The jasmonate ZIM-domain (JAZ) proteins are repressors that function in the regulation of plant growth, development, and response to stimulation of different signals in the JA signaling pathway. Erigeron breviscapus is characteristic of sporophyte self-incompatibility (SSI). However, whether JA signaling is involved in regulation of development processes in E. breviscapus is unclear. In this study, the JAZ homolog EbJAZ1 was isolated and characterized from E. breviscapus. EbJAZ1 was localized to the nucleus, and expressed in roots, stems, leaves and flowers. Ectopic expression of EbJAZ1 in Arabidopsis resulted in shorter filament and silique length, and lower seed fertility. In addition, MeJA-induced root growth inhibition was compromised in transgenic plants. Further qRT-PCR analysis indicated that expression patterns of marker genes for VSP1, VSP2, JAZ1, JAZ5, JAZ8, JAZ10, MYC2, and bHLH17 were downregulated in transgenic plants compared to wild-type, suggesting that EbJAZ regulates the development of flower organs, seed fertility, and primary root growth through the JA signaling pathway. Thus, our results indicate that EbJAZ1 is one of the important regulators possibly involved in SSI and other developmental processes in Erigeron breviscapus.

The phytohormone jasmonoyl-L-isoleucine (JA-Ile) regulates many stress responses and developmental processes in plants. A co-receptor complex formed by the F-box protein Coronatine Insensitive 1 (COI1) and a Jasmonate (JA) ZIM-domain (JAZ) repressor perceives the hormone. JA-Ile antagonists are invaluable tools for exploring the role of JA-Ile in specific tissues and developmental stages, and for identifying regulatory processes of the signaling pathway. Using two complementary chemical screens, we identified three compounds that exhibit a robust inhibitory effect on both the hormone-mediated COI–JAZ interaction and degradation of JAZ1 and JAZ9 in vivo. One molecule, J4, also restrains specific JA-induced physiological responses in different angiosperm plants, including JA-mediated gene expression, growth inhibition, chlorophyll degradation, and anthocyanin accumulation. Interaction experiments with purified proteins indicate that J4 directly interferes with the formation of the Arabidopsis (Arabidopsis thaliana) COI1–JAZ complex otherwise induced by JA. The antagonistic effect of J4 on COI1–JAZ also occurs in the liverwort Marchantia polymorpha, suggesting the mode of action is conserved in land plants. Besides JA signaling, J4 works as an antagonist of the closely related auxin signaling pathway, preventing Transport Inhibitor Response1/Aux–indole-3-acetic acid interaction and auxin responses in planta, including hormone-mediated degradation of an auxin repressor, gene expression, and gravitropic response. However, J4 does not affect other hormonal pathways. Altogether, our results show that this dual antagonist competes with JA-Ile and auxin, preventing the formation of phylogenetically related receptor complexes. J4 may be a useful tool to dissect both the JA-Ile and auxin pathways in particular tissues and developmental stages since it reversibly inhibits these pathways.One-sentence summary: A chemical screen identified a molecule that antagonizes jasmonate perception by directly interfering with receptor complex formation in phylogenetically distant vascular and nonvascular plants.  相似文献

Although a wealth of information is available regarding resistance induced by plant growth-promoting rhizobacteria (PGPR), not much is known about plant growth-promoting fungi (PGPF). Hence, the goal of the present research was to provide more information on this matter. In Arabidopsis thaliana L., root colonizing PGPF Penicillium sp. GP16-2 or its cell free filtrate (CF) elicited an induced systemic resistance (ISR) against infection by Pseudomonas syringae pv. tomato DC3000 (Pst), leading to a restriction of pathogen growth and disease development. We demonstrate that signal transduction leading to GP16-2-mediated ISR requires responsiveness to JA and ET in a NPR1-dependent manner, while CF-mediated ISR shows dispensability of SA, JA, ET and NPR1-dependent signaling (at least individually). In addition, root colonization by GP16-2 is not associated with a direct effect on expression of known defense-related genes, but potentiates the activation of JA/ET-inducible ChitB, which only becomes apparent after infection by Pst. However, CF-mediated ISR was partly associated with the direct activation of marker genes responsive to both SA and JA/ET signaling pathways and partly associated with priming, leading to activation of JA-/ET-inducible ChitB and Hel genes. These suggest that CF may contain one or more elicitors that induce resistance by way where at least SA, JA and ET may play a role in defense signaling in Arabidopsis. Therefore, defense gene changes and underlying signaling pathways induced by Penicillium sp. GP16-2 root colonization and its CF application are not the same and only partially overlap.  相似文献

The native auxin, indole-3-acetic acid (IAA), is a major regulator of plant growth and development. Its nonuniform distribution between cells and tissues underlies the spatiotemporal coordination of many developmental events and responses to environmental stimuli. The regulation of auxin gradients and the formation of auxin maxima/minima most likely involve the regulation of both metabolic and transport processes. In this article, we have demonstrated that 2-oxindole-3-acetic acid (oxIAA) is a major primary IAA catabolite formed in Arabidopsis thaliana root tissues. OxIAA had little biological activity and was formed rapidly and irreversibly in response to increases in auxin levels. We further showed that there is cell type–specific regulation of oxIAA levels in the Arabidopsis root apex. We propose that oxIAA is an important element in the regulation of output from auxin gradients and, therefore, in the regulation of auxin homeostasis and response mechanisms.  相似文献

茉莉酸信号分子参与植物生长发育众多生理过程的调控,尤其是作为环境信号分子能有效地介导植物对生物及非生物胁迫的防御反应。迄今已知具有信号分子生理功能的至少包括茉莉酸(jasmonic acid,JA)以及茉莉酸甲酯(methyl jasmonate,Me JA)和茉莉酸-异亮氨酸复合物(jasmonoyl-isoleucine,JA-Ile)等茉莉酸衍生物,统称为茉莉酸类化合物(jasmonates,JAs)。从环境信号分子角度介绍了茉莉酸信号的启动(环境信号感知与转导、茉莉酸类化合物合成)、传递(局部传递、维管束传输、空气传播)和生物学功能(茉莉酸信号受体、调控的转录因子、参与的生物学过程)。  相似文献
