Cells of the trophoblast lineage make up the epithelial compartment of the placenta, and their rapid development is essential for the establishment and maintenance of pregnancy. A diverse array of specialized trophoblast subtypes form throughout gestation and are responsible for mediating implantation, as well as promotion of blood to the implantation site, changes in maternal physiology, and nutrient and gas exchange between the fetal and maternal blood supplies. Within the last decade, targeted mutations in mice and the study of trophoblast stem cells in vitro have contributed greatly to our understanding of trophoblast lineage development. Here, we review recent insights into the molecular pathways regulating trophoblast lineage segregation, stem cell maintenance, and subtype differentiation.  相似文献

Tissue-resident stem cells (SCs) are critical players in the maintenance of tissue homeostasis. SCs reside in complex and uniquely anatomically organized microenvironments (SC niches), that carefully control SC lineage outputs depending on localized tissue needs. Upon environmental perturbations and tissue stressors, SCs respond and restore the tissue to homeostasis, as well as protect it from secondary assaults. Critical to this function are two key processes, SC lineage plasticity and SC memory. In this review, we delineate the multifactorial determinants and key principles underlining these two remarkable SC behaviors. Understanding lineage plasticity and SC memory will be critical not only to design new regenerative therapies but also to determine how these processes are altered in a multitude of pathologies such as cancer and chronic tissue damage.  相似文献

利用随机扩增多态性DNA技术(randomamplifiedpolymorphicDNA,RAPD)对广东省2001年度稻瘟病菌群体的遗传结构进行了分析。以相似性系数为0.70阈值时,可将采集于广东省三大生态稻作区、早稻和晚稻生长季节的96个菌株划分为12个遗传宗谱;其中宗谱8和9的菌株数各占总数的25%和18.8%,为优势宗谱。从稻作区来看,宗谱3和8为各个稻作区的共同宗谱;而宗谱1和2,7和11,以及9、10和12则依次是粤北、粤中和粤南稻作区的特异性宗谱。从生长季节来看,来源于早、晚季的菌株完全分属于宗谱图的上、下两个半区,彼此之间不存在共同的宗谱;而且两个优势宗谱都集中于晚季供试亚群体。结合前两次实验的结果,作者提出了如下两个假说来解释广东省稻瘟病菌群体所表现的遗传特性:一个地区或生长季节的病原菌群体,其优势宗谱所占的比例越高,该地区或生长季节病害发生就越严重;在长期的水稻栽培历史中,稻瘟病菌群体可能逐步地形成了早季宗谱(小种)和晚季宗谱(小种)的遗传分化。如何进一步验证上述两个假说是值得我们进一步探讨的重要课题。  相似文献

The morphogenesis of blood cell lineages in channel catfish Ictalurus punctatus , from head and trunk kidney and spleen imprints as well as from blood smears of bled and control fish, showed that early maturation stages resembled those in higher vertebrates. The erythroid lineage consisted of the proerythroblast, erythroblasts (basophilic, polychromatic, orthochromic), young erythrocyte and erythrocyte. The rare bilobed erythrocyte seemed to be a cell in apoptosis while old erythrocytes and erythroplastids represented remnants of this process. Maturation stages of neutrophils and basophils encompassed the granuloblast, young progranulocyte, progranulocyte and metagranulocyte. The basophilic lineage was regularly present in kidneys, rare in spleen and absent from blood. It contained large Sudan Black and PAS-negative, water soluble granules and small PAS-positive ones. Lymphocytes with azurophilic granulation occured regularly in kidneys and spleen. Monoblasts and promonocytes in kidneys preceded monocytes. A phagocytic lineage devouring apoptotic blood cell remnants was present in kidneys and spleen. Its youngest identified stage (promacrophage) resembled more a granuloid cell without granules than a monocytoid one. The larger, young macrophages contained a few to several ingestions and the very large mature macrophages were loaded with them. The latter two stages corresponded to cells in melano-macrophage centres (macrophage aggregates). Precursor stages of the thrombocyte were not identified.  相似文献

One obstacle in the development of a coherent theoretical framework for the process of animal domestication is the rarity of domestication events in human history. It is unclear whether: (1) many species are suitable for domestication, the limiting factor being the requirement of people for new domestic animals; or (2) very few species are preadapted for domestication. Comparisons between 16 species and subspecies of small cats (Felidae) kept in zoos indicated that affiliative behaviour towards people, an important preadaptation to domestication, is widely, if patchily, distributed throughout this taxon, and is not concentrated in species closely related to the domestic cat, Felis silvestris catus . The highest proportion of individuals showing affiliative behaviour was found in the ocelot lineage, which is estimated to have diverged from the rest of the Felidae between 5 and 13 Mya. The domestication of F. silvestris alone among felids is therefore likely to have been the result of a specific set of human cultural events and requirements in the Egyptian New Kingdom, rather than the consequence of a unique tendency to tame-ness in this subspecies.© 2002 The Linnean Society of London, Biological Journal of the Linnean Society , 75 , 2002, 361–366.  相似文献

During development, neural progenitor cells or neuroblasts generate a great intra- and inter-segmental diversity of neuronal and glial cell types in the nervous system. In thoracic segments of the embryonic central nervous system of Drosophila, the neuroblast NB6-4t undergoes an asymmetric first division to generate a neuronal and a glial sublineage, while abdominal NB6-4a divides once symmetrically to generate only 2 glial cells. We had earlier reported a critical function for the G1 cyclin, CyclinE (CycE) in regulating asymmetric cell division in NB6-4t. Here we show that (i) this function of CycE is independent of its role in cell cycle regulation and (ii) the two functions are mediated by distinct domains at the protein level. Results presented here also suggest that CycE inhibits the function of Prospero and facilitates its cortical localization, which is critical for inducing stem cell behaviour, i.e. asymmetric cell division of NB6-4t. Furthermore our data imply that CycE is required for the maintenance of stem cell identity of most other neuroblasts.  相似文献

It is thought that small intestinal epithelial stem cell progeny, via Notch signaling, yield a Hes1-expressing columnar lineage progenitor and an Atoh1 (also known as Math1)-expressing common progenitor for all granulocytic lineages including enteroendocrine cells, one of the body's largest populations of endocrine cells. Because Neurogenin 3 (Neurog3) null mice lack enteroendocrine cells, Neurog3-expressing progenitors derived from the common granulocytic progenitor are thought to produce the enteroendocrine lineage, although more recent work indicates that Neurog3+ progenitors also contribute to non-enteroendocrine lineages. We aimed to test this model and better characterize the progenitors leading from the stem cells to the enteroendocrine lineage. We investigated clones derived from enteroendocrine precursors and found no evidence of a common granulocytic progenitor that routinely yields all granulocytic lineages. Rather, enteroendocrine cells are derived from a short-lived bipotential progenitor whose offspring, probably via Notch signaling, yield a Neurog3+ cell committed to the enteroendocrine lineage and a progenitor committed to the columnar lineage. The Neurog3+ cell population is heterogeneous; only about 1/3 are slowly cycling progenitors, the rest are postmitotic cells in early stages of enteroendocrine differentiation. No evidence was found that Neurog3+ cells contribute to non-enteroendocrine lineages. Revised lineage models for the small intestinal epithelium are introduced.  相似文献

In this review, several features of the cells originating from the lateral borders of the primitive neural anlagen, the neural crest (NC) are considered. Among them, their multipotentiality, which together with their migratory properties, leads them to colonize the developing body and to participate in the development of many tissues and organs. The in vitro analysis of the developmental capacities of single NC cells (NCC) showed that they present several analogies with the hematopoietic cells whose differentiation involves the activity of stem cells endowed with different arrays of developmental potentialities. The permanence of such NC stem cells in the adult organism raises the problem of their role at that stage of life. The NC has appeared during evolution in the vertebrate phylum and is absent in their Protocordates ancestors. The major role of the NCC in the development of the vertebrate head points to a critical role for this structure in the remarkable diversification and radiation of this group of animals. One, three, seven, and six of polymorphic chloroplast microsatellite (cpSSR) markers were developed from four mangrove species, Acanthus ilicifolius, Aegiceras corniculatum, Avicennia marina, and Lumnitzera racemosa, respectively. Characterization of 229, 509, 369, and 216 individuals of A. ilicifolius, A. corniculatum, A. marina, and L. racemosa, collected from different natural mangrove populations (A. ilicifolius, 6; A. corniculatum, 14; A. marina, 10; L. racemosa, 6) in the southern coastline of China showed that these loci provide cpSSR markers with polymorphisms ranging from two to four alleles per locus and gene diversity between 0.005 and 0.675. Combining the polymorphic cpSSR loci of each species, 3, 5, 11, and 4 of cpSSR haplotypes were separately detected in populations of A. ilicifolius, A. corniculatum, A. marina, and L. racemosa in the southern coastline of China. These cpSSR markers will be useful for analyzing the maternal lineage distributions and population genetic structures of these four species.  相似文献

Myelomonocytic lineage cells express anM r 60 000 mannose specific lectin, MR60 (Pimpaneauet al. (1991),Carbohydr Res 213: 95–108). Under non-reducing conditions, this protein migrates as a 120 000 protein. MR60 does not contain anyN-glycan moiety cleavable by the action ofN-glycanase. MR60 induces a sugar selective aggregation of beads coated with glycosylated albumin: beads bearing -d-mannosyl residues are aggregated while beads bearing -d-glucosyl residues are not. A monoclonal antibody Lec101B, specific for MR60, recognizes a singleM r 60 000 protein by Western blotting. This monoclonal antibody does not label the cell surface of cells expressing MR60, but decorates intracellular vesicles upon permeabilization of these cells.  相似文献

Summary Cell lineage relationships observable inDrosophila gynandromorphs have been used to locate the primordia of the individual thoracic disc relative to each other in the embryo. Three observations indicate that the borders of the individual disc primordia lie very close to each other, separated by few if any non-disc cells.First, the frequency of mosaicism within each disc indicates large primordia, of similar magnitude to the distances between the centres of adjacent primordia.Second, very few XX/XO-borders cut between adjacent discs without also cutting into one or the other disc.Third, sturt distances between points on adjacent discs are often much smaller than distances measured within individual discs. Summary Monoclonal antibodies specifically recognizing cardiac and extraocular muscle myosin heavy chains of the quail (Coturnix coturnix japonica) were used to determine the patterns of expression of these isoforms in clonal cultures of embryonic quail myoblasts. Myoblasts prepared from 9 day embryonic pectoralis are virtually homogeneous in their ability to form clones expressing both cardiac and extraocular isoforms. The majority of myoblasts obtained from day 5 embryos also formed clones which co-express the cardiac and extraocular isoforms, but a small percentage of the clones expressed only cardiac isoforms.  相似文献
