Summary Homologous high molecular weight storage prolamins were purified from grain of wheat, rye and barley using combinations of gel filtration, ion-exchange chromatography and preparative isoelectric focusing. Sodium dodecylsulphate polyacrylamide gel electrophoresis showed that the components were single bands with apparent mol.wts. of above 100,000. Molecular weights determined by sedimentation equilibrium ultracentrifugation were considerably lower; 54,700, 67,600 and 69,600 for the components from barley, rye and wheat respectively. Amino acid analysis showed the presence of 13.6 to 16.5 mol% glycine, 29.6 to 34.0 mol% glutamate + glutamine, 11.4 to 13.7 mol% proline and a total of 4.0 to 5.7 mol% basic amino acids. Automated N-terminal amino acid sequencing of the component from wheat showed the presence of cysteine residues at positions 5 and 10, and this is discussed in relation to the possible role of these proteins in the visco-elastic gluten network.  相似文献

The antigenic relationships between the prolamins of barley, rye and wheat have been studied by examining the specificity of an antibody to C hordein in a quantitative study using a laser nephelometer. The antibody reacts weakly with B hordein and strongly with 75-kdalton and 40-kdalton -secalins from rye and 3 some -gliadins from wheat. Absorption experiments and immunodiffusion tests indicate that there are shared antigenic determinants for most of the prolamins. All the species with reacting prolamins belong to the sub-family Festucoideae of the Gramineae. The prolamins of maize, pearl millet and sorghum, species of the sub-family Panicoideae, do not react. The results confirm the known lack of homology between the prolamins of the two sub-families and also indicate the presence of relationships not yet established between C hordein, the 75-kdalton and 40-kdalton -secalins and also 3 gliadin.Abbreviations HMW high molecular weight - PAGE polyacnylamide-gel electrophoresis - PBS phosphate-buffered saline - RLS relative light scattering - SDS sodlum dodecyl sulphate  相似文献

Summary Long-term cultures of four different cultivars of barley (Hordeum vulgare L.) have been established. Both callus and suspension cultures formed embryogenic structures at high frequency even after more than 18 months of culture. These compact proembryogenic cell clusters synthesize seed storage globulins whereas loose cell aggregates in callus culture and suspension cultures of fine dispersed consistency were free of globulins. Globulin synthesis was especially intense in compact structures of callus cultures established from suspension culture-derived protoplasts. Within the cells storage globulins are deposited in the vacuolar compartment as in zygotic embryos. 小麦醇溶蛋白组成可以作为小麦品种鉴定的指纹图谱,其分离方法有酸性电泳、反相高压液相色谱(RP-HPLC)和毛细管电泳(CE)等手段,3种方法相互补充,而CE分辨率最高。对醇溶蛋白酸性电泳条件的改良和完善仍在进行中,利用最新的分离技术对小麦醇溶蛋白基因进行染色体定位和遗传行为分析是近年来醇溶蛋白研究的另一领域。小麦高分子量麦谷蛋白亚基(HMW-GS)与小麦面包烘烤质量密切相关,关于它的研究目前主要集中在3个方面;对各个迁3移率较近的亚基进行快速,准确分离方法的研究,HMW-GS与小麦面包烘烤质量关系的研究和通过基因工程来改良小麦的品质、提高面粉的加工特性等。低分子量麦保蛋白(LMW-Glutenin)影响小麦面粉的特性,截止目前已经获得了17个该基因的克隆,并对其基因结构进行了描述,有些低分子量麦谷蛋白亚基(LMW-GS)加入碱性面粉后改变了面筋的性质,报道了小麦醇溶蛋白,高分子量麦谷蛋白亚(HMW-GS)、低分子量麦谷蛋白亚基(LMW-GS)3个方面的最新研究进展。  相似文献

This work was undertaken to determine the kinds and amount of substances that would account for the previously demonstrated differential growth of Claviceps purpurea on guttation fluids from Rosen rye, Genesee wheat, and Traill barley seedlings. Chromatographic methods were used for determining amino acids and sugars, spot tests and spectrometric methods for inorganic materials, and microbiological methods for vitamins.

Total sugar content is about equal in rye and barley fluids, but lower in wheat. Glucose is the principal sugar component of the rye and barley fluids and galactose highest in wheat. Most of the amino acid in all 3 fluids is aspartic acid or asparagine. Barley fluid is far higher than the other 2 in total amino acids, with wheat the lowest. Most inorganic elements are found to be highest in barley and lowest in wheat, with the exception of iron where rye is highest and barley lowest. Barley fluid is highest in choline, p-aminobenzoic acid, thiamine, and uracil, while rye is highest in inositol and pyridoxine. Wheat is much lower than the other 2 in choline and inositol.

High yield isolation of mesophyll protoplasts from wheat, barley and rye   总被引:1,自引:0,他引:1  
Efficient procedures are described for high-yield isolation of mesophyll protoplasts from spring wheat ( Triticum aestivum L. cv. Glenlea), winter wheat ( Triticum aestivum L. cv. Frederick), barley ( Hordeum vulgare L. cv. Bruce) and rye ( Secale cereale L. cv. Puma). Factors such as plant age, composition of the incubation medium during isolation, purification procedures and culture medium affect protoplast yield, viability and metabolic competence, as measured by light-dependent CO2 fixation. Optimal osmolarity of the isolation medium was equivalent to 1.8 times that measured in the leaves of all plant material used. The presence of 2 m M ascorbic acid in the preincubation and isolation medium increased the yield by 50% and conserved viability and metabolic competence. The protoplasts were stable for up to 48 h without loss of either viability or of original activity of CO2 fixation, which was in the order of 100 μmol CO2 (mg chl)−1h−1.
In our MC-56 liquid medium these protoplasts regenerated cell walls within 72 h and a few divided.  相似文献

Summary Legumin and vicilin were purified from seeds of Vicia faba L. var. Scuro, characterized in different electrophoretic systems, and used to produce polyclonal antibodies in rabbits. Two-dimensional electrophoretic studies showed a wide range of heterogeneity in the subunits of both legumin and vicilin. Legumin was found to be composed of 29 disulphide-linked subunit pairs with different molecular weight and/or isoelectric point. Western blot analysis of legumin of several mutants revealed molecular polymorphism based on a corresponding gene family. Three different -major legumin patterns were found, and inheritance studies showed that the 34.3-kD legumin polypeptide is the product of one locus, Lg-1, which is the first legumin genetic locus described in Vicia faba. Vicilin was found to be composed of as many as 59 subunits distributed in a molecular weight range of 65.7 to 42.8 kD (major polypeptides) and 37.2 to 15.2 kD (minor polypeptides), with different isoelectric points. A model is proposed that explains the possible formation of the minor subunits and the major subunits of 48.2 and 46 kD molecular weight (MW) from proteolytic cleavages and/or glycosilation of precursor polypeptides. Summary Genetic maps of the homoeologous group-2 chromosomes were constructed, comprising 114 loci in wheat and 34 loci in rye. These include the genes coding for sucrose synthase, sedoheptulose-1,7-bisphosphatase, a bZIP protein (EmBP-1), a peroxidase and an abscisic acid-induced protein (#7). Overall, gene orders are highly conserved in the genomes of wheat, barley and rye, except for the distal ends of chromosome arms 2BS and 2RS, which are involved in interchromosomal, probably evolutionary, translocations. Clustering of loci in the centromeric regions of the maps, resulting from the concentration of recombination events in the distal chromosomal regions, is observed in wheat and rye, but not in barley. Furthermore, loci for which homoeoloci can be detected in rye and barley tend to lie in the centromeric regions of the maps, while non-homoeologous and wheat-specific loci tend to be more evenly distributed over the genetic maps. Mapping of the group-2 chromosomes in the intervarietal Timgalen x RL4137 cross revealed that the T. timopheevi chromosome segment introgressed into chromosome 2B in Timgalen is preferentially transmitted. Regeneration potential of different wheat, rye and barley species in leaf explant culture. Comparative analysis of the induction ability of morphogenetic processes in vitro has been carried out in 16 wheat genotypes, 4 barley species and 6 rye genotypes. It has been shown that tetra- and hexaploid wheat species as well as wild barley species exhibited the highest embryogenic potential in the leaf explant culture while diploid wheat species and rye genotypes showed the lowest one. Genotypic dependence of processes of callus formation, induction of embryogenic calli and regeneration was revealed in the studied species.  相似文献

Summary Eight leaf peroxidase isozymes were distinguished by IEF in Chinese Spring. Two genes which control the production of three of these isozymes were located on chromosome arms 1BS and 1DS by nullisomic analysis. These loci probably form part of a homoeoallelic series and have been designated Per-B1 and Per-D1 respectively. Analysis of chromosome 1B short arm terminal deletion stocks indicated that the Per-B1 locus is located between the nucleolar organiser region and another isozyme marker, Hk-B1. Two variant leaf peroxidase phenotypes were distinguished in a small sample of hexaploid wheat varieties. Analysis of wheat-alien addition and substitution lines identified homoeologous loci in rye (Per-R1) and barley (Per-H1).  相似文献

We have purified the protein synthesis inhibitors from wheat (tritin) and investigated their antigenic relationship to inhibitors from other grains and more distantly related plants. Three forms of tritin (tritin 1, 2, and 3) indistinguishable with regards to their molecular weight and enzymatic properties were found. Antiserum against the most abundant tritin, tritin 2, cross reacted with tritin 1, tritin 3, and with the translation inhibitors from barley and rye, but not with other plant-derived translational inhibitors. The results indicate that only closely related plants contain antigenically similar protein synthesis inhibitors.  相似文献

Soybean (Glycine max L.) storage proteins are composed mainly of two major components, beta-conglycinin and glycinin. Electrophoretic variants of the beta subunit of beta-conglycinin and the A3 polypeptide of glycinin were detected on SDS-PAGE, and designated them as beta* and A3*, respectively. beta* and A3* exhibited higher and lower mobilities, respectively, than the common beta subunit and A3 polypeptide. The N-terminal nine and 10 amino acid sequences of beta* and A3* were completely identical to the previously reported sequences of the beta subunit and the A3 polypeptide, respectively. Analysis using concanavalin A-horseradish peroxidase and treatment with N-glycosidase indicated that glycans were not responsible for the difference in electrophoretic mobility of beta* or A3*. Furthermore, five clones of beta* or beta and three clones of A3*, respectively, were sequenced but we could not detect deletions and insertions except for a single or a few amino acid substitutions as compared with the common beta subunit and A3 polypeptide. These results indicate that a single or a few amino acid substitution affects the electrophoretic mobilities of beta* and A3*.  相似文献

Arabinoxylans that interfere with growth of ice crystals have been purified from rye (Secale cereale L., Rosen) and two varieties of wheat (Triticum aestivum L., Genesee and Hillsdale) seed. The most active polysaccharide from each seed type was homogeneous in the sense that all the molecules were in the same size range, they contained the same sugar residues, and they reacted similarly in chemical characterization experiments. Structural studies showed that the polysaccharides consist of a xylan chain to which are attached side-chains that contain a single, terminal arabinose residue. The polysaccharides differ with respect to the number of arabinose residues. The xylose:arabinose ratios in the most active fractions from rye, Genesee wheat, and Hillsdale wheat are 1.26, 1.54, and 2.08, respectively. Gel-permeation column chromatography showed that the most active polysaccharide from each seed type has a molecular weight greater than 2 x 10(6) and that the rye polysaccharide is slightly larger than the Hillsdale wheat polysaccharide. The rye polysaccharide is a better inhibitor of ice-crystal growth than is the Hillsdale wheat polysaccharide.  相似文献

The objective of the current work was to analyse the variability of high and low molecular mass (HMM and LMM) glutenin subunits, along with some morphological characteristics in sixty Spanish accessions of rivet wheat (Triticum turgidum L. ssp. turgidum). The lines were grouped in sixteen botanical varieties and five additional types, according the morphological criteria. Up to 13 allelic variants (four alleles for the Glu-A1 locus and nine alleles for the Glu-B1 locus) and 34 B-LMMGs patterns were found in the evaluated lines. The current data indicated a clear reduction of morphological variability, along with an asymmetric distribution of the alleles and patterns for seed storage proteins. This polymorphism could be useful for enlarging the genetic background of modern durum wheat.  相似文献

The segregation of the 75K gamma secalin locus (Sec-2) in combination with five interchanges (reciprocal translocations) and two marker genes was analyzed. The translocations involved chromosome arms 1RL, 1RS, 2RL, 2RS, 4RL, 5RL, 5RS, 6RL and 6RS. The gene loci were both on 2R, but the arm was not known. Although the Sec-2 locus was expected to be on chromosome 2RS, no linkage between Sec-2 and any of the markers was found. This is concluded to be the result of exceptionally frequent recombination between Sec-2 and the break point of one of the translocations, which is the only marker in 2RS.  相似文献

81 wild forms and 110 cultivated cowpea,Vigna unguiculata, accessions from 21 countries of Africa were screened for variability in seed storage proteins. Total seed proteins, albumin and globulin fractions were investigated by means of sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and isoelectric focusing (IEF) of nonreduced and/or reduced samples in one- and two-dimensional procedures. The globulin fraction is heterogeneous in molecular weight and contains both legumin-like components and three to six nondisulfide-linked subunits. Three globulin subunits, with molecular weights 110, 76, and 41 kD were found to be composed of disulfide-linked polypeptides. In the nondisulfide-linked fraction, both cultivated and wild forms exhibited patterns of four types (A–D). This fraction contains polypeptide subunits of molecular weights 62, 56, and 52 kD for A type, 62, 56, 54, and 52 kD for B type, 62, 56, 52, and 50 kD for C type, and at least 62, 56, 54, 52, 50, and 49 kD for D type. These subunits present similar multiple charge forms but C and D types possess more basic specific 50 and 49 kD nondisulfide linked components. Major albumin fraction contains subunits of 94, 86, 32, and 24kD. No infraspecific variation was observed in albumin or legumin-like fractions. The discussion is focussed on the relations between genetic variability assessed by storage protein coding genes and phenotypic variability.  相似文献
