The North American beluga whale Delphinapterus leucas population has been divided into a number of putative geographical stocks based upon migration routes and areas of summer concentration. Nucleotide sequences of the mitochondrial DNA (mtDNA) control region were used to assess whether these geographical stocks are genetically distinct. Beluga whale samples from 25 sites were collected primarily from aboriginal subsistence hunts across North America from 1984 to 1994. Thirty-nine mtDNA haplotypes were identified in 628 beluga samples. No differences were found in the distribution of haplotypes between male and female beluga whales at any sampling site. These haplotypes segregated into two distinct assemblages in both a haplotype network and a neighbour-joining tree. The haplotype assemblages had a geographically disjunct distribution that suggests postglacial recolonization of the North American Arctic from two different refugia.
An analysis of molecular variance based on haplotype relationships and frequency indicated genetic heterogeneity among beluga whale summering groups ( P ≤ 0.001). Sequence divergence estimates between sampling sites also indicated geographical differentiation, particularly between samples taken at east Hudson Bay or St Lawrence River and the western or central Arctic. The results of this study show a high degree of philopatry to specific summering areas by this highly mobile animal.  相似文献

Most larger individuals of migratory striped bass Morone saxatilis from the two major Atlantic coast stocks, the Chesapeake Bay and Hudson River, appear to winter in mid‐Atlantic coastal waters. But it is not known whether they exhibit differential wintertime distributions in accordance with the latitudinal differences in locations of these two estuaries. Mixed‐stock analyses were conducted based on mitochondrial DNA and nuclear DNA genotypic frequencies on wintertime collections of striped bass from coastal waters. No significant differences (P > 0.05) were seen in the proportions of striped bass from the two stocks between collections made from the Delaware Bay mouth and Cape Hatteras in 1997. However, there was a substantially higher Hudson contribution to a 1995 collection from coastal New Jersey (0.349, SD = 0.136) than to the combined 1997 Delaware Bay mouth and Cape Hatteras collection (0.157, SD = 0.072), suggesting this question deserves further study. Additionally, use of the original four reference samples from Chesapeake Bay tributaries (Choptank, Potomac, Rappahannock, Upper Bay) proved adequate alone in characterizing the Chesapeake Bay stock in simulations in which additional tributary collections (Nanticoke, Patuxent, Pocomoke) were added.  相似文献

Results from our previous investigation of beluga whale mitochondrial DNA (mtDNA) revealed genetic heterogeneity among beluga from different summering locations that was interpreted as representing a high degree of summering site philopatry. However, mtDNA is maternally inherited and does not reflect mating that may occur among beluga from different summering locations in wintering areas or during annual migrations. To test the possibility that breeding occurs among beluga from different summering locations, genetic variability at five nuclear DNA (nDNA) microsatellite loci was examined in the same animals tested in the mtDNA study. Beluga samples ( n = 640) were collected between 1984 and 1994 from 24 sites across North America, mostly during the summer. Whales from the various sites were categorized into eight summering locations as identified by mtDNA analysis, as well as four hypothesized wintering areas: Bering Sea, Hudson Strait (Hudson Strait, Labrador Sea, southwest Davis Strait), Baffin Bay (North Water, east Davis Strait), and St Lawrence River. Microsatellite allele frequencies indicated genetic homogeneity among animals from summering sites believed to winter together but differentiation among whales from some of the wintering areas. In particular, beluga from western North America (Bering Sea) were clearly distinguished from beluga from eastern North America (Hudson Strait, Baffin Bay, and St Lawrence River). Based upon the combined data set, the population of North American beluga whales was divided into two evolutionarily significant units. Beluga whale hunting is one of the most social subsistence hunting activities to take place in the Canadian Arctic. Through the harvest, distribution and consumption of beluga whales, Inuit identity and social relationships are affirmed. The whale-hunting complex is influenced by beliefs that beluga whales are sentient beings who inhabit a shared social space with humans. Yet, across the region beluga whales are perceived by wildlife managers as scarce resources and as such require protection through the imposition of management plans. There is currently no management of whales on the west coast of Hudson Bay, in Nunavut. In 2002, Inuit there were requested to sell part of their whale harvest to Inuit in Nunavik, northern Quebec, where hunting quotas exist. The outcome of this event was concern in Nunavut for the future of the whale hunt, and a deepening sense of powerlessness in Nunavik due to the management of the whale harvest.

Defining subpopulations using genetics has traditionally used data from microsatellite markers to investigate population structure; however, single‐nucleotide polymorphisms (SNPs) have emerged as a tool for detection of fine‐scale structure. In Hudson Bay, Canada, three polar bear (Ursus maritimus) subpopulations (Foxe Basin (FB), Southern Hudson Bay (SH), and Western Hudson Bay (WH)) have been delineated based on mark–recapture studies, radiotelemetry and satellite telemetry, return of marked animals in the subsistence harvest, and population genetics using microsatellites. We used SNPs to detect fine‐scale population structure in polar bears from the Hudson Bay region and compared our results to the current designations using 414 individuals genotyped at 2,603 SNPs. Analyses based on discriminant analysis of principal components (DAPC) and STRUCTURE support the presence of four genetic clusters: (i) Western—including individuals sampled in WH, SH (excluding Akimiski Island in James Bay), and southern FB (south of Southampton Island); (ii) Northern—individuals sampled in northern FB (Baffin Island) and Davis Strait (DS) (Labrador coast); (iii) Southeast—individuals from SH (Akimiski Island in James Bay); and (iv) Northeast—individuals from DS (Baffin Island). Population structure differed from microsatellite studies and current management designations demonstrating the value of using SNPs for fine‐scale population delineation in polar bears.  相似文献

Narwhal and beluga whales are important species to Arctic ecosystems, including subsistence hunting by Inuit, and little is understood about their mating ecology. Reproductive tract metrics vary across species in relation to mating strategy, and have been used to infer mating ecology. Reproductive tracts from beluga and narwhal were collected between 1997 and 2008 from five beluga stocks and two narwhal stocks across the Canadian Arctic. Tract length for males and females, relative testes mass for males, and tusk length for male narwhal were measured. We assessed variation relative to species, body size, stock, maturity, and season. Significant variation was found in testes mass across month and stock for beluga, and no significant difference between stock or date of harvest for narwhal. Beluga had significantly larger testes relative to body size than narwhal, suggesting they were more promiscuous than narwhal. A significant relationship was found between narwhal tusk length and testes mass, indicating the tusk may be important in female mate choice. No significant differences were found between narwhal and beluga reproductive tract length for males or females. The mating systems suggested for narwhal and belugas by our results mean the two species may respond differently to climate change.  相似文献

Brent geese (called brant in North America) are among the smallest and the most marine of all goose species, and they have very long migration routes between high Arctic breeding grounds and temperate wintering grounds. Like all other geese, brent geese are almost entirely herbivorous. Because of these ecological characteristics they have a high food demand and are strongly dependent on stopover sites to ”refuel” during the migration period. Three subspecies of brent geese are distributed around the Holarctic, forming seven populations with distinct migration routes. Most or all of these populations make heavy use of Zostera spp. during migratory stopovers on spring and/or autumn migration. Examples of Zostera stopover areas being used by large numbers of brent geese for several weeks each year are Izembek Lagoon (Alaska), lagoons in Baja California, the German/Danish Wadden Sea, the Golfe du Morbihan (France), British estuaries, and the White Sea (Western Russian Arctic). Brent geese feed on Zostera wherever they can, but they can only reach the plants at low tide or in shallow water. Changes in Zostera abundance affect brent goose distribution, and the ”wasting disease” affecting Atlantic Zostera stocks during the 1930s was at least partly responsible for a steep decline in brent goose population sizes on both sides of the Atlantic. While Zostera is of outstanding importance as food for brent geese, the impact of the geese on Zostera stocks seems to be less important – at many sites, the geese consume only a small amount of the available Zostera, or, if they consume more, the seagrass can regenerate fully until the following season. Japanese anchovy (Engraulis japonica) is a migratory marine fish of high economic significance in Taiwan. The adult Japanese anchovies migrate from the East China sea to spawn in coastal waters of Taiwan; the larvae then drift back to the East China Sea to complete their life cycle. We developed six highly polymorphic microsatellites for E. Japonica (expected heterozygosity ranging from 0.751 to 0.971) and these microsatellites can be used as genetic markers for identifying stocks to establish regulations in fishing management. Moreover, the markers will be useful in inferring the stock origins and migration routes in the future.  相似文献

In this study the morphometric characteristics based on the truss network system were constructed from the body shapes and used to investigate the population structure of the small yellow croaker (Larimichthys polyactis Bleeker, 1877). From April to June 2013, a total of 431 fish samples were collected from commercial catches acquired with bottom trawlers (20 × 20 mm) in seven main spawning grounds along the coast of China. The results of principal component and discriminant function analyses indicated a clear distinction among the regional stocks and within the Haizhou Bay (HZB) stock. Observed differences primarily involved head measurements, body length and tail size, which were associated with environmental factors. Latitudinal gradient of the stock distribution and the relationship among the stocks provided further evidence for the existence of subpopulations and the re‐division of the East China Sea group. Moreover, the temporal inter‐structure of the HZB stock is a potential reason for the contrast in results from previous studies. Additional thorough studies of migration trajectories are required for a conclusive and improved understanding of the population structure.  相似文献

Humpback whales wintering in tropical waters along the Atlantic and Pacific coasts of the South American continent are thought to represent distinct populations or “stocks.” Here we present the first analysis of genetic differentiation and estimates of gene flow between these breeding stocks, based on both mitochondrial DNA (mtDNA) control region sequences (465 bp) and 16 microsatellite loci from samples collected off Brazil (n = 277) and Colombia (n = 148), as well as feeding areas near the western Antarctic Peninsula (n = 86). We found significant differentiation between Brazilian and Colombian breeding grounds at both mtDNA (FST = 0.058) and microsatellite (FST = 0.011) markers and corroborated previous studies showing genetic similarity between humpbacks from Colombia and those from Antarctic Peninsula feeding areas. Estimates of long‐term gene flow between Brazil and Colombia were low to moderate, asymmetrical, and mostly mediated by males. Assignment procedures detected some cases of interchange and individuals of admixed ancestry between breeding grounds, indicating limited mixing of individuals between these stocks. Overall, results highlight the differentiation of humpback whale breeding populations with adjacent feeding grounds. This appears to be a remarkable example of fidelity to seasonal habitat in the absence of any contemporary barriers.  相似文献

Migratory connectivity plays an important role in conservation of long-distance migrant birds. Here, we study migratory links of dunlin (Calidris alpina), focusing on a stopover and wintering region (Portugal) where it is known that migration routes of dunlin from a broad geographic range (three subspecies) converge, and populations occur simultaneously or separated in time. We combine three methods (ringing recoveries, morphometrics and molecular genetics) to assess breeding origins and extent of temporal segregation of dunlin assemblages. Ringing recoveries show temporal separation of dunlin from different migration routes. Birds found in Portugal during August and September, migrating via Britain, reveal links to breeding areas in Iceland and Greenland. In October, a clear shift to more eastern migration routes occurs, with most Portuguese winter records from stopover sites along migration routes of populations from northern Scandinavia and Russia. Mitochondrial DNA (mtDNA) of Portuguese dunlin was compared with breeding populations. Spring and autumn migrants in Portugal corresponded to C. a. schinzii and C. a. arctica populations, while the Portuguese winter population clearly differs by including mtDNA haplotypes of C. a. alpina. For genetically sexed individuals, we found significant differences in morphology (bill and tarsus length) supporting the temporal separation of populations/subspecies revealed by recoveries and mtDNA. Our results give evidence for migratory connectivity of dunlin populations between geographic areas previously not considered connected. They confirm the existence of clear differences in breeding origin between birds in Portugal at different times of year. These results are important in the consideration of future long-term conservation plans.  相似文献

Caribou (Rangifer tarandus) has been fundamental to the diet and culture of Arctic Indigenous Peoples for thousands of years. Although caribou populations observe natural cycles of abundance and scarcity, several caribou herds across the Circumpolar North have experienced dramatic declines in recent decades due to a range of interrelated factors. Broadly, the objectives of this study are to examine food and nutrition security in relation to wildlife population and management status across Inuit Nunangat (the Inuit homeland, consisting of four regions across the Canadian Arctic). Specifically, we: (1) characterize the contribution of caribou to Inuit nutrition across northern Canada and (2) evaluate the population and management status of caribou herds/populations harvested by Inuit. Dietary data were derived from the 2007–2008 Inuit Health Survey, which included dietary information for Inuit adults (n?=?2097) residing in thirty-six communities, spanning three regions (the Inuvialuit Settlement Region, Nunavut, and Nunatsiavut) of the Canadian North. Published information regarding the range, abundance, status, and management status of caribou herds/populations was collected through document analysis and was validated through consultation with northern wildlife experts (territorial governments, co-management, and/or Inuit organizations). While caribou contributed modestly to total diet energy (3–11% of intake) across the regions, it was the primary source of iron (14–37%), zinc (18–41%), copper (12–39%), riboflavin (15–39%), and vitamin B12 (27–52%), as well as a top source of protein (13–35%). Restrictions on Inuit subsistence harvest (harvest quotas or bans) are currently enacted on at least six northern caribou herds/populations with potential consequences for country food access for over twenty-five Inuit communities across Canada. A holistic multi-sectorial approach is needed to ensure the sustainability of wildlife populations, while supporting Inuit food and nutrition security in the interim.  相似文献

The spatial variation in zooplankton biomass, abundance andspecies composition in relation to hydrography and chlorophylla (Chl a) was studied in the subarctic waters of Hudson Bayand Hudson Strait. Sampling was carried out in early September1993 at 21 stations arranged along a transect following theQuébec coast from James Bay, in Hudson Bay, to the vicinityof Ungava Bay in Hudson Strait. Both the biomass and the abundanceof total zooplankton were low along the lower part of HudsonBay (averaging 1.6 g DM m–2 and 9432 ind. m–2) andincreased sharply toward the upper end of the Bay and in HudsonStrait (averaging 6.0 g DM m–2 and 40 583 ind. m–2).A total of 80 zooplankton taxa was identified in the samples.Copepods were clearly numerically dominant at all sampling stations,accounting for more than 85% and 93% of the zooplankton communityin the Bay and the Strait, respectively. Clustering samplesby their relative species composition revealed four groups distributedalong well defined environmental gradients characterizing thedistribution of physical variables and Chl a. The first group,located in the most southern region of Hudson Bay and fartheroffshore, northwest of the Belcher and Sleeper Islands, wasstrongly influenced by freshwater run-off from James Bay andother major rivers around the Bay, and was characterized bythe presence of two euryhaline copepod species (Acartia longiremisand Centropages hamatus). The second and the third groups occupiedthe largest region along the sampling transect, from the middleof Hudson Bay to the western region of Hudson Strait, and werecharacterized by a typical arctic zooplankton fauna relatedto the cyclonic circulation in central Hudson Bay. The fourthgroup was located in the easternmost part of the sampling transectin Hudson Strait where the highest phytoplankton biomass valueswere observed (Chl a ~220 mg m–2). The zooplankton assemblagethere showed an important increase in the abundance of the largeherbivorous copepod Calanus glacialis/finmarchicus, which werenumerically four times more abundant in the central Strait region(averaging 15 251 ind. m–2) than in the western side ofHudson Strait and in Hudson Bay (3629 ind. m–2). Theseresults support the hypothesis that the structure in the localbiological community is influenced by the local hydrodynamicfeatures which, through their action on surface water temperature,salinity, stratification and mixing conditions, lead to spatialdifferentiation of the phytoplankton and zooplankton communities.  相似文献

There is a need for biological information to support current stock designations of bottlenose dolphins (Tursiops truncatus) in the Gulf of Mexico. The existence of many inshore, resident “communities” raises questions as to the relationship these dolphins may hold with dolphins inhabiting neighboring inshore and coastal areas. In this study, population subdivision was examined among four resident, inshore bottlenose dolphin stocks (Sarasota Bay, FL, Tampa Bay, FL, Charlotte Harbor, FL and Matagorda Bay, TX) and one coastal stock (1–12 km offshore) in the Gulf of Mexico. Evidence of significant population structure among all areas was found on the basis of both mitochondrial DNA (mtDNA) control region sequence data and nine nuclear microsatellite loci. Estimates of relatedness showed no population contained a significantly high number of related individuals, while separate AMOVAs for males and females indicated that both sexes exhibit a significant level of site philopatry. Results presented here provide the first genetic evidence of population subdivision between the coastal Gulf of Mexico and adjacent inshore areas along the central west coast of Florida. Such strong genetic subdivision is surprising given the short geographical distance between many of these areas and the lack of obvious geographic barriers to prevent gene flow. These findings support the current, separate identification of stocks for bottlenose dolphins inhabiting the eastern coastal and inshore areas of the Gulf of Mexico.  相似文献

Mitochondrial DNA (mtDNA) from endosymbiote-free stocks of Paramecium tetraurelia was isolated by 2 procedures. the buoyant density of the mtDNA in neutral CsCI was 1.702 gm/cm3. a value consistent with the melting temperature of the mtDNA. Only linear molecules were observed by electron microscopy. These molecules were homogeneous in size with a monomer molecular weight of 25.6 × 106 daltons. the size of the mtDNA determined after digestion with the restriction endonucleases EcoRI or Hind III agreed with the value obtained by electron microscopy. These studies also revealed that the digestion pattern of mtDNA from stock 172 differed from that of the other 3 stocks (51, 127. 203) examined. Some mtDNA molecules exhibited snapback reassociation following denaturation.  相似文献

The brown-marbled grouper, Epinephelus fuscoguttatus, is a long-lived, late-maturing protogynous species listed as Near Threatened by the IUCN. In Pohnpei, Micronesia, reproductively active brown-marbled grouper were tagged with acoustic and spaghetti-type tags at a multi-species fish spawning aggregation (FSA) site to establish patterns of movement, residency and seasonality. Telemetry confirmed the use of common reproductive migratory corridors and significant sex-specific variations in residency at the FSA. Combined underwater visual census and telemetry data verified a 3-month peak aggregation period, with aggregations forming and persisting over ca. 12 days prior to full moon between January and May. FSA formation coincided with seasonally low and relatively stable seawater temperatures. Some males frequented the FSA site during each aggregation month over two consecutive years. Conversely, most females were present at the FSA during only a single aggregation period, with the month of visitation consistent among years. Nearly two-thirds of tagged fish were relocated or recaptured within 11 km of the aggregation site, with a maximum detected distance of 26 km and a minimum estimated catchment area of 100–175 km2. Findings highlight the need for a combined approach to management that prohibits the capture and sale of reproductive adults and protects both spawning sites and common reproductive migratory corridors during aggregation periods.  相似文献

The worldwide distribution of blue whales (Balaenoptera musculus) has not prevented this species from becoming endangered due to twentieth century whaling. In Australia there are two known feeding aggregations of blue whales, which most likely are the pygmy subspecies (B. m. brevicauda). It is unknown whether individuals from these feeding aggregations belong to one breeding stock, or multiple breeding stocks that either share or occupy separate feeding grounds. This was investigated using ten microsatellite loci and mitochondrial DNA control region sequences (N = 110). Both sets of markers revealed no significant genetic structure, suggesting that these whales are likely to belong to the same breeding stock.  相似文献

Populations of anadromous European smelt Osmerus eperlanus (L.) are declining across its geographical range in northern Europe, but no practical culture techniques exist to develop stock enhancement programmes for this species. In this study, a culture protocol is described to rear fish from fertilised eggs to mature adults in 2 years involving the use of ‘green water’, live feed and artificial diets. The sequence of embryonic development for eggs incubated at 10°C/0 ppt was described and photographed. To determine the optimum conditions for larval culture, fertilised eggs were reared at a range of salinities (0–20 ppt) and temperatures (5–18°C) until first feeding. Best hatching success (ca. 97%), size at hatch (ca. 0.8 mm) and survival to first feeding (ca. 96%) of larvae were achieved under combined conditions of low salinity (0–0 ppt) and temperature (5–10°C). No larvae survived a salinity of 20 ppt. The time taken from fertilisation to hatch (FtH) and hatching duration (HD) were temperature-dependent ranging from 42 days FtH and 10 days HD at 5ºC, to 10 days FtH and 2 days HD at 18°C irrespective of salinity. The results indicate that conservation programmes could utilise existing salmonid hatchery facilities (i.e. freshwater, ≤10°C water temperature) for stock enhancement. Since on-growing of smelt involves the logistical and technical problems of live feed production, it is recommended that smelt enhancement programme utilise freshwater hatchery facilities to rear fish until hatching, and then stock out onto known spawning grounds in rivers allowing hatched larvae to drift into estuaries to complete the larval and juvenile phases. This approach would minimise the time spent in the hatchery post-hatching, eliminate the need for live food production, prevent the development of predator-naïve fish, and hence would mimic the natural life cycle of the species as closely as possible.  相似文献

The spatial distribution of genetic diversity has been long considered as a key component of policy development for management and conservation of marine fishes. However, unraveling the population genetic structure of migratory fish species is challenging due to high potential for gene flow. Despite the shallow population differentiation revealed by putatively neutral loci, the higher genetic differentiation with panels of putatively adaptive loci could provide greater resolution for stock identification. Here, patterns of population differentiation of small yellow croaker (Larimichthys polyactis) were investigated by genotyping 15 highly polymorphic microsatellites in 337 individuals of 15 geographic populations collected from both spawning and overwintering grounds. Outlier analyses indicated that the locus Lpol03 might be under directional selection, which showed a strong homology with Grid2 gene encoding the glutamate receptor δ2 protein (GluRδ2). Based on Lpol03, two distinct clusters were identified by both STRUCTURE and PCoA analyses, suggesting that there were two overwintering aggregations of L. polyactis. A novel migration pattern was suggested for L. polyactis, which was inconsistent with results of previous studies based on historical fishing yield statistics. These results provided new perspectives on the population genetic structure and migratory routes of L. polyactis, which could have significant implications for sustainable management and utilization of this important fishery resource.  相似文献
