  • 1.1. Superoxide was generated during the auto-oxidation of the antituberculous drug, isonicotinic acid hydrazide (INH), but not with its meta-isomer, nicotinic acid hydrazide (NH). During Fe2+-stimulated oxidation of INH and NH, aromatic hydroxylation occurred which was inhibited by the chelating agent, phytic acid.
  • 2.2. A mixture of myeloperoxidase (MPO) and a hydrazide induced formation of compound III (oxyperoxidase) and aromatic hydroxylation which was stimulated by phytic acid. INH was considerably more potent than NH.
  • 3.3. Co-oxidation of a hydrazide and thyroxine (T4) in the MPO system resulted in the formation of a pink-coloured product (maximum absorbance at 504 nm) which was more stable with NH than with INH.
  • 4.4. The hydrazides and Cl acted synergistically on MPO haem modification when co-oxidised in the MPO-H2O2 system. INH was more destructive than NH.
  • 5.5. The different oxidative pathways of the hydrazides are consistent with the fact that an acyl intermediate of INH, unlike that of NH, is resonance stabilized.

  • 1.1. The oxygen consumption rates for three sympatric species of marine gastrotrichs (anatomically similar, except that one contains hemoglobin) were measured with a Cartesian diver microrespirometer.
  • 2.2. The rates for the two species without hemoglobin, Turbanella ocellata and Dolichodasys carolinensis, were 307.2 μl O2 g−1 hr−1 and 108.0 μl O2 g−1 hr−1, respectively, while the rate for the hemoglobin-containing species, Neodasys, was 208.9 μl O2 g−1 hr−1.
  • 3.3. The possession of hemoglobin by Neodasys (14% by volume) cannot be explained by an unusually high demand for oxygen.
  • 4.4. Instead, the hemoglobin may be useful as an oxygen store providing continued aerobic metabolism in anoxic conditions, thus allowing Neodasys to exploit a different niche.

  • 1.1. Effect of controlled up- and down-shifts of growth temperature on the antioxidant enzymes activities and lipid peroxidation were investigated in intact cells of Cyanobacterium synechocystis PCC 6803 acclimated at different growth temperature.
  • 2.2. Algal cells grown at 36°C were treated at 20 and 43°C as down- and upward-shifts of growth temperature for 24 hr, respectively. At the down-shift of growth temperature the superoxide dismutase, catalase and glutathione peroxidase were significantly increased with concomitant decrease in protein content.
  • 3.3. These parameters showed similar temperature dependencies in the up-shift of growth temperature, they were decreased significantly.
  • 4.4. The increased hydroxyl (HO) radical and malonyldialdehyde (MDA) formation, when algal cells exposed to down-shift of growth temperature, supposedly due to stimulated production of superoxide radicals (O2) and hydrogen peroxide (H2O2) at lower temperature.

  • 1.1. u.v. radiations and copper acetate, as free radical generating systems, determine a significant diminishing of glucose-6-phosphate dehydrogenase activity in the homogenates of Saccharomyces cerevisiae.
  • 2.2. The inactivation is proportional to the concentration of the formed free radicals, existing a direct dependence on the action time of the free radicals generating systems and on the irradiation dose. The decrease of the enzyme catalytic activity is correlated with the increase of the malondialdehyde concentration.
  • 3.3. The affinity for the substrate of the enzyme under the action of free radicals does not change significantly compared to the native enzyme: the Km value for NADP is halved, whilst that for glucose-6-phosphate remains unchanged.

  • 1.1. The effects of prostaglandin (PG) E1, and I2 analogs (OP-41483 and OP-2507) on the Superoxide generation of human neutrophil NADPH oxidase (EC 1.6.99.6) in both whole-cell and cell-free systems were investigated.
  • 2.2. In a whole-cell system, OP-2507 inhibited the Superoxide generation by neutrophils exposed to phorbol myristate acetate concentration-dependently through its superoxide-scavenging action.
  • 3.3. The concentration of the drug required for 50% inhibition of the oxidase (IC50) was 21 μM.
  • 4.4. In a cell-free system, however, the drug in concentrations of < 100 μM did not inhibit the activation of NADPH oxidase by sodium dodecyl sulfate because of its inactivation by the detergent.
  • 5.5. Although PGE1 and OP-41483 did not inhibit the Superoxide production by stimulated neutrophils in a whole-cell system, they both inhibited the activation of NADPH oxidase in a cell-free system concentration-dependently, with IC50 values of 44 and 170 μM, respectively.
  • 6.6. In addition, in the cell-free system, the Km value for NADPH of the oxidase was unchanged by PGE1.
  • 7.7. The results suggest that the PGI2 analog, OP-2507, is a possible superoxide-scavenger and that PGE1 inhibits the NADPH oxidase activation by sodium dodecyl sulfate in a cell-free system concentration-dependently.

  • 1.1. An endoxylanase (EC 3.2.1.8) was purified from an Escherichia coli strain carrying a xylanase gene from the extreme thermophile “Caldocellum saccharolyticum strain Tp8T6.3.3.1. It was found to have an Mr of 42,000 and an isoelectric point of approx. 5.0.
  • 2.2. The enzyme showed optimum activity at pH 5.0–7.7 and had an activation energy of 44 kJ mol−1. It was stable at room temperature at pH 4.5–11.5 in the presence of 0.5 mg ml−1 bovine serum albumin. The half-life of the enzyme at 75°C was 20 min at pH 6.0 in the presence of 0.5 mg ml−1 bovine serum albumin.
  • 3.3. The xylanase had highest activity on oat spelts xylan, releasing xylobiose and some xylotriose. The Km for oat spelts xylan was 0.021% (w/v) at pH6.0.
  • 4.4. The enzyme had high activity on sugar cane bagasse hemicelluloses A and B, lower activity on larchwood xylan and also hydrolysed carboxymethylcellulose, 4-methylumbelliferyl β-D-cellobioside and p-nitrophenyl β-D-cellobioside, but could not hydrolyse xylobiose.
  • 5.5. It showed transferase activity on p-nitrophenyl β-D-xylopyranoside. Xylose did not inhibit the enzyme.

  • 1.1. In the presence of insulin, 10−5 M 3,3',5-triiodothyronine (T3) treatment for 1/2 hr decreased fatty acid synthesis 35% only in adipocytes from lean rats, whereas at 10−11 M through 10−7M T3 the obese adipocytes had nearly a 20% increase in fatty acid synthesis.
  • 2.2. A 2 hr pretreatment of adipocytes with 10−9 and 10−7 M T3 decreased insulin-stimulated fatty acid synthesis by nearly 20% in both lean and obese adipocytes.
  • 3.3. In the absence of insulin, the 2 hr pretreatment with 10−9 M T3 resulted in a 45% increase in lean adipocyte fatty acid synthesis, though the obese adipocytes required at least 10−7 M T3 for 2 hr to increase the non-insulin-stimulated fatty acid synthesis by 50%.
  • 4.4. At 10−9M T3 concentrations non-insulin-stimulated fatty acid synthesis was increased by 200% in lean adipose tissue explants, but obese adipose expiants were not significantly affected under these conditions.
  • 5.5. The addition of 10−9 M T3 plus insulin to the explant media decreased fatty acid synthesis by 35% in both the lean and obese tissues.
  • 6.6. The results also imply that the low T3 status of the obese rat may be contributory to the elevated fatty acid synthesis observed in obese adipocytes.

  • 1.1. The binding of O2 to goldfish haemoglobin showed a strong pH dependence P50=5.5 mmHg; n = 2.4 at pH 8.0 and P50 = 170 mmHg; n = 1.0 at pH 5.5 such that the protein is only 50% saturated in a solution of air equilibrated buffer at pH 5.5.
  • 2.2. The binding of CO is cooperative at high pH (n = 2.8; L = 1000; KR = 0.1 μM; KT = 4 μM) and non-cooperative (n = 1) at pH 5.5.
  • 3.3. The rate of O2 dissociation is extremely fast and pH dependent; being 30 sec−1 at pH 8.0 and 400 sec−1 at pH 6.0 at 1°C. At 23°C the rate of this process is too fast to obtain accurate data using stopped-flow techniques.
  • 4.4. Partial photolysis of the oxyhaemoglobin species leads to homogeneous recombination kinetics at pH 8.0 with an associated rate constant of 4.7 × 107 M−1 sec−1. At pH < 7.5 the recombination process occurs in two steps. One rate is equal to that observed at pH 8.0. The slower process is favoured at low pH.
  • 5.5. Photolysis of the CO haemoglobin complex indicates that, at high pH, combination of CO with deoxyhaemoglobin is cooperative, whilst recombination with Hb(CO)3 is non-cooperative and occurs at a rate of 1.2 × 106 M−1 sec−1.
  • 6.6. At neutral pH recombination of CO with partially linganded haemoglobin occurs in a two-step process. The proportion contributed by each of these two steps in pH dependent.
  • 7.7. The functioning of this Root effect haemoglobin is discussed in terms of the two state-model of cooperativity in which the αβ chain heterogeneity is minimal

  • 1.1. Unidirectional Na+ influx in lamprey red blood cells was determined using 22Na as a tracer.
  • 2.2. Total Na+ uptake and amiloride-inhibitable Na+ influx increased in a saturable fashion as a function of external Na+ concentration (Nae).
