Considerable tension among the subfields has existed within the discipline of anthropology. As a result, some anthropology departments have splintered, and the hallmark "holistic approach" of anthropology has been considered more myth than reality. However, as promoted by the American Anthropological Association and the American Anthropologist for over one hundred years, enhancing the holistic nature of anthropology remains an important and necessary endeavor. This article provides an introduction to this special issue of the American Anthropologist , which focuses on the subfield of biological anthropology. Hopefully, as a result, increased connections among the subfields will be fostered, for the betterment of both biological anthropology and anthropology in general. The underlying theme of this article and the subtext for the entire special issue is clear: Biological anthropology needs anthropology, and anthropology needs biological anthropology. Many researchers set up an experiment, make measurements, do an analysis of variance, calculate the mean response for each treatment, and then try to decide if the treatment means are significantly different and why. Much too frequently, Duncan's multiple range test is used to test differences among means. It is only one of a number of techniques that can be used to examine treatment means. Some researchers are unaware of the different techniques and that the interpretation of the results of an experiment is strongly influenced by the technique used i.e. using two different techniques might produce two different interpretations of the results. Selection of the appropriate technique to use for a particular experiment depends upon the nature of the treatments and the objectives of the research. Univariate and bivariate time series analysis techniques have enabled new insights into neural processes. However, these techniques are not feasible to distinguish direct and indirect interrelations in multivariate systems. To this aim multivariate times series techniques are presented and investigated by means of simulated as well as physiological time series. With biomedical literature increasing at a rate of several thousand papers per week, it is impossible to keep abreast of all developments; therefore, automated means to manage the information overload are required. Text mining techniques, which involve the processes of information retrieval, information extraction and data mining, provide a means of solving this. By adding meaning to text, these techniques produce a more structured analysis of textual knowledge than simple word searches, and can provide powerful tools for the production and analysis of systems biology models.  相似文献

The upheaval in the General Medical Council two decades ago came from doctors not the consumers the council was set up to protect. Since then there have been repeated calls for doctors to improve their self regulation by amending the disciplinary procedures. Private member''s bills have failed and the GMC has now proposed performance procedures to deal with doctors who exhibit a "pattern of poor performance." After months of wide consultation in and outside the medical profession the GMC will decide next week whether to endorse the procedures, which unlike the conduct hearings will be inquiries by peers. Professor Margaret Stacey suggests that the procedures lack clarity, smacking of that "trust me" principle whose subtext is "but I''m not telling you what I''m up to."  相似文献

In this study, two new microsatellites HAU02 and HAU06, which are screened and cloned. HUA02 and HAU06 are repectively mapped in lq23-27 and 6q11-21 by means of PRINS (primed in situ labeling) and somatic cell hybrid (SCH) panel techniques, we discussed their advantage and disadvantage in gene mapping. These two means provide us more techniques in gene mapping, and the locations of these new microsatellites accumulate more resource for Porcine Physical Map.  相似文献

It has been proposed that the mechanism of formation of the chorion of Oryzias latipes involves the transfer of precursor material from the Golgi bodies of the oocyte to the chorion by means of a population of dense cored vesicles. Thin sectioned ovarian material was investigated by means of several techniques available for the resolution of periodate reactive material at the ultrastructural level. The techniques employed were modifications of the thiocarbohydrazide, thiosemicarbazide, alkaline bismuth subnitrate and silver methenamine reactions. These all demonstrated a positive reaction in the material of the chorion and that of the dense cored vesicles. This finding is in accord with the hypothesis that the dense cored vesicles are transporting material from the Golgi to the growing chorion.  相似文献

Derivation of haplotypes from pedigree data by means of likelihood techniques requires large computational resources and is thus highly limited in terms of the complexity of problems that can be analyzed. The present paper presents 20 rules of logic that are both necessary and sufficient for deriving haplotypes by means of nonstatistical techniques. As a result, automated haplotype analysis that uses these rules is fast and efficient, requiring computer memory that increases only linearly (rather than exponentially) with family size and the number of factors under analysis. Some error analysis is also possible. The rules are completely general with regard to any system of completely linked, discrete genetic markers that are autosomally inherited. There are no limitations on pedigree structure or the amount of missing data, although the existence of incomplete data usually reduces the fraction of haplotypes that can be completely determined.  相似文献

Although used as a therapeutic for 50 years, it is only recently that the application of molecular techniques has provided a basis for understanding growth hormone's (GH) clinical actions. This article reviews progress in our current knowledge of the molecular mechanism of growth hormone (GH) receptor activation based on a number of physicochemical techniques, and documents insights gained into the means used by the activated GH receptor to control the expression of genes regulating growth and metabolism. These findings are related to disorders of short stature, and the therapeutic consequences are summarized.  相似文献

The origin, production, and means of confirming the authenticity of some historic dyes are described. The underlying chemistry is revealed. The evolution of analytical techniques from early times, when the main criterion for a good textile dye was fitness for use, to more modern chromatographic techniques, where the emphasis is on chemical identity, is illustrated. Recent developments have led to smaller sample size requirements, greater speed of analysis and have led to a more detailed knowledge of the chemical components of historic dyes.  相似文献

Microscale processing techniques are rapidly emerging as a means to increase the speed of bioprocess design and reduce material requirements. Automation of these techniques can reduce labour intensity and enable a wider range of process variables to be examined. This article examines recent research on various individual microscale unit operations including microbial fermentation, bioconversion and product recovery techniques. It also explores the potential of automated whole process sequences operated in microwell formats. The power of the whole process approach is illustrated by reference to a particular bioconversion, namely the Baeyer-Villiger oxidation of bicyclo[3.2.0]hept-2-en-6-one for the production of optically pure lactones.  相似文献

定量蛋白质组学是蛋白质研究的前沿学科。目前常用的定量蛋白质组学研究技术有荧光差异凝胶电泳(DIGE)、同位素亲和标记(ICAT)等。同位素标记相对和绝对定量(iTRAQ)技术是近年来最新开发的一种新的蛋白质组学定量研究技术。结合非凝胶串联质谱技术,该技术可对复杂样本、细胞器、细胞裂解液等样本进行相对和绝对定量研究,具有较好的定量效果、较高的重复性,并可对多达四种不同样本同时进行定量分析。本文对 iTRAQ 技术的原理、实验方法及应用进展进行了综述。  相似文献

Besides availability, there are numerous advantages to using mice for meiotic studies: (1) techniques exist for enriching the population of spermatocytes at particular meiotic stages; (2) spermatogenesis in mice is highly synchronized and testis sections afford a highly accurate method of staging meiotic events; (3) knock-out mice provide a rich source of meiotic mutants. Coupled with antibody localization techniques these tools, singularly and in combination, provide multiple means of temporally and physically dissecting meiosis.  相似文献

There are many statistical techniques that require the assumption that the population being studied is normally distributed--regression analysis, multivariate analysis, time series analysis, and so on. Unfortunately, as the development of survey sampling has long acknowledged, large human populations are usually stratified into several different subpopulations. Since the boundaries between the strata are somewhat blurred, they are not independent, so the overall distribution of the population tends to be multimodal rather than normal. In this paper, a technique is developed to find these multimodal techniques using nonparametric density estimation. Its effectiveness is demonstrated by means of an example.  相似文献

Research in the mechanism of endocrine disorders in functional disturbances of the central regulatory apparatus requires the development of an appropriate model of the affection. The employment of an experimental neurosis would seem to be promising in this regard. The Pavlov school has thoroughly elaborated techniques of inducing experimental neuroses in dogs, but when small laboratory animals (mice, rats) are employed in large-scale experiments, existing techniques do not provide a means of inducing a long-lasting pathological condition of the higher centers of the central nervous system.  相似文献

土壤微量元素测试在其发展历史过程中,不断改进技术,逐渐深入地揭示土壤供给微量元素的能力,对指导施肥和保护生态环境起着积极的作用。现代测试手段发展到广泛采用原子吸收光谱(AA)和电感耦合等离子体发射光谱(ICP),但比色法(光度法)和极谱法不仅设备比较简便,而且新的显色剂、催化系统等方面的研究进展,使其对某些元素的测定灵敏度和准确度超过AA和ICP,从而在微量元素测试技术中仍占据一定地位。土壤溶液抽取技术虽然有所改进,但有效态微量元素仍然主要是选用适当提取剂来提取测定。临界值和分级标准的确定是应用测定值的桥梁。作者曾采用全幅分级标准分级制图,确定缺素面积和需肥区域,再根据土壤含量水平和增产幅度之间的函数关系预测增产效果和投入的经济效益。  相似文献
