Among the Agrobacterium T-DNA genes, rolB, rolC, orf13, orf8, lso, 6b and several other genes encode weakly homologous proteins with remarkable effects on plant growth. The 6b oncogene induces tumors and enations. In order to study its properties we have used transgenic tobacco plants that carry a dexamethasone-inducible 6b gene, dex-T-6b. Upon induction, dex-T-6b plants develop a large array of morphological modifications, some of which involve abnormal cell expansion. In the present investigation, dex-T-6b-induced expansion was studied in intact leaves and an in vitro leaf disc system. Although T-6b and indole-3-acetic acid (IAA) both induced expansion and were non-additive, T-6b expression did not increase IAA levels, nor did it induce an IAA-responsive gene. Fusicoccin (FC) is known to stimulate expansion by increasing cell wall plasticity. T-6b- and FC-induced expansion were additive at saturating FC concentrations, indicating that T-6b does not act by a similar mechanism to FC. T-6b expression led to higher leaf osmolality values, in contrast to FC, suggesting that the T-6b gene induces expansion by increasing osmolyte concentrations. Metabolite profiling showed that glucose and fructose played a major role in this increase. We infer that T-6b disrupts the osmoregulatory controls that govern cell expansion during development and wound healing.  相似文献

Loss of dehydration tolerance coincides with a shift from heterotrophy to autotrophy during post-germination growth of spring wheat seedlings. This critical stage falls on the fifth day following imbibition. Till the sixth day of experiment light had no effect on dry weight of the seedlings but the survival of six day old seedlings was reduced by half upon dehydration. Germinating seeds in the presence of 5 mM glucose, fructose, mannose or sucrose did not promote seedling growth but either increase (glucose, fructose) or decreased (mannose, sucrose) the survival of dehydrated seedlings. Protection against dehydration by the former sugars was correlated, irrespective of the seedling age, with the decrease of sugar pool in seeds and increase in shoots (coleoptile and first leaf) and roots. The opposite changes were provoked by the sugars hampering seedling survival. Generally, survival of wheat seedlings was not correlated with the size of soluble sugar pool but its distribution and composition. Lower mobilisation of soluble sugars in seed, lower proportion of reduced sugars to sucrose and higher share of raffinose is characteristic for the tolerant four day old seedlings and those grown in the media containing glucose or fructose. The results presented indicate that higher proportion of reduced sugars to sucrose and lower share of raffinose in six day old seedlings seems to be associated with the loss of dehydration tolerance of these seedlings, despite heterotrophic character of growth.  相似文献

The identity, localization and physiological significance of enzymes involved in sugar uptake and accumulation were determined for endocarp tissue of pods of Kentucky Wonder pole beans (Phaseolus vulgaris). An intracellular, alkaline invertase (pH optimum, 8) was assayed in extracted protein, as well as enzymes involved in sucrose synthesis, namely, uridinediphosphate (UDP-glucose pyrophosphorylase and UDP-glucose-fructose transglucosylase). Indirect evidence indicated the presence also of hexokinase, phosphohexoseisomerase and phosphoglucomutase. The data suggested that sucrose synthesis occurred in the cytoplasm, and that both sugar storage and an alkaline invertase occurred in the vacuole. The latter functions to hydrolyze accumulated sucrose. An outer space invertase (pH optimum, 4.0) was detected, but was variable in occurrence. Although its activity at the cell surface enhanced sucrose uptake, sucrose may be taken up unaltered.

Over a wide range of concentrations of exogenous glucose the sucrose/reducing sugar ratio of accumulated sugars remained unchanged at about 20. Synthesis of sucrose appears to be requisite to initial accumulation from glucose or fructose, as free hexoses do not increase at the apparent saturating concentration for uptake. Sucrose accumulation from exogenous hexose represents a steady-state value, in which sucrose is transported across the tonoplast into the vacuole at a rate equivalent to its rate of synthesis. Evidence indicates that this component of the accumulation process involves active transport of sucrose against a concentration gradient. The ratio of sucrose/reducing sugars in the accumulated sugars immediately after a period of uptake was inversely related to the level of inner space invertase. Within 16 hours after a period of accumulation, practically all of the sugar occurs as glucose and fructose.

The absence of competition among hexoses and sucrose indicated that a common carrier was not involved in their uptake. From a series of studies on the kinetics of uptake of glucose and fructose, including competition studies, the effects of inhibitors, radioactive assay of accumulated sugars and the distribution of label in accumulated sucrose it appeared that rate limitation for glucose or fructose uptake resides in the sequence of reactions leading to sucrose synthesis, rather than in a process mediated by a carrier protein.

To gain a better understanding of the mechanism of cold induced sweetening, sugar accumulation in potato, Solanum tuberosum cv Bintje, was compared to the maximum activity of inorganic pyrophosphate (PPi):fructose 6-phosphate 1-phosphotransferase (EC 2.7.1.90) and the concentration of two regulatory metabolites. Mature tubers accumulated reducing sugars and sucrose at an almost linear rate of 13.4 and 5.2 micromole per day per gram dry weight at 2°C and 4.5 and 1.3 micromole per day per gram dry weight, respectively, at 4°C. During storage at 8°C sugar accumulation was nil. Sugar accumulation was preceded by a lag phase of about 4 days. The accumulation of reducing sugars persisted for at least 4 weeks, whereas sucrose accumulation declined after 2 weeks of storage. The ratio of glucose:fructose changed concomitantly with sugar increase from 65:35 to equimolarity. The maximum activity of PPi:fructose 6-phosphate 1-phosphotransferase was 2.51 and 2.25 units per gram dry weight during storage at 2 and 8°C, respectively. The temperature coefficient of this enzyme from potatoes kept at 2 or 8°C was 2.12 and 2.48, respectively. The endogenous concentration of fructose 2,6-biphosphate increased from 0.15 to 1 nanomole per gram dry weight during storage at 2 and 4°C but remained the same throughout storage at 8°C. After exposure to 2°C an initial increase in the concentration of PPi was observed from 4.0 to 5.6 nanomoles per gram dry weight. Pyrophosphate concentration did not change during storage at 4°C but decreased slightly at 8°C. All observed changes became annulled after transfer of cold stored tubers to 18°C. These data strongly indicate that PPi:fructose 6-phosphate 1-phosphotransferase can be fully operational in cold stored potato tubers and the lack of increase in PPi concentration supports the functioning of this enzyme during sugar accumulation.  相似文献

This study investigated the accumulation of osmotic solutes in citrus (Poncirus trifoliata) seedlings colonized by Glomus versiforme subjected to drought stress or kept well watered. Development of mycorrhizae was higher under well watered than under drought-stressed conditions. Arbuscular mycorrhizal (AM) seedlings accumulated more soluble sugars, soluble starch and total non-structural carbohydrates in leaves and roots than corresponding non-AM seedlings regardless of soil-water status. Glucose and sucrose contents of well-watered and drought-stressed roots, fructose contents of well-watered roots and sucrose contents of drought-stressed leaves were notably higher in AM than in non-AM seedlings. K+ and Ca2+ levels in AM leaves and roots were greater than those in non-AM leaves and roots, while AM symbiosis did not affect the Mg2+ level. AM seedlings accumulated less proline than non-AM seedlings. AM symbiosis altered both the allocation of carbohydrate to roots and the net osmotic solute accumulations in response to drought stress. It is concluded that AM colonization enhances osmotic solute accumulation of trifoliate orange seedlings, thus providing better osmotic adjustment in AM seedlings, which did not correlate with proline but with K+, Ca2+, Mg2+, glucose, fructose and sucrose accumulation.  相似文献

Previously, we reported that chlorogenic acid (CGA) facilitated root hair formation at pH 4.0 in lettuce (Lactuca sativa L. cv. Grand Rapids). Light was essential for this process. In the present study, we determined relationships between CGA, light, and sugar during root hair formation in lettuce seedlings. The amount of CGA increased with white light in intact seedlings. Exogenously applied CGA restored root hair formation in dark-grown intact seedlings at pH 4.0. However, no root hair formation was induced in decapitated seedlings regardless of light exposure and CGA application. Application of sucrose or glucose induced both root hair formation and CGA synthesis in light-grown decapitated seedlings at pH 4.0. Blue light was the most effective for both root hair formation and CGA synthesis when supplied with sucrose to decapitated seedlings. Addition of sucrose and CGA together induced root hair formation at pH 4.0 in dark-grown decapitated seedlings. Results suggest that light induced CGA synthesis from sugar in the roots. Sugar was also required for root hair formation other than starting material of CGA synthesis. In addition, an unknown low pH-induced factor was essential for lettuce root hair formation.  相似文献

The kinetics of substrate uptake and product formation in the process of citric acid accumulation by Aspergillus niger on sucrose as a sole carbon source are presented. The experiments are aimed at studying if glucose and fructose obtained from the hydrolysis of sucrose are equivalent carbon sources for A. niger and how the presence of the two different carbon substrates might influence the citric acid formation process. Beet sugar was used as a sole carbon source in the first series of experiments conducted in two types of bioreactors: stirred tank and air-lift. The fructose uptake rate was significantly lower than the glucose uptake rate in the late idiophase. A substrate utilisation breakpoint occurred when a large amount of citric acid was accumulated in the fermentation broth. A similar phenomenon was also detected in repeated fed-batch fermentation. This phenomenon was confirmed by the second series of parallel shake culture runs, in which fungal growth and citric acid accumulation by A. niger was simultaneously tested on the media containing the following carbon sources: sucrose, glucose and fructose, with and without addition of concentrated citric acid solution. Finally, it was shown that high concentration of citric acid strongly depleted fructose uptake rate.  相似文献

The effects of increasing concentrations of nickel sulfate, NiSO4 (200 and 400 μM) in the growth medium on the content of starch and sugars and activity levels of enzymes involved in starch and sugar metabolism were examined in seedlings of the two Indica rice cvs. Malviya-36 and Pant-12. During a 5–20 day growth period of seedlings in sand cultures, with Ni treatment, no definite pattern of alteration in starch level could be observed in the seedlings. In both roots and shoots of the seedlings Ni treatment led to a significant decrease in activities of starch degrading enzymes α-amylase, β-amylase, whereas starch phosphorylase activity increased. The contents of reducing, non-reducing, and total sugars increased in Ni-treated rice seedlings with a concomitant increase in the activities of sucrose degrading enzymes acid invertase and sucrose synthase. However, the activity of sucrose synthesizing enzyme sucrose phosphate synthase declined. These results suggest that Ni toxicity in rice seedlings causes marked perturbation in metabolism of carbohydrates leading to increased accumulation of soluble sugars. Such perturbation could serve as a limiting factor for growth of rice seedlings in Ni polluted environments and accumulating soluble sugars could serve as compatible solutes in the cells under Ni toxicity conditions.  相似文献

Suspension cultures of Coleus blumei were characterized with respect to growth and rosmarinic acid formation in media with different sugars and various sugar concentrations. Sucrose is the sugar with the highest stimulating effect on growth and rosmarinic acid accumulation, followed by glucose and fructose. The sugar alcohol mannitol cannot be metabolized by the plant cells. Sucrose is cleaved into glucose and fructose by the Coleus cells. Sucrose concentrations from 1 to 5% have an increasing positive effect on growth and rosmarinic acid synthesis in the cell cultures with a maximum rosmarinic acid content of 12% of the dry weight in medium with 5% sucrose; in medium with 6% sucrose rosmarinic acid accumulation obviously did not reach its highest level in the culture period of 14 days. A very high yield of rosmarinic acid (2 mg ml-1 suspension) could also be achieved by maintaining a sucrose concentration of 2% during the whole culture period. The start of rosmarinic acid synthesis by the cell cultures seems to be regulated by the growth limitation when a nutrient, e.g. phosphate is depleted from the medium. The growth of excised tomato roots in sucrose is accompaniedby the appearance in the medium of glucose and fructose. Thequantitative relations between this appearance of glucose andfructose in the medium, total sugar absorption by the roots,and the decrease in the sucrose concentration of the mediumdo not suggest any causal relationship between sucrose uptakeand glucose and fructose appearance in the medium. Excised tomato roots exhibit surface invertase activity witha pH optimum at 4.0–4.4. Alterations of external pH, whichdid not affect sucrose absorption, drastically altered the levelsof glucose and fructose appearing in the medium. Glucose is preferentially absorbed from mixtures of glucoseand fructose, and by adjustment of the ratio of the two sugars,a mixture can be obtained from which equimolar absorption ofthe two sugars occurs. Root growth in this mixture is, however,very poor compared with that occurring in presence of sucrose. The results are discussed in the light of earlier studies onsucrose uptake by cultured tomato roots.  相似文献

Sugars supplied to germinating seedlings of maize (Zea mays L.) regulate the secretion of polysaccharides by the outer cells of the root cap. The polysaccharide secreted by these cells adheres to the root tip as a droplet and the size of the droplet was used to quantitate polysaccharide secretion. The polysaccharide contains glucose, galacrose, and galacturonic acid residues with smaller quantities of mannose, arabinose, xylose, fucose and rhamnose. These sugars supplied to maize seedlings had marked effects on the rate of polysaccharide secretion by root tips. The effects on secretion were independent of the growth rates of the roots. Glucose, fucose and xylose increased droplet size 1.5–2 fold (as did sucrose, maltose, lacrose, fructose and ribose) whereas galactose, arabinose and galacturonic acid were inhibitory. Mannose increased dropler size 5–7 fold. The marked effect of mannose on polysaccharide secretion was due to an increased rate of secretion combined with a longer phase of extrusion of polysaccharide into the forming droplet. The effect of mannose was partially reversed by inorganic phosphate and other sugars (except for fucose which had no effect or promoted secretion in the presence of mannose). In contrast to sucrose, mannose stimulated secretion in a maize variety having a high sugar endosperm (high endogenous sugar). The results suggest that regulation of secretion by mannose is due to an alteration of normal sugar metabolism; whereas stimulation of secretion by sucrose and other sugars may be due to an increased availability of sugars for metabolism.  相似文献

植物根系如何响应环境因子变化是植物发育和营养吸收研究的重要科学问题。丙酮酸激酶OsPK1在根部的表达主要在根尖成熟区和根毛区,其表达水平变化有可能影响水稻对外源糖分的吸收。采用日本晴和水稻突变体ospk1,通过改变1/2 MS培养基中蔗糖含量,探索水稻幼苗对外源糖分的吸收和响应。通过GC-MS的方法检测了水稻幼苗叶片、叶鞘和根中蔗糖、葡萄糖、果糖和半乳糖的含量。发现根与培养基中糖分接触能明显提高幼苗中的糖含量。并且这些幼苗的根系长度大于那些不加蔗糖的培养基培养的幼苗,表明外源糖分被吸收后能促进根的伸长。OsPK1表达下调影响了糖代谢和外源糖分的吸收。半定量RT-PCR结果显示,幼苗根与糖分的直接接触明显上调根中OsPIP2;4,OsPIP2;5和OsTIP2;1三个水孔蛋白基因的表达。  相似文献

以‘台农1号’芒果为材料,测定了果实生长发育过程中淀粉、蔗糖、葡萄糖和果糖含量以及淀粉酶、蔗糖代谢相关酶———酸性转化酶(AI)、中性转化酶(NI)、蔗糖合成酶(SS)和蔗糖磷酸合成酶(SPS)的活性,并对果实中糖组分与酶活性的关系进行了分析.结果显示,(1)台农1号芒果果实属于单S型生长曲线,发育前期主要积累淀粉、葡萄糖和果糖,果实成熟软化时,淀粉酶活性降至最低,淀粉水解,蔗糖快速积累.(2)酸性转化酶活性在果实整个发育过程中维持最高,完熟时略有降低;蔗糖磷酸合成酶在果实发育前期略有降低,完熟时升至最高;蔗糖合成酶和中性转化酶活性在整个发育期一直很低且较稳定.(3)淀粉含量与淀粉酶活性呈显著正相关,与SPS活性呈极显著负相关,蔗糖、葡萄糖含量均与SPS、SS呈显著、极显著的正相关;果糖含量与SS呈极显著的正相关.研究表明,芒果成熟时淀粉分解、酸性转化酶活性的降低,且蔗糖合成酶和蔗糖磷酸合成酶活性的增加是引起果实蔗糖积累的主要因子.  相似文献

As osmolytes and signaling molecules, soluble sugars participate in the response and adaptation of plants to environmental stresses. In the present study, we measured the effect of chilling (12 °C) stress on the contents of eight soluble sugars in the leaves, cotyledons, stems, and roots of Jatropha curcas seedlings, as well as on the activities of eight rate-limiting enzymes that are critical to the metabolism of those soluble sugars. Chilling stress promoted both starch hydrolysis and soluble sugar accumulation. The soluble sugar contents of the leaves and cotyledons were affected more than that of the stems and roots. Meanwhile, the activities of the corresponding metabolic enzymes (e.g., β-amylase, uridine diphosphate glucose phosphorylase, and sucrose phosphate synthase) also increased in some organs. The gradual increase of soluble neutral alkaline invertase activity in the four studied organs suggested that sucrose catabolic production, such as glucose and fructose, was especially important in determining resistance to chilling stress and hexose signal transduction pathway. In addition, the substantial accumulation of raffinose family oligosaccharides and increase in corresponding metabolic enzyme activity suggested that galactinol and raffinose play an important role in determining the chilling resistance of J. curcas. Together, these findings establish a foundation for determining the relationship between the chilling resistance and soluble sugar accumulation of J. curcas and for investigating the mechanisms underlying sugar signaling transduction and stress responses.  相似文献

13C NMR was used to detect sugars in detached fibrous and storage roots of carrots ( Daucus carota L. cv. Chantenay Red-Cored). The relative amounts of sucrose, glucose and fructose in storage roots of two ages and in fibrous roots were similar to those detected by a destructive, enzymatic sugar determination method. In tap roots all three sugars were present with glucose being marginally predominant in 8-week-old roots and sucrose being the major sugar present in 12-week-old roots. Glucose was the major sugar present in fibrous roots with small amounts of sucrose and no fructose detected. The results indicate that with some instrumental modification, 13C NMR could be used as a non-destructive method for measuring sugar levels in intact storage root systems during development.  相似文献

The effects of three conditions likely to affect soluble carbohydrate pools, namely drought, expression of barley sucrose: fructan 6-fructosyl transferase (6-SFT, EC 2.4.1.10) and the establishment of the arbuscular mycorrhizal symbiosis with Glomus mosseae were studied in a multifactorial experiment using tobacco ( Nicotiana tabacum ). Tobacco, a plant naturally unable to form fructan, accumulated fructan in leaves, and to a larger extent in the roots, when transformed with 6-SFT. Under drought conditions, growth was considerably reduced, but neither expression of 6-SFT nor mycorrhiza formation had an effect on growth rate. However, in response to drought, carbon partitioning was significantly altered towards accumulation of soluble sugars. In plants exposed to drought, pools of sucrose were greater than those of unstressed plants, particularly in their roots. In the transgenic plants expressing 6-SFT, there were also increased contents of the products of 6-SFT, namely fructan, most probably because of the increased availability of the substrate, sucrose. These effects were the same in the presence or absence of mycorrhiza. Hexoses (glucose and fructose) also increased in response to drought, primarily in the leaves. This effect of drought was little affected by the expression of 6-SFT, except that it slightly enhanced drought-induced glucose accumulation in roots. However, the presence of mycorrhiza led to a considerable reduction in drought-induced accumulation of hexoses in the leaves. The content of the fungal disaccharide trehalose was greatly increased in the roots of all mycorrhizal plants upon exposure to drought, particularly in some of the transgenic plants expressing 6-SFT.  相似文献

Red beet hairy root cultures, obtained after genetic transformation with Agrobacterium rhizogenes, are completely heterotrophic and synthesize betalaines (BNs). Upon subjecting the hairy roots to treatments containing different sugars (3% w/v) it was found that sucrose was rapidly utilized, followed by maltose, and a very limited use of glucose, but the other hexoses – fructose, lactose, xylose and galactose or glycerol totally suppressed both growth and BN synthesis. No habituation or adaptability to maltose or glucose occurred, evidenced by the lack of growth upon re-culture in respective medium. Glycerol, was not taken up alone, but was utilized to a considerable extent in the presence of low levels of sucrose for growth only but not BN synthesis. Red beet hairy root culture did not exogenously hydrolyse sucrose to hexoses, as there were only traces of reducing sugar present in the medium soon after inoculation, without an increase later, confirmed by HPLC. There was an increase in medium osmolarity in the presence of fructose indicating the exudation of certain compounds from the roots. Red beet hairy roots appear useful as a model system to study sugar metabolism/signalling due to their sensitivity to different sugars that may directly link to morphological changes and BN synthesis.  相似文献

河套蜜瓜果实发育过程中糖积累与蔗糖代谢相关酶的关系   总被引:3,自引:0,他引:3  
以河套蜜瓜为试材,采用外部形态观测与内部生理指标测定相结合的方法,对其果实发育过程中果实生长模式以及果实中蔗糖、果糖、葡萄糖和淀粉含量以及蔗糖代谢相关酶活性进行测定,以揭示河套蜜瓜果实生长发育过程中糖的代谢积累与相关酶的关系.结果显示:(1)河套蜜瓜果实生长速率呈单"S"曲线,果实发育早期以积累葡萄糖为主,进入成熟期后蔗糖积累量迅速增加,最终由蔗糖和己糖共同构成果实品质.(2)在河套蜜瓜果实成熟期前,蔗糖磷酸合成酶(SPS)活性维持较低水平,进入成熟期后,SPS活性迅速升高;蔗糖合成酶(SS)活性在成熟期前为分解活性大于合成活性,成熟期后表现为合成活性大于分解活性;在整个果实发育期,酸性转化酶(AI)活性较低,中性转化酶(NI)活性始终高于AI.(3)在果实整个发育期,蔗糖含量与蔗糖代谢酶的净活力呈极显著正相关,蔗糖代谢相关酶共同作用决定果实中蔗糖含量.研究表明,在河套蜜瓜果实发育前期,以蔗糖分解代谢为主,且蔗糖合成酶和中性转化酶是催化蔗糖分解的关键酶;果实成熟期间,蔗糖代谢转为合成方向为主,蔗糖合成酶和蔗糖磷酸合成酶在蔗糖积累中起主导作用.  相似文献
