In view of the need to exploit saline water resources in agriculture in arid zones, we investigated the salt tolerance of Opuntia ficus-indica in plants growing in solution culture. Salt (NaCl) was added in concentrations ranging from 5 (control) to 200 mol m-3. Cladode growth was sensitive to salinity, being 60% of the control at 50 mol m-3 NaCl. The root-to-stem ratio decreased significantly only at 200 mol m-3. Various other parameters were studied, such as water content, Na, K and Cl content, osmotic pressure, and CO2 uptake. Of these parameters the decreases in cladode water content and CO2 uptake were related to the decrease in cladode growth. Raised salinity increased cladode osmotic pressure, which was associated with tissue dehydration. We concluded that osmotic adjustment does not occur in prickly pear under salt stress.  相似文献

The cold-water extract from the skin of Opuntia ficus-indica fruits was fractionated by anion-exchange chromatography. The major fraction, which was purified by size exclusion chromatography, consisted of a polysaccharide composed of galactose and arabinose residues in the ratio 6.3:3.3, with traces of rhamnose, xylose and glucose, but no uronic acid. The results of methylation analysis, supported by (13)C NMR spectroscopy, indicated that this polysaccharide corresponded to an arabinogalactan having a backbone of (1-->4)-linked beta-D-galactopyranosyl residues with 39.5% of these units branched at O-3. The side-groups consisted either of single L-arabinofuranosyl units or L-arabinofuranosyl alpha-(1-->5)-linked disaccharides. This polysaccharide is thus an arabinogalactan that can be classified in the type I of the arabinogalactan family.  相似文献

After removal of the mucilage with water at room temperature, pectic polysaccharides were solubilized from Opuntia ficus-indica fruit skin, by sequential extraction with water at 60 degrees C (WSP) and EDTA solution at 60 degrees C (CSP). Polysaccharides with neutral sugar content of 0.48 and 0.36 mol/mol galacturonic acid residue were obtained, respectively, in the WSP and CSP extracts. These pectic polysaccharides were de-esterified and fractionated by anion-exchange chromatography, yielding for each extract five fractions, which were thereafter purified by size-exclusion chromatography. Two of these purified fractions were characterized by sugar analysis combined with methylation and reduction-methylation analysis. The study was then supported by (1)H and (13)C NMR spectroscopy. The results showed that the water-soluble fraction WSP3 and the EDTA soluble fraction CSP3, consisted of a disaccharide repeating unit -->2)-alpha-l-Rhap-(1-->4)-alpha-d-GalpA-(1--> backbone, with side chains attached to O-4 of the rhamnosyl residues. The side chains contained highly branched alpha-(1-->5)-linked arabinan and short linear beta-(1-->4)-linked galactan.  相似文献

Xylans were isolated from the pericarp of prickly pear seeds of Opuntia ficus-indica (OFI) by alkaline extraction, fractionated by precipitation and purified. Six fractions were obtained and characterized by sugar analysis and NMR spectroscopy. They were assumed to be (4-O-methyl-D-glucurono)-D-xylans, with 4-O-alpha-D-glucopyranosyluronic acid groups linked at C-2 of a (1-->4)-beta-D-xylan. The sugar composition and the 1H and 13C NMR spectra showed that their chemical structures were very similar, but with different proportions of D-Xyl and 4-O-Me-D-GlcA. Our results showed that, on average, the water soluble xylans have one nonreducing terminal residue of 4-O-methyl-D-glucuronic acid for every 11 to 14 xylose units, whereas in the water non-soluble xylans, xylose units can varied from 18 to 65 residues for one nonreducing terminal residue of 4-O-methyl-D-glucuronic acid.  相似文献

We tested the effects of above- and belowground competition and evaluated insect herbivory on the growth of a common cactus (Opuntia fragilis) in Sandhills prairie. Our purpose was to determine the independent and joint contribution of these factors to the variation observed in cactus size and local abundance. We manipulated ambient prairie vegetation and surface water availability in a full factorial design. When Opuntia fragilis was released from competition with surrounding vegetation, it grew significantly larger in the second growing season, both in terms of number and size of new cladodes. Ramets grown within live prairie vegetation averaged no net growth. These ramets were also more frequently fed upon by the larvae of two internally feeding cactus insects. As a result, insect herbivory reinforced the competitive suppression of the cactus by grasses. Water supplementation had no significant effect, either on cactus growth or on insect herbivory. We conclude that the mechanism by which dense prairie vegetation influenced net growth of O. fragilis was both direct, through competition for nonwater resources such as light, and indirect, through its mediation of feeding by specialized insect herbivores. Furthermore, since the direct growth response to treatment did not precede the indirect effect of habitat-mediated variation in feeding by insects, our results contradict the current expectation that indirect effects require more time to be expressed than do direct effects.  相似文献

Opuntia ficus-indica is a long-domesticated cactus crop that is important in agricultural economies throughout arid and semiarid parts of the world. The biogeographic and evolutionary origins of this species have been obscured through ancient and widespread cultivation and naturalization. The origin of O. ficus-indica is investigated through the use of Bayesian phylogenetic analyses of nrITS DNA sequences. These analyses support the following hypotheses: that O. ficus-indica is a close relative of a group of arborescent, fleshy-fruited prickly pears from central and southern Mexico; that the center of domestication for this species is in central Mexico; and that the taxonomic concept of O. ficus-indica may include clones derived from multiple lineages and therefore be polyphyletic.  相似文献

Of prickly-pear cacti occurring in Sicily, the most widespread and economically important isOpuntia ficus-indica (L.) Miller. In Sicily it has, since its introduction, played an important role in the exploitation of marginal areas. The Sicilian experience is described with reference to the historical outlines and the present intensive production of late fruit. Due to the pronounced morphological variation and geographical distribution of Galápagos' Opuntia cacti, numerous hypotheses have been advanced regarding their radiation, diversification, and classification. The currently accepted classification is based on morphology and recognizes six species and fourteen varieties, but the plasticity of many of the characteristics renders any morphological taxonomy problematic. Our analysis of previously published morphological data agrees only partially with the current classification. We present the first molecular phylogeny of these plants. Multiple DNA sequences indicate little genetic distinction among the currently identified species, despite restricted gene flow and limited long distance dispersal within the archipelago. No clear relationship exists between morphological and genetic differences. The Opuntia (prickly pear) genus contains over 200 species. Six of them are endemic to the Galapagos archipelago. Although these cacti are ‘keystone’ species of the Galapagos’ semi‐arid ecosystem, they have never been studied in detail. Because of their current threatened status and their important role in the ecosystem, we developed 16 microsatellite markers to study the population genetic structure of some of these species. These markers display a high level of polymorphism with numbers of alleles per locus ranging from six to 53. Results also revealed possible polyploidy in these cacti.  相似文献

Pereskia aculeata Miller (Cactaceae) is an invasive alien species in South Africa that is native in Central and South America. In South Africa, P. aculeata outcompetes native plant species leading to a reduction in biodiversity at infested sites. Herbicidal and mechanical control of the plant is ineffective and unsustainable, so biological control is considered the only potential solution. Climatic matching and genotype matching indicated that the most appropriate regions in which to collect biological control agents were Santa Catarina and Rio de Janeiro provinces in Southern Brazil. Surveys throughout the native distribution resulted in 15 natural enemy species that were associated with the plant. Field host range data, as well as previous host plant records, were used to prioritise which of the species were most likely to be suitably host specific for release in South Africa. The mode of damage was used to determine which species were most likely to be damaging and effective if released. The most promising species prioritised for further study, including host specificity and impact studies, were the stem-wilter Catorhintha schaffneri Brailovsky & Garcia (Coreidae); the stem boring species Acanthodoxus machacalis Martins & Monné (Cerambycidae), Cryptorhynchus sp. (Curculionidae) and Maracayia chlorisalis (Walker) (Crambidae) and the fruit galler Asphondylia sp. (Cecidomyiidae). By prioritising the potential biological control agents that are most likely to be host-specific and damaging, the risk of conducting host specificity testing on unsuitable or ineffective biological control agents is reduced.  相似文献

We examined levels of genetic variation and genetic structure in the leafy cactus (Pereskia guamacho) in arid and semiarid zones in Venezuela. We surveyed genetic diversity within 17 populations using 19 allozyme loci. Genetic diversity was relatively high at both the species (P(s) = 89%, A(s) = 3.26, AP(s) = 3.53, H(es) = 0.24) and population (P(p) = 63%, A(p) = 1.90, AP(p) = 2.42, H(ep) = 0.20) levels. A significant deficit of heterozygote individuals was detected within populations in the Paraguana Peninsula region (F(IS) = 0.301). Relatively low levels of population differentiation were detected at macrogeographic (G(ST) = 0.112) and regional levels (G(ST) = 0.044 for peninsula region and G(ST) = 0.074 for mainland region), suggesting substantial genetic exchange among populations; however, gene flow in this species seems to be regulated by the distance among populations. Overall, estimates of genetic diversity found in P. guamacho are concordant with the pattern observed for other cacti surveyed, namely high levels of polymorphism and genetic diversity with one common allele and several rare alleles per locus. Differences in gene dispersal systems between this species and other cacti studied were not reflected in the patterns of genetic diversity observed. The concentration of the highest estimates of genetic variation in northwestern Venezuela suggests a potential reservoir of plant genetic diversity within xerophilous ecosystems in northern South America.  相似文献

Partial hydrolysis of the mucilage of O. ficus-indica affords O-β-d-galactopyranosyl-(1→6)-d-galactose, the polymer-homologous trisaccharide, and fourteen oligosaccharides that contain arabinose and most of which have xylosyl end-groups. O-β-d-Xylopyranosyl-(1→5)-l-arabinofuranose and O-β-d-xylopyranosyl-(1→5)-O-α-l-arabinofuranosyl-(1→5)-l-arabinofuranose were the oligosaccharides isolated in greatest amount. 1. Until recently, neither the phycitid moth Cactoblastis cactorum nor the cochineal insect Dactylopius opuntiae have been satisfactory biological control agents of Opuntia stricta in South Africa.
2. In marked contrast, both of these agents have kept O. stricta under biological control for many decades in Australia.
3. In an attempt to improve the situation in South Africa, a stock of D. opuntiae was obtained from O. stricta in Australia during 1996.
4. Host-specificity tests confirmed that the newly imported D. opuntiae from Australia is a different biotype to the one already established in South Africa.
5. The Australian ('stricta') biotype thrives on O. stricta but is unable to develop satisfactorily on O. ficus-indica , while the converse is true for the South African ('ficus') biotype, which thrives on O. ficus-indica but fares poorly on O. stricta .
6. The integrity of the host-plant specificity of the two biotypes of D. opuntiae has important implications for biological control of Cactaceae in South Africa, and has greatly enhanced prospects that O. stricta can be brought under biological control successfully.  相似文献

A historical perspective on the use and production of species of Dactylopius (Hemipetera: Dactylopiidae) and Opuntia (Cactaceae: Opuntioidae), information on their origin, diversity and distribution in Mexico are reviewed, and aspects of their conservation are discussed. The use and exploitation of both genera are part of Mexican cultures since prehistory. Opuntia species were among the main components of human diet during pre-agricultural times. Cochineal was used and probably cultivated at least from the Tenth century. During the colonial period, cochineal generated significant benefits to the Spaniard colonizers and Mexico was the world’s first producer of insects and dyes until the mid Nineteenth century. Currently, Mexico is the main producer of Opuntia cladodes and prickly pear, but cochineal cultivation is marginal and only maintained in traditional indigenous systems. Mexico is one of the main areas of diversity of Opuntia, having 83–104 out of nearly 200 species worldwide. More than 50 species are used mainly as food, fodder and medicine and 20 species are cultivated with different degrees of domestication. The genus Dactylopius includes nine species, with five of them naturally occurring in Mexico. Only D. coccus has been cultivated and domesticated but other wild species have been used throughout history. Arid and semiarid areas of Mexico are among the most important reservoirs of biological diversity for both genera, particularly for D. coccus. Specific measures for protection of such biodiversity and generic resources are required. Strategies for in situ conservation combined with re-established use and cochineal production may enhance conservation policies.  相似文献

Explants from young joints of mature plants of tuna (Opuntia ficus-indica Mill.) were cultured on Murashige and Skoog (MS) medium containing 8.8 M benzyladenine (BA) and 0.5 M naphthaleneacetic acid (NAA). Shoots produced were utilized as secondary explants. Each shoot was cut longitudinally from apex to base into two explants, and some of these explants were cut transversely into proximal and distal explants. The size and number of shoots produced was affected by size and position of the explant within its source. To examine whether spininess evolved at random or differently in various life forms and plant organs, we characterized spiny, thorny and prickly organs in the entire wild flora of Israel (294 such species). Of the species, 63.3% defended their reproductive organs (the most‐defended organ) and 67.0% defended various non‐reproductive organs. Ninety‐three species defended both their reproductive organs and at least one other part; 48.3% defended their leaves and 36.4% their stems and branches. Spiny wings defended stems and branches only in herbaceous (annual or perennial) species. There were clear differences between the life forms. Annuals and perennial herbs defended mostly their reproductive organs (95.7 and 83.0%, respectively), dwarf shrubs defended mostly their leaves (54.2%) and shrubs and trees mostly their branches (89.7 and 76.2%, respectively). Trees do not defend their reproductive organs by associated sharp appendages. The differences in defence on various organs among different life forms may influence the results of meta‐analysis studies of the optimal defence allocation if such differences are not taken into account. We noted spine, thorn and prickle colours for 167 species with yellow, red, orange and white being the dominant, supporting hypotheses about spines being visually aposematic. © 2012 The Linnean Society of London, Botanical Journal of the Linnean Society, 2012, 168 , 344–352.  相似文献
