The effect of dietary selenium yeast, a source of organic selenium, on heat shock protein 70 (hsp70) responses, redox status, growth and feed utilization were evaluated either in enteropathogenic Escherichia coli-challenged (EPEC) or in heat-stressed (HS) male broiler chickens grown to 42 days of age. One day-old chicks in experiment 1 were challenged orally with EPEC (10(6) cfu/chicken on day 1 and boosted by water application on days 2, 3, and 4) and fed diets with or without selenium yeast. Body weight (BW), feed conversion ratio (FCR), and total mortality were determined at 42 days of age, and this was followed by collection of ileal tissue for the quantification of total glutathione (TGSH), reduced glutathione (GSH), oxidized glutathione (GSSG), and hsp70 in randomly selected chickens from each treatment. In experiment 2, male broiler chickens were fed diets with or without selenium yeast under a thermoneutral rearing condition. At four weeks of age, blood and hepatic tissue were collected from chickens maintained in the thermoneutral environment and from chickens subjected to HS (40 degrees C for 1 h) and analyzed for TGSH, GSH, GSSG, and hsp70. Selenium yeast improved BW, FCR, and decreased mortality in both control and EPEC-challenged chicks. Selenium yeast significantly attenuated hsp70 expression in EPEC-challenged chickens and in those subjected to HS. The EPEC challenge increased TGSH and GSSG levels and decreased GSH/GSSG ratio. However, GSSG level accumulated in chickens fed diets without selenium supplementation resulting in a lower GSH/GSSG ratio in the selenium yeast-fed group. Heat stress increased GSSG level and decreased GSH/GSSG ratio. Selenium yeast-fed groups maintained higher levels of GSSG before and after HS with a resultant lower GSH/GSSG ratio. The hsp70 response was significantly less in those chickens fed selenium yeast and challenged with either EPEC or HS than in those chickens given no supplemental selenium. The results of this study suggest that selenium yeast supplementation had imparted resistance to oxidative stress associated with enteric bacteria infection and to high temperature exposure. It is believed that the resistance to the stressors was due to an improved redox status of the selenium yeast-fed chickens. 相似文献

A feeding experiment was carried out to compare the effects of supplementing a poultry meal-based diet with selenium as sodium selenite or selenium yeast on broiler chickens. Three groups with three replicates of broiler chickens (mean weight 710 ± 5.3 g) were given a basal diet either unsupplemented (control) or supplemented with 0.2 mg Se kg−1 as sodium selenite (trial 1) or selenium yeast (trial 2) respectively, for 21 days. There was significant difference (P<0.05) in Feed Conversion Ratio (FCR) of trials 1 and 2 compared with the control. However, there were no significant differences (P>0.05) in FCR between trials 1 and 2. Final weight, survival rate and Daily Gain (DG) were not affected by the dietary Se source. Chickens fed the basal diet showed lower (P<0.05) selenium content in muscle, kidney, liver and pancreas compared to that fed selenium supplements (trials 1 and 2). Furthermore, trial 2 showed the highest value (P<0.05) among these treatments. However, there was no significant difference (P>0.05) in muscle selenium content of chickens between trials 1 and 2. Glutathione peroxidase (GSH-Px) activities in broiler chickens plasma and liver of all selenium treatment groups (trials 1 and 2) were significantly different (P<0.05) from that of the control. The GSH-Px activity in plasma was higher (P<0.05) in trial 2 compared with trial 1 and the control. However, there was no difference (P>0.05) in hepatic glutathione peroxidase between trials 1 and 2 although the average value of GSH-Px activity in trial 2 presented the trend of increase. 相似文献

Heat shock protein 70 (hsp70) family of proteins, which functions as molecular chaperones, has been associated with tolerance to stressors in avian species. Selenium (Se) is an essential trace mineral incorporated into the seleno-enzymes such as glutathione peroxidase (GSHpx). GSHpx reduces oxidized glutathione (GSSG) to reduced glutathione (GSH) in the GSH/GSSG antioxidant system and protects cells from oxidative damage. This study was conducted to examine if the relationship between dietary supplementation of selenium to turkey (Meleagris gallopavo) hens and the embryonic expression of hsp70 and GSHpx activity in heat stressed embryos. Livers of embryos developing in eggs from turkey hens fed diets with or without supplemental Se were analyzed for hsp70 concentration and GSHpx activity before and after recovery from a heating episode. Before heat stress, hsp70 concentrations were equivalent in each treatment, but GSHpx activity was maximized in the SE treatment group. After recovery from the heating episode, hsp70 concentrations were significantly higher (P<0.05) in the non-Se-supplemented groups, but in the Se-supplemented groups the hsp70 concentrations were not different from pre-stress concentrations. In the pre-stress Se-supplemented group, liver GSHpx activity was significantly higher than GSHpx activity in the non-Se-supplemented embryo livers, and in the livers from embryos recovering from heat stress, GSHpx activity in the non-Se-supplemented group was lower than the pre-stress activity and significantly lower than the GSHpx activity in liver from Se-supplemented embryos recovering from heat distress. Se supplementation to the dams resulted in a significant increase in their embryos and that condition would facilitate a decreased incidence of oxidative damage to cells. A more reduced redox status in embryos from Se-supplemented dams decreased the need for cellular protection attributed to stress induced hsp70 and presumably allows heat distressed embryos to resume normal growth and development than embryos from dams with inadequate selenium nutrition. 相似文献

Effect of heat stress and drinking water salt supplements on plasma electrolytes and aldosterone concentration in broiler chickens 总被引:3,自引:0,他引:3
An experiment was conducted to evaluate the effects of supplementing drinking water with isomolar (0.067 mol/l) KCl or NaCl on mass gain, food and water consumption, rectal temperature, and plasma concentrations of aldosterone, Na+, and K+ in broiler chickens reared in thermoneutral and cycling heat stressing environments. Heat stress decreased (P0.05) mass gain, food consumption, and plasma concentrations of Na+ and K+, while increases (P0.05) in plasma concentrations of aldosterone, rectal temperature, and water consumption were observed. Drinking water supplemented with either KCl or NaCl increased (P0.05) broiler mass gain and water consumption, but had no effect (P>0.1) on the other variables evaluated. The results of this study indicate that broiler chickens in a heat stress environment are under osmotic stress and supplementing drinking water with 0.067 mol/1 KCl or NaCl does not lessen this stress. 相似文献

Two trials were conducted to evaluate the effects of short-term administration of corticosterone (CORT) on the induction of oxidative injury in broiler chickens (Gallus gallus domesticus). Twelve broiler chickens of 30 and of 40 days of age were respectively employed in Trial 1 and 2. Half of the chickens were administered subcutaneously with CORT (4 mg/kg body weight [BW] in corn oil), while another half served as controls (corn oil) in each trail. In Trial 1, a blood sample was obtained from each chicken immediately before administration and at 1 and 3 h after injection. In Trial 2, the liver and heart were obtained after 3 h of CORT exposure. Short-term administration of CORT resulted in enhanced proteolysis and gluconeogenesis. There were no obvious changes in lipid peroxidation status of the heart and liver, whereas a decrease in lipid peroxidation in the plasma was observed after acute CORT exposure. The significantly increased plasma nonenzymatic antioxidants (uric acid [UA] and total antioxidant capacity) in concert with the enhanced enzymatic antioxidant activity (SOD in heart) during short-term CORT administration indicate preventive changes to counteract the oxidative injury, and these may be tissue specific. 相似文献

Present study was carried out with the objective of investigating the role of green synthesized nano Se (GNS) in growth performance, digestibility of minerals, immunity, stress alleviation, antioxidant status, and body Se content of broiler chicken raised under hot and humid environment with respect to market nano Se (MNS) and inorganic Se. The experimental design was 3 × 3 factorial, in which three levels (0.15, 0.20, and 0.25 ppm) and three sources (inorganic, green nano, and market nano) of Se resulted in nine treatments viz. IS-0.15, GNS-0.15, MNS-0.15, IZ-0.20, GNS-0.20, MNS-0.20, IS-0.25, GNS-0.25, and MNS-0.25 (IS: inorganic Se, GNS: green nano Se, MNS: market nano Se). A total of 432 broiler chicken were divided among nine treatments with six replicates of birds per treatment (8 birds/replicate). Results of present study revealed significantly better growth performance of birds supplemented with 0.25 ppm nano Se. The supplementation of 0.25 ppm nano Se improved the immune response and lymphoid organ development of birds. Significantly higher Se and nitrogen digestibility coefficients, serum antioxidant activity and decline of Heterophil: Lymphocyte ratio and expression of HSP70 gene were observed in birds supplemented with 0.25 ppm Se and nano source of Se compared to inorganic Se. Significantly higher Se concentration in liver and breast muscle and higher serum Se concentration were observed in birds fed 0.25 ppm nano Se. The liver Se concentration was much higher than that of breast muscle. However, the nano Se synthesized by green method in this study did not differ significantly from the chemically synthesized nano Se. It was concluded that 0.25 ppm Se and nano form of Se are superior to lower levels and inorganic form of Se, respectively, in improving the immunity, growth, antioxidant status, and in stress alleviation of broiler chicken. In order to evaluate the metabolic responses of laying hens induced by high temperature at later laying stage, nine 60-wk-old laying hens (Gallus gallus domesticus) were employed in the present study. The hens were exposed to 32 degrees C for 21 d and blood samples were obtained before and at 1, 7, 14 and 21 d of heat exposure. The reactive oxygen species (ROS) formed in blood during heat exposure were estimated by the ex vivo spin-trapping method. Body temperature and plasma concentrations of glucose, urate, creatine kinase (CK), triiodothyronine (T(3)), thyroxine (T(4)), corticosterone (CORT), thiobarbituric acid reacting substances (TBARS), ferric/reducing antioxidant power (FRAP) and superoxide dismutase (SOD) activity were measured. Plasma levels of glucose, CK and CORT were not significantly influenced by heat exposure at any time point. The circulating concentrations of T(3) were decreased while plasma T(4) levels changed in the opposite way. The formation of ROS was significantly augmented by heat exposure in laying hens though the body temperature was not significantly altered. The enhanced enzymatic and non-enzymatic antioxidant systems acted in concert to alleviate the heat stress evoked oxidative damage. 相似文献

This paper describes the effects of ambient temperatures on whole blood viscosity and plasma protein concentration in broiler chickens. Whole blood viscosity and haematocrit, compared at 7 and 20°C, decreased significantly at or above 25°C of ambient temperature. However, no marked difference were found between 7 and 20°C or between the temperatures of 25, 30 and 35°C. The heat-induced decrease of whole blood viscosity was found after exposure for 1 h. These results suggest that the heat-induced decrease in blood viscosity is only a level change, which occurs between thermoneutral and high ambient temperature. 相似文献

BackgroundHigher environmental temperature is a major abiotic stress factor for animals and human beings. The selenium (Se) is an important trace mineral having diverse health promoting effects under stress conditions. However, studies on dietary requirement of selenium under prolonged heat stress condition are lacking. Present study discern the effect of higher dietary Se levels on antioxidant, cytokine, haemato-biochemical profile, and immune response, and the selenoproteins mRNA expression in rats under prolonged heat stress (HS) condition.MethodsWeaned Wistar rats (4 wk age; 67.6 ± 1.53 g BW; n = 72) housed under thermoneutral (TN) or HS conditions and fed with purified diets containing three graded Se levels were divided in six experimental groups. The groups were 1) TN control with 138 ppb Se (TN_CON), 2) HS control with 138 ppb Se (HS_CON), 3) TN with higher Se @ 291 ppb (TN_Se1), 4) HS with higher Se @ 291 ppb (HS_Se1) 5) TN with higher Se @ 460 ppb (TN_Se2), 6) HS with higher Se @ 460 ppb (HS_Se2). Rats in all the six groups were maintained in TN environmental conditions (57.3 ± 0.22 temperature humidity index; THI) for initial 28 days period. Subsequently, rats of HS groups were exposed to 77.0 ± 0.11 THI for 6 h/d in a psychrometric chamber for last fourteen days.ResultsHigher dietary Se (291 and 460 ppb) significantly improved the blood hemoglobin concentration and reduced serum alanine aminotransferase activity of rats under HS conditions. The serum triiodothyronine and insulin levels were significantly higher in high dietary Se groups irrespective of the environmental conditions. Similarly, the serum reduced glutathione levels, and catalase and superoxide dismutase enzyme activity were increased and malondialdehyde levels were reduced in high dietary Se groups irrespective of stress conditions. The glutathione peroxidase (GPx) activity was significantly higher in 460 ppb dietary Se groups as compared to other groups. The serum pro-inflammatory cytokine interleukin (IL)− 1 was declined, whereas the anti-inflammatory cytokine IL-10 level was increased in high dietary Se fed rats under both HS and TN conditions with 460 ppb dietary Se groups showing pronounced effects. Further, there was heat stress- and dietary Se level dependent- up regulation in hepatic GPx and iodothyronine deiodinase-II mRNA expression and similar pattern was noticed in hepatic thioredoxin reductase mRNA expression. The selenoprotein-P mRNA expression was up regulated in 460 ppb Se fed HS group as compared to CON and Se1_C groups. High dietary Se improved the humoral immune response 7d after antigen inoculation under HS conditions whereas cell-mediated immune response was augmented in rats fed higher Se under TN condition.ConclusionIt is concluded that under prolonged heat stress conditions the dietary requirement of Se may be increased to 460 ppb for improving the antioxidant status and humoral immune response, cytokine levels, modulating the thyroid and insulin hormone, and the selenoproteins mRNA expression of rats. 相似文献

The objective of this study was to investigate the kinetics of Hsp60, Hsp70, Hsp90 protein, and messenger RNA (mRNA) expression levels and to correlate these heat shock protein (Hsp) levels with tissue damage resulting from exposure to high temperatures for varying amounts of time. One hundred broilers were heat-stressed for 0, 2, 3, 5, and 10 h, respectively, by rapidly increasing the ambient temperature from 22 +/- 1 degrees C to 37 +/- 1 degrees C. Obvious elevations of plasma creatine kinase indicate damage to myocardial cells after heat stress. Hsp70 and Hsp90, and their corresponding mRNAs in the heart tissue of heat-stressed broilers, elevated significantly after 2 h of heat exposure and decreased quickly with continued heat stress. However, the levels of hsp60 mRNA in the heart of heat-stressed broilers increased sharply (P < 0.01) at 2 h of heat stress but then decreased quickly after 3 h, while the level of Hsp60 protein in the heart increased (P < 0.01) at 2 h of heat stress and maintained a high level throughout heat exposure. The results indicate that the elevation of the three Hsps, especially Hsp60 in heart, may be important markers at the beginning of heat stress and act as protective proteins in adverse environments. The reduction of Hsp signals in the cytoplasm of myocardial cells implies that myocardial cell lesions may have an adverse impact on the function of Hsps during heat stress. Meanwhile, the localization of Hsp70 in blood vessels of broiler hearts suggests another possible mechanism for protection of the heart after heat exposure. 相似文献

Effects of commercial probiotic (Bactocell®) on growth performance and blood parameters were evaluated. A total of 800 one day-old Ross broiler chicks were raised over 42 days. Chicks were wing-banded, weighed individually and randomly allocated into four equally major groups each having two replicates. Chicks of group 1 (control group) were fed the starter and finisher diets that did not supplemented with probiotic. The chicks of groups 2, 3, and 4 were fed the control starter and finisher diets supplemented with 1.6 g, 1 g and 0.8 g of probiotic per kg feed, respectively. Weekly body weight, feed consumption and feed conversion were measured. Blood parameters at 1, 4 and 6 weeks of age including packed cell volume (PCV), haemoglobin (Hb), total protein, albumin, total lipid and cholesterol were determined. All birds were kept under similar environmental, managerial and hygienic conditions. The results of the current study revealed that there was no significant change for Hb and PCV concentrations among different groups at all studied times. Also, total protein, lipids and albumin concentrations were not affected by probiotic supplementation. Chicken fed a diet containing various levels of probiotic showed a significant decrease (p ⩽ 0.05) in cholesterol concentration compared to control group. Probiotic supplementation significantly increased the body weight and daily weight gain of broiler chicks at late ages (3–6 weeks). Also, the birds fed on probiotic levels 1 and 0.8 g/kg diet exhibited higher body weight among chicken groups at 6 weeks of age. Improved feed conversion was noticed in birds fed a diet supplemented with probiotic. There was no significant difference in mortality rate among groups. We concluded that use of selected commercial probiotic resulted in improved performance parameters and reduced serum cholesterol in broiler chickens. Moreover, supplementation of the probiotic to broilers in the levels of 1 and 0.8 g/kg diet was found to be better than control and 1.6 g/kg level indicating that increasing dietary probiotic level does not has the best performance. 相似文献

This experiment was conducted to evaluate the effects of zinc (ZnSO4H2O) and vitamin A (retinol) supplementation on performance, carcass characteristics, and serum concentrations of glucose, cholesterol,
total protein, and malondialdehyde (MDA) as an indicator of lipid peroxidation in broiler chickens (Ross) reared at a high
temperature (34°C). One hundred twenty 10-d-old male broilers were randomly assigned to 4 treatment groups, 3 replicates of
10 birds each. The birds were fed either a basal diet or the basal diet supplemented with either 30 mg Zn/kg diet, 4.5 mg
(15,000 IU) retinol/kg diet, or 30 mg Zn+4.5 mg retinol/kg diet. Supplemental zinc and vitamin A significantly increased live
weight gain and improved feed efficiency (p<0.05). However, a combination of zinc and vitamin A, rather than each separately, provided a greater performance. Hot and
chilled carcass weights and yields and the weights of internal organs with the exception of abdominal fat were greater for
each supplement (p<0.05) compared to the control group. Abdominal fat decreased (p<0.05) upon dietary zinc and vitamin A supplementation. Supplemental treatments resulted in an increased total serum protein
but decreased glucose, cholesterol, and MDA concentrations. The results of the study show that, separtely or as a combination,
zinc and vitamin A supplementation resulted in an improved live weight gain, feed efficiency, and carcass traits, as well
as a decrease in serum MDA concentrations. The results of the present study also suggest that zinc and vitamin A have similar
effects and that a combination of zinc and vitamin A may offer a potential protective management practice in preventing heat-stress-related

Two trials were conducted to investigate the effect of corticosterone (CORT) on protein metabolism and the amino acid composition in muscle tissues of broiler chickens (Gallus gallus domesticus). In Trial 1, two groups of 30 broiler chickens were subjected to control or CORT treatment (30 mg/kg diet) from 28 to 39 days of age. In Trial 2, three groups of chickens of 28 days of age were randomly subjected to one of the following treatments for 7 days: CORT (30 mg/kg diet), pair-fed (maintaining the same feed intake as CORT treatment) and control treatments. The body mass gain and feed efficiency was significantly decreased by CORT treatment, while the food intake was decreased. The breast and thigh masses (% body mass) were significantly suppressed by CORT treatment, while the abdominal fat and liver masses (%) were obviously increased. The plasma levels of glucose, urate and total amino acid were significantly elevated by CORT treatment. The capacity for protein synthesis, estimated by RNA:protein ratio, were significantly suppressed by CORT in M. pectoralis major and M. biceps femoris. The 3-methylhistidine concentrations were significantly increased in both M. pectoralis major and M. biceps femoris of CORT chickens, compared to control but not the pair-fed chickens. The amino acid composition of M. pectoralis major and M. biceps femoris was not significantly affected by CORT treatment. In conclusion, the arrested growth in skeletal muscles induced by CORT administration has tissue specificity. The CORT treatment retards the growth of skeletal muscle by suppressed protein synthesis and augmented protein catabolism. 相似文献

An investigation was carried out to verify whether the heat stress hyperthermia response of broilers is prostaglandin-dependent.
Male broiler chickens of the Hubbard-Petterson strain, aged 35–49 days, were used. Chickens were injected with indomethacin
(1 mg/kg intraperitoneally ) 15 min before or 2 h after heat exposure (at 35°C for 4 h), and rectal temperature was measured
before injection and up to 4 h thereafter. Birds were separated into two groups with and without access to water during heat
stress. The increase in rectal temperature was lower (P<0.05) in birds with access to drinking water during heat exposure. All birds injected with indomethacin exhibited an increase
in rectal temperature, irrespective of whether indomethacin was administered before or in the course of the rise in temperature.
The results revealed that the increase in rectal temperature during heat exposure is not prostaglandin-dependent, and that
the use of cyclooxigenase inhibitors is not recommended to attenuate heat stress hyperthermia in broiler chickens.

Different toxic doses of selenium and monensin preparations were administered to broiler chickens. The two substances were given by oral route, alone or concurrently, for variable periods. Erythrocyte glutathione peroxidase. (GSH-Px) activity was found to be elevated after the administration of the drugs. This increase was considerably higher when selenium and monensin were administered concurrently, indicating the occurrence of strong interaction between them. Administration of selenium led to a rapid increase in the liver selenium concentration. This increase, in turn, was enhanced by concurrent application of monensin. Monensin given alone did not have any significant effect on the changes of liver selenium concentration. Further results suggest that administration of monensin increases erythrocyte GSH-Px activity, even in the absence of supplemental selenium or during increased liver selenium concentration. 相似文献
