The impact of hypophysectomy on the state of the interrenal gland and ultrastructure of chloride cells of gills is investigated in 18-month old juveniles of starred sturgeon Acipenser stellatus in the process of its adaptation to artificial sea water (14.6‰). Hypophysectomized juveniles, similarly to intact juveniles, are able to support a relative stability of osmolarity of blood serum in the course of adaptation to sea water by transition from hyperosmotic to hypoosmotic type of osmoregulation. Changes in the investigated parameters of cells of the interrenal gland (volume of nuclei, areas of cells and of lipophilic vacuoles) occurring in the hypophysectomized and in intact specimens in the process of adaptation to sea water are generally similar, but have different dynamics. In contrast to many teleostean species, in acipenserids the hypophyectomy does not cause atrophy of the interrenal gland. The latter is incorporated in the process of regulation in the course of adaptation of fish to sea water. Hypophyecotmy results in partial destruction of organoids in some chloride cells of gills. However, when the fish are transferred to sea water, the structural changes occur in chloride cells characteristic of their transition to the excretory state. This may happen only at activation of the transport enzyme Na+/K+-ATPase of these cells by cortisol produced by the interrenal gland. In the absence of hypophysis, the functional connection of organs of the axis hypothalamus (ACTH-immunopositive cells of tuberal nucleus) → the interrenal gland → chloride cells is realized in the fish.  相似文献

The ultrastructure of the mid-intestine of four sturgeon species and their hybrids was studied. Apart from enterocytes and glandular goblet cells, the cells with microvilli, endocrine cells, and immunocompetent elements were revealed. Specific cells responsible for the primary immune response, analogous to mammalian M-cells, were discovered in sturgeon fishes. The ultrastructural characteristics common for the mid-intestinal epithelium of all the sturgeon fishes studied were determined. Species-specific features of intestinal epithelium of the parent species, which were characteristic of the hybrids, were revealed.  相似文献

The salinity tolerance, and hydromineral regulation capabilities of three size groups (small 110–170 g; medium 230–290 g, large 460–700 g; n=48 for each group) of 13-month-old juvenile Gulf of Mexico sturgeon were investigated. Fish (n=6 for each salinity) were transferred directly from freshwater (FW) to a series of experimental salinity treatments (0, 5, 10, 15, 20, 25, 30, and 35 parts per thousand (ppt)). Fish were also acclimated in brackish water (20 ppt) for 2 weeks and transferred to a salinity of 34 ppt. In this condition juvenile Gulf of Mexico sturgeon adapted to saltwater (SW) and maintained their hydromineral balance. FW adapted sturgeon (n=6) had an average blood hemotocrit of 28.2±0.8%, plasma osmolality of 260.7±1.6 mOsm kg−1 H2O, and plasma ion concentrations of 135.7±1.2 mM l−1 Na+, 106.9±1.9 mEq l−1 Cl, and 2.9±0.1 mM l−1 K+. In SW adapted sturgeon (n=8) blood parameters averaged 26.9±0.7% for hematocrit, 294.2±2.3 mOsm kg−1 H2O for osmolality, 152.0±1.7 mM l−1 Na+, 149.2±1.4 mEq l−1 for Cl, and 3.1±0.1 mM l−1 K+. The method of transfer (abrupt or slow acclimation) directly affected fish survival and the time they took to achieve ionic and osmotic regulation. This SW adaptation appears to be related to body size, the larger the fish the easier the adaptation process. A threshold size of about 170 g was apparent for the fish to adapt to saltwater after 2 weeks of acclimation. Chloride cells were present in both FW and SW adapted sturgeon with SW and brackish water fish having chloride cells significantly (P<0.05) more numerous (561±53 and 598±45 cells mm−2) and larger in size (41.0±3.85 and 34.2±4.49 μm2) than FW adapted sturgeon (10±1.0 cells mm−2 and 22±2.53 μm2). Few chloride cells were observed in the opercular membrane, however, none were found in the pseudobranch and spiracle.  相似文献

For the first time by the example of juveniles of Russian sturgeon Acipenser gueldenstaedtii Brandt, the role of growth hormone of hypophysis in the osmotic regulation of acipenserids was studied, also the process of transformation of reserve chloride was described. In Russian sturgeon juveniles, administration of growth hormone of carp Cyprinus carpio at a dose of 1 mg per 1 g body weight causes changes in the fine structure of numerous reserve (undifferentiated) gill epithelial cells and thus stimulates their transformation into mature chloride cells involved in transport of monovalent ions. Increase in the number of chloride cells, on account of reserve cells, was accompanied by higher enzymatic activity of Na+/K+-ATPase in gill homogenates of the injected fish adapting to a hyperosmotic environment of salinity 12.5‰ (403 mosm/l) than in the intact fish under the same conditions but without hormonal treatment. Morphophysiological changes observed in the injected juveniles provided the more efficient dynamics of blood serum osmolarity under saline exposure than that in the intact fish. Their serum osmolality during the maximum rise (24 hours after the transfer of fish into the water of 12.5‰ salinity) was lower (p <0.01), than that of the intact fish. The inclusion of growth hormone in the osmoregulatory process was established.  相似文献

Cells with the ultrastructure typical of steroid cells of vertebrates were found in the theca and stroma of the Russian sturgeon ovary. Mitochondria with the tubulo-vesicular internal structure, smooth and rough endoplasmic reticulum, lipid droplets or liposomes were observed in the cytoplasm of these cells. After the ovulation of fishes, the volumetric density and mean diameter of cell mitochondria and of the reticulum are found to increase; besides, mitochondria are seen vacuolized. The ultrastructural changes may indicate that the production of steroid sex hormones is more intensive in fish during ovulation. These suggestions are in accordance with biochemical data available in the literature.  相似文献

Electron-microscopic examinations of the sturgeon gut were performed. Oesophageal goblet cells were abundant in the stratified epithelium. The ultrastructural features of the secretory granules of the oesophageal and intestinal goblet cells were quite similar to those of other vertebrates. Lobules of multilocular adipose tissue were observed in the deep tunica propriasubmucosa of the oesophagus, in close association with vasculature and large fibre bundles of myelinated and unmyelinated axons. Similarly composed nerve fibre bundles were observed in the cardiac stomach, too. The presence of myelinated axons is an unusual feature in the vertebrate enteric nervous system. Cardiac and fundic zones of the stomach showed an epithelium with columnar ciliated and non-ciliated cells, the latter equipped with fuzzy microvilli. Cells lining the tubular gastric proper glands were markedly granulated. Intestinal superficial epithelium was columnar and contained ciliated, as well as non-ciliated and goblet cells. In the tunica propria all over the intestine, the presence and ultrastructure of granulated cells was in addition described. Intraepithelial granulated leukocytes were seen throughout the alimentary canal. Various types of endocrine cells were seen both in the stomach and in the intestine, the size of their granules was measured and their ultrastructure described and compared to that of mammalian cell types.  相似文献

The aim of the present work was to compare the structure and protein composition of centrioles from spermatozoa of sturgeon and salmon fishes. The total protein content of the extracted fractions was studied by Na-SDS electrophoresis. Proteins with molecular weights from 15 to 170 kDa were detected. In both cases the major protein of centrioles is a protein with a molecular weight equal to that of tubulin. A protein with the molecular weight corresponding to actin was also detected. In both cases the ATPase activity stimulated by Ca2+ and Mg2+ ions was revealed. Electron microscopic studies showed differences in the ultrastructure of centrioles from sturgeon and salmon spermatozoa.  相似文献

The Chinese sturgeon (Acipenser sinensis Gray 1835) is an endangered anadromous sturgeon inhabiting the Yangtze River in China. In this study, the ultrastructure and morphology of spermatozoa was studied using transmission and scanning electron microscopy with a cryo-holder. The spermatozoon consisted of an elongated head with a distinct acrosome and nucleus region, a midpiece and a flagellum. The mean length of the head and midpiece, the flagellum and total length of spermatozoon were 4.48, 33.3 and 37.8 microm, respectively. The nucleus was an elongated trapezoid shape with anterior (acrosome) end narrower than the posterior. Granular material and an actin filament were observed within the anterior acrosome. Three to five endonuclear canals were present. The midpiece was eudipleural along its longitudinal axis. Compared to other sturgeon species, the data from the present study suggest a more recent evolutionary linkage between Chinese sturgeon and white sturgeon (Acipenser transmontanus Richardson 1836).  相似文献

The sturgeon stock in the Sea of Azov is presently maintained by artificial reproduction allowing for the harvest of sturgeons in this basin. Shortened spawning migrations in regulated rivers and long-term artificial selection of the ripest fish at the hatcheries have resulted in high functional maturity of sturgeon spawners in the sea near the river mouth. The traditional technology of artificial sturgeon reproduction is non-effective under modern ecological conditions. The use of ecological and hormonal methods to control seasonal propagation of stellate sturgeon, A.stellatus , Russian sturgeon, A.gueldenstaedti , beluga, Huso huso , and sterlet, A.ruthenus , make it possible to stagger the sexual cycle of breeders from the various biological groups for early (5 months) or later (6 months) spawning. Use of these methods preserves the natural heterogeneity of sturgeon populations maintained through stocking, and also allows for a considerable increase in the quantity of fry reared at the same production base.
The use of the synthetic analogue of mammalian gonadotropin-releasing hormone (Gn-RHa) is better for inducing maturity of the ripest brood fishes, especially for stellate sturgeon, in comparison with the traditional pituitary injections. The schedule of injections was determined for the various species and seasons of spawning.
Short-term rearing of larvae in ponds (no more than 15 days) and subsequent adaptation to sea water in brackish lagoons (1–5 %o) allows for repeated use of nursery ponds in sturgeon hatcheries and a considerable increase in the quantity and survival of sturgeon juveniles in the sea.
The restoration of the sturgeon population structure in the Sea of Azov is not possible without developing a genetic collection of the fish under a controlled culture regime.  相似文献

The physiological state of the pigment was dependent on the level of light/dark adaptation. The oocytes of many organisms, including frogs and fish, contain a distinct cytoplasmic organelle called the Balbiani body. Because of the scarcity of published information and the tremendous variability in the appearance, ultrastructure, and composition of Balbiani bodies between species, the function of the Balbiani body and its inter-species homology remain a mystery. In Xenopus laevis, the Balbiani body is known to play a role in transporting germ cell determinants and localized RNAs to the oocyte vegetal cortex. In fish, however, the molecular composition of the Balbiani body has not been studied to date, and its function remains completely unknown. We have studied the ultrastructure and molecular composition of previtellogenic oocytes of the sturgeon, Acipenser gueldenstaedtii, by using electron microscopy, in situ hybridization, and immunostaining. We have found that sturgeon oocytes contain two distinct zones of cytoplasm: homogeneous (organelle-free) and granular (organelle-rich). We have also found that the granular ooplasm, which we term the Balbiani cytoplasm, shares important homologies, in both ultrastructure and molecular composition, with Xenopus Balbiani bodies. Photosynthesis is known to provide nearly all the carbon and chemical energy needed for plant growth, it depends on many environmental factors and alternates when these factors fluctuate. The degree of the chloroplast membrane system development can be, to a certain extent, an indicator of the organelles' photosynthetic activity. To-date, changes in chloroplast size and ultrastructure as well as starch and pigment content in leaf mesophyll cells in microgravity have been found in variety of the angiosperm species investigated in this respect. However, available data are very limited and contradictory. Taking into account the importance of studying the photosynthesis process to elucidate the possibilities of plant physiological adaptation in altered gravity that is the basis for working out the technologies of space planting in controlled ecological life-support systems, we conducted the investigations of ultrastructure and state of the photosynthetic apparatus in Arabidopsis thaliana leaf mesophyll cells at the different stages of plant development under clinorotation.  相似文献

Effect of the tretionine (retinoid) and aluminum chloride (neurotoxin) on the growth and differentiation of neuroblastoma cells in culture after their introduction into the medium separately and in combination was studied. The introduction of these substances creates a new information field in the medium, which becomes apparent by the reactions of neuroblastoma found on the populational and cellular levels of its organization. The presence of tretionine stimulates proliferation and induces differentiation of the cells into astrocytes. Aluminum chloride inhibits cell proliferation and enhances the process of their destruction in the monolayer. The variety of the reactions of neuroblastoma cells to the presence of these substances in the medium indicates the existence and functioning of a mechanism that selects from the information introduced only the portion which may contribute to adaptation of neuroblastoma cells to the changed culture conditions.  相似文献

A comparative analysis of the cellular ultrastructure and pigment content of both underwater and above-water Sagittaria sagittifolia leaves was conducted by means of the method of transmission electron microscopy and spectrophotometric analysis. A comparative study of the levels of heterogeneity and polymorphism of albumins, the most important functionally specialized blood proteins, has been carried out. The albumin system of Russian sturgeon undergoes distinct changes while the fish change habitat during the spawning migration from sea to river. They are expressed as an increased level of heterogeneity, an increased content of serum albumins in fish during the river period of life as compared to the sea period, and an increased share of a slow component of albuminograms. These changes suggest a significant role of the blood albumin system in adaptation of the Caspian sturgeon migratory species to fresh water life conditions.  相似文献

Study of the dilate sea-water tolerance of the pipefish Syngnathus acusimilis indicated that it successfully acclimatized to an abrupt change in water salinity from 5 to 32‰. Larvae in the brood pouch also successfully acclimatized to salinity change, and, after several days, juveniles left the brood pouch and continued to live. In freshwater, adult and fry perished during several minutes. In the gill epithelium of S. acusimilis, chloride cells of only a marine type were found. A comparison of the ultrastructure of chloride cells of gill epithelium and mitochondrion-rich cells of the brood pouch of S. acusimilis demonstrated their high similarity. The ultrastructure of cells of the brood pouch indisputably attests to their involvement in maintaining an osmotic and ionic homeostasis in the cavity of the brood pouch.  相似文献

Calcium regulation in sturgeon is of special interest because they are a representative of the ancient fishes possessing mainly cartilaginous skeletons and a supposedly low calcium demand. The present study aimed to characterize the effect of a chronic absence of dietary calcium and the effect of parathyroid hormone-related protein (PTHrPA) (1-34) (7) on calcium balance in juvenile sturgeon (Acipenser naccarii). At rest, sturgeon juveniles are in net positive calcium balance, since whole body calcium uptake is significantly higher than efflux and calcium accumulates in the body. To study the importance of dietary calcium, the sturgeon were kept on a calcium-free diet for 8 wk. This manipulation impaired growth as measured by failure to gain weight or increase in length and indicates that dietary calcium is important for growth in sturgeon. An increased whole body calcium uptake partially compensated dietary calcium deficiency and was associated with increased gill chloride cell number in lamellae and filaments in parallel with increased gill Na(+)K(+)-ATPase activity. In addition, a single injection of piscine PTHrP(1-34) significantly increased whole body calcium uptake and decreased whole body calcium efflux. Administration of PTHrP significantly increased circulating plasma calcium 4-24 h postinjection. The increase in net calcium transport and increased plasma levels of calcium is consistent with the actions of a hypercalcemic factor. It would appear that the sturgeon rely on calcium for growth and tightly regulate calcium transport. The action in calcium balance is consistent with PTHrP acting as a hypercalcemic factor in sturgeon.  相似文献

Changes were measured in the numbers of chloride cells and the levels of (Na+ + K+)-DEPENDENT ATPase in the gills of immature, yellow eels and mature, silver eels during adaptation from freshwater to seawater. The percentage of chloride cells in yellow eels more than doubled after six days in seawater; at this time the specific activity and concentration of (Na+ + K+)-dependent ATPase in gills start to increase in parallel to reach maxima after two weeks that are 2.5 times the starting values. It is concluded that adaptation of yellow eels to seawater involves an increase in the numbers of chloride cells in gills as well as an increased amount of (Na+ + K+)-dependent ATPase per chloride cell. Mature silver eels in freshwater had essentially the same numbers of chloride cells and the same specific activity of the enzyme in the gills as yellow eels fully adapted to seawater. Transferring silver eels to seawater did not alter the percentage of chloride cells in gills although the level of (Na+ + K+)-dependent ATPase and its specific activity increased slightly. Thus, although the silver eel is better prepared for life in seawater than the yellow eel, it still has to attain an increased level of (Na+ + K+)-dependent ATPase in its chloride cells to be fully adapted to seawater.  相似文献

The paper presents results of a cytochemical study of localization of phosphatase responsible for hydrolysis of inositol 1,4,5-triphosphate (ITP) in the olfactory lining of true sturgeons (the sturgeon, starred sturgeon, and sterlet). Reaction products as a dark discrete granules are localized in the apical parts of epithelium, practically in the same manner in all the species studied. The precipitate is found on the plasma membranes of cilia, microvilli, and clava of the olfactory cells. Occasionally, the precipitate is also found in the cilia, basal bodies, and rootlets of microvillar cells. The ITP-hydrolyzing phosphatase is supposed to restrict development of transduction process by removing excess messengers from the operating system. The data obtained indicate that in the true sturgeons, the phospholipase cascade of olfactory transduction is concentrated predominantly in the cilia and microvilli of olfactory cells.  相似文献

The critical thermal maximum (CTmax) and the associated hematological response of juvenile (~145 g, n = 8 for both species) Atlantic Acipenser oxyrinchus and shortnose Acipenser brevirostrum sturgeons acclimated to 15°C were determined using a heating rate of 8°C h?1. The critical thermal maximum averaged 30.8°C and 31.6°C for Atlantic and shortnose sturgeon, respectively, and values fell within the range noted for other sturgeon species. Oxygen‐carrying capacity (hemoglobin and hematocrit) measures were generally unaffected by thermal stress. Plasma lactate levels increased from 0.5 mm to 4 mm following temperature stress in both species. Both plasma glucose and potassium levels increased following CTmax, however, these levels were about double in the shortnose sturgeon. Lastly, plasma sodium and chloride levels were significantly depressed (by more than 10%) following thermal stress in shortnose sturgeon, whereas only chloride levels decreased in Atlantic sturgeon. Taken together, while CTmax values were similar, thermal stress resulted in different hematological profiles; these differences are consistent when compared to other stressors, and may be related to the phylogenetic position and thus could reflect the evolutionary history of these two species.  相似文献
