When grown in defined media, 15 of 21 laboratory and clinical strains ofNeisseria gonorrhoeae produced one or more factors (cytotaxins) chemotactic for human polymorphonuclear leukocytes obtained from normal individuals. Production of the cytotaxin(s) depended upon the media in which the bacteria were grown and the growth phase of the cultures, as well as upon the strain of gonococcus.  相似文献

In the present study we examined whether endothelin-1 stimulation of human monocytes causes release of chemotactic factors. It was found that monocytes released neutrophil- and monocyte-chemotactic activity in a dose- and time-dependent manner in response to ET-1. ET-1 did not show any chemotactic activity by itself. NCA was detected in monocyte supernatants in response to ET-1 (0.01-100 nM) after 1, 4, 8 and 24 h stimulation. MCA was detected only after 24 h stimulation with ET-1 (0.1-100 nM). Preincubation of the monocyte cultures with the lipoxygenase inhibitors nordihydroguaiaretic acid (10(-4) M) or diethylcarbamazine (10(-9) M) completely abolished the appearance of NCA and MCA. NCA was neutralized by > 75% using a polyclonal antibody against human interleuktn-8. The ET-1 induced release of IL-8 was confirmed by IL-8 ELISA. A monoclonal antibody against human monocyte chemotactic protein-1 neutralized MCA by > 80%. It is concluded that ET-1 stimulation of monocytes in vitro causes release of neutrophil- and monocyte-chemotactic activity identified as IL-8 and MCP-I respectively. An intact lipoxygenase pathway is crucial for this effect of ET-1 to occur.  相似文献

In order to establish the species cross-reactivity of the human neutrophil attractant/activation protein-1 (interleukin-8, NAP-1/IL-8) and find which experimental species are responsive to the human cytokine, blood polymorphonuclear leukocytes (PNMLs) were isolated from chicken, dog, goat, guinea-pig, monkey, mouse, pig, rabbit, and rat and their in vitro migration in response to this cytokine was investigated. PMNLs from all of the tested species migrated in response to recombinant human NAP-1/IL-8 (rhNAP-1/IL-8). The potency of rhNAP-1/IL-8 for the PMNLs of different species varied and was considerably lower than its potency for human cells. The morphological study combined with the leukocyte enumeration in the intradermal rhNAP-1/IL-8 injection sites established an in vivo proinflammatory potency of rhNAP-1/IL-8 for rabbit and rat that was comparable to the observed in vitro chemotactic potency of rhNAP-1/IL-8 for neutrophils of these species.  相似文献

The affinity-based Nα-amino protecting group tetrabenzo [a,c,g,i]fluorenyl-17-methoxycarbonyl (Tbfmoc) has been utilized as a hydrophobic probe to allow the simple, quick and highly effective isolation of a 76 residue cysteine-containing protein (MCP-1). The base-labile Tbfmoc group can be removed under very mild conditions, which preserve the thiol-con taining protein in the reduced state. E-selectin (CD62E) is an endothelial specific glycoprotein belonging to the selectin family of adhesion molecules. Because a high expression of this molecule at intestinal mucosal surfaces in inflammatory bowel disease (IBD) has been described earlier, the aim was to assess serum levels of E-selectin (sE-selectin) and to correlate it to disease activity, and further to evaluate its chemotactic properties at physiological concentrations. Levels of sEselectin were measured by a sandwich ELISA technique in 31 IBD patients together with 15 healthy volunteers. In ulcerative colitis the median value was 0.46 nM (0.16-0.75), in Crohn's disease 0.47 nM (0.22-1.24), and in healthy controls 0.34 nM (0.22-0.83). No statistically significant differences in sE-selectin were revealed between these groups (p > 0.05). The in vitro chemotactic capabilities of E-selectin (in the concentration range of 0.10-31.4 nM) were assessed using the leading front technique. A significantly increased migratory response was found at concentrations of 1.00 (p < 0.05) and 3.14 nM (p < 0.02). It is concluded that sE-selectin in contrast to sICAM-1 does not act as a sensitive indicator of local immune activation in IBD. However, E-selectin may be important for recruitment and accumulation of neutrophilic granulocytes and other phagocytes involved in the inflammatory process seen in IBD. Future investigations are encouraged in order to reveal its in vivo effects.  相似文献

In non-alcoholic fatty liver disease (NAFLD) and insulin resistance, hepatic de novo lipogenesis is often elevated, but the underlying mechanisms remain poorly understood. Recently, we show that CDK8 functions to suppress de novo lipogenesis. Here, we identify the mammalian target of rapamycin complex 1 (mTORC1) as a critical regulator of CDK8 and its activating partner CycC. Using pharmacologic and genetic approaches, we show that increased mTORC1 activation causes the reduction of the CDK8-CycC complex in vitro and in mouse liver in vivo. In addition, mTORC1 is more active in three mouse models of NAFLD, correlated with the lower abundance of the CDK8-CycC complex. Consistent with the inhibitory role of CDK8 on de novo lipogenesis, nuclear SREBP-1c proteins and lipogenic enzymes are accumulated in NAFLD models. Thus, our results suggest that mTORC1 activation in NAFLD and insulin resistance results in down-regulation of the CDK8-CycC complex and elevation of lipogenic protein expression.  相似文献

Catalase activity in crude extracts ofEscherichia coli has two pH optima: one at pH 6.8 and the other at pH 10.5. The former is inducible by H2O2 and is the major species in exponential cells. The latter is constitutive and is the major species in stationary cells. Both activities are repressed by glucose. Catalase-negative mutants that are impaired in the pH-6.8 activity were isolated by the inability of aerobically grown mutagenized colonies to decompose H2O2. A capillary assay was employed to quantify chemotactic responses in the chemoautotrophic bacterium,Thiobacillus thioparus. NH4Cl, KNO3, and Na2S2O3 were strong attractants. The minimum concentration of each of these inorganic chemicals needed to elicit an observable response was approximately 10–4 M.T. thioparus did not respond to carbohydrates or amino acids.  相似文献

Earlier studies have suggested that myogenic cells of somite origin migrate into the developing limb, but little is known about the factors affecting the pattern of migration. In order to understand the migratory behavior of myogenic cells, embryonic skeletal muscle cells were tested for their ability to migrate chemotactically using a modified Boyden chamber assay system. It is shown here, for the first time, that embryonic skeletal muscle cells have the capacity to migrate toward a gradient of platelet-derived growth factor (PDGF) and PDGF-like factors present in serum and chick embryo extract (CEE). On the other hand, nonmyogenic limb mesenchyme cells do not exhibit such a response. A hypothesis is proposed here that chemotactic factors from the already patterned vasculature might influence the distribution of skeletal muscle cells during early limb development.  相似文献

The behavior of spiroplasmas in diffusion-generated concentration gradients of attractants and repellents was examined microscopically. Motility was strongly biased in a direction parallel to attractant gradients, and examination of individual cells showed that the characteristic flexing of the cell body took place less frequently under these conditions, suggesting that flexing is associated with changes in the direction of swimming.  相似文献
