Effects of low‐temperature stress, cold acclimation and growth at high irradiance in a spring (Triticum aestivum L. cv. Katepwa) and a winter wheat (Triticum aestivum L. cv. Monopol) were examined in leaves and crowns with respect to the sucrose utilisation and carbon allocation. Light‐saturated and carbon dioxide (CO2)‐saturated rates of CO2 assimilation were decreased by 50% in cold‐stressed spring and winter wheat cultivars. Cold‐ or high light‐acclimated Katepwa spring wheat maintained light‐saturated rates of CO2 assimilation comparable to those of control spring wheat. In contrast, cold‐ or high light‐acclimated winter wheat maintained higher light and CO2‐saturated rates of CO2 assimilation than non‐acclimated controls. In leaves, during either cold stress, cold acclimation or acclimation to high irradiance, the sucrose/starch ratio increased by 5‐ to 10‐fold and neutral invertase activity increased by 2‐ to 2.5‐fold in both the spring and the winter wheat. In contrast, Monopol winter wheat, but not Katepwa spring wheat, exhibited a 3‐fold increase in leaf sucrose phosphate synthase (SPS) activity, a 4‐fold increase in sucrose:sucrose fructosyl transferase activity and a 6.6‐fold increase in acid invertase upon cold acclimation. Although leaves of cold‐stressed and high light‐grown spring and winter wheat showed 2.3‐ to 7‐fold higher sucrose levels than controls, these plants exhibited a limited capacity to adjust either sucrose phosphate synthase or sucrose synthase activity (SS[s]). In addition, the acclimation to high light resulted in a 23–31% lower starch abundance and no changes at the level of fructan accumulation in leaves of either winter or spring wheat when compared with controls. However, high light‐acclimated winter wheat exhibited a 1.8‐fold higher neutral invertase activity and high light‐acclimated spring wheat exhibited an induction of SS(d) activity when compared with controls. Crowns of Monopol showed higher fructan accumulation than Katepwa upon cold and high light acclimation. We suggest that the differential adjustment of CO2‐saturated rates of CO2 assimilation upon cold acclimation in Monopol winter wheat, as compared with Katepwa spring wheat, is associated with the increased capacity of Monopol for sucrose utilisation through the biosynthesis of fructans in the leaves and subsequent export to the crowns. In contrast, the differential adjustment of CO2‐saturated rates of CO2 assimilation upon high light acclimation of Monopol appears to be associated with both increased fructan and starch accumulation in the crowns.  相似文献

The effect of hardening on the composition of phenolic compounds in winter wheat (Triticum aestivum L.) leaves was studied. It was shown that green tissues contained mainly flavonoids, especially flavons (C-and O-glycosides of apigenin and luteolin), and also ferulic acid derivatives. Among flavons, derivatives of luteolin dominated, including isoorientin, which comprised approximately a half of the content of all identified phenolic compounds. Low temperature induced the accumulation of phenolic compounds in winter wheat leaves, whereas their qualitative composition was not practically changed.  相似文献

The formation of soluble phenolic compounds, including flavonols, was studied in winter (Erythrospermum , Lutescens 230, and R 47-28) and spring cultivars (Lada) of wheat (Triticum aestivum L.). The contents of soluble phenolic compounds and flavonols were 1.8–2.6 and 0.5–1.3 mg/g fresh weight, respectively. These results illustrate the similarity of phenol metabolism in leaves of winter and spring wheat cultivars. The exception was the cultivar R 47-28 that accumulated the maximum amount of phenolic compounds (e.g., flavonols). In this cultivar the ratio of flavonols reached 50% of total soluble phenol content. In other cultivars, this parameter did not exceed 25–35%. The data indicate that the cultivar R 47-28 differs from other wheat cultivars in metabolism of phenolic compounds. Abrupt temperature reduction in winter wheat at either autumn seedling stage prior to vernalisation or early spring crown stage can cause severe crop damage and reduce production. Many studies have reported the physiological and molecular mechanisms underlying cold acclimation in winter wheat by comparing it with spring wheat. However, processes associated with abrupt temperature reduction in autumn seedling stage prior to vernalisation in winter wheat are less understood. In this study, physiological and molecular responses of winter wheat seedlings to abrupt low temperature (LT) stress were characterised in the relatively LT‐tolerant winter wheat cultivar Shixin 828 by comparing it with the relatively LT‐sensitive cultivar Shiluan 02‐1 using a combination of physiological, proteomics and biochemical approaches. Shixin 828 was tolerant to abrupt LT stress, while Shiluan 02‐1 exhibited high levels of reactive oxygen species (ROS) and leaf cell death. Significant increases in relative abundance of antioxidant‐related proteins were found in Shixin 828 leaves, which correlate with observed higher antioxidant enzyme activity in Shixin 828 compared to Shiluan 02‐1. Proteomics analysis also indicated that carbohydrate metabolism‐related proteins were more abundant in Shiluan 02‐1, correlating with observed accumulation of soluble sugars in Shiluan 02‐1 leaves. Amino acid analysis revealed a strong response to LT stress in wheat leaves. A negative effect of exogenous sucrose on LT tolerance was also found. This study indicates that high ROS scavenging capacity and high abundance of photosynthesis‐related proteins might play a role in winter wheat response to abrupt LT stress. In contrast, excess accumulation of soluble sugars might be disadvantageous for LT tolerance in the wheat cultivar Shiluan 02‐1.  相似文献

Phytochemical investigation of the stem bark and leaves of Hymenostegia afzelii resulted in the isolation of three new flavonoids, named afzelin A-C (1–3), together with five known compounds: 7,6-(2″,2″-dimethylpyrano)-3,5,4′-trihydroxyflavone, apigenin, 3-O-α-l-rhamnopyranosylcincholic acid, 3-O-β-d-glucopyranosylcincholic acid, and dodecanoic acid 1,1′-[(1S)-1-(hydroxymethyl)-1,2-ethanediyl] ester. The structures of 1–3 were determined by means of spectroscopic methods. Compounds 1–3 were tested in vitro for their preliminary cytotoxicity using the Artemia salina assay.  相似文献

Resistance to powdery mildew infection was compared in 18 winter wheat cultivars under field conditions in three seasons, 1977 to 1979. In each year, natural infection occurred from populations of Erysiphe graminis f. sp. tritici containing host-specific pathogenicity characters matching identified cultivar resistance. Leaves, ears and stems were assessed separately. The expression of resistance in leaves appeared to be independent of that in ears. There were few consistent differences between cultivars in stem infection. Resistance in adult leaves did not necessarily correspond with that in seedling leaves; resistance in ears did not appear to be correlated with plant height. Quantitative differences between cultivars having the same identified resistance characters occurred in both leaves and ears; Flinor, Avalon and Bounty were most resistant on leaves whereas Iona, Wizard and Bounty were most resistant on ears. Differences in background resistance were considered to be responsible for these observations.  相似文献

Investigation of the aqueous alcoholic extract of Pyruscalleryana Decne. leaves led to the isolation of two new phenolic acids glycosides, namely protocatechuoylcalleryanin-3-O-β-glucopyranoside (1) and 3′-hydroxybenzyl-4-hydroxybenzoate-4′-O-β-glucopyranoside (2), together with nine known compounds among them lanceoloside A and methylgallate, which have been isolated for the first time from the genus Pyrus. Structures of the isolated compounds were established by spectroscopic analysis, including UV, IR, HRESI-MS, and 1D/2D NMR. The total extract and some isolated compounds were determined against DPPH (2,2-diphenyl-1-(2,4,6-trinitrophenyl) hydrazinyl radical, for their free radical scavenging activity, the total alcoholic extract showed strong antioxidant activity while the two new compounds showed weak antioxidant activity.  相似文献

The increase in concentrations of phenolic compounds in boron (B) deficiency has been demonstrated in many herbaceous plant species, but information on woody plants is scarce. It has been suggested that accumulation of phenolic compounds plays a role in the development of cold hardiness in herbaceous plants but also that B deficiency decreases winter hardiness. Here we study the effects of B nutrition on phenolic compounds in Norway spruce (Picea abies L.) in the course of cold acclimation. Phenolic compounds were analysed in Norway spruce seedlings from three different B-fertilisation treatments in two harvests: non-acclimated and cold-acclimated seedlings. Norway spruce phenolic compounds consisted mainly of condensed tannins. During B deficiency, condensed tannins and monocoumaroyl–astragalin der. 1 increased in non-acclimated seedlings. The increase in tannins was 21%, which was nearly significant. However, the effect of B on phenolic compounds was almost absent in cold-acclimated seedlings. The condensed tannin concentration increased much more with time in the simulated autumn than due to B deficiency, and we conclude that the B effect was probably not large enough to be important for the hardening of the seedlings. The total phenolic concentrations more than doubled during the course of cold hardening suggesting that phenolics have a role in the winter hardiness in Norway spruce.  相似文献

Transgenic rice (Oryza sativa) plants were engineered to express a N-(hydroxycinnamoyl)transferase from pepper (Capsicum annuum), which has been shown to have hydroxycinnamoyl-CoA:tyramine N-(hydroxycinnamoyl)transferase activity, a key enzyme in the synthesis of hydroxycinnamic acid amides, under the control of constitutive maize (Zea mays) ubiquitin promoter. The transgenic rice plants require foliar application of amines to support synthesis of hydroxycinnamic acid amides, suggestive of limiting amine substrates in rice shoots. In addition, when T2 homozygous transgenic rice plants were grown in the presence of amines or phenolic acids, two novel compounds were exclusively identified in the leaves of the transgenic plants. These compounds eluted earlier than p-coumaroyltyramine and feruloyltyramine during HPLC chromatography and were identified as p-coumaroylserotonin and feruloylserotonin by liquid chromatography/mass spectrometry and other methods. To test whether the unpredicted production of serotonin derivatives is associated with the pepper N-(hydroxycinnamoyl)transferase, the substrate specificity of the pepper enzyme was analyzed again. Purified recombinant pepper N-(hydroxycinnamoyl)transferase exhibited a serotonin N-hydroxycinnamoyltransferase (SHT) activity, synthesized p-coumaroylserotonin and feruloylserotonin in vitro, and demonstrated a low K(m) for serotonin. SHT activity was inhibited by 10 to 50 mm tyramine. In addition, SHT activity was predominantly found in the root tissues of wild-type rice in parallel with the synthesis of serotonin derivatives, suggesting that serotonin derivatives are synthesized in the root of rice. This is the first report of SHT activity and the first demonstration, to our knowledge, that serotonin derivatives can be overproduced in vivo in transgenic rice plants that express serotonin N-(hydroxycinnamoyl)transferase.  相似文献

Understorey shade plants are seasonally exposed to dramatic changes in light conditions in deciduous forests related with the dynamics of the overstorey leaf phenology. These transitions are commonly followed by changes in herb plant communities, but shade-tolerant evergreen species must be able to adapt to changing light conditions. In this work we checked the photoprotective responses of evergreen species to acclimate to the shady summer environment and reversibly de-acclimate to a more illuminated environment after leaf fall on deciduous overstoreys. For that purpose we have followed the process of light acclimation in leaves of common box (Buxus sempervirens) during the winter to spring transition, which decrease irradiance in the understorey, and conversely during the transition from summer to autumn. Four parameters indicative of the structure and degree of acclimation of the photosynthetic apparatus have been studied: chlorophyll a/b ratio which is supposed to be inversely proportional to the antenna size, α/β-carotene which increases in shade acclimated leaves and the pools of α-tocopherol and xanthophyll cycle pigments (VAZ) which are two of the main photoprotection mechanisms in plants. Among these parameters, chlorophyll a/b ratio and VAZ pool responded finely to changes in irradiance indicating that modifications in the light harvesting size and photoprotective capacity contribute to the continuous acclimation and de-acclimation of long-lived evergreen leaves.  相似文献

Contents of soluble proteins, proline and chlorophyll in winter wheat (Triticum aestivum cv. Doğu-88) and cabbage leaves (Brassica oleracea convar. acephala) during acclimation to low temperature were investigated. When both of the plants species were cold acclimated, soluble proteins, proline and chlorophyll contents were higher than in the controls (non-acclimated). Flavonoid compounds are ubiquitous in nature. They constitute an important part of the human diet and act as active principles of many medicinal plants. Their O-methylation increases their lipophilicity and hence, their compartmentation and functional diversity. We have isolated and characterized a full-length flavonoid O-methyltransferase cDNA (TaOMT2) from a wheat leaf cDNA library. The recombinant TaOMT2 protein was purified to near homogeneity and tested for its substrate preference against a number of phenolic compounds. Enzyme assays and kinetic analyses indicate that TaOMT2 exhibits a pronounced preference for the flavone, tricetin and gives rise to three methylated enzyme reaction products that were identified by TLC, HPLC and ESI-MS/MS as its mono-, di- and trimethyl ether derivatives. The sequential order of tricetin methylation by TaOMT2 is envisaged to proceed via its 3′-mono- → 3′,5′-di- → 3′,4′,5′-trimethyl ether derivatives. To our knowledge, this is the first report of a gene product that catalyzes three sequential O-methylations of a flavonoid substrate.  相似文献

Time-courses of the development of freezing tolerance and the expression of a cold-responsive gene wlt10 were monitored during cold acclimation in wheat (Triticum aestivum L.). Bioassay showed that cold acclimation conferred much higher freezing tolerance on a winter cultivar than a spring cultivar. Northern blot analysis showed that the expression of wlt10 encoding a novel wheat member of a cereal-specific LT-COR protein family was specifically induced by low temperature. A freezing-tolerant winter cultivar accumulated the mRNA more rapidly and for a longer period than a susceptible spring cultivar. The increase in the amount of mRNA was temporary but the peak occurred at the time when the maximum level of freezing tolerance was attained. The mRNA accumulated more in the leaves than in the roots, and different light/dark regimes modulated the level of mRNA accumulation. Genomic Southern blot analyses using the nulli-tetrasomic series showed that the wlt10 homologues were located on the homologous group 2 chromosomes.  相似文献

A variety of volatile phenylpropenes, C6‐C3 compounds are widely distributed in the plant kingdom, whereas prenylated phenylpropenes are limited to a few plant species. In this study, we analysed the volatile profiles from Illicium anisatum leaves and identified two O‐prenylated phenylpropenes, 4‐allyl‐2‐methoxy‐1‐[(3‐methylbut‐2‐en‐1‐yl)oxy]benzene [O‐dimethylallyleugenol ( 9 )] and 5‐allyl‐1,3‐dimethoxy‐2‐(3‐methylbut‐2‐en‐1‐yl)oxy]benzene [O‐dimethylallyl‐6‐methoxyeugenol ( 11 )] as major constituents. The structure–activity relationship of a series of eugenol derivatives showed that specific phenylpropenes, including eugenol ( 1 ), isoeugenol ( 2 ) and 6‐methoxyeugenol ( 6 ), with a phenolic hydroxy group had antifungal activity for a fungal pathogen, whereas guaiacol, a simple phenolic compound, and allylbenzene had no such activity. The eugenol derivatives that exhibited antifungal activity, in turn, had no significant toxicant property for mite oviposition. Interestingly, O‐dimethylallyleugenol ( 9 ) in which the phenolic oxygen was masked with a dimethylallyl group exhibited a specific, potent oviposition deterrent activity for mites. The sharp contrast in structural requirements of phenylpropenes suggested distinct mechanisms underlying the two biological activities and the importance of a phenolic hydroxy group and its dimethylallylation for the structure‐based design of new functional properties of phenylpropenes.  相似文献

Adult rove beetles (Staphylinidae) were sampled every 7–14 days from one winter wheat field located in each of the four major wheat growing regions of Oklahoma during the 1999–2000 and 2000–2001 growing seasons. The number of cereal aphids per tiller, wheat plant growth stage, and wheat tiller density also were estimated. A total of 12 genera representing 13 species of beetles were collected from the field. The density of rove beetles was generally low, ranging from 0.003 beetles per m2 in fall to 0.106 beetles per m2 in spring. Rove beetle communities differed among seasons. After accounting for the effect of season, there was no statistically significant association between rove beetle community structure and field location, aphid density, wheat plant growth stage, or wheat plant density. Most rove beetle species showed no association with a particular season, however, Aleochara notula Erichson, Lathrobium sp., and Oxypoda sp. were present predominantly in fall, while Bisnius inquitus Erichson was associated with winter. Oxypoda sp. was the most abundant rove beetle in winter wheat fields in spring and was relatively abundant in winter, but was not collected from wheat fields in fall. Tachyporus jocosus Say was present in wheat fields during all seasons. T. jocosus was the most abundant rove beetle species in the winter wheat fields in fall and winter and was the second most abundant species during spring.  相似文献

Ether lipids     
The naturally occurring 1-O-alkyl-sn-glycerols and their methoxylated congeners, 1-O-(2′-methoxyalkyl)-sn-glycerols, are biologically active compounds, ubiquitously found in nature as diacyl glyceryl ether lipids and phosphoether lipids. The chief objective of this article is to provide a comprehensive and up to date review on such ether lipids. The occurrence and distribution of these compounds in nature are extensively reviewed, their chemical structure and molecular variety, their biosynthesis and chemical synthesis and, finally, their various biological effects are described and discussed. An unprecedented biosynthesis of the 2′-methoxylated alkylglycerols is proposed. The first synthesis of enantiopure (Z)-(2′R)-1-O-(2′-methoxyhexadec-4′-enyl)-sn-glycerol, the most prevalent 2′-methoxylated type alkylglycerol present in cartilaginous fish, is described. The objective of this study was to identify plasma membraneproteins that are specifically induced by cold acclimation inwheat (Triticum aestivum L.). Two cultivars with a marked differencein the genetic ability to cold-acclimate, namely, spring wheat(cv. Chinese Spring) and winter wheat (cv. Norstar), were usedas the experimental material. After four weeks of growth ina cold chamber, the freezing tolerance in the shoots of winterwheat increased to –18°C, whereas it increased onlyto –8°C in the shoots of spring wheat. In the caseof roots from both cultivars, freezing tolerance increased onlyslightly after the growth in the cold environment. Cold acclimationinduced remarkable changes in the electrophoretic patterns ofplasma membrane proteins which depended on both the cultivarand the tissue examined. Levels of polypeptides with molecularmasses from 22 to 31 kDa decreased in both the root and shootplasma membranes from both cultivars. Among these polypeptides,levels of those of 28 and 26 kDa decreased abruptly after oneweek of cold acclimation. By contrast, levels of polypeptidesof 89, 83, 52, 23, 18 and 17 kDa increased specifically in theshoots of winter wheat. The increases in the levels of the 23-,18- and 17-kDa polypeptides were proportional to the developmentof freezing tolerance. Freeze-fracture electron microscopy ofplasma membranes from shoot cells revealed that the number ofintramembrane particles on the fracture faces decreased markedlyin winter wheat after cold acclimation, but to a lesser extentin spring wheat. The success of P. juliflora, an evergreen woody species has been largely attributed to temperature acclimation and stomatal control of photosynthesis under wide range of environmental conditions prevalent in India. We studied the contribution of the enzyme ribulose-1,5 bisphosphate carboxylase/oxygenase (Rubisco) in diurnal and seasonal photosynthesis changes in P. juliflora. The changes observed in photosynthesis under natural conditions could be effected by the growth temperatures, which ranged from 10–30 °C in winter to 30–47 °C in summer. However, the Total Rubisco activity displayed a constant diurnal pattern and showed a maximum at 1200 in all seasons namely spring, summer, monsoon and winter irrespective of the changes in temperature. The Total Rubisco activity from two cohorts of leaves produced in spring and monsoon appeared to be down-regulated differentially at low PPFD during the evening. The in vivo and in vitro measurements of carboxylation efficiency of Rubisco showed wide variation during the day and were correlated with the photosynthesis rate. The light activation of Rubisco showed the acclimation to moderately high temperatures in different seasons except in summer. The exceptionally high temperatures (>45 °C) in summer, though not affecting Total activity, severely inhibited the light activation of Rubisco and also modulated the recovery process for the activation of Rubisco. Our studies suggest that the modulation of Rubisco driven by Rubisco activase and not Rubisco per se was crucial for the diurnal regulation of photosynthesis. NBRI Publication No.: 528  相似文献
