The complex post-translational processing of concanavalin A (Con A) in maturing jackbeans is unique because the non-glycosylated mature active protein is circularly permuted in primary sequence relative to its own inactive precursor (glycosylated pro-Con A) and to other legume lectins. We show here that non-glycosylated pro-Con A expressed in bacteria from recombinant cDNA (rec-pro-Con A) folds in vivo and in vitro to a stable form which is active without further processing. N-glycosylation alone must therefore be sufficient to inactivate pro-Con A--a novel role for glycosylation in regulating activity during protein maturation.  相似文献

Non-degradative ubiquitylation plays a crucial role in many cellular signaling pathways, including the DNA damage response. Two ubiquitin ligases, RNF8 and RNF168, in combination with the E2 ubiquitin conjugating enzyme UBC13 catalyze the formation of K63-linked ubiquitin chains at sites of DNA double-strand breaks to promote their faithful repair. However, little is known about their negative regulation. A recent study identifies a deubiquitylating enzyme, OTUB1, which counteracts RNF8/RNF168-dependent ubiquitin chain formation at break sites. Surprisingly, this enzyme carries out its function not by cleavage of polyubiquitin chains, but by targeting UBC13. This non-canonical role for a deubiquitylating enzyme has implications for the regulation of ubiquitylation not just in DNA repair, but potentially in many other cellular signaling processes.  相似文献

Biofuels are an attractive means to prevent a further increase of carbon dioxide emissions. Currently, gasoline is blended with ethanol at various percentages. However, butanol has several advantages over ethanol, such as higher energy content, lower water absorption, better blending ability, and use in conventional combustion engines without modification. Like ethanol, it can be produced fermentatively or petrochemically. Current crude oil prices render the biotechnological process economic again. The best-studied bacterium to perform a butanol fermentation is Clostridium acetobutylicum. Its genome has been sequenced, and the regulation of solvent formation is under intensive investigation. This opens the possibility to engineer recombinant strains with superior biobutanol-producing ability.  相似文献

Multiple states of differentiation developed within the same undivided egg cytoplasm of ascidian zygotes cleavage-arrested with cytochalasin B. Complex ultrastructural traits of up to four quite diverse cell lineage components were observed in regions of the common cytoplasm in such multinucleate homokaryons of Ciona intestinalis: epidermal, muscle, notochordal, and neural. Almost all specimens among those selected as showing differentiation contained two such features, half of them had at least three, and a few expressed all four. The histospecific morphological characteristics noted were the extracellular test material of epidermal cell origin, muscle myofilaments and myofibrils, sheath components (leaflets and filaments) associated with notochordal cells, and the particular localized combinations of microtubules, filamentous structures, and cilia indicative of neural tissues. Cleavage-arrested one-celled embryos of Ascidia ceratodes served to demonstrate that those which were found cytochemically to contain muscle acetylcholinesterase always had myofibrils and myofilaments. Other arrested zygotes of Ascidia (unstained specimens) also had quite fully formed test material as well as myofilaments and myofibrils. The occurrence within the same cell of so many specific markers of diverse pathways of development is consistent with a theory about a primary level of regulation based on autonomous gene activation factors already present in the fertilized egg. If further investigation substantiates a real cytoplasmic continuity within these cleavage-arrested embryos, other theories that invoke cell interactions, temporal sequences of metabolically distinct microenvironments, and gradients of substances as causes of determinative change seem inadequate to account for the coexisting expressions of differentiation described here.  相似文献

Photoreaction of zinc methyl 20-substituted meso(pyro)pheophorbide-a prepared by modifying naturally occurring chlorophyll-a in the presence of oxygen molecules gave its C19-C20 oxidative cleavage (1-carbonyl-19-oxo-bilatrienes) as the major products and the regioisomeric C1-C20 cleavage (19-carbonyl-1-oxo-bilatrienes) as the minor products. The resulting zinc complexes of linear tetrapyrroles took a helical conformation and the P-conformers were preferential over the M-stereoisomers due to the presence of their 17S,18S-chiral centers. The helical conformers (diastereomers) of the corresponding nickel complexes were separated by reverse-phase or chiral HPLC and their conformational changes were observed in solution.  相似文献

Sexually selected male courtship displays often involve multiple behavioural and physical traits, but little is known about the function of different traits in mate choice. Here, we examine female courtship behaviours to learn how male traits interact to influence female mating decisions. In satin bowerbirds (Ptilonorhynchus violaceus), successful males give highly aggressive, intense behavioural displays without startling females. Males do this by modulating their displays in response to female crouching, which signals the display intensity that females will tolerate without being startled. Females typically visit multiple males for multiple courtships before choosing a mate, and females show differing tolerance for intense displays during their first courtship with each male. We test three hypotheses that may explain this: (i) familiarity with the courting male; (ii) the order of the courtship in mate-searching; and (iii) the attractiveness of the courting male. We found that females are more tolerant of intense displays during first courtships with attractive males; this increased female tolerance may allow attractive males to give higher intensity courtship displays that further enhance their attractiveness. We then examined why this is so, finding evidence that females are less likely to be startled by males with better physical displays (bower decorations), and this reduced startling then contributes to male courtship success. This role of physical displays in facilitating behavioural displays suggests a novel mechanism by which multiple physical and behavioural traits may influence female choice.  相似文献

Fluctuating asymmetry in bilateral traits has been proposed to reflect aspects of individual quality, and has hence been suggested to act as a cue in mate choice. Since sexual selection generally acts more strongly on males, numerous studies have focused on female preferences for symmetrical partners, while very little is known about potential male symmetry preferences. In the present experiment, we tested whether bluethroat males are sensitive to symmetry in an artificial ornament. Using different combinations of blue and orange leg bands, females were made symmetrical or asymmetrical. In outdoor aviaries placed in breeding habitat, males were allowed to choose between a symmetrical and an asymmetrical female. We found that males associated more with symmetrical than asymmetrical females, indicating a preference for symmetry. The magnitude of the symmetry preference of bluethroat males was similar to that of females of the same species.  相似文献

In Escherichia coli cells carrying wild-type ovalbumin cDNA, some of the recombinant protein was secreted into the periplasmic space. In contrast, a signal-region mutant form of ovalbumin (deletion, Gly1 to Ala39) was not detected in the periplasm despite being synthesized at the same level as the wild-type protein. Chemical and spectroscopic analyses showed that periplasmic ovalbumin assumes a conformation indistinguishable from that of native egg white ovalbumin. We concluded that a process resembling the secretion of ovalbumin process in the oviduct occurs also in bacteria.  相似文献

Activation of unfertilized Chaetopterus eggs by treatment with an excess of KCl may lead to the production of unicellular ciliated larvae (Lillie's differentiation without cleavage). The effects of a number of inhibitors of protein (puromycin, cycloheximide, emetin), RNA (actinomycin D), and DNA (hydroxyurea) synthesis of differentiation without cleavage and on normal development have been studied in Chaetopterus. Incorporation of radioactive leucine, uridine, and thymine has been followed by biochemical methods and by autoradiography. The DNA content of the large polyploid nucleus has been estimated by cytophotometry. The initial pseudocleavage period of differentiation without cleavage is characterized by a burst in DNA and protein synthesis; the inhibitors have little or no effect on this burst and on pseudocleavage itself. Protein and DNA synthesis levels off during the following phase (segregation), but the inhibitors become more effective. RNA synthesis is almost linear for 20 h. These results are compared with those obtained on eggs from other species.  相似文献

Synthetic myosin filaments with regular projections at intervals of 430 Å along their entire lengths have been observed using the electron microscope. The filaments were formed following dialysis in or rapid dilution to 0.3 m-KC1, 0.01 m-imidazole or phosphate buffer, pH 7.0. The effects of aphidicolin - a powerful inhibitor of DNA polymerase alpha and of DNA replication - on normal development and on differentiation without cleavage of Chaetopterus eggs have been studied with cytological, cytochemical, and biochemical methods. The experiments show that the initial period of pseudocleavage can take place in the absence of nuclear DNA synthesis, but further development (segregation, hatching, ciliation) requires DNA synthesis. However ciliated unicellular larvae can be obtained under conditions where the DNA content of the embryos in only 40% of the controls. In fertilized eggs, aphidicolin immediately stops cleavage. The significance of these results is discussed.  相似文献

1. A behavioural bioassay, based on antennal movement responses, was developed using Glossina morsitans morsitans Westwood for screening chemical attractancy to tsetse. 2. Chemicals found to be attractive to male tsetse were acetone, formaldehyde, methylethylketone, methylvinylketone, 1-octen-3-ol and pentanal but not acetophenone, hexanal, lactic acid or urea. 3. Female tsetse also responded to all these chemicals in a similar fashion. Overall responses of females were, however, less than those of males. 4. These laboratory bioassay findings agree with field observations on tsetse responses to certain chemical odours. Therefore this behavioural bioassay should be a useful laboratory test procedure for screening attractants.  相似文献

Urea treatment of the big form of somatostatin obtained from rat pancreas resulted in a conversion into the small form of somatostatin. Further dissociation does not occur with mercaptoethanol. The results indicate that the existence of big somatostatin is dependent upon the formation of a non-covalent bond of the tetradecapeptide with another peptide fragment.  相似文献

AtT-20 cells, which were derived from a murine pituitary tumor and produce ACTH, have until now been considered to originate from pituitary corticotrophs. Here we show that AtT-20 cells constitutively express several neuronal features. First, AtT-20 cells develop cytoplasmic processes whose fine structure is essentially identical to that of neurites and neuronal growth cones. These growth cones (i) are characterized by an extensive membranous reticulum which is derived from the endoplasmic reticulum (ER) since it contains immunoglobulin heavy chain binding protein, protein disulfide isomerase and glucose-6-phosphatase; (ii) are a major site of endocytosis; (iii) form cell-to-cell contacts resembling immature synapses. Second, AtT-20 cells, in contrast to pituitary corticotrophs, contain neurofilaments and express all three neurofilament polypeptides. They also contain the high molecular weight form of microtubule-associated protein 2 and tau protein. Third, AtT-20 cells express the neuron-specific phosphoprotein synapsin I which accumulates in the growth cones prior to contacts forming between growth cones and cells. Our results show that AtT-20 cells exhibit several properties of peptidergic neuronal cells and that the constitutive expression of a variety of these properties is compatible with continuous cell division.  相似文献

An analog of 20alpha-hydroxycholesterol, (20R)-20-phenyl-5-pregnene-3beta,20-diol, which is completely substituted at C-22 was prepared with radioisotopes at various positions. The analog labeled with 3H at C-M and 14C at C-4 and C-IU was converted into radioactive pregnenolone by an enzyme preparation derived from adrenal mitochondria. Cleavage of the phenyl analog labeled with 3H in the aromatic ring by the same enzyme preparation led to the formation of [3H]phenol. Using the substrate doubly labeled with 14C at C-4 and 3H in the aromatic ring, it appeared that the products of the reactions, pregnenolone and phenol, were formed in equal amounts. During incubation of the side chain labeled substrate, another labeled fragment was formed. It was identified as acetophenone, a product resulting from cleavage of the C17,20 bond. The steroidal fragment corresponding to this C8 ketone was traced using nuclear label analog. From its nonpolar chromatographic properties it appears to be a C-17-deoxy-C19 steroid.  相似文献
