Fluorescent labeling of the flagellar apparatus of Tetraselmis (Prasinophyceae) and Dunaliella (Polyblepharidaceae, Chlorophyceae) were successfully performed using fluorescein isothiocyanate–labeled lectins from Arachis hypogaea and Glycine maxima. These lectins specifically bound to the flagella and kinetosome of the cell but did not bind to the cell surface. Lectin binding on the flagellar apparatus remained constant under different culture media, temperatures, irradiances, cell division cycle, and culture aging. All the Tetraselmis and Dunaliella analyzed (five species, 20 clones) showed intense labeling of the flagellar apparatus. In contrast, no other species analyzed (46 clones of 25 species from four classes) showed binding to their flagellar apparatus. After the lectin treatment, many cells remained alive, and they were able to swim with the flagellar apparatus intensely labeled. The lectin binding indicates that the flagella and kinetosome of Tetraselmis are rich in Gal and GalNH2 moieties and that the flagella of Dunaliella are rich in Gal and GalNAc moieties. Apparently, this feature seems to be specific to these species. 相似文献

Thirty-five green algal species, representing 17 orders in 5 classes, were examined for the presence of the xanthophyll loroxanthin by reversed-phase high-performance liquid chromatography. Of these, 16 possessed loroxanthin, including at least one member of each class of green algae. The distribution of loroxanthin as established by this and previous studies is disjunct within the Chlorophyta and does not appear to have any taxonomic significance. Most of the green algae examined possessed one or more xanthophyll pigments that have not been found in land plants. 相似文献

Isolated cell walls (thecae) from the scaly flagellate green alga Tetraselmis striata Butcher contain the unusual 2-keto-sugar acids 3-deoxy-manno-2-octulosonic acid (Kdo), 3-deoxy-5-O-methyl-manno-2-octulosonic acid (5OMeKdo), and 3-deoxy-lyxo-2-heptulosaric acid (Dha). In addition, galacturonic acid, galactose, gulose, and arabinose are present. EDTA-extraction yielded an insoluble fraction that retains the shape of the cell walls and contains no 2-keto-sugar acids. Methylation analysis demonstrated the presence of terminal hexose, GalA, Dha, and Kdo as well as 2-substituted hexose, 4-or 8-substituted Kdo, and 4,8-disubstituted Kdo. However, most of the carbohydrate material (about 60%) was not methylated. Periodate oxidation of the cell wall preparation showed the presence of 2-substituted Gul, 4- or/and 5-substituted and 7- or/and 8-substituted Kdo, which is in agreement with the methylation analysis. Again, a significant amount of carbohydrate material was not degraded, indicating complex substitution patterns. Oligosaccharides were generated by partial hydrolysis and fractionated using gel permeation chromatography and high-pH anion-exchange chromatography. Oligosaccharides contained either GalA and Kdo, or Gal, Kdo, Dha, and Gul, respectively. The structure of a GalA and Kdo containing disaccharide was established using 1 H NMR spectroscopy. 相似文献

The effects of mercury and cadmium on the intracellular level of nonproteinaceous thiols in a unicellular green alga Tetraselmis tetrathele (West) Butcher (Prasinophyceae) were investigated by using a fluorescent dye, 5-chloromethylfluorescein (5CMF), as a probe for nonproteinaceous thiols. The 5CMF fluorescence was observed in cytoplasm, and the intensity of the fluorescence was decreased by exposure of the cells to HgCl2 . Analysis of the fluorescent intensity of 5CMF by flow cytometry made it possible to distinguish cells in three states during the dying process caused by HgCl2 : a normal state, a thiol-depleted state, and a dead state. Depletion of nonproteinaceous thiols began within 30 min, and they were completely depleted at 2 h. Most cells died after 24 h of exposure to more than 3.0 μM HgCl2 , whereas exposure up to 1.0 mM CdCl2 did not cause depletion of nonproteinaceous thiols or cell death within 48 h. HPLC analyses revealed that glutathione was a major nonprotein thiol in T. tetrathele and that it was oxidized by exposing the cells to HgCl2 . Phytochelatins, which play a great role in the tolerance to heavy metals of higher plants and many algae, could not be found in T. tetrathele. However, a tripeptide, Arg-Arg-Glu, was found to be abundant, and it showed ability to bind Hg2+ , suggesting that it functions to scavenge heavy metals as well as thiol molecules. 相似文献

Marine planktonic algae are frequently exposed to metallic contaminants. Because heavy metals can be assimilated and accumulated by algal cells, they can then be transferred to higher trophic levels of food chains. We studied the effects of cadmium on protein production and the growth of the marine prasinophyte Tetraselmis gracilis (Kylin) Butcher. By means of toxicological assays, we estimated the LC50 of cadmium as 3.2 ppm and 1.8 ppm after 48 h and 96 h of exposure to this heavy metal, respectively. The growth curves and survival percentages of cell cultures in the presence of cadmium were determined, and a proportional reduction of both parameters with increasing metal concentrations was found. When chronically exposed to sublethal concentrations of cadmium, T. gracilis contained high levels of superoxide dismutase (SOD) activity, one of the main enzymes of the cell's antioxidant defense mechanism. Under these growth conditions, total SOD activity in crude extracts was increased by 41% (at 1.5 ppm) and 107% (at 3.0 ppm). Assays of SOD activity in nondenaturing polyacrylamide gels also showed a similar induction by cadmium. These results show that cadmium has potentially toxic properties since it significantly inhibited the growth of T. gracilis at low concentrations and promoted the induction of SOD activity, suggestive of an oxidative stress state. Besides being the first report of SOD in T. gracilis, this work describes experimental evidence of SOD induction by cadmium in this species. 相似文献

Forty-seven species of desmids, representing all four families, were examined for the presence of the xanthophyll loroxanthin by reverse-phase high-performance liquid chromatography. In the Desmidiaceae 28 of the 35 species examined possessed loroxanthin, and in the Mesotaeniaceae two of the six examined had loroxanthin present. All six species of the families Peniaceae and Closteriaceae examined possessed loroxanthin. Although the distribution of loroxanthin appears to be disjunct in the desmids and does not have strict taxonomic significance, it does follow a coherent pattern consistent with current ideas on desmid phylogeny. This pattern suggests that loroxanthin synthesis probably evolved once in the desmid lineage, with one or more subsequent reversals. 相似文献

Phylogenetic analyses of 19 strains representing five species of Eudorina, one strain of Pleodorina indica, and seven strains of Yamagishiella unicocca were carried out by sequencing the internal transcribed spacer region (ITS 1 and ITS 2) of the nuclear ribosomal DNA (rDNA) repeats. The sequence data resolved five phylogenetic groups, one consisting of Y. unicocca and the other four encompassing all the Eudorina species. Two isolates, Eudorina sp. (ASW 05157) and Pleodorina indica (ASW 05153), were of uncertain affiliation. Whereas one monophyletic group included strains of E. elegans only, the other strains of E. elegans appeared alongside E. cylindrica, E. illinoisensis, and E. unicocca var. unicocca in the other Eudorina clades. The distribution pattern of the carotenoid loroxanthin ([3R,3'R,6'R]-β,ε-carotene-3,19,3'-triol), a systematically useful biochemical marker within chlorophycean flagellates, was shown to match the evaluated molecular data. Whereas it was either totally absent or universally present in six of the deduced phylogenetic lines, it occurred randomly in the E. elegans clade containing only E. elegans isolates. The results substantiated the current hypothesis that the unique vegetative morphology of E. elegans has independently arisen at various times during evolution and that it is not a marker of a monophyletic group. 相似文献

Ostreococcus tauri Courties et Chrétiennot-Dinet is the smallest described autotrophic eukaryote dominating the phytoplanktonic assemblage of the marine Mediterranean Thau lagoon (France). Its taxonomic position was partly elucidated from ultrastructure and high-pressure liquid chromatography (HLPC) pigment analysis. The sequence analysis of the 18S rDNA gene of O. tauri measured here is available in EMBL Nucleotide Sequence Database (accession number: Y15814) and allowed to clarify its phylogenetic position. O. tauri belongs to the Prasinophyceae and appears very close to Mantoniella, a typical scaly Prasinophyceae, morphologically very different from the naked and coccoid Ostreococcus. An electrophoretic analysis of O. tauri shows that the nucleus contains 10.20 mbp. This small genome, fragmented into 14 chromosomes ranging in size from 300 to 1500 kbp, confirms the minimalist characteristics of Ostreococcus tauri. 相似文献

Nephroselmis spinosa Suda sp. nov. is described based on LM and EM observations. Two strains of N. spinosa (S222 and SD959‐3) were isolated from sand samples collected from the northwest coast of western Australia. The cells were remarkably right–left flattened and appeared ellipse or bean‐shaped when viewed from their right or left side. A single, parietal, crescent chloroplast was pale green to yellowish green and contained one conspicuous eyespot in its anterior ventral edge near the base of the short flagellum. A pyrenoid with three starch plates was located at the dorsal of the chloroplast. The cells divided by transverse binary cell division, as is common in other species of this genus. This alga possessed four types of body scales, and three scale types were similar to N. olivacea Stein and N. astigmatica Inouye & Pienaar. However, the fourth and outermost scale type was distinctive because although it was a spiny stellate scale with nine spines, one of them extended about 1 μm and was slightly curved with a hook at the end. This scale morphology, an important taxonomic characteristic, has never been described for the genus Nephroselmis. The cell's morphology, pyrenoid structure, hair scales, and cell size were distinctive from previously described Nephroselmis species, and its unique scale characteristic led me to name this newly proposed species “spinosa,” meaning spiny. 相似文献

Diel changes in mean cell volume, cellular carbon (carbon content per cell), cellular Chl a, C/N ratio, Chl a/carbon ratio and pigment composition were determined for an axenic clonal culture of Pyramimonas parkeae Norris et Pearson through three 12:12 h LD cycles in a laboratory culture tank of 1 m3. Mean cell volume and cellular C, N and most pigments increased during the light period as a result of photosynthesis and decreased with an increase in cell density by phased cell division during the dark period. Chi a and Chi b increased in a parallel manner during the light period. Increases in the diel synthesis pattern of carotenoids varied. Violaxanthin and lutein content increased for a few hours at the beginning of the light period and preceeded that of neoxanthin. The diel synthesis pattern of neoxanthin was similar to that of Chi a. Increases of loroxanthin and its ester form were slower than that of Chi a at the beginning of the light period. A net increase of α-carotene was observed during the dark period. Mass spectroscopy of carotenoid structure showed a new xanthophyll, loroxanthin dodecenoate, in this species. 相似文献

A freeze fracture analysis of the cell wall of Tetraselmis convolutae (Parke et Manton) revealed the existence of a crystalline median layer consisting of regular repeating subunits of 27 nm. These circular subunits lie in curved, interlocking, longitudinal rows with some irregular discontinuities appearing in the subunit pattern of the crystalline lattice. A comparison with the cell walls of other green algal flagellates is presented, revealing similarities and suggesting evolutionary patterns. 相似文献

A new punctate species of Pyramimonas Schmarda from South Africa is described. This species is somewhat anomalous and, like P. chlorina Sym et Pienaar (subgenus Pyramimonas McFadden), has characters that bridge the divide between the subgenera Pyramimonas and Punctatae McFadden. Features of these two species, together with the lack of exclusive character sets for either subgenus derived from other species of these subgenera, lead to the conclusion that Punctatae now should be subsumed formally into the subgenus Pyramimonas. 相似文献

Carotenoid composition and spectroscopic characteristics were analyzed for Pterosperma cristatum Schiller, one of the most primitive known members of green algae. This alga contained a substantial amount of carotenoid esters, siphonaxanthin C14:1 trans‐Δ2 ester and 6′‐OH siphonaxanthin C14:1 trans‐Δ2 ester, but lacked lutein. This is the first report of carotenoid C14:1 trans‐Δ2 esters from phototrophic organisms. In vivo absorption spectra and excitation spectra of the cells revealed that these carotenoids absorbed blue‐green light and could transfer energy to chl a. These carotenoids were concluded to function as antenna pigments in P. cristatum. 相似文献

A 1089-basepair fragment (approx. 75%) of the large subunit of the chloroplast-encoded gene, ribulose-1,5-bis-phosphate carboxylase/oxygenase (rbcL), was sequenced from 16 species of the genus Pyramimonas Schmarda. Electron microscopic and biochemical studies of Pyramimonas, one of the most morphologically diverse genera within the potential sister groups to the chlorophyll a- and b-containing plants, suggest that this genus consists of at least four separate subgenera. Using the homologous sequence of rbcL from Cymbomonas tetramitiformis Schiller (Halosphaeraceae) as an outgroup and applying the maximum likelihood method, we show that the inferred topology is congruent with traditional delimitations of the taxa based on observations of periplast, internal ultrastructure, and biochemical features. A bootstrap analysis also supports division at the subgeneric level; however, the low bootstrap support associated with the deep nodes precludes resolution of these branches. A maximum likelihood relative rate test revealed that the rbcL gene in these single-celled green flagellates has a heterogeneous rate of substitution. The rbcL gene in species of the subgenus Pyramimonas has evolved at an accelerated rate relative to that of congenerics. 相似文献

A new marine microalga from the Mediterranean Sea, Crustomastix stigmatica Zingone, is investigated by means of LM, SEM, TEM, and pigment and molecular analyses (nuclear‐encoded small subunit [SSU] rDNA and plastid‐encoded rbcL). Pigment and molecular information is also provided for the related species Dolichomastix tenuilepis Throndsen et Zingone. Crustomastix stigmatica has a bean‐shaped cell body 3–5 μm long and 1.5–2.8 μm wide, with two flagella four to five times the body length. The single chloroplast is pale yellow‐green, cup‐shaped, and lacks a pyrenoid. A small bright yellow stigma is located in the mid‐dorsal part of the cell under the chloroplast membrane. An additional accumulation of osmiophilic globules is at times seen in a chloroplast lobe. Cells lack flat scales, whereas three different types of hair‐like scales are present on the flagella. The main pigments of C. stigmatica are those typical of Mamiellales, though siphonein/siphonaxanthin replaces prasinoxanthin and uriolide is absent. The pigment pool of D. tenuilepis is more similar to that of Micromonas pusilla (Butcher) Manton et Parke and of other Mamiellales. The nuclear SSU rDNA phylogeny shows that the inclusion of C. stigmatica and D. tenuilepis in the Mamiellales retains monophyly for the order. The two species form a distinct clade, which is sister to a clade including all the other Mamiellales. Results of rbcL analyses failed to provide phylogenetic information at both the order and species level. No unique morphological or pigment characteristics circumscribe the mamiellalean clade as a whole nor its two daughter clades. 相似文献

Pyramimonas mucifera sp. nov., a punctate species of the genus, is unusual both behaviorally and at the fine structural level. It forms two distinct populations in culture, one benthic and one planktonic. Planktonic forms are more conventional for the genus, but benthic forms are found in loosely packed mucilage, have flagellar rather than ciliary beating of the flagella, and display a higher degree of metaboly. Ultrastructurally this species is unusual in that it has a unique scale complement and the cells contain numerous muciferous vesicles, leaving only small pockets of cytoplasm containing the usual organelles. This species has a 3–1 type flagellar apparatus but has an additional fibrillar band, a 4–3-2–3 microtubular root system and a flexible synistosome. The discovery of a mucilage-producing species of Pyramimonas draws attention to possible links with other prasinophytes (Halosphaera) and green algae of questionable affiliation (Oltmannsiellopsis and Hafniomonas). It also provides a model of the primitive pyramimonad. 相似文献

The pigmentation of 23 species of Neospongiococcum Deason was studied in the stationary phase of nitrogen-limited cultures on agar with or without glucose or in liquid medium. Fourteen species gradually developed an orange or brown color, and in all, secondary carotenoids were detected by thin-layer chromatography. Among these, the keto-carotenoids echinenone, canthaxanthin and esterified astaxanthin were identified, although the last named was not always present. When aging on glucose agar, the remaining nine species developed only a yellow-green or yellow color and a pronounced tendency to bleach. In these, the aforementioned keto-carotenoids were lacking, and β-carotene and lutein lucre dominant. From this study it is not clear whether secondary carotenoid formation is species-specific in Neospongiococcum. 相似文献

Cell division in Pyramimonas parkeae is described and compared with some other green algae. The first indication of mitosis is division of the chloroplast, accompanied by growth of a prominent microbody, followed by replication of the 4 basal bodies. Also closely timed with this is the replication of the Golgi and other components of the basal body complex. Two basal body complexes separate, each taking a position at either pole of the nucleus which has migrated to a characteristic position just beneath the plasmalemma of a broadened and flattened flagellar pit. Cytokinesis is accomplished by the fusion of ducts and vesicles with the simultaneous release of scales to the newly formed exterior. Cells swim throughout division. 相似文献

Carotenoid compositions were analyzed for ten strains of Nephroselmis (Prasinophyceae) containing four described and three undescribed species. Based on the distribution pattern of the siphonaxanthin series, five carotenoid types were recognized in the examined strains/species: type I (N. astigmatica Inouye et Pienaar, N. pyriformis (N. Carter) Ettl, and Nephroselmis sp1. MBIC 11158) had siphonaxanthin C12:1 and C14:1 esters as well as 6′‐OH siphonaxanthin C12:1 and C14:1 esters, type II (Nephroselmis sp2. MBIC 11149) had siphonaxanthin C8:1 ester, type III (Nephroselmis sp3. NIES 486, NIES‐PS 535, and MBIC 10871) had 19‐methoxy siphonaxanthin and siphonaxanthin C12:1 and C14:1 esters, type IV (N. spinosa Suda) had only a small amount of siphonaxanthin C12:1 ester, and type V (N. olivacea Stein) had lutein as a major carotenoid but completely lacked the siphonaxanthin series. 19‐Methoxy siphonaxanthin was a novel and very unique carotenoid, that is, it contains a methoxy group and was found for the first time in photosynthetic eukaryotes. Additionally, carotenoids containing a primary methoxy group had previously never been found in any group of organisms. Siphonaxanthin C8:1 ester, which was only known as a trace carotenoid in Chlamydomonas parkeae Ettl, was first discovered as a major carotenoid in Nephroselmis sp2. (MBIC 11149). Based on these results and comparison of the phylogenetic relationships of the Nephroselmis species used, we discuss the taxonomic significance of the carotenoid types and evolutionary process of the photosynthetic antenna systems in green plants. 相似文献
