Originally described in insect viruses, cellular proteins with Baculoviral IAP repeat (BIR) motifs have been thought to function primarily as inhibitors of apoptosis. The subsequent finding that a subset of IAPs that contain a RING domain have ubiquitin protein ligase (E3) activity implied the presence of other functions. It is now known that IAPs are involved in mitotic chromosome segregation, cellular morphogenesis, copper homeostasis, and intracellular signaling. Here, we review the current understanding of the roles of IAPs in apoptotic and nonapoptotic processes and explore the notion that the latter represents the primary physiologic activities of IAPs.  相似文献

Telomeres are nucleoprotein complexes that cap the end of eukaryotic chromosomes. They are essential for the functions and the stability of the genomes. There is now compelling evidences that telomerase, the enzyme that adds telomeric DNA repeats to chromosome end, is an important player in oncogenesis. The absence of telomerase in somatic tissues is thought to promote genome instability at initial stages of oncogenesis, favoring the emergence of cancer-associated chromosomal abnormalities \; restablishment of telomerase activity is expected afterwards if long term cell cycling is to occur. In addition to telomerase, various factors control the structure and function of telomeres, suggesting that additional telomeric components play important roles during oncogenesis.  相似文献

Despite its failure to predict variation in secondary chemistry for many metabolites and taxa, the Carbon/Nutrient Balance (CNB) hypothesis continues to be invoked with regularity. According to Lerdau and Coley (2002) , the value of the hypothesis is that it explains phytochemical phenotypic plasticity outside of an evolutionary context. We contend that the CNB hypothesis impedes a more comprehensive understanding of plant biology and that an approach divorcing plant chemistry from function is inherently limited in its ability to predict responses of plants to the environmental conditions in which they have evolved. Although it is possible for a mechanistically flawed hypothesis to predict phenomena, such hypotheses almost always suffer a lack of generality. Lerdau and Coley argue that the limitations of the CNB model are its strengths, not its weaknesses, and set forth refinements that, in essence, further limit its applicability. We address these refinements and explore alternative adaptive explanations for adjustments of secondary metabolism in relation to resource availability and environmental variation. In view of its flawed nature and the existence of alternative approaches, CNB no longer warrants consideration as a viable hypothesis.  相似文献

There is currently a great deal of interest in the relative contributions of external and internally sequestered Ca(2+) in various physiological responses. However, this field is losing clarity due to inattentive use of terms. In particular, the terms "receptor-operated channels" and "store-operated channels" (ROCs and SOCs, respectively) are becoming ambiguous through over-use. In this paper, we will first consider basic principles of channel gating in order to set the stage for defining criteria which can be used to distinguish precisely between different channel-related functions. We will then focus on recurring examples of adventurous use of terminology, or of blurring of the distinctions between channel types, and propose solutions to this quandary.  相似文献

The term 'type III secretion' has seen widespread use. However, problems persist in nomenclature. We propose that the standard abbreviation for this kind of secretion should be 'T3S' and that 'type III secretion system' should be abbreviated to 'T3SS'. There is also a need for a new terminology to distinguish flagellar and non-flagellar type III secretion systems that reflects their common evolutionary ancestry but does not obscure their distinctive features. Finally, the use of the term 'type III secretion' to cover cytolysin-mediated translocation is to be deprecated because an authentic type III secretion system has already been described in gram-positive bacteria, namely the flagellar protein export apparatus.  相似文献
