This trial studied the effect of including mannanoligosaccharides (MOS, Bio-Mos®, Alltech Inc., USA) in the diet on the caecal volatile fatty acids (VFA) and pH of rabbits from 34 to 90 days of age. Three experimental diets were compared: control diet, zinc bacitracin (ZnBac) diet (control diet with 0.1 g ZnBac/kg feed) and MOS diet (control diet with 2.0 g MOS/kg feed). Rabbits were slaughtered at 34, 48, 69 and 90 days of age and caecal contents were collected and analyzed for dry matter, pH and VFA concentration. The empty caecum and caecal contents weights relative to live weight were also determined.Age affected (P<0.0001) VFA concentration and pH values in the caecum. The pH decreased with age whereas VFA concentration increased. Rabbits fed MOS had higher (P<0.05) VFA and tended (P=0.098) to had lower pH in the caecum than rabbits fed ZnBac and control diets. Acetic, propionic and butyric acids concentrations in the caecum increased with MOS but its molar proportions were similar among diets. Diet had no effect on empty caecum weight and caecal contents weight and dry matter concentration. There was no interaction effect between diet and age. From 34 to 90 days of age, VFA production was higher (P<0.05) in the caecum of rabbits fed MOS than in those fed ZnBac diet and control diet.The addition of MOS to the diet increased the VFA concentration in the caecum of growing rabbits from 34 to 90 days of age. 相似文献

An experiment was conducted to determine the effects of level of fibre on production performance, gastrointestinal tract development, caecum fermentation and fibrolytic activity in weaner to 2-month-old New Zealand rabbits. One hundred weaner rabbits were allocated in individual cages for five treatments in which they were fed each diet with ADF at 130 (NDF, 274.1; ADL, 39.7; CP, 158.4; EE, 26.8; starch, 252.5 g/kg), 160 (NDF, 292.9; ADL, 41.9; CP, 160.3; EE, 25.6; starch, 249.4 g/kg), 190 (NDF, 312.4; ADL, 50.5; CP, 157.6; EE, 24.2; starch, 218.2 g/kg), 220 (NDF, 343.9; ADL, 51.2; CP, 158.5; EE, 22.2; starch, 217.6 g/kg), and 250 g/kg (NDF, 375.7; ADL, 66.0; CP, 156.9; EE, 22.4; starch, 220.8 g/kg) original matter basis, respectively. The results were: ADG and F/G deteriorated with increasing fibre (P<0.05); ADI increased with increasing dietary ADF (P=0.002). The stomach relative weight (g/kg LW), small intestine relative weight (g/kg LW), caecum weight, caecum content weight and caecum relative weight (g/kg LW) all increased with increasing ADF (P=0.003, 0.007, 0.002, 0.010 and <0.001, respectively). Quadratic effects of level of ADF on the villus height, villus height/crypt depth of the duodenum and the villus height/crypt depth of the jejunum were obtained; cubic effects of level of ADF on the crypt depth of the jejunum and the villus height/crypt depth of the ileum were obtained. Cubic effects of level of ADF on the pH, the butyric acid (% total VFA) and C2/C4 were obtained. The NH3–N concentration dropped and the acetic acid (% total VFA) increased when the dietary ADF increased (P<0.001). The fibrolytic activity measured in the caecal contents increased when the dietary ADF increased (P<0.001). Providing a ration consisting mainly of clover meal, wheat straw and peanut hulls, the optimum dietary ADF content for weaner to 2-month-old growing meat rabbits was shown to be 190 g/kg feed. 相似文献

Two experiments were conducted to evaluate the effects of live yeast supplementation on performance, nutrient digestibility, enteric microbial populations and volatile fatty acid (VFA) concentration of weanling pigs, receiving diets supplemented with aureomycin and elevated doses of CuSO4. In experiment 1, 90 crossbred pigs (7.20 +/- 0.44 kg, 28 d of age) were randomly allotted to one of five dietary treatments containing either 0, 4.0 x 10(6), 9.0 x 10(6), 2.6 x 10(7), or 5.1 x 10(7) cfu Saccharomyces cerevisiae per gram with six replicate pens per treatment and three pigs per pen. BWG and feed intake increased quadratically during days 1-14 and days 1-28 as live yeast levels increased (p < 0.01). Pigs fed the diet containing 2.6 x 10(7) cfu yeast per gram had the highest BWG and feed intake among the treatments. In experiment 2, 48 crossbred pigs (7.64 +/- 0.72 kg, 28 d of age) were fed diets containing live yeast at 0 or 3.2 x 10(7) cfu of S. cerevisiae per g with six replicate pens per treatment and four pigs per pen. The yeast supplementation improved BWG and feed intake during days 1-14 (p < 0.01) and days 1-28 (p < 0.05). Treatment differences were not observed in any of the bacterial populations, yeast numbers or VFA concentrations, at any of the sites of the gastrointestinal tract tested. Total tract nutrient digestibility was also not different between treatments. Overall, dietary supplementation of live yeast had a positive effect on BWG and feed intake of weanling pigs, receiving diets supplemented with aureomycin and elevated doses of CuSO4. The improvement in BWG appears to be partly related to an increase in feed intake. The mechanism of yeast improving feed intake of piglets needs to be explored. 相似文献

The effects of dietary cholesterol and fatty acids on the plasma cholesterol level and rates of very low density lipoprotein (VLDL) cholesterol secretion and low density lipoprotein (LDL) transport through LDL receptors in the liver of the hamster were investigated. Increases of plasma VLDL- and LDL-cholesterol levels and VLDL-cholesterol secretion from hepatocytes were observed in animals fed a diet enriched with 0.1% cholesterol for 2 weeks in comparison with animals fed a control diet. The addition of dietary palmitic acid accelerated the effect of dietary cholesterol on plasma VLDL- and LDL-cholesterol levels and VLDL-cholesterol secretion from hepatocytes. Dietary linoleic acid accelerated the effect of dietary cholesterol on VLDL-cholesterol secretion from hepatocytes and diminished the effect on the plasma LDL-cholesterol level. Hepatic LDL receptor activity was considerably suppressed by a control diet containing 0.05% cholesterol and a further small suppression was induced by a diet enriched with 0.1% cholesterol with or without 5% palmitic acid. However, dietary linoleic acid diminished the effect of dietary cholesterol on the suppression of hepatic LDL receptor activity. These results suggest that dietary palmitic acid augments the effect of dietary cholesterol in elevating the plasma LDL-cholesterol level through acceleration of VLDL-cholesterol secretion from the liver, and that dietary linoleic acid diminishes the effect of dietary cholesterol in elevating the plasma LDL-cholesterol level by preventing the suppression of hepatic LDL receptor activity induced by cholesterol. 相似文献

Previous studies from our laboratory established that low-fat diets prevent immunosuppression and reduce oxidative stress after a thermal injury. The purpose of the present study was to test the hypothesis that the type of dietary fatty acid influences splenocyte proliferation and oxidative stress following a burn injury. Female C3H/HeN mice were fed ad libitum six experimental diets (5% w/w lipids) differing in fatty acid composition for 10 days following a burn injury. Compared to the controls, burned mice fed whichever diet showed lower lymphoproliferative responses to concanavalin-A (Con-A) and lipopolysaccharide (LPS) (p<0.01), but not to an anti-T cell receptor monoclonal antibody (H-57). In burned animals, nitric oxide (NO) concentration was negatively correlated to the proliferation induced by Con-A (p<0.01) or LPS (p<0.05). These results suggest that: (1) dietary fatty acid type does not influence the splenocyte proliferation or oxidative stress and (2) NO production is involved in the immunosuppression following burn injury. 相似文献

The in vitro caecal fermentation of five substrates low in starch and protein content [d-(+)-glucose (GLU), d-cellobiose (CEL), sugar beet pectin (PEC), sugar beet pulp (SBP) and wheat straw (WS)] was investigated using soft faeces from rabbits receiving different levels of cellobiose and soluble fibre as inoculum. A total of 24 rabbits were supplemented 3 levels of cellobiose in the drinking water (0.0, 7.5, 15.0 g/l) and fed two experimental diets containing either low soluble fibre (LSF) or high soluble fibre (HSF) levels (84.0 and 130 g/kg dry matter). All substrates were subjected to a two-step pepsin/pancreatin in vitro pre-digestion, and the whole residue was used as substrate for the in vitro incubations. Gas production was measured until 144 h, and volatile fatty acid (VFA) production was determined at 24 h incubation. Experimental treatments did not affect SBP fermentation and had only a subtle influence on fermentation of WS and GLU. In contrast, cellobiose supplementation × donors’ diet interactions were detected for most gas production parameters for CEL. Both the fractional gas production (k) and maximal gas production rates were linearly increased (p ≤ 0.042) and the initial delay in the onset of gas production (Lag) linearly decreased (p < 0.001) by cellobiose supplementation with the HSF inoculum, with no differences between the 7.5 and 15.0 doses. In contrast, with the LSF inoculum cellobiose supplementation only affected k values, which were quadratically increased (p = 0.043) and had maximal values for the 7.5 dose. A quadratic effect (p ≤ 0.018) of cellobiose supplementation was observed for total VFA production at 24 h when CEL and PEC were fermented, obtaining the maximal VFA production for the 7.5 dose of cellobiose. Total VFA production for CEL was greater with LSF than with HSF inoculum (20.7 vs. 12.9 mmol/l; p = 0.014), but the opposite was found for WS (3.97 vs. 6.21 mmol/l; p = 0.005). The use of LSF inoculum for CEL fermentation sharply reduced acetate (p = 0.001) and increased butyrate proportions (p ≤ 0.001) compared with the HSF inoculum. A positive relationship between total VFA caecal concentrations in rabbits receiving the same experimental treatments and in vitro values was only observed when WS was used as substrate (r = 0.90; p = 0.015; n = 6). The results suggest that experimental factors influenced the fermentative activity of caecal digesta, but the observed response differed with the incubated substrate, being the CEL the most affected. 相似文献

The study aimed to investigate the effects of insoluble dietary fibre differing in lignin content on performance and parameters of gut microbiology in 48 weanling piglets (8.5 kg mean body weight) fed common diets ad libitum. Fibre sources tested were wheat bran (low lignin) and pollen from Chinese Masson pine (Pinus massoniana) as model of fibre rich in lignin, added to diets as follows: no addition (control), 3.0% wheat bran, 1.27% pine pollen and 2.55% pine pollen. In colonic chyme, bacterial colony counts remained unaffected by treatment, but ammonia contents were reduced by fibre additions (up to -38%). The effects on ammonia were best explained by added cellulose and lignin. Fibre additions reduced apparent (faecal) digestibility of dry matter (DM) and crude protein (CP) up to -3.5 and -4.3% units with cellulose and lignin being the major determinants for changes in CP digestibility. 相似文献

To verify if non-invasive collection of soft faeces (SF) from rabbits can be used as an index of bacterial biodiversity in caecal contents (CC), 24 weaned rabbits were given diets with low (LI) and high (HI) levels of insoluble fibre (neutral detergent fibre, NDF) and low (LS) and high (HS) levels of soluble fibre (neutral detergent soluble fibre, NDSF). After 21 days, animals were fitted with neck collars for SF collection. Two days later, animals were slaughtered and CC sampled. Total bacterial concentration quantified by real time PCR (log10 ng DNA/mg DM) was higher in SF than CC (2.615 vs. 2.383). Among diets, in CC it was (P = 0.059) lowest in LILS diet, whereas in SF it decreased (P = 0.025) with the NDF level. DGGE profiles showed that structure of bacterial communities of SF was close to that of CC; however, similarity was higher in LI than HI diets (0.82 vs. 0.74). Diversity indexes in CC decreased with NDSF (P < 0.05), whereas the effect of NDF (P < 0.05) was also appreciated in SF. Soft faeces can be an alternative to surgery or slaughter techniques to monitor changes in caecal bacterial community; however, high dietary NDF may decrease similarity between both communities. 相似文献

Effect of substrate and feed antibiotics on in vitro production of volatile fatty acids and methane in caecal contents of chickens 总被引:5,自引:0,他引:5
An attempt was done to identify some factors influencing the caecal fermentation pattern in poultry. Experiments were conducted to study effects of carbohydrate substrates (feed components and supplements) and antibiotics on the formation of volatile fatty acids (VFA) and methane in in vitro incubations of the caecal contents of 7-week-old chickens. Stoichiometry of fermentation differed in cultures with different carbohydrates. Fermentation pattern characterized by high propionate and low acetate production was found in cultures with lactose (0.447 and 0.376 mol/1 mol of VFA produced, respectively) and, to a lesser extent, also in cultures with raffinose. Acetate was the predominant metabolite of starch, pectin and xylan (0.727, 0.773 and 0.685 mol/1 mol of VFA produced, respectively). Fermentation of inulin resulted in high proportion of butyrate, 0.221 mol/1 mol of VFA. Other polysaccharides produced only 0.060–0.111 mol of butyrate per 1 mol of VFA. Oligosaccharides (lactose, raffinose) were fermented more rapidly than polysaccharides. Fermentation of inulin yielded more VFA than fermentation of starch, pectin and xylan. No production of VFA from carboxymethylcellulose was observed. On average, 11 mols of VFA were produced per mol of methane. Lasalocid significantly increased molar proportion of propionate, which is potentially beneficial from the point of view of salmonellae control. The magnitude of improvement, however, was small. Other feed antibiotics tested (avoparcin, bacitracin, lincomycin, spiramycin, tylosin, virginiamycin) produced only non-significant or marginal fermentation shifts. The aim of the study was to evaluate the effect of a temporary quantitative feed restriction on growth performance, nutrient digestibility and carcass criteria of rabbits. A total of 80 weaned male Californian rabbits (30 d of age) were randomly assigned to four treatments of 20 rabbits each. The Control group was fed ad libitum during the whole experimental period (days 30–72 of age). For the three restricted fed groups the feed intake was reduced by 15%, 30% and 45% compared to the Control group, respectively. The feed restriction was applied after weaning and lasted for 21 d. Thereafter, at 51 d of age, in all treatments the feed supply returned to ad libitum intake till 72 d of age (AL period). The feed restriction decreased the body weight gain of rabbits (during the restriction period and the whole experimental period, p < 0.001) and improved feed conversion ratio during all tested periods (p < 0.001). In the AL period, the daily body weight gain of all groups was similar. After the AL period, the digestibility of all measured nutrients was significantly higher for animals fed restrictively. Furthermore, feed restrictions significantly decreased the proportion of perirenal and scapular fat and increased relative weight and length of the gastrointestinal tract. Therefore, it can be concluded that the applied feed restriction improved feed conversion, nutrient digestibility and reduced fat at the slaughter age of Californian rabbits, but the reduced body weight gain could not be compensated by a subsequent ad libitum feeding for 3 weeks. 相似文献

1. Immature hens were preconditioned to a standard diet containing either 0, 100, 200 or 400 g/kg of added dried grass, 200 g/kg powdered cellulose, or 200 g/kg grass with an enzyme supplement, and were killed at either 10.30, 12.30 or 15.30 hr (after measurement of intestinal sugar absorption reported elsewhere). 2. Contents of caeca removed from these birds immediately after death were weighed and analysed for pH, uric acid, free sugars and volatile fatty acids, and the results related to dietary fibre level and time of day (of death). 3. Wet weights and uric acid concentrations of caecal contents both increased with increasing grass in the diet; neither measure varied with time, thus supporting the proposal that filling of caeca is continuous. Values of pH were all close to neutrality. 4. Mean molar concentrations of glucose, galactose, mannose, xylose, arabinose, fucose and rhamnose were in the proportions 36:2:3:1:4:1:1, respectively. Xylose and rhamnose declined with increasing grass; as did glucose, galactose and mannose with added cellulose; glucose, mannose and arabinose levels changed with time. The relative abundance of glucose in caecal contents should be taken into account when estimating contributions of fermentation products to energy balance. 5. Mean concentrations of acetate, propionate, butyrate, iso-butyrate, valerate and iso-valerate were in the proportions 72:22:16:1:2:2. In order to test whether monounsaturated fatty acids (MUFA, M) would lower plasma cholesterol and/or triglycerides (TG), 14 young men were studied in a metabolism ward. They were given two experimental diets with the same P/S ratio (P: polyunsaturated; S:saturated fatty acids) but different P + M/S ratios in composition of dietary fatty acids. Six men were in experiment I. In two groups of three, each group was given either the high or the low P + M/S ratio diet for 6 weeks. In experiment II, two groups of four men were subjected to a 3-week cross-over feeding trial (total of 6 weeks). We found in the crossover study that when the dietary P/S ratio was fixed at 1.0, those subjects given a diet with the higher P + M/S ratio of 4.5 showed significantly higher plasma total TG, very low density lipoprotein (VLDL)-TG, low density lipoprotein (LDL)-TG, and LDL-cholesterol than when given a diet with a lower P + M/S ratio of 1.5. Plasma total cholesterol, apoprotein A-I, and apoprotein B-100 were not significantly different between the two dietary periods. Plasma fatty acid patterns showed a significantly higher plasma MUFA level during the period of a higher P + M/S diet. In contrast to some recent reports, our studies indicate that a large amount of dietary MUFA may raise some fractions of plasma lipids in humans. 相似文献

The effects of feeding n-6 and n-3 fatty acids to broiler hens on cardiac ventricle fatty acid composition, and prostaglandin E2 (PGE2) and thromboxane A2 (TXA2) production of hatched chicks were investigated. Fertile eggs obtained from hens fed diets supplemented with 3.5% sunflower oil (Low n-3), 1.75% sunflower+1.75% fish oil (Medium n-3), or 3.5% fish oil (High n-3) were incubated. The hatched chicks were fed a diet containing 18:3 n-3, but devoid of longer chain n-6 and n-3 fatty acids for 42 days. Arachidonic acid content was lower in the cardiac ventricle of High n-3 and Medium n-3 compared to Low n-3 birds for up to 2 weeks (P<0.002). Long chain n-3 fatty acids were higher in the cardiac ventricle of chicks from hens fed High and Medium n-3 diets when compared to chicks from hens fed the Low n-3 diet. Differences in long chain n-3 fatty acids persisted up to four weeks of age (P<0.001). Peripheral blood mononuclear cells (PBMNC) of 7-day-old High n-3 broilers produced significantly lower PGE2 and TXA2 than PBMNC from Low n-3 and Medium n-3 birds. These results indicate that maternal dietary n-3 fatty acids increases cardiac ventricle n-3 fatty acids while reducing arachidonic acid and ex vivo PGE2 and TXA2 production during growth in broiler chickens. 相似文献

Apart from being an energy source, dietary fibre is also discussed to act as anti-nutritional factor reducing apparent precaecal protein and amino acid (AA) digestibility due to reduced absorption or increased endogenous secretion or both. However, the amounts of protein and AA of endogenous origin determined at the terminal ileum in cannulated animals do not represent the total losses associated with endogenous secretion. A high proportion of secreted protein is reabsorbed and does not reach the terminal ileum, and losses occur during synthesis of endogenous protein. Therefore, the present study used an alternative indirect approach, taking the reduction of nitrogen (N) retention in a threonine (Thr) limited diet as a sensitive indicator for fibre-associated Thr losses. Two experiments were conducted with 12 castrated male pigs each between 37 and 75 kg body weight to measure the effect of the intake of Thr and 150 and 300 g/d fibre from wheat bran (Exp. 1), or 150 g/d fibre from rape seed, cassava leaves, and cassava root peels, respectively (Exp. 2), on N retention. During two (Exp. 1) and three (Exp. 2) balance periods the animals were subjected to the dietary treatments according to a cross-over design. All animals received 1350 g/d of a wheat-soybean-based diet supplemented with free AA to ensure Thr being the first-limiting AA. To determine the effect of Thr on N retention, intake of the basal diet was reduced to 1150 g/d and supplemented with corn starch to reach equal energy intake and an unchanged AA pattern. With increasing BW additional starch was added to all diets to ensure a constant energy intake of 1.25 MJ ME/kg BW(0.75). Since the fibre sources contained small amounts of Thr, N retentions were corrected for precaecal digestible Thr intake originating from the fibre sources according to the Thr effect on N retention as determined in experiment 1. Corrected N retentions were affected by fibre level (p = 0.007) and source (p < 0.001). Fibre-associated Thr losses amounted to 3.3, 3.2, 1.2, and 1.1 g/kg fibre from wheat bran, rapeseed, cassava leaf, and cassava root peel, respectively. The effects of fibre in a pelleted diet on food intake, digestibility of crude fibre, dry matter and energy, on passage time and consistency of faeces were studied in 2 species of Callithricidae, Callithrix jacchus and Saguinus fuscicollis. Coarse cellulose, microcellulose, wheat bran and shrimp meal (chitin = crude fibre) were tested in diets containing 2,4 and 6% total crude fibre, respectively. Digestibility and passage time were determined by inclusion of 0.5% Cr2O3 in the diet. Both celluloses had little influence on the digestibility of energy and dry matter. Digestibility of crude fibre was very low. Wheat bran led to evident depression of energy and dry matter digestibility. High digestibility of crude fibre occurred at the higher levels of inclusion in the diet. Shrimp meal was highly digested with little influence on digestibility of energy and dry matter, indicating considerable degradation of chitin. Wheat bran showed a marked effect, while microcellulose had no effect on passage time, consistency and volume of faeces. 相似文献

In animals, the composition of fatty acids (FAs) in body pools reflects dietary intake. This paper reviews evidence that the manipulation of tissue lipids of farmed fish, by feeding them different natural oils, can have significant effects on their respiratory and cardiovascular physiology. Sturgeon and eels with tissue lipids rich in highly unsaturated FAs of the n-3 series (n-3HUFAs, accumulated from dietary menhaden oil) had significantly lower metabolic rates than fish with tissues rich in saturated FAs (SFAs, from coconut oil), although they grew equally well. In sturgeon, the difference in metabolism influenced tolerance of hypoxia. Degrees of hypoxia that depressed oxygen uptake and spontaneous activity in fish rich in SFAs had no such effects on fish rich in n-3HUFAs. In the isolated sturgeon heart working in vitro, reduced oxygen supply depressed the performance of hearts with lipids rich in SFAs but not that of hearts rich in n-3HUFAs. In salmon fed diets with graded mixtures of menhaden and canola oils, there was no relationship between tissue n-3HUFA content (from menhaden oil) and any measured aspect of swimming performance, but a linear relationship between maximum sustainable swimming speed and muscle oleic acid levels (from canola oil). Such exploratory studies indicate that an animal's responses to its environment may be profoundly affected by the oils and FAs it consumes in its diet. 相似文献
