以湿地松×洪都拉斯加勒比松(Pinus elliottii×P.caribaea var.hondurensis)及亲本为实验材料,采用甲基化敏感扩增多态性技术对其基因组中CCGG位点的甲基化相对水平及遗传变异模式进行了初步分析。结果表明,杂种及亲本CCGG总甲基化相对水平介于77.74%~81.75%,CG甲基化相对水平略低于CNG甲基化水平,CG/CNG甲基化相对水平高于亲本。杂种遗传自亲本的CG与CNG甲基化位点数之比接近1:1,遗传自母本的甲基化位点数与遗传自父本的CCGG甲基化位点数比例为1:1;杂种产生的全新甲基化与完全去甲基化位点数之比接近7:1,初步推测大量甲基化变异促进了杂合体的生长发育。  相似文献

Genetic maps for individual Pinus elliottii var. elliottii and P. caribaea var. hondurensis trees were generated using a pseudo-testcross mapping strategy. A total of 329 amplified fragment length polymorphic (AFLP) and 12 microsatellite markers were found to segregate in a sample of 93 interspecfic F(1) progeny. The male P. caribaea var. hondurensis parent was more heterozygous than the female P. elliottii var. elliottii parent with 19% more markers segregating on the male side. Framework maps were constructed using a LOD 5 threshold for grouping and interval support threshold of LOD 2. The framework map length for the P. elliottii var. elliottii megagametophyte parent (1,170 cM Kosambi; 23 linkage groups) was notably smaller than the P. caribaea var. hondurensis pollen parent (1,658 cM Kosambi; 27 linkage groups). The difference in map lengths was assumed to be due to sex-related recombination variation, which has been previously reported for pines, as the difference in map lengths not be accounted for by the larger number of markers mapping to the P. caribaea var. hondurensis parent - 109 compared with 78 in P. elliottii var. elliottii parent. Based on estimated genome sizes for these species, the framework maps for P. elliottii var. elliottii and P. caribaea var. hondurensis covered 82% and 88% of their respective genomes. The pseudo-testcross strategy was extended to include AFLP and microsatellite markers in an intercross configuration. These comprehensive maps provided further genome coverage, 1,548 and 1,828 cM Kosambi for P. elliottii var. elliottii and P. caribaea var. hondurensis, respectively, and enabled homologous linkage groups to be identified in the two parental maps. Homologous linkage groups were identified for 11 out of 24 P. elliottii var. elliottii and 10 out of 25 P. caribaea var. hondurensis groups. A higher than expected level of segregation distortion was found for both AFLP and microsatellite markers. An explanation for this segregation distortion was not clear, but it may be at least in part due to genetic mechanisms for species isolation in this wide cross.  相似文献

The rotifer Brachionus plicatilis can grow in a wide range of salinities and temperatures, but rapid shifts in both salinity and temperature may result in immobilized, non-swimming rotifers. The goal of this study was to examine the effect of perturbations in temperature and salinity on the swimming pattern of the rotifer.Only slight changes in mobility were observed when rotifers were exposed to changes in temperature (from 20 °C to 8–30 °C) and to an increase in salinity (from 20% to 30%). When the salinity was reduced to 15% and 5%, the proportion of mobile rotifers was reduced to 50% and 5%, respectively. The rotifers were throughout more resistant to perturbations in temperature than to those of salinity.Combined temperature and salinity perturbations compared to perturbations in each factor separately suggested a synergetic effect of temperature and salinity on the rotifers locomotion. Transfer from cultivation conditions to low salinity (5%) and high temperature (28 °C) resulted in very low percent of mobile rotifers (0–10%). However, if the temperature was reduced to 8 °C concomitant with the changes in salinity, the percent of mobile rotifers was 85%.Rotifers use a high share of their metabolic energy for locomotion, and it is therefore not surprising that perturbations in salinity and temperature may result in partial or complete immobilization.  相似文献

Streptococcus faecium var. casseliflavus is a gram-positive, spherical cell. The cells occur chiefly as pairs within chains and elongate to ogive-shaped cells during growth. Growth is good on 5% bile salts-agar and in broth at 10 C, and in broth adjusted to pH 9.6 or containing 6.5% NaCl, but many strains fail to grow at 45 C. Litmus is reduced rapidly prior to formation of an acid curd. Few strains release ammonia from arginine or serine. The organism is not proteolytic and does not produce H(2)S or acetylmethylcarbinol, reduce nitrate, decarboxylate tyrosine, or produce slime on sucrose-agar. Most strains survive heating to 60 C for 30 min. It produces gray colonies on potassium tellurite agar, reduces 2,3,5-triphenyltetrazolium-HCl to a pink color, and ferments cellobiose, dextrin, maltose, mannose, and sorbitol, thus resembling S. faecalis. Like S. faecium, it produces peroxidase but not catalase on heated blood media, dissimilates malate, and ferments arabinose, melibiose, and salicin, but not melezitose. Like both species, it ferments dextrose, galactose, lactose, mannitol, sucrose, trehalose, and citrate. Properties peculiar to the variant include the high pH limiting initiation and termination of growth; the fermentation of alpha-methyl-d-glucoside, raffinose, and xylose; motility; and growth without blue button formation in ethyl violet broth. The water-soluble, pale lemon-yellow pigment is released into the aqueous phase only after the cell envelope is altered by fat solvents. The bacterium thrives as an epiphyte on plants.  相似文献

The essential oil of the leaves of P. racemosa var. terebinthina and P. racemosa var. grisea were examined by GC and GC/MS. The major constituents were alpha-terpineol acetate (27%), alpha-terpineol (20%) and 4-methoxy eugenol (12.6%) for Pracemosa var. terebinthina and 4-methoxy-isoeugenol (75.2%) and 4-methoxy-eugenol (4.5%) for P. racemosa var. grisea.  相似文献

Zusammenfassung Rotalia beccarii var. flevensis nov. var. zeigt eine typische Trimorphie (Formen a1, A2 und B). Diese Trimorphie ist der Ausdruck eines Generationswechsels, der mit den Jahreszeiten gleichen Tritt hält. Die B-Form ist die Dauerform im Winter und bildet im Frühjahr durch asexuelle Vermehrung Plasmodiosporen, welche zur A1-Form heranwachsen. Die A1-Form bildet ihrerseits im Sommer die A2-Form, welche gekennzeichnet ist durch den Besitz eines Chromidiums. In dieser Form entwickeln sich die Mikrosporen durch mitotische Teilungen der aus dem Chromidium entstandenen kleinen Kerne, nachdem die Individuen sich enzystiert haben. Die Kopulation der Mikrosporen liefert im Herbst wieder die mikrosphärische B-Form.Ich habe eine ausführliche Studie publiziert über die Fortpflanzung der Foraminiferen in: Siboga-Reports 4, Part II, S. 79–104. Daselbst ist diese Rotalia noch als Pulvinulina repanda angeführt. The ability of phytopathogenic fungi to overcome the chemical defense barriers of their host plants is of great importance for fungal pathogenicity. We studied the role of cyclic hydroxamic acids and their related benzoxazolinones in plant interactions with pathogenic fungi. We identified species-dependent differences in the abilities of Gaeumannomyces graminis var. tritici, Gaeumannomyces graminis var. graminis, Gaeumannomyces graminis var. avenae, and Fusarium culmorum to detoxify these allelochemicals of gramineous plants. The G. graminis var. graminis isolate degraded benzoxazolin-2(3H)-one (BOA) and 6-methoxy-benzoxazolin-2(3H)-one (MBOA) more efficiently than did G. graminis var. tritici and G. graminis var. avenae. F. culmorum degraded BOA but not MBOA. N-(2-Hydroxyphenyl)-malonamic acid and N-(2-hydroxy-4-methoxyphenyl)-malonamic acid were the primary G. graminis var. graminis and G. graminis var. tritici metabolites of BOA and MBOA, respectively, as well as of the related cyclic hydroxamic acids. 2-Amino-3H-phenoxazin-3-one was identified as an additional G. graminis var. tritici metabolite of BOA. No metabolite accumulation was detected for G. graminis var. avenae and F. culmorum by high-pressure liquid chromatography. The mycelial growth of the pathogenic fungi was inhibited more by BOA and MBOA than by their related fungal metabolites. The tolerance of Gaeumannomyces spp. for benzoxazolinone compounds is correlated with their detoxification ability. The ability of Gaeumannomyces isolates to cause root rot symptoms in wheat (cultivars Rektor and Astron) parallels their potential to degrade wheat allelochemicals to nontoxic compounds.  相似文献

Summary Protoplasts isolated from seedling cotyledons of yellow passionfruit (Passiflora edulis var. flavicarpa Deg.) and two related wild species, P. amethystina Mikan. and P. cincinnata Mast., divided in culture and produced calli. Shoot regeneration was obtained in MS medium (Murashige and Skoog 1962) containing 2.0 mg/l 6-benzylaminopurine (BAP). Regenerated plants produced roots in half-strength hormone-free MS medium and could be transferred to soil after being acclimatized.  相似文献

Among the genus Populus, the sections Populus (white poplar), Aigeiros Duby (black poplar) and Tacamahaca Spach contain many tree species of economical and ecological important properties. Two parental maps for the inter-specific hybrid population of Populus adenopoda × P. alba (two species of Populus section) were constructed based on SSR and SRAP markers by means of a two-way pseudo-test cross mapping strategy. The same set of SSR markers developed from the P. trichocarpa (belonging to Tacamahaca section) genome which were used to construct the maps of P. deltoides and P. euramericana (two species of Aigeiros section) was chosen to analyze the genotype of the experimental population of P. adenopoda × P. alba. Using the mapped SSR markers as allelic bridges, the alignment of the white and black poplar maps to each other and to the P. trichocarpa physical map was conducted. The alignment showed high degree of marker synteny and colinearity and the closer relationship between Aigeiros and Tacamahaca sections than that of Populus and Tacamahaca. Moreover, there was evidence for the chromosomal duplication and inter-chromosomal reorganization involving some poplar linkage groups, suggesting a complicated course of fission or fusion in one of the lineages. A poplar consensus map based on the comparisons could be constructed will be useful in practical applications including marker assisted selection.  相似文献

The diseases Phytophthor a crown and root rot consist of the most important problems in cherry cultivation. In this study, the susceptibility of 30 cherry genotypes to Phytophthora cactorum , P. citrophthora, P. parasitica and P. citricola was evaluated by using excised twig assay, excised shoot method and stem inoculation method. The results showed that all cherry genotypes tested were susceptible to all Phytophthora isolates used. Phytophthora parasitica and P. citrophthora were the most aggressive species. Host specificity of the Phytophthora isolates used in this study was not found although these isolates were from different plant species. In conclusion, because none of the cherry genotype showed a level of resistance to these pathogens, caution should be taken when these genotypes are used in locations, where these diseases are endemic.  相似文献

Cellular fatty acid analysis was employed to differentiate three varieties of Rhizoctonia circinata ; var. circinata , var. oryzae and var. zeae . Eight fatty acids including myristic (14 : 0), pentadecanoic (15 : 0), palmitic (16 : 0), palmitoleic (16 : 1  cis 9), stearic (18 : 0), oleic (18 : 1  cis 9), linoleic (18 : 2  cis 9,12) and linolenic (18 : 3  cis 9,12) acids were present in isolates of all three varieties of R. circinata . Heptadecanoic acid (17 : 0) was detected in isolates of R. circinata var. zeae but not in isolates of R. circinata var. circinata or R. circinata var. oryzae . Palmitic, oleic and linoleic acids were the major fatty acids found, comprising 94–98% of the whole-cell fatty acid content. The remaining fatty acids were present in small amounts. Based on the composition (%) of fatty acids, isolates of R. circinata var. circinata , R. circinata var. oryzae and R. circinata var. zeae were clearly differentiated into three groups as shown by principal component and cluster analyses. This finding agrees well with the grouping of R. circinata into three varieties based on differences in colony morphology of the vegetative state. In principal component and cluster analysis, isolates of R. circinata var. circinata from Japan and Alaska were indistinguishable.  相似文献

In the present work, three varieties of Teucrium orientale, var. orientale, var. puberulens, and var. glabrescens, were collected and investigated for chemical composition of the oils. Subsequent gas chromatography (GC-FID) and gas chromatography coupled to mass spectrometry (GC/MS) revealed high abundance of sesquiterpenes in the essential oils analyzed. All the oils contained β-caryophyllene (22.6, 8.5, and 6.3%, resp.) and hexadecanoic acid (7.9, 12.8, and 13.1%). Germacrene D (24.6 and 33.4%) and bicyclogermacrene (6.7 and 8.5%) were found to be the main constituents of var. orientale and var. puberulens, respectively. The high percentages of β-cubebene (26.9%), α-cubebene (9.0%), and α-copaene (7.2%) established the diversity of var. glabrescens. The qualitative difference between the essential oils allowed the differentiation between the varieties in agreement with the morphological observations described in Flora of Turkey for each variety studied. In addition, a cluster analysis of twelve Teucrium taxa based on the essential-oil composition has been carried out. Hovewer, the analysis did not clearly reflect the infrageneric classification of the genus, it largely confirmed the relationships between the infraspecific taxa of Teucrium orientale and T. chamaedrys.  相似文献
