Glassy-winged sharpshooter (GWSS), Homalodisca vitripennis (Germar), is a vector of the xylem-inhabitant bacterium Xylella fastidiosa Wells et al., which causes Pierce’s disease of grapevines. Current GWSS control strategies in California, USA include area-wide insecticide applications and mass release of mymarid egg parasitoids, including Gonatocerus ashmeadi Girault. Gas chromatography–mass spectrometry was used to identify (E)-β-ocimene and (E,E)-α-farnesene as volatiles emitted from grapevines on which GWSS had previously fed and oviposited. Attractiveness of female G. ashmeadi to sugar-based formulations containing either (E)-β-ocimene, (E,E)-α-farnesene, or a mixture of both was evaluated using Y-tube olfactometry. When exposed to synthetic formulation containing a mixture of (E)-β-ocimene and (E,E)-α-farnesene vs. blank control, 61% of G. ashmeadi females initially chose the synthetic formulation. After the initial choice for a Y-tube arm, females visited the Y-tube arm connected to the source of formulation more often than it did to the arm connected to a blank control. There was no difference in the female’s time spent in the arm connected to the formulation. When testing formulations containing either (E)-β-ocimene or (E,E)-α-farnesene alone, there was a 1:1 ratio between the proportion of parasitoid’s first choice, visits, and residence time. Results suggest that synthetic formulations containing mixtures of certain plant volatiles may be used to localize GWSS egg parasitoids in vineyard systems. Results are discussed in the context of potential applications in GWSS biological control programs.  相似文献

Grapholita molesta (Busck) (Lepidoptera: Tortricidae) is a notorious pest of many Rosaceae crops worldwide. Enhancement of trap efficiency of its sex pheromone was devised by addition of E-β-ocimene and E-β-farnesene. The addition of E-β-ocimene or E-β-farnesene to sex pheromone increased electroantennogram response of male G. molesta compared to sex pheromone alone. Blend of pheromone and E-β-ocimene or E-β-farnesene in 1:0.1 increased the upwind flight and landing behaviors. Furthermore, field experiments showed that sex lures with E-β-ocimene, or /and E-β-farnesene, enhanced trapping efficiency compared to sex pheromone alone. These results may provide the basis for the development of efficient pest management systems against G. molesta using plant volatiles and insect sex pheromones.  相似文献

The volatiles emitted in vivo by different organs (leaf, bud flower, flower, green and red fruits) of Ziziphus jujuba were analysed using HS-SPME-GC/MS. A total of 144 compounds were identified corresponding to 94.6–99.4% of the whole aroma profile of jujube samples. The main constituents detected were (E, E)-α-farnesene, (E)-β-ocimene, perillene, γ-terpinene, cis-sabinene-hydrate, trans-sabinene-hydrate and 4-terpineol. The SPME analyses of the collected samples showed different patterns of emission and can contribute to understand their ecological interactions and fruit production management.  相似文献

Plants and insects often use the same compounds for chemical communication, but not much is known about the genetics of convergent evolution of chemical signals. The terpene (E)-β-ocimene is a common component of floral scent and is also used by the butterfly Heliconius melpomene as an anti-aphrodisiac pheromone. While the biosynthesis of terpenes has been described in plants and microorganisms, few terpene synthases (TPSs) have been identified in insects. Here, we study the recent divergence of 2 species, H. melpomene and Heliconius cydno, which differ in the presence of (E)-β-ocimene; combining linkage mapping, gene expression, and functional analyses, we identify 2 novel TPSs. Furthermore, we demonstrate that one, HmelOS, is able to synthesise (E)-β-ocimene in vitro. We find no evidence for TPS activity in HcydOS (HmelOS ortholog of H. cydno), suggesting that the loss of (E)-β-ocimene in this species is the result of coding, not regulatory, differences. The TPS enzymes we discovered are unrelated to previously described plant and insect TPSs, demonstrating that chemical convergence has independent evolutionary origins.

This study identifies a novel terpene synthase involved in production of an anti-aphrodisiac pheromone by the butterfly Heliconius melpomene. Maize plants attacked by lepidopteran larvae emit a volatile mixture that consists mostly of the sesquiterpene olefins, (E)-α-bergamotene and (E)-β-farnesene. These volatiles are produced by the herbivore-induced terpene synthase TPS10 and attract natural enemies to the damaged plants. A survey of volatiles in maize lines and species of teosinte showed that the TPS10 products (E)-α-bergamotene and (E)-β-farnesene are consistently induced by herbivory, indicating that release of TPS10 volatiles is a defense trait conserved among maize and its wild relatives. Sequence comparison of TPS10 from maize and its apparent orthologs from four teosinte species demonstrated stabilizing selection on this defense trait. The teosinte volatiles and the enzymatic activity of the apparent TPS10 orthologs were not completely uniform but varied in the ratio of (E)-α-bergamotene to (E)-β-farnesene products formed. We identified a single amino acid in the active center which determines the ratio of (E)-α-bergamotene to (E)-β-farnesene and has changed during the evolution of maize and teosinte species. Feeding experiments with the substrate (Z,E)-farnesyl diphosphate revealed that this amino acid controls the rate of isomerization of the (E,E)-farnesyl carbocation intermediate to the (Z,E)-configuration.  相似文献

Newly hatched Laspeyresia pomonella larvae were attracted to only two of six synthetic stereo-isomers of the acyclic sesquiterpene farnesene. These were (E,E)-α-farnesene and (Z,E)-α-farnesene which together comprise a natural attractant for the insect. Two other α-isomers and two β-isomers had no influence on larval behaviour. The polyphagous planthopper Hyalesthes obsoletus Signoret is considered to be the principal vector of stolbur phytoplasma, which is associated with yellow diseases of grapevine. To explore the possibility of developing novel control strategies, the behavioral responses to six synthetic mixtures and nine single compounds, previously identified from the headspace of Vitex agnus-castus L. (chaste tree) and Urtica dioica L. (nettle), were investigated in Y-tube bioassays. Choice tests revealed differences in the behavioral responses of males and females to the volatiles that they were exposed to. Males were attracted to a mixture containing (E)-β-caryophyllene, 1,8-cineole, (E,E)-α-farnesene, (E)-β-farnesene, and methyl salicylate (mixture 2). The addition of methyl benzoate to this five-compound mixture (mixture 3) did not attract males but elicited positive responses in females. Furthermore, females were attracted to a mixture containing (E)-β-caryophyllene, (E,E)-α-farnesene, (Z)-3-hexenyl acetate, (Z)-3-hexen-1-ol, and benzothiazole (mixture 4), but here addition of methyl salicylate (mixture 5) did not attract females. Neither males nor females showed attractivity or repellency toward the singly tested compounds. This study enhances knowledge on the interaction of insect behaviorally effective constituents in complex plant volatile mixtures. The attractive mixtures of plant volatiles identified suggest the possibility of using them in monitoring and management of H. obsoletus.  相似文献

Abstract

The oriental fruit moth (OFM) is one of the most serious pests of commercial fruit orchards worldwide. Newly planted peach trees in particular, can be very attractive for mated OFM females for oviposition. Samples of airborne host plant volatiles from intact young peach shoot tips and old leaves of the same potted plants were collected and analyzed with a gas chromatograph-mass spectrometer. Chemicals present in young shoot tips, but not in old leaves, were used for field trials. Moth capture by traps with the synthetic chemicals was compared to that of the standard terpinyl acetate (TA) food trap. The TA food trap caught OFM males and mated females, but tested synthetic chemicals trapped only OFM males. We observed that the mixture of (Z)-3-hexenyl acetate: (E)-β-ocimene: (E)-β-farnesene in proportion 1:2:2 attracted OFM males. Further, 1 mg of (E)-β-ocimene, and that of (E)-β-farnesene also attracted OFM males.  相似文献

The exotic wood-boring pest, emerald ash borer (EAB), Agrilus planipennis Fairmaire (Coleoptera: Buprestidae), has been threatening North American ash (Fraxinus spp.) resources, this being recognized since its first detection in Michigan, USA and Ontario, Canada in 2002. Ash trees are killed by larval feeding in the cambial region, which results in disruption of photosynthate and nutrient translocation. In this study, changes in volatile and non-volatile foliar phytochemicals of potted 2-yr-old black ash, Fraxinus nigra Marshall, seedlings were observed in response to EAB larval feeding in the main stem. EAB larval feeding affected levels of six compounds [hexanal, (E)-2-hexenal, (Z)-3-hexenyl acetate, (E)-β-ocimene, methyl salicylate, and (Z,E)-α-farnesene] with patterns of interaction depending upon compounds of interest and time of observation. Increased methyl salicylate emission suggests similarity in responses induced by EAB larval feeding and other phloem-feeding herbivores. Overall, EAB larval feeding suppressed (Z)-3-hexenyl acetate emission, elevated (E)-β-ocimene emission in the first 30 days, but emissions leveled off thereafter, and generally increased the emission of (Z,E)-α-farnesene. Levels of carbohydrates and phenolics increased overall, while levels of proteins and most amino acids decreased in response to larval feeding. Twenty-three amino acids were consistently detected in the foliage of black ash. The three most abundant amino acids were aspartic acid, glutamic acid, glutamine, while the four least abundant were α-aminobutyric acid, β-aminoisobutyric acid, methionine, and sarcosine. Most (16) foliar free amino acids and 6 of the 9 detected essential amino acids decreased with EAB larval feeding. The ecological consequences of these dynamic phytochemical changes on herbivores harbored by ash trees and potential natural enemies of these herbivores are discussed.  相似文献

Four Old World species of Pheidole ants contain different mixtures of farnesene-type hydrocarbons in their poison apparatus, and the mixture is different between the minor and major workers within a species. A bishomofarnesene (C17H28) provides approximately half of the secretion of the Dufour glands of minor workers of Pheidole pallidula. (Z,E)-α-Farnesene constituted 96% of the Dufour secretion of major workers of P. pallidula, but only 20% of that of minors. The Dufour glands of minor workers of Pheidole sinaitica contain a mixture of farnesene homologues with (Z,E)-α-farnesene and the bishomofarnesene also found in P. pallidula predominant. The mixture in major workers was similar but had, in addition, a small amount of (E)-β-farnesene. The Dufour glands of Pheidole teneriffana minors contain chiefly the same bishomofarnesene found in P. pallidula and P. sinaitica while major workers contain (Z,E)-α-farnesene. Pheidole megacephala minor workers contained small amounts of eight farnesenes, while major workers contained essentially no farnesenes. The poison glands of minor workers of P. pallidula contain 3-ethyl-2,5-dimethylpyrazine. No pyrazine compounds were found in the major workers of P. pallidula or the minor workers of P. sinaitica. The poison glands of the major workers of P. sinaitica contained larger amounts of tetra-substituted pyrazines. No pyrazines were found in the poison reservoirs of major or minor workers of P. teneriffana or P. megacephala.  相似文献

The production of quince (Cydonia oblonga Miller) is affected worldwide by codling moth (Cydia pomonella L.), an oligophagous pest. In this study, volatile compounds in healthy and infested fruits were collected from four cultivars of quince: Champion, Portugal, Smyrna and INTA 147. Volatiles were extracted by SPME and analyzed by GC-MS. Up to 30 compounds were identified in the volatile profiles of uninfested quinces, ranging from 90.9% to 98.4% of the total relative abundance. The volatile profile of all four cultivars of quince was similar, with (E,E)-α-farnesene and octanoic acid- ethyl ester as the main components. Infested fruits presented increased levels of (E,E)-α-farnesene, while minor compounds decreased significantly. Champion and Smyrna varieties might be more susceptible cultivars to infestation as their healthy fruits presented the highest contents of (E,E)-α-farnesene and Pear Ester, both codling moth kairomones. The role of these highly recognized kairomones in the chemical ecology of codling moth in quince is discussed.  相似文献

After herbivore damage, many plants increase their emission of volatile compounds, with terpenes usually comprising the major group of induced volatiles. Populus trichocarpa is the first woody species with a fully sequenced genome, enabling rapid molecular approaches towards characterization of volatile terpene biosynthesis in this and other poplar species. We identified and characterized four terpene synthases (PtTPS1-4) from P. trichocarpa which form major terpene compounds of the volatile blend induced by gypsy moth (Lymantria dispar) feeding. The enzymes were heterologously expressed and assayed with potential prenyl diphosphate substrates. PtTPS1 and PtTPS2 accepted only farnesyl diphosphate and produced (−)-germacrene D and (E,E)-α-farnesene as their major products, respectively. In contrast, PtTPS3 and PtTPS4 showed both mono- and sesquiterpene synthase activity. They produce the acyclic terpene alcohols linalool and nerolidol but exhibited opposite stereospecificity. qRT-PCR analysis revealed that the expression of the respective terpene synthase genes was induced after feeding of gypsy moth caterpillars. The TPS enzyme products may play important roles in indirect defense of poplar to herbivores and in mediating intra- and inter-plant signaling.  相似文献

Nineteen host plant volatiles (HPVs) were screened for attractivity to adult codling moth Cydia pomonella (L.) as a fourth component of core blends (3K) including (E,Z)-2,4-ethyl decadienoate, (E)-4,8-dimethyl-1,3,7-nonatriene and acetic acid. Each new quaternary combination was compared with a previously reported attractive bisexual lure (4K), consisting of the 3K blend plus 6-ethenyl-2,2,6-trimethyloxan-3-ol (pyranoid linalool oxide, pyrLOX). All lure evaluations were conducted in apple, Malus domestica (Borkhausen). Several compounds were found to significantly lower total and/or female catches when added to the 3K blend, including (Z)-3-hexenol, (E)-2-hexanal and hexyl butanoate (female and total moths), and (Z)-3-hexenyl acetate and linalool (female moths). Other compounds when added to the 3K blend did not increase or decrease moth catches, including methyl salicylate, (E)-β-ocimene, limonene, β-caryophyllene, butyl hexanoate, farnesol, terpineol, terpinen-4-ol and α-pinene. A few added compounds significantly increased moth catches compared with the 3K blend, including β-pinene (male moths), (Z)-jasmone (male and total moths), (E)-β-farnesene and β-myrcene (female and total moths), and (E,E)-α-farnesene (male, female, and total moths). In addition, each of these five compounds when added to the 3K core blend performed similarly to the 4K lure (male, females, and total moths). Further studies should expand these results through tests of these and other new blends with a range of component ratios and total loading amounts. Field trials should also be replicated within all host crops of codling moth and across major geographical production regions.  相似文献

In response to injury, plants produce volatile organic compounds (VOCs) that usually differ depending on the type of damage they have suffered (e.g., mechanical damage, herbivory, and oviposition). The objectives of this study were to identify and compare the bouquet of volatiles emitted by passion vine plants (Passiflora edulis) after injury caused by mechanical damage (MD), herbivory (HB), and oviposition (OV) by the lepidopteran, Heliconius erato phyllis. Following injury, extracts of plant emissions were collected from each treatment every 24 h for three days and were analyzed by GC and GC/MS. Results show that plants emitted 12 volatiles before and after damage, namely terpenoids, ketones, and aldehydes. Although no significant differences were detected between the three treatments individually, if the entire bouquet of volatiles is analyzed, samples collected at 24 h were different from samples collected at 48 and 72 h. However, terpenoid emission increased significantly in HB plants after 24 h. HB plants emitted approximately 6300, 50, 46, 11, 6, and 3.6 times more (3E,7E)-4,8,12-trimethyl-1,3,7,11-tridecatetraene, (E)-4,8-dimethyl-1,3,7-nonatriene (DMNT), (E)-β-ocimene, (Z)-β-farnesene, (E)-β-caryophyllene, and farnesane, respectively, compared to control plants. OV plants displayed a peak of emission of (E)-β-ocimene after 72 h, which distinguished them from HB plants. MD plants showed a general increase of VOCs versus undamaged control plants. Furthermore, it has been suggested that (E)-β-ocimene may be sequestered by larvae of H. erato phyllis as a component of the odoriferous bouquet of the abdominal scent glands present in adult males, which play a role in sexual communication.  相似文献

The effects of plant-derived chemicals (volatiles) on the attraction of the Spodoptera litura moth to sex pheromones were evaluated using an electroantennogram (EAG). Neuronal responses of male moths to sex pheromone mixtures (SPs) (a 9:1 mixture of synthetic (9Z,11E)-9,11-tetraddecadienyl acetate (Z9E11-14:OAc) and (9Z,12E)-9,12-tetradecadienyl acetate (Z9E12-14:OAc)) and to SPs mixtures with eight plant volatiles (benzaldehyde, (E)-β-caryophyllene, phenylacetaldehyde, 2,6-nonadienal, benzyl alcohol, racemic linalool, longifolene, and (E)-β-ocimene) were also measured. Then, wind tunnels and field trapping bioassays were conducted to determine the influence of plant volatiles on S. litura moth behavioral responses to SPs. The results indicated that benzaldehyde, phenylacetaldehyde, and benzyl alcohol significantly enhanced, and longifolene, (E)-β-caryophyllene, and (E)-β-ocimene had no significant effect on the attractions to SPs, whereas racemic linalool significantly decreased the attraction of male S. litura moths to SPs throughout the olfactory pathway. 2,6-Nonadienal significantly enhanced olfactory responses, but had no significant effect on output behavior. These findings provide foundations in utilization of plant volatiles and sex pheromones to manage the pest and other agricultural pests.  相似文献

Cotton plants accumulate gossypol and related sesquiterpene aldehydes, which function as phytoalexins against pathogens and feeding deterrents to herbivorous insects. However, to date little is known about the biosynthesis of volatile terpenes in this crop. Herein is reported that 5 monoterpenes and 11 sesquiterpenes from extracts of a glanded cotton cultivar, Gossypium hirsutum cv. CCRI12, were detected by gas chromatography–mass spectrometry (GC–MS). By EST data mining combined with Rapid Amplification of cDNA Ends (RACE), full-length cDNAs of three terpene synthases (TPSs), GhTPS1, GhTPS2 and GhTPS3 were isolated. By in vitro assays of the recombinant proteins, it was found that GhTPS1 and GhTPS2 are sesquiterpene synthases: the former converted farnesyl pyrophosphate (FPP) into β-caryophyllene and α-humulene in a ratio of 2:1, whereas the latter produced several sesquiterpenes with guaia-1(10),11-diene as the major product. By contrast, GhTPS3 is a monoterpene synthase, which produced α-pinene, β-pinene, β-phellandrene and trace amounts of other monoterpenes from geranyl pyrophosphate (GPP). The TPS activities were also supported by Virus Induced Gene Silencing (VIGS) in the cotton plant. GhTPS1 and GhTPS3 were highly expressed in the cotton plant overall, whereas GhTPS2 was expressed only in leaves. When stimulated by mechanical wounding, Verticillium dahliae (Vde) elicitor or methyl jasmonate (MeJA), production of terpenes and expression of the corresponding synthase genes were induced. These data demonstrate that the three genes account for the biosynthesis of volatile terpenes of cotton, at least of this Upland cotton.  相似文献

Sandalwood, Santalum album (Santalaceae) is a small hemi-parasitic tropical tree of great economic value. Sandalwood timber contains resins and essential oils, particularly the santalols, santalenes and dozens of other minor sesquiterpenoids. These sesquiterpenoids provide the unique sandalwood fragrance. The research described in this paper set out to identify genes involved in essential oil biosynthesis, particularly terpene synthases (TPS) in S. album, with the long-term aim of better understanding heartwood oil production. Degenerate TPS primers amplified two genomic TPS fragments from S. album, one of which enabled the isolation of two TPS cDNAs, SamonoTPS1 (1731 bp) and SasesquiTPS1 (1680 bp). Both translated protein sequences shared highest similarity with known TPS from grapevine (Vitis vinifera). Heterologous expression in Escherichia coli produced catalytically active proteins. SamonoTPS1 was identified as a monoterpene synthase which produced a mixture of (+)-α-terpineol and (−)-limonene, along with small quantities of linalool, myrcene, (−)-α-pinene, (+)-sabinene and geraniol when assayed with geranyl diphosphate. Sesquiterpene synthase SasesquiTPS1 produced the monocyclic sesquiterpene alcohol germacrene D-4-ol and helminthogermacrene, when incubated with farnesyl diphosphate. Also present were α-bulnesene, γ-muurolene, α- and β-selinenes, as well as several other minor bicyclic compounds. Although these sesquiterpenes are present in only minute quantities in the distilled sandalwood oil, the genes and their encoded enzymes described here represent the first TPS isolated and characterised from a member of the Santalaceae plant family and they may enable the future discovery of additional TPS genes in sandalwood.  相似文献

Feeding of Helicoverpa zea larvae on cotton (Gossypium hirsutum L.) flower buds (squares) for 24 or 48 h induced the release of a number of terpenes [(E)--ocimene, linalool, (E)--farnesene, (E,E)--farnesene, (E)-4,8-dimethyl-1,3,7-nonatriene, (E,E)-4,8,12-trimethyl-1,3,7,11-tridecatetraene], isomeric hexenyl butyrates, 2-methylbutyrates, indole and (Z)-3-hexenyl acetate. These compounds are not released in significant amounts from undamaged squares and freshly damaged squares. The release of inducible compounds was not limited to the damaged squares themselves. The compounds were also released systemically from the upper undamaged leaves of the same plant after 72 h. However, the composition of the blend of systemically released volatiles differed from the blend released by damaged squares. The compounds that were systemically released from undamaged leaves in response to feeding on the squares were (Z)-3-hexenyl acetate, (E)--ocimene, linalool, (E)-4,8-dimethyl-1,3,7-nonatriene, (E)--farnesene, (E,E)--farnesene, and indole. This study shows that insect damage inflicted to the reproductive parts of a plant causes a systemic emission of volatiles from its vegetative parts.  相似文献
