Summary With the indirect immunofluorescence technique of Coons and collaborators somatostatin-like immunoreactivity (SLI) was observed in certain neurons of the central and peripheral nervous system of the rat. In the cell bodies a strong SLI was observed with a distribution resembling that of the Golgi apparatus. In addition a weak SLI was diffusely distributed in the cytoplasm. After photography the sections processed for immunocytochemistry were stained with the thiamine pyrophosphatase technique of Novikoff and Goldfischer. The latter technique is assumed to be a specific marker for the Golgi complex. It was found that the strong SLI and the thiamine pyrophosphatase activity had an identical distribution. Summary Neurones in the suboesophageal ganglion of the locust Schistocerca gregaria were stained with an antiserum raised against gamma amino butyric acid (GABA). This ganglion consists of the fused mandibular, maxillary and labial neuromeres. Immunoreactive cell bodies of similar size and distribution occur in the lateral, ventral and middorsal regions of all three neuromeres. Approximately 200 cell bodies stain in both the mandibular and maxillary neuromeres and 270 in the labial neuromere. A few distinctly larger cells occur in the ventral groups and one large pair occurs in the lateral group of the maxillary neuromere. Dorsal commissures DCIV and DCV are composed mainly of stained fibres, while DCI–DCIII are largely unstained. A ventral commissure also stains in the maxillary neuromere. All longitudinal tracts contain both stained and unstained fibres. Many processes within the neuropil are also immunoreactive. A stained axon is found in the posterior tritocerebral commissure which enters the anterior dorsal region of the mandibular neuromere. The salivary branch of the 7th nerve contains one stained axon and two axons stain in nerve 8 which innervates neck muscles.  相似文献

Samples were incubated with an antiserum specific for GABA coupled to BSA with glutaraldehyde. Anti-rabbit IgG-HRP was processed by the two step method (Eckert and Ude 1983). GABA-positive varicose fibers and also nerve cell bodies were revealed within the myenteric plexus. The density of GABA-immunoreactive neurons was not higher than 4-8 cell/cm2, which is approximately 1% of the total nerve cell number in the myenteric plexus.  相似文献

Summary The distribution of gamma-aminobutyric acid (GABA)-like and glutamate decarboxylase (GAD)-like immunoreactivity was studied in the cervical sympathetic ganglion complex of rats, including the intermediate and inferior cervical ganglia and the uppermost thoracic ganglion. GABA-positive axons may enter the ganglion complex via its caudal end. Others apparently arise from small GABA-positive cell bodies which are scattered among principal neurons, within clusters of SIF cells and in bundles of GABA-negative axons. The majority of these cells is located in the lower half of the ganglion complex. Principal neurons did not react with antibodies against GABA or GAD. An unevenly distributed meshwork of GABA-immunoreactive axons was seen in each of the ganglia. Immunoreactive axons formed numerous varicosities. Some of them were aggregated in a basket-like form around a subpopulation of GABA-negative principal ganglion cell bodies. Electron-microscopic immunocytochemistry revealed that GABA-positive nerve fibers establish asymmetric synaptic junctions with dendritic and somatic spines of principal neurons, whereas postsynaptic densities are inconspicous or absent on dendritic shafts and somata. The results suggest that in the cervical sympathetic ganglion complex principal neurons are not GABAergic, but are innervated by axons which react with both antibodies against GAD and/ or GABA antibodies and originate from a subpopulation of small neurons.  相似文献

Summary The development of GABA-like immunoreactivity was investigated in embryonic and juvenile locusts using an antibody raised against GABA-protein conjugates. GABA-like immunoreactivity was first detectable in the neuropile of embryonic ganglia at 55% development, and in neuronal somata at 62% development. The total number of immunoreactive somata increased between 62% and 85% embryonic development, and followed an anterio-posterior pattern of expression. At 85% development, the number of immunoreactive somata reached adult levels and no change in number was then seen. In embryonic stages and first and second juvenile instars two dorsal and four ventral groups of somata were labeled in all three thoracic ganglia, whilst in later juvenile instars one of the dorsal groups was visible as a separate entity only in the metathoracic ganglion. These early patterns were modified by alterations in the positions of some of the groups during late embryogenesis and during juvenile development to produce the adult pattern. The results show that the development of GABA expression is similar to that of other neurotransmitters. The characteristics of the development of immunoreactivity indicate that some of these immunoreactive clusters may be derived from clonally related neurones. Summary Application of two methods for the selective staining of neurons, Golgi impregnation and intracellular marking with Procion Yellow, has revealed the anatomical arrangements of the horizontal cells in the carp retina. There are two basic horizontal-cell types, those with axons and these without. The former can be subdivided into three groups on the basis of the pattern of branching of the dendrites. These three structural groups are also functionally distinct, as shown by the electrical recordings made during Procion-Yellow injection. The axons of these three types of cell project into the proximal part of the inner nuclear layer, where they expand to form morphologically indistinguishable terminals. Fine horizontal processes leave the surfaces of these axon terminals. The functional behavior of a terminal resembles that of the associated soma. Dopamine-like immunoreactivity was demonstrated histochemically in about ten local interneurons in the antennal lobe of Periplaneta americana. The somata of these neurons are within the ventrolateral group of cell bodies. Additional immunohistochemical tests revealed that the same neurons also have a GABA-like immunoreactivity. Immunohistochemical dopamine staining (preembedding) of preparations in which the antennal receptor fibers had been caused to degenerate showed that in the glomerular neuropil these antennal fibers form output synapses on dopamine-immunoreactive neurons. Summary In crickets, a deutocerebral motoneuron sends axon collaterals to 6 of the 7 antennal muscles. Previous results indicated that this neuron exerts inhibition on these muscles and thus may be a common inhibitory motoneuron. In our present study, we show by doublelabelling, i.e. retrograde cobalt-filling and GABA-immunocytochemistry, that this neuron is GABA-immunoreactive, thus demonstrating that one common inhibitory motoneuron is part of the antennal motor system of crickets.  相似文献

Summary An antiserum raised against gamma aminobuyric acid (GABA) was used to stain the thoracic nervous system of the locust. It stained both neuronal somata and processes within the neuropile. Among the stained somata, those of the three pairs of common inhibitory motor neurones could be identified in each of the three thoracic ganglia. In the pro- and mesothoracic ganglia five discrete groups of somata are stained, four ventral and one dorsal. In the metathoracic neuromere, an additional second dorsal group can be identified. In the abdominal neuromeres of the metathoracic ganglion both dorsal and ventral somata are stained but the latter cannot be divided into discrete populations. In each ganglion, dorsal commissures (DC) IV and V are composed of stained neurites, DCVII, the supramedian commissure, the perpendicular tract, and all the longitudinal tracts contain both stained and unstained neurites. DCI, II, III and VI, the T and I tracts are unstained. An abundance of GABA-like immunoreactive processes is found throughout the neuropile except for the anterior ventral association centre where stained processes are sparser. Summary The transmitter content of identified inhibitory interneurons in the flight system of the locust, Locusta migratoria, has been characterized using antibodies raised against protein-conjugated gamma aminobutyric acid. Identified flight neurons were filled with the fluorescent dye, Lucifer Yellow. Serial sections of dye-filled neurons were incubated with an antibody to gamma aminobutyric acid which was subsequently tagged with a fluorescent marker. Excitatory motoneurons to wing muscles and 13 flight interneurons (3 excitatory, 7 inhibitory, and 3 with unknown synaptic effect) were examined. Neither the moto-neurons nor any of the 3 excitatory interneurons contained immunoreactive material. Six of the 7 inhibitory interneurons did contain immunoreactive material. All the neurons which contained immunoreactive material and whose synaptic effect is known were inhibitory. We conclude that most of the inhibitory flight interneurons which have been described use gamma aminobutyric acid as their transmitter. Interestingly, at least 1 set of interneurons known to be inhibitory does not use gamma aminobutyric acid. We predict that the 2 interneurons which do contain immunoreactive material and whose synaptic effect is not yet known will be found to have inhibitory roles in the operation of the flight circuitry.  相似文献

Individual olfactory neurons in the antennal lobes of Periplaneta americana were investigated for their morphological and neurochemical properties by means of intracellular cobalt injection and indirect peroxidase-antiperoxidase immunohistochemistry. Whole-mounts of 1-day-old chicken midgut were incubated with an antiserum against GABA-glutaraldehyde-BSA conjugate. The immunoreaction was visualized by using the peroxidase-antiperoxidase method, and processed for consecutive light and electronmicroscopic observation. GABA was selectively localized in some of the varicose and nonvaricose nerve fibres of the myenteric plexus. The varicose fibres formed dense networks within the myenteric ganglia, some of which--mainly in duodenum--also contained immunopositive nerve cell bodies. Some of the varicose fibres projected out from the myenteric plexus into the circular muscle layer. At the electronmicroscopic level, labelled axon terminals formed synaptic contact with unlabelled perikarya and vica versa. At the same time, no labelled terminals were found on immunostained cells. In a few cases, axon terminals with GABA positivity were situated close to the smooth muscle cells in the circular muscle layer, suggesting a prejunctional GABA effect on the neighbouring nerve terminals on the release of their transmitters.  相似文献
