Synopsis Acid phosphatase activity is demonstrated employingp-nitrophenyl phosphate as substrate and lead acetate as coupler. The fine structural localization of the enzyme in starved planarian tissues is described. The method is used to pin-point starvation-induced acid phosphatase activity in relation to autophagy and crinophagy in the gland cells; autophagy, autolysis and cell death in parenchymal and gastrodermal cells and basement membrane lysis. Attention is also payed to the demonstration of muscle lysis. The histochemical implications of the method are discussed.  相似文献

A combination of microscopical, cytochemical, and biochemical techniques have been employed to study the changes occurring during the first seven days of blastema formation and regeneration after decapitation in adult Polycelis tenuis worms. Fine structural data reveal evidence of cell fragmentation, selective cell deletion, and phagocytosis at and below the wound surface. Initially, (0–12 h regeneration) cell debris is phagocytosed by intact parenchymal and gastrodermal cells near the cut surface which is later sealed (24 h) by a stretching of marginal epidermal cells. Wound sealing is followed by a migration of newly differentiated rhabdite cells into the epithelium. Morphological evidence of a selective cell autolysis precedes evidence of an accumuluation of lipid and glycogen reserves in the parenchymal and gastrodermal cells and the later (48 h regeneration time) aggregation of undifferentiated mitotically active neoblasts beneath the wound.
Biochemical data reveal an early period of high acid phosphatase (p-nitrophenyl phosphatase and sodium-β-glycerophosphatase) activity 3–12 h after injury, followed by a further intense period of activity at 44–48 h after decapitation. The coincident cytochemical data show an increased level of acid phosphatase activity associated with cell lysis and death in the wound and blastema zone and also with the digestion of phagocytosed cell debris.  相似文献

A chromic acid oxidation-silver technique was used to localize polysaccharide material in Polycelis tenuis at the electron microscope level. In the epithelium, staining was observed within apical vacuoles and on the free surfaces of the cells. A similar staining was observed in relation to the glycocalyx of the pharyngeal epithelia and that of the flame cells. Silver was deposited in the basement membrane. In the parenchyma, the major components giving a positive reaction were the cyanophil and mucous gland cells. Particularly strong silver staining (confirmed by X-ray microanalysis) was observed in the granules and Golgi apparatus of the cyanophil cells. IDPase activity was also found in relation to the Golgi apparatus and its secretory products. The overall distribution of mucopolysaccharide material was confirmed with the PAS and Alcian blue techniques. The fine structural localization of the Alcian blue was also determined using electron microscopy and X-ray microanalysis.  相似文献

Cell death and tissue remodeling in planarian regeneration   总被引:1,自引:0,他引:1  
Many long-lived organisms, including humans, can regenerate some adult tissues lost to physical injury or disease. Much of the previous research on mechanisms of regeneration has focused on adult stem cells, which give rise to new tissue necessary for the replacement of missing body parts. Here we report that apoptosis of differentiated cells complements stem cell division during regeneration in the planarian Schmidtea mediterranea. Specifically, we developed a whole-mount TUNEL assay that allowed us to document two dramatic increases in the rate of apoptosis following amputation—an initial localized response near the wound site and a subsequent systemic response that varies in magnitude depending on the type of fragment examined. The latter cell death response can be induced in uninjured organs, occurs in the absence of planarian stem cells, and can also be triggered by prolonged starvation. Taken together, our results implicate apoptosis in the restoration of proper anatomical scale and proportion through remodeling of existing tissues. We also report results from initial mechanistic studies of apoptosis in planarians, which revealed that a S. mediterranea homolog of the antiapoptotic gene BCL2 is required for cell survival in adult animals. We propose that apoptosis is a central mechanism working in concert with stem cell division to restore anatomical form and function during metazoan regeneration.  相似文献

Cell wall degradation in the autolysis of filamentous fungi   总被引:1,自引:0,他引:1  
A systematic study on autolysis of the cell walls of fungi has been made on Neurospora crassa, Botrytis cinerea, Polystictus versicolor, Aspergillus nidulans, Schizophyllum commune, Aspergillus niger, and Mucor mucedo. During autolysis each fungus produces the necessary lytic enzymes for its autodegradation. From autolyzed cultures of each fungus enzymatic precipitates were obtained. The degree of lysis of the cell walls, obtained from non-autolyzed mycelia, was studied by incubating these cell walls with and without a supply of their own lytic enzymes. The degree of lysis increased with the incubation time and generally was higher with a supply of lytic enzymes.Cell walls from mycelia of different ages were obtained. A higher degree of lysis was always found, in young cell walls than in older cell walls, when exogenous lytic enzymes were present.In all the fungi studied, there is lysis of the cell walls during autolysis. This is confirmed by the change of the cell wall structure as well as by the degree of lysis reached by the cell wall and the release of substances, principally glucose and N-acetylglucosamine in the medium.  相似文献

Regenerating planarians of the species Polycelis tenuis have been studied with respect to the two distinct phases which occur during regeneration, viz., the first 24 h involving cellular activation, and the following days when differentiation of the blastema occurs after a period of cellular proliferation. We have studied particularly the biochemical events that control regeneration with respect to the nature of the signals which induce cellular activation, the membrane receptors of these signals, and the consequences of these signals for the cellular metabolism of DNA, RNA and proteins. The roles of neurohormones such as serotonin, dopamine, and noradrenaline are assessed, and a provisional model of the process of cellular activation is proposed which takes account of all the information that is now available concerning planarian regeneration.  相似文献

Log phase cells of Micrococcus lysodeikticus (luteus) IFO 3333 autolyzed when incubated at 37 C in 0.01 M sodium-phosphate buffer pH 7.5. The enzyme involved in the autolysis was recovered mainly in an aqueous phase from cytoplasmic membranes and cytoplasmic materials treated with n-butanol, and proved to be an N-acetylmuramyl-L-alanine amidase. The autolysis of log phase cells suspended in autolyzing buffer was depressed by the addition of trypsin to the buffer.  相似文献

1 The zoophytophagy of Nesidiocoris tenuis (Reuter) (Heteroptera: Miridae) was characterized in relation to prey availability and environmental factors by: (i) monitoring its population dynamics in tomato greenhouses; (ii) analysis of the influence of N. tenuis and whitefly density, temperature and humidity on the intensity of N. tenuis plant feeding; and (iii) laboratory assays under controlled conditions to determine the intensity of plant feeding in relation to prey availability, temperature and humidity. 2 A negative relationship was found between plant feeding and predated whiteflies in tomato greenhouses. Plant feeding was directly related to N. tenuis density and temperature and inversely related to whitefly density. The significance of prey availability and temperature was corroborated in laboratory assays. The intensification of plant feeding at low prey density indicates switching from zoophagy to phytophagy as prey become scarce. 3 Nesidiocoris tenuis showed a typical predator dynamic in relation to variance in prey density. Populations increased after whitefly outbreaks and decreased after whitefly had been depleted. The rapid decrease of N. tenuis populations after whitefly decreased, however, suggests that plants are a poorer nutrient source than whitefly for this species.  相似文献
