Two treatments were employed to influence the amount of amino nitrogen (N) transport in phloem. In walnut trees (Juglans regia L.), developing fruit significantly reduced the efflux of foliar-applied 15N-enriched urea from treated spurs over a 33-day period in comparison with similarly-treated defruited spurs. Those data suggest that local aboveground demand for N influences vascular transport of amino N. In another experiment, a 1% urea solution was applied foliarly to 5-year old `Mission' almond trees [Prunus dulcis (Mill.) D. A. Webb] to increase the concentration of amino N in the phloem. The effect of foliar N treatments on a) the transport and distribution of labelled urea N within the trees over the experimental period and b) the uptake of soil-applied labelled N were determined by replicated whole tree excavation, fractionation into various tree components and mass spectrometric analyses of the 14N/15N ratios. Concentrations and composition of amino acids in the phloem and xylem saps of control trees and trees receiving foliar-applied urea were also determined. In foliar urea-treated trees, the amino acid concentrations increased significantly in leaf and bark phloem exudate, within 24 and 96 h, respectively. Foliar-applied urea N was translocated to the roots of almond trees over the experimental period and decreased soil N uptake. The results of these experiments are consistent with the hypothesis that aboveground N demand affects the amount of amino N cycling between shoots and roots, and may be involved in the regulation of soil N uptake. Background  

Although the effects of P deficiency on tea (Camellia sinensis (L.) O. Kuntze) growth, P uptake and utilization as well as leaf gas exchange and Chl a fluorescence have been investigated, very little is known about the effects of P deficiency on photosynthetic electron transport, photosynthetic enzymes and carbohydrates of tea leaves. In this study, own-rooted 10-month-old tea trees were supplied three times weekly for 17 weeks with 500 mL of nutrient solution at a P concentration of 0, 40, 80, 160, 400 or 1000 μM. This objective of this study was to determine how P deficiency affects CO2 assimilation, Rubisco, carbohydrates and photosynthetic electron transport in tea leaves to understand the mechanism by which P deficiency leads to a decrease in CO2 assimilation.  相似文献

Pedunculate oak (Quercus robur L.) was germinated and grown at ambient CO2 concentration and 650 μmol mol?1 CO2 in the presence and absence of the ectomycorrhizal fungus Laccaria laccata for a total of 22 weeks under nonlimiting nutrient conditions. Sulphate uptake, xylem loading and exudation were analysed in excised roots. Despite a relatively high affinity for sulphate (KM= 1.6 mmol m?3), the rates of sulphate uptake by excised lateral roots of mycorrhizal oak trees were low as compared to herbaceous plants. Rates of sulphate uptake were similar in mycorrhizal and non-mycorrhizal roots and were not affected by growth of the trees at elevated CO2. However, the total uptake of sulphate per plant was enhanced by elevated CO2 and further enhanced by elevated CO2 and mycorrhization. Sulphate uptake seemed to be closely correlated with biomass accumulation under the conditions applied. The percentage of the sulphate taken up by mycorrhizal oak roots that was loaded into the xylem was an order of magnitude lower than previously observed for herbaceous plants. The rate of xylem loading was enhanced by mycorrhization and, in roots of mycorrhizal trees only, by growth at elevated CO2. On a whole-plant basis this increase in xylem loading could only partially be explained by the increased growth of the trees. Elevated CO2 and mycorrhization appeared to increase greatly the sulphate supply of the shoot at the level of xylem loading. For all treatments, calculated rates of sulphate exudation were significantly lower than the corresponding rates of xylem loading of sulphate. Radiolabelled sulphate loaded into the xylem therefore seems to be readily diluted by unlabelled sulphate during xylem transport. Allocation of reduced sulphur from oak leaves was studied by flap-feeding radiolabelled GSH to mature oak leaves. The rate of export of radioactivity from the fed leaves was 4–5 times higher in mycorrhizal oak trees grown at elevated CO2 than in those grown at ambient CO2. Export of radiolabel proceeded almost exclusively in a basipetal direction to the roots. From these experiments it can be concluded that, in mycorrhizal oak trees grown at elevated CO2, the transport of sulphate to the shoot is increased at the level of xylem loading to enable increased sulphate reduction in the leaves. Increased sulphate reduction seems to be required for the enhanced allocation of reduced sulphur to the roots which is observed in trees grown at elevated CO2. These changes in sulphate and reduced sulphur allocation may be a prerequisite for the positive effect of elevated CO2 on growth of oak trees previously observed.  相似文献

Summary A gas transport system based upon the physico-chemical effect of thermo-osmosis of gases in described for the black alder, Alnus glutinosa (L.) Gaertn. Air is transported through the alder's stem to the roots, thus improving O2 supply to respiring tissues of the root system. The gas transport system is investigated by means of a tracer gas technique (11% ethane in air, v/v). Gas transport depends on any source of radiant heat generating a temperature difference between the tree's stems and the atmosphere. The amount of gas transported in leafless trees is four times higher than the amount of gas reaching the roots by gas diffusion. Two-thirds of the gas is transported in the wood, only one-third in the bark. Intercellular spaces inside the porous lenticels of the bark are responsible for this kind of gas transport. Their diameters are estimated by the effusion rates of different tracer gases to be in the range of 1 m.  相似文献

Environmental sensitivity of gas exchange in different-sized trees   总被引:1,自引:0,他引:1  
The carbon isotope signature (δ13C) of foliar cellulose from sunlit tops of trees typically becomes enriched as trees of the same species in similar environments grow taller, indicative of size-related changes in leaf gas exchange. However, direct measurements of gas exchange in common environmental conditions do not always reveal size-related differences, even when there is a distinct size-related trend in δ13C of the very foliage used for the gas exchange measurements. Since δ13C of foliage predominately reflects gas exchange during spring when carbon is incorporated into leaf cellulose, this implies that gas exchange differences in different-sized trees are most likely to occur in favorable environmental conditions during spring. If gas exchange differs with tree size during wet but not dry conditions, then this further implies that environmental sensitivity of leaf gas exchange varies as a function of tree size. These implications are consistent with theoretical relationships among height, hydraulic conductance and gas exchange. We investigated the environmental sensitivity of gas exchange in different-sized Douglas-fir (Pseudotsuga menziesii) via a detailed process model that specifically incorporates size-related hydraulic conductance [soil–plant–atmosphere (SPA)], and empirical measurements from both wet and dry periods. SPA predicted, and the empirical measurements verified, that differences in gas exchange associated with tree size are greatest in wet and mild environmental conditions and minimal during drought. The results support the hypothesis that annual net carbon assimilation and transpiration of trees are limited by hydraulic capacity as tree size increases, even though at particular points in time there may be no difference in gas exchange between different-sized trees. Maximum net ecosystem exchange occurs in spring in Pacific Northwest forests; therefore, the presence of hydraulic limitations during this period may play a large role in carbon uptake differences with stand-age. The results also imply that the impacts of climate change on the growth and physiology of forest trees will vary depending on the age and size of the forest.  相似文献

Oxygen uptake measurements have shown that pressurized gas transport, resulting from the physical effect of thermo-osmosis of gases, improves oxygen supply to the roots of the seedlings in two alder speciesAlnus japonica (Thunb.) Steud. andAlnus hirsuta (Spach) Rupr., which are both native in Japan. When gas transport conditions were established by irradiation of the tree stems the internal aeration was increased to a level nearly equal to the oxygen demand of the root system in leafless seedlings ofA. hirsuta, but was higher inA. japonica so that excess oxygen was excreted into the environment. An increase of superoxide dismutase (SOD) activity, which protects plants from toxic oxygen radicals and post-anoxic injury, has been observed in root tissues ofA. japonica when the seedlings were flooded for 3 days. The increase of SOD activity, in concert with high gas transport rates, may enable this tree species to grow in wet sites characterized by low oxygen partial pressure in the soil and by varying water tables. A less effective gas transport, flood-induced reduction of SOD activity in root tissues, and reduced height growth in waterlogged soil may be responsible for the fact thatA. hirsuta is unable to inhabit wettland sites.  相似文献

Carbon uptake by forests constitutes half of the planet’s terrestrial net primary production; therefore, photosynthetic responses of trees to rising atmospheric CO2 are critical to understanding the future global carbon cycle. At the Swiss Canopy Crane, we investigated gas exchange characteristics and leaf traits in five deciduous tree species during their eighth growing season under free air carbon dioxide enrichment in a 35-m tall, ca. 100-year-old mixed forest. Net photosynthesis of upper-canopy foliage was 48% (July) and 42% (September) higher in CO2-enriched trees and showed no sign of down-regulation. Elevated CO2 had no effect on carboxylation efficiency (V cmax) or maximal electron transport (J max) driving ribulose-1,5-bisphosphate (RuBP) regeneration. CO2 enrichment improved nitrogen use efficiency, but did not affect leaf nitrogen (N) concentration, leaf thickness or specific leaf area except for one species. Non-structural carbohydrates accumulated more strongly in leaves grown under elevated CO2 (largely driven by Quercus). Because leaf area index did not change, the CO2-driven stimulation of photosynthesis in these trees may persist in the upper canopy under future atmospheric CO2 concentrations without reductions in photosynthetic capacity. However, given the lack of growth stimulation, the fate of the additionally assimilated carbon remains uncertain.  相似文献

Photosynthetic induction in leaves of four-month-old Eucalyptus urograndis seedlings and of cuttings obtained from adult trees that were previously dark-adapted was studied by the in vivo and in situ Open Photoacoustic Cell Technique. Results for the gas exchange component of the photoacoustic (PA) signal were interpreted considering that the gas uptake component would have a phase angle nearly opposite to that of the oxygen evolution component. By subtracting the thermal component from the total PA signal, we studied the competition between gas uptake and oxygen evolution during the photosynthetic induction. Seedlings presented a net oxygen evolution prior to cuttings, but cuttings reached a higher steady-state photosynthetic activity. The chlorophyll (Chl) a/b ratio and the Chl fluorescence induction characteristic Fv/Fm were significantly higher for cuttings, while there was no difference between samples in stomata density and leaf thickness. Thus the differences in PA signals of seedlings and cuttings are associated to differences between the photosystem 2 antenna systems of these samples.  相似文献

To assess hydraulic architecture and limitations to water transport across scion–rootstock combinations (Prunus avium L. cultivar Van grafted on five differing size-controlling rootstocks: P. avium (vigorous) > CAB 11E > Maxma 14 > Gisela 5 > Edabriz (dwarfing)), we compared xylem anatomy, and calculated relative hydraulic conductivity (RC) and vulnerability index (VI) of roots (small, medium and large diameter) and stems. Water relations, leaf gas exchange and variations in growth were also determined. Roots exhibited larger-diameter xylem conduits (VD), greater RC and VI than stems in all Van–rootstock combinations. Moreover, there was a significantly higher vessel frequency (VF), lower VD, RC and VI in dwarfed trees, especially grafted on Gisela 5 than trees on the invigorating rootstocks, P. avium L., CAB 11E and Maxma 14. Anatomical constraints on water status imposed by the smaller VD (and/or in lower xylem thickness and root system length) of dwarfed trees imply a series of negative feedbacks, like a decrease in RC, stem water potential, leaf gas exchange and growth. On the other hand, Van grafted on CAB 11E and Maxma 14, with wide vessels and high VI, could be more susceptible to embolism, especially during periods of severe water stress, than trees on dwarfing rootstocks, which had small vessels and low VI.  相似文献

Irrigation effects on whole-plant sap flow and leaf-level water relations were characterised throughout a growing season in an experimental olive (Olea europaea L.) orchard. Atmospheric evaporative demand and soil moisture conditions for irrigated and non-irrigated olive trees were also monitored. Whole-plant water use in field-grown irrigated and rain fed olive trees was determined using a xylem sap flow method (compensation heat-pulse velocity). Foliage gas exchange and water potentials were determined throughout the experimental period. Physiological parameters responded diurnally and seasonally to variations in tree water status, soil moisture conditions and atmospheric evaporative demand. There was a considerable degree of agreement between daily transpiration deduced from heat-pulse velocity and that determined by calibration using the Penman–Monteith equation in the field. Summer drought caused decreasing leaf gas exchange and water potentials, and a progressive increase in hydraulic conductance (stronger in non-irrigated than irrigated trees), probably attributable to modifications in hydraulic properties at the soil-root interface. Negligible hysteresis, attributable to low plant capacitance, was observed in the relationship between leaf water potential and sap flow. A proportional decrease in maximum daily leaf conductance with increasing vapour pressure deficit was observed, while mean daytime canopy stomatal conductance decreased with the season. As a result, plant water use was limited and excessive drought stress prevented. Non-irrigated olive trees recovered after the summer drought, showing a physiological behaviour similar to that of irrigated trees. In addition to physiological and environmental factors, there are endogenous keys (chemical signals) influencing leaf level parameters. Olive trees are confirmed to be economical and sparing users of soil water, with an efficient xylem sap transport, maintenance of significant gas exchange and transpiration, even during drought stress.  相似文献

Much research has focused on effects of plant parasites on host-plant physiology and growth, but little is known about effects of host physiological condition on parasite growth. Using the parasitic dwarf mistletoe Arceuthobium vaginatum subsp. cryptopodum (Viscaceae) and its host Pinus ponderosa, we investigated whether changes in host physiological condition influenced mistletoe shoot development in northern Arizona forests. We conducted two studies in two consecutive years and used forest thinning (i.e., competitive release) to manipulate host physiological condition. We removed dwarf mistletoe shoots in April, before the onset of the growing season, and measured the amount of regrowth in the first season after forest thinning (Study I: n=38 trees; Study II: n=35 trees). Thinning increased tree uptake of water and carbon in both studies, but had no effect on leaf N concentration or δ13C. Mistletoe shoot growth was greater on trees with high uptake of water and carbon in thinned stands than trees with low uptake in unthinned stands. These findings show that increased resource uptake by host trees increases resources to these heterotrophic dwarf mistletoes, and links mistletoe performance to changes in host physiological condition.  相似文献

The uptake of water by apple trees (Malus × domestica‘Golden Delicious’) was measured in a pot experiment and was found to be much greater in apple trees carrying many fruits than in comparable specimens without fruits. The main reason seemed to be differences in the transpiration rates of the leaves. This was corroborated by microscopic investigations which showed a higher degree of stomatal opening in the fruiting trees. In some cases minor reductions in water uptake per tree were caused by reducing the supplies of potassium and nitrogen.  相似文献

Summary The effect of cold soils on stem sap flow, shoot gas exchange and water potential of Picea engelmannii (Parry) was investigated during the snowmelt period in the Medicine Bow Mountains, Wyoming, USA. Shoot net photosynthetic rates were higher in young trees (1.5–1.8 m in height) growing in cold soils (<3.5° C) associated with snowpack, than trees in warm soils until about 1500 h. Higher shoot photosynthetic rates of trees in cold soils continued after snow was removed and could not be completely explained by higher visible irradiance over highly reflective snow. Following soil warming higher photosynthetic rates were evident in these trees for five days. High nutrient availability associated with snowmelt may improve shoot nutrient status leading to higher gas-exchange rates during snowmelt. Shoot conductance to water vapor was higher in trees in cold soil until midday, when declining shoot conductance led to lower intercellular CO2 concentrations. Midday through afternoon shoot water potentials of trees in cold soils were similar or higher than those of trees in warm soils and the lower afternoon shoot conductances in cold soils were not the result of lower bulk shoot water potentials. Decline in net photosynthesis of trees in cold soils at 1500 h paralleled increases in intercellular CO2 concentrations, implying a nonstomatal limitation of photosynthesis. This scenario occurred consistently in mid-afternoon following higher morning and midday photosynthesis in cold soils, suggesting a carbohydrate feedback inhibition of photosynthesis. Diurnal patterns in stem sap flow of all trees (cold and warm soils) reflected patterns of shoot conductance, although changes in stem sap flow lagged 1–3 h behind shoot conductance apparently due to stem water storage. Total daily stem sap flow was similar in trees in cold and warm soils, although diel patterns differed. The morning surge and night-time drop in sap flow commenced 1–2 h earlier in trees in cold soils. Overnight stem sap flow was lower in trees in cold soils, possibly due to higher resistance to root water uptake in cold soils, which may explain lower predawn shoot water potentials. However, midday shoot water potentials of trees in cold soils equalled or exceeded those of trees in warm soils. Higher resistance to root water uptake in P. engelmannii in cold soils was apparently overshadowed by transpirational forces and significant shoot water deficits did not develop.  相似文献

Prunus avium trees were grown in sand culture for one vegetative season with contrasting N supplies, in order to precondition their N storage capacities. During the spring of the second year a constant amount of 15N was supplied to all the trees, and the recovery of unlabelled N in the new biomass production was used as a direct measure of N remobilization. Destructive harvests were taken during spring to determine the pattern of N remobilization and uptake. Measurements of both xylem sap amino acid profiles and whole tree transpiration rates were taken, to determine whether specific amino acids are translocated as a consequence of N remobilization and if remobilization can be quantified by calculating the flux of these amino acids in the xylem. Whereas remobilization started immediately after bud burst, N derived from uptake by root appeared in the leaves only 3 weeks later. The tree internal N status affected both the amount of N remobilization and its dynamics. The concentration of xylem sap amino acids peaked shortly after bud burst, concurrently with the period of fastest remobilization. Few amino acids and amides (Gln, Asn and Asp) were responsible for most of N translocated through the xylem; however, their relative concentration varied over spring, demonstrating that the transport of remobilized N occurred mainly with Gln whereas transport of N taken up from roots occurred mainly with Asn. Coupling measurements of amino acid N in the xylem sap with transpiration values was well correlated with the recovery of unlabelled N in the new biomass production. These results are discussed in relation to the possibility of measuring the spring remobilization of N in field‐grown trees by calculating the flux of N translocation in the xylem.  相似文献

Rapid Effects of Abscisic Acid on Ion Uptake in Sunflower Roots   总被引:1,自引:0,他引:1  
Short-term effects of ABA, ABA + kinetin and kinetin on ion (86Rb-potassium and phosphate) and water uptake in sunflower plants (Helianthus annuus var. californicus) were examined with a continuous-recording technique. Ion uptake in the roots and transport to the shoots were also investigated by conventional tracer uptake experiments and by sap bleeding experiments with excised roots. After addition of 5 × 10?6-4 × 10?5M ABA to the root medium there was an immediate decrease (30–70%) in the rate of ion uptake which lasted 30–70 min. The rate of water uptake was not significantly affected as measured with this method. Ion transport to the shoots and to the bleeding sap of excised roots was decreased by ABA. ABA-induced inhibition of ion uptake was abolished by the presence of kinetin, and uptake was slightly stimulated by 2 × 10?5M kinetin alone. We suggest that concentration gradients of ABA or rapid changes in the ABA-kinetin balance in the roots affect ion uptake and transport.  相似文献

The Caco-2 cell line was used as a model to determine if the type of fatty acid, unsaturated versus saturated, differentially altered the uptake and transport of iron in the human intestine and if the changes were the result of alterations in monolayer integrity and paracellular transport. Cells were cultured in either a lower-iron or normal-iron medium and incubated with a bovine serum albumin control, linoleate, oleate, palmatate, or stearate. Oleate, palmatate, and stearate enhanced (p<0.05) iron uptake in cells grown in lower iron. The fatty acid effect on iron uptake by cells grown in normal iron was not as pronounced. Iron transport was not affected (p>0.05) by an interaction between the type of medium (iron concentration) and the type of fatty acid. Iron transport was enhanced (p<0.05) with palmatate and stearate. Various indicators of monolayer integrity and paracellular transport were also affected by the fatty acids, thus impacting iron uptake and transport. These results indicate that oleate, palmatate, and stearic can enhance iron uptake and transport; however, this enhancement may be the result of alterations in the integrity of the intestine. Sulfate transport processes and its regulation were studied in roots of poplar trees (Populus tremula x P. alba). From the exponential increase in sulfate uptake with temperature an activation energy (Ea) of 9.0±0.8 kJ mol–1 was calculated. In the concentration range 0.005–10 mM sulfate uptake showed biphasic Michaelis-Menten kinetics with a Km of 3.2±3.4 M and a Vmax of 49±11 nmol SO42– g–1 FW h–1 for the high-affinity uptake system (phase 1) and a Km of 1.33±0.41 mM and a Vmax of 255±25 nmol SO42– g–1 FW h–1 for the low-affinity system (phase 2). Xylem loading decreased linearly with temperature and remained unchanged within the sulfate concentration range studied. Regulation of sulfate uptake and xylem loading by O-acetyl serine (OAS), Cys, reduced glutathione (GSH), Met and S-methylmethionine (SMM) were tested by perfusion into the xylem sap with the pressure probe and by addition to the incubation medium. When added directly to the transport medium, Cys and GSH repressed, and OAS stimulated sulfate uptake; xylem loading was stimulated by Cys, repressed by GSH and only slightly affected by OAS. When perfused into the xylem, none of the compounds tested affected sulfate uptake of excised roots, but xylem loading was stimulated by SMM and OAS and repressed by Met. Apparently, the site of application strongly determined the effect of regulatory compounds of sulfate transport processes.  相似文献

The uptake of 2-ketogluconate is inducible in Pseudomonas putida: 2-ketogluconate, glucose, gluconate, glycerol and glycerate were each good nutritional inducers of this ability. 2-Ketogluconate uptake obeyed saturation kinetics (apparent K min 2-ketogluconate-grown cells was 0.4 mM). 2-Ketogluconate was transported against a concentration gradient, apparently in an unchanged state, and the process required metabolic energy, all of which indicate an active transport system.A number of independently isolated mutants with deranged activity of a common glucose-gluconate uptake system were found to be also defective in 2-ketogluconate transport. Strains unable to transport 2-ketogluconate which grew readily on glucose and gluconate were also isolated. These results suggest that 2-ketogluconate transport is governed by at least two genetic elements: one which is also required to take up glucose and gluconate and another which appears to be specific for 2-ketogluconate transport. Nitrate uptake in right-side out plasma membrane vesicles isolated from cucumber roots was characterized. Nitrate uptake into vesicles was driven by an artificially imposed pH gradient. The uptake was strongly inhibited by phenylglyoxal, an arginyl residue modificator. Only a slight repression of NO 3 transport in vesicles was observed in the presence of NEM, a thiol group reagent. pCMBS, an other thiol reagent and DEPC, an effector of histidine residue, had no effect on the nitrate transport in plasma membranes. ATP-driven proton transport in vesicles was not significantly affected in the presence of both, phenylglyoxal and DEPC, whereas pCMBS and NEM abolished it almost completely. The possible role of the particular amino acids residues in the active nitrate transport is discussed. NO 3 uptake into vesicles isolated from both, nitrate-induced and nitrate-depleted plant material was higher than that observed in the vesicles obtained from uninduced plants. Thus, isolated vesicles reflect the well-known in vivo response of intact plants on the exogenous nitrogen regime.  相似文献

Analyses of amino acid transport systems in JapaneseParamecium symbiont F36-ZK were performed using14C-amino acids. Kinetic analyses of amino acid uptake and competitive experiments revealed three transport systems; a basic amino acid transport system, which catalyzed transport of L-Arg and L-Lys, a general amino acid transport system, which had broad specificity for 19 amino acids (but not L-Arg), and an alanine transport system. These three systems were considered to be capable of active transport. Amino acid-proton symport was indicated by the following data: decreases in pH of the medium observed during L-Ser and L-Ala uptake, and uptake of L-Arg, L-Ser and L-Ala being inhibited by carbonyl cyanide m-chlorophenylhydrazone, sodium azide and vanadate. The optimal pH for uptake of neutral amino acids and L-Arg was around 5 and 5 to 6.5, respectively. Uptake of L-Asp and L-Glu was very sensitive to pH and little uptake of L-Asp was measured above pH 6.0. Amino acid uptake was not inhibited by nitrate or ammonium, and cultured cells with ammonium also possessed constitutive uptake systems.  相似文献
