电镜原位观察结合图象分析研究了多头绒泡菌Physarum polycephalum Schw间期细胞核和中期染色体中银染蛋白的形状、大小和分布。结果看到,银染蛋白主要呈颗粒状存在于间期细胞核和中期染色体中。银粒的大小不一,分布不均匀。间期细胞核中存在众多直径在5~15nm的银粒,其中10nm以上的较大银粒主要分布于核仁,集缩染色质和核基质部分10nm以上银粒不多。中期细胞核内10nm以上的较大银粒主要分布于染色体中。染色体中除含有一些较大银粒外,多数银粒的直径为5~10nm。本文结果提示,构成染色体骨架的嗜银蛋白可能来自间期细胞核的染色质、核基质和核仁。  相似文献

Summary In the gastric mucosa of two teleost species, the perch (Perca fluviatilis) and the catfish (Ameiurus nebulosus) three endocrine cell types were found, located predominantly between the mucoid cells of the gastric mucosa. A fourth cell type is present in the gastric glands of catfish. Each cell type was defined by its characteristic secretory granules. Type-I cells were predominant in both fish. These cells contained round or oval granules with a pleomorphic core. The average diameter of granules was 400 nm for the perch and 270 nm for the catfish. Type-II cells of both species displayed small, highly osmiophilic granules about 100 nm in diameter. The secretory granules of type-III cells (260 nm in the perch and 190 nm in the catfish) were round or slightly oval in shape and were filled with a finely particulate electron-dense material. Type-IV cells of the catfish were found in the gastric glands only. Their cytoplasm was filled with homogeneous, moderately electron-dense granules averaging 340 nm in diameter. The physiological significance of these different morphological types of gastric endocrine cells requires further investigation.  相似文献

Abstract

Eighteen plant species belonging to 11 families commonly growing in the Pathratola, Dhorli and Pipardhar areas of the Malanjkhand Copper Province were studied. High Cu values were observed in the leaves of the abundant species of Terminalia alata in Pipardhar area. The leaves of T. alata also show wide pre- and post-monsoon seasonal variations in their Cu contents (average: 593-1,713 μg g-1). Amongst several samples from various species, most of the leaf samples of T. alata, collected from the Pipardhar area showed high Cu values that lie within and occupy the high ends of cumulative probability curves. Anomalous Cu values of > 200 and > 497 μg g-1were obtained for most of the T. alata leaves of the Pipradhar area for pre- and post-monsoon periods, respectively. The correlation coefficient values between Cu contents of T. alata leaves and substrate soil do not show a significant relationship for any area in either season. However, t- test results between Cu contents of T. alata leaves and substrate soil samples indicated a significant relationship for the samples collected from the Pipardhar and Pathratola areas during the pre-monsoon period. For post-monsoon period, the t-test results show non-significant relationship in the Pipardhar area. Another sampling area selected adjacent to the Pipardhar area had significance in the t-test results between the two sample periods. Polynomial curves revealed that 465 and 480 μg g-1 of Cu in the soil are the “toxicity threshold limits” for the pre- and post-monsoon periods, respectively. Possible reasons attributed to the high bioassay values found in the T. alata leaves have been discussed in the paper.  相似文献

Ultrastructural studies of cereal anthers found intranuclear bundles of microfilaments in pollen mother cells (PMCs) but not elsewhere. The ultrastructure, distribution, and behaviour of this fibrillar material (FM) are described. FM was seen in all 19 genotypes studied comprising Aegilops, Triticum, Secale, Hordeum and Avena species, which together included haploid, diploid and allo-and autopolyploid, and natural and synthetic polyploid examples. Detailed studies in diploid S. cereale, and hexaploid T. aestivum and Triticale showed that FM was present in PMC nuclei during premeiotic interphase, leptotene and zygotene but not at pachytene and later meiotic stages. Moreover, it was most abundant at late premeiotic interphase in T. aestivum, and at leptotene in S. cereale and Triticale, when it occurred in up to 100% of sampled PMC nuclei in an anther. Although FM and synaptonemal complex (SC) occurred together in some PMC nuclei at later stages, FM was present long before SC, and reached its peak of abundance before SC did. Bundles of FM often formed links at their ends between either two masses of chromatin, or more rarely, between chromatin and the nuclear membrane. Individual bundles of FM varied in length but showed roughly similar ranges of lengths and widths in these three species. They were up to about 0.2 m in diameter and about 3 m in length, equivalent to about 20% of the maximum diameter of the nuclei containing them. Reconstructions of PMC nuclei indicated that FM was never associated with centromeres but was sometimes, and perhaps usually, associated with telomeric or sub-telomeric chromosome segments.The function of FM is unknown but its possible role is discussed in relation to (1) previously described intranuclear inclusions in meiocytes and (2) the cytogenetics and developmental behaviour of meiotic nuclei in the wheat comparium. As FM was a constant and characteristic structural component of PMC nuclei, its presence is probably of functional significance to the meiotic process. If so, it may function before, and over greater distances, than SC in establishing or maintaining the coorientation of chromosomes prerequisite for normal chromosome pairing. As FM was most abundant at stages when major chromosome movements occur, yet its distribution was non-centromeric, it is suggested that it may function in the attachment and movement of telomeres at the nuclear membrane formed after premeiotic mitosis. The possibility that a bundle of FM normally links corresponding sites on two homologues is considered.  相似文献

Carthamus divaricatus (Beg. et Vacc.) Pamp., found only in Libya, has 11 pairs of chromosomes, a new chromosome number in the genus. The species is distinct morphologically. It has yellow, purple, and white corollas, yellow pollen, dark-purple striped anthers, horizontal branches, and strongly divaricate outer involucral bracts. The terminal portion of the middle involucral bracts is dentate and reddish brown. It is self-incompatible. Meiosis is regular in the different corolla-color types of C. divaricatus and their intraspecific hybrids. C. divaricatus was crossed to six species with n = 12, to three species with n = 10, to C. lanatus with n = 22, and to two species with n = 32 chromosomes. The morphological characteristics and cross-ability of the parental species plus the pollen viability, seed-set, and meiotic behavior of the hybrids involving C. divaricatus and other Carthamus species indicated that C. divaricatus is very closely related to species with n = 10, closely related to C. lanatus with n = 22, and less closely related to C. tinctorius with n = 12 chromosomes. C. divaricatus seems to be distantly related to C. nitidus (n = 12). It is proposed that C. divaricatus be included provisionally with 10-chromosome species in Section II. Alternative hypotheses for the development of the three basic chromosome numbers are discussed.  相似文献

ABSTRACT. Ultrastructural cytochemical techniques were used to analyze the nucleus and the kinetoplast of epimastigotes of Trypanosoma cruzi. With the use of ethanolic phosphotungstic acid, which detects basic proteins, reaction product was seen in the chromatin and at the periphery of the kinetoplast. Thallium alcoholate, which interacts with DNA, stained strongly the whole kinetoplast and the chromatin. With the use of a silver impregnation method that detects acidic nucleolar proteins, silver granules were seen preferentially located in the central region of the nucleolus. With the EDTA method, which reveals the presence of ribonucleoproteins, staining was observed in the nuclear pores. Also 6–8 nm fibrils, 25 nm and 40 nm granules, which correspond to the perichromatin fibers, interchromatin granules and the perichromatin granules, respectively, were identified in the nucleus. The EDTA method also revealed the presence of 40 nm granules in the kinetoplast. These granules were seen mainly at the two extremities of the kinetoplast. Freeze-fracture images indicate that the nuclear membrane contains ca. 9 pores/μm2 of nuclear surface area. The mean diameter of the pores was 80 nm. All these results suggest that epimastigotes of T. cruzi have a very active nucleus and a high rate of nucleocytoplasmic interchange.  相似文献

The evolutionary and population demographic history of marine red algae in East Asia is poorly understood. Here, we reconstructed the phylogeographies of two upper intertidal species endemic to East Asia, Gelidiophycus divaricatus and G. freshwateri. Phylogenetic and phylogeographic inferences of 393 mitochondrial cox1, 128 plastid rbcL, and 342 nuclear ITS2 sequences were complemented with ecological niche models. Gelidiophycus divaricatus, a southern species adapted to warm water, is characterized by a high genetic diversity and a strong geographical population structure, characteristic of stable population sizes and sudden reduction to recent expansion. In contrast, G. freshwateri, a northern species adapted to cold temperate conditions, is genetically relatively homogeneous with a shallow population structure resulting from steady population growth and recent equilibrium. The overlap zone of the two species roughly matches summer and winter isotherms, indicating that surface seawater temperature is a key feature influencing species range. Unidirectional genetic introgression was detected at two sites on Jeju Island where G. divaricatus was rare while G. freshwateri was common, suggesting the occurrence of asymmetric natural hybrids, a rarely reported event for rhodophytes. Our results illustrate that Quaternary climate oscillations have left strong imprints on the current day genetic structure and highlight the importance of seawater temperature and sea level change in driving speciation in upper intertidal seaweed species.  相似文献

 Cytoplasmic somatic hybrids (cybrids) between the two sexually incompatible species Nicotiana tabacum and Nicotiana alata were constructed. A total of 33 green regenerants were obtained after fusion of protoplasts from a tobacco cytoplasmic chlorophyll-deficient mutant and gamma irradiation-inactivated leaf protoplasts of N. alata. Twenty nine of them were male sterile and displayed an altered stamen morphology (formation of petaloid and stigmoid structures instead of stamens). Southern-blot analyses of eight CMS plants using N. alata-specific nuclear repetitive DNA and cpDNA probes revealed that they contained nuclear genetic material of N. tabacum and chloroplasts from N. alata. Restriction-enzyme analysis of mitochondrial DNAs of the cybrids in question showed different patterns consisting of an incomplete mix of mtDNA fragments from both parents, as well as new fragments. Southern-blot analysis of mtDNAs with a sunflower atpA probe gave the same recombinant hybridization pattern for all analyzed cybrids, indicating that high-frequency specific recombination occurs in the atpA region. Analysis of the progeny from three successive backcrosses of the studied cybrids with N. tabacum demonstrated a strict cytoplasmic inheritance of the male-sterile phenotype. Fine structure of gametocytes and oocyst formation of Sarcocystis sp. from Quiscalus quiscula Linnaeus grown in cultured embryonic bovine kidney cells was studied. Microgametocytes measured up to ~5 μm diameter. During nuclear division of the microgametocyte, dense plaques were found adjacent to the nucleus just beneath the pellicle; occasionally microtubules were present within these plaques. These microtubules subsequently formed 2 basal bodies with a bundle of 4 microtubules between them. Microgametocytes also contained numerous mitochondria, micropores, granules, vacuoles, and free ribosomes. Each microgamete was covered by a single membrane and consisted of 2 basal bodies, 2 flagella, a bundle of 4 microtubules, a perforatorium, a mitochondrion, and a long dense nucleus which extended anteriorly and posteriorly beyond the mitochondrion. The bundle of 4 microtubules is thought to be the rudiment of a 3rd flagellum. Macrogametes were covered by a double membrane pellicle, and contained a large nucleus (~2.5 μm), vacuoles, and a dilated nuclear envelope connected with the rough endoplasmic reticulum (ER). In young macrogametes (~4 μm), the ER was arranged in concentric rows in the cortical region, and several sizes of dense granules were found in the cytoplasm. However, in later stages (~8 μm) the ER was irregularly arranged and was dilated with numerous cisternae; only large dark granules remained and a few scattered polysaccharide granules were found. No Golgi apparatus or micropores were observed. After the disappearance of dark granules 5 concentric membranes appeared. Four of these fused to form an oocyst wall composed of a dense outer layer (~66 nm thick) and a thin inner layer (~7 nm). The 5th or innermost membrane surrounded the cytoplasmic mass which was covered by a 2-layered pellicle and contained a nucleus, small amounts of ER, large vacuoles, and mitochondria. Using electron spectroscopic imaging, a new type of small granular structural constituent has been observed in the extrachromosomal zone of the polytene nucleus of the salivary gland cells ofChironomus thummi andChironomus tentans. These granules appear isolated or in small clumps and are often seen to be connected with surrounding thin fibrils. They are stained by the EDTA procedure, which is preferential for nuclear ribonucleoprotein (RNP) constituents, and by the bismuth oxynitrate method for visualizing phosphorylated compounds. The granules are 15–23 nm in diameter and are digested by prolonged post-embedding RNAse hydrolysis. These structural elements contain the highest concentration of phosphorus in the interchromosomal space as revealed by electron energy loss spectroscopy. The small granules exhibit several morphological and cytochemical features in common with interchromatin granules, but they are not labeled with antibodies directed against extranucleolar small nuclear RNPs (snRNPs), as are interchromatin granules.  相似文献

The effect of cell cycle mutation ff3 on chromosome segregation was studied on fixed cells of neural ganglia of Drosophila melanogasterlarvae. The cell distributions by diameter of interphase nuclei and by distance between sister chromatid sets were compared at anaphase and telophase. In the control wild-type strain Lausenne, the cell distribution by distance between sister chromatids in anaphase was similar to their distribution by nuclear size. The mean distance between segregating chromatids at anaphase (l av) coincided with the mean diameter of interphase nuclei (d av) and was 8.3 m. Cells passed to telophase when chromatids were at least 10 m apart. The mutant ff3 strain differed from the control strain Lausenne in cell distribution by interphase nuclear diameter and distance between sister chromatids in anaphase; the mean nuclear diameter and mean distance between segregating chromatids similarly increased to 9.3 m. A specific feature of mitosis in mutant strain ff3 was a premature beginning of telophase chromatin reorganization. This caused the occurrence of cells with abnormally short (less then the interphase nuclear diameter) distance between sister chromatid sets in telophase but not in anaphase, as if these cells had passed from anaphase to telophase prematurely, during the chromatid movement toward poles in anaphase A.  相似文献

Summary— The ultrastructural organization of the interphase nucleus of the green alga Chlamydomonas reinhardtii was investigated and found to be largely dependent on the fixation conditions. In specimens stained with bismuth, densely contrasted granules ranging from 25 to 45 nm in diameter were localized throughout the interchromatin space and often formed clusters. These granules were labeled by RNase A-gold complexes and may represent the counterparts of animal and higher plant cll interchromatin granules. Within the nucleolus the Ag-NOR and pyroantimonate stains and, to a lesser extent, the bismuth stain reacted with the nucleolar dense fibrillar component (DFC). When cells were subjected to a heat shock at 42°C, the nucleolar DFC was found to progressively separate from the nucleolus and, after 3 h, appeared as a continuous meandering thread about 0.1 μm in width. Within the nucleolus, labeling on conventional preparations occurred as small clusters with antibodies to H3 histones or to DNA whereas RNase A-gold complexes labeled most of it including fibrillar centers. Improved ultrastructural preservation in cryofixed, cryosubstituted specimens gently fixed in glutaraldehyde permitted to localize nucleolar DNA predominantly at the outer edge of fibrillar centers and to a lesser extent within the neighbouring DFC. Our results indicate that the structure and composition of Chlamydomonas interphase nuclei are comparable, despite particularities, to those of animal and higher plant nuclei.  相似文献

The interphase nucleus of the chloromonadophycean alga, Gonyostomum semen (Ehrenberg) Diesing, has a highly distinctive appearance. Interphase chromatin is readily distinguishable in both light and electron microscope preparations. It extends throughout the neucleus and frequently makes contact with the nucleoli and with the nuclear envelope. Among the chromatin filaments are large numbers of 35-46 nm diameter granules which occur singly or in clusters. The nucleoli are characteristically located in the posterior half of the nucleus and are composed of granular and non-granular components. Nuclear pores occur in slight depressions of the nuclear surface; their lumen has a diameter of approximately 75 nm and contains electron-dense material. The chromatin and the large numbers of nuclear granules are unusual and warrant further investigation.  相似文献

Phylogenetic relationships of Oceanian staple yams (species of Dioscorea section Enantiophyllum) were investigated using plastid trnL‐F and rpl32‐trnL(UAG) sequences and nine nuclear co‐dominant microsatellites. Analysis of herbarium specimens, used as taxonomic references, allowed the comparison with samples collected in the field. It appears that D. alata, D. transversa and D. hastifolia are closely related species. This study does not support a direct ancestry from D. nummularia to D. alata as previously hypothesized. The dichotomy in D. nummularia previously described by farmers in semi‐perennial and annual types was reflected by molecular markers, but the genetic structure of D. nummularia appears more complex. Dioscorea nummularia displayed two haplotypes, each corresponding to a different genetic group. One, including a D. nummularia voucher from New Guinea, is closer to D. tranversa, D. alata and D. hastifolia and encompasses only semi‐perennial types. The second group is composed of semi‐perennial and annual yams. However, some of these annual yams also displayed D. alata haplotypes. Nuclear markers revealed that some annual yams shared alleles with D. alata and semi‐perennial D. nummularia, suggesting a hybrid origin, which may explain their intermediate morphotypes and the difficulty met in classifying them.  相似文献

Electron microscope studies on the premetaphase stages of micronuclear divisions of Paracineta limbata and Ichthyophtirius multifiliis showed that spindle material also exists during interphase. In the case of I. multifiliis scattered microtubule fragments persist in the nuclear space; in P. limbata the micronuclei contain a small paracrystalloid which is suggested to be microtubular protein. Wide microtubules, varying in diameter from 300 to 400 Å develop during intranuclear prophase near the nuclear envelope in both cases. There are good reasons to assume that they function as a kind of stem body during the enlargement of the surface area of the nuclear envelope. Later micronuclear prophase stages of both species show a some-what different development. In I. multifiliis, there are scattered groups of short microtubular segments, partly in parallel array, whereas in P. limbata the wide tubules are transformed into normal microtubules of 180–200 Å diameter. The nuclei of both species are similar at late prophase and prometaphase stages. Bundles of interpolar microtubules run between the chromosomes, and single microtubules, presumably induced by the chromosomes, cross them at different angles. The chromosome-induced microtubules appear a short time after the interpolars. At prometaphase stage all microtubules show a highly parallel arrangement and therefore it is suggested that chromosomal tubules reach their final polar orientation by the action of cross-bridges.  相似文献

【目的】明确阔叶丰花草在荔枝园定植对杂草群落特征及物种多样性的影响,为阔叶丰花草在果园应用提供参考依据。【方法】通过调查广东茂名高州荔枝国家现代农业产业园的荔枝园中90个有、无阔叶丰花草样方中杂草群落组成、群落高度以及物种多样性,探究阔叶丰花草定植对荔枝园杂草群落结构及物种多样性的影响。【结果】阔叶丰花草在荔枝园定植后,杂草种类减少,由对照样方的42种减少到试验样方的30种;杂草群落结构显著改变,牛筋草、白花鬼针草等恶性杂草重要值降低,薇甘菊、香附子、火炭母等杂草消失;阔叶丰花草的重要值与杂草群落高度和物种多样性指数为显著性负相关(P0.001),且均呈显著三项式回归关系。阔叶花草在荔枝园定植后可建立结构较为单一,且低矮的杂草群落。【结论】阔叶丰花草有望成为荔枝园生草的有益草种。  相似文献

Unicellular autofluorescent picoplankton ranging from 0.4 to 1.5 µm in diameter were found to be a significant component of phytoplankton in the North Basin of Lake Biwa during early summer in 1989 and 1990. The abundance of these picoplankton varied seasonally by about three orders of magnitude with one maximum of up to 106 cells ml–1. Bloom-forming picoplankton were isolated by dilution and further cultivated in liquid medium. Three clones were found to be representative species of the bloom. Using epifluorescence and electron microscopy as well as absorption and fluorescence emission spectroscopy, we examined these clones according to shape and pigment composition. They have ringlike thylakoids, are photosynthetically active and have no nuclear envelope. The cyanobacterial clones isolated represent three types containing phycobilisomes with either phycocyanin or phycoerthrin as the dominant accessory pigment. They are described here as three new species, two phycoerythrin-rich types and one phycocyanin-rich type, all of them belonging to the Synechococcus group. The differences found by fluorescence emission of isolated clones are discussed with respect to in situ strain identification.  相似文献

Modern methods for species delimitation provide biologists with the power to detect cryptic diversity in nearly any system. To illustrate the application of such methods, we collected data (21 sequence loci) from a carnivorous plant in southeastern North America and applied several recently developed methods (Gaussian clustering, Structurama, BPP, spedeSTEM). The pale pitcher plant Sarracenia alata inhabits the southeastern USA along the northern coast of the Gulf of Mexico. Sarracenia alata populations are separated by the Mississippi River and Atchafalaya Basin, a known biogeographical barrier in this region, but the cohesiveness of S. alata as currently classified has not been tested rigorously. Multiple analytical approaches (including allelic clustering and species trees methods) suggest that S. alata comprises two cryptic lineages that correspond to the eastern and western portions of the plant's distribution. That such clear genetic evidence for cryptic diversity exists within S. alata and is in conflict with other sources of data (e.g. morphology, environmental differentiation) illustrates a conundrum faced by those who investigate species boundaries: genetic data are often the first type of data to accumulate evidence of differentiation, but most existing taxonomic treatments are based on nongenetic data. Our results suggest that S. alata as currently described contains two cryptic species, and we recommend the elevation of the western populations to species status. © 2013 The Linnean Society of London, Biological Journal of the Linnean Society, 2013, 109 , 737–746.  相似文献

The morphology of vitellogenic oocytes and the batch fecundity of five tropical forage species belonging to the family Characidae, were studied in 104 late‐maturing ovaries. Significant morphological differences between vitellogenic oocytes and ovarian follicles were found. The lowest batch fecundity values were recorded in Hemigrammus marginatus (480 ± 163) and Orthospinus franciscensis (1701 ± 562), which were the smaller species in terms of total length, body weight and oocyte diameter. The highest batch fecundity value was observed in Tetragonopterus chalceus (8384 ± 3944) having the highest GSI and oocyte diameter. Batch fecundity and gonad weight was highly correlated followed by body weight and total length. Relative fecundity was estimated per unit total length, body weight and gonad weight. The wide variation in fecundity observed between the specimens and the species analysed is possibly related to the multiple spawning reproductive strategy of these fishes. Since T. chalceus have higher values of batch and relative fecundity, it is concluded that this species has a higher reproductive potential than the other forage species studied.  相似文献

Summary The organisation of the spermatozoa ofAnthopharynx sacculipenis is described, based on electron-microscopical observations. The male gametes are fili-form in shape. They are totally enclosed by cortical microtubules and possess two free cilia. Special features are dot-like dense granules arranged in regular rows and terraced elaborations of the nuclear membrane. Such terraced elaborations are not known in any other species of flatworms whereas dot-like dense granules are described for some other taxa of the Rhabdocoela.
