Somatic embryos were obtained from 1% of cotyledon pieces and hypocotyls of mature embryos of Eucalyptus globulus Labill. cultured on media containing a high concentration of picloram or IBA. 2,4-D and other synthetic auxins did not yield somatic embryos or embryogenic callus. Somatic embryos arose indirectly via callus, being visible after four months, and directly, where little callus or adventitious root initiation occurred. Somatic embryos, formed directly from explants, were visible within five weeks. Various structural abnormalities of somatic embryos were observed, especially after induction on media containing picloram. Only two out of fifteen somatic embryos showed hypocotyl and radical elongation, but plantlets did not develop further.  相似文献

以Sb33高粱非胚性、胚性愈伤组织和体胚为材料,用传统石蜡切片法对各组织材料进行组织化学染色,对高粱胚性与非胚性愈伤组织以及体胚进行组织细胞学观察。结果表明:高粱非胚性愈伤组织无淀粉粒积累,高粱胚性愈伤组织淀粉粒积累较多,而与胚性愈伤组织相比,高粱体胚淀粉粒积累更多,这说明淀粉粒的积累与高粱体细胞的胚胎发生密切相关。此外,高粱可通过鱼雷胚基部产生球形胚的方式实现体胚的增殖,高粱离体再生途径以体细胞胚发生为主,并同时存在少量器官发生途径。在高粱体细胞胚胎发生中,外起源和内起源同时存在。本研究为高粱体细胞胚胎发生提供细胞学理论基础。  相似文献

Induction of somatic embryogenesis in leaf explants from young mangosteen seedlings using different concentrations and combinations of 6-benzylaminopurine (BAP) and thidiazuron (TDZ) was investigated. The best medium inducing the formation of globular structures (40 %) was Murashige and Skoog medium with 0.7 mg dm−3 BAP and 0.7 mg dm−3 TDZ. For their further development, subculturing onto different maturation media was carried out, but these globular structures did not develop futher stages of somatic embryogenesis. However, they developed shoots after 90 d of culture on the original medium. Morphological and histological analyses were performed, and showed that the globular structures resembled closely the undifferentiated structure of the mangosteen seed. We propose that the development of mangosteen somatic embryos does not follow the typical course of somatic embryogenesis, but the course of development that is natural for mangosteen seed, where procambium is the only structure observed and there is no differentiated embryo.  相似文献

Embryogenic callus was obtained from bulb segments of Iris pseudacorus on Murashige and Skoog (MS) medium with 2,4-dichlorophenoxyacetic acid (2,4-D) alone or in combination with kinetin. When early globular somatic embryos were subcultured onto MS medium with 4.52 μM 2,4-D, high frequency of somatic embryogenesis was obtained. Deprivation of 2,4-D was required for maturation. Mature somatic embryos had an elongated scutellum with a notch on the base of scutellum. Separation of embryos from embryo clusters was necessary to enhance the frequency of germination. Germination was stimulated by separation of embryos from embryo clusters and transfer onto fresh half-strength MS medium with 3% sucrose. After acclimation in artificial soil in greenhouse for 2 months, 96.4% of plantlets survived.  相似文献

In monocots, the zygotic embryo is protected and nourished by an endosperm. In the present study starch deposition and amylase accumulation was noticed during somatic embryogenesis in stem callus of a bamboo, Dendrocalamus hamiltonii. SEM studies revealed that starch grains were clearly visible in the scutellum during the maturation stage of the somatic embryo. As the somatic embryo developed further, the scutellum got reduced with corresponding increase in amylase. The amylase activity was tested periodically at different developmental stages of embryos. The role of scutellum in somatic embryos for starch deposition and amylase accumulation is discussed.  相似文献

Background and Aims The date palm is a dioecious perennial species of the Arecaceae for which in vitro micropropagation is essential to ensure the renewal of palm plantations. This study presents a histocytological analysis of the traditional Mauritanian Amsekhsi cultivar beginning from the initial callogenesis and continuing up to the establishment of the cellular embryogenic cell suspensions. The formation of somatic embryos and their development into rooted plants are also described.• Methods Foliar segments of seedlings cultured in the presence of 2,4-D produced primary calli that were chopped to produce fine friable granular calli that subsequently produced cellular suspensions when transferred to liquid medium. The somatic proembryos that developed after removal of the 2,4-D were plated on agar medium where they developed into rooted plants. Thin sections of tissue fragments taken at each stage of the process were stained using Periodic Acid Schiff and Naphthol Blue-Black.• Key Results The first cellular divisions were localized close to the vascular vessels of the leaf. The primary calli were obtained within 2 months. Fine friable granular calli grew quickly after the primary calli were chopped. Individual embryogenic cells were identified that rapidly started to divide and developed into globular proembryos. In addition, in the microcalli, breaking zones appeared in the thick pectocellulosic walls which delimited the pluricellular proembryos. The anatomy of somatic embryos is similar to that of zygotic embryos despite a deficit in the accumulation of intracellular proteins. When rooted with NAA, the vitroplants developed a strong orthotropic taproot.• Conclusions This study contributes to understanding the whole process of somatic embryogenesis, but two specific questions remain to be answered: what factors are involved in the reactivation of the somatic cells at the beginning of the initial callogenesis, and why do the somatic embryos not accumulate proteins in their tissues during maturation?  相似文献

Eucalyptus grandis and Eucalyptus globulus are important species for the Brazilian forestry industry. E. grandis plantations are mainly found in tropical regions, yet E. globulus plants are usually cultivated under moderate to low temperature conditions. As temperature seems to be a key factor for the planting of these species, we revisited our previously generated shotgun proteomics dataset to identify the main patterns of proteome regulation induced by thermal stimulus and to pinpoint specific proteins involved in the environmental response. Large-scale analysis has pointed out the different proteomic responses of E. grandis and E. globulus under temperature stimulus, with 296 proteins considered to be differentially regulated in the stems of Eucalyptus spp. grown at different temperatures. A stringent filtering approach was used to identify the most differentially regulated proteins. Through the stringent criteria, 66 proteins were found to be enriched in the plant species. Cultivation of E. globulus plants in low-temperature conditions induced the highest number of differentially regulated proteins. Additionally, metabolic proteins were mostly down-regulated, while stress-related proteins were majorly up-regulated in both species. Finally, the subset of the most differentially regulated proteins comprised new candidates of protein markers of temperature stress.  相似文献

Cotyledon explants from zygotic embryos of Panax ginseng produced somatic embryos on Murashige and Skoog basal medium without growth regulators. Somatic embryos developed directly from epidermal cells at the cotyledon base. Somatic embryos were always formed from the side of the cotyledon opposite to the one attached to the medium surface regardless of cotyledon orientation. The frequency of somatic embryo formation from the abaxial epidermis (66%) was much higher than that from the adaxial epidermis (12%). Differences in embryogenic response were likely related to cell structure. Abaxial epidermal cells were filled with reserve materials (lipid bodies), while adaxial epidermal cells were devoid of any prominent reserves. During germination, the reserve materials in the cells of the cotyledons disappeared rapidly. At the same time, the competency of somatic embryo formation from cotyledon explants declined rapidly to zero. Upon culture of the cotyledon explants (for somatic embryo induction), lipid bodies slowly disappeared, but starch grains accumulated prominently. Reserve materials disappeared after commencement of embryogenic cell division. During germination, lipid bodies rapidly disappeared, and chloroplasts developed instead of starch grains. Leaf explants of the HBsAg-transgenic cherry tomato (Solanum lycopersicum) mutant were cultured on Murashige and Skoog (MS) basal medium, supplemented with 1.0 mg/L 6-BA and 0.05 mg/L IAA for callus induction, to clarify the physiological and biochemical characteristics of morphogenesis development. Therefore, the physiological and biochemical changes during the development of organogenic shoots and somatic embryos in the mutant were studied. Superoxide dismutase (SOD) activities of the mutant had only one peak value on the 21st day. Peroxidase (POD) activities of the mutant declined less sharply since the explants were cultured. IAA oxidase activity of the mutant increased steadily until 42 days from culturing and then decreased sharply. Malondialdehyde (MDA) of the mutant showed a significant decreasing trend after 21 days from culturing. Growth rate of the mutant was at times lower than that of the control during its callus differentiation, and the soluble protein content of the mutant callus decreased from explant cultivation until the 28th day of culture. The mutant had greater values of chlorophyll a, carotenoid and Chlorophyll contents than the control after 14 days of culturing, and Chlorophyll b content of the mutant showed a declining trend. The electrical conductivity trend of the mutant was consistent with that in the control. It indicated that in terms of the organogenesis or somatic embryogenesis pattern, protein synthesis and catabolism were very active, and a number of antioxidant enzyme activities were consistent in the early development stages of the two regeneration systems. These findings were useful for the regeneration of the mutant.  相似文献

Soybean somatic embryos were developed as a model for investigating the developmental relationships of lipid biosynthesis and accumulation in this important crop. Batch cultures of embryos grown for 8 wk in liquid culture medium exhibited typical sigmoidal growth kinetics as they passed through characteristic globular, heart, torpedo, and cotyledon stages. Exponential growth occurred for the first 4 wk in culture with net growth terminating when total embryo fresh weight per culture flask reached a maximum of 4–4.5 g at 6 wk. This was followed by a slight decrease in embryo fresh weight (FW) and the onset of apparent tissue senescence as judged by yellowing and browning of embryos. On a FW basis, embryos accumulated up to 4% protein, 2.5% soluble sugars, 1.9% starch, and 1.5% lipid relatively early in development. Levels decreased to 0.8% protein, 0.5% soluble sugars, 0.03% starch, and 0.09% lipid at the end of the culture period. On a mass percent basis, lipid extracts were comprised of approximately 80–90% polar lipid early in embryo development. This shifted to 56% storage lipid (triacylglycerol) and 44% polar lipid after 4–5 wk in culture and then reverted back to 91% and 9% polar vs. storage lipid, respectively, by the end of the 8-wk culture period. On the average, polar and storage lipid fractions were comprised of 24% palmitic acid, 7% stearic acid, 8% oleic acid, 36% linoleic acid, and 26% linolenic acid. However, the amounts of linoleic and linolenic acids declined sharply during embryo senescence at the end of the culture period, with corresponding increases in the combined amounts of palmitic and stearic acids. This is the first report that documents the progress of storage reserve accumulation in soybean somatic embryos in relation to their continuous growth in liquid batch cultures.  相似文献

Eucalypt cultivation is an important economic activity worldwide. In Portugal, Eucalyptus globulus plantations account for one-third of the total forested area. The nutritional requirements of this crop have been well studied, and nitrogen (N) is one of the most important elements required for vegetal growth. N dynamics in soils are influenced by microorganisms, such as diazotrophic bacteria (DB) that are responsible for biological nitrogen fixation (BNF), so the aim of this study was to evaluate and identity the main groups of DB in E. globulus plantations. Samples of soil and root systems were collected in winter and summer from three different Portuguese regions (Penafiel, Gavião and Odemira). We observed that DB communities were affected by season, N fertilization and moisture. Furthermore Bradyrhizobium and Burkholderia were the most prevalent genera in these three regions. This is the first study describing the dynamic of these bacteria in E. globulus plantations, and these data will likely contribute to a better understanding of the nutritional requirements of eucalypt cultivation and associated organic matter turnover.  相似文献

Understanding the factors affecting variation in phenology within a species is important as flowering time constitutes one of the major barriers to gene flow. We studied the genetic and environmental control of flower initiation and anthesis time in E. globulus ssp. globulus. For 5 years, flower initiation and anthesis were monitored in a seed orchard containing clones of 63 genotypes from four different regions of the species’ natural distribution. Anthesis occurred over a long period each year, spanning as much as 9 months in 2008. This variation was under strong genetic control with little genotype by year interaction (broad-sense heritability, Ĥ 2 = 0.78 ± 0.04). There were highly significant differences among regions; anthesis occurred earlier for Furneaux and Tasmania than Strzelecki and Otways each year. Surprisingly though, there was little variation in flower initiation time between regions and genotypes, and this was under weak genetic control (Ĥ 2 = 0.06 ± 0.05). The average anthesis time in the orchard varied from year to year, and there was evidence that heat sum was a major driver of this environmental variation. Anthesis time is controlled by both genetic and environmental factors, with the responses to each being predictable to some extent, and unrelated to the timing of flower initiation.  相似文献

Summary Flooding for up to 40 days induced morphological changes and reduced growth of 6-week-old Eucalyptus camaldulensis and Eucalyptus globulus seedlings. However, the specific responses to flooding varied markedly between these species and with duration of flooding. Both species produced abundant adventitious roots that originated near the tap root and original lateral roots, but only E. camaldulensis produced adventitious roots on submerged portions of the stem. Flooding induced leaf epinasty and reduced total dry weight increment of seedling of both species but growth of E. globulus was reduced more. In both species dry weight increment of shoots was reduced more than dry weight increment of roots, reflecting compensatory growth of adventitious roots. The described protocol for repetitive somatic embryogenesis (SE) in Eucalyptus globulus produced more somatic embryos than the primary SE protocol. Primary somatic embryos (induced on MS3NAA) were transferred to the same medium, leading to new cycles of somatic embryos, for at least 2 years. The influence of medium (MS and B5), plant growth regulators (auxins and cytokinins), and light on secondary SE was tested. The MS medium without growth regulators (MSWH) was more efficient for cotyledonary embryo formation and germination than the B5 medium. Reducing auxin (NAA) levels increased the proliferation of globular somatic embryos and allowed SE competence to be maintained on medium free of plant growth regulators. The addition of two cytokinins (BAP and KIN) to the MS medium did not improve proliferation of globular secondary embryos, but was crucial during later stages of the SE process (germination and conversion). Data also show that light may influence the quality of the process, depending on its stage. Darkness should be maintained until the cotyledonary stage is reached, after which exposure to light is recommended.  相似文献

This review focuses on investigation in acquisition of embryogenic competence during somatic embryogenesis in the last five decades. In tissue culture, differentiated somatic cells acquire embryogenic competence and proliferate as embryogenic cells during the induction phase. These embryogenic cells are important because they differentiate to form somatic embryos at a later time. Various molecular and structural markers for detecting embryogenic cells or enhancing embryogenic competence are summarized and implications of the findings are discussed.  相似文献

Summary Adventitious buds and shoots of Eucalyptus globulus Labill. (Tasmanian Bluegum) have been regenerated from cotyledons and hypocotyls from mature embryos and seedlings. Adventitious buds, were induced at high frequency with 0.05 μM thidiazuron in combination with 0.2 μM 2,4-dichlorophenoxyacetic acid or 5 μM α-naphthaleneacetic acid. Culture of explants in the dark inhibited bud induction, but up to 86% of cotyledons, longitudinally split just prior to culture, produced adventitious buds, in the light. Development of buds into shoots occurred only at low frequency, after transfer to media containing N6-benzylaminopurine.  相似文献

In vitro cultures of Leucojum aestivum are considered as an alternative for the production of galanthamine, which is used for the symptomatic treatment of Alzheimer’s disease. We studied the effects of auxins 2,4-dichlorophenoxyacetic acid (2,4-D), 4-amino-3,5,6-trichloropicolinic acid (picloram), 3,6-dichloro-o-anisic acid (dicamba) at concentrations of 25 and 50 µM on the induction of embryogenic callus and its capacity to induce somatic embryogenesis and alkaloid accumulation. The embryogenic response of the explants was from 30% for 25 µM of dicamba to 100% for picloram (for both 25 and 50 µM). 2,4-D (50 µM) stimulated greater callus proliferation and somatic embryo induction as compared to the other auxins. Polyethylene glycol (PEG) stimulated somatic embryo maturation. Callus grown on media containing 50 µM of auxins produced fewer phenolic compounds as compared with callus grown on media containing 25 µM of auxins. GC-MS analyses showed seven alkaloids in the in vivo bulbs and two to four in callus culture. Galanthamine was detected in callus cultivated with 2,4-D (25, 50 µM), picloram (25 µM), and dicamba (50 µM). Other alkaloids, trisphaeridine, tazettine, and 11-hydroxyvittatine were accumulated only in callus growing on medium with picloram (50 µM).  相似文献

The plant growth retardant paclobutrazol was applied by foliar spray to seedlings of Eucalyptus globulus (Labill.) to determine its effects on reproductive activity. It was shown to induce flowering in vegetatively juvenile seedlings and increase reproductive activity in vegetatively adult material. The former case represented a 50% (three year) reduction in generation time for commercial seedlings. Paclobutrazol treatment did not appear to have a negative effect on any aspect of reproductive development, germination of new seed, or early seedling growth, while having a distinct positive effect on reproductive output. This suggests that paclobutrazol may be an effective tool enabling tree breeders to reduce generation time and increase reproductive output in E. globulus. Foliar spraying during autumn was shown to be an effective means and timing of paclobutrazol application, with results evident in the following flowering season. Considerable persistence of paclobutrazol effects was noted in the second season after application. Pot size and growth temperatures also play roles in determining the extent of new reproductive activity each season in E. globulus seedlings.  相似文献
