Analysis for gibberellin-like substances (GA) and cytokinins in cold and non-cold treated fall-bearing and cold treated June-bearing cultivars of red raspberries (Rubus idaeus L.) was made at the 10 and 20 node stages of growth (at 10 nodes, all plants were vegetative; at 20 nodes, only cold treated plants of fall-bearing cultivars were flower induced). Quantitative changes in GAs, as analyzed by the lettuce hypocotyl bioassay, were observed only in flower induced plants. In fall-bearing cultivars at the 20 node stage of growth, there was a 100% greater activity in cold than non-cold treated plants. Likewise, cold treated plants had a 60% greater activity at the 20 than at the 10 node stage of growth. Major qualitative differences in GAs were observed between the 10 and 20 node stage of growth in cold treated fall-bearing cultivars but not in cold treated plants of June-bearing cultivars. Changes in compounds at Rf 0.3 and 0.5 accounted for 35 to 54% of the total increase in GA activity between flower induced and non-induced stages of growth. Cytokinin activity tested by the tobacco callus bioassay was greater at the 20 than at the 10 node stage of growth in all cultivars and treatments. However, cold treated plants of the fall-bearing type had 70% greater cytokinin activity than non-cold treated plants at both stages of growth. It is suggested that gibberellins, possibly acting synergistically with cytokinins, are involved inflower induction in red raspberries.  相似文献

The relationship between age-related resistance of peper plants to Phytophthora capsici and contents of carbohydrates, amino acids, phenolics and mineral nutrients in pepper stems was studied using two pepper cultivars, Hanbyul (susceptible) and Kingkun (resistant). With increasing age of pepper plants, the two cultivars, which differ in their susceptibility to Phytophthora blight, became gradually resistant to the disease. The cultivar Kingkun distinctly showed the age-related resistance to Phytophthora blight at the second branch stage. The weight of dry matter in healthy stems of pepper plants at the second branch stage was twice that at the six leaf stage. The resistant cultivar Kingkun contained lower levels of fructose, glucose and sucrose in stems than the susceptible cultivar Hanbyul at the different developmental stages. No consistent differences between the developmental stages of the plants were recognized with regard to their glucose content. However, the contents of fructose and sucrose in the cultivar Hanbyul greatly increased at the second branch stage. The levels of inositol reduced in both pepper cultivars during plant development. In view of the fact that there were only slight changes in the amount of total amino acids, it seems unlikely that there is a relationship between the amino acid metabolism and the retardation of Phytophthora infection during plant development. The amounts of total phenolic compounds in pepper stems were relatively low at the later growth stages of the plants and also in the resistant cultivar Kingkun. The contents of macroelemental nutrients such as nitrogen, phosphorus, potassium, calcium and magnesium were drastically reduced in pepper stems at the later plant growth stage. No significant differences between the cultivars or the plant growth stages were found in the silicon and microelemental nutrients such as sodium, iron, zinc and manganese. These results suggest that the expression of age-related resistance of pepper plants may be due to the morphological and nutritional changes in tissues of pepper stems during ageing, i.e. the pronounced increase in weight of dry matter, the significant decrease in amounts of mineral nutrients such as nitrogen, phosphorus, potassium, calcium and magnesium, and the tow contents of fructose, glucose and sucrose in the stem tissues.  相似文献

Interactions between Brassica cultivars (cvs) and adult and larval stages of Plutella xylostella L. were examined. In six-way choice oviposition experiments, glasshouse-grown Chinese cabbage cv. Tip Top was preferred to five outdoor-grown temperate cabbage cultivars but was similar to four out of five tropical cultivars. Bionomic studies with neonate larvae on temperate cabbage cultivars showed that mean larval duration was longer and percentage survival less on older compared with younger plants but that plant age or cultivar had little effect on pupal weight. The fecundity of P. xylostella reared on outdoor-grown cabbage cultivars varied ten-fold between the poorest and the best cultivar but plant age had little effect on P. xylostella reared on Chinese cabbage. In a glasshouse experiment, survival of P. xylostella on Chinese cabbage was significantly greater compared with field-grown glossy-leaved, normal bloom green cabbage and red cabbage. In bioassays with neonate larvae, when the leaves were placed vertically instead of horizontally a significant level of intrinsic resistance was revealed for glasshouse and field-grown glossy-leaved cabbage but not for Chinese cabbage. Observed differences in host status to P. xylostella are discussed in relation to the potential for partial plant resistance in control programmes.  相似文献

Myzus persicae transmitted soybean mosaic virus (SMV) most efficiently following 30 or 60 s acquisition probes on infected plants. There were no differences in susceptibility to SMV infection of soybean plants 1 to 12 wk old, but symptoms were more severe in plants inoculated when young than when old. Soybeans inoculated between developmental stages R3 and R6 only showed yellowish-brown blotching on one or more leaves. There were no observable differences in the time of appearance or type of symptoms shown by soybean seedlings inoculated either by sap or by aphids; infected plants became acquisition hosts for aphids 5–6 days after inoculation. There was no change in the efficiency with which M. persicae transmitted SMV from source plants up to 18 wk after inoculation. M. persicae transmitted SMV from leaves of field-grown soybeans when plants were inoculated at developmental stages V6, R2, and R3 and tested as sources 57–74 days after inoculation but not from plants inoculated at R5 and tested as sources 14 to 32 days after inoculation. M. persicae acquired SMV from soybean buds, flowers, green bean pods, and unifoliolate, trifoliolate, and senescent leaves. Middle-aged and deformed leaves were better sources of the virus than buds, unfolding and old symptomless leaves. The results are being incorporated into a computer model of SMV epidemiology.  相似文献

Enzyme-linked immunosorbent assay was used to measure the concentration of potato leafroll virus (PLRV) antigen in different parts of field-grown secondarily infected plants of three potato genotypes known to differ in resistance to infection. The antigen concentration in leaves of cv. Maris Piper (susceptible) was 10–30 times greater than that in cv. Pentland Crown or G 7445(1), a breeder's line (both resistant). Differences between genotypes in antigen concentration were smaller in petioles and tubers (5–10-fold) and in above-ground stems (about 4-fold), and were least in below-ground stems, stolons and roots (about 2-fold). PLRV antigen, detected by fluorescent antibody staining of tissue sections, was confined to phloem companion cells. In Pentland Crown, the decrease in PLRV antigen concentration in leaf mid-veins and petioles, relative to that in Maris Piper, was proportional to the decrease in number of PLRV-containing companion cells; this decrease was greater in the external phloem than in the internal phloem. The spread of PLRV infection within the phloem system seems to be impaired in the resistant genotypes. Green peach aphids (Myzuspersicae) acquired < 2800 pg PLRV/aphid when fed for 4 days on infected field-grown Maris Piper plants and < 58% of such aphids transmitted the virus to Physalis floridana test plants. In contrast, aphids fed on infected Pentland Crown plants acquired <120 pg PLRV/aphid and <3% transmitted the virus to P. floridana. The ease with which M. persicae acquired and transmitted PLRV from field-grown Maris Piper plants decreased greatly after the end of June without a proportionate drop in PLRV concentration. Spread of PLRV in potato crops should be substantially decreased by growing cultivars in which the virus multiplies to only a limited extent.  相似文献

The growth and development of field-grown Avena fatua plants were studied for autumn and spring sowings in two consecutive years. The duration of various growth stages from sowing until anthesis was quantified in terms of thermal time (accumulated degree days) or photothermal time (degree days modified by photoperiod). Base temperatures and photoperiods for developmental phases were estimated as those which minimised the coefficient of variation among sowing dates. Relationships were derived between leaf emergence, canopy height, plant leaf area, and photothermal time. Stem extension and flowering occurred earlier in autumn-sown plants than spring-sown plants. Autumn-sown plants produced more leaves on the main stem, and had greater leaf area and above-ground biomass at anthesis than spring-sown plants.  相似文献

An efficient in vitro protocol was established for developing pathogen-free plantlets in Cucurbita pepo through meristem culture. Meristems of about 0.3–0.5 mm in size were isolated from shoot tips of 25–30 day old in vitro grown plants. For primary establishment of isolated apical meristem, MS liquid medium supplemented with 2.0 mgl KIN and 0.5 mg/l GA3 was found to be most effective in both cultivars. MS semisolid medium containing 2.0 mg/l BAP were found to be most effective for shoot development from primarily established meristem in both cultivars. A good number of shoots were not concomitant with good rooting. The best root induction was found in media having 1.0 mg/l IBA in cv. Bulum. It was found that cv. Bulum was better than cv. Rumbo in all stages of meristem culture. The presence of virus in plantlets was achieved by DAS-ELISA test, where 68–81% plantlets have been proved to be virus free among the studied viruses. Healthy growth and vigour was observed in meristem derived plants over their source plants after cultivation under natural conditions.  相似文献

In order to study the species composition of endophytes from wheat healthy plants in Buenos Aires Province (Argentina) and to determine their infection frequencies from leaves, stems, glumes and grains, wheat plants were collected from five cultivars at five growth stages from crop emergence to harvest. A total of 1,750 plant segments (leaves, stems, glumes and grains) were processed from the five wheat cultivars at five growth stages, and 722 isolates of endophytic fungi recovered were identified as 30 fungal genera. Alternaria alternata, Cladosporium herbarum, Epicoccum nigrum, Cryptococcus sp., Rhodotorula rubra, Penicillium sp. and Fusarium graminearum were the fungi that showed the highest colonization frequency (CF%) in all the tissues and organs analysed. The number of taxa isolated was greater in the leaves than those in the other organs analysed.  相似文献

In Brazil, the silverleaf whitefly (SLW), Bemisia tabaci biotype B, is a serious soybean pest. SLW management is difficult and new control strategies, such as host–plant resistance, are required. Herein, we aimed to evaluate the biology of SLW, from eggs to adults, on seven soybean cultivars. The emergence of adult insects was monitored daily. Defense-related compounds were identified and quantified from the V3 to the V8 stages in SLW-infested and non-infested plants. The rates of emergence of SLW adults were lower in cultivars ‘IAC 17,’ ‘IAC 19’, and ‘IAC 24’ compared with the susceptible cultivar ‘IAC Holambra Stwart’. Rutin, genistin, genistein, and salicylic acid were identified and quantified in plant extracts. The rutin, genistin, and genistein levels decreased after SLW infestation. Rutin concentrations increased in infested plants of ‘IAC 17’ (V6) and ‘IAC 19’ (from V5 to V8). ‘Barreiras’ showed the highest genistein content in non-infested plants; from V5 growth stage, it was not detected in cultivars Doko (infested), Vencedora, ‘IAC 17’, and ‘IAC 24’ (non-infested). High levels of salicylic acid were observed in ‘IAC ‘19’-infested plants (at V3 and V5). The results suggest that rutin can be related to SLW adults’ emergence only when the feeding source was ‘IAC 19’. Consequently, further studies are needed to access the associated gene expressions and the effect of other secondary metabolites, mainly volatile compounds from SA pathway, including its consequences on feeding preference and mostly in relation to IAC cultivars.  相似文献

Efficient protocols for shoot regeneration from leaf explants suitable for micropropagation as well as for the development of transgenic plants were developed for blueberry (Vaccinium corymbosum) and lingonberry (Vaccinium vitis-idaea) cultivars. Nodal segments were used to initiate in vitro shoot cultures of lingonberry cultivar ‘Red Pearl’ and southern highbush blueberry cultivar ‘Ozarkblue’. In order to develop an optimized regeneration procedure, different types and concentrations of plant growth regulators were tested to induce adventitious shoot regeneration on excised leaves from micropropagated shoots of both cultivars. The effect on percentage regeneration and number of shoots per explant was investigated. Results indicated that zeatin was superior to TDZ and meta-topolin in promoting adventitious shoot formation. A concentration of 20 μM zeatin was most effective in promoting shoot regeneration in both cultivars, in case of ‘Red Pearl’ along with 1 μM NAA. Shoots were either allowed to root in vitro on medium containing IBA or NAA or ex vitro in a fog tunnel. IBA was superior to NAA for induction of root development in vitro in both Vaccinium cultivars. Ex vitro rooting under high humidity was tested with cuttings from mature field-grown plants, from acclimatized tissue culture derived plants and with unrooted in vitro proliferated shoots planted directly. It was found that in vitro shoots rooted better under fog than cuttings from the other plant sources and rooting was equivalent to that achieved in vitro.  相似文献

Field and greenhouse experiments were conducted to assess the nitrogen fixation rates of four cultivars of common bean (Phaseolus vulgaris L.) at different growth stages. The 15N isotope dilution technique was used to quantify biological nitrogen fixation. In the greenhouse, cultivars M4403 and Kallmet accumulated 301 and 189 mg N plant–1, respectively, up to 63 days after planting (DAP) of which 57 and 43% was derived from atmosphere. Under field conditions, cultivars Bayocel and Flor de Mayo RMC accumulated in 77 DAP, 147 and 135 kg N ha–1, respectively, of which approximately one-half was derived from the atmosphere. The rates of N2 fixation determined at different growth stages increased as the plants developed, and reached a maximum during the reproductive stage both under field and greenhouse conditions. Differences in translocation of N were observed between the cultivars tested, particularly under field conditions. Thus, the fixed N harvest index was 93 and 60 for cultivars Flor de Mayo and Bayocel, respectively. In early stages of growth, the total content of ureides in the plants correlated with the N fixation rates. The findings reported in the present paper can be used to build a strategy for enhancing biological N2 fixation in common bean.  相似文献

Summary Germplasm collections of vegetatively propagated crops are usually maintained as plants in fields or potted in greenhouses or screened enclosures. Safety duplication of these collections, as duplicate plants or separate collections, is costly and requires large amounts of space. Cryopreservation techniques which were recently developed for long-term storage of pear germalasm may offer an efficient alternative to conventional germplasm collection maintenance. Pear (Pyrus L.) germplasm may now be stored as seeds (species), dormant buds or pollen from field-grown trees, or shoot tips fromin vitro-grown plants (cultivars). Pear germplasm may now be cryopreserved and stored for long periods (> 100 yr) utilizing slow-freezing or vitrification ofin vitro-grown shoot-tips. Dormant bud freezing, pollen, and seed cryopreservation of other lines are being developed to complete the base collection forPyrus. This cryopreserved collection provides base (long-term) storage for the field-grown pear germplasm collection at the National Clonal Germplasm Repository, Corvallis, Oregon. Impact of initial density of cowpea aphid Aphis craccivora Koch (Aphididae) at infestation on the growth and yield of aphid-susceptible cowpea cultivar ICV-1 and aphid-resistant cultivar ICV-12, was investigated. Plants at the seedling, flowering and podding stages of development were infested with five aphid densities consisting of 0, 2, 5, 10 and. 20 aphids per plant and maintained for 22 days. Extended leaf heights of plants and aphid counts were recorded at 7, 12, 17 and 22 days after infestation. Two crop growth parameters (biomass duration and leaf area duration), and two plant yield parameters (number of pods per plant and number of seeds per pod) were recorded. Due to the occurrence of parthenogenesis and changes in population dynamics during infestations, aphid densities were converted into cumulative cowpea aphid-days, to facilitate data analyses and interpretation. ANOVA indicated that there was significant (P=s 0.05) difference in aphid-day accumulations between the two cultivars when infested at the seedling stage. Accumulations on cv. ICV-1 were greater than on cv. ICV-12. However, no such differences between the cultivars were detected when plants were infested at flowering and podding stages. Therefore, the seedling stage was used for comparisons of the impact of cowpea aphid-days on the growth and yield parameters of the two cultivars. At the 95% confidence intervals, ICV-12 plants were consistently taller than ICV-1 plants. Infested ICV-1 seedlings showed stunting and other growth deformities which were not observed on ICV-12 plants. Regression analyses revealed substantial reductions in the growth and yield parameters of ICV-1 relative to ICV-12. Overall, cowpea aphid-days provided a convenient and reliable method for studying the aphid population dynamics and the subsequent impact on plant growth and yield performance.  相似文献

An experiment was conducted under greenhouse conditions to evaluate the effect of mineral nitrogen on N2 fixation of two cultivars of Phaseolus vulgaris L., Puebla 152 and Negro Argel. Nitrogen application was 0, 2.5, 12.5 and 25 mg N Kg–1 of a vermiculite-sand-mixture at planting time. Shoot and root growth were elevated by nitrogen application at all growth stages. During vegetative growth (V 5) nodule dry weight and nitrogenase activity (acetylene reducing activity) per plant were reduced by nitrogen supply in both cultivars, but less in Negro Argel than in Puebla 152. At later stages nodulation in nitrogen-treated Puebla 152 did not differ from that in non-treated plants, whereas increased nodule number was found in Negro Argel at high nitrogen levels. The influence of mineral N on the total amount of nitrogen fixed in the two bean cultivars was only slightly different.  相似文献

Diuraphis noxia infestations were monitored in early winter and winter wheat during 1983–1985. The aphid population increase was logarithmic in all three years and on both cultivars 100 % infestation levels were recorded. The period of exponential increase in both percentage infestation and in the number of D. noxia per stem took place when the plants had reached the stem elongation stage. The mean number recorded at peak population levels was 150–160 aphids per stem. During the period of plant senescence numbers dropped rapidly. Large reductions in yield were recorded on all infested plants. Single-plant data showed that yield reductions in the early winter cv. Betta were related to the duration of infestation. Relative to sprayed controls, reduction in kernel mass ranged from 25–80% and in 1000-kernel mass from 15–45%. However, the yield reductions of the faster-growing cv. Flamink, appeared to be dependent on the number of aphids per plant during the later growth stages and not the duration of infestation.  相似文献

Six rice cultivars showing various types of resistance or susceptibility to Pyricularia oryzae in the field were compared under controlled environmental conditions. The resistance of the cultivars with adult-plant resistance was race-specificat early growth stage. On all cultivars tested, blast infection became increasingly reduced on either leaves of adult plants or older leaves, as observed in rice plants of different leaf stages infected with different individual races. Their increase inhost resistance was marked by an apparent transition in infection type and reduced blast severity depending on leaf age and developmental stage of plants. The ranking of disease severity of the adult-plantresistant cultivars to different races was constant during plant development, whereas that of the susceptible cultivars was differential. It is suggested that different degrees of resistance in rice cultivars may exist in seedlings and be consistently maintained during plant development, probably becoming more distinct as rice plants mature.  相似文献

The influences of culture period on growth, plant survival rate and content of phenolic compounds were investigated in vitro and in acclimatized field-grown plants of Hypericum polyanthemum. The growth kinetics of micropropagated plantlets cultured in MS modified medium and the concomitant transference to ex vitro conditions showed that cultures achieved maximum biomass and yield of bioactive compounds after 12 weeks of in vitro growth, with field-grown plants displaying the same survival pattern. Differences in yield among plants cultured in vitro for 8 and 12 weeks that were acclimatized and followed over two years showed that the physiological age of the in vitro cultures influenced biomass production. However, benzopyrans and total phenolic compounds (TPC) contents did not vary significantly, with the exception of the 5-hydroxy-6-isobutyryl-7-methoxy-2,2-dimethyl-benzopyran (HP3) concentration in the reproductive parts, which was higher in the plants grown in vitro for 12 weeks over the two years of the study. All analyzed plant parts from the spring harvest accumulated lower benzopyran levels than plants harvested after 18 weeks of growth in both treatments, except for the levels of 6-isobutyryl-5,7-dimethoxy-2,2-dimethyl-benzopyran (HP1) and 7-hydroxy-6-isobutyryl-5-methoxy-2,2-dimethyl-benzopyran (HP2) in the new vegetative parts of the plants, which did not vary. The concentration of TPC, which was detected at low levels in the old vegetative parts in both treatments, was not altered in other plant parts. The information provided by this work will help structure plant growth and collection periods designed to optimize the yield of each required bioactive metabolite.  相似文献

Root density distribution of plants is a major Indicator of competition between plants and determines resource capture from the solh This experiment was conducted in 2005 at Anyang, located in the Yellow River region, Henan Province, China. Three cotton (Gossyplum hlrsutum L.) cultivars were chosen: hybrid Btcultlvar CRI46, conventional Btcultlvars CRI44 and CRI45. Six planting densities were designed, ranging from 1.5 to 12.0 plants/m^2. Root parameters such as surface area, diameter and length were analyzed by using the DT-SCAN Image analysis method. The root length density (RLD), root average diameter and root area Index (RAI), root surface area per unit land area, were studied. The results showed that RLD and RAI differed between genotypes; hybrid CRI46 had significantly higher (P 〈0.05) RLD and RAI values than conventlonal cultlvars, especially under low planting densities, less than 3.0 plants/m^2. The root area index (RAI) of hybrid CRI46 was 61% higher than of CRI44 and CRI45 at the flowering stage. The RLD and RAI were also significantly different (P = 0.000) between planting densities. The depth distribution of RAI showed that at Increasing planting densities RAI was Increasingly distributed in the soil layers below 50 cm. The RAI of hybrid CRI46 was for all planting densities, obviously higher than other cultivars during the flowering and boll stages. It was concluded that the hybrid had a strong advantage in root maintenance preventing premature senescence of roots. The root diameter of hybrid CRI46 had a genetically higher root diameter at planting densities lower than 6.0 plants/m^2. Good associations were found between yield and RAI In different stages. The optimum planting density ranged from 4.50 plants/m^2 to 6.75 plants/m^2 for conventional cultlvars and around 4.0-5.0 plants/m^2 for hybrids.  相似文献

The concentrations and composition of free amino acids in phloem sap from two cultivars of oats and barley, both susceptible to the aphid Rhopalosiphum padi, were determined by means of high performance liquid chromatography. Sap was collected from excised aphid stylets at three developmental stages (seedlings, tillering plants and plants undergoing stem elongation) from plants given or not given fertiliser and grown outdoors. In connection, the growth of individual R. padi nymphs was estimated at the same phenological stages on plants grown in the greenhouse. The content of free amino acids was consistently higher in seedlings than in plants at the early tillering stage. Only in seedlings did the addition of fertiliser increase amino acid levels. Barley phloem sap contained more free amino acids than that of oats when fertiliser was added and at later developmental stages. Phloem sap of oats and barley showed similar patterns in their composition of free amino acids at the seedling stage, but as the plants grew older the patterns became increasingly different. Plants given fertiliser had higher amounts of dicarboxylic amino acids (glutamic and aspartic acid) than unfertilised plants. The concentrations of γ-amino butyric acid, glycine, histidine, and methionine were very low in all treatments. The relative growth rates of R. padi nymphs were low when amino acid content was low and vice versa. The results are discussed in relation to host plant suitability and plant resistance mechanisms.  相似文献

The bacterial diversity and population dynamics in the rhizosphere of two maize cultivars (Nitroflint and Nitrodent) grown in tropical soils was studied, by traditional cultivation techniques and 16S rRNA gene-based molecular analysis of DNA directly extracted from soil and rhizosphere samples. Rhizosphere and soil samples were taken at three different plant growth stages. Total aerobic bacterial counts were determined. Fingerprints of the most dominant bacterial population were generated by TGGE separation of 16S rRNA gene fragments amplified from total community DNA using eubacterial specific primers. To reduce the complexity of TGGE fingerprints or to analyse less abundant populations, primers specific for different phylogenetic groups have been used. A comparison of the cfu obtained for rhizosphere of both cultivars indicated significant differences only for rhizosphere and soil samples taken 40 days after sowing. However, a comparison of TGGE patterns indicated that the composition of the bacterial community analysed at different plant growth stages for both cultivars was similar. A comparison of -, -proteobacterial and actinomycete TGGE patterns of both cultivars confirmed this observation. The eubacterial TGGE profiles reflected strong seasonal population shifts in the bacterial rhizosphere community of both maize cultivars which could be also observed in the TGGE patterns of - and -proteobacteria and to a lesser extent for actinomycetes. The rhizosphere effect was much more pronounced for young roots compared to samples taken from mature maize plants. The rhizosphere fingerprints showed a reduced complexity for young plants with up to five dominating bands while for mature plants patterns similar to those of soil were observed. Sequencing of dominant clones indicated that the dominant population found at all plant growth stages can be assigned to Arthrobacter populations.  相似文献
