Despite large genetic differentiation among neighbouring populations of many freshwater zooplankton species, a macrogeographical homogeneity of allozyme variation is generally observed. A study on breeding systems in Scandinavian populations of Daphnia pulex suggested a latitudinally related cline in breeding system with both diploid cyclic parthenogens and diploid obligate parthenogens at the latitude of 60-61 degrees N. Variation at neutral markers may be more affected by selection at linked loci in such species than in strictly sexual species. In this paper I present a study of variation at five microsatellite loci in a total of 34 populations from small ponds and rockpools on both sides of the Baltic Sea at 60-61 degrees N. Two major groups, which may represent different species of the D. pulex complex, are defined with the microsatellites. Neighbouring populations show both similar and well differentiated genetic composition. Populations separated by larger geographical distances show only a large differentiation and a macrogeographic pattern. The large differentiation observed at small distances can be explained with small effective population size: variation at the microsatellite loci has been shaped by population bottlenecks followed with expansion in size, and possibly by selection. No conclusive evidence is found for obligative parthenogenesis.  相似文献

Phenotypic plasticity is one major source of variation in natural populations. Inducible defences, which can be considered threshold traits, are a form of plasticity that generates ecological and evolutionary consequences. A simple cost-benefit model underpins the maintenance and evolution of these threshold, inducible traits. In this model, a rank-order switch in expected fitness, defined by costs and benefits of induction between defended and undefended morphs, predicts the risk level at which individuals should induce defences. Here, taking predator-induced morphological defences in Daphnia pulex as a threshold trait, we provide the first comprehensive investigation into the costs and benefits of a threshold trait, and how they combine to reflect fitness and predict the switchpoint at which induction should occur. We develop reaction norms that show genetic variation in switchpoints. Further experiments show that induction can confer a survival benefit and a cost in terms of lifetime reproductive success. Together, these two traits combine to estimate expected fitness and can predict the switchpoint between an undefended and a defended strategy. The predictions match the reaction norm data for clones that experience these costs and benefits, and correspond well to independent field data on induction. However, predictions do not, and cannot, match for clones that do not gain a benefit from induction. This study confirms that a simple theory, based on life history costs and benefits, is a sufficient framework for understanding the ecology and evolution of inducible, threshold traits.  相似文献

In this laboratory experiment, swarms of D. pulex were artificially created by placing thirty individuals in small chambers containing 130 ml of water. The swimming behaviour of the animals was measured using a video camera and a programme to digitize observations. The D. pulex swimming in water with fish kairomones had a more uniform swimming speed compared with animals in control water. If aggregated prey individuals benefit from a reduced predation risk, and this risk is further reduced by uniformity of swarm members, the uniformity of swimming speed can be interpreted as an behavioural adjustment to minimize the vulnerability to predation. Daphnia pulex is a keystone species for aquatic habitats and an ecological/evolution model organism. Although significant progress has been made on characterizing its genome, the D. pulex proteome remains largely uncharacterized partially due to abnormally high protein degradation during homogenization and emphasis on genomic analysis. In this study, various sample preparation and mass spectrometry acquisition methods are performed for the purpose of improving D. pulex proteome exploration. Benefits for employing both in‐gel and in‐solution methods of trypsin digestion are observed. Furthermore, acquisition methods employing ion mobility separation greatly increase peptide identification and more than doubled the proteome coverage. Bioinformatic analysis suggests that mitochondrial and hydrolytic activities are enriched in D. pulex compared to closely related invertebrates or Homo sapiens. Also, novel D. pulex proteins possessing putative genome modifying functional domains are identified. Data are available via ProteomeXchange with identifier PXD008455.  相似文献

Laboratory populations of cloned Daphnia pulex at three population phases (growing phase, density peak, declining phase) were exposed to the insecticide carbaryl at 15 μg L?1, which killed juveniles but not adults. 2. The population decline was largest and recovery was slowest when the chemical was applied at the density peak, indicating that the population was most sensitive to the chemical at that stage. Populations in the declining phase were least affected by the treatment. 3. We propose that food condition, which varies with population density, was a major factor affecting vulnerability to carbaryl. In that case, Daphnia populations in lakes would be most vulnerable to toxic chemicals during food‐limited condition such as the spring clear‐water phase.  相似文献

  1. Organisms in the wild are faced with multiple threats and a common response is a change in behaviour. To disentangle responses to several threats, we exposed two differently sized species of the freshwater invertebrate Daphnia to solar ultraviolet radiation (UVR) and predation from either moving pelagic or benthic ambush predators.
  2. Using an advanced nanotechnology-based method, we tracked the three-dimensional movements of those mm-sized animals at the individual level. Each behavioural trial was performed both under conditions resembling night (no UVR) and day (UVR) and we examined patterns of the depth distribution and swimming speed by Daphnia across three treatments: no predator (control); bottom-dwelling damselfly (Calopteryx sp.); and fish (stickleback, Pungitius pungitius) predators. We also quantified the actual predation rate by the two predators on the two Daphnia species, Daphnia manga and Daphnia pulex.
  3. We show that individual Daphnia are able to identify predators with different feeding habitats, rank multiple and simultaneously occurring risks and respond in accordance with the actual threat; complex responses that are generally associated with larger animals.
  4. In a broader context, our results highlight and quantify how a cocktail of everyday threats is perceived and handled by invertebrates, which advances our understanding of species distribution in space and time, and thereby of population dynamics and ecosystem function in natural ecosystems.

The distribution of the planktonic crustacean Daphnia pulex was tested in a ring-formedflow-through chamber divided into eleven sections. Thedistribution of the animals under both homogeneousfood conditions and in a food gradient were studied.The distribution of repeated registrations of singleanimals was randomly distributed in a homogeneous,low food environment. Single daphnids exposed to afood gradient tended to reside during 88% of the timeat the highest or next highest food concentration,suggesting that daphnids can detect food gradients.When a group of approximately 100 co-occurring animalswere given low homogeneous food conditions, they alsotended to be randomly distributed. In one out of threecases, however, they were slightly less aggregatedthan expected from a random distribution. Exposed toa food gradient (0.5–0.0015 mg C l-1), thedistribution of the daphnids approximated the idealfree distribution. A very high maximum density in thefood gradient (2 mg C l-1) resulted in less strongaggregations than expected from the ideal freedistribution.  相似文献

1. The influences of dietary phosphorus (P) and food concentration on the calcium (Ca) balance in Daphnia magna were examined in this study at two different ambient Ca concentrations (0.5 and 10 mg Ca L?1). Daphnia were grown by feeding the young adults differentially under contrasting dietary P conditions [molar C : P ratio = c. 900 and c. 90 as low P (LP) and high P (HP), respectively], ambient Ca concentrations [0.5 mg and 10 mg Ca L?1 as low Ca (LCa) or high Ca (HCa), respectively] and food levels [0.15 or 1.5 mg C L?1 as low food (LF) or high food (HF), respectively] for 5 days. 2. The specific Ca contents of daphniids (1.9–6.5% of dry weight?1) increased with increasing Ca concentration, food level and dietary P content, although the food level did not affect the Ca content in the HPHCa treatment. A radioactive tracer method showed that the food level did not affect the influx of Ca from the water under LP conditions, but the Ca influx under HP conditions doubled with a HF level. A LP condition also led to a decrease in Ca influx with a HF level. 3. During the 3 days of efflux, generally only a small proportion of Ca (2.6–3.3%) was retained by the daphniids, but this retention increased (14–23%) under low ambient Ca concentrations and under P‐limitation. Excretion was the most important pathway for Ca loss (accounting for 50–60% of body Ca), followed by moulting (20–47%), but the relative contribution of these two pathways (excretion and moulting) did not vary among all the different treatments. The absolute loss of Ca through excretion and moulting, on the contrary, differed with different ambient Ca concentrations and dietary P conditions. A HF level led to an increase in the loss rates in most cases. 4. Our study strongly suggested that there is an interaction between an essential metal (Ca) and macronutrients (C and P) in freshwater crustaceans with HCa and P contents. The results imply that variation in environmental nutrient conditions may change the Ca budget in crustaceans and may affect the dynamics of Ca in the epilimnion of freshwaters.  相似文献

Short-term toxicity tests with Daphnia magna are reasonably reproducible. The sensitivity of Daphnia pulex, Daphnia cucullata and Daphnia magna proved to be about the same in short-term tests with 15 different chemical compounds.Reproduction toxicity experiments with Daphnia magna can easily be carried out within three weeks. Summary Esterase patterns were examined in three populations of Daphnia pulex. The total number of bands showing esterase activity was 17. Three major genes Est-1, Est-2, Est-3 controlling esterase synthesis were identified and genetically studied. These genes were found to be located in the same linkage group. It was shown that two or three homologous chromosomes differing in sets of esterase alleles predominantly occur in the populations considered.  相似文献

Genetic variants of mitochondrial DNA at the individual (heteroplasmy) and population (polymorphism) levels provide insight into their roles in multiple cellular and evolutionary processes. However, owing to the paucity of genome-wide data at the within-individual and population levels, the broad patterns of these two forms of variation remain poorly understood. Here, we analyze 1,804 complete mitochondrial genome sequences from Daphnia pulex, Daphnia pulicaria, and Daphnia obtusa. Extensive heteroplasmy is observed in D. obtusa, where the high level of intraclonal divergence must have resulted from a biparental-inheritance event, and recombination in the mitochondrial genome is apparent, although perhaps not widespread. Global samples of D. pulex reveal remarkably low mitochondrial effective population sizes, <3% of those for the nuclear genome. In addition, levels of population diversity in mitochondrial and nuclear genomes are uncorrelated across populations, suggesting an idiosyncratic evolutionary history of mitochondria in D. pulex. These population-genetic features appear to be a consequence of background selection associated with highly deleterious mutations arising in the strongly linked mitochondrial genome, which is consistent with polymorphism and divergence data suggesting a predominance of strong purifying selection. Nonetheless, the fixation of mildly deleterious mutations in the mitochondrial genome also appears to be driving positive selection on genes encoded in the nuclear genome whose products are deployed in the mitochondrion.  相似文献

Laboratory microcosm experiments tested the intermediate disturbance hypothesis, which states that the highest level of diversity (e.g. species diversity) will be maintained at intermediate scales of disturbance. The effects of disturbance on the maintenance of clonal diversity and on competitive interactions among clones of the obligately parthenogenetic freshwater cladoceran, Daphnia pulex were examined. No significant effect of disturbance size (i.e. dilution volume) on clonal diversity was noted. However, frequency of disturbance had a pronounced effect on clonal diversity, with the highest clonal diversity maintained at low to intermediate disturbance frequencies. Competitive hierarchies among clones were often invariant within a given experiment. Generally, one or two clones dominated, with several less abundant clones persisting throughout an experiment. Results suggest that low to intermediate disturbances could be important in the maintenance of genetic variation in natural populations (i.e. through pre-emption of competitive exclusion between genotypes). This could have a direct bearing on the maintenance of both intra- and interspecific diversity.  相似文献

In order to examine the usefulness of detoxifying genes as molecular markers in different chemical environments, isolation of cytochrome P450 genes (CYPs) belonging to the CYP4 family was performed in different samples from two subalpine populations of Daphnia pulex. The use of degenerate primers allowed us to isolate seven cDNAs. Four of them were assigned to the CYP4C subfamily, and were closely related to previously isolated crustacean CYP4s while the others were assigned to new CYP4AN and CYP4AP subfamilies. Expression studies, using semiquantitative polymerase chain reaction (PCR) followed by Southern blot hybridization with specific probes revealed differences in CYP4C32 and CYP4AP1 expressions between the two populations, which differ in the polyphenol richness of the vegetation surrounding their aquatic habitat. Further exposure to toxic dietary polyphenols showed different CYP induction patterns. Taken together, these preliminary results suggest a possible involvement of CYP4s in the ecological differentiation of subalpine D. pulex populations related to the polyphenol richness of the environmental vegetation. CYP4s may thus be considered as possible molecular markers in aquatic environmental bioreporting.  相似文献

Laboratory experiments tested the hypothesis that a toxic strain of Microcystis aeruginosa decreases the ability of Daphnia pulex to interfere with Keratella cochlearis. To test a variety of conditions, juvenile and adult Daphnia were exposed to the cyanobacterium for different time periods prior to, and during the experiments. Adult Daphnia not only suppressed rotifers over successive two-day intervals, but also had a significant impact within a 24-hour period. However, the presence of Microcystis (5 × 105 cells ml–1) decreased the Daphnia effect in both experiments. Although juvenile Daphnia also significantly suppressed Keratella population growth, the presence of Microcystis (105 and 5 × 105 cells ml–1) caused a significant reduction in daphniid body size and decreased the ability of both nonacclimated and acclimated daphniids to suppress rotifers. Keratella inhalation and mortality are positively correlated with filtering rates and body size of Daphnia. Therefore, the feeding rates and size structure of a Daphnia population will determine its potential to interfere with vulnerable rotifers. In all experiments the presence of Microcystis significantly decreased the ability of Daphnia to interfere with this rotifer despite the fact that Keratella was also inhibited. In the field this effect might be augmented if Microcystis colonies are more easily ingested by cladocerans than by the rotifers.  相似文献

Whether exotic species invade new habitats successfully depends on (i) a change in the invaded habitat that makes it suitable for the invader and (ii) a genetic change in the invading taxon that enhances its fitness in the new habitat, or both. We dissect the causes of invasions of Swiss lakes, north of the Alps, by Daphnia galeata (a zooplankter typical of eutrophic lakes, e.g. those south of the Alps, which are also warmer) by comparing the fitness performance of eight geographically distributed clones that were fed algal-food typical of oligotrophic versus eutrophic conditions at two temperatures. Daphnia longispina, native to oligotrophic Swiss lakes, served as a reference. Daphnia galeata requires eutrophic food to persist, whereas D. longispina survives and grows on oligotrophic food but does even better on eutrophic food. Invasion by D. galeata is further explained because invading clones from the north perform better on eutrophic food and at cooler temperatures than native clones from the south, suggesting a local response to countergradient selection. Our data support the hypothesis that populations of the invader in northern lakes are dominated by well-adapted genotypes. Our results illustrate how environmental change (i.e. eutrophication) and local adaptation can act together to drive a successful invasion.  相似文献

We investigate the role of recombination in transposable element (TE) proliferation in the cyclical parthenogen, Daphnia pulex. Recombination provides a mechanism by which the rate of both TE gain and loss can be accelerated, a duality that has long intrigued many biologists interested in the influence of sex on mutation accumulation. We compared TE loads among populations of D. pulex where sex occurs regularly (cyclical parthenogens or ‘sexuals’) with those in which the ability to reproduce sexually has been completely lost (obligate ‘asexuals’) for six different families of DNA transposons. Transposon display assays showed that sexuals have more TEs than asexuals, contrary to the expectations under Muller''s ratchet but consistent with the idea that sex facilitates TE spread. Sexuals also exhibit higher insertion site polymorphism among lineages, as predicted because recombination accelerates rates of loss and gain. Asexuals, however, have proportionally more singletons (loci occupied in a single isolate), which differs from previous studies where selfing and outcrossing were used as a proxy for high and low recombination. Our multi-element survey reveals that the impact of sex on TE proliferation is consistent among different Class II TE families and we discuss the genomic consequences of different reproductive strategies over long time periods.  相似文献

Using Southern blot analysis, we have characterized restriction fragment patterns of a transposable element, Pokey, in obligately and cyclically parthenogenetic populations of the cladoceran crustacean Daphnia pulex. We show that the element is most likely active in cyclically parthenogenetic populations but is, for the most part, inactive in obligate parthenogens. This result is consistent with theory suggesting that transposable element dynamics are likely to change with a change in reproductive mode. Such changes could have important consequences for the long-term evolutionary potential of obligate parthenogens and may also be informative with regard to the underlying mechanisms that regulate transposable element frequencies in sexual organisms. Seasonal variation of egg size and number was examined in a Daphnia pulex population inhabiting a vernal pond. In this population, size at maturity declines at midseason, probably as an adaptive response to size-selective predation by larvae of the salamander Ambystoma. The larger early season individuals produce more and larger eggs than the smaller late season individuals. Age at maturity does not vary between seasons. Laboratory experiments indicate that temperature may affect egg size, egg number and size at maturity. However, field data suggest that temperature accounts for only a small fraction of the total variation in egg size and number. Indirect measures of nutrition indicate that food limitation does not cause the seasonal decline in egg size and number. The seasonal change in reproductive traits is well correlated with changes in invertebrate and vertebrate predation. Examination of predator feeding preferences and their impact on Daphnia mortality indicate that variation of reproductive traits is most likely a complex adaptation to changing predation regimes.  相似文献

作为入侵物种,北美水貂(Neovison vison)在欧洲引起了一系列生态问题,侵占了欧亚水獭(Lutra lutra)的生态空间,其入侵性对当地生物多样性和生态系统构成了严重威胁。水貂引入我国东北地区已有70多年的历史,然而国内对其野外种群却鲜有研究。掌握水貂种群的入侵范围、入侵影响因素以及与本地具有相似生态位的欧亚水獭之间的竞争关系,对水貂的入侵管理和东北地区的生物多样性保护具有重要意义。本研究利用实地调查和文献资料获取的分布信息,通过集合模型识别水貂和水獭的潜在分布区,评估水貂对水獭在地理空间上的入侵风险,并通过主成分分析(principal component analysis,PCA)评估其生态位重叠和影响因素。结果表明:(1)我国东北地区水貂的潜在分布区面积为61,944.57 km2,水獭的潜在分布区面积为83,590.94 km2,两者重叠区域面积为50,544.21 km2,占水獭潜在分布区面积的60.47%;(2)从各省分布情况来看,黑龙江省水獭受水貂入侵的风险最高,潜在分布区重叠的比例达到78.9...  相似文献

The presence of plankton predators may induce altered morphology in their potential prey. To date, the mechanism of induction and adaptive value of such defensive responses have been examined in the laboratory. This study investigated the morphological defense structures induced by the invertebrate predator Chaoborus in two coexisting Daphnia species, D. pulex and D. rosea, in the field. In Piscivore Lake (Gr?fenhain, Germany), continuous and intense biomanipulation had led to near elimination of planktivorous fish and greatly increased abundances of Chaoborus (up to >10 larvae l–1). Here, the density of Chaoborus was manipulated within the lake by an enclosure/exclosure setup and resulting morphological responses of Daphnia spp. were investigated in situ. Three replicate enclosures (4.6 m3) contained no Chaoborus (predator exclusion bags), whereas Chaoborus entered three others at ambient densities (predator enclosures). In both species of Daphnia, formation of neckteeth and elongation of the tail spine were recorded in the predator enclosures, but not in the predator exclusion treatments. Additionally, D. rosea responded to predator inclusion with an increase of the size at first reproduction. Despite the induced defense structures, the presence of Chaoborus caused increased mortality of both Daphnia species. In addition, Chaoborus affected the coexistence of the two populations of Daphnia by causing higher relative mortality in D. rosea. Neckteeth formation was always more pronounced in D. pulex than in D. rosea of the same size. Neckteeth were induced specifically in vulnerably sized juvenile instars of D. pulex, but were not found in all vulnerable instars of D. rosea. In D. rosea, neckteeth were few or absent in the ephippial hatchlings, and neckteeth formation ceased before juveniles reached a body size outside the range that larger larval stages of Chaoborus could ingest. This study provides the first experimental demonstration in the field of the inducibility of morphological defense structures in Daphnia at ambient densities of Chaoborus larvae, and quantifies these in situ responses. This expands on earlier observations of a correlation between predator density in the field and the expression of neckteeth in Daphnia. The term ”maximum size for neckteeth formation” (MSNF) is defined as the limit in body size above which no production of neckteeth was evident. This limit was used to distinguish the size classes of Daphnia that show a sensitive response to Chaoborus kairomone. This new term may be used for further comparisons among species and among different types of predator-induced responses as well as for the evaluation of the adaptive value of defense structures.
