Gladiolus bud explants propagated in agitated liquid medium, in the presence of the growth retardants daminozide, ancymidol, paclobutrazol or Majic, proliferated into massive bud aggregates without leaves. The buds developed into protocorms and after subculture to agar-solidified medium formed cormlets 8–10 mm in diameter. The corms were not dormant and, after transplanting to pots in the greenhouse, developed into plantlets, 5–6 months after explant isolation.  相似文献

The effects of AMO-1618, AY-9944 and SKF-7997 on the growth of Nicotiana tabacum and on the biosynthesis of acidic and neutral isopentenoids in the seedlings were examined. All three compounds significantly retarded stem elongation but not dry wt of the plants. Incorporation of radioactivity from [2- 14C]-mevalonate into acidic and neutral isopentenoids showed a direct relationship between the radioactivity in the two fractions and stem growth. All three compounds inhibited steps before as well as after squalene formation, but the acylic and polycyclic isopentenoids which accumulated as a result of the inhibition were not necessarily the same in each of the treatment groups. This indicates that stem growth is probably influenced not by a single product of isopentenoid biosynthesis, but rather by several products, which may even act independently in their effects on developmental processes.  相似文献

The effects of different growth regulators on induction and growth of callus ofAsparagus densiflorus cv. Sprengeri were studied. Calluses grew more rapidly on Murashige and Skoog basal medium supplemented with 5.4 μM p-chlorophenoxyacetic acid (pCPA) and 4.4 μM 6-benzylaminopurine (BA) (medium 1) as compared to the same medium with 11.3 μM 2,4-dichlorophenoxyacetic acid (2,4-d) and 4.6 μM kinetin (medium 2). Calluses on medium 1 were soft and friable, whereas, compact, hard calluses originated on medium 2. Different concentrations and combinations of BA and/or kinetin were also used to study their effects on shoot regeneration. Kinetin was found to be less effective than BA in the initiation of shoots (1.8 shoots/callus). High numbers of shoots were produced in the presence of 0.4 μM BA alone (3.3 shoots/callus). The addition of ancymidol (5 μM) in MS with 0.4 μM BA enhanced multiplication of shoots (9.8 shoots/explant) and also produced well-developed crowns.  相似文献

The germination of whole seeds of celery (Apium graveolens L.) was inhibited by paclobutrazol, ancymidol and lower concentrations of uniconazole. The growth retardants daminozide, AMO 1618 and chlormequat chloride inhibited the germination of cut seeds only, indicating that the seed coat prevents the penetration of these compounds at the examined concentrations. Application of a mixture of the gibberellins A4 and A7 (GA 4/7) reversed the inhibition of all the examined growth retardants. Cytokinins (artificial or natural) had no effect when applied alone and did not interact with GA4/7 in the light. However, in the dark the cytokinins at some concentrations and GA4/7 had a synergistic effect in reversing the inhibition caused by growth retardants to whole seeds or in promoting the germination of whole seeds. A procedure was developed to reduce premature bolting and flowering in Chinese cabbage grown in the field during the winter. This involved (a) the use of geothermic water as an inexpensive heat source for heating the transplants in the nursery; (b) application of the growth retardants daminozide (Alar) and paclobutrazol (PP333) to the heated plants before removing them to the field; and (c) application of daminozide to the plants in the field. This procedure markedly reduced stem elongation and the length of the internal stem in relation to the head without affecting the size of the head. Furthermore, application of daminozide in the field minimized the severity of tipburn, a common physiological disorder in Chinese cabbage fields. The result of such treatment was the production of high-quality Chinese cabbage heads which otherwise are unobtainable during the winter.Contribution from the Agricultural Research Organization, The volcani Center, Bet Dagan, Israel. The growth of Brassica campestris, Solanum tuberosum L., Lycopersicum esculentum and Brassica junce in the field were promoted by the degraded soybean meal products (DSP). The root hair number of Brassica campestris was increased when 10 l DSP (containing 30 mg peptides + amino acids ml–1) were added to 10 ml plant growth medium. A chemical fertilizer and an acid-hydrolyzed DSP did not show such an effect.  相似文献

The impact of asparagus virus I (AV-I), a potyvirus, and asparagus virus II (AV-II), an ilarvirus, on micropropagation of field-grown asparagus was studied. Apical shoot tips excised from singly or doubly-infected plants were slow to develop roots and had a 15 to 75% reduction in survival in culture, respectively, compared to those excised from virus-free plants. The four virus infection groups were ranked: virus-free >AV-II>AV-I>AV-I & II for capacity of explants to both root and survivein vitro. Micropropagated plants infected with AV-II exhibited slight reductions in fresh and dry weights, with greater reductions associated with infection with AV-I and double infection, compared to the virus-free controls. Eighty-one virus-infected apical shoot tips yielded 7 (8.6%) virus-free clones, as determined by rub inoculation on indicator plants.  相似文献

The effects of growth retardants, 4-hydroxy-5-isopropyl-2-methylphenyltrimethylammonium chloride-1-piperidine carboxylate (AMO-1618 or AMO) and 2-chloroethyltrimethylammonium chloride (CCC), applied with and without gibberellic acid (GA3), on -[3H]alanine uptake and leakage from pea (Pisum sativum L.) and betacyanin efflux from beetroot (Beta vulgaris L.) tissue were examined. Both compounds decreased the amount of -[3H]alanine taken up into pea leaf discs, and increased the quantity of radioactive label that subsequently leaked out of this tissue. Efflux of betacyanin from slices of beetroot was also found to be promoted by treatment with CCC or AMO-1618. In no case were these effects reversed by application of GA3. Germination of lettuce seeds cv. May Queen iscompletely prevented either with 10 µM tetcyclacisor with continuous FR illumination. GA3 and the N-substituted phtalimide, AC 94,377, werepartially effective in overcoming tetcyclacis-inducedinhibition but ineffective on photoinhibited seeds. FCcompletely reversed tetcyclacis inhibition and inducedca. 60% germination in continuous FR light. Aninteraction between FC and GA3 (as well asbetween FC and AC 94,377) was evident in stimulationof germination under both inhibitory conditions.Interaction was calculated as a ratio of thepercentage of seeds germinated under the simultaneousaction of stimulators compared to their additiveeffect. This was 2.54 for tetcyclacis- and 2.95 forphotoinhibited seeds. It is concluded that thistype of interaction is promotive synergism. Themagnitude of the interaction was highest if theapplication of FC was delayed after GA3application, and the optimal time lag was 6 h fortetcyclacis-inhibited, or 24 h for photoinhibited seeds.  相似文献

Effects of gibberellins A1, A4/7, A9, A19 and A20 and growth retardants were studied on shoot elongation in seedlings of Salix pentandra L. The growth-retarding effects of CCC and ancymidol were antagonized by all the gibberellins tested. The novel plant growth regulator prohexadione (free acid of BX-112), which is suggested to block 3β-hydroxylation of gibberellins, effectively prevented shoot elongation in seedlings grown under long photoperiod. Initiation of new leaves was only slightly reduced. GA1, but not GA19 and GA20, was active in overcoming the inhibition of stem elongation of seedlings, treated with prohexadione, GA19, GA20 and GA1 are native in S. pentandra , and the results are compatible with the hypothesis that GA1 is active per se in shoot elongation, and that the effect of GA19 and GA20 is dependent on their conversion to GA1.
A mixture of GA4 and GA7 was as active as GA1 in promoting shoot elongation in seedlings treated with prohexadione, while GA9 showed slight activity only when applied at high doses.  相似文献

Aphid-induced reduction of shoot and root growth in Douglas-fir seedlings   总被引:2,自引:0,他引:2  
1. The short‐ and long‐term effects of photosynthate removal by sap‐sucking herbivores on plant growth were examined by experimentally manipulating densities of an aphid Cinara pseudotsugae (Wilson) on 2‐year‐old Douglas‐fir seedlings Pseudotsuga menziesii (Mirb.) Franco under greenhouse conditions. An 18‐week test was conducted to determine short‐term effects. Effects of long‐term aphid feeding were examined by exposing seedlings to aphid feeding for two consecutive growing seasons. A third experiment evaluated the extent of recovery during 1 year following a single season of aphid feeding. At least 35 seedlings were used in each test. 2. Volume and dry weight of both shoots and roots decreased significantly with increasing aphid feeding in all three experiments. 3. The most significant and severe aphid effect was reduced root tissue density, suggesting carbohydrate depletion due to translocation from roots to shoots. 4. There was no sign of recovery, of either root or shoot growth, during the year following one season of feeding. 5. The results of this study indicate that short‐term feeding by aphids can affect plant growth and structure for a relatively long time.  相似文献

The growth retardant chlormequat stimulated microtuber formation by a recalcitrant cultivar of potato (Solanum tuberosum), but reduced microtuber fresh weight in a cultivar that tuberised readily in its absence. Inhibition of microtuber growth by high concentrations of chlormequat was confirmed using a different in vitro system where all cultivars tuberised in the absence of growth retardants.Alternative growth retardants were tested. Daminozide also had a detrimental effect on microtuber fresh weight, but ancymidol and paclobutrazol did not inhibit microtuber growth at the concentrations required for stimulation of tuberisation by recalcitrant cultivars. 目的:研究渐狭蜡蚧菌产生的几丁质酶及其对芦笋茎枯病菌天门冬拟茎点霉的抑制效果。方法:通过乙酰葡萄糖胺法、对硝基苯酚法和活性电泳法测定渐狭蜡蚧菌CGMCC5328的产几丁质酶特性及其粗酶液对芦笋茎枯病菌的抑菌效果。结果:渐狭蜡蚧菌CGMCC5328几丁质酶粗提取液的几丁质降解酶系活性第4 d达到高峰期;几丁质外切酶活性第6 d达到高峰期;活性染色检测到5条几丁质酶谱带,相对分子质量分别为32.9×103、42.1×103、54.1×103、65.6×103和79.6×103。用该几丁质酶粗酶液处理芦笋茎枯病菌,与灭活酶液处理相比较,抑菌效果明显,酶液处理导致菌丝畸形。结论:渐狭蜡蚧菌CGMCC5328产生的几丁质酶对芦笋茎枯病菌有抑制作用。  相似文献

Applications of the growth promotive gibberellins (GAs) GA4 and 2,2-dimethyl GA4, and of C-16,17 endo-dihydro GA5, which is known to promote flowering while inhibiting stem growth in the long-day grass Lolium temulentum, were made to micropropagated plants of Metrosideros collina cv. Tahiti, a highly ornamental cultivar with an intermittent flowering pattern. Gibberellin A4 and 2,2-dimethyl GA4 stimulated vegetative growth both in elongating shoots, and internodes of shoots developing from buds that were quiescent at the time of GA application. Abscission of the apices of expanding shoots, a feature of mature Metrosideros plants, was inhibited by these GAs, the rejuvenation of micropropagated plantlets being enhanced. However, C-16,17 endo-dihydro GA5 differed from GA4 and 2,2-dimethyl GA4 by having no promotive effects on vegetative growth, and no inhibition of apical abscission. Notwithstanding this contrasting effect on vegetative growth, high doses of GA4 or C-16,17 endo-dihydro GA5 similarly reduced flowering on shoots to which either GA was applied. Reduced flowering in response to applied GAs is common in many woody angiosperms, and in this instance was probably the combined result of abortion of developing floral structures in quiescent buds, and a preferential inhibition of bud break for floral buds relative to vegetative buds, particularly by GA4. Finally, both C-16,17 endo-dihydro GA5 and GA4 strongly inhibited bud break in this woody angiosperm, although GA4 could initially stimulate bud break when applied to vegetative buds close to the expansion stage. The above findings, in toto, highlight the sensitivity of Metrosideros to both classes of GA in a variety of growth and development processes.  相似文献

AIMS: The purpose of this Botanical Briefing is to stimulate reappraisal of root growth, root/shoot partitioning, and analysis of other aspects of plant growth under heterogeneous conditions. SCOPE: Until recently, most knowledge of plant growth was based upon experimental studies carried out under homogeneous conditions. Natural environments are heterogeneous at scales relevant to plants and in forms to which they can respond. Responses to environmental heterogeneity are often localized rather than plant-wide, and not always predictable from traditional optimization arguments or from knowledge of the ontogenetic trends of plants growing under homogeneous conditions. These responses can have substantial impacts, both locally and plant-wide, on patterns of resource allocation, and significant effects on whole-plant growth. Results from recent studies are presented to illustrate responses of plants, plant populations and plant communities to nutritionally heterogeneous conditions. CONCLUSIONS: Environmental heterogeneity is a constant presence in the natural world that significantly influences plant behaviour at a variety of levels of complexity. Failure to understand its effects on plants prevents us from fully exploiting aspects of plant behaviour that are only revealed under patchy conditions. More effort should be invested into analysis of the behaviour of plants under heterogeneous conditions.  相似文献

Summary Calluses from five asparagus genotypes G14, G32, G171, G203, and G447 and hybrid Jersey Giant (JG) were incubated at three temperature regimes (24, 27, and 30°C) on embryo induction medium to assess somatic embryo development and conversion to plantlets. The calluses from three genotypes (G14, G32, and G171) were not responsive, failing to produce somatic embryos at any temperature regime. For three responsive genotypes (G203, G447, and JG), both incubation temperature and genotype significantly affected the numbers of somatic embryos produced. The calluses produced the most and the least numbers of total, bipolar, and globular embryos when incubated at 27°C and 24°C, respectively. When incubated at 27°C, G203 produced the highest numbers of total and globular embryos, 178 g−1 callus and 142 g−1 callus, respectively while G447 produced the highest number of bipolar embryos, 77 g−1 callus. Incubation temperature but not genotype significantly affected the conversion of somatic embryos to plantlets. The somatic embryos recovered from the three responsive genotypes incubated at 27°C also converted to plantlets at the highest frequencies, 60–63% of the bipolar embryos and 42–43% of the globular embryos converted to plantlets, while the somatic embryos recovered from the calluses incubated at 24°C converted to plantlets at the lowest frequencies.  相似文献

Addition of abscisic acid (ABA) to the nutrient solution increased the root to shoot ratio of hydroponically-grown cauliflower plants by reducing the dry weight of the shoot and increasing that of the root. At concentrations higher than 10–7 M, ABA increased root branching and root hair formation. Root extension was inhibited in plants kept continuously in solutions containing high ABA concentrations but following removal from the ABA solution root elongation was increased in comparison with plants given no ABA treatment. This elongation was greatest in plants with increased root branching caused by higher ABA concentrations.  相似文献

In field experiments with Japanese mint using different growth retardants, chlormequat chloride (2-chloroethyltrimethylammonium chloride) increased the oil content significantly and inhibited growth to some extent only, whilst ethephon (2-chloroethyl phosphonic acid) at 0.06% concentration significantly decreased growth but had no significant effect on oil content. Among the two oil components studied, menthone content only was significantly increased by 0.06% ethephon whereas the other growth retardants were ineffective. Correlation studies indicated a strong negative relationship of leaf/stem ratio with plant height and herb yield while a strong positive relationship was obtained between the latter two characters.
