The CoASTAL cohort represents the first community cohort assembled to study a HAB-related illness. It is comprised of three Native American tribes in the Pacific NW for the purpose of studying the health impacts of chronic, low level domoic acid (DA) exposure through razor clam consumption. This cohort is at risk of DA toxicity by virtue of their geographic location (access to beaches with a history of elevated DA levels in razor clams) and the cultural and traditional significance of razor clams in their diet. In this prospective, longitudinal study, Wave 1 of the cohort was comprised of 678 members across the human lifespan, with both sexes represented within child, adult, and geriatric age groups. All participants were followed annually with standard measures of medical and social history; neuropsychological functions, psychological status, and dietary exposure. DA concentrations were measured at both public and reservation beaches where razor clams are acquired. Multiple metrics were piloted to further determine exposure. Baseline data indicated that all cognitive and psychological functions were within normal limits. In addition, there was considerable variability in razor clam exposure. Therefore, the CoASTAL cohort offers a unique opportunity to investigate the potential health effects of chronic, low level exposure to DA over time.  相似文献

A clam bed of 21 km2 was discovered at Gormanstown on the East coast of Ireland in 1997. Between July 1998 and August 1999, dredge samples were collected from vessels fishing there and these provided material for biological analysis. E. siliqua occurs with an inshore invertebrate fauna; occasional occurrences of large numbers of Donax vittaus, Pharus legumen and Lanice conchilega were associated with razor clams of less than 6 cm long. As harvesting proceeded smaller individuals appeared in the samples; these may have immigrated onto the bed from the periphery of high densities of larger clams. On the clam bed the animals ranged from 0+ to 19+ years old. Ensis siliqua grew at a slower rate on the west coast of the Irish Sea than on the coast of North Wales. Males grew slightly faster and longer than females and males were more numerous in the landings. The clams are estimated to commence maturation at approximately 4 years of age. They have a similar gonadal cycle to those in Portugal and a similar spawning period from mid-May to the end of July or early August. Condition reflects gonad development, reaching a peak in May. Harvesting by hydraulic dredging causes breakages which are higher in quarters 1 and 4 than in quarters 2 and 3. Bruised razor clams do not eliminate sand in desanding tanks and so they are rejected by processors. The total biomass of the Gormanstown bed is estimated at approximately 1500 tonnes of which 2/3 had been removed by July 1999, two years after exploitation commenced.  相似文献

Domoic acid (DA), the toxin responsible for amnesic shellfish poisoning (ASP) can accumulate in king scallop Pecten maximus leading to extensive fishery closures. Approximately 59% of the total value of all fish and shellfish landed in the Isle of Man in 2004 comprised king scallop, hence the economy of the Manx marine sector is particularly susceptible to impacts from this biotoxin. Scallop from fishing grounds around the Isle of Man were sampled in October 2003, June 2004 and October 2004 to determine levels of inter-animal and spatial variability in DA concentration and factors that might influence toxin concentration such as scallop size and water depth. Mean DA concentrations in hepatopancreas ranged from 296.3 μg g−1 to below the detection limit, in gonad from 27.8 μg g−1 to below the limit of detection and in adductor muscle from 7.3 μg g−1 to below the limit of detection. High levels of inter-animal variability of DA concentration in hepatopancreas were recorded; CVs ranging from 16.1% to 70.0%. DA concentrations above 20 μg g−1 were recorded in gonads on all three sampling dates. Scallops from fishing grounds on the east of the Isle of Man were significantly less contaminated than those from the west and southwest. A significant positive correlation between DA concentration and shell length was recorded in some sites, but there was no relationship with water depth. The high inter-animal, spatial and seasonal variability in toxin concentration highlighted the importance of understanding field variability for the development of reliable sampling and management protocols.  相似文献

The annual reproductive cycle of the Antarctic soft-shelled clam, Laternula elliptica, in Marian Cove, King George Island was studied over a 2-year period from February 1998 to January 2000. Annual changes in the gametogenesis were investigated by measuring the percentage of area occupied by oocytes in a follicle [follicle index (FI)] and the oocyte size. In 1998, the monthly mean FI increased significantly from October to November, peaked in December, and decreased rapidly from December to January. In February and March 1999, degenerated eggs were observed in the spent follicles. Degeneration and resorption of residual eggs by phagocytosis occurred mostly in February and March in both 1998 and 1999, although the resorption process was observed year-round. The histology indicated that complete vitellogenic growth of L. ellpitica at Marian Cove takes at least a year and the clams spawn annually during the austral summer. The ripening and subsequent spawning of clams at Marian Cove in 1998 and 1999 coincided with the algal blooming (September–October 1998 and December and January 1999–2000) suggesting that in coastal Antarctica food supply is a crucial factor that governs gonad maturation and subsequent spawning along with the water temperature.  相似文献

Concentrations of domoic acid (DA) above the regulatory limit in Washington coast razor clams are usually higher on northern beaches from summer to fall. Recent field studies have confirmed that the primary source of toxic Pseudo-nitzschia (PN) cells in those seasons is a semi-retentive topographically trapped seasonal eddy located offshore and north of the clamming beaches. Another semi-retentive coastal feature, Heceta Bank, that has been shown to support toxic PN cells in summer, is located south of Washington's clamming beaches. In this paper we present evidence to demonstrate that Heceta Bank, although not a likely source of toxic cells to Washington in summer due to the prevailing southward seasonal currents, may be a source of cells in springtime before the southward currents develop. In contrast to summer and fall seasons, concentrations of DA in razor clams are typically higher at southern beaches in spring. The likelihood of a southern source is explored using biological and transport data surrounding a period of toxic razor clams in April 2005. In particular, satellite-derived chlorophyll data confirm that a bloom occurred over Heceta Bank in March of that year, just prior to a period of strong storm-driven northward transport. PN cells of the same species observed in the April bloom on Washington beaches and in offshore waters were documented in Oregon offshore waters on the northern edge of Heceta Bank. That species, P. australis, has been shown to be highly toxic in this region; shore-based data show that razor clams on Oregon beaches were also toxic at the time when P. australis was observed offshore. Both measured and modeled currents show that transport was more than sufficient to move cells from the vicinity of Heceta Bank, Oregon to southern Washington beaches by the time the toxic cells were observed on those beaches. The rapid transport was due in part to the presence of the buoyant plume from the Columbia River, a common feature in winter and spring in nearshore waters of the U.S. Pacific Northwest.  相似文献

The mangrove clam Anodontia edentula is highly prized in thePhilippines for its flavor and large size. Because this infaunal species isfound down to one meter deep in mangrove areas, harvesting the clam reportedlydamages mangrove stands. To evaluate such reports, a survey of collectionmethods was undertaken in Panay and Guimaras, central Philippines in August1997–December 1999. Host to chemosynthetic bacterial symbionts thatutilize sulfide as energy source, A. edentula are strategically situated insulfide-rich anoxic substrates but also gain access to oxygenated seawaterthrough a ventilation burrow or tube. By locating the opening of this burrow,collectors can detect the presence of a buried clam and harvest itnondestructively with a blade or bare hands. In contrast, the indiscriminatetilling of wide mangrove areas can damage mangrove plants. Most collectors were40–45 years old with 22–30 years collection experience, married with5–7 children, and had low educational attainment. They sold clams directlyin the local markets or through middlemen (to restaurants and beach resorts);sales provided from 10% to 100% of daily family income. Concentrations of domoic acid (DA), the biotoxin responsible for amnesic shellfish poisoning (ASP), exceeding the regulatory limit of 20 μg g−1 have caused restricted harvesting and closures of wild king scallop fisheries. Toxin monitoring programmes have reported significant inter-animal variation in DA concentration between scallops from the same area. For the development of reliable sampling and management protocols an understanding of the magnitude and causes of inter-animal variation in toxin concentration are important. Ten samples were collected from an aquaculture site in Clew Bay, Co. Mayo off the west coast of Ireland between February 2003 and February 2004, each sample comprising 12 scallops of each of the following size groups: small (70–85 mm), medium (85–100 mm), large (100–115 mm) and very large (>115 mm). DA concentration in each hepatopancreas and in composite samples of both gonad and adductor muscle from each size group on each sampling occasion were measured. High inter-animal variability in DA concentration in hepatopancreas was recorded; CVs ranging from 12.5% to 82.5%. One negative correlation (R2 = 0.7079) between DA concentration in hepatopancreas and scallop shell length, three positive but weak correlations (R2 = 0.4536, 0.3459 and 0.4665) and six no correlations were exhibited. Negative correlations were attributed to faster DA uptake by smaller scallops, positive correlations to faster DA depuration by smaller scallops. If only scallops greater than or equal to 100 mm shell length, the minimum commercial size of this species were considered, no correlation occurred on any of the 10 sampling occasions.  相似文献

Sexual reproduction is documented for the first time in field populations of the pennate diatoms Pseudo‐nitzschia australis Freng. and P. pungens (Grunow ex Cleve) Hasle (var. cingulata Villac and hybrids between var. cingulata and var. pungens). A bloom dominated by these species began on June 26, 2006, along Kalaloch Beach, Washington, USA, coincident with a drop in the Si(OH)4:NO3 ratio to below two. Multimodal size distributions were detected for both species, and synchronous auxosporulation occurred within the smallest size class during a 3‐week window. Auxospores and initial cells created a new class of large cells, and cells in the intermediate size classes increased in abundance during auxosporulation. Mating cells of both species were attached to colonies of surf‐zone diatoms. Paired gametangia, gametes, zygotes, auxospores, and large initial cells were found. Auxosporulation began first for P. pungens (June 30), apparently once a critical, high cell concentration was reached, followed by P. australis (July 5), when the total Pseudo‐nitzschia cell concentration reached 929,000 cells · L?1. Low frequencies of auxosporulation occurred throughout the bloom but increased 4‐fold for P. australis and 3‐fold for P. pungens when macronutrients were reduced to low levels on July 11. A 2‐year life cycle was estimated for P. australis and 3 years for P. pungens, both with annual auxosporulation. Domoic acid (DA) in razor clams reached a maximum of 38 μg DA · g?1 on July 18. A significant relationship existed between the percent of cells within the new size range and DA concentrations in razor clams on the same beach.  相似文献

Several species of the toxigenic diatom Pseudo-nitzschia, together with low concentrations of domoic acid (DA) in shellfish have been observed in Puget Sound, Washington State, since 1991. However, for the first time in September 2003, high-density blooms of Pseudo-nitzschia forced the closure of recreational, commercial, and tribal subsistence shellfish harvesting in Puget Sound. Here we report on the environmental conditions associated with shellfish closures in two inland waterways of Washington State during the Fall 2005. In Sequim Bay, shellfish harvest losses occurred on September 12 following the measurement of elevated macronutrient levels on September 2, and a bloom of P. pseudodelicatissima (up to 13 million cells/L) on September 9. Ambient NH4 concentrations >12 μM (measured on September 2) were likely due to anthropogenic sources, ostensibly from sewage inputs to Sequim Bay. The closure of a Penn Cove commercial shellfish farm on October 16 was caused by a bloom of P. australis that followed a period of sustained precipitation, elevated Skagit River flow, and persistent southeasterly winds. The relative importance of a number of environmental factors, including temperature, stratification caused by rivers, and nutrient inputs, whether natural or anthropogenic, must be carefully studied in order to better understand the recent appearance of massive blooms of toxigenic Pseudo-nitzschia in the inland waterways of Washington State.  相似文献

We examined recent temporal trends in the abundance of fingernial clamsMusculium transversum (formerlySphaerium transversum) in the upper Mississippi River. Historical data on densities of fingernail clams were obtained from regional scientists and published literature. We also sampled benthos in six navigation pools in summer 1991, finding very few fingernail clams. The combined data set, including historical data and sampling results, extended from 1973 to 1992 and was sufficient to statistically evaluate trends in densities of fingernail clams in eight pools. Populations of fingernail clams declined significantly in five of the eight pools examined (Pools 2, 5, 7, 9, and 19), which spanned a 700-km reach of river from St. Paul, Minnesota, to Keokuk, Iowa. Densities in Pool 19, which had the longest historical record on fingernail clam abundance, averaged 30 000 m−2 in 1985 and progressively declined to zero in 1990. Combined data from all eight pools showed a significant decline in abundance of fingernail clams. An evaluation of potential causal factors led us to hypothesize that the population declines in Pools 2 to 9 were linked to point-source pollution rather than to dredging activity or commercial navigation traffic. In Pool 19, the declines of fingernail clams may have resulted from low-flow conditions during drought periods, but the causal mechanisms by which low flow influences fingernail clam abundance are unclear. The decrease in fingernail clam populations may adversely affect certain fish and wildlife, such as migrating lesser scaupAythya affinis, which feed heavily on the small mollusk. Moreover, the decreases in populations of this pollution-sensitive mollusk may signal a large-scale deterioration in the health of this riverine ecosystem. This study represents a preliminary effort to examine the potential impacts of chronic, low level domoic acid (DA) exposure on memory in the CoASTAL cohort over the first four years of data collection (Wave 1). Five hundred and thirteen adult men and women representing three Native American Tribes were studied annually with standard measures of cognition and razor clam consumption (a known vector of DA exposure) over a four-year period. In addition, a pilot metric of DA concentration exposure was used which took into consideration average DA concentration levels in source beaches, as well as the amount consumed. Based upon generalized estimating equations (GEE) analysis, controlling for age, sex, race, year, education level, tribe, and employment status, findings indicated that high razor clam consumers (15 or more per month) had isolated decrements on some measures of memory (p = 0.02–0.03), with other cognitive functions unaffected. The relatively lower memory scores were still within normal limits, and were thus not clinically significant. The pilot DA exposure metric had no association with any other aspect of cognition or behavior. There is a possible association between long-term, low-level exposure to DA through heavy razor clam consumption and memory functioning.  相似文献

Plankton samples from three inland embayments and several outer coastal sites of Washington State were collected from 1997 through 1999 and were examined for the presence of diatoms of the genus Pseudo‐nitzschia and levels of the toxin, domoic acid (DA). Seven species were observed, including Pseudo‐nitzschia pungens (Grunow ex Cleve) Hasle, P. multiseries (Hasle) Hasle, P. australis Frenguelli, P. fraudulenta (Cleve) Hasle, P. cf. heimii Manguim, P. pseudodelicatissima (Hasle) Hasle, and P. delicatissima (Cleve) Heiden. The coastal Pseudo‐nitzschia species assemblages differed significantly from those observed within embayments. The dominant species observed at coastal sites were P. pseudodelicatissima and P. cf. heimii. Pseudo‐nitzschia assemblages found in embayments included one or more of the following species: P. pungens, P. multiseries, P. australis, P. pseudodelicatissima, and P. fraudulenta. The nuclear large subunit rRNA gene was sequenced for six of the seven species identified. This sequence revealed that P. multiseries, P. pungens, P. australis, and P. heimii were genetically similar to those found in California, whereas P. delicatissima and P. pseudodelicatissima were distinct from the California isolates. Although the concentrations of DA in razor clams along Washington State coasts have exceeded regulatory limits several times since 1991, levels of DA in shellfish from Washington State embayments have not yet exceeded regulatory limits. The widespread presence of toxin‐producing Pseudo‐nitzschia species suggests, however, that toxic blooms are likely to occur within embayments in the future. In conjunction with the monitoring of environmental conditions conducive to toxic bloom formation, the development of species‐specific probes for rapid and accurate detection of potentially toxic Pseudo‐nitzschia species in this region would enable the forecasting of a toxic event before DA accumulates in shellfish, thereby reducing the impacts to coastal communities.  相似文献

The amnesic shellfish poison domoic acid is produced by marine algae of the genus Pseudo-nitzschia. We have developed a portable surface plasmon resonance (SPR) biosensor system for the detection of domoic acid. Because of concerns with domoic acid contamination of shellfish, there is a need for rapid field quantification of toxin levels in both shellfish and seawater. Antibodies were raised against domoic acid and affinity purified. These antibodies were used to develop competition- and displacement-based assays using a portable six-channel SPR system developed in our laboratories. Standard curves for detection of domoic acid in phosphate buffered saline and in diluted clam extracts analyzed by the competition-based SPR assay demonstrated a limit of detection of 3 ppb (10 nM) and a quantifiable range from 4 to 60 ppb (13–200 nM). Comparison of analyses for domoic acid levels in Pacific razor clams, Siliqua patula, containing moderate to high levels of domoic acid by the standard HPLC analysis protocol and the SPR-based assay gave an excellent correlation. This same technology should also function for detection of domoic acid in concentrated algal extracts or high dissolved levels in seawater.  相似文献

Ecological impacts of vehicle traffic are a significant environmental management issue on many sandy shores. Impacts usually focus on lethal effects of vehicles to organisms, but sub-lethal effects which could reduce the fitness of macrofauna populations are equally possible but unknown. Consequently, we measured changes in body condition and burrowing performance of the beach clam Donax deltoides subjected to vehicle traffic on sandy shores in eastern Australia. Body mass index of clams on beaches open to traffic was 16% lower, but gonadosomatic index and relative valve thickness were not consistently linked to vehicle access to beaches. By contrast, off-road vehicles significantly impaired the burrowing performance of clams. After experimental exposure to ORV traffic (30 passes) and dislodgement from the sediment, the time taken for clams to re-bury into the sand doubled irrespective of the vehicle weight used. Because burrowing is such a critical function in the behavioural repertoire of sandy beach animals, the traffic-induced changes to organisms' performance found in this study may increase mortality by causing displacement to less favourable habitats by swash, and by intensifying the risk of predation and desiccation. When assessing the ecological impacts of vehicles on beach fauna, it is thus important to consider both lethal and sub-lethal effects.  相似文献

The lady crab (Ovalipes ocellatus) is one of the most common native species of swimming crab (Portunidae) of the Atlantic Coast of North America but most populations occur south of Cape Cod, Massachusetts. There is a disjunct population in Northumberland Strait (southern Gulf of St Lawrence), which was the focus of this study. Adult lady crabs were collected by trawling in water >4 m deep from May to October 1999 to 2005 to determine abundance, distribution, and diet. Lady crab occurred only in a small area (about 2,500 km2) in the central part of Northumberland Strait where bottom water temperature was >18°C during summer, and the substrate was mainly sand or sandy gravel. Male lady crab attained a maximum carapace width (CW) of 112 mm compared to 92 mm CW for females. The summer and autumn diet consisted mainly of infauna. The principal prey (each >5% of diet by weight) were: small bivalve molluscs (primarily Atlantic razor clam Siliqua costata and Macoma sp.; 43%), small rock crab (Cancer irroratus; 13%), polychaetes (11%), fish remains (9%), and small lady crab (9%). All stomachs collected during May (water temperature ≤10°C) were empty. There was little evidence of any difference in feeding intensity between 0700 h and 1900 h.  相似文献

The effects of particle concentration and season on the filtration rates of the freshwater clamSphaerium striatinum Lamarck were assessed by measuring clearance rates of small (2.02 μm) latex beads from dilute suspensions. Filtration rates decreased as particle concentration increased over a range of 2–64 mg 1−1, with rates decreasing in similar proportion for clams of all sizes. For a 1-mg clam, rates decreased from approximately 8.4 to 0.57 ml clam−1 h−1. Seasonal filtration rates for adult clams peaked during periods of greatest reproduction. The patterns for smaller clams are similar, though proportional changes in filtration rates differ for various sizes of clams. It is estimated that clams occupying 1 m2 of stream substrate removed about 3.67 gCa−1. This is equivalent to 0.0004% of the carbon that flows past them annually. Filter-feeding provided only 24% of the calculated energy needs of the population, suggesting that another mode of feeding (e.g. deposit-feeding) may provide an important energy source for these forms. Management responses to reconcile declining fisheries typically include closed areas and times to fishing. This study evaluated this strategy for a beach clam fishery by testing the hypothesis that changes in the densities and size compositions of clams from before to during harvesting would differ between commercially fished and non-fished beaches. Sampling was spatially stratified across the swash and dry sand habitats on each of two commercially fished and two non-fished beaches, and temporally stratified across three six-week blocks: before, early and late harvesting. Small-scale spatio-temporal variability in the densities and sizes of clams was prevalent across both habitats and the components of variation were generally greatest at the lowest levels examined. Despite this, differences in the densities and sizes of clams among individual beaches were evident, but there were few significant differences across the commercially fished versus non-fished beaches from before to during harvesting. There was no evidence of reduced densities or truncated size compositions of clams on fished compared to non-fished beaches, contrasting reports of some other organisms in protected areas. This was probably due to a combination of factors, including the current levels of commercial harvests, the movements and other local-scale responses of clams to ecological processes acting independently across individual beaches. The results identify the difficulties in detecting fishing-related impacts against inherent levels of variability in clam populations. Nevertheless, continued experimental studies that test alternate management arrangements may help refine and determine the most suitable strategies for the sustainable harvesting of beach clams, ultimately enhancing the management of sandy beaches.  相似文献

Since 1998, king scallops (Pecten maximus) obtained from Scottish offshore sites have been monitored for domoic acid (DA) and epi-domoic acid (epi-DA), the principal toxic compounds associated with amnesic shellfish poisoning (ASP). The presence of these toxins in king scallops harvested from Scottish waters at concentrations exceeding the current regulatory limit (20 μg g−1 shellfish flesh) is a recurrent event. However, little information was available to determine the effects that different storage conditions experienced during sample transportation to the monitoring laboratory may have on the toxin concentrations, which are subsequently detected. Furthermore, the stability of DA and epi-DA in the solvents (methanol:water (1:1, v/v) and citric acid buffer (0.5 M, pH 3.2)) routinely used for their extraction from shellfish has not previously been assessed. Results from this study demonstrate that when king scallop samples were stored for 2–3 days at 12 °C, a significantly higher toxin concentration was detected in the gonad than when samples were stored at 4 °C and analysed within 48 h. This implies that monitoring programmes must consider transport and storage conditions between harvest and analysis. Stability studies showed rapid decomposition of DA and epi-DA in aqueous methanol extracts while DA and epi-DA seem acceptably stable when stored refrigerated in citrate buffer.  相似文献

Declining Chesapeake Bay harvests of softshell clams, together with historical and emerging reports of epizootic diseases in Mya arenaria, prompted a survey in summer 2000 of the health status of selected commercial clam populations. All sampled populations (8 M arenaria softshell clam, 2 Tagelus plebeius razor clam) were infected by Perkinsus sp. protozoans at prevalences ranging from 30 to 100% of sampled clams. Nucleotide sequences for the internal transcribed spacer (ITS) region of the rRNA gene complex were determined for clonal in vitro Perkinsus sp. isolates propagated from both M. arenaria and T plebeius. Multiple polymorphic sequences were amplified from each isolate, but phylogenetic analysis placed all sequences into 2 clades of a monophyletic group, which included both recently described clam parasites P. chesapeaki and P. andrewsi. Sequences amplified from each clonal isolate were found in both sister clades, one containing P. andrewsi and the other P. chesapeaki. Most (7 of 8) M. arenaria samples were also affected with disseminated neoplasia (DN), at prevalences of 3 to 37%, but neither T. plebeius sample showed DN disease. Disease mortalities projected for sampled clam populations, especially those affected by both diseases, may further deplete subtidal commercial clam populations in mesohaline portions of Chesapeake Bay.  相似文献

Trends in wrack composition and biomass, and its relationship with the anthropogenic impact were studied along a coastal area in Nuevo Gulf (south Patagonia) in front of Puerto Madryn city. Beach-cast macroalgae composition was sampled from 1992 to 1999 in the Puerto Madryn beaches and in several other nearby beaches in 1993, 1996 and 1998. Historical information was based on local knowledge and observations reported by marine biologists who worked in the area. The botanical composition of the beach-cast macroalgae in Puerto Madryn indicates a succession in the dominance from Codium spp. to Ulva spp. during the 1990s and from Ulva to Undaria pinnatifida since 1998, accompanied by a significant decrease in biomass of Gracilaria gracilis and Macrocystis pyrifera. The increase of the opportunist species such as Ulva may be supported by the continuous delivery of waste waters into the Nuevo Gulf while the dominance of U. pinnatifida may be associated with port activities. During the sampling period the highest wrack biomass values were recorded in spring and summer. The beach-cast seaweed biomass harvested by the municipality of Puerto Madryn during beach cleaning operation, ranged between 2500 and 12000 t year–1 ( 200 and 960 t dry weight). Wrack harvesting produces an environmental impact by removing sand from the beach and affecting coastal communities. Composting of wrack is proposed as one of the environmental alternatives to land disposal.  相似文献
