To determine whether or not the changes in the orientation ofmicrotubules (MTs) that are induced by GA3 and ABA result fromchanges in the rate of epicotyl elongation caused by these hormones,we examined the effects of GA3 and ABA on the orientation ofMTs in epidermal cells of decapitated epicotyls of the dwarfpea (Pisum sativum cv. Little Marvel), in which neither GA3nor ABA causes changes in the rate of epicotyl elongation. Cuttings taken from GA3-pretreated seedlings were decapitatedand treated with ABA. ABA eliminated the GA3-induced predominanceof transverse MTs and treatment with ABA resulted in a predominanceof longitudinal MTs in the decapitated cuttings. However, ABAdid not reduce the rate of epicotyl elongation in these samples.Cuttings taken from ABA-pretreated seedlings were decapitatedand treated with GA3. GA3 caused the orientation of MTs to changefrom longitudinal to transverse in the decapitated cuttings.However, GA3 had no promotive effect on elongation of theseepicotyls. The results indicate that both ABA and GA3 have the abilityto change the orientation of MTs by mechanisms that do not involvechanges in the rate of cell elongation. The sensitivity of light-grown cucumber hypocotyl sections toIAA and GA3 depends on the degree of aging of the tissue. Agreater response to GA3 was obtained with young tissue, whilethat to IAA was obtained with relatively old tissue. The responseto IAA reached a maximum at about 15 hr of incubation; the youngerthe tissue the earlier the time of maximum response. The responseto GA3 continued for more than 70 hr with a constant growthrate. Very young tissue started to respond to GA3 without lagtime; the older the tissue the later the start of the response. Sucrose (2%) inhibited IAA-induced elongation, while there wasa distinct synergism between GA3 and sucrose. The promotiveeffect of sucrose on GA3-induced elongation was also obtainedwhen sections were pretreated with sucrose, then transferredto GA3. Summary The effects of abscisic acid (ABA) on the orientation and cold stability of cortical microtubules (MTs) in epidermal cells of epicotyls of the dwarf pea,Pisum sativum L. cv. Little Marvel, were examined by immunofluorescence microscopy. The effect of ABA on the elongation of epicotyls and on the orientation of cortical MTs was opposite to that of gibberellin A3 (GA3). Treatment with ABA, which reduced the promotion of epicotyl elongation by GA3, eliminated the GA3-induced predominance of transverse MTs and resulted in a predominance of longitudinal MTs. The effect of ABA on the cold stability of cortical MTs was also opposite to that of GA3. ABA increased the cold stability of MTs, while GA3 decreased it. The predominance of longitudinal MTs brought about by ABA may have some relationship to ABA-induced inhibition of the elongation of the epicotyl. A synthetic brassinosteroid, 22,23(S,S)-homobrassinolide (hBR),was examined for its interaction with IAA and GA3 in the elongationof hypocotyl sections of light-grown cucumber (Cucumis salivusL. cv. Aonagajibai) seedlings. hBR alone was less active thanIAA. Its optimal concentration was around 10 µM and thelowest effective concentration between 10 and 100 µM,which is more than 100 times higher than that of brassinolide.hBR was more active in sections from younger seedlings. Itsgrowth-promoting effect was negated or greatly reduced by inhibitorsof auxin-induced elongation such as p-chlorophenoxyisobutyricacid and kinetin. hBR acted synergistically with IAA and 2,4-Dbut not with GA3 showing only an additive effect. Sequentialtreatment of sections with hBR and then with IAA also resultedin synergistic enhancement of auxininduced elongation, but whenthe order of treatment was reversed, hBR was inactive. The synergisticeffect was obtained with 1 h pretreatment with hBR and couldbe reduced by subsequent washing with water. There was no sequentialinteraction between hBR and GA3. The synergistic pretreatmenteffects of hBR and GA3 were simply additive to each other. Amembrane-bound ATPase inhibitor, dicyclohexylcarbodiimide, inhibitedthe hBR-induced elongation, but did not affect GA3-induced elongation.The findings led to the conclusion that brassinosteroids enhanceauxin action and possess growth-promoting activity which isindependent of that of gibberellin. IAA-induced growth of light-grown cucumber hypocotyl sectionsis markedly enhanced by GA3-pretreatment of the sections; thereis a distinct synergism between IAA and GA3. Water pretreatmentalso enhances IAA-induced growth. On the other hand, IAA-pretreatedsections showed practically no further growth in response topost treatment with GA3. The enhancing effect of GA3 is obtainedwith only 30 min pretreatment, the maximum effect occuring with2 hr pretreatment. Pretreatment longer than 8 hr is less effective.This enhancing effect of GA3 can be observed soon after posttreatment with IAA. The response of GA3-pretreated sectionsto IAA is greater in pretreatment with higher concentrationsof GA3, and higher degrees of synergism between IAA and GA3are obtained at IAA concentrations less than 10-4 M. This synergisticinteraction between GA3 and IAA is more marked in aged hypocotylsections than in young sections. From these results we concludedthat gibberellin sensitizes hypocotyl cells to the subsequenteffect of auxin on cell elongation. (Received October 6, 1973; )  相似文献

In azuki bean (Azukia angularis = Vigna angularis) epicotylsections, gibberellin A3 (GA3) enhanced the growth promotingeffect of indoleacetic acid (IAA), but showed no growth effectwhen applied alone. Sections showed practically no cell division.The promoting effect of GA3 on section growth seems to be dueto its promoting effect on cell elongation. The diameters of sections treated with IAA increased, but thediameters of sections treated with GA3 together with IAA remainedconstant. GA3 seems to suppress cell expansion in a directiontransverse to the cell axis. Colchicine at a concentration with no inhibiting effect on IAA-inducedelongation almost completely reversed the effect of GA3 On the basis of these results, the participation of wall microtubulesin GA3-induced elongation is discussed. Phaseolus multiflorus plants at three stages of developmentwere decapitated either immediately below the apical bud orlower down at a point 1 cm above the insertion of the primaryleaves. Growth regulators in lanolin were applied to the cutstem surface. IAA always inhibited axillary bud elongation anddry-matter accumulation, and enhanced internode dry weight butnot elongation. GA3 applied below the apical bud greatly increasedinternode elongation and dry weight, but simultaneously reducedbud elongation and dry-weight increase. Application of GA3 1cm above the buds had no effect on bud elongation in the youngestplants, but enhanced their elongation in the two older groups.IAA always antagonized GA3-enhancement of internode extensiongrowth, whereas its effects on GA3-enhanced dry-matter accumulationdepended on the stage of internode development. Bud elongationwas greater in plants treated with GA3+IAA than in plants treatedonly with IAA, except in the youngest plants decapitated immediatelybelow the apical bud, where GA3 caused a slight increase inIAA-induced bud inhibition. GA3 increased inhibition of buddry weight by IAA in the two youngest groups of plants, butslightly reduced it in the oldest plants. No simple compensatorygrowth relationship existed between internode and buds. It wasconcluded that, (1) auxin appears to be the principal growthhormone concerned in correlative inhibition, and (2) availabilityof gibberellin to internode and buds is of importance as a modifyingfactor in auxin-regulated apical dominance by virtue of itslocal effects on growth in the internode and in the buds.  相似文献

Effects of GA3 on the stability of cortical microtubules (MTs)were studied in mesocotyl cells of etiolated maize seedlings.Propyzamide, an MT-disrupting agent specific for plant tubulin,disrupted cortical MTs in cells in the upper regions of mesocotylsof GA3-untreated seedlings and caused swelling of the cells.GA3 prevented propyzamide from disrupting MTs and from causingsuch swelling. Chilling of mesocotyls at 4°C for 60 minbrought about the disruption of cortical MTs in cells in theupper regions of mesocotyls of GA3-untreated seedlings, butnot in cells in corresponding regions of GA3-treated seedlings,suggesting that treatment with GA3 increased the stability ofcortical MTs in maize mesocotyl cells. Cortical MTs in protoplastsisolated from mesocotyls of GA3-untreated seedlings failed towithstand chilling at 0°C for 90 min, while those in protoplastsisolated from mesocotyls of GA3-treated seedlings withstoodchilling successfully. The interaction of kinetin with IAA and GA3 on the elongationof hypocotyl sections of Cucumis sativus L. cv. National Picklingwas studied. Kinetin in the concentration range of 10–7M to 10–4 M markedly inhibited IAA-induced elongation,while in a lower range from 10–10 M to 10–8 M, itsynergistically enhanced IAA-induced elongation. Kinetin alonein this range had no effect. A 5-to 15-min pulse treatment seemsenough to induce the maximum effect for both inhibition andpromotion. Since the magnitude of the maximum inhibition dependedon the concentration and not on the duration of treatment, thereaction in the cell caused by kinetin seemed to be completedwithin a short period. Washing of the sections with distilledwater after kinetin treatment (30 min) did not significantlyeliminate the kinetin effect. This probably indicates that thebinding of kinetin molecules to a supposed acceptor is not reversible.Interaction of kinetin with GA3 in their pretreatment effectson IAA-induced elongation shows that in the inhibitory concentrationrange, the kinetin effect was partly overcome by GA3, and thatin the promotive range, the magnitude of the enhancement wasdetermined by kinetin regardless of the presence of GA3. Rishitin, a norsesquiterpene alcohol, found in infected, resistantpotato-tuber tissue completely inhibited zoospore germinationand germtube elongation of Phytophthora infestans (MONT.) DEBARY at 10–3M. There was little difference in sensitivityto rishitin among races of Phytophthora infestans. IAA-inducedelongation of Avcna coleoptile sections and GA3-induced elongationof wheat leaf sections were also inhibited by rishitin. Theinhibition of IAA-induced elongation of Avena coleoptiles wasrelieved to some extent by increasing IAA concentration. However,little relief of the inhibition of GA3-induced elongation ofwheat leaf sections was obtained by increasing GA3 concentration.No plant injury was observed at this concentration of rishitin(10–3M). Examination of a series of rishitin derivatives indicated thatthe hydroxyl group at C-3 is indispensable for antifungal activity.This activity was intensified by saturating the double bondbetween the rings of rishitin and/or that of the isopropenylgroup at C-7, though activity decreased when oxygenated functionalgroups were introduced into the side chain. Aromatization of the A ring did not lower biological activities.The antifungal activities of most rishitin derivatives almostparalleled their activities as plant growth retardants. Segments excised from the upper and the lower parts of cowpea(Vigna unguiculata L.) hypocotyls were compared in terms oftheir responses to exogenous indole-3-acetic acid (IAA) in relationto their endogenous levels of gibberellin. Growth of the segmentswas measured continuously during xylem perfusion with a lineardifferential transformer. IAA induced a burst of elongationin the upper segments but only slight promotion of growth inthe lower segments. Treatment with uniconazole, a potent inhibitorof the biosynthesis of gibberellins, reduced the responsivenessof the upper segments to exogenous IAA to about one half ofthe control value. Pre-perfusion with GA3 of such segments fortwo hours prior to application of IAA, partially restored theresponsiveness to IAA. Analysis by GC/MS identified GA1, GA4,GA9 GA20 and GA51 as native gibberellins in the hypocotyls ofcowpea seedlings. Analysis by GC/SIM also showed that the physiologicallyactive gibberellins (GA1 and GA4) were located mainly in theupper part of the hypocotyl and the treatment with uniconazolemarketly reduced the endogenous level of gibberellins thereto less than 11% of the control level. Longitudinal microtubules are predominant in epidermal cellsof the 3rd internodes of dwarf pea (Pisum sativum L. cv. LittleMarvel) seedlings. In more than 50% of the cells, cortical microtubulesare running parallel to the cell axis. GA3 promotes elongation of the internodes and gives rise toa predominance of transverse microtubules. In more than 60%of the GA3-treatd cells, cortical microtubules are running transverseto the cell axis. Longitudinal microtubules in the GA3-untreated cells are resistantto low-temperature treatment, but transverse microtubules inthe GA3-treated cells are sensitive to it. Longitudinal microtubulesare present in GA3-treated epidermal cells with low frequency.They are resistant to low-temperature treatment. Longitudinal, oblique and transverse microtubules are presentwith almost the same frequency in epidermal cells of the 3rdinternodes of tall pea (cv. Early Alaska) seedlings. GA3 promoteselongation of the internodes also in tall pea seedlings, butit does not alter the direction of cortical microtubules sodistinctly as it does in dwarf pea seedlings. As in dwarf pea seedlings, longitudinal microtubules are resistantto low-temperature treatment, and transverse microtubules aresensitive to it in tall pea seedlings. Auxin activates pumping of protons from the symplast to theapoplast and causes hyperpolarization of the symplast membranein the elongation zone of Vigna stems prior to the accelerationof growth. This auxin-induced hyperpolarization has been studiedin most cases in hypocotyl segments excised from the elongationzone. In the present study, mature-zone segments were perfusedwith IAA by the xylem perfusion technique in an effort to determinewhether or not IAA has any effects in the mature zone. Althoughno hyperpolarization of the symplast membrane was observed uponthe perfusion with auxin alone, auxin-induced hyperpolarizationwas observed when mature-zone segments had been pretreated withGA3, in the absence of an increase in the growth rate. Theseresults suggest that cells in the mature zone have lost theability to activate the proton-pumping machinery in responseto auxin but that this ability can be restored by treatmentwith GA3. This effect of GA3 suggests the possibility that theconcentration of gibberellin in a tissue controls one of thecell's responses to auxin, namely, activation of the protonpump. The initial events in tuberization were examined in single-nodestem segments of potato, in which the tuberization was easilyregulated in culture. The addition of 8% sucrose to the culturemedium caused the cessation of elongation of lateral shootsand the swelling of the sub-apical region of each shoot. Swellingwas first induced by lateral cell expansion, which was followedby periclinal cell division. The divided cells then expandedlaterally. The alteration in the direction of growth was accompaniedby the reorientation of arrays of cortical microtubules (MTs),which was monitored by immunofluorescence microscopy. Cellsin the sub-apical region of elongating shoots had prominenttransverse arrays of MTs. The MTs in swelling cells were orientedlongitudinally with respect to the axis of the shoot. Finally,the arrays of MTs became completely disorganized. By contrast,the elongation of lateral shoots continued in GA3-treated segmentsand the cells in the sub-apical region of such shoots retainedconspicuous transverse arrays of MTs during culture, even inthe presence of a high concentration (8%) of sucrose. Darkness and GA3 stimulate the elongation of the first internodebut inhibit the production of roots while IAA inhibits internodalelongation but promotes the production of roots on epiphyllousbuds of Bryophylhum tubiflorum. Cycloheximide inhibits both,implicating the synthesis of proteins in the growth of bothroot and shoot. Even pre-treatment of buds with cycloheximidefor 4 h inhibits rooting as well as internodal elongation whenthese are subsequently transferred to IAA or GA3. On the otherhand, pre-treatment with IAA or GA3 even for 8 h does not alleviatethe inhibitory effect of cycloheximide suggesting that thereis a lag period between the application of these regulatorsand the synthesis of proteins caused by them.  相似文献

Fresh and dry weights and leaf size of Poa pratensis were reducedwhen treated with 6-azauracil (AzU), (2-chloroethyl)phosphonicacid (CEPA), or (2-chloroethyl)trimethylammonium chloride (CCC).AzU and CEPA inhibited epidermal cell division without inhibitingcell elongation, while CCC inhibited mainly cell elongationand cell division to a small extent. The ratio of blade lengthto sheath length and the blade length/width ratio were reduced,but leaf emergence and tillering were increased by AzU and CEPA.CCC affected only the latter three features. Like GA3, CEPAinduced stem formation, but internodes were shorter. GA3 was ineffective in preventing leaf-growth inhibition byAzU, which inhibited Ga3-induced cell elongation. The inhibitoryeffect of CEPA on leaf growth was apparently reversed by GA3,but this was due solely to increased cell elongation, the reductionin cell number being unaffected. Ga3 reversed the effect ofCCC on leaf length, as well as on cell size and number. Simultaneousapplication of the inhibitors produced a complex interactionin reducing leaf length and number and size of epidermis cells.It is postulated that AzU, CEPA, and CCC have different modesof action because they have specific effects on plant growthand different effects on GA3-induced cell elongation.  相似文献

The relationship between the dwarf habit of growth and corticalmicrotubule (MT) orientation as affected by gibberellin wasexamined using a gibberellin responding dwarf mutant of Zeamays L. (d5). The 4 mm portion of the mesocotyl below the coleoptilarnode of dark-grown seedlings was divided into four 1 mm segments.MTs were observed by means of immunofluorescence microscopyon the tangential surface of the epidermal cells (EP-t), theradial surfaces of epidermal (EP-r) and cortex cells (C-r) inboth normal and d5 seedlings. MT arrangement in EP-t was transverse,oblique and/or longitudinal. In lower regions, cells with transverselyoriented MTs decreased, while those with obliquely/longitudinallyoriented MTs increased. The frequency of the occurrence of transverselyoriented MTs was much higher in normal than in d5. In EP-r almostall the cells of the four regions had transversely orientedMTs. In d5 seedlings only a few percent of cells had obliquelyoriented MTs. In C-r all the cells of the four regions of normalseedlings had transversely oriented MTs, while in d5, 20% ofthe uppermost region cells showed oblique/longitudinal orientationand more in lower regions. The kinetics of extension induced by GA31 in the hypocotyl ofintact seedlings of Lactuca sativa are similar in the dark andin the light, and differs fundamentally from the kinetics ofelongation in the dark without GA3. Both in continuous lightand in the dark, GA3-induced promotion starts 24 h after incubation.In the dark, even low concentrations of GA3, which do not affectthe length measured after 6 d when the extension of hypocotylalmost ceases, remove the lag period of 48 h which precedesextension, and prolong the high rate of elongation. FollowingGA3 supply the hypocotyl length in the dark and in the lightdoes not differ until 48 h; thereafter the rate of elongationin the light is less, so that the final length of the hypocotylis 40 per cent shorter than that of the dark-grown seedlingswithout GA3. IAA supplied apically to light-grown seedlings induces a weakpromotion at a concentration of 1 mg l–1 only. With anincreasing concentration of GA3 supplied simultaneously, theconcentration of IAA inducing a significant promotion decreases.A combined supply of both these regulators, however, does notrestore the light-mediated inhibition of hypocotyl elongationcompletely. The maximum decrease in hypocotyl length induced by the growthretardants AMO-1618, CCC, and B-9 supplied from the beginningin the dark does not exceed 70 per cent. Saturating doses ofGA3 supplied in combination with any one of the retardants compensateonly a fraction of the decrease. The interaction of light, gibberellic acid (GA3), and phlorizinin the growth of lettuce (Lactuca sativa L. cv. ‘GrandRapids’) hypocotyls was investigated. At all concentrationsof GA3, phlorizin enhanced GA3-induced growth at luminous intensitiesabove 50 ft-c (continuous light). Without GA3, phlorizin hadno effect on hypocotyl growth in the light but it inhibitedgrowth in the dark. Both seedlings and hypocotyl sections respondedto phlorizin in the presence of GA3. There was no iteractionbetween phlorizin and KCl. Water-growth was severly inhibitedby light. GA3,-induced growth was slightly inhibited by light,and then only at luminous intensities above 50 ft-c. Thus, relativeto H2O-growth, GA3-induced growth increased with increasingluminous intensity up to 450 ft-c, where it reached saturation.It seems that a synergism may exist between light and GA3 aswell as between phlorizin and GA3.
