The genus Dasypyrum contains two species: the annual and widespread D. villosum (2x = 2n = 14) and the perennial and generally rare D. breviaristatum (2x = 2n = 14 and 4x = 2n = 28). The origin of the latter and its genome constitution have been subject of several studies. There is agreement that the genome of the diploid D. villosum (VV) is different from the diploid cytotype of D. breviaristatum (VbVb), but there is no agreement of the constitution of the tetraploid cytotype, specifically whether is it an autotetraploid or an allotetraploid. This is a long-standing disagreement that this study aims to resolve using the 5S nrDNA as a genomic marker. Our studies suggest that the 4x D. breviaristatum is an allotetraploid (VVVbVb).  相似文献

The present work includes detailed male meiotic studies on 46 species of grasses falling into 59 accessions from different localities of Parvati Valley in Kullu district of Himachal Pradesh in the altitudinal range of 1,100 to 2,750 m. All the species have been studied cytologically for the first time from the study area. The meiotic chromosome count of n = 14 for Calamagrostis emodensis is the first ever chromosome report. Three species, namely Agrostis alba (n = 21), Avena byzantina (n = 21) and Bromus inermis (n = 14) have been studied cytologically for the first time from India. New intraspecific diploid/polyploid cytotypes have been reported for Arthraxon serrulatus (2n = 4x = 32), Iseilema laxum (2n = 12x = 60), Digitaria albudens (2n = 8x = 72), Festuca kashmiriana (2n = 2x = 14) and Stipa orientalis (2n = 2x = 20). The existence of variable number of B-chromosomes (2n = 60 + 0-5B) has been reported for the first time in the 12x cytotype of Iseilema laxum. Secondary associations of chromosomes in the tetraploid cytotype of Cymbopogon martini (n = 20) indicated its secondary polyploid nature. As many as 18 species showed various meiotic anomalies such as the phenomenon of cytomixis involving inter PMC migration of chromatin material, chromatin stickiness, interbivalent connections, abnormal spindle activity, presence of bridges and laggards during anaphases and telophases and abnormal sporads. These meiotic abnormalities consequently yielded sterile and heterogeneous-sized fertile pollen grains. The polyploidy and aneuploidy have played an active role in the evolution of grasses.  相似文献

Genus Helianthus comprises diploid and polyploid species. An autoallopolyploid origin has been proposed for hexaploid species but the genomic relationships remain unclear. Mitotic and meiotic studies in annual Helianthus annuus (2n = 2x = 34) and perennial Helianthus resinosus (2n = 6x = 102) as well as the F1 hybrids between both species were carried out. Chromosome counting confirmed the hybrid origin of the latter plants and their tetraploid condition. Bivalents in hybrids ranged from 12 to 28 ( $ \bar{x} $  = 20.8). Univalents, trivalents and quadrivalents were also observed. Meiotic products comprised dyads, triads and normal tetrads and pollen grains were heterogeneous in size. These observations suggest the occurrence of 2n pollen in addition to the expected n. Genomic in situ hybridization (GISH) of total H. annuus DNA on H. resinosus chromosomes rendered weak but uniform signals; similar hybridization pattern was observed using three other annual species. Hybridization with H. annuus probe performed on root tip cells of F1 H. annuus × H. resinosus hybrids revealed 17 chromosomes with a strong hybridization signal. GISH in hybrid meiocytes distinguished chromosomes from parental species and revealed autosyndetic pairing of H. resinosus chromosomes, allosyndetic pairing in bivalents, trivalents and quadrivalents, and the presence of univalents derived from parents, H. annuus and H. resinosus. Results obtained from classical and molecular cytogenetics do not support H. annuus as a direct ancestor of H. resinosus. The occurrence of allosyndetic pairing and the relatively high fertility of the F1 hybrids point to the possibility that useful genes could be transferred from H. resinosus to cultivate sunflower, although the effective rate of recombination has not been evaluated. GISH method proved effective to recognize parental chromosomes in H. annuus × H. resinosus progeny.  相似文献

Silene vulgaris is an important ethnobotanical species, but so far no thorough meiotic analysis has been performed, despite its wide occurrence in temperate and alpine Himalayas, this prompted us to undertake present study. We investigated original meiotic chromosome number and effect of cytomixis on meiotic course, microsporogenesis and pollen grain size and fertility. The paper also summarizes the previously published chromosome counts in the species and cytogeography of 2x and 4x cytotypes in the Indian Himalayas and elsewhere. Meiocytes preparations were made through standard squashed technique in 1 % acetocarmine. Two ploidy levels have been determined during the cytomorphological surveys, diploid (2n = 2x = 24) and tetraploid (2n = 4x = 48). Most of the presently scored populations were found to be diploid (2n = 2x = 24). Morphologically 2x and 4x individuals were indistinguishable from each other. Meiosis was perfectly normal in 4x cytotype. However, some of the 2x plants showed the phenomenon of cytomixis and associated meiotic abnormalities. Consequent to cytomixis, hypo-, hyperploid and enucleated pollen mother cells (PMCs) were resulted. Such PMCs combined with other meiotic abnormalities yielded sterile and variable sized pollen grains. On the basis of overall information, it is clear that 2x cytotype is the most widely distributed cytotype in the Indian Himalayas as well as elsewhere. On the other hand, the 4x cytotype seems to be less frequently present in the Indian Himalayas and outside of India.  相似文献

Cytological studies were carried out on two populations of Trigonobalanus verticillata in Yinggeling, Hainan Province, China (YGL), and Fraser’s Hill, Malaysia (FH). In the two populations, the pattern of interphase nuclei was of the simple chromocentre type, the mitotic prophases were of the proximal interstitial type and chromosome numbers were 2n = 2x = 14 (YGL) and 2n = 6x = 42 (FH), representing diploid and hexaploid respectively. The basic chromosome number of the species was x = 7; this is a unique number in the Fagaceae and in the Fagales as well. The characteristic karyotype of the diploid population in China was 2n = 14 = 8m + 4sm + 2st. Based on the new finding of the chromosome number in this study, we clarify the previous dispute about the basic chromosome number and propose that the two Asian Trigonobalanus species have closer affinity.  相似文献

Chromosome number variations play an important role in the genus Medicago. In addition to polyploidy there are cases of dysploidy as evidenced by two basic numbers, x = 8 and x = 7, the latter limited to five annual species having 2n = 14. Annuals are diploid with the exception of Medicago scutellata and Medicago rugosa which have 2n = 30 and are considered the result of crosses between the 2n = 16 and 2n = 14 species. However, this hypothesis has never been tested. This study was carried out to investigate the 2n = 14 and 2n = 30 karyotypes and verify the allopolyploid origin of M. scutellata and M. rugosa. Fluorescence in situ hybridization (FISH) of rDNA probes and genomic in situ hybridization (GISH) were performed. FISH showed that all five diploids with 2n = 14 have one pair of 45S and one pair of 5S rDNA sites. M. scutellata displayed four sites of 45S and four sites of 5S rDNA, while in M. rugosa only one pair of each of these sites was found. GISH did not produce signals useful to identify the presumed progenitors with 14 chromosomes. This result suggests alternative evolutionary pathways, such as the formation of tetraploids (2n = 32) and subsequent dysploidy events leading to the chromosome number reduction.  相似文献

Abstract

Solanum × michoacanum (Bitter.) Rydb. (mch) is a wild diploid (2n = 2x = 24) potato species derived from spontaneous cross of S. bulbocastanum and S. pinnatisectum. This hybrid is a 1 EBN (endosperm balance number) species and can cross effectively only with other 1 EBN species. Plants of mch are resistant to Phytophthora infestans (Mont) de Bary. To introgress late blight resistance genes from mch into S. tuberosum (tbr), genepool somatic hybridization between mch and susceptible diploid potato clones (2n = 2x = 24) or potato cultivar Rywal (2n = 4x = 48) was performed. In total 18,775 calli were obtained from postfusion products from which 1,482 formed shoots. The Simple Sequence Repeat (SSR), Cleaved Amplified Polymorphic Sequences (CAPS) and Random Amplified Polymorphic DNA (RAPD) analyses confirmed hybrid nature of 228 plants and 116 autofused 4x mch. After evaluation of morphological features, flowering, pollen stainability, tuberization and ploidy level, 118 somatic hybrids and 116 autofused 4x mch were tested for late blight resistance using the detached leaf assay. After two seasons of testing three somatic hybrids and 109 4x mch were resistant. Resistant forms have adequate pollen stainability for use in crossing programme and are a promising material useful for introgression resistance from mch into the cultivated potato background.  相似文献

The Solanum brevicaule complex contains about 20 species of diploids (2n = 2x = 24), tetraploids (2n = 4x = 48) and hexaploids (2n = 6x = 72), distributed from central Peru south to northwestern Argentina. The complex is defined entirely by morphological similarity of its constituent members, which are very similar to each other and to some landraces of the cultivated potato, Solanum tuberosum. Conflicting taxonomic treatments are common among authors. Species boundaries within the complex have been studied with morphological phenetics from germplasm accessions planted in a field plot in the north central US, and with molecular marker data from RAPDs, low-copy nuclear RFLPs, and AFLPs. The present study compares these results with an additional replicated morphological study of the same germplasm accessions in a greenhouse environment in the high Andes of central Peru. The results support extensive reduction of species in the complex.  相似文献

In Myrtaceae, reports regarding the nuclear DNA content are scarce. The aim of this study is to present genome size data for fleshy-fruited Myrteae, and to test their relation with chromosome number and ploidy, the available data for cytoevolutionary studies in Myrtaceae. Thirty species out of ten genera were investigated for chromosome number and genome size using flow cytometry. Twenty-eight species were diploid with 2n = 2x = 22 and two species were tetraploid with 2n = 4x = 44. All genome sizes measured are new. Among the diploid species, a gradual and small variation in 2C-values (0.486 pg in Gomidesia schaueriana to 0.636 pg in Eugenia multicostata) was observed, whereas the tetraploid genomes of Psidium acutangulum and P. cattleianum had about twice as much DNA (1.053 and 1.167 pg, respectively). The total interspecific variation of C-values was 2.45-fold. The fleshy-fruited Myrteae have smaller holoploid genomes than the capsular-fruited Eucalypteae and Melaleuceae.  相似文献

During the present study, 360 populations of 174 species belonging to 82 genera and 23 families of dicots have been cytologically worked out from different localities of District Kangra (Himachal Pradesh) between 550 and 3,800 m altitude. As many as 10 species including Berberis ceratophylla (2n = 28), Caltha alba (2n = 32), Corydalis meifolia (2n = 16), C. thyrsiflora (2n = 16), Impatiens reidii (2n = 14), Indigofera hamiltonii (2n = 16), Potentilla thomsonii (2n = 14), Sedum trifidum (2n = 36), Stellaria semivestita (2n = 26) and Viola canescens (2n = 12) have been cytologically worked out for the first time at world level. New intraspecific diploid cytotypes have been recorded in 11 species while new intraspecific tetraploid cytotypes are noticed in 15 species. B-chromosomes have also been reported in 3 species viz. Anemone obtusiloba, Clematis grata and Ranunculus diffusus for the first time at world level. The species of the area are active state of evolution, depicting heterogeneity in chromosome numbers involving polyploidy (48 species, 28.18 %) as well as irregular meiotic behavior. The meiotic abnormalities have been witnessed in 69 species (39.65 %) showing different types of irregularities such as cytomixis, interbivalant connections, unoriented bivalents, chromatin stickiness, chromosomal laggards and bridges as well as abnormal microsprogenesis. All these meiotic abnormalities lead to reduced pollen fertility and formation of variable sized pollen grains in most of these species.  相似文献

Eighteen species belonging to family Convolvulaceae occurring in northeastern Brazil were analyzed to identify the karyotypic variability among rupicolous and non-rupicolous species. The count of 2n = 56 for Evolvulus sp., is the first report of polyploidy for the genus. The similarities among the karyotypes of species of Ipomoea, Merremia, and Operculina suggest that x = 15 is the basic secondary number for these genera, while x = 13 has been observed in Evolvulus. The genus Jacquemontia, with 2n = 18 is karyologically distinct from the other genera of the family. Intraspecific numerical chromosomal variations were not detected, nor were any karyological variations related to the rocky habitat characteristic of inselbergs. The occurrence of other plant groups with polyploid forms linked to this type of habitat suggests that the family Convolvulaceae is karyologically stable. We discuss variations in chromosome numbers, base numbers, and their implications in the evolution of the principal groups within the Convolvulaceae.  相似文献

Three endemic Centaurea species from North Africa are investigated for the first time by chromomycin fluorochrome banding for GC-rich DNA distribution, fluorescence in situ hybridization for physical mapping of rRNA genes, and flow cytometry for genome-size assessment. Investigated species belong to three different sections and possess three basic chromosome numbers: C. tougourensis subsp. tougourensis 2n = 4x = 36 (x = 9), C. musimonum 2n = 2x = 20 (x = 10), and C. maroccana 2n = 2x = 24 (x = 12). The number and distribution of chromomycin positive bands (CMA+) and 18S-5.8S-26S (35S) rDNA loci were different among investigated species and ranged from 6 to 80 chromomycin bands and from 2 to 6 35S rDNA loci. The three species have just one 5S rDNA locus at intercalary position on a separate chromosome pairs, except in the case of C. musimonum in which both rDNA loci were localized on the same chromosome. All rDNA loci were co-localized with CMA+ bands, except three 35S in C. musimonum. Genome size ranged from 2C = 1.66 to 2C = 2.86 pg in diploid species (C. musimonum and C. maroccana, respectively) and to 2C = 4.51 pg in tetraploid C. tougourensis subsp. tougourensis.  相似文献

The only Balkan endemic of the genus Picris, Picris hispidissima, was studied in detail using morphological and karyological methods. The species was shown to be morphologically distinct from the closest taxon, Picris hieracioides, by the pectinate–ciliate indumentum of involucral bracts, dilatation of the peduncle, length of the outer and inner bracts, and indumentum colour. No morphological variation that would require taxonomic classification was found within this species. Despite the diploid chromosome number (2n = 2x = 10) being confirmed for P. hispidissima, variation of up to 9.5% in genome size was found. The likely explanation for this variation is hybridization and introgression with closely related P. hieracioides. The most convincing evidence for this hypothesis is the detection of plants with two clearly different DNA contents arising from a single capitulum found in the location where P. hispidissima and P. hieracioides co-occur.  相似文献

Detailed male meiosis, critical morphological observations and distribution pattern of diploid as well as tetraploid cytotypes of the Western Himalayan species, Bupleurum lanceolatum have been evaluated at present. A diploid (n = 8) cytotype is reported from Kashmir, whereas, both diploid (n = 8) and tetraploid (n = 16) cytotypes are available from two districts Kangra and Sirmaur of Himachal Pradesh. Out of these, the tetraploid cytotype makes new addition for the species on a worldwide basis. As per behavior, a tetraploid cytotype is characterized by abnormal meiosis leading to high pollen sterility and size variation of the pollen grains. Morphologically, tetraploids are noted to be luxuriant in comparison to the diploids.  相似文献

Fish of the genus Selene, known as lookdowns or moonfish, are one of the most morphologically derived groups of the family Carangidae, whose phylogenetic relationships are still largely unknown. In this study, we discuss karyoevolutionary aspects of three representatives of this genus from the Western Atlantic: Selene brownii (2n = 48; FN = 48), Selene setapinnis (2n = 46; FN = 48), and Selene vomer (2n = 48; FN = 50). Their body patterns were also investigated and compared to one another and in relation to two other species of different genera. Two mechanisms of karyotypic evolution seem to have acted in the diversification of this genus, namely pericentric inversions and centric fusions. Mapping of rDNA sequences showed that chromosome pairs bearing 5S rDNA sites are similar, whereas those bearing 18 rDNA sites are morphologically distinct while apparently also exhibiting interspecies synteny. Although the nucleolar organizer-bearing chromosomes are extremely efficient cytotaxonomic markers among Selene species, others cytogenetic patterns of these species are relatively conserved. Hybridization with telomeric probes (TTAGGG)n did not exhibit interstitial telomeric sites (ITS), especially in S. setapinnis, where, along with a reduction in diploid number, a large metacentric pair derived from centric fusion is present. Data obtained by geometric morphometrics enable a clear morphological distinction among the three species, as well as in relation to two other species of the genus Caranx and Oligoplites. Data obtained suggest that morphologic evolution in Selene species was primarily dissociated from visible changes that occurred at the chromosomal level.  相似文献

Abstract

Cultivated potato (Solanum tuberosum L.) lacks resistance to bacterial wilt caused by Ralstonia solanacearum. Interspecific symmetric protoplast fusion was conducted to transfer bacterial wilt resistance from eggplant (S. melongena, 2n = 2x = 24) into dihaploid potato (2n = 2x = 24). In total, 34 somatic hybrids were obtained, and of these, 11 rooted and were tested for genome components and resistance to race 1 of R. solanacearum. The hybrids exhibited multiple ploidy levels and contained the dominant nuclear genome from the potato parent. Three types of nuclear genomes were identified in the hybrids through genomic in situ hybridization (GISH) and simple sequence repeat (SSR) analysis, including (1) the potato type of the tetraploids in which eggplant chromosomes could not be detected by GISH but their nuclear DNA was confirmed by SSR, (2) the biased type of the hexaploids in which the chromosome dosage was 2 potato:1 eggplant, and (3) the chromosome translocation type of the mixoploids and aneuploids that was characterized by various rates of translocations of nonhomologous chromosomes. Cytoplasmic genome analysis revealed that mitochondrial DNA of both parents coexisted and/or recombined in most of the hybrids. However, only potato chloroplast DNA was retained in the hybrids speculating a compatibility between cpDNA and nuclear genome of the cell. The pathogen inoculation assay suggested a successful transfer of bacterial wilt resistance from eggplant to the hybrids that provides potential resistance for potato breeding against bacterial wilt. The genome components characterized in present research may explain partially the inheritance behavior of the hybrids which is informative for potato improvement.  相似文献

Chromosome data are important for elucidating intergeneric relationships and delimiting infrafamilial tribes in plants. This paper reports karyological data for 12 Seriphidium (Bess.) Poljak. species from China, of these, five count as new reports and the others have been reported elsewhere. We also report the tetraploid level in S. schrenkianum for the first time. The karyotype formulae and types for the five new reports are: S. schrenkianum (Ledeb.) Poljak. 2n = 4x = 36 = 22m + 12sm + 2st, S. sublessingianum (Kell.) Poljak. 2n = 2x = 18 = 14m + 4sm and S. transiliense (Poljak.) Poljak. 2n = 2x = 18 = 4M + 6m + 2m(SAT) + 4sm + 2sm(SAT), belong to 2A type; S. cinum (Berg. ex Poljak.) Poljak. 2n = 2x = 18 = 8m + 10sm and S. sawanense Y. R. Ling et C. J. Humphries 2n = 2x = 18 = 10m + 2m(SAT) + 6sm, belong to 2B type. Furthermore, we estimated karyotype asymmetry in the 12 species by using an intrachromosomal asymmetry index and an interchromosomal asymmetry index. The data increase information about the karyosystematics of Seriphidium.  相似文献

Abstract

In the present study 16 populations belonging to 13 species of the genus Vernonia Schreb. were examined cytologically. In total, six different chromosome numbers, which represent three basic numbers: x = 10, x = 16 and x = 17, were found. These results include the first chromosome number reports for the following four species: V. lanifera Cristóbal & Dematt. (2n = 2x = 32), V. membranacea Gardner (2n = 2x = 34), V. salzmannii DC. (2n = 2x = 20) and V. scabrifoliata Hieron. (2n = 2x = 128). Besides, a new chromosome number was found in V. saltensis Hieron. (2n = 2x = 32), for which only tetraploid populations have been previously recorded. The data obtained in this work, along with the information available from the literature, show that the genus Vernonia in South America is heterogeneous with basic chromosome numbers that range between x = 9 and x = 19. These numbers suggest that a combination of polyploidy and aneuploidy has played an important role in the evolution of the genus.  相似文献

Our study (survey, atlas of 136 microphotographs and 67 drawings) points out the actual chromosome numbers of 82 taxa of the genus Pinguicula L. They were gathered from literature and critically examined. In addition, numerous counts are published for the first time. They represent about 80% of all the taxa known. The basic chromosome numbers are x = 6, 8, 9, 11, and 14; the ploidy levels are 2n (diploid), 4n (tetraploid), 8n (octoploid) and 16n (hexadecaploid). The basic number x = 6 is a one-off, x = 8 and 11 are the most frequent in the genus; x = 14 indicates a hybridogenous differentiation process in the past. The caryological differentiation—chromosome numbers and ploidy level—is discussed with regard to distribution pattern, growth type, and infrageneric classification (at the level of sections).  相似文献
