本研究探讨了NO对深层培养的桦褐孔菌积累抗氧化酚类化合物的影响。在桦褐孔菌的培养基中分别加入0.01mmol/L、0.1mmol/L以及1mmol/L的NO供体硝普钠,并测定总酚的含量及其抗氧化活性。发酵产物的抗氧化活性以清除DPPH和羟自由基活力表示。加入0.1mmol/L的硝普钠可使桦褐孔菌胞内外酚类化合物分别达到最高水平67mg/g和677mg/L。不同培养时期产生的胞内外酚类化合物都具有抗氧化活性,尤其是添加0.1mmol/L硝普钠的菌丝体。因此NO可用于上调深层发酵培养的桦褐孔菌酚类化合物的积累。  相似文献

黑色素合成是桦褐孔菌Phaeoporus obliquus次生代谢过程中的一个重要事件。黑色素的过多合成势必影响其他酚类的积累。本文以茉莉酸和双氧水为调控因子,开展了该突变体深层发酵过程酚类化合物积累研究。正常培养条件下,每升发酵液积累酚类化合物最高可达317.45mg/L,其中包括苯甲酸衍生物(45.3mg/L),黄酮苷元(74.4mg/L),黄酮苷(160mg/L)以及硬毛素类化合物(18.5mg/L)。加入茉莉酸后,总酚最大值增加到620.45mg/L,其中苯甲酸衍生物增加到230.26mg/L,黄酮苷元增加到166.86mg/L,而黄酮苷和硬毛素类化合物却分别降低到98.5mg/L和7.1mg,双氧水的加入没有提高总酚的最大积累(315.69mg/L)。因此,与野生型相比,黑色素缺失使总酚的积累有所增加,随茉莉L酸的加入总酚积累最大值增加近一倍。虽然双氧水的加入提高了苯丙氨酸裂解酶活性,但总酚的积累却没有增加,这可能是由于一部分酚类化合物在抗双氧水氧化胁迫过程中被消耗。  相似文献

研究了p H(4.0~7.0)对桦褐孔菌(Phaeoporus obliquus)H011菌株深层培养菌丝体和滤液甲醇提取物抗氧化活性、自由基清除能力及抗氧化成分的影响。结果表明:p H显著影响桦褐孔菌菌丝体和滤液甲醇抽提物的抗氧化能力、自由基清除能力、总酚和多糖产量;当p H为6.0时,滤液甲醇提取物的抗氧化能力、对自由基的清除能力最强,显著高于其他组(P0.05);当p H为6.0时,菌丝体甲醇抽提物多酚产量和多糖产量达到最高,分别是189.9和47.2 mg·g-1,均分别显著高于其他组的含量(P0.05),桦褐孔菌菌丝体和滤液甲醇抽提物的抗氧化能力、自由基清除能力与总酚、多糖产量有关;深层培养的抗氧化能力比子实体高。因此,可通过桦褐孔菌深层培养生成抗氧化剂来取代固体培养。  相似文献

黄酮类化合物是桦褐孔菌菌丝体中多酚类化合物的重要组成部分,也是该菌治疗众多疾病的有效成分之一。然而人工培养桦褐孔菌黄酮等酚类化合物积累甚少,导致药理活性的明显下降。为此,我们研究了3种氨基酸和4种霉菌水提物对深层发酵桦褐孔菌黄酮的积累及其抗氧化能力的影响。在所试验的3种氨基酸和4种霉菌水提物中,L-酪氨酸,黄曲霉和毛霉水提物能有效地增加该菌黄酮的积累。人工培养菌体中的黄酮至少由4种黄酮苷组成,苷元分别是槲皮素、柚皮素、山奈酚和异鼠李素。深层发酵菌丝体具有一定的抗氧化能力,并与总黄酮的含量呈正相关。由L-酪氨酸,黄曲霉和毛霉水提物调控生长的桦褐孔菌菌丝体,能有效地清除超氧阴离子、羟基自由基和DPPH自由基。  相似文献

为评价桦褐孔菌和松口蘑发酵菌粉的降血糖作用,本文观察了经不同浓度桦褐孔菌和松口蘑菌粉溶液灌胃后,小鼠正常血糖、给予葡萄糖小鼠以及四氧嘧啶糖尿病小鼠血糖的变化.结果表明:桦褐孔菌、松口蘑菌粉溶液(500、1000 mg/kg)对小鼠正常血糖值、正常小鼠糖耐量没有明显影响(P>0.05);对于糖尿病小鼠血糖,桦褐孔菌低剂量组(500 mg/kg)和桦褐孔菌高剂量组(1000 mg/kg)的降糖效果较为明显(P<0.01),用药前与用药后第21 d的血糖值分别为(14.94±1.85)、(10.43±2.22)和(14.89±1.65)、(10.17±2.14) mmol/L,降糖率分别为30.23% 和31.7%.桦褐孔菌和松口蘑菌粉对于小鼠正常血糖、正常小鼠糖耐量无明显影响,对于四氧嘧啶糖尿病小鼠具有一定的降血糖效果.  相似文献

以桦褐孔菌总酚得率为指标,在单因素试验基础上,根据中心组合试验设计原理,采用三因素三水平的响应面设计方法对超声提取过程中影响总酚得率的乙醇浓度、超声温度及时间进行优化。结果表明桦褐孔菌总酚超声提取的最佳工艺条件为:采用55%乙醇,料液比1∶30,在52℃下超声提取40 min,在此最佳条件下,总酚得率为19.42 mg/g,实验结果与模型预测值相符,相对误差为+0.78%。  相似文献

旨在研究真菌激发子对桦褐孔菌多酚合成的影响,采用液体摇瓶培养方式,将真菌激发子加入到桦褐孔菌液体培养物中,检测桦褐孔菌次级代谢产物的积累,以及真菌激发子对桦褐孔菌一氧化氮合酶(NOS)活性的影响。结果表明,适量添加真菌激发子能够提高桦褐孔菌的代谢速率,提高多酚类化合物的合成,胞内和胞外多酚最高可达102.15 mg/L和311.91 mg/L,并能显著提高桦褐孔菌NOS的活性,其活性最高可达151.93 U/mg prot,发酵液一氧化氮(NO)含量最高可达256.28μmol/L。这表明,真菌激发子能够有效激活桦褐孔菌的次级代谢产物合成途径。  相似文献

作者旨在阐明真菌激发子对桦褐孔菌多酚积累的影响。以摇瓶法培养桦褐孔菌,在其培养液中加入真菌激发子和一氧化氮合酶抑制剂氨基胍,观察桦褐孔菌多酚和一氧化氮的积累并测定菌丝体内一氧化氮合酶和苯丙氨酸解氨酶的活性。多酚以Folin-Ciocalteu法测定,一氧化氮的积累量用硝酸还原酶法测定,一氧化氮合酶和苯丙氨酸解氨酶活性均以分光光度计法测定。结果表明,添加45μg/mL的激发子可使桦褐孔菌菌丝体多酚的积累量达到46.5mg/g,显著地高于正常培养的菌丝体多酚积累量34.6mg/g;同时加入45μg/mL的激发子和10mmol/L氨基胍则使菌丝体多酚积累的最高水平降为34.8mg/g。此外,激发子的加入显著促进了一氧化氮的产生并提高了苯丙氨酸解氨酶活性,而这种促进和提升作用为氨基胍所抑制。这表明真菌激发子能显著地提升桦褐孔菌多酚类化合物的积累,而一氧化氮可能是这种提升作用的信号传导分子。  相似文献

为提取桦褐孔菌菌质各活性组分并研究其体外抗氧化活性,将桦褐孔菌菌质醇提后分别梯度萃取获得各极性组分,将残渣沸水浸提醇沉得粗多糖;将获取的各活性组分别进行DPPH自由基清除率、羟基自由基清除率和总抗氧化能力的测定,并测定各组分中总多酚、总三萜的含量;同时与桦褐孔菌菌核进行对比。研究结果表明菌质和菌核乙酸乙酯、正丁醇组分对DPPH自由基、OH自由基清除能力及其总抗氧化能力明显优于相应的其他组分(P0.01);菌质的乙酸乙酯、正丁醇组分对DPPH自由基的清除能力与菌核相应组分比较无显著性差异(P0.05),菌质和菌核的乙酸乙酯、正丁醇部位的总三萜和总多酚含量较高。结果显示桦褐孔菌菌质的乙酸乙酯、正丁醇组分的体外抗氧化活性较好,总多酚和总三萜含量较高,抗氧化活性的强弱可能与该二者活性成分的含量相关。通过双向固体发酵技术制得的桦褐孔菌菌质体外抗氧化作用较好。  相似文献

Folin-Ciocalteu比色法测定桦褐孔菌多酚的条件优化   总被引:3,自引:1,他引:2  
以没食子酸为标准品,通过正交试验和单因素实验研究了Folin-Ciocalteu 比色法测定桦褐孔菌中多酚含量的适宜条件。结果表明,优化后的显色条件为Folin-Ciocalteu试剂用量0.3mL、10% Na2CO3溶液0.6mL,25℃时避光反应30min,于750nm处测定其吸光值。多酚质量浓度在0.0032-0.0256mg/mL范围内与吸光值有良好的线性关系。根据拟合的线性回归方程对桦褐孔菌多酚进行定量测定,野生桦褐孔菌和人工培养桦褐孔菌中多酚含量最高分别为(72.05±0.08)mg/g、(52.76±0.06)mg/g(n=6)。不同加标水平的加标回收率测定实验获得的平均回收率为98.95%,RSD为1.27%。该法测定桦褐孔菌多酚具有快速、准确,稳定性强,重现性好,精密度高的特点,可应用于实际样品的测定。  相似文献

Phenolic compounds produced by sclerotia of Inonotus obliquus are the active constituents responsible for antioxidant activities. In this study, I. obliquus was grown in a continuously stirred tank reactor (CSTR) to explore how it accumulates phenolic compounds in different culture media and whether these compounds possess antioxidant activities. Phenolic compounds produced by I. obliquus in the control medium consisted of melanins, flavonoids, polyphenols and small phenolics. Their accumulation was affected by adding H(2)O(2) to the medium, where increased levels of total intracellular phenols (TIP) and melanins, but less total extracellular phenol (TEP) occurred. Simultaneous exposure to H(2)O(2) and arbutin resulted in a further increase in TIP production and reduced accumulation of TEP. Both TIP and TEP obtained at different culture ages and media were active in scavenging superoxide anion and DPPH radicals. Therefore, production of phenolic compounds by I. obliquus is enhanced by imposing oxidative stress, which might allow it to be exploited as a reliable source of pharmaceutically important phenolic compounds.  相似文献

Phenolic compounds from field-grown Inonotus obliquus sclerotia (Chaga) consist mainly of hispidin analogs and melanins, and are thought to be the active constituents to treat several human diseases. In submerged cultures of the fungus, however, no information is currently available on the production of phenolic compounds and their corresponding pharmacological functions. In this study, phenolic compounds from Chaga and submerged cultures of the fungus were assayed for their composition and immune-stimulating effects. Phenolic compounds produced by I. obliquus in submerged cultures mostly consist of flavonoids, together with small amounts of hispidin analogs and melanins. This is quite contrary to the situation in Chaga, where flavonoids are determined as trace elements. Furthermore, phenolic compounds from Chaga show capacity about two-fold higher than those produced in submerged cultures in inhibiting cyclophosphamide-induced reduction of bodyweight, spleen index and viability of peripheral lymphocytes in test mice. Thus less production of hispidin analogs and melanins is likely to be responsible for less immune-stimulating effects in phenolic compounds from submerged cultures, and additional factors should be imposed during submerged cultures of I. obliquus to regulate biosynthesis of phenolic compounds directed to the composition similar to Chaga.  相似文献

Needles of Larix gmelinii seedlings grown under different irradiances (100, 52, and 26 % of natural sunlight) were collected from June to August 2009. The content of phenolic compounds and condensed tannins in needles were strongly affected by different irradiances. The highest content of phenolic acids occurred under the lowest irradiance. Chlorogenic acid and syringic acid were detected only under the shade. In contrast, the needles under full irradiance showed the highest content of condensed tannins.  相似文献

The fungus Inonotus obliquus (Pers.) Pil. synthesised high-molecular-weight phenolic pigments that were assigned to melanins according to their physicochemical properties. It was showed that copper ions (0.008%), pyrocatechol (1.0 mM), and tyrosine (20.0 mM) stimulated the melanogenesis. The production of melanin correlated with the synthesis of o- and p-diphenoloxidases. The fungal melanin had strong antioxidant and genoprotective effects.  相似文献

The common green alga Scenedesmus obliquus may respond morphologicallyto the presence of natural enemies. Exposure to water-bornecues from the herbivorous zooplankton Ceriodaphnia reticulata,Daphnia galeata x hyalina, Daphnia magna and Daphnia pulicariastimulated the formation of protective colonies in S. obliquus.This response seemed strongly related to the amount of algaegrazed upon, because a highly significant correlation betweenthe amount of S. obliquus consumed and the induced colony formationwas found. However, when exposed to medium that had been inhabitedby the carnivorous zooplankton Bythotrephes longimanus and Leptodorakindtii, no colony formation occurred. A similar result wasobtained in two different experiments when S. obliquus was exposedto filtrate from cultures of the freshwater fish ide (Leuciscusidus) and perch (Perca fluviatilis). These results support thehypothesis that S. obliquus responds to a herbivorous zooplanktonchemical cue, rather than to a more general animal excretoryproduct. No support was obtained for the hypothesis that cuesfrom the enemy of their enemy could serve as signals to thealgae: despite the presence of filtrate from fish culture, filtratefrom Daphnia cultures induced the formation of colonies in S.obliquus. The biological activity seems to be linked to thealga–grazer interaction, which ensures a reliable cuethat evokes the morphological response of S. obliquus only whennecessary. The grazer-induced colony formation can be viewedas an adaptive reaction in habitats with variable grazing pressurefrom an assemblage of many different herbivores to pare downmortality through grazing.  相似文献

采用基于核磁共振代谢组学的方法研究了两株地理隔离的桦褐孔菌中国牡丹江菌株(IOM)和芬兰万塔菌株(IOV)在深层培养过程中主要代谢产物组成的差异。IOM和IOV两菌株在培养过程中积累菌丝体生物量、菌丝体酚类(TMP)以及三萜化合物(TMT)的过程相似,但IOM菌株产生较高水平的TMP。代谢产物组成动力学比较结果表明,IOM在第3天含有较高水平的硬毛素类似物、桦褐孔菌二糖和TMT,但在第5天均有所下降。导致这些差异的代谢产物为fuscoparianolD,21,24-羊毛甾-7,9(11),23-三烯-3β,22,25-三醇,羊毛甾-8,23-二烯-3β,22,25-三醇,23,24-环戊羊毛甾-3β,21,25-三醇-8-烯,桦褐孔菌二糖,phelligridinsC,D及H,甲基地花菌素A和C,地花菌素B,davallialactone及methyl davallialactone。这些代谢产物在IOV的提取物中均参与清除自由基,而在IOM的提取物中主要是酚类化合物参与自由基的清除。两菌株的这些差异反应了环境条件对桦褐孔菌生长及代谢的影响,即不仅影响其次生代谢产物的合成,还影响代谢产物在防止细胞氧化损伤过程中所起的作用。  相似文献

The composition and concentrations of phenolic compounds were studied in the first true leaves, cotyledons, stems and roots of 2.5-week-old seedlings of mountain birch ( Betula pubescens ssp. czerepanovii ). The differences in secondary compounds among these plant parts were both qualitative and quantitative. In all parts, condensed tannins accounted for more than 50% of the phenolics. In the first true leaves and cotyledons, chlorogenic acid was the most abundant of the HPLC phenolics. The main components in stems were (+)-catechins and rhododendrins whereas in roots, the main components were ellagitannins. The seedlings were grown at three levels of nitrogen supply (very low-N, low-N, moderate-N), and the effect of nitrogen on concentrations of phenolic compounds was studied in all plant parts. The dry weight of all plant parts, except the roots, increased with increased nitrogen. In all parts, the concentration of condensed tannins was higher at lower levels of nitrogen than at moderate-N. The concentrations of total HPLC phenolics and also those of the compound groups of HPLC phenolics were, however, affected only in the first true leaves and roots. The concentrations in the first true leaves were generally higher in seedlings grown at very low-N and low-N than in seedlings grown at moderate-N. The concentrations in roots were highest at low-N. Not all compounds responded to nitrogen supply in the same manner. The changes in concentrations cannot be exclusively interpreted as changes in the accumulation of phenolic compounds, due to dilution caused by the increase in biomass in better nitrogen availability. There were differences in carbon allocation between condensed tannins and HPLC phenolics in seedlings grown at different nitrogen levels.  相似文献
