Rice fields are important substitute wetlands for frogs. Traditionally, rice fields in Japan were supplied with water and drained via shallow earth ditches (old-style). In the last 30 years, however, more than 80% of rice fields have been converted to a new irrigation system in which water is typically supplied through underground pipes via taps and is drained into deep, concrete-sided ditches (new-style). We compared the occurrence of frogs in paired areas of old- and new-style rice fields at six locations in Ibaraki Pref., central Japan, from May to August 1995. The Japanese tree frog (Hyla japonica) did not differ in abundance between the two types of rice fields, but the Japanese brown frog (Rana japonica) and the Tokyo daruma pond frog (Rana porosa porosa) preferred the old-style rice fields. These findings suggest that the status ofRana species has been adversely affected by the conversion of rice fields to the new irrigation system. Modification of the deep, U-shaped concrete ditches and the water management regime during the spawning season is needed to safeguard these species.  相似文献

Tadpoles of some ranid species appear to possess chemical defenses against fish predators, but the chemicals have not been characterized. Here, we evaluated the vulnerability of three Japanese anuran tadpole species (Glandirana rugosa, Pelophylax nigromaculatus, and Hyla japonica) to a fish (Gnathopogon elongatus elongatus) and analyzed the defensive chemicals extracted from the unpalatable tadpoles. Additionally, we examined the defensive behavior of unpalatable tadpoles in response to fish chemical cues. The fish rejected both G. rugosa (83%) and P. nigromaculatus (48%), but not H. japonica (0%). Many of the rejected tadpoles survived (60–80%). Possible defensive chemicals were extracted by methanol from the skin of G. rugosa, but were not identifiable by gas chromatography–mass spectrometry because of small quantities. The chemicals have high polarity and non-volatility. When exposed to fish chemical cues, P. nigromaculatus decreased activity presumably as a defensive behavior, but G. rugosa did not. We demonstrated the presence of chemical defenses in at least two of these species and revealed that G. rugosa releases more effective or greater amounts of defense chemicals than P. nigromaculatus with respect to this fish predator. The increased efficacy of chemical defenses may correlate with decreasing defensive behavior.  相似文献

Molecular weights of the skeletal muscle myosin, actin, troponin C and tropomyosin were compared in two frog species, Hyla japonica and Xenopus tropicalis, by SDS-PAGE and Western blot. Polyclonal antibody was produced using H. japonica skeletal muscle as the antigen. Polyclonal antibodies to nematode (Caenorhabditis elegans), mold slime (Physarum polycephalum), crab (Pagurus japonicus) and chicken skeletal muscle were also used. In H. japonica, the molecular weights of skeletal myosin, actin, troponin C and tropomyosin were 230, 42, 19 and 38 kDa, respectively, by using anti-C. elegans paramyosin, anti-P. polycephalum actin, anti-crab troponin C and anti-chicken gizzard tropomyosin antibodies. Molecular weights of the same proteins in X. tropicalis detected by the same antibodies were 230, 43, 20 and 40 kDa, respectively. In total, 29 protein bands were detected in H. japonica skeletal muscle and 24 bands in X. tropicalis by SDS-PAGE. The results revealed interspecific differences in molecular weights of selected skeletal muscle proteins and in the total skeletal muscle protein profiles between the two frog species.  相似文献

Giant water bug Kirkaldyia (=Lethocerus) deyrolli (Belostomatidae: Heteroptera) nymphs consume many individuals of anuran larvae (tadpoles) before they reach adulthood. However, no quantitative data on the number of tadpoles consumed have been made available until now, despite the importance of this data for the conservation of this species. The number of tree frog, Hyla japonica (Hylidae: Anura), tadpoles consumed by nymphs of K. deyrolli was examined under laboratory conditions. The number increased according to the developmental stage of the nymphs; the fifth instar nymphs were the highest consumers. The total numbers consumed by the nymphs throughout development were 155.0 ± 12.8 (average ± SD) and 172.8 ± 9.6 for the males and the females, respectively. Thus, it might be important to maintain high densities of frogs in fields to conserve the population of K. deyrolli into the future.  相似文献

Microtubule arrays associated with simultaneous cytokinesis in the coenocytic microsporocytes of Lonicera japonica and Impatiens sultani were studied by indirect immunofluorescence. The future division planes are not predicted prior to meiosis by either a preprophase band of microtubules or cytoplasmic lobing. Cleavage planes appear to be determined by position of the four haploid nuclei and the development of postmeiotic microtubule systems. Perpendicular second division spindles in Lonicera result in tetrahedrally arranged tetrads while parallel spindles in Impatiens result in tetragonal arrangement. Immediately following meiosis bands of microtubules, the secondary spindles, develop between both sister and nonsister nuclei. These arrays give way to systems of microtubules that radiate equally from each of the four nuclei in the coenocytic sporocyte. Simultaneous cytokinesis is initiated by centripetal wall deposition at the periphery of the sporocyte and proceeds along planes marked by interaction of the opposing arrays of nuclear-based microtubules.  相似文献

Allozyme variation was examined in three diploid taxaChionographis japonica var.japonica, var.kurokamiana, andC. koidzumiana and three tetraploid taxaC. japonica var.kurohimensis, ssp.hisauchiana, and ssp.minoensis. Results show thatC. japonica var.kurokamiana is genetically closer toC. koidzumiana than to var.japonica. In the tetraploid taxa, fixed heterozygosities were found at several loci, and this supports the hypothesis that these taxa are allotetraploids. Furthermore, the tetraploid taxa have many unique alleles not found in the diploid taxa. This suggests that sufficient time has passed since the origin of tetraploids for new mutations to have been fixed.  相似文献

Some myrmecophilous animals show myrmecomorphy, however, its adaptive significance is still controversial. We investigated a possible benefit of Batesianmimicry between a myrmecophilous staphylinid beetle, Pella comes, and its host ant, Lasius (Dendrolasius) spathepus, by using a common ant predator, the Japanese treefrog, Hyla japonica. In the field, H. japonica were found to feed on numerous ants and other insects, but in laboratory experiments they refused feeding on L. spathepus. L. spathepus was highly repellent to these frogs, while P. comes was potentially palatable. After repeated contacts with L. spathepus which led to its avoidance the treefrogs started to reject P. comes as well . This suggests that myrmecomorphy is beneficial to P. comes, reducing the risk of predation, and that it , may represent a case of Batesian mimicry. may represent a case of Batesian mimicry. In adult amphibian skin, Na+ crosses from outside to inside. This Na+ transport can be measured as the amiloride-blockable short-circuit current (SCC) across the skin. We investigated the effects of arginine vasotocin (AVT) and mesotocin (MT), and those of antagonists of the vasopressin and oxytocin receptors, on the SCC across Hyla japonica skin. (1) Both AVT (100 pmol/L or more) and MT (1 nmol/L or more) increased the SCC. (2) The AVT- and MT-induced increases in SCC recovered with time (downregulation). (3) These AVT/MT-induced effects were blocked by application of OPC-31260 (vasopressin V2-receptor antagonist). (4) The OPC-31260 concentration needed to block the AVT-induced response was lower upon post-application (after application of agonist) than upon pre-application (before application of agonist), suggesting the number of receptors may have decreased after AVT application. (5) Upon repeated application of AVT (100 pmol/L), the induced SCC increase did not differ significantly between the 1st and 2nd applications. (6) The time to reach the half-maximum value of the AVT-induced or MT-induced increase in SCC was not significantly different between washout and post-application of OPC-31260, suggesting that post-application of OPC-31260 cleared AVT and MT from their receptors. The effects of AVT, MT, and their antagonists in H. japonica, which is adapted to a terrestrial habitat, are compared with our previously published data on Rana catesbeiana (=Lithobates catesbeianus), which is adapted to a semiaquatic habitat.  相似文献

Nuptial pads, secondary sexual characteristics of male frogs, develop on the first digit of the hand of Hyla japonica in the family Hylidae and of Rhacophorus schlegelii in the family Rhacophoridae, and on both the first and second digits of the rhacophorids Buergeria buergeri and B. japonica. By scanning electron microscopy (SEM), it was seen that numerous mounds covered the surface of the pads. Mounds were similarly hemispherical in R. schlegelii and B. buergeri and flat in B. japonica. The mounds of H. japonica were irregular in shape and size and some of them stood in rows. Fine columnar protuberances were present all over the surfaces of the mounds in this species. Numerous peg-like protuberances projected radially upward from the top of the mounds in B. buergeri and B. japonica. Irregular-shaped, leaf-like protuberances bearing knobby or rod-shaped apical protrusions were present on the top of the mounds in R. schlegelii. In pads observed by transmission electron microscopy, the outermost epithelial monolayer of the mounds was generally thick, especially at the top, compared to that of the rest of the skin. Epidermal cells in this layer were well keratinized, devoid of organelles, and contained closely packed, fine filaments within a dense matrix. Apical accessory protuberances projecting from the outermost cells were also packed with dense filamentous materials, showing rigid comb-like structures. © 1994 Wiley-Liss, Inc.  相似文献

The food habits of the endangered giant water bug, Lethocerus deyrolli, were studied in the rice fields of Nose, in the north of Osaka Prefecture, Japan. Field observations revealed that frogs were the most important prey item. Frogs represented 86.4% and 78.6% in the diet of L. deyrolli in spring and summer, respectively. Among seven species of three families (Hylidae, Rhacophoridae, and Ranidae) exploited by L. deyrolli, the most important food item was adult Hyla japonica in spring and juvenile Rana nigromaculta in summer. Fish and aquatic arthropods were not considered important foods for L. deyrolli. The frog‐dependent food habits indicate that the recovery and conservation of frogs should be prioritized to protect L. deyrolli from extinction.  相似文献

The regenerative capacity of limbs was investigated by amputation of limbs at the zeugopodium in postmetamorphic froglets and adults of various sizes in four species of Japanese frogs, all of which showed some regeneration at these ages. In Hyla arborea japonica and Rana brevipoda porosa most young froglets regenerated their limbs well; however, the rate of regeneration decreased with the age of amputation, and the limb became nonregenerative in adults. Limbs of adults in Rana rugosa and R. japonica, on the other hand, exhibited good regeneration. All of the regenerates in the four species were heteromorphic, consisting histologically of well-developed cartilaginous rods surronded by connective tissue and skin. Limited development of muscle was appartment in regenerates of the three ranid species. The relations between body size, innervation of limbs, and regenerative capacity are discussed.  相似文献

We have examined morphological and chromosomal variation inFallopia sect.Reynoutria in Korea to clarify their taxonomic identities and to determine whether their morphological variability is associated with ploidy levels. Principal components analysis (PCA) of individuals from 21 populations, using major distinguishing characters, revealed the presence of four major entiries of sect.Reynoutria in Korea; these includeF. sachalinensis, F. japonica var.japonica, F. forbesii, and the Nonsan population consisting of presumed hybrids. Based on morphology, it is hypothesized that the Nonsan population was probably derived from multiple hybridization events involving the three named taxa. The results also indicate thatF. forbesii is distinct fromF. japonica var.japonica. Polyploidy is more prevalent in sect.Reynoutria than has been previously recognized.Fallopia sachalinensis in Korea occurs as dodecaploids with 2n=132; our count is the first dodecaploid count for the species, and represents the highest chromosome number known in the genus.Fallopia japonica var.japonica occurs as tetraploids (2n=44), hexaploids (2n=66), and octoploids (2n=88), whileF. forbesii occurs as hexaploids (2n=66) and octoploids (2n=88); our counts appear to be the first reported chromosome numbers forF. forbesii. Morphological analysis indicates that there is no apparent correlation between the ploidy levels in these taxa and the morphological characters that we have considered in this study except that the tetraploids ofF. japonica var.japonica tend to have somewhat thicker leaves.  相似文献

Negative interactions between non-indigenous and native species has been an important research topic of invasion biology. However, interactions between two or more invasive species may be as important in understanding biological invasions, but they have rarely been studied. In this paper, we describe three field experiments that investigated interactions between two non-indigenous plant species invasive in the eastern United States, Lonicera japonica (a perennial vine) and Microstegium vimineum (an annual grass). A press removal experiment conducted within a deciduous forest understory community indicated that M. vimineum was a superior competitor to L. japonica. We tested the hypothesis that the competitive success of M. vimineum was because it overgrew, and reduced light available to, L. japonica, by conducting a separate light gradient experiment within the same community. Shade cloth that simulated the M. vimineum canopy reduced the performance of L. japonica. In a third complementary experiment, we added experimental support hosts to test the hypothesis that the competitive ability of L. japonica is limited by support hosts, onto which L. japonica climbs to access light. We found that the abundance of climbing branches increased with the number of support hosts. Results of this experiment indicate that these two invasive species compete asymmetrically for resources, particularly light.  相似文献

The endoparasitoid wasp Asobara japonica has highly poisonous venom: the host Drosophila larvae are killed by envenomation at a dose that is naturally injected by the female wasp at parasitism. This insecticidal venom is neutralized, however, because A. japonica introduces lateral oviduct components soon after venom injection at oviposition. Although the venom and lateral oviduct components of this parasitoid have been partially characterized, how the venom components favor successful development of wasp eggs and larvae in the host remains ambiguous. Here, we demonstrated that A. japonica venom did not affect host humoral immune responses, determined as expression of antimicrobial peptide (AMP) genes, but significantly diminished two cellular responses, spreading and phagocytosis, by host hemocytes. Moreover, venom components drastically elevated a serine protease‐like activity 4 h after its injection. The lateral oviduct components did not negate the detrimental effects of the venom on host cellular immunities, but significantly reduced the venom‐induced elevation of protease activity. Both active factors in venom and lateral oviduct components were roughly characterized as heat‐labile substances with a molecular mass of at least 10 kDa. Finally, venom of A. japonica, with a wide host range, was found to be much more toxic than that of Asobara rossica, which has a limited host range. These results reveal that A. japonica venom toxicity allows exploitation of a broader range of host insects because it is essential to overcome cellular immune responses of the host for successful parasitism.  相似文献

Telomeres generally consist of short repeats of minisatellite DNA sequences and are useful in chromosome identification and karyotype analysis. To date, telomeres have not been characterized in the economically important brown seaweed Saccharina japonica, thus its full cytogenetic research and genetic breeding potential has not been realized. Herein, the tentative sequence of telomeres in S. japonica was identified by PCR amplification with primers designed based on the Arabidopsis‐type telomere sequence (TTTAGGG)n, which was chosen out of three possible telomeric repeat DNA sequences typically present in plants and algae. After PCR optimization and cloning, sequence analysis of the amplified products from S. japonica genomic DNA showed that they were composed of repeat units, (TTTAGGG)n, in which the repeat number ranged from 15 to 63 (n = 46). This type of repeat sequence was verified by a Southern blot assay with the Arabidopsis‐type telomere sequence as a probe. The digestion of S. japonica genomic DNA with the exonuclease Bal31 illustrated that the target sequence corresponding to the Arabidopsis‐type telomere sequence was susceptible to Bal31 digestion, suggesting that the repeat sequence was likely located at the outermost ends of the kelp chromosomes. Fluorescence in situ hybridizations with the aforementioned probe provided the initial cytogenetic evidence that the hybridization signals were principally localized at both ends of S. japonica chromosomes. This study indicates that the telomeric repeat of the kelp chromosomes is (TTTAGGG)n which differs from the previously reported (TTAGGG)n sequence in Ectocarpus siliculosus through genome sequencing, thereby suggesting distinct telomeres in brown seaweeds.  相似文献

Calls produced by hybrids resulting from laboratory crosses of tetraploid Hyla versicolor females and either diploid Hyla chrysoscelis (type I) or Hyla arborea (type II) males were induced through manipulation of environmental conditions. Type I hybrids produced trilled calls similar in note repetition to H. versicolor, but more similar in dominant frequency to H. chrysoscelis. Mean duration was shorter than in both parent calls. Type II hybrids produced calls which were longer in duration and lower in note repetition rate than H. versicolor, but shorter in duration and higher in note repetition rate than H. arborea. Dominant frequency of type II hybrids was lower than in H. arborea but not significantly different than in H. versicolor. Hybrid calls were not strictly intermediate, and may provide information regarding parental relationships.  相似文献

Invasion of alien species has been shown to cause detrimental effects on habitats of native species. Insect pollinators represent such examples; the introduction of commercial bumble bee species for crop pollination has resulted in competition for an ecological niche with native species, genetic disturbance caused by mating with native species, and pathogen spillover to native species. The European honey bee, Apis mellifera, was first introduced into Japan for apiculture in 1877, and queen bees have been imported from several countries for many years. However, its effects on Japanese native honey bee, Apis cerana japonica, have never been addressed. We thus conducted the survey of honey bee viruses and Acarapis mites using both A. mellifera and A. c. japonica colonies to examine their infestation in native and non-native honey bee species in Japan. Honey bee viruses, Deformed wing virus (DWV), Black queen cell virus (BQCV), Israeli acute paralysis virus (IAPV), and Sacbrood virus (SBV), were found in both A. mellifera and A. c. japonica colonies; however, the infection frequency of viruses in A. c. japonica was lower than that in A. mellifera colonies. Based on the phylogenies of DWV, BQCV, and SBV isolates from A. mellifera and A. c. japonica, DWV and BQCV may infect both honey bee species; meanwhile, SBV has a clear species barrier. For the first time in Japan, tracheal mite (Acarapis woodi) was specifically found in the dead honey bees from collapsing A. c. japonica colonies. This paper thus provides further evidence that tracheal-mite-infested honey bee colonies can die during cool winters with no other disease present. These results demonstrate the infestation of native honey bees by parasite and pathogens of non-native honey bees that are traded globally.  相似文献
