Wild bank voles (Clethrionomys glareolus) may develop diabetes in laboratory captivity. The aim of this study was to test whether bank voles develop type 1 diabetes in association with Ljungan virus. Two groups of bank voles were analyzed for diabetes, pancreas histology, autoantibodies to glutamic acid decarboxylase (GAD65), IA-2, and insulin by standardized radioligand-binding assays as well as antibodies to in vitro transcribed and translated Ljungan virus antigens. Group A represented 101 trapped bank voles, which were screened for diabetes when euthanized within 24 hours of capture. Group B represented 67 bank voles, which were trapped and kept in the laboratory for 1 month before being euthanized. Group A bank voles did not have diabetes. Bank voles in group B (22/67; 33%) developed diabetes due to specific lysis of pancreatic islet beta cells. Compared to nondiabetic group B bank voles, diabetic animals had increased levels of GAD65 (P < .0001), IA-2 (P < .0001), and insulin (P = .03) autoantibodies. Affected islets stained positive for Ljungan virus, a novel picorna virus isolated from bank voles. Ljungan virus inoculation of nondiabetic wild bank voles induced beta-cell lysis. Compared to group A bank voles, Ljungan virus antibodies were increased in both nondiabetic (P < .0001) and diabetic (P = .0015) group B bank voles. Levels of Ljungan virus antibodies were also increased in young age at onset of newly diagnosed type 1 diabetes in children (P < .01). These findings support the hypothesis that the development of type 1 diabetes in captured wild bank voles is associated with Ljungan virus. It is speculated that bank voles may have a possible zoonotic role as a reservoir and vector for virus that may contribute to the incidence of type 1 diabetes in humans.  相似文献

Long-term field studies on parasite communities are rare but provide a powerful insight into the ecological and evolutionary processes shaping host–parasite interactions. The aim of our study was to identify the principal factors regulating long-term trends in the haemoparasite communities of bank voles, and to this end, we sampled three semi-isolated populations of bank voles (n?=?880) in 1999, 2002, 2006 and 2010 in the Mazury lake district region of NE Poland. Overall, 90.8 % of the bank voles harboured at least one of the species of haemoparasites studied. Whilst overall prevalence (all species combined) did not vary significantly between the surveys, different temporal changes were detected among voles in each of the three sites. In voles from Urwita?t, prevalence increased consistently with successive surveys, whereas in Ta?ty, the peak years were 2002 and 2006, and in Pilchy, prevalence oscillated without a clear pattern. Across the study, bank voles harboured a mean of 1.75?±?0.034 haemoparasite species, and species richness remained stable with no significant between-year fluctuations or trends. However, each of the five constituent species/genera showed a different pattern of spatio-temporal changes. The overall prevalence of Babesia microti was 4.9 %, but this varied significantly between years peaking in 2006 and declining again by 2010. For Bartonella spp., overall prevalence was 38.7 %, and this varied with year of study, but the temporal pattern of changes differed among the three sites. The overall prevalence of Haemobartonella (Mycoplasma) was 68.3 % with an increase in prevalence with year of study in all three sites. Hepatozoon erhardovae had an overall prevalence of 46.8 % but showed a marked reduction with each successive year of the study, and this was consistent in all three sites. The overall prevalence of Trypanosoma evotomys was 15.4 % varying significantly between sites, but showing temporal stability. While overall prevalence of all haemoparasites combined and species richness remained stable over the period of study, among the five haemoparasites, the pattern of spatiotemporal changes in prevalence and abundance of infections differed depending on parasite species. For some genera, host age was shown to play an important role, but a significant effect of host sex was found only for Haemobartonella spp.  相似文献

Current theory and supporting data regarding population regulationand cycling in microtine rodents needs to be reviewed in lightof a new, season-sensitive model of social behavior for meadowvoles, Microtus pennsylvanicus. The finding of a clear socialimperative among meadow voles during most of the fall, winterand spring conflicts with the prediction of high levels of socialintolerance in the higher density populations commonly existinglate in the year. The general rarity of adult dispersal, exceptduring periods of declining densityin winter and early spring,and the contact-seeking nature of this dispersal,are generallycontradictory to predictions based on "intrinsic’ theories.Finally,published data on meadow vole reproduction, recruitmentand dispersal, and hence demography, are probably biased asa result of effects produced by the spread of rodent pheromonesby investigators, prolonged entrapment of individual voles,and inappropriate behavioral measures in the field.  相似文献

Relatively little is known about the mechanisms of communication during the non-breeding season in species that are seasonal breeders. Previous work with meadow voles, Microtus pennsylvanicus, housed under long photoperiods, has shown that they prefer the odors of opposite-sex conspecifics. In this paper, we investigated the effects of short photoperiod on preferences for sex-specific odors and the production of such odors. Short-photoperiod females preferred anogenital and fecal scents of other short-photoperiod females over those of males, but did not show sexual preferences for three other scents. Short-photoperiod males did not exhibit sexual preferences for any of the odors. Furthermore, scents from short-photoperiod voles did not elicit sex-specific preferences in long-photoperiod voles, and scents from long-photoperiod voles did not elicit preferences from short-photoperiod voles. These and previous results indicate that both the odors and responses to odors change seasonally and that long-photoperiod voles respond selectively to scents from long-photoperiod voles and short-photoperiod voles respond selectively to scents from short-photoperiod voles. Taken together, these results suggest the co-evolution of seasonal changes in scents and in perceptual or other response mechanisms.  相似文献

Bank voles free living in a contaminated environment have been shown to be more sensitive to cadmium (Cd) toxicity than the rodents exposed to Cd under laboratory conditions. The objective of this study was to find out whether benzo(a)pyrene (BaP), a common environmental co-contaminant, increases Cd toxicity through inhibition of metallothionein (MT) synthesis-a low molecular weight protein that is considered to be primary intracellular component of the protective mechanism. For 6 weeks, the female bank voles were provided with diet containing Cd [less than 0.1 μg/g (control) and 60 μg/g dry wt.] and BaP (0, 5, and 10 μg/g dry wt.) alone or in combination. At the end of exposure period, apoptosis and analyses of MT, Cd, and zinc (Zn) in the liver and kidneys were carried out. Dietary BaP 5 μg/g did not affect but BaP 10 μg/g potentiated rather than inhibited induction of hepatic and renal MT by Cd, and diminished Cd-induced apoptosis in both organs. The hepatic and renal Zn followed a pattern similar to that of MT, attaining the highest level in the Cd + BaP 10-μg/g group. These data indicate that dietary BaP attenuates rather than exacerbates Cd toxicity in bank voles, probably by potentiating MT synthesis and increasing Zn concentration in the liver and kidneys.  相似文献

采用水培实验,研究重金属镉对草莓幼龄期叶、根矿质元素含量和分布的影响。结果表明,高量镉降低草莓根中K、Ca、Mg、P、Cu、Zn、Mn、Fe的积累,叶片相应元素以及B、Mo含量也随镉浓度增大而呈降低趋势;但镉浓度较低时,K、Mg、P、Zn在叶、根中的含量均增加,而后才随镉处理浓度提高而降低。在镉胁迫下,同一元素在草莓叶、根之间的分布也受到影响,而且某些矿质元素含量间有显著相关性。  相似文献

以镉(Cd)积累潜力不同的2个籼稻品种为材料,研究了籽粒发育过程中各器官中Cd含量的动态变化及其与土壤中Cd含量的关系。结果表明,在含Cd的生长环境中,水稻根系中的Cd含量在整个生育期中保持稳定增长的趋势,而茎叶、穗轴和稻壳等器官在营养生长阶段积累了大量的Cd,然后在籽粒充实过程中向外输出,其中旗叶和稻壳中的Cd输出率在50%以上,明显高于其他营养器官。根系和叶片中的Cd含量与土壤中的Cd含量呈高度线性相关,茎秆和籽粒中的Cd含量与土壤中的Cd含量呈抛物线相关,说明根系、茎秆、叶片等营养器官对Cd有储存和“过滤”作用。低积累品种‘X24’穗轴中的Cd含量明显低于高积累品种‘T705’,说明营养器官中的Cd向籽粒中转运的速率是决定籽粒中Cd积累量的关键因素。但是,只有当土壤中的Cd含量为0.3~1.2 mg&#183;kg-1时,低积累品种精米中的Cd含量才会显著低于高积累品种;当土壤中的Cd含量高于2.4 mg&#183;kg-1时,2个品种精米中的Cd含量没有显著差异。  相似文献

Background and Aims

Exercise-induced iron deficiency is a common finding in endurance athletes. It has been suggested recently that hepcidin may be an important mediator in this process.

Objective

To determine hepcidin levels and markers of iron status during long-term exercise training in female runners with depleted and normal iron stores.

Methods

Fourteen runners were divided into two groups according to iron status. Blood samples were taken during a period of eight weeks at baseline, after training and after ten days’ recovery phase.

Results

Of 14 runners, 7 were iron deficient at baseline and 10 after training. Hepcidin was lower at recovery compared with baseline (p<0.05). The mean cell haemoglobin content, haemoglobin content per reticulocyte and total iron binding capacity all decreased, whereas soluble transferrin receptor and hypochromic red cells increased after training and recovery (p<0.05 for all).

Conclusion

The prevalence of depleted iron stores was 71% at the end of the training phase. Hepcidin and iron stores decreased during long-term running training and did not recover after ten days, regardless of baseline iron status.  相似文献

通过组织学方法,对产后1 d、10 d、25 d、45 d、60 d及70 d的棕色田鼠Lasiopodomys mandarinus睾丸和附睾发育进行了观察,以探讨其精子发生特点.结果 发现,1 d棕色田鼠的生殖细胞主要是生殖母细胞和前精原细胞;10 d出现大量精原细胞,睾丸间质细胞明显;25 d出现精子细胞;45 d有少量精子出现;60 d和70 d具有各级生精细胞,睾丸生精小管和附睾内出现大量成熟精子.睾丸生精小管管径和生精上皮厚度随日龄增加,于60 d达到最大;附睾管腔直径和附睾上皮厚度也于60 d达到最大.这些结果表明,棕色田鼠在生后45 d左右进入青春期,60 d左右达到性成熟,精子的产生及成熟与附睾的发育同步.  相似文献

Altitudinal localities of the northern India are associated with high seasonal changes. Drosophila melanogaster flies are darker during the winter season as compared to the autumn season. We tested the hypothesis whether there are altitudinal clines for mating related traits. We observed negative cline for mating latency and positive for copulation period along altitude in D. melanogaster. We further tested if seasonally varying body melanisation is correlated with mating propensity in D. melanogaster. Thus, we examined the D. melanogaster flies collected during autumn and winter season for changes in body melanisation and mating-related traits. Flies from the winter season show high melanisation, copulation duration and fecundity/day as compared to the autumn season flies. By contrast mating latency is longer during autumn as compared to winter season. Based on within- and between-population analysis, body melanisation shows positive correlation with copulation duration and fecundity/day, while negative correlation with mating latency. Within-population analyses show no correlation between body size and ovariole number with body melanisation. Thus, our data suggest that seasonal changes in body melanisation are correlated with mating latency, copulation duration and fecundity/day, but no correlation with body size and ovariole numbers. Further, we observed that seasonal changes in these clines, although have some component of plasticity, have strong genetic basis as the seasonal and population differences were maintained for various traits after 8 generations in the laboratory.  相似文献

Background

Coronary artery disease is very common in Pakistani population. Some of the studies carried out on Western populations have shown a relationship between body iron status as determined by the ratio of concentrations of serum soluble transferrin receptor (sTfR) to ferritin and the risk of acute myocardial infarction (AMI). In order to investigate whether increased body iron status has any relationship with the risk of premature AMI in Pakistani population, a case-control study was carried out.

Methodology/Principal findings

In this case-control study, 203 consecutive AMI patients [146 males and 57 females; age range 18–45 years] admitted to the National Institute for Cardiovascular Diseases, Karachi, were enrolled with informed consent. In addition, 205 healthy controls whose gender and age (within 3 years) matched the patients, and who had a similar socio-economic background were recruited. Fasting venous blood was obtained and assessed for plasma/serum folate, vitamin B12, homocysteine, total cholesterol, triglycerides, LDL-cholesterol, HDL-cholesterol, sTfR and ferritin and blood lead. It was found that serum concentration of ferritin and blood lead levels were significantly higher in AMI patients compared to their age and gender-matched healthy controls (p value <0.05), while the concentrations of vitamin B12 and HDL-cholesterol were significantly lower in AMI patients compared to controls (p value <0.01). The ratio of sTfR to ferritin was significantly lower in AMI patients compared to controls [mean±SD/median (IQR) values 84.7±295/28.9 (38.4) vs 255±836/49.4 (83.8), respectively; p value <0.001]. Compared with the highest quartile of sTfR/ferritin (low body iron status), the OR for the risk of AMI was 3.29(95% CI, 1.54–7.03) for the lowest quartile (quartile 1) when the model was adjusted for vitamin B12 and HDL-cholesterol (p value for trend <0.01).

Conclusions/Significance

This study shows a positive association between total body iron status and risk of premature AMI in a Pakistani population.  相似文献

Fuschna Spring in the Swiss Alps (Engadin region) is a bicarbonate iron(II)-rich, pH-neutral mineral water spring that is dominated visually by dark green microbial mats at the side of the flow channel and orange iron(III) (oxyhydr)oxides in the flow channel. Gradients of O2, dissolved iron(II), and bicarbonate establish in the water. Our goals were to identify the dominating biogeochemical processes and to determine to which extent changing geochemical conditions along the flow path and seasonal changes influence mineral identity, crystallinity, and microbial diversity. Geochemical analysis showed microoxic water at the spring outlet which became fully oxygenated within 2.3 m downstream. X-ray diffraction and Mössbauer spectroscopy revealed calcite (CaCO3) and ferrihydrite [Fe(OH)3] to be the dominant minerals which increased in crystallinity with increasing distance from the spring outlet. Denaturing gradient gel electrophoresis banding pattern cluster analysis revealed that the microbial community composition shifted mainly with seasons and to a lesser extent along the flow path. 16S rRNA gene sequence analysis showed that microbial communities differ between the flow channel and the flanking microbial mat. Microbial community analysis in combination with most-probable-number analyses and quantitative PCR (qPCR) showed that the mat was dominated by cyanobacteria and the channel was dominated by microaerophilic Fe(II) oxidizers (1.97 × 107 ± 4.36 × 106 16S rRNA gene copies g−1 using Gallionella-specific qPCR primers), while high numbers of Fe(III) reducers (109 cells/g) were identified in both the mat and the flow channel. Phototrophic and nitrate-reducing Fe(II) oxidizers were present as well, although in lower numbers (103 to 104 cells/g). In summary, our data suggest that mainly seasonal changes caused microbial community shifts, while geochemical gradients along the flow path influenced mineral crystallinity.  相似文献

This study was undertaken to demonstrate the synthesis of glutathione (GSH) in Mercenaria mercenaria brown cells to test the hypothesis that failure to achieve 100% mortality with metal treatment is the result of high concentrations of GSH heterogeneously distributed in the brown cell population and to determine the effect of Cd2+, Cu2+, and Pb2+ on the GSH status in brown cells. The monochlorobimane (MCB) assay appeared to be selective for GSH in brown cells and a close relationship between the levels of GSH measured by MCB and a standard enzymatic method was found. The fluorescent GSH-bimane adduct, once formed within the cell, was not released from the cell. The technique was used to establish that GSH was synthesized in brown cells and was heterogeneously distributed in the brown cell population. Metal concentrations as high as 40 mM cadmium, 6.0 μM copper, or 20 mM lead did not deplete but caused decreases in brown cell GSH concentration that differed significantly (P < 0.05) from controls. The decrease caused by cadmium, copper, and lead was not in a dose dependent manner, whereas, the decrease caused by N-ethylmaleimide (NEM) was. It appears that the partial kill phenomenon associated with metal toxicity may be due to the heterogeneous distribution of GSH in the brown cell population.  相似文献

In order to assess the susceptibility of bank voles to chronic wasting disease (CWD), we inoculated voles carrying isoleucine or methionine at codon 109 (Bv109I and Bv109M, respectively) with CWD isolates from elk, mule deer and white-tailed deer. Efficient transmission rate (100%) was observed with mean survival times ranging from 156 to 281 days post inoculation. Subsequent passages in Bv109I allowed us to isolate from all CWD sources the same vole-adapted CWD strain (Bv109ICWD), typified by unprecedented short incubation times of 25–28 days and survival times of ∼35 days. Neuropathological and molecular characterisation of Bv109ICWD showed that the classical features of mammalian prion diseases were all recapitulated in less than one month after intracerebral inoculation. Bv109ICWD was characterised by a mild and discrete distribution of spongiosis and relatively low levels of protease-resistant PrPSc (PrPres) in the same brain regions. Despite the low PrPres levels and the short time lapse available for its accumulation, end-point titration revealed that brains from terminally-ill voles contained up to 108,4 i.c. ID50 infectious units per gram. Bv109ICWD was efficiently replicated by protein misfolding cyclic amplification (PMCA) and the infectivity faithfully generated in vitro, as demonstrated by the preservation of the peculiar Bv109ICWD strain features on re-isolation in Bv109I. Overall, we provide evidence that the same CWD strain was isolated in Bv109I from the three-cervid species. Bv109ICWD showed unique characteristics of “virulence”, low PrPres accumulation and high infectivity, thus providing exceptional opportunities to improve basic knowledge of the relationship between PrPSc, neurodegeneration and infectivity.  相似文献
