The purpose of the present study was to investigate the effects of exogenous ovarian steroid treatment, which is known to induce follicular cyst experimentally in cows, on ovarian activity in goats. Eleven female Shiba goats with the length of the normal oestrous cycle (approximately 21 days) received subcutaneously either 1 ml of ethanol (control group, n=4) or 4 mg of progesterone and 2mg of oestradiol (treatment group, n=7) daily for 7 days beginning on day 14 of the oestrous cycle (day 0=ovulation). Ultrasonographic images of the ovary and blood samples were collected daily to monitor the ovarian activity. Ovulation was observed before 1 day after the end of treatment in the control group. In the treatment group, no detectable structures of follicles or corpus luteum (static ovarian condition) were found for 6.0+/-1.4 days (mean+/-S.D.) after the end of treatment. Then, detectable follicles appeared and ovulation was observed in all animals of the treatment group. There was no significant difference in the maximum diameter of the ovulatory follicle between the control and treatment group (4.7+/-0.4mm versus 5.1+/-0.7 mm). The large non-ovulatory follicles, which grew more than 10mm in diameter were observed after the static ovarian condition in one goat of the treatment group, whereas no turnover of the cystic follicular structures was found. The length of the inter-ovulatory intervals in the treatment group was significantly longer than that in the control group (38.4+/-7.4 days versus 20.3+/-0.5 days, P<0.05). The present results demonstrated that the exogenous treatment of progesterone and oestradiol, which was adapted from the follicular cyst model in cows, did not induce follicular cysts in goats, suggesting that there is/are different mechanism(s) mediating the occurrence of follicular cysts between cows and goats.  相似文献

In this experiment we used nine goats to which we administered GnRH in fractioned doses at a pulse like rhythm in order to obtain follicular growth and oestrus. The average length of treatment was 5.5 days; the animals were injected with a daily amount of 0.05 mg of GnRH subdivided in three doses (0.017 mg each). All the 9 experimental goats came into oestrus and became pregnant. The GnRH treatment in fractioned and repeated daily doses proves a valid method to induce follicular growth and ovulation in anoestrus goats.  相似文献

The effect of exogenous progesterone exposure early in the oestrous cycle on the duration of the interovulatory interval was studied in dairy goats. A controlled intravaginal drug release (CIDR-G) device was inserted for 5 days starting at day 0 (D0 group, n=6) or day 3 (D3 group, n=5) postovulation. A third group was composed of untreated control goats (control group, n=7). Daily transrectal ultrasound was carried out during the interovulatory interval to assess the ovarian dynamics. Oestrous behaviour was checked twice a day and serum progesterone levels were assayed in daily jugular blood samples. Treated goats showed two different responses. In three D0 goats and one D3 goat, progesterone concentrations fell immediately after CIDR withdrawal and this was followed by oestrus and ovulation between days 8 and 11 (short cycles). In the other three D0 goats and in four D3 goats the treatment significantly reduced the interovulatory interval (18.3+/-0.3 and 18.5+/-0.3 days, respectively) (shortened cycles) compared with the control group (20.0+/-0.2 days; P<0.05), but the intervals with progesterone concentrations over 1 ng/ml were not different (15.7+/-0.3, 15.8+/-0.7 and 16.0+/-0.5 days for D0, D3 and control goats, respectively). In all D0 goats with a short cycle response, the ovulatory follicle arose from the first follicular wave but in the D3 goat with a short cycle it arose from the second follicular wave. These results showed that premature progesterone exposure early in the ovulatory cycle of the goat affected its length inducing short or shortened cycles. The effect of progesterone could either affect luteotropic support of the corpus luteum (CL) and/or stimulate a premature release of the luteolysin.  相似文献

Oestrus and ovulation were recorded for eight oestrous cycles after sheep and goats were put onto maintenance or supramaintenance levels of nutrition. Oestrous cycles were longer and more variable in goats than in sheep (20.3 ± 0.3 and 17.4 ± 0.2 days respectively), but in neither species was the incidence of oestrus affected by level of nutrition. There were no significant differences between sheep and goats in average body weight or mean ovulation rate. Animals fed at a maintenance level showed no significant changes in average body weight (19.0 ± 0.3 kg) or mean ovulation rate (1.69 ± 0.01). At the supramaintenance level, on the other hand, mean body weight increased progressively to a maximum of 24.8 ± 0.5 kg, and mean ovulation rate increased to a maximum of 2.65 at the sixth oestrus after the beginning of supramaintenance feeding. The effect of supramaintenance feeding on ovulation rate was reflected in subsequent lamb and kid production.  相似文献

Active immunization against oxytocin significantly prolonged the oestrous cycle in 3 out of 4 goats; the mean (+/- s.e.m.) cycle length was 29.1 +/- 1.7 days (n = 12) compared to 19.4 +/- 0.6 days (n = 9) in control animals. During Days 10-21 of the cycle in the 3 responsive goats, peripheral plasma concentrations of progesterone and oxytocin were steady and those of 13,14-dihydro-15-keto-prostaglandin F-2 alpha were very low (50-100 pg X ml-1) with no marked pulsatile activity. The major effect of immunization would appear to be suppression of the synthesis of the uterine luteolysin PGF-2 alpha, thus confirming that endogenous oxytocin has a facilitatory role in luteolysis via prostaglandin production.  相似文献

Intrauterine glass balls are used to prevent oestrous signs in sports mares, but the mechanism of action is unknown. It has been suggested that the glass ball can mimic an embryo or act via an induced chronic uterine inflammation and absent or continuous low-grade prostaglandin (PG) release. The purpose of this study was to induce prolonged luteal function in mares using a small intrauterine device (IUD) and to study the mechanisms behind prolonged IUD-induced luteal function. A uterine swab and a biopsy specimen were obtained in early oestrus. A water-filled plastic ball, diameter 20mm and weight 3.6g, was inserted into the uterus 2-4 days after ovulation; the control mares underwent similar cervical manipulation without ball insertion. The mares were examined three times per week until day 23 and twice weekly thereafter until they returned to oestrus (transrectal palpation, ultrasonography and progesterone determination). The location of the IUD was recorded and ultrasound scans were video-recorded to assess the frequency of uterine contractions. When the mare returned to oestrus, a uterine swab and biopsy specimen were obtained and the bacteriological, cytological and histological (inflammation and glandular dilation) results compared with the samples obtained before the IUD insertion. The PG F(2alpha) metabolite levels were measured in the plasma of four control mares and eight IUD mares on days 11-16. The IUD induced a prolonged luteal phase in 75% of the mares (9/12; IUD-P); the mean dioestrous length was 57.0 days. The three mares that did not respond to the IUD (IUD-N) showed a mean dioestrous length of 15.7 days and the 12 control mares 16.1 days. The inflammation and glandular dilation scores were not significantly different in pre- and post-manipulation biopsy specimens. Although locational changes of the IUD were observed, they occurred over very small distances and were mostly limited within the body-bifurcation area. The IUD-N and control mares showed increased uterine contractility 11-16 days post-ovulation, whereas the IUD-P mares did not. The control mares (n=4) and IUD-N mares (n=2) showed increased PG levels from day 14 post-ovulation, while the IUD-P mares (n=6) showed basal levels only. We concluded that the IUD did not cause continuous PG release and suggest that close contact of the IUD with the endometrium may prevent the endometrial cells from releasing PGF(2alpha).  相似文献

Fifteen nulliparous and nine multiparous Serrana goats were used, through two successive oestrous cycles, in order to characterize their ovulation time with regard to the number of ovulations after induced and natural oestrus during the breeding season. The onset of oestrus was detected by the amount of vasectomized bucks after oestrus synchronization with prostaglandin, given 10 days apart, and in the following two expected natural oestrus. The preovulatory LH peak was determined from blood samples collected 0, 4, 8, 12, 16, 20 and 24 h after onset of oestrus. A transrectal ovarian ultrasound scanning was performed 20, 24, 28, 32, 36, 40, 44 and 60 h after onset of oestrus, for the detection of ovulations by means of the disappearance of large follicles (>4 to 5 mm). Single ovulations were observed in 76% of oestrous periods in nulliparous goats and in 18% of nulliparous goats. The onset of oestrus to LH peak interval was lower in nulliparous (12.1 ± 0.9 h, n = 38) than in multiparous (15.6 ± 1.0 h, n = 22, P < 0.05) goats with no oestrus interaction effects (P > 0.05). The LH peak to first ovulation interval was higher after natural (18.9 ± 0.7 h, n = 36) than after induced (15.8 ± 1.2 h, n = 24, P < 0.05) oestrus. The onset of oestrus to total ovulation interval was influenced by parity (P < 0.01) and oestrus type (P < 0.05) with a length of 30.1 ± 1.1 h (n = 15) and 33.4 ± 1.5 h (n = 9) for induced oestrus of nulliparous and multiparous goats, respectively, and 32.5 ± 1.0 h (n = 23) and 36.5 ± 1.1 h (n = 13) for natural oestrus of nulliparous and multiparous goats, respectively. The onset of oestrus to first ovulation interval was not influenced by parity, but an interval of 8.0 ± 1.6 h was observed between the first and second ovulations in polyovulatory oestrus. Consequently, nulliparous goats that are predominantly monovular ovulate earlier than multiparous goats that are predominantly polyovulatory. In conclusion, significant differences occurred in the number and time of ovulations between nulliparous and multiparous goats. More research is necessary for a deeper understanding of the mechanisms regulating monovularory and polyovulatory oestrous cycles regarding the parity of goats.  相似文献

Boer goats were superovulated by immunization against the recombinant human inhibin α-subunit. The immune response to the antigen during the oestrous season varied greatly between animals, as judged by serum inhibin binding determined by enzyme-linked immunosorbent assay (ELISA). A booster injection administered during the anoestrous season elicited a prompt and rather uniform response and serum inhibin binding activity remained elevated in all goats throughout the remainder of the experiment. Both during the oestrous and anoestrous season, immunization increased the number of corpora lutea and unovulated follicles. Immunization shortened cycle length in some animals. The embryo yield from immunized goats during the oestrous and anoestrous season was over three times the number of embryos recovered from control animals. The recovery rates were 61% and 50%, respectively. Plotting the number of follicles and corpora lutea observed during the oestrous season against inhibin binding activity gave a positive correlation. Transfer of embryos recovered from the immunized animals resulted in 41% live births. These results indicate that immunization against the recombinant human inhibin α-subunit may be a useful alternative to the conventional approach of superovulation.  相似文献

The aim of this study was to determine if there is a seasonal pattern of sexual activity in female Payoya goats and if this seasonality could be modulated by nutrition. During the experimental period of 20 months, 43 non-pregnant adults goats were penned under natural photoperiod at latitude 37 degrees 15'N. At the onset of the experiment, the animals were allocated to three experimental groups differing in the level of nutrition and whether the animals were entire or ovariectomized does. The high nutrition group (H, n = 16 entire does) receiving 1.5 times maintenance requirements. The low nutrition group (L, n = 16 entire does) and an ovariectomized and oestradiol treated group (OVX, n = 11 ovariectomized does) received a diet supporting their maintenance requirements. The groups were balanced for live weight (LW) and body condition score (BCS) at the beginning of the study. In entire goats, oestrus was tested daily using aproned males, ovulation rate was assessed by laparoscopy 7 days after identification of oestrus and plasma samples were obtained twice per week for progesterone assay. OVX goats were isolated from the other groups and bucks, plasma samples were assayed twice per week for LH and there were four intensive sampling periods during the year to determine LH pulsatility. LW and BCS were recorded for all animals once a week. A clear circannual cycle in live weight change was observed in all experimental groups, being relatively stable or slightly decreasing in summer and autumn and increasing during winter and spring. The effect of exposure to high (H) rather than low (L) nutrition was to cause earlier onset of ovarian activity (5 versus 17 August; P < 0.05), and expression of oestrous (16 August versus 2 September; P < 0.01) and later cessation of reproductive activity (ovulation 11 February versus 17 January; P < 0.01). Consequently, seasonal anoestrus was 32 days shorter in does on the higher plane of nutrition (P < 0.01). The seasonality of reproductive activity was confirmed in the OVX does, with reduced LH concentrations during spring and summer, and increased LH concentrations in autumn and winter. There was no effect of nutrition on ovulation rate. These results demonstrate that the female Payoya goat exhibits marked reproductive seasonality which is modulated by nutrition but possibly not ovulation rate.  相似文献

Information on the use of buffalo follicular fluid (buFF) in modulation of ovarian functions in farm animals is scanty compared to other species. This is an attempt to investigate the effect of direct administration and active immunization of 30 kDa and above buFF proteins on ovarian functions in goats. Treatment of goats (n = 6) with steroid free 30 kDa and above buFF protein fraction during late-luteal phase for 4 days (days 12 or 13 to days 15 or 16) of the natural cycle, delayed the onset of estrus by 24 h compared to control although the mean duration of estrus was unaffected. A 71% increase (P = 0.06) in mean ovulation number was also observed following treatment. However, the population of large (> or =5 mm diameter) follicle was not affected. The ovarian activity calculated as total of ovulation and large follicles increased (1.6 times) significantly (P = 0.02) in treated animals. Active immunization of goats (n = 5) against these proteins did not affect the onset and duration of estrus. Similarly, the ovulation rate, number of large follicles and the ovarian activity did not differ significantly between immunized and control groups. The study revealed that 30 kDa and above buffalo follicular fluid contains some factor(s) that cause delay in the onset of estrus in goats and increase the ovulation rate. Active immunization against these proteins in goat did not show any effect either on onset, duration of estrus or ovulation rate and large follicle population. Detailed study on these buffalo follicular fluid proteins may help to use them further for modulation of ovarian function in farm animals.  相似文献

The reproductive performance of Coopworth ewes after administration of zearalenone was determined in two trials. In Trial I, zearalenone was administered to groups of 33 ewes at rates of 0, 1.5, 3.0, 6.0, 12.0 and 24.0 mg/ewe/day for 10 days, starting on Day 7 of the oestrous cycle before mating. There was a linear decline (P less than 0.001) in ovulation rate with dose of zearalenone; also cycle length decreased and duration of oestrous increased with increasing dose levels. Reductions in the incidence of ovulation and in fertilization were seen only at doses of 12 and 24 mg. In Trial 2, groups of 50 ewes were given the same range of doses of zearalenone for 10 days, starting 5 days after mating to entire rams. There was no effect of zearalenone treatment after mating on pregnancy rate or embryonic loss. These results indicate that the effects of zearalenone, administered orally, on ewe reproduction, at the dose levels examined, were restricted to ewes exposed before mating. Intakes of zearalenone of 3 mg/ewe/day or more during this period would be reflected as depressed ovulation rates and lower lambing percentages.  相似文献

Studies were conducted to determine the effect of iodine infusion on the luteal function of goats, as evident by blood progesterone concentration, and on plasma PGF2a levels. Ten cycling mixed breed goats were synchronized for estrus by PGF2a (5 mg) and given a single intrauterine iodine infusion on day 5 and on day 15 of the estrous cycle. Iodine infusion on day 5 (group II) resulted in shorter estrous length (8.2 days) and a 7-fold increase in plasma PGF2a concentration as compared to control animals (group I) given distilled water infusion. Similar infusion on day 15 (group III), on the other hand, failed to alter the estrous cycle length but induced a moderate increase in PGF2a concentration which lasted only for a brief period. The progesterone levels declined concomitantly as PGF2a levels rose after iodine infusion in group II animals but failed to decline until after 24 hours in group III animals. The studies indicate that the endometrium reacts to the chemical stimuli and releases PGF2a which, in turn, alters the luteal function.  相似文献

Fifty-three anestrous cows of Hariana crossed with Holstein, Friesian, Brown Swiss or Jersey half-breed crosses were treated with Buserelin acetate aqueous solution 0.0042 mg/ml, administered as a single intramuscular (i.m.) dose of 21 ng. Twelve (22.64%) animals exhibited complete follicular development, ovulation and behavioural expression of oestrus. Twenty-eight (52.83%) cows showed only partial follicular development with no ovulation or oestrous manifestations. Thirteen (24.53%) animals did not respond. Of eight control animals, three (37.50%) showed partial follicular development and no ovulation or symptoms of oestrus while five (62.50%) did not respond.In GnRH-treated cows showing follicular development, oestrous manifestation, and ovulation, nine animals responded between 4 to 10 d after administration. Three animals took from 14 to 155 d. The mean time from treatment to response for this group was 29.17 d. In animals showing partial follicular development and no oestrous behaviour, the interval was 3 to 11 d in 28 animals. In the control group, three animals showed partial follicular development on 94 d average. The administration of GnRH in anestrous cows promoted the development of follicles to a certain stage as evident from the clinical observations, but the follicles failed to grow beyond a particular stage. The reasons for failure of follicular development beyond a particular stage in GnRH-treated animals are not clear. It appeared that the response to GnRH may be expected to peak by the eleventh day and a second dose may then be tried for a favourable response.  相似文献

Border Leicester x Merino ewes were fasted for the first six days or the second six days of the oestrous cycle preceding mating in the fall season. When not fasting, ewes were grazed with a control group on irrigated pasture. Compared with control ewes, fasting had no effect on oestrous occurrence or fertilization but did reduce the ovulation rate, especially where fasting occurred early in the oestrous cycle.  相似文献

The effect of fasting during oestrous cycle on the occurrence of oestrous and concentration of leptin and steroid hormones was investigated in goats. Sixteen Ardi goats of 10-12 month of age were split into two groups (control and fasting). Oestrous was synchronized with intravaginal progesterone sponges and detected 24h after sponge removal. Blood samples were collected at the days 5, 10, 15 of each cycle. Fasting of mature goats twice for 4 days starting on day 10 of two successive oestrous cycles inhibited oestrous behaviour and resulted in reduced concentration of leptin, progesterone and testosterone with different timing. Day 5 of the second cycle showed significant decrease in the plasma level of leptin (1.6+/-0.15 ng/ml) and progesterone (1.6+/-0.1 ng/ml) as compared to control group (3.2+/-0.15 ng/ml and 4.1+/-0.2 ng/ml, respectively). Testosterone started to decrease from day 10 of the second cycle (35.0+/-12.0 pg/ml) as compared to control group (65.0+/-15.0 pg/ml); the decrease in this hormone was significant in day 15 of the second cycle (65.0+/-16.0 pg/ml) as compared to the control (320.0+/-50.0 pg/ml). These data suggest that fasting-induced inadequate corpus luteum function, hence, lowering progesterone plasma level may partly be more leptin-dependent than the following decrease in plasma level of testosterone.  相似文献

Subcutaneous administration of oxytocin between days 3-6, inclusive, significantly reduced the length of the oestrous cycle in goats. Concomitant oral administration of the prostaglandin synthetase inhibitor, meclofenamic acid, completely prevented this effect. Oxytocin treatment did not affect the duration of the posttreatment cycles.  相似文献

Reproductive cycles were studied in a group of tame Père David's deer hinds. The non-pregnant hind is seasonally polyoestrous and, in animals studied over 2 years, the breeding season began in early August (2 August +/- 3.3 days; s.e.m., N = 9) and ended in mid-December (18 December +/- 5.7 days; N = 8) and early January (6 January +/- 3.2 days; N = 11) in consecutive years. During the anoestrous period, plasma progesterone concentrations were low (0.2 +/- 0.01 ng/ml) or non-detectable. There was a small, transient increase in progesterone values before the onset of the first cycle of the breeding season. In daily samples taken during an oestrous cycle in which hinds were mated by a marked vasectomized stag, progesterone concentrations remained low (less than 0.5 ng/ml) for a period of about 6 days around the time of oestrus, showed a significant increase above oestrous levels by Day 4 (Day 0 = day of oestrus) and then continued to increase for 18 +/- 2.8 days to reach mean maximum luteal levels of 3.5 +/- 0.6 ng/ml. The plasma progesterone profiles from a number of animals indicated that marking of the hinds by the vasectomized stag did not occur at each ovulation during the breeding season and therefore an estimate of the cycle length could not be determined by this method. In the following year, detection of oestrus in 5 hinds was based on behavioural observations made in the absence of the stag. The ovaries of 18 post-partum beef suckler cows were examined daily, using ultrasound, from Day 5 post partum until a normal oestrous cycle was completed. Periods of growth and regression of medium-sized (5-9 mm) follicles were identified before one medium follicle became dominant (single large follicle greater than or equal to 10 mm). The mean (+/- s.e.m.) number of days from parturition to detection of the first post-partum dominant follicle was 10.2 +/- 0.5. The first post-partum dominant follicle ovulated in 2/18 (11%) cows. The interval from calving to first ovulation (mean +/- s.e.m. = 35.9 +/- 3.3 days) was characterized by the growth and regression of a variable number (mean = 3.2 +/- 0.2; range 1-6) of dominant follicles. The maximum diameter of the dominant follicle increased as the cows approached first ovulation (P less than 0.05). Behavioural oestrus was not detected in 16/18 (89%) cows at first ovulation. Following first ovulation, the length of the subsequent cycle was short (mean = 9.7 +/- 0.5 days; range 8-15 days) in 14/18 (78%) cows and was characterized by the development and ovulation of a single dominant follicle. During oestrous cycles of normal length (mean = 20.6 +/- 0.5 days; range 18-23 days) one (N = 2), two (N = 7) or three (N = 8) dominant follicles were identified. The growth rate, maximum diameter or persistence of non-ovulatory dominant follicles before first ovulation or during oestrous cycles were not different (P greater than 0.05). These data show that, in beef suckler cows, follicular development and formation of a dominant follicle occur early after parturition and the incidence of ovulation of the first dominant follicle is low. The number of dominant follicles that develop before first ovulation is variable; first ovulation is rarely associated with oestrus and short cycles are common after first ovulation. It is concluded that prolonged anoestrus in post-partum beef suckler cows is due to lack of ovulation of a dominant follicle rather than delayed development of dominant follicles.  相似文献

The influence of season and exposure to buck on the length of the estrous cycle were determined in 32 normally cycling native goats of different ages and parity. The estrous cycle durations were classified as short, medium and long. Ten goats (31%) exhibited medium length estrous cycles; 22 animals (69%) exhibited a combination of short, medium and long estrous cycles. Of the 155 estrous cycles studied, 15% were short, 72% were medium and 13% were long. Short estrous cycles were significantly shorter (P < 0.01) in estrus duration than medium or long estrous cycles. Significant differences (P < 0.05) for estrous cycle length and estrus duration were found between does but not within does. The presence of a buck for 8 to 16 h showed no significant effect on the length of the estrous cycle or on estrus duration, but a 24-h exposure period to a buck yielded shorter estrous cycles and estrus duration than found in the unexposed control group. Estrous cycles were significantly shorter (P < 0.01) in October, when rainfall is 57.9% of the annual total, and significantly longer (P < 0.01) in February, when rainfall is low at 0.2% of the annual total. A negative linear relationship (r = -0.87) was observed between estrous cycle and rainfall. Twenty-eight (90.3 %) of the short estrous cycles were ovulatory. The short cycles had a relatively lower ovulation rate than medium and long estrous cycles. The results indicate that the short estrous cycle in goats is associated with ovulation, and its occurrence in the tropics is related to rainfall.  相似文献
