The steady-state fluxes of Na+, K+, and NH4+ carried by nonactin and trinactin across thin lipid membranes have been measured as functions of ion activity, carrier concentration, and the applied potential. In agreement with earlier studies the conductance, G(O), is found to be proportional to the carrier concentration and, for low activities, to the ion activity. The determination of the dependence of G(O) on activity at high activities is, however, apparently obscured by changes in the concentration of carrier in the membrane. Using the values for the rate constants at zero potential which were determined in the preceding paper, it is possible to adjust the potential dependence of the constants so as to achieve a reasonable fit to the current-voltage relations. The data presented provide further evidence that a single molecule of nonactin or trinactin acts cyclicly as a carrier of univalent cations.  相似文献

For trinactin the data presented allow the estimation of the rate constants of the carrier process (in the limit of zero potential) in a manner which does not assume any particular variation with potential for the constants. Using empirically determined functions of potential, a complete set of values is also available for nonactin. The curve fitting which is necessary is described in the following paper. The experimental steady-state current-voltage relations for low concentrations of a neutral carrier and an ion may be fitted theoretically either by assuming a form for the potential dependence of the rate of transfer of complex across the membrane and adjusting the proposed nature of the association-dissociation reactions or by assuming equilibrium for the association and adjusting the potential dependence of the transfer process. Different dependences for the rate of transfer correspond, at least formally, to different shapes for the potential energy barrier which the complex must cross. By comparing measurements of the current-voltage relations for non-actin with Na+, K+, and NH4+, it is possible to distinguish between the effeects of the various rates. For black lipid membranes made from glycerolmonooleate+n-hexadecane, the potential energy barrier is high with a narrow top, but the rate of association still becomes increasingly limiting for Na+, K+ and NH4+, in the order given. For bacterial phosphatidylethanolamine, with n-decane the barrier is much wider and no effect of the rate of association can be detected.  相似文献

The conductance of black lipid membranes in the presence of 2,4,6-trinitrophenol (or 2,4-dinitrophenol) is considerably enhanced, if the cation carriers valinomycin, enniatin B or nonactin are added. The effect is, however, largely independent of the cation concentration and is identical for the cations Li+, Na+ and Ba2+. This finding, as well as the sign and magnitude of the diffusion potential in the presence of a gradient of picrate are consistent with the assumption that the transport of picrate anions is facilitated by the above-mentioned macrocyclic compounds, but that cations are not directly involved. A model is suggested which, based on the generation of mobile defect structures by the incorporation of large molecules, allows one to explain facilitated transport without the assumption of stable chemical bonds between a carrier and its transported substrate.If K+ is present in the aqueous phase, the conductance is largely determined by the permeation of the cation complexes of valinomycin and nonactin. The conductance is, however, increased by adsorption of picrate anions to the membrane surface. The negative surface potential generated by the adsorption layer seems to be responsible for the saturation of the conductance at high picrate concentrations in the absence of valinomycin and nonactin.  相似文献

Resealed chromaffin-granule ;ghosts' were used to study the steady-state kinetics of catecholamine transport. The pump has a high affinity for (-)-noradrenaline, (-)-adrenaline, tyramine and 5-hydroxytryptamine (serotonin), but a lower affinity for (+)-noradrenaline. The measured rates of incorporation do not conform to Michaelis-Menten kinetics, but affinity constants for the former substrates are in the range 8-18mum. Reserpine is a potent inhibitor. Incorporation as a function of ATP concentration also fails to show simple kinetics; the affinity constant for ATP is deduced to be about 3mm at 1mm-MgCl(2). Adenylyl (betagamma-methylene)diphosphonate is a competitive inhibitor at low concentrations, but inhibits more strongly at high concentrations. The pump has a transition temperature at 29 degrees C and does not seem to be identical with the Mg(2+)-stimulated adenosine triphosphatase of chromaffin granules.  相似文献

1. Conditions of incubation of everted sacs of rat small intestine were selected to ensure that absorption of d-glucose by mucosal tissue from the incubation medium, intracellular metabolism of the absorbed glucose and transport of glucose through the intact intestinal tissue proceeded linearly with respect to time of incubation within stated time intervals. 2. Under these experimental conditions, steady intracellular concentrations of glucose and lactate were demonstrated. 3. The quantitative translocational and metabolic fate of absorbed glucose was determined under these steady-state conditions. About 25% of glucose absorbed from the external mucosal solution was accumulated (temporarily) within mucosal tissue and about 25% transported through the intact tissue into the external serosal solution; the remainder (about 50%) of the absorbed glucose was metabolized, 90% to lactate and 10% to CO2. Concomitant respiration rates were comparable with those reported for several other preparations of intestine and were stoicheiometrically in excess of the O2 metabolism required to account for the production of CO2 from the absorbed glucose. 4. Water transport through the everted sacs proceeded at an optimum rate under the experimental conditions selected. 5. Some other observations are recorded which influenced the design of the experiments and the interpretation of results; these include the initial physiological state of the animal, the anaesthetic used and the ionic composition of the incubation medium.  相似文献

Activation of potassium ion transport in mitochondria by cadmium ion   总被引:2,自引:0,他引:2  
Low levels of Cd2+ (1-5 microM) produce rapid swelling of mitochondria, which is respiration-dependent and uncoupler-sensitive. No cation requirement is apparent, since the swelling occurs in a medium containing only sucrose and the respiratory substrate. The swelling is inhibited by ruthenium red, suggesting that this effect of Cd2+ requires its entry into mitochondria. In medium containing 9 mM K+, addition of Cd2+ along with ruthenium red increases the rate of K+ influx threefold. In the presence of K+, Rb+ or Li+, but not of Na+, addition of Cd2+ produces first efflux of H+ into the medium followed by discharge of the pH gradient or uncoupling. Only the latter effect is inhibited by ruthenium red, showing that the efflux and influx of H+ are independent reactions. The H+ efflux appears to be an antiport response to the induced K+ entry. Its activation by Cd2+ is similar to the known effect of p-chloromercuriphenyl sulfonate. The H+ influx or uncoupling appears to result from binding of Cd2+ to some matrix-facing membrane site, perhaps the dithiol group on coupling factor B, and may relate to apparent permeability changes associated Cd2+-induced swelling.  相似文献

4-Hydroxy-2,3-trans-nonenal (HNE), a major lipid peroxidation product, has been shown to react with specific amino acid residues of proteins and alter their function. In vitro exposure of erythrocyte ghosts and neutrophil membranes to HNE results in the inhibition of ion transport ATPases. Neutrophil membrane Ca2+-ATPase is strongly inhibited by micromolar concentrations of HNE, while HNE is considerably less effective against neutrophil Mg2+-ATPase and the erythrocyte ghost enzymes.  相似文献

It is a misconception to think that extracellular electric currents of biologic origin are carried by all of the ions in the extracellular medium. It is established that, for certain reasonable boundary conditions, only those ion species that are transported across the plasma membranes of the biological system, or that chemically derive from or contribute to these species, can contribute to the extracellular electric current at steady state. In the absence of convection, the extracellular current will be carried largely by diffusion of the transported ion species, at steady state. Extracellular electric potential gradients are shown to arise in a secondary manner, as a result of the electroneutrality condition. Preincubating pig erythrocyte membranes with ATP enhances their ability to accumulate Ca2+ against a concentration gradient. The extent of this increase is dependent on preincubation time over the period 0–60min. As the accessibility of outside membrane markers is decreased by preincubation and as accumulated Ca2+ is not removed by EGTA [ethanedioxybis(ethylamine)tetra-acetate], it is suggested that ATP causes the formation of sealed inside-out vesicles which can transport Ca2+ inward. The transport system requires ATP and Mg2+ and exhibits an apparent dissociation constant for Ca2+ of approx. 100μm. Since the dissociation constant for Ca2+-sensitive ATPase (adenosine triphosphatase) in these preparations is similar, it is concluded that this ATPase is responsible for Ca2+ transport. Polyphosphoinositide concentrations are also increased during incubation with ATP; however, there is no change in their rate of synthesis or breakdown during Ca2+ transport.  相似文献

Suspensions of mitochondria are turbid and scatter light. An increase in the matrix volume (swelling) due to the influx of permeable solutes results in a decrease in the amount of light scattered. This property can be used to study solute fluxes across the mitochondrial inner membrane. A rapid method for isolating mitochondria is presented along with three swelling experiments using energized and non-energized mitochondria to illustrate ion transport across energy transducing membranes.  相似文献
