsistent with the view that hair follicle stem cells reside in the matrix area of the hair bulb, we found that label-retaining cells exist exclusively in the bulge area of the mouse hair follicle. The bulge consists of a subpopulation of outer root sheath cells located in the midportion of the follicle at the arrector pili muscle attachment site. Keratinocytes in the bulge area are relatively undifferentiated ultrastructurally. They are normally slow cycling, but can be stimulated to proliferate transiently by TPA. Located in a well-protected and nourished environment, these cells mark the lower end of the "permanent" portion of the follicle. Our findings, plus a reevaluation of the literature, suggest that follicular stem cells reside in the bulge region, instead of the lower bulb. This new view provides insights into hair cycle control and the possible involvement of hair follicle stem cells in skin carcinogenesis.  相似文献
