Abstract: The potential of some isolates of Metarhizium anisopliae and Beauveria bassiana for use in the integrated management of Culex quinquefasciatus was evaluated. Metarhizium anisopliae isolate 1037 was selected in initial bioassays with a 50% lethal concentration (LC50) of 1.97 × 104 conidia/ml. This fungus caused higher mosquito larva mortality when applied as a conidial suspension to the surface of the water than as dry conidia, with a time to 50% lethal (LT50) of 1 day compared with 3.6 days for the dry conidial application. However, results with UV- and heat-inactivated conidia did not confirm a possible role of fungal toxins in causing mortality when ingested by C. quinquefasciatus larvae. Metarhizium anisopliae did not remain active for as long as the bacterium Bacillus sphaericus isolate 2362. At 24 h after application, the fungus-induced mortality on mosquito larvae was significantly lower than the mortality caused by the bacterium. By the second day, almost no activity by the fungus was observed. Results suggest that M. anisopliae isolate 1037 has potential for use in mosquito control programmes.  相似文献

Abstract:  The two-spotted stink bug, Perillus bioculatus (F) (Hem., Pentatomidae) is considered an important predator of Colorado potato beetle egg masses and small larvae. The susceptibility of P. bioculatus second instar nymphs to six isolates of Beauveria bassiana (Balsamo) Vuillemin, which are highly pathogenic to Colorado potato beetle, was tested in the laboratory. Five isolates were highly pathogenic and caused more than 77% mortality after 8 days at a concentration of 106 conidia/ml. However, isolate IPP46 showed low pathogenicity and caused only 11% mortality of nymphs. The pathogenicity of isolates was independent of their host, source or country of origin. Isolate IPP46 was tested at six different concentrations. A linear relationship between the concentration of B. bassiana and the mortality of nymphs was observed. The 50% lethal concentration (LC50) value was 3.4 × 107 conidia/ml. The LT50 decreased with increase of conidia concentration used. The present results suggest that isolate IPP46 has good potential as a biological control agent within an integrated pest management programme.  相似文献

Laboratory and net house assays of the entomopathogenic fungus, Beauveria bassiana CS-1, were conducted to examine the possibility of using this strain to control the diamondback moth (DBM), Plutella xylostella in Korea. Treatment of conidial suspensions at various concentrations indicated that corrected mortality at 1 × 108 conidia/ml was 86.23% with LT50 of 1.63 days under the laboratory conditions and 66.5% with LT50 of 3.61 days in the net house assay. Assessment of rigorous dose–response showed that at least 1800 conidia of the strain CS-1 should adhere to a single DBM larva to induce successful infection. These results indicated that the strain CS-1 has the capability to lower the DBM larval population rapidly as well as effectively.  相似文献

Abstract:  The efficacy of methyl iodide (MI) as a fumigant against all developmental stages of the maize weevil, Sitophilus zeamais Motsch. was investigated. Tests were conducted with concentrations of 1.5, 1.8, 2.1, 2.4, 2.7 and 3.0 mg/l, for a 6-h exposure period. Values of LC50, LC95 and LC99 of MI for immatures and adult stages were determined. The present laboratory tests showed that MI was toxic to various life stages of S. zeamais at relatively short exposure periods. At the LC50 and LC95 levels, the most susceptible stage was the egg stage followed by larvae, pupae and adults (1-day mortality). The egg was found to be most susceptible to MI, requiring 0.81 and 2.16 mg/l for 50 and 99% mortality, respectively, while the adult was most tolerant, requiring 2.30 and 3.02 mg/l for 50 and 99% mortality, respectively, based on 1-day mortality count. Pupae were less susceptible to MI than egg and larvae, requiring 1.47 and 3.19 mg/l for 50 and 99% mortality, respectively. Based on the present toxicity tests, MI has the potential for use as a fumigant to control all developmental stages of the maize weevil, S. zeamais .  相似文献

Abstract. A treated-surface test was used in a bioassay to detect insecticide resistance to the organophosphorus compounds diazinon and propetamphos, in larvae of field strains of Lucilia cuprina in New Zealand during 1990/91 and 1991/92 (December-April). Mean LC50s for both diazinon and propetamphos showed a significant increase between seasons although this is not necessarily indicative of a trend. Mean LC50s for diazinon increased from 1.16 to 1.60 mg/1 (P< 0.001) and for propetamphos from 0.49 to 0.59 mg/1 (P< 0.005) in pooled populations. Diazinon and propetamphos resistance was significantly correlated in both seasons, suggesting side-resistance between the two insecticides.  相似文献

Two pesticides, the insecticide imidacloprid and the herbicide acetochlor, were evaluated for their insecticidal and genotoxic effects in Drosophila melanogaster . Their insecticidal effects were assessed by calculating LC50 values after acute or chronic exposure of larvae and adults to different concentrations of the test compounds. After acute exposure, the LC50 of imidacloprid was 7.59 × 10−5 M for larvae and 1.43 × 10−4 M for adults, and after chronic exposure, it was 2.67 × 10−5 M and 6.09 × 10−5 M for larvae and adults, respectively. On the contrary, the herbicide acetochlor showed no acute or chronic insecticidal effect against either larvae or adults, even at very high concentrations (8 × 10−3 M). For the evaluation of genotoxic properties of the two pesticides, the Somatic Mutation and Recombination Test in D. melanogaster was used. Our results suggest that neither imidacloprid nor acetochlor exhibits mutagenic or recombinogenic activity at applicable concentrations.  相似文献

In this work, a protocol for zoospores production of Phytophthora cryptogea , an economically important plant pathogen was optimized. Five different concentrations of zoospores (5 × 105, 5 × 104, 5 × 103, 5 × 102, 5 × 101 zoospores/ml) from four different isolates of P. cryptogea (Maria 1, Maria 2, S3 1-A, Amazzone) were used as inoculum on pot marigold ( Calendula officinalis ) and gerbera ( Gerbera jamesonii ) plants. Maria 1 was the most virulent isolate both on pot marigold and gerbera plants according to disease severity. A rapid and sensitive pathogen DNA extraction protocol suitable for large quantities of plant samples was adopted. Conventional polymerase chain reaction (PCR) was able to detect the pathogen in artificially inoculated symptomless pot marigold (collected day 12) and gerbera plants (day 8) after pathogen inoculation, with the suspension of 5 × 105, 5 × 104, 5 × 103 P. cryptogea  zoospores/ml. Real-time PCR showed the possibility to detect the pathogen in artificially inoculated symptomless pot marigold (collected day 8) and gerbera plants (day 4) after pathogen inoculation, with the suspension of 5 × 105, 5 × 104 P. cryptogea  zoospores/ml. The first symptoms appeared on pot marigold plants 14 days after pathogen inoculation and on gerbera plants 10 days after inoculation. Real-time PCR showed the possibility to detect the pathogen 4 days before conventional PCR and 6 days before the appearance of disease symptoms both on pot marigold and gerbera plants.  相似文献

SUMMARY 1. Temporary ponds are inhabited by a variety of invertebrates, of which anostracans are an important group. We studied the lifetables of male and female anostracan Streptocephalus mackini at 3 algal concentrations (0.5 × 106, 1.0 × 106 and 1.5 × 106 cells mL−1).
2. Regardless of sex, S. mackini showed better survivorship at lower food levels. The longest average lifespan observed was 85 ± 2 days for males fed Chlorella at 0.5 × 106 cells mL−1.
3. Both net reproductive rate and generation time decreased with increasing food level. The highest net reproductive rate was about 120 cysts per female. The longest generation time of about 40 days, observed at 0.5 × 106 cells mL−1, was more than three times that at 1.5 × 106 cells mL−1.
4. The rate of population increase ( r ) was nearly the same (0.31 ± 0.06) at high (1.5 × 106 cells mL−1) and intermediate (1.0 × 106 cells mL−1) food levels. The r -value at low food level (0.5 × 106 cells mL−1 of Chlorella ) was 0.20 ± 0.01 per day.  相似文献

The relative toxicity (LC50 values based on µg oil/cm2) is evaluated of aqueous n C24 agricultural mineral oil (AMO) emulsions to the egg, six-legged nymph (larva), eight-legged protonymph and adult stages of two-spotted mite ( Tetranychus urticae ) and its predator, Phytoseiulus persimilis , on French bean leaf discs, using a Potter spray tower to apply of the oil. The egg of P. persimilis was the least susceptible stage (LC50 444.84) and its LC50 was significantly higher than all other stages tested of either P. persimilis or T. urticae . The LC50 for adult female T. urticae (LC50 63.89) was significantly lower than the larva (LC50 93.86); however, there was no significant difference in response between the protonymph (LC50 70.44) and the larva, which were both higher than T. urticae eggs (LC50 17.55). LC50s for P. persimilis larva (LC50 43.87), protonymph (LC50 41.55) and adult female (LC50 53.34) were similar. Scanning electron microscopy showed that the egg surface of T. urticae is usually well covered with fine silk that may trap more oil and increase AMO efficacy. Other possible differences in AMO efficacy between T. urticae and P. persimilis may be due to differences in egg size, egg incubation period, egg surface structure and the presence of vulnerable respiratory cones in T. urticae eggs. Dose of 0.2–0.3% (w/w) is considered to be the most appropriate for n C24 AMOs use against T. urticae in combination with P. persimilis in integrated pest management programs.  相似文献

Aims:  Concentration of pathogens diluted in large volumes of water is necessary for their detection. An automated concentration system placed online in drinking water distribution systems would facilitate detection and mitigate the risk to public health.
Methods and Results:  A prototype concentrator based on dead-end hollow fibre ultrafiltration was used to concentrate Bacillus atrophaeus spores directly from tap water. Backflush was used to recover accumulated particulates for analysis. In field tests conducted on a water utility distribution system, 3·2 × 104–1·4 × 106 CFU ml−1 (6·1 × 106–3·0 × 108 CFU) were recovered from the filter when 2·9 × 107–1·0 × 109 CFU were spiked into the system. Per cent recovery ranged from 21% to 68% for flow volumes of 15–21 l. Tests using spore influent levels <10 CFU l−1 (spike < 1000 CFU) yielded 23–40% recovery for volumes >100 l.
Conclusions:  B. atrophaeus spores at levels <10 CFU l−1 were concentrated directly from tap water using an automated dead-end hollow-fibre ultrafiltration system.
Significance and Impact of the Study:  The prototype concentrator represents a critical step towards an autonomous system that could be installed in drinking water distribution lines or other critical water lines to facilitate monitoring. Recovered samples can be analysed using standard or rapid biosensor methods.  相似文献

Acute toxicity studies of emamectin and spinosad against Helicoverpa armigera revealed that the pest is highly susceptible to both the insecticides. The median lethal dose (LD50) of emamectin is 3.86 × 10−3 µg per larva. The median lethal concentrations (LC50) of emamectin and spinosad were found to be 0.09 and 2.94 ppm, respectively. The discriminating doses were fixed based on the LC95 of the susceptible population of H. armigera as 0.80 ppm for emamectin and 10 ppm for spinosad. Resistance was not observed when the discriminating doses of emamectin and spinosad were applied on field-collected populations of H. armigera from two intensive cotton growing areas, Coimbatore and Madurai, India.  相似文献

The effects of environmental factors on infection of the entomopathogenic fungus, Nomuraea rileyi , isolated from the corn earworm, Helicoverpa armigera , in Taiwan, to its host insect were studied in the laboratory. The fungus caused higher larval mortality at 20°C than at 30°C when 5 × 106 conidia/ml were sprayed on the fourth instar. However, mortality of the fifth instar injected with 1 × 103 conidia/larva was not significantly different when the inoculated larvae were incubated from 15 to 30°C. The fungal development in inoculated larvae was best at 20 and 25°C after shifting from 20°C to either lower or higher temperatures. The germination rate was higher at 20 and 25°C than at 30 or 35°C. Conidial germination was better on the wash-off of insect cuticle than on Sabouraud maltose agar with yeast extract. Sporulation on chill-dried cadavers was maximal at 95 or 100% relative humidity than at lower levels of relative humidity. The time required for sporulation was 2 days less at 100% than at 95% relative humidity. Although photoperiod did not affect fifth instar mortality caused by N. rileyi , the median lethal time (LT50) values were shorter upon incubating under light than in darkness. Incubation of infected cadavers under 12 or 24 h light resulted in 20-fold more conidial production than under full darkness. Therefore, illumination is necessary for development of this isolate on insect cadavers.  相似文献

Abstract:  In laboratory bioassays, the efficacy of the entomopathogenic fungus Beauveria bassiana against the spruce bark beetle, Ips typographus , was tested under various conditions. Four of the tested isolates and the commercial product Boverol® caused 99–100% mortality when tested at a concentration of 1.0 × 107 conidia/ml at 25°C. Using B. bassiana isolate 138 at a concentration of 1.0 × 106, the median survival time (MST) was 6.1 d and significantly longer compared with the MST of 4.2 and 4.0 d at 1.0 × 107 and 1.0 × 108 conidia/ml, respectively. In the next experiment, the beetles were maintained on spruce bark, filter paper or artificial diet during the bioassay with Boverol®, and significant differences in the MST of 3.6, 2.5 and 5.3 d, respectively, were noticed. The experiment with Boverol® at different temperatures showed that the beetles lived significantly longer at 15°C (MST 8.7 d) than at 20, 25, 30 and 35°C. At 25°C, the beetles died most rapidly (MST 3.5 d). At different relative humidities (RH) of 40, 70 and 100%, nearly all beetles were dead after treatment with a suspension of Boverol® at 1.0 × 107 conidia/ml. At 40% RH, 49% of the untreated beetles died after 7 d. The best effects were achieved with the following bioassay: beetles were fed for three days on artificial diet, then dipped into a solution of 1.0 × 107 conidia/ml and transferred on a piece of spruce bark in Petri dishes at 25°C and 70% RH.  相似文献

Several insect growth inhibitors (IGIs) and regulators (IGRs) were tested in the laboratory for their ovicidal and larvicidal properties on the codling moth C. pomonella , by dipping apples in solutions of them. The IGIs which block chitin synthesis – diflubenzuron, hexaflumuron and teflubenzuron – were noticeably more effective against eggs than on newborn larvae with preventive ovicidal 50% lethality concentrations (LC50) values of approximately 0.6, 1.3 and 15 p.p.m., respectively, and larvicidal LC50 values of 104, 1208 and 204 p.p.m. Flufenoxuron, on the other hand, is almost as effective on larvae (LC50 : 9.9 p.p.m.) as on eggs (LC50 : 5.4 p.p.m.). Fenoxycarb, an IGR juvenile hormone analogue, acts as an excellent ovicidal product with an LC50 value of 0.05 p.p.m. Tebufenozide, an IGR ecdyson (moulting hormone) agonist, is exclusively larvicidal with an LC50 at 0.4 p.p.m. Methoxyfenozide, an IGR of the same family and currently being developed, acts as effectively on eggs as on larvae with ovicidal and larvicidal LC50 values of about 0.6 and 0.8 p.p.m., respectively. When ovicidal products are applied as a curative treatment on eggs less than 24 h old, their effectiveness is much lower than that obtained from preventive application.  相似文献

Abstract: Based on surveys of species of museum pest insects commonly found in Finland, Norway, Denmark and Sweden, six species were selected for a study of the effect of exposure to anoxic treatment on the larval stage. An oxygen level of 0.3% (the rest, nitrogen) was applied and lethal exposure times were determined. Anthrenus museorum L. was the most susceptible species (LT99 = 32.2 h), while Attagenus woodroffei Halst. & Green and Attagenus smirnovi Zhantiev (LT99 = 88.1 h) were the most tolerant species. The LT99 value of At. woodroffei was not calculated, as only 50% of the larvae were killed by the treatment. The results indicate that large intraspecific variation is present in these two Attagenus species. Larvae of Anthrenus verbasci (L.) (LT99 = 43.9 h), Reesa vespulae (Milliron) (LT99 = 53.6 h), and Trogoderma angustum (Solier) (LT99 = 57.2 h) showed intermediate tolerance.  相似文献

Abstract. This study aims to investigate engraftment of human cord blood and foetal bone marrow stem cells after in utero transplantation via the intracoelomic route in the sheep. Here, we performed transplantation in 14 single and 1 twin sheep foetuses at 40–47 days of development, using a novel schedule for injection. (i) Single injection of CD34+ human cord blood stem cells via the coelomic route (from 10 to 50 × 104) in seven single foetuses. (ii) Single injection of CD34+ foetal bone marrow stem cells via the intracoelomic route with further numbers of cells (20 × 105 and 8 × 105, respectively) in three single and in one twin foetuses. (iii) Double fractioned injection (20–30 × 106) via the coelomic route and 20 × 106 postnatally, intravenously, shortly after birth of CD3-depleted cord blood stem cells in four single foetuses. In the first group, three single foetuses showed human/sheep chimaerism at 1, 8 and 14 months after birth. In the second group, the twin foetuses showed human/sheep chimaerism at 1 month after birth. In the third group, only two out of four single foetuses that underwent transplantation showed chimaerism at 1 month. While foetal bone marrow stem cells showed good short-term engraftment (1 month after birth), cord blood stem cells were able to persist longer in the ovine recipients (at 1, 8 and 14 months after birth).  相似文献

In trials conducted in Benin, conidia of Beauveria bassiana were evaluated as a control method against the cowpea weevil, Callosobruchus maculatus , in stored cowpea. In the first trial using a high artificial infestation of C.   maculatus in 8-kg batches of cowpea in jerry cans under typical conditions, concentrations of 1 × 109 and 1 × 1011 conidia kg−1 were compared with lemon grass oil at 2 mL kg−1 and the synthetic pesticide mixture of 1.5% pirimiphos methyl + 0.05% deltamethrin at 0.5 g kg−1. After 2 months of storage, seed losses (SD) were 20.63 (5.3)% in the untreated control, 8.04 (3.2)% in the low-dose B.   bassiana group, 3.12 (1.3)% in the high-dose B.   bassiana group, 2.52 (0.4)% in the lemon grass oil group and 0% in the pirimiphos methyl + deltamethrin group. In a second trial with natural infestations in 50-kg batches of cowpea in 200-L drums, treatment with B.   bassiana 1 × 1011 conidia kg−1 was compared with pirimiphos methyl + deltamethrin at 0.5 g kg−1. After 6 months of storage on six farms, losses reached 30.76 (1.5)% in the control, 1.28 (0.2)% in the pirimiphos methyl + deltamethrin group and 3.69 (0.6)% in the B.   bassiana group.  相似文献

Aims:  To use experimental design techniques and a multiple logistic regression model to optimize a microbiological inhibition test with dichotomous response for the detection of Penicillin G in milk.
Methods and Results:  A 23 × 22 robust experimental design with two replications was used. The effects of three control factors (V: culture medium volume, S: spore concentration of Geobacillus stearothermophilus , I: indicator concentration), two noise factors (Dt: diffusion time, Ip: incubation period) and their interactions were studied. The V, S, Dt, Ip factors and V × S, V × Ip, S × Ip interactions showed significant effects.
Conclusions:  The use of 100  μ l culture medium volume, 2 × 105 spores ml−1, 60 min diffusion time and 3 h incubation period is recommended. In these elaboration conditions, the penicillin detection limit was of 3·9  μ g l−1, similar to the maximum residue limit (MRL). Of the two noise factors studied, the incubation period can be controlled by means of the culture medium volume and spore concentration.
Significance and Impact of the Study:  We were able to optimize bioassays of dichotomous response using an experimental design and logistic regression model for the detection of residues at the level of MRL, aiding in the avoidance of health problems in the consumer.  相似文献

Abstract  Hexane, petroleum ether, chloroform, ethyl acetate, methanol and water extracts of Clausena anisata [(Willd.) Hook F. Ex Benth] leaves and roots were evaluated against Helicoverpa armigera (Hübner) for antifeedant activities. Antifeedant activity was confirmed, and was found to be higher in root extracts than those of the leaf. Chloroform and petroleum ether extracts of the root showed strongest antifeedant activities (DC50s [concentration (C) causing 50% deterrence compared with the control] 0.014% and 0.016% respectively), and root extracts were fractionated using silica gel column chromatography. One fraction of the chloroform and one of the petroleum ether root extracts was active; and on the basis of mass spectroscopy and 1H and 13C nuclear magnetic resonance spectral data, the active compounds in the two fractions were confirmed to be identical, and identified as osthol [2H-1-Benzopyran-2-one, 7-methoxy-8-(3-methyl-2-butenyl)]. The highest concentration of osthol was found in the chloroform root extract. Antifeedant activities of the root extracts, as measured by DC50 values, were highly correlated with their osthol contents. Approximately 99% of the variation in bioactivity of the root extracts could be accounted for by variation in osthol content; osthol therefore, appeared to be an antifeedant component of C . anisata to H. armigera . This may provide a new approach to managing this pest.  相似文献

One laboratory and three greenhouse experiments were conducted to study the pathogenicity and efficacy of Finnish isolates of entomopathogenic hyphemycetous fungi against cabbage root flies. In Petri dishes, exposure to 1.5 × 1010 spores of Metarhizium anisopliae per dish caused 40–50% mortality of undifferentiated second- and third-stage larvae of Delia floralis , and 1 × 107 spores per dish caused 40–50% mortality of Delia radicum larvae. In one greenhouse test, 1 × 108 and 1 × 109 spores of M. anisopliae and Paecilomyces fumosoroseus reduced the root damage of head cabbage by 20–70% compared with untreated controls, although this was not accompanied by significant reductions in the number of pupae. Only M. anisopliae consistently grew out of larvae and pupae of D. floralis during incubation that followed their recovery from the soil at the end of an experiment testing different formulations of M. anisopliae and Beauveria bassiana , but the frequency of the latent infections of the pest by M. anisopliae was not associated with reduced severity of damage to seedlings of head cabbage.  相似文献
